ExoU, uma citotoxina produzida pelo patógeno oportunista Pseudomonas aeruginosa e translocada para o citossol de células hospedeiras via sistema de secreção do tipo III, é associada à gravidade de infecções agudas. Estudos anteriores realizados em nosso laboratório relataram a potente atividade pró-inflamatória de ExoU, responsável por um intenso recrutamento de neutrófilos para o sítio de infecção. No presente trabalho, o efeito de ExoU na modulação da ativação do fator transcricional NF-κB e na regulação da expressão e da secreção da quimiocina para neutrófilos IL-8 foi avaliado em culturas de células epiteliais respiratórias e endoteliais humanas infectadas com a cepa PA103 de P. aeruginosa (produtora de ExoU) ou com a mutante deletada no gene exoU, PA103κexoU. Análises por RT-PCR semi-quantitativo mostraram que a infecção pela cepa produtora de ExoU levou ao aumento dos níveis de mRNA de IL-8, enquanto ensaios de alteração da mobilidade eletroforética (EMSA), supershift e com gene repórter mostraram que ExoU induziu a translocação nuclear do heterodímero transativador p65/p50 de NF-κB e a ativação da transcrição de genes dependente deste fator transcricional. Adicionalmente, o tratamento das culturas celulares com um inibidor de NF-κB antes da infecção bacteriana reduziu significativamente os níveis de mRNA de IL-8 e da secreção desta quimiocina. Em conjunto, estes resultados mostram que ExoU ativa NF-κB e, consequentemente, estimula a expressão e a secreção de IL-8 por células epiteliais respiratórias e células endoteliais infectadas com P. aeruginosa

Background: Neonatal trials remain difficult to conduct for several reasons: in particular the need for study sites to have an existing infrastructure in place, with trained investigators and validated quality procedures to ensure good clinical, laboratory practices and a respect for high ethical standards. The objective of this work was to identify the major criteria considered necessary for selecting neonatal intensive care units that are able to perform drug evaluations competently. Methodology and Main Findings: This Delphi process was conducted with an international multidisciplinary panel of 25 experts from 13 countries, selected to be part of two committees (a scientific committee and an expert committee), in order to validate criteria required to perform drug evaluation in neonates. Eighty six items were initially selected and classified under 7 headings: "NICUs description - Level of care'' (21), "Ability to perform drug trials: NICU organization and processes (15), "Research Experience'' (12), "Scientific competencies and area of expertise'' (8), "Quality Management'' (16), "Training and educational capacity'' (8) and "Public involvement'' (6). Sixty-one items were retained and headings were rearranged after the first round, 34 were selected after the second round. A third round was required to validate 13 additional items. The final set includes 47 items divided under 5 headings. Conclusion: A set of 47 relevant criteria will help to NICUs that want to implement, conduct or participate in drug trials within a neonatal network identify important issues to be aware of. O presente estudo tem como tema central o ganho de peso e o consumo alimentar no período reprodutivo. A tese está dividida em três partes: i) revisão da literatura científica sobre os fatores de risco para o ganho de peso excessivo na gestação e para a sua retenção no pós-parto; ii) análise quantitativa e qualitativa do consumo alimentar (energia, nutrientes e alimentos) do período gestacional para o pós-parto; e iii) avaliação prospectiva da associação entre a dieta hiperproteica (≥ 1.2 g/kg) e a variação de peso pós-gestacional. A revisão da literatura apontou como principais determinantes da variação de peso no pós-parto os seguintes fatores: ganho de peso gestacional, peso pré-gestacional, dieta, atividade física, lactação, idade, escolaridade, renda, paridade e raça. As partes ii e iii correspondem a dois artigos, sendo que a população do estudo empregada nas duas análises foi baseada em uma coorte de mulheres no pós-parto atendidas no Centro Municipal de Saúde (CMS) Marcolino Candau, localizado no Município do Rio de Janeiro. Dentre as 709 mulheres convidadas para participar da pesquisa, 479 ingressaram na coorte. As mulheres foram recrutadas através dos serviços de pré-natal e na rotina pediátrica de imunização do BCG, oferecidos pelo CMS, e imediatamente após o parto, na maternidade central de referência. A coleta de dados ocorreu entre maio de 1999 e abril de 2001, sendo 15 meses de recrutamento e 9 meses de seguimento. As mulheres foram entrevistadas aos 15 dias, 2, 6 e 9 meses do pós-parto. Foram consideradas elegíveis para as duas analises as mulheres com idade entre 18 e 45 anos de idade, entrevistadas até 30 dias do pós-parto, sem doenças crônicas pré-existentes, com gravidez de 35 ou mais semanas gestacionais e sem gestação gemelar. O estudo sobre a composição da dieta do período gestacional para o pós-parto contou com 276 mulheres que responderam aos dois questionários de freqüência de consumo alimentar (QFCA) aplicados aos 15 dias e aos 6 meses do seguimento. Os resultados mostraram que as mulheres consumiram dietas mais energéticas na gravidez e aquelas que mais restringiram o consumo de energia no pós-parto, apresentaram um incremento na densidade de proteínas da dieta. As análises das medidas repetidos do peso foram realizadas para avaliar o efeito da dieta hiperproteica (≥ 1.2 g/kg) sobre a variação de peso no pós-parto, para as quais haviam dados de uma amostra de 430 mulheres. Os resultados mostraram que as mulheres com dieta hiperproteica perderam mais peso do que as mulheres com dieta normo ou hipoproteica (< 1.2 g/kg) (226 g/mês versus 123 g/mês). As duas analises indicam que um pequeno incremento no consumo de proteínas promove maior perda de peso. Estudos longitudinais futuros devem incluir avaliação relativa a segurança das dietas hiperproteicas no pós-parto.

Dynamin-Related Protein 1 (Drp1), a large GTPase of the dynamin superfamily, is required for mitochondrial fission in healthy and apoptotic cells. Drp1 activation is a complex process that involves translocation from the cytosol to the mitochondrial outer membrane (MOM) and assembly into rings/spirals at the MOM, leading to membrane constriction/division. Similar to dynamins, Drp1 contains GTPase (G), bundle signaling element (BSE) and stalk domains. However, instead of the lipid-interacting Pleckstrin Homology (PH) domain present in the dynamins, Drp1 contains the so-called B insert or variable domain that has been suggested to play an important role in Drp1 regulation. Different proteins have been implicated in Drp1 recruitment to the MOM, although how MOM-localized Drp1 acquires its fully functional status remains poorly understood. We found that Drp1 can interact with pure lipid bilayers enriched in the mitochondrion-specific phospholipid cardiolipin (CL). Building on our previous study, we now explore the specificity and functional consequences of this interaction. We show that a four lysine module located within the B insert of Drp1 interacts preferentially with CL over other anionic lipids. This interaction dramatically enhances Drp1 oligomerization and assembly-stimulated GTP hydrolysis. Our results add significantly to a growing body of evidence indicating that CL is an important regulator of many essential mitochondrial functions.

English: For nearly a century, fisheries scientists have studied marine fish stocks in an effort to understand how the abundances of fish populations are determined. During the early lives of marine fishes, survival is variable, and the numbers of individuals surviving to transitional stages or recruitment are difficult to predict. The egg, larval, and juvenile stages of marine fishes are characterized by high rates of mortality and growth. Most marine fishes, particularly pelagic species, are highly fecund, produce small eggs and larvae, and feed and grow in complex aquatic ecosystems. The identification of environmental or biological factors that are most important in controlling survival during the early life stages of marine fishes is a potentially powerful tool in stock assessment. Because vital rates (mortality and growth) during the early life stages of marine fishes are high and variable, small changes in those rates can have profound effects on the properties of survivors and recruitment potential (Houde 1989). Understanding and predicting the factors that most strongly influence pre-recruit survival are key goals of fisheries research programs. Spanish: Desde hace casi un siglo, los científicos pesqueros han estudiado las poblaciones de peces marinos en un intento por entender cómo se determina la abundancia de las mismas. Durante la vida temprana de los peces marinos, la supervivencia es variable, y el número de individuos que sobrevive hasta las etapas transicionales o el reclutamiento es difícil de predecir. Las etapas de huevo, larval, y juvenil de los peces marinos son caracterizadas por tasas altas de mortalidad y crecimiento. La mayoría de los peces marinos, particularmente las especies pelágicas, son muy fecundos, producen huevos y larvas pequeños, y se alimentan y crecen en ecosistemas acuáticos complejos. La identificación los factores ambientales o biológicos más importantes en el control de la supervivencia durante las etapas tempranas de vida de los peces marinos es una herramienta potencialmente potente en la evaluación de las poblaciones. Ya que las tasas vitales (mortalidad y crecimiento) durante las etapas tempranas de vida de los peces marinos son altas y variables, cambios pequeños en esas tasas pueden ejercer efectos importantes sobre las propiedades de los supervivientes y el potencial de reclutamiento (Houde 1989). Comprender y predecir los factores que más afectan la supervivencia antes del reclutamiento son objetivos clave de los programas de investigación pesquera.

English: We describe an age-structured statistical catch-at-length analysis (A-SCALA) based on the MULTIFAN-CL model of Fournier et al. (1998). The analysis is applied independently to both the yellowfin and the bigeye tuna populations of the eastern Pacific Ocean (EPO). We model the populations from 1975 to 1999, based on quarterly time steps. Only a single stock for each species is assumed for each analysis, but multiple fisheries that are spatially separate are modeled to allow for spatial differences in catchability and selectivity. The analysis allows for error in the effort-fishing mortality relationship, temporal trends in catchability, temporal variation in recruitment, relationships between the environment and recruitment and between the environment and catchability, and differences in selectivity and catchability among fisheries. The model is fit to total catch data and proportional catch-at-length data conditioned on effort. The A-SCALA method is a statistical approach, and therefore recognizes that the data collected from the fishery do not perfectly represent the population. Also, there is uncertainty in our knowledge about the dynamics of the system and uncertainty about how the observed data relate to the real population. The use of likelihood functions allow us to model the uncertainty in the data collected from the population, and the inclusion of estimable process error allows us to model the uncertainties in the dynamics of the system. The statistical approach allows for the calculation of confidence intervals and the testing of hypotheses. We use a Bayesian version of the maximum likelihood framework that includes distributional constraints on temporal variation in recruitment, the effort-fishing mortality relationship, and catchability. Curvature penalties for selectivity parameters and penalties on extreme fishing mortality rates are also included in the objective function. The mode of the joint posterior distribution is used as an estimate of the model parameters. Confidence intervals are calculated using the normal approximation method. It should be noted that the estimation method includes constraints and priors and therefore the confidence intervals are different from traditionally calculated confidence intervals. Management reference points are calculated, and forward projections are carried out to provide advice for making management decisions for the yellowfin and bigeye populations. Spanish: Describimos un análisis estadístico de captura a talla estructurado por edad, A-SCALA (del inglés age-structured statistical catch-at-length analysis), basado en el modelo MULTIFAN- CL de Fournier et al. (1998). Se aplica el análisis independientemente a las poblaciones de atunes aleta amarilla y patudo del Océano Pacífico oriental (OPO). Modelamos las poblaciones de 1975 a 1999, en pasos trimestrales. Se supone solamente una sola población para cada especie para cada análisis, pero se modelan pesquerías múltiples espacialmente separadas para tomar en cuenta diferencias espaciales en la capturabilidad y selectividad. El análisis toma en cuenta error en la relación esfuerzo-mortalidad por pesca, tendencias temporales en la capturabilidad, variación temporal en el reclutamiento, relaciones entre el medio ambiente y el reclutamiento y entre el medio ambiente y la capturabilidad, y diferencias en selectividad y capturabilidad entre pesquerías. Se ajusta el modelo a datos de captura total y a datos de captura a talla proporcional condicionados sobre esfuerzo. El método A-SCALA es un enfoque estadístico, y reconoce por lo tanto que los datos obtenidos de la pesca no representan la población perfectamente. Además, hay incertidumbre en nuestros conocimientos de la dinámica del sistema e incertidumbre sobre la relación entre los datos observados y la población real. El uso de funciones de verosimilitud nos permite modelar la incertidumbre en los datos obtenidos de la población, y la inclusión de un error de proceso estimable nos permite modelar las incertidumbres en la dinámica del sistema. El enfoque estadístico permite calcular intervalos de confianza y comprobar hipótesis. Usamos una versión bayesiana del marco de verosimilitud máxima que incluye constreñimientos distribucionales sobre la variación temporal en el reclutamiento, la relación esfuerzo-mortalidad por pesca, y la capturabilidad. Se incluyen también en la función objetivo penalidades por curvatura para los parámetros de selectividad y penalidades por tasas extremas de mortalidad por pesca. Se usa la moda de la distribución posterior conjunta como estimación de los parámetros del modelo. Se calculan los intervalos de confianza usando el método de aproximación normal. Cabe destacar que el método de estimación incluye constreñimientos y distribuciones previas y por lo tanto los intervalos de confianza son diferentes de los intervalos de confianza calculados de forma tradicional. Se calculan puntos de referencia para el ordenamiento, y se realizan proyecciones a futuro para asesorar la toma de decisiones para el ordenamiento de las poblaciones de aleta amarilla y patudo.

A presente pesquisa comparativa mostra por que ao longo do século XX se consolidou o sindicalismo docente de base na Argentina, no Brasil e no México e explica as particularidades nacionais desse processo. A categoria sindicalismo docente de base, desenvolvida no contexto da presente investigação, pretende captar um fenômeno que aparece com clareza na segunda metade do século XX: as organizações de professores tem reivindicações fundamentalmente trabalhistas, legitimidade para organizar medidas coletivas de pressão sobre os governos (particularmente greves) e, além disso, a base da categoria tem uma importante gravitação sobre as entidades que pretendem representá-la. A comparação histórica e sociológica permite identificar três processos sucessivos que foram fundamentais para a afirmação do sindicalismo docente de base: a propagação das associações da categoria, a implantação das organizações na base docente e, finalmente, a consolidação do sindicalismo docente de base. Esses processos constituem conjunturas críticas e as características particulares que as práticas sindicais adquiriram nesses contextos tendem a se reproduzir basicamente por dois mecanismos: a tradição sindical e a regulamentação estatal da atividade sindical e do trabalho docente. As práticas sindicais docentes são estruturadas pelas características dos professores e das suas condições de existência. Também são mediadas pelas particularidades das organizações docentes, pela tradição sindical e pela ação governamental perante a atividade reivindicativa e associativa dos trabalhadores. A reconstrução desses elementos estruturantes e dessas mediações contribui para explicar quando as conjunturas críticas aconteceram e quais características particulares apresentaram. No México, a situação política geral e a relação que estabeleceram os quadros docentes com os governos pós-revolucionários em particular, permitiram uma rápida consolidação das associações docentes e uma implantação na base através do Estado na primeira metade do século. Também nesse período, no México, o professorado perdeu parcialmente as suas características femininas e o confronto com as autoridades como forma de pressão coletiva legitimou-se. Isso contrasta com os outros países, nos quais a organização da categoria se generalizou sem apoio estatal decisivo. A concentração da categoria em escolas e cidades fortaleceu a afirmação do sindicalismo docente de base cujas consequências já podem ser vistas nos três países em finais da década de 1950. Nesse contexto, as organizações docentes argentinas se implantaram na base (depois de que falisse a tentativa do governo de implantar o sindicalismo docente através do Estado), mas as brasileiras não. As organizações brasileiras só se implantariam na base após 1978. Processos que na década de 1950 já estavam em desenvolvimento (como a consolidação do professor como trabalhador de base de sistemas burocráticos dirigidos por especialistas, a regulamentação e burocratização da carreira, a perda de importância das recompensas simbólicas como incentivo para exercer o professorado) e outros que apareceriam nas décadas seguintes (como a incorporação crescente da mulher no mercado de trabalho, a radicalização do movimento estudantil e o recrutamento do professorado entre as camadas mais pobres da população) explicam a consolidação do sindicalismo docente de base entre as décadas de 1970 e 1980

A medula óssea adulta possui duas populações de células-tronco importantes no tratamento de diversas doenças hepáticas: células-tronco hematopoiéticas (CTHs) e células-tronco mesenquimais. A regeneração do fígado após a hepatectomia é um processo complexo que requer a proliferação de todas as células hepáticas. Fatores de crescimento, citocinas e componentes da matriz extracelular são elementos-chave nesse processo. As lamininas são uma família de proteínas de matriz extracelular, com funções adesivas e quimiotáticas pelo recrutamento de integrinas e outros receptores de superfície celular. No fígado normal, a laminina é expressa nas veias porta e centrolobular. O objetivo desse estudo foi investigar a expressão de laminina durante a regeneração hepática induzida por hepatectomia parcial e após o transplante de células mononucleares de medula óssea. As células mononucleares de medula óssea foram obtidas dos fêmures e tíbias de ratos, isoladas, marcadas com DAPI e injetadas pela veia porta em ratos recém-hepatectomizados. Os fígados foram coletados 15 minutos, 1 dia e 3 dias após a hepatectomia e o transplante de células de medula óssea e congelados. Os cortes foram imunomarcados com anticorpos primários anti-CD34 e anti-laminina de rato e observados em microscópio confocal de varredura a laser. Os resultados mostraram que 15 minutos após a hepatectomia parcial, as células-tronco hematopoiéticas CD34+ transplantadas foram encontradas em contato com a laminina localizada nas veias porta e centrolobular, indicando que a laminina poderia participar na adesão inicial das células-tronco a esses vasos logo após o seu transplante. Além disso, 1 e 3 dias após a hepatectomia, as células mononucleares de medula óssea transplantadas foram observadas nos sinusóides hepáticos expressando laminina. Esses resultados sugerem que a laminina pode ser um componente da matriz extracelular importante para a adesão e enxerto de células de medula óssea no fígado após uma lesão. Nós também analisamos a expressão de osteopontina (OPN) em células de medula óssea e CTHs. Os resultados por microscopia confocal demonstraram que a maioria das células mononucleares de medula óssea recém-isoladas expressa quantidades variáveis de OPN. Além disso, algumas CTHs CD34+ também expressam OPN. Após 1 e 4 dias de cultura, observamos uma diminuição de células expressando CD34, e um aumento na expressão de OPN pelas células mononucleares de medula óssea.

As part of the River Lune juvenile salmonid investigation, a number of sites on the River Wenning catchment were electrofished annually from 1981 - 1985. Particularly low Salmon parr populations were evident for much of the Wenning catchment which has caused some concern. All the Wenning catchment electrofishing results are reported in this paper and comparisons are made with designated groups of sites on the remainder of the Lune catchment. These groups of sites are: River Lune and tributaries, upstream Tebay, Upper Middle Lune, Lower Middle Lune, Lower Lune, Birk & Borrow Becks, Chapel & Crosdale Becks, Rawthey system, Barbon & Leek Becks and the Greta system. The general scarcity of Salmonids in large sections of the main river Wenning probably reflects the apparent limited natural spawning areas on the catchment. Details of Salmon fry and ova planting on the Wenning catchment since 1981 have been collated and whilst survival through the 0+ stage appears to be quite good, recruitment to the 1++ stage appears to be poor. Water quality does not appear to be responsible for the very low salmonid densities at some main river sites but is possibly a factor in apparently preventing any Salmon run in Keasden Beck. Recommendations for future Fisheries and Biological work are given.

This study takes a preliminary step towards investigating the reports of poor rod catches of coarse fish in the River Lune. These poor catches and concern over the possible loss of coarse fish from the Lower Lune is investigated, using creel census data from 1991. Historical data is used where available for comparative studies. The conclusion that this report comes to is that there is a problem with the recruitment of coarse fish within the population. Dace, roach and bream all show good growth rates but only poor if any recruitment. Historical investigations suggest that roach and bream may never have successfully recruited without artificial induced conditions. The situation with dace is of more concern as populations in other rivers in Northern England are also in decline, whilst historically they were naturally recruiting.

Background: Limited information is available about predictors of short-term outcomes in patients with exacerbation of chronic obstructive pulmonary disease (eCOPD) attending an emergency department (ED). Such information could help stratify these patients and guide medical decision-making. The aim of this study was to develop a clinical prediction rule for short-term mortality during hospital admission or within a week after the index ED visit. Methods: This was a prospective cohort study of patients with eCOPD attending the EDs of 16 participating hospitals. Recruitment started in June 2008 and ended in September 2010. Information on possible predictor variables was recorded during the time the patient was evaluated in the ED, at the time a decision was made to admit the patient to the hospital or discharge home, and during follow-up. Main short-term outcomes were death during hospital admission or within 1 week of discharge to home from the ED, as well as at death within 1 month of the index ED visit. Multivariate logistic regression models were developed in a derivation sample and validated in a validation sample. The score was compared with other published prediction rules for patients with stable COPD. Results: In total, 2,487 patients were included in the study. Predictors of death during hospital admission, or within 1 week of discharge to home from the ED were patient age, baseline dyspnea, previous need for long-term home oxygen therapy or non-invasive mechanical ventilation, altered mental status, and use of inspiratory accessory muscles or paradoxical breathing upon ED arrival (area under the curve (AUC) = 0.85). Addition of arterial blood gas parameters (oxygen and carbon dioxide partial pressures (PO2 and PCO2)) and pH) did not improve the model. The same variables were predictors of death at 1 month (AUC = 0.85). Compared with other commonly used tools for predicting the severity of COPD in stable patients, our rule was significantly better. Conclusions: Five clinical predictors easily available in the ED, and also in the primary care setting, can be used to create a simple and easily obtained score that allows clinicians to stratify patients with eCOPD upon ED arrival and guide the medical decision-making process.

A summary of results obtained from 1969 to 1977 is given. It concerns the biology of the species (ecology and distribution of the adults, behaviour and diel variation of catch rates, reproduction and larval migration, juveniles migration and recruitment at sea, sexual maturity, growth and mortality by marking experiments) and the history of the fishery (catches, efforts, seasonal variations of catch rates). The combined use of a dynamic pool model of Ricker and a production model of Fox leads to the evaluation of the potential of the stock. The simulation of different and combined fishery strategies on adults at sea and juveniles in lagoons, allows the evaluation of the consequences (in yield, value, biomass and potential fecundity) of the different proposed management procedures (reductions in fishing effort, closed seasons on both fisheries).

In relation to the hydroclimate prevailing off Congo (B) and Côte d'Ivoire, synchronic variations are described in abundance and distribution of Pseudotolithus senegalensis V., economically the most important species in the West African trawl fishery. Although this fish prefers the warm surface layer, it is relatively indifferent to hydrological conditions, since it may also occur in the thermocline down to the higher levels of 'Central South Atlantic Water'. The oxygen concentration appears to have an important effect upon their distribution, especially the low concentrations occurring with the upwelling. The main biological functions, such as spawning and recruitment times, condition factor, diet and ring appearance on otoliths, also follow cycles, which are parallel with the hydroclimate one. Therefore, the ring-shaped structures revealed by burning previously ground otoliths could be easily interpreted. Thus, an accurate method for ageing tropical fish is now available. From the age determinations for the Congolese stock, it appears that growth is fast and total mortality rate high. An influence of fishing effort, which increased 3 times during the exploited phase of sampled specimens, appears both in growth and in total mortality. From there an estimate for the fishing mortality could be given. The estimates of dynamic parameters for the Congolese stock are then used in the Beverton-Holt model. A valuable increase of the yield could be expected, if mesh size is widened and effort restricted.

Yorkshire Water Services (YWS) are currently granted a Time Limited Licence (TLL) for abstraction at Kilgram Bridge which is due for renewal in 1999. The Environment. Agency requires information on fish populations with regards to drought conditions and any possible effects that abstraction may have when considering licence renewal. In' order to evaluate any effects of drought and abstraction a three year study was instigated to examine fish populations. Surveys were conducted at nine main River Ure sites and two tributaries in which the triennial rolling programme formed the basis of site selection. Multi-method sampling techniques were carried out at several sites in order to evaluate capture efficiency. High densities of brown trout juveniles were observed in the tributaries with an indication that fish had become crowded as a result of low flows. Recruitment of brown trout in the tributaries was not directly related to flow levels in the main R. Ure. However, it is concluded that salmonids are at risk during drought flows and high temperatures from increased susceptibility to disease, predation, poor water quality and the direct lethal effect of high temperatures in shallow water.
