The main objective of this research was the study of the soil nematode community, and in particular plant parasitic nematodes (PPN), from a field located in Portugal’s southern region, used for sugarbeet production. The study was performed from February to July 2003, covering part of the fallow period previous to tomato cultivation, the alternative crop in the rotation. The end of the fallow period in March and the soil preparation period in May were marked by a significant reduction in the numbers of PPN, whereas their numbers increased on the following tomato crop. The genus Helicotylenchus stood out as the most representative group, forming 90% of all PPN counted each month. The genus Heterodera was relatively abundant in the months following the previous sugarbeet crop, and numbers of the genus Meloidogyne increased during the tomato crop. The correlations between these group and environmental parameters show that, apart from the direct influence of the host, pH, organic matter, temperature and soil moisture significantly influenced nematode abundance and community composition.

Indústrias metalúrgicas de produção de ligas metálicas podem por em risco toda a área envolvente, nomeadamente a qualidade das águas (superficiais e subterrâneas), dos solos e do ar, sempre que não existam infraestruturas e planos adequados de gestão destes resíduos. No caso em estudo, a unidade industrial situa-se na bacia do rio São Francisco, no estado brasileiro de Minas Gerais, e nos mais de quarenta anos de funcionamento tem produzido inúmeros problemas ambientais. Este estudo baseia-se nos primeiros resultados referentes aos solos da envolvente, tendo como objetivos a identificação dos principais contaminantes e a definição da área contaminada, de forma a avaliar a eficácia de projetos futuros de recuperação.

Mining and metallurgical industries are the most responsible for heavy metal contamination. These contaminants are often associated to poor management strategies and the lack of suitable containment areas. This is the case of one of the largest metal alloy producers in the São Francisco river basin, in Minas Gerais state, Brazil. During operation, the tailings of this activity have been accumulated in the surrounding area, leading to heavy metal contamination (Mn, Cu, Zn, Ni, Pb, Cd, Cr and As), much above the critical levels, which accumulate in the sediments of the nearby waterways, in the soils of the drainage area, and in the water column. In this work we are going to discuss both the contamination level and its extent in the Consciência river, a small tributary of the São Francisco river, the longest river entirely Brazilian, by analyzing the geochemistry and mineralogy of the accumulated sediments. It will be also discuss the reclamation strategies of this site and the impact on the environment.

O presente trabalho é constituído por uma revisão bibliográfica e uma série de estudos científicos experimentais visando a melhoria do conhecimento da ecologia de germinação e resposta ao estresse hídrico em arbustos autóctones. Estes estudos pretendem contribuir para o conhecimento da ecologia de espécies autóctones na fase de germinação e estabelecimento inicial das plântulas através da comparação da resposta de sementes de diferentes proveniências. Os primeiros três capítulos focam-se no estudo dos efeitos de pré-tratamentos na germinação de espécies arbustivas com diferentes estratégias de regeneração e dormência da semente, enquanto os dois últimos capítulos incidem sobre o efeito de diferentes condições de disponibilidade de água na germinação e estabelecimento inicial de plântulas com potencial para restauro ecológico. Este estudo procura abordar aspetos importantes para o estudo e validação do uso de espécies alvo para o restauro ecológico em zonas Mediterrânicas, contribuindo deste modo para a melhoria do conhecimento da ecologia destas espécies. A germinação foi estimulada por pré-tratamentos de calor em quatro das cinco espécies cuja época de dispersão da semente na primavera-verão. As sementes provenientes do sul tenderam a germinar melhor, sendo ao mesmo tempo mais pequenas. Por outro lado, a germinação em espécies cuja dispersão da semente ocorre no outono-inverno variou de acordo com o tipo de dormência da semente. A germinação foi em geral favorecida pela estratificação a frio na espécie apresentando dormência fisiológica mas foi negativamente afetada na espécie que não apresenta dormência. Esta resposta não foi, no entanto, inequívoca pois foi dependente da proveniência estudada. O efeito da estratificação a frio teve em geral um efeito mais positivo ou menos negativo nas sementes provenientes do norte em comparação com as outras proveniências. O stress-osmótico teve um efeito negativo consistente na germinação de Pistacia lentiscus e outro fator que também afetou significativamente a germinação foi a variabilidade intra-populacional entre plantas mãe. As duas proveniências apresentaram diferentes graus de associação entre a germinação e/ou caracteres morfológicos da semente ou planta mãe. Quanto à fase de estabelecimento inicial, o baixo conteúdo em água teve um claro efeito negativo no investimento relativo em biomassa acima do solo e um efeito positivo no investimento relativo em biomassa abaixo do solo em Arbutus unedo. Esta resposta não foi inequívoca, uma vez que as proveniências diferiram em adaptações morfológicas ao baixo conteúdo em água. As plântulas da proveniência mais húmida revelaram uma taxa de crescimento relativo superior mas, ao mesmo tempo, uma fraca adaptação às condições de baixa disponibilidade de água, quando comparadas com as outras proveniências. Ao contrário, as plântulas da proveniência seca com verão mais quente apresentaram semelhantes diâmetro do colo da raiz, peso seco de folhas e performance fisiológica sob os dois regimes de irrigação. Os resultados obtidos revelam diferenças significativas na germinação e desenvolvimento inicial entre as distintas proveniências que poderão estar relacionadas com adaptações ao clima dos locais de origem. Os desenvolvimentos futuros do estudo dos mecanismos subjacentes às adaptações observadas poderiam contribuir para a melhoria das previsões de sucesso do estabelecimento inicial em diferentes populações de plantas, permitindo aumentar a confiança e efetividade de custos nas decisões relacionadas com ações de restauro ecológico em cenários de alterações climáticas.

Extensive green roofs are becoming a popular tool for restoring green infrastructure in urban areas, particularly biodiverse habitats such as post-industrial/brownfield sites. This study investigated the use of six recycled lightweight aggregates and combinations of them in green roof growing substrate, to determine their effectiveness for enhancing plant abundance and species diversity. In two separate experiments, we examined the roles of substrate type and depth on the establishment of a perennial wildflower mix over a 15-month period. We found that some of the alternative substrates are comparable to the widely used crushed red brick aggregate (predominantly found in commercial green roof growing substrate) for supporting plant establishment. For some materials such as clay pellets, there was increased plant coverage and a higher number of plant species than in any other substrate. Substrates that were produced from a blend of two or three aggregate types also supported higher plant abundance and diversity. Generally, increasing substrate depth improved plant establishment, however this effect was not consistent across substrates. We conclude that recycled materials may be viable constituents of growing substrate for green roofs and they may improve green roof resilience, through increased plant cover and diversity. The results could provide evidence to support the construction of mosaic habitat types on single roofs using various substrate blends.

Dissolved oxygen concentration is one of the most limiting factors in aerobic cultures, due to the poor solubility of oxygen in aqueous media. In many processes, the microorganisms growth and production can be affected as a result of insufficient oxygen supply to the broths [1, 2]. To increase oxygen solubility, some methods can be used, such as the increment of aeration or agitation rates or decrease of the solution temperature.

Grapevine leafroll disease (GLRD) is one of the most important virus diseases of grapevines worldwide, causing major economical impact. The disease has a complex aetiology and currently eleven phloem-limited viruses, termed in general Grapevine leafroll-associated virus (GLRaVs), have been identified. Two of the GLRaVs, GLRaV-1 and GLRaV-3, are included in the European certification scheme of propagation material. However, the flawed notion that GLRaV-3 is more frequent than GLRaV-1 and that all other GLRaVs are possibly not as relevant for GLRD, has until now precluded the development of specific serological and molecular detection assays and limited the scope of molecular characterization of the viruses known to be associated with the disease. Hence, few studies have addressed the phylodynamics of GLRaVs or even characterized the genetic structure of their natural populations. This generalized lack of molecular information, in turn underlie the deficient capacity to detect the viruses. The phylogenetic analyses were conducted on the basis of the heat shock protein 70 homologue (HSP70h) and the coat protein (CP) genes for GLRaV-1 and the HSP70h, the heat shock protein 90 homologue (HSP90h) and the CP genes for GLRaV-5. The data obtained for GLRaV-1 contributed 83 new CP sequences. This information was combined with previous analysis by other authors and used for the production of new polyclonal IgG, capable of detecting CP variants from all the phylogroups observed. Successful testing of this new tool included tissue print immunoblotting (TPIB) and in situ immunoassay (ISIA). The data obtained for GLRaV-5, contributed 61 new CP and 28 new HSP90h gene sequences. Eight phylogenetic groups were identified on the basis of the CP. Characterization of the genetic structure of the isolates revealed a higher diversity than previously reported and allowed the identification of dominant virus variants. For both GLRaV-1 and GLRaV-5, the effect of vegetative propagation on the virus transmission dynamics was addressed.

Total phenol, hydroxycinnamic acid derivatives, flavone/flavonol and flavanones/dihydroflavonol contents of hydro-alcoholic extracts, obtained by sonication, from the aerial parts of Artemisia campestris L., Anthemis arvensis L., Haloxylon scoparium Pomel, Juniperus phoenicea L., Arbutus unedo L., Cytisus monspessulanus L., Thymus algeriensis Boiss et Reut, Zizyphus lotus L (Desf.) collected in Djebel Amour (Sahara Atlas, Algeria) were quantified by spectrophotometric methods. The chemical composition of the essential oils obtained by hydrodistillation from Artemisia campestris L. and Juniperus phoenicea I aerial parts were also evaluated by gas chromatography (GC) and gas chromatography coupled to mass spectrometry (GC-MS). The antioxidant activity of the extracts and essential oils was assessed measuring the capacity for preventing lipid peroxidation using two lipidic substrates (egg yolk and liposomes), the capacity for scavenging DPPH, ABTS, superoxide anion radicals, hydroxyl radicals and peroxyl radicals. Anti-inflammatory activity was assessed by measuring the capacity for inhibiting lipoxygenase. Reducing power and chelating capacity were also assayed. The results showed different amounts of total phenols depending on the method used: A. campestris extract had the highest levels of total phenols when the measurement was made at lambda = 280 nm, whereas H. scoparium and A. unedo extracts showed the highest levels of total phenols with Folin-Ciocalteau. C. monspessulanus had the highest levels of flavones/flavonols and flavanones/dihydroflavonols. The essential oils of A. campestris and J. phoenicea were mainly constituted by alpha-pinene, beta-pinene and sabinene; and a-pinene, respectively. The methods used for assaying the capacity for preventing lipid peroxidation revealed to be inadequate for extracts due to the great interferences detected. The essential oils were more active than the generality of extracts for scavenging peroxyl radicals and for inhibiting lipoxygenase, whereas A. unedo extract was the most active for scavenging ABTS, DPPH, superoxide anion radicals and it also had the best reducing capacity. In a general way, the great majority of the antioxidant activities correlated well with the phenol content although such correlation was not so clear with the flavonoid content. The cell cycle comprise the four phases of, G1, S-phase, G2 and mitosis. Two critical transitions are G1/S and G2/M; the latter is regulated by WEE1 kinase and CDC25 phosphatases. The scope of this thesis was to investigate the regulation of the G2/M transition of the cell cycle by WEE1 and CDC25, and how these genes interface with plant growth regulators in Arabidopsis thaliana. In Arabidopsis roots, the frequency of lateral roots was found to be increased by ectopic expression of Schizosaccharomyces pombe (Sp)cdc25e and reduced by Arath;WEE1 expression. I examined the effect of Arath;WEE1 and Spcdc25 on induction of shoots and roots in Arabidopsis hypocotyls in vitro. Hypocotyl explants from two over-expressing WEE1 lines , three T-DNA insertion lines and two expressing cdc25 (Spcdc25e) lines together with wild type (WT) were cultured on two-way gradients of kinetin (Kin) and naphthyl acetic acid (NAA). Below a threshold concentration of NAA (100 ng ml-1), WEE1 repressed morphogenesis in vitro, whereas at all NAA/Kin combinations Spcdc25 promoted morphogenesis (particularly root formation) over and above that in WT. Loss of function wee1-1 cultures were very similar to WT. Quantitative data indicated a significant increase in the frequency of root formation in Spcdc25e cultures compared with WT particularly at low Kin concentrations, and WEE1oe’s repressive effect was overcome by NAA but not Kin. In conclusion, WEE1 has a repressive effect on morphogenesis in vitro that can be overcome by auxin whereas Spcd25 by-passes a cytokinin requirement for the induction of morphogenesis in vitro. The role of CDC25 and WEE1 in DNA damage responses was also analysed. Two over-expressing Arath;CDC25 lines and T-DNA mutants showed no difference to WT either in standard conditions or zeocin-supplemented treatments. However, root length was longer in Arath;CDC25oe lines treated with hydroxyurea (HU) and lateral root number was increased compared to WT. This suggests a differential response of Arath;CDC25oe in the DNA replication (HU-induced) and DNA damage (zeocin-induced) checkpoints (Chapter 5). Finally the roles of WEE1 and CDC25 in cell cycle regulation were examined using tobacco TBY-2 cell cultures expressing Arath;WEE1, Nicotiana tabacum (Nicta)WEE1 or Arath;CDC25. Whilst Nicta;WEE1 lengthened G2 of the cell cycle, Arath;WEE1 had an unusual effect of shortening G2 phase and Arath;CDC25 had no observable effect (Chapter 6).

Oomycetes form a deep lineage of eukaryotic organisms that includes a large number of plant pathogens that threaten natural and managed ecosystems. We undertook a survey to query the community for their ranking of plant pathogenic oomycete species based on scientific and economic importance. In total, we received 263 votes from 62 scientists in 15 countries for a total of 33 species. The Top 10 species and their ranking are: (1) Phytophthora infestans; (2, tied) Hyaloperonospora arabidopsidis; (2, tied) Phytophthora ramorum; (4) Phytophthora sojae; (5) Phytophthora capsici; (6) Plasmopara viticola; (7) Phytophthora cinnamomi; (8, tied) Phytophthora parasitica; (8, tied) Pythium ultimum; and (10) Albugo candida. The article provides an introduction to these 10 taxa and a snapshot of current research. We hope that the list will serve as a benchmark for future trends in oomycete research.

In this study the inhalation doses and respective risk are calculated for the population living within a 20 km radius of a coal-fired power plant. The dispersion and deposition of natural radionuclides were simulated by a Gaussian dispersion model estimating the ground level activity concentration. The annual effective dose and total risk were 0.03205 mSv/y and 1.25 x 10-8, respectively. The effective dose is lower than the limit established by the ICRP and the risk is lower than the limit proposed by the U.S. EPA, which means that the considered exposure does not pose any risk for the public health.
