Under greenhouse conditions, Epidendrum nocturnum Jacq. plants produce fruits by both self-fertilization and cleistogamy. Although adapted to these reproductive processes the species respond also to cross-pollination. Seeds without embryos and with one embryo are usual but occasionally seeds with two, three or four embryos are produced. Multiple embryos are formed by polyembryony and apomixis. © 1985 Annals of Botany Company.

Superovulation was induced in 15 Nelore cows with porcine follicle stimulating hormone (FSH-P) or pregnant mare serum gonadotropin (PMSG), and embryos were collected to compare the response of this breed of cattle to these hormones. FSH-P was given in 30-and 40-mg doses for 5 d as a single daily injection or fractionated into two daily injections. PMSG was given in doses of 1000 and 2000 IU. The animals were killed immediately after embryo collection and the ovaries and genitalia were examined clinically. PMSG proved to be more effective in inducing superovulation than FSH-P, probably because PMSG caused no stress since it was administered as a single dose. No differences were observed between the 30-and 40-mg dose of FSH-P or between the application as a single or fractionated dose. Differences did occur, however, between the number of ovulations and embryos obtained at each collection. On the basis of postmortem analysis, we concluded that lack of egg uptake by the infundibulum had occurred in cases of increased ovulation, with excessive increase in volume of the ovary. We also recommend using smaller doses of FSH-P and suggest that avoiding stress in handling is essential for a good response to hormonal stimulation by Zebu cattle. © 1986.

In negatively photoblastic seeds of Cucumis anguria L. the scarification of micropyle overcame the inhibitory effect of white light , although far-red radiation inhibited the germination in both intact and scarified seeds. The moistened testa powder transmitted more far-red than red radiation. Thus the testa could act as a filter of radiation and maintain a high far-red/red ratio at the photosensitive site of the embryo, which must be localized near the micropyle. © 1988 Academia.

Bovine interferon-alpha I1 (bIFN-alpha) may be useful for enhancing fertility in sheep and cattle because it has extensive sequence homology with ovine and bovine trophoblast protein-1 and, like those proteins, extends corpus luteum lifespan. To test the effectiveness of bIFN-alpha to enhance fertility, several experiments were performed in which inseminated heifers were given i.m. injections of bIFN-alpha approximately at the time of embryo-mediated signals that result in maintenance of the corpus luteum. In Exp. 1, heifers given 20 mg of bIFN-alpha daily from d 14 to 17 tended (P less than .07) to have lower pregnancy rates at d 110 to 112 of gestation (36/75; 48% vs 43/72; 60%). Similar results were obtained in Exp. 2 when heifers received a single injection of 40 mg of bIFN-alpha or placebo at d 13 after estrus; pregnancy rates at d 42 were 39/104 (38%) for bIFN-alpha and 47/98 (48%) for placebo. In Exp. 3, heifers were given gradually increasing doses of bIFN-alpha or placebo from d 11 to 19, because such a regimen had been shown to reduce the number of heifers experiencing hyperthermia after bIFN-alpha injection. Pregnancy rates were 42/95 (44%) for bIFN-alpha and 62/111 (56%) for placebo. Across all three experiments, pregnancy rates were lower (P less than .01) for heifers treated with bIFN-alpha (117/274; 43%) than for heifers treated with placebo (152/281; 54%). In conclusion, these results demonstrate that, under the administration systems used, bIFN-alpha does not increase pregnancy rate, but rather tends to reduce it.

Murine and bovine embryos were sexed by using polyclonal and monoclonal anti-H-Y antibodies in cytotoxic and immunofluorescence assays. The specificity of 3 different sources of antibodies are compared. Sexing was accomplished by complement-mediated cytotoxicity or by H-Y antigen detection using a anti-mouse Ig fluorescein conjugate. Cytogenetic ascertainment for correct sex identification yielded a score of about 85% in both species, while the immunofluorescence assay yielded a score of 83%. © 1993.

Maternal alcoholism (ethanol and sugar cane brandy) during gestation induces delayed cellular growth and differentiation in fetal rat palate epithelium, with increased nuclear, cytoplasmic and cellular volumes, increased epithelial and keratin thickness and decreased cellular numerical density.

The influence of fetal calf serum alone (FCS) or associated with proestrous (FCS+PCS), estrous (FCS+ECS) or metaestrous (FCS+MCS) cow serum added to the culture medium and of the steroids produced by co-cultured granulosa cells were evaluated in terms of the in vitro maturation (IVM) and fertilization (IVF) of bovine oocytes. Supplementation of the medium with FCS+ECS and FCS+MCS resulted in higher proportions of oocytes that reached metaphase II (96.0% and 93.3%, respectively) and in higher proportions of embryos that reached the four- and eight-cell/morula stages (51.9% and 65.6%, respectively), whereas the supplementation with FCS and FCS+PCS resulted in only 79.2% and 67.5%, respectively, of matured oocytes and 26.7% and 34.3%, respectively, of cleaved embryos. These findings show that the best IVM and IVF were obtained at lower concentrations of estradiol produced by co-cultured granulosa cells (supplementation with FCS+ECS: 10.3 ng/ml and FCS+MCS: 2.1 ng/ml), whereas the worst results in IVM and IVF occurred at higher concentrations of estradiol that were obtained with FCS (33.1 ng/ml) and FCS+PCS (19.9 ng/ml) supplementation. These data suggest an inhibitory effect of estradiol on resumption of oocyte meiosis in vitro.

Mast cells are present in the eye of chick embryos from the 14th day onward, displaying metachromatic granules, mainly in the iris anterior surface and pectinate ligament. Ultrastructurally these cells show electron-dense granules and a few thin and short cytoplasmic projections in close contact with fibroblasts. Sometimes these contacts are extensive, with long fibroblast projections partially involving the mast cells. Gap junctions between mast cells and fibroblasts are observed only in the eyes of 16- and 20-day-old embryos. These intercellular specializations are represented by a close apposition of cytoplasmic membranes with an extension up to 300 nm. Gap junctions between mast cells and fibroblasts were not observed previously in vivo or in vitro, although in vitro studies have shown that a number of functionally critical interactions may occur between these cells. Our morphological findings suggest that, in vivo, fibroblasts interact with mast cells and may influence their maturation.

Six or 7-day-old equine embryos were divided into 4 groups; Group 1, n = 15, Day 7 embryos destined for immediate transfer; Group 2, n = 15, Day 6 embryos destined for deep-freezing with glycerol plus sucrose as cryoprotectant; Group 3, n = 10, Day 6 embryos destined for deep-freezing with glycerol plus 1,2-propanediol as cryoprotectant and Group 4, n = 3, fresh embryos destined for ultrastructural analysis. All the frozen/thawed embryos were transferred to recipient mares, except 3 embryos in Group 3 that were subjected to ultrastructural analysis. After thawing the cryoprotectants were removed by successive dilutions in PBS + 15% v:v fetal calf serum (FCS) containing decreasing concentrations of the cryoprotectants. Pregnancy was diagnosed ultrasonographically in 53.3%, 13.3% and 0% of the mares in Groups 1, 2 and 3 respectively. Ultrastructural analysis showed differences between frozen/thawed and fresh embryos. In the former, embryonic cells were deformed and showed dilation of the intercellular and perivitelline spaces, a decrease of desmosome number in the junctional complexes, few microvilli on the apical surface of the trophectoderm and an almost total absence of pinocytotic vesicles. Most of the mitochondria showed regions containing dilation and irregularities on the cristae, which appeared electron-dense. The results obtained with Groups 2 and 3 embryos showed that the cryoprotectants employed were not effective in protecting the embryos against damage during freezing and thawing. Indeed, the ultrastructural changes observed in the Group 3 embryos explained the absence of any established pregnancies in this group of mares.

The objective of this study was to estimate the relative effects of genetic and phenotypic factors on the efficacy and efficiency of superovulation for Holstein-Friesian cows reared in Brazil. A database, established by the Associacao Brasileira de Criadores de Bovinos da Raca Holandesa, consisting of a total of 5387 superovulations of 2941 cows distributed over 473 herds and sired by 690 bulls was used for the analysis. The records were analyzed by MTDFREML (Multiple Trait Derivative-Free Restricted Maximum Likelihood), using a repeatability animal model. The fixed effects included in the model were contemporaneous group (veterinarian, herd, year and season of the superovulation); number of semen doses; cow age; and superovulation order. The estimated repeatability of the number of the transferable embryos was low (0.13), and the estimated heritability was 0.03. These results indicate that environmental factors play a critical role in the response of a cow to a superovulation treatment. There is little evidence that future responses to superovulation by individual females can be predicted by previous treatment(s) or that superovulation response is an heritable trait.

Guazuma ulmifolia is as popular reforestation tree all over Latin America. It is characteristic of the initial stages of the secondary sucession and presents potential utility in the restoring of degraded areas. There is no information about fruit, seed and seedling morphology, which is of fundamental importance for identification, extraction, management and seed germination as well as for the characterization of post-seminal development and normal seedling pattern. To obtain such information, external fruit, and external and internal seed structures were studied considenng shape, size, micropile and embryo localization, and tegumentar structures. All stages of this work were conduced in the Universidade Estadual Paulista (UNESP), Campus of Jaboticabal city. The fruits were collected in a mixed plantation in Jaboticabal city, State of São Paulo, Brazil. For the biometric study eight repetitions of ten fruits and eight repetitions of 100 seeds were utilized. For seed internal traits study, 50 seeds were drenched in a distiled water, cut, and observed with a scanning electron microscope and a stereomicroscope. For post-seminal study ten repetitions of seven seeds were scarificated chemically with sulphuric acid during 50 min, and placed to germinate in a culture medium, at 30°C, and eight hours of photoperiod. We found elipsoid, woody, indehiscent, pentacarpelar fruits, with a mean lenght of 22.61 mm (diameter 24.88 mm) and 64.0 seeds per fruit. Seed shape varies, mean length is 3.07 mm (width of 2.36 mm).The seed is bitegumented, tegmic, with a continuous, axial and curved embryo. The germination is epigeal and the seedlings are fanerocotiledoneus. Drawings of all stages are included.

In order to identify the desiccation tolerance of E. edulis seeds, four fruitlots were collected from the Campinas Agronomic Institute Palmae collection, located at Ubatuba, state of Sao Paulo, Brazil, and taken in moisture-proof containers to the College of Agriculture in Botucatu, state of Sao Paulo. There, the fruits were shelled and the seeds were dried, either under silica gel conditions or in a drying chamber. The effects of dehydration were evaluated by measurements of the germination, embryo protrusion, germinative button, plumule, speed of emergence, seedling length, seed dry matter and water content. Differences among seedlots and between harvest season for most of the traits were observed. Euterpe edulis seedlots seem to tolerate water content reductions until 39% without significant reduction in germination and vigor. Below this value, most of the evaluated traits were negatively affected. Total loss of germination was observed whenever seed water content was reduced to values below 21%.

Calcium chloride concentrations from 0.0 to 12.12 mM were added to the culture medium and calcium content in calluses were determined directly by X-ray fluorescence spectrometry, a non-destructive method, allowing the processing of the same tissue for histological analysis. A multivariate statistical analysis (PCA - Principal Components Analysis) grouped the treatments into 5 blocks and indicated the most responsive group. Lack of calcium supply caused a complete absence of a morphogenic process and tissue collapse. An increase in calcium concentration gave higher total protein and sugar contents, an increase in peroxidase specific activity and changes in the histological characteristics. It was possible to verify that calcium stimulated globular somatic embryo formation at concentration of 6.62 mM.

An antigen-competitive enzyme-linked immunosorbent assay (Ag-C-ELISA) was developed for the detection of infectious bronchitis virus (IBV) antigens, M41 strain, in tissues from experimentally infected chickens, or in allantoic fluid harvested from inoculated embryonated eggs. The detection limit of IBV in the Ag-C-ELISA was 104.1 median embryo infective doses (EID50)/well. Tracheal and lung samples from chickens vaccinated with 102.5 EID50 of live attenuated infectious bronchitis (H120) vaccine were negative in the direct detection Ag-C-ELISA. The results indicate that the Ag-C-ELISA has the potential to detect IBV, either directly in tissue samples or when combined with the passage of material in embryonated eggs, thereby constituting an alternative method for the diagnosis of IBV.
