The antifungal "paradoxical effect" has been described as the reversal of growth inhibition at high doses of echinocandins, most usually caspofungin. This microbiological effect appears to be a cellular compensatory response to cell wall damage, resulting in alteration of cell wall content and structure as well as fungal morphology and growth. In vitro studies demonstrate this reproducible effect in a certain percentage of fungal isolates, but animal model and clinical studies are less consistent. The calcineurin and Hsp90 cell signaling pathways appear to play a major role in regulating these cellular and structural changes. Regardless of the clinical relevance of this paradoxical growth effect, understanding the specific actions of echinocandins is paramount to optimizing their use at either standard or higher dosing schemes, as well as developing future improvements in our antifungal arsenal.

The use of carbon fibers to develop new sensors and biosensors has received great attention due to its characteristics and electrochemical properties. A brief presentation about history, properties, characteristics, composition and structure of the carbon fibers are shown in this paper. Several applications of the carbon fibers in electroanalytical chemistry for determination of metals and organic molecules in environmental and clinical samples are also described.

Työn tarkoituksessa on luoda kokonaisvaltainen kuva Venäjän sähkösektorista ja siellä suoritettavasta reformista. Työ johdattelee lukijan aiheeseen kuvaamalla Venäjän sähkösektorin- ja markkinoiden tilaa ennen reformia. Tässä on tärkeää ymmärtää, kuinka kulutus ja tuotanto kohtasivat. Tämän jälkeen siirrytään selvittämään reformin syitä, suuntia ja päämääriä. Reformin kolme pääkohtaa ovat lainsäädännöllisen viitekehyksen luominen, sähkösektorin uudelleenjärjestely sekä uusien sähkömarkkinoiden kehittäminen ja täytäntöönpano. Uudelleenjärjestely on valtava prosessi, joka koskettaa koko Venäjän sähkömarkkinoita. Tämän prosessin keskeisenä tekijänä tosin on lähes monopoliasemaa nauttiva Unified Energy System of Russia. Tätä prosessia pyritään kuvaamaan alusta asti tähän päivään saakka. Toinen merkittävä prosessi on ollut uuden sähkömarkkinamallin kehittäminen ja tämän käyttöönotto. Työssä kuvataan tämän uuden mallin toimintaa ja rakennetta, ja työn loppupuolella pyritään selvittämään Venäjän sähkösektorin tulevaisuuden suunnitelmia, haasteita ja ennusteita.

Immobilized Metal Ion Affinity Cromatography - IMAC - is a group-specific based adsorption applied to the purification and structure-function studies of proteins and nucleic acids. The adsorption is based on coordination between a metal ion chelated on the surface of a solid matrix and electron donor groups at the surface of the biomolecule. IMAC is a highly selective, low cost, and easily scaled-up technique being used in research and commercial operations. A separation process can be designed for a specific molecule by just selecting an appropriate metal ion, chelating agent, and operational conditions such as pH, ionic strength, and buffer type.

Production of ethanol from biomass fermentation has gained much attention recently. Biomass cellulosic material is first converted into glucose either by chemical or by enzymatic process, and then glucose is fermented to ethanol. Considering the current scenario, where many efforts are devoted for the search of green routes to obtaining ethanol from renewable sources, this review presents the relationship between structure and properties of cellulosic material, pre-treatments and hydrolysis of cellulosic material, and structure and function of cellulase enzyme complex.

Suomessa rautatiellä käytetään muutamia tuhansia sähköisesti ohjattavia vaihteita, joiden toiminta varmistetaan määräajoin tehtävillä huolloilla. Työssä on tutkittu, onko kehitteillä olevalla vaihteenohjaimella mahdollista mitata ja analysoida vaihteen kunnonvalvontaan liittyviä suureita. Tutkimuksessa on tutustuttu rautatietekniikkaan sekä vaihdetta ohjaavaan vaihteenkääntölaitteeseen. Tutkimuksen perusteella on kehitetty sulautettuun järjestelmään perustuva mittaus- ja analysointijärjestelmäsovellus, joka tekee mittauksia, havaintoja ja johtopäätöksiä vaihteen toiminnasta. Mittaukset on suoritettu moottoria ohjaavista johtimista ja mittaustulokset on tallennettu analysointia ja jatkokäsittelyä varten. Ohjelmiston toimintaa on varmistettu mittauksilla sekä laboratorio-olosuhteissa että oikealla vaihteella testausympäristössä. Saatujen tulosten perusteella voidaan todeta, että kehitetyllä laitteistolla on mahdollista toteuttaa vaihdetta diagnosoivia mittauksia riittävän laadukkaasti ja tarkasti. Työssä kehitetty järjestelmäalusta soveltuu käytettäväksi vaihteen diagnostiikan jatkokehittämiselle.

The correct utilization of non-wood raw material allows reducing tree cutting and reduces emissions of carbon dioxide from burning of non-wood plants on farmers fields. Also it allows increasing economical situation in regions that non-wood plants are grown and where they are converted into pulp and paper. Also it gives positive effect on population pressure of work by addition of working place. In the literature survey included an overview of the historical meaning of non-wood pulp on developing paper production and structure of non-wood pulps. Moreover, anatomical and chemical composition of straw, reed and bamboo were studied more detailed. Also, an overview of the utilization of non-wood pulp in papermaking was made. Especially tissue, tree-free and release papers were reviewed. In the experimental part the goal was to investigate suitability of non-wood pulp like wheat straw pulp and bamboo pulp for different fiber products. Finally release and tree-free paper products were selected for experimental studies. It was discovered that wheat straw, especially screened wheat straw, showed good results for release paper. Also utilization of wheat straw and bamboo pulp in tree-free paper showed good results and suitability of these non-wood pulps for tree-free paper production. Also it was noticed that addition of wheat straw pulp gave positive effect on initial wet strength for release and tree-free paper.

The control of coating layer properties is becoming increasingly important as a result of an emerging demand for novel coated paper-based products and an increasing popularity of new coating application methods. The governing mechanisms of microstructure formation dynamics during consolidation and drying are nevertheless, still poorly understood. Some of the difficulties encountered by experimental methods can be overcome by the utilisation of numerical modelling and simulation-based studies of the consolidation process. The objective of this study was to improve the fundamental understanding of pigment coating consolidation and structure formation mechanisms taking place on the microscopic level. Furthermore, it is aimed to relate the impact of process and suspension properties to the microstructure of the coating layer. A mathematical model based on a modified Stokesian dynamics particle simulation technique was developed and applied in several studies of consolidation-related phenomena. The model includes particle-particle and particle-boundary hydrodynamics, colloidal interactions, Born repulsion, and a steric repulsion model. The Brownian motion and a free surface model were incorporated to enable the specific investigation of consolidation and drying. Filter cake stability was simulated in various particle systems, and subjected to a range of base substrate absorption rates and system temperatures. The stability of the filter cake was primarily affected by the absorption rate and size of particles. Temperature was also shown to have an influence. The consolidation of polydisperse systems, with varying wet coating thicknesses, was studied using imposed pilot trial and model-based drying conditions. The results show that drying methods have a clear influence on the microstructure development, on small particle distributions in the coating layer and also on the mobility of particles during consolidation. It is concluded that colloidal properties can significantly impact coating layer shrinkage as well as the internal solids concentration profile. Visualisations of particle system development in time and comparison of systems at different conditions are useful in illustrating coating layer structure formation mechanisms. The results aid in understanding the underlying mechanisms of pigment coating layer consolidation. Guidance is given regarding the relationship between coating process conditions and internal coating slurry properties and their effects on the microstructure of the coating.

I det lilla sammanhanget synliggörs de stora frågorna om både pedagogik, kultur och struktur och om samverkan mellan dem. Studiet av de minsta enheterna i den finländska utbildningen ger kunskap om det viktiga i pedagogiska relationer och i skolgemenskaper, men synliggör också samhällsskeendets inbyggda konflikter om mål och värderingar. Det övergripande syftet med studien är att fördjupa kunskapen om de små skolornas pedagogiska, kulturella och strukturella innebörd och betingelser. Genom djupintervjuer med 12 finlandssvenska lärare i byskolor med färre än 30 elever, analys av skolindragningsdebatt och utbildningspolitiska styrdokument, samt genom teoretiskt förankrade reflektioner skapas en förståelse av såväl lärares arbete och pedagogiska tänkande som skolans funktion i samhället. Avhandlingen byggs upp enligt ett hermeneutiskt och narrativt tänkande. I studien framkommer att byskollärares pedagogiska tänkande syftar till det enskilda barnets optimala och balanserade helhetsutveckling i en gemenskap, där det är centralt att finna den pedagogiska balansen och möjligheten inom kontinuum mellan bl.a. elevbemyndigande och beledsagande. De pedagogiska intentionerna bär syftet att ge rötter och vingar, samhörighet och frihet. I lärarnas beskrivningar uttrycks en sammanvävning mellan deras pedagogiska intentioner och de kontextuella möjligheterna att realisera dessa. Byskollärarna är bärare av en pedagogisk professionalitet som utvecklas i relation till arbetets betingelser. Det praktiska yrkeskunnandet innefattar en balansgång mellan å ena sidan systematik och organisering och å andra sidan flexibilitet och frihet. Att samma lärare handhar eleverna en lång tid är en pedagogisk möjlighet och utmaning. I lärarnas berättelser aktualiseras vad balansgången i en god pedagogisk relation innebär. Både lärare och elever fostras in i en särskild skolkultur och undervisningskultur, i en växelverkan. Yrkeskulturen präglas av olika former av samverkan, både med skolans hela personal som ett teamarbete och med lokalsamhället. Lärarna uttrycker god arbetstrivsel, men friheten och ansvaret i arbetet kan vara både stimulerande och betungande. I en metaanalys skapas teoretiska modeller för vad arbete i närhetens och litenhetens spänningsfält kan innebära och hur det kan påverka lärares professionella utveckling och ork. Den lilla byskolan relaterad till en större samhällskontext öppnar för frågor om vad kvalitet innebär och vilka värderingar som är riktgivande inom utbildningsplanering. Kampen för kontinuiteten i byskolans berättelse tolkas som en kamp för det lokala rummet, för gemenskap, existens, framtid, likvärdighet och trygghet. Kampen för byskolan är ett försvar av både lokal livskvalitet och pedagogisk kvalitet för den enskilde eleven. I det övergripande kulturella sammanhanget synliggörs motsättningar. Det verkar finnas en inbyggd konflikt i utbildningsplaneringens föresatser att samtidigt uppnå jämlikhet, kostnadseffektivitet och kvalitet. En teoretisk modell synliggör hur pedagogik, kultur och struktur samverkar inom utbildning/byskola och påverkar lärares och elevers handlingsutrymme.

The time required to regrowth a forest in degraded areas depends on how the forest is removed and on the type of land use following removal. Natural regeneration was studied in abandoned old fields after intensive agricultural land use in areas originally covered by Brazilian Atlantic Forests of the Anchieta Island, Brazil in order to understand how plant communities reassemble following human disturbances as well as to determine suitable strategies of forest restoration. The fields were classified into three vegetation types according to the dominant plant species in: 1) Miconia albicans (Sw.) Triana (Melastomataceae) fields, 2) Dicranopteris flexuosa (Schrader) Underw. (Gleicheniaceae) thickets, and 3) Gleichenella pectinata (Willd.) Ching. (Gleicheniaceae) thickets. Both composition and structure of natural regeneration were compared among the three dominant vegetation types by establishing randomly three plots of 1 x 3 m in five sites of the island. A gradient in composition and abundance of species in natural regeneration could be observed along vegetation types from Dicranopteris fern thickets to Miconia fields. The gradient did not accurately follow the pattern of spatial distribution of the three dominant vegetation types in the island regarding their proximity of the remnant forests. A complex association of biotic and abiotic factors seems to be affecting the seedling recruitment and establishment in the study plots. The lowest plant regeneration found in Dicranopteris and Gleichenella thickets suggests that the ferns inhibit the recruitment of woody and herbaceous species. Otherwise, we could not distinguish different patterns of tree regeneration among the three vegetation types. Our results showed that forest recovery following severe anthropogenic disturbances is not direct, predictable or even achievable on its own. Appropriated actions and methods such as fern removal, planting ground covers, and enrichment planting with tree species were suggested in order to restore the natural forest regeneration process in the abandoned old fields.

Current industrial atomic layer deposition (ALD) processes are almost wholly confined to glass or silicon substrates. For many industrial applications, deposition on polymer substrates will be necessary. Current deposition processes are also typically carried out at temperatures which are too high for polymers. If deposition temperatures in ALD can be reduced to the level applicable for polymers, it will open new interesting areas and applications for polymeric materials. The properties of polymers can be improved for example by coatings with functional and protective properties. Although the ALD has shown its capability to operate at low temperatures suitable for polymer substrates, there are other issues related to process efficiency and characteristics of different polymers where new knowledge will assist in developing industrially conceivable ALD processes. Lower deposition temperature in ALD generally means longer process times to facilitate the self limiting film growth mode characteristic to ALD. To improve process efficiency more reactive precursors are introduced into the process. For example in ALD oxide processes these can be more reactive oxidizers, such as ozone and oxygen radicals, to substitute the more conventionally used water. Although replacing water in the low temperature ALD with ozone or plasma generated oxygen radicals will enable the process times to be shortened, they may have unwanted effects both on the film growth and structure, and in some cases can form detrimental process conditions for the polymer substrate. Plasma assistance is a very promising approach to improve the process efficiency. The actual design and placement of the plasma source will have an effect on film growth characteristics and film structure that may retard the process efficiency development. Due to the fact that the lifetime of the radicals is limited, it requires the placement of the plasma source near to the film growth region. Conversely this subjects the substrate to exposure byother plasma species and electromagnetic radiation which sets requirements for plasma conditions optimization. In this thesis ALD has been used to modify, activate and functionalize the polymer surfaces for further improvement of polymer performance subject to application. The issues in ALD on polymers, both in thermal and plasma-assisted ALD will be further discussed.

This thesis discusses adaption of new project management tool at ABB Oy Motors and Generators business unit, Synchronous Machines profit centre. Thesis studies project modeling in general and buries in the Gate Model used at ABB Synchronous Machines. It is essential to understand Gate Model because this new project management tool, called Project Master Document, is created on the base of the existing project model. Thesis also analyzes goals and structure of Project Master Document in order to ease implementation of this new tool. Project Master Document aims to improved customer order fulfillment by clearing order handover interface. Office process, especially responsibilities and target dates, become also clearer after Master Document implementation. The document is built to be frame for whole order fulfillment process including check points for each gate of project model and updated memos from all project meetings. Furthermore, project progress will be clearly stated by status markings and visualized with colors.

Parametric cost modeling is a technique, where cost estimating relationships are built to meet products’ parameters. Parameters are directly defined from product features, when it is possible to solve product cost exact before even a single product is manufactured and calculated with general cost accounting. The parametric model can be used in product design, sourcing and comparing product cost of similar products. The model reveals the cost origin more clear than general accounting. The purpose of this thesis was to find out parameters for modeling elevator doors and validate the parameters to meet actual costs. The other target was to simulate cost impact and changes in the cost structure. The results were compared to previous calculations and actual costs and model was tested in new product design. The results of the calculations revealed, that material consumption is the most significant issue in design effectiveness as well as the complexity of the components and structure. To develop the model more research is needed for other continents cost structure and waste calculation principles.

Hoitotyössä infektioiden torjunnan painopistealueena on ollut lisätä potilaiden osallistuvuutta voimavaraistavalla potilasohjauksella. Potilasohjauksen apuna käytetään usein kirjallisia potilasohjeita, joiden rakenne ja laatu tulee olla hyvä. Lisäksi potilasohjeiden tulee omata valmiuksia tukea voimavaraistavaa potilasohjausta. Kirjalliset potilasohjeet ovat hyvä potilasohjauksen keino, mutta systemaattinen tieto potilasohjeiden laadusta, valmiuksista tukea voimavaraistavaa potilasohjausta sekä infektioiden torjunnan sisällöstä puuttuu. Tutkimuksen tarkoituksena oli analysoida Suomen yliopistosairaaloiden kirurgisten potilasohjeiden infektioiden torjuntaan liittyvää sisältöä. Lisäksi oltiin kiinnostuneita potilasohjeiden laadusta hyvien kirjallisten potilasohjeiden ominaispiirteiden toteutumisessa sekä potilasohjeiden valmiuksista tukea voimavaraistavaa potilasohjausta. Tutkimuksen aineistona oli viiden yliopistosairaalan kirurgisille aikuispotilaille tarkoitetut potilasohjeet (N = 237). Organisaatioittain joka viides potilasohje (n = 50) analysoitiin potilasohjeiden laadun osalta sisällönerittelyllä ja aikaisemmin kehitetyn mittarin avulla (Salanterä ym. 2004). Deduktiivisella sisällönanalyysillä analysoitiin potilasohjeiden valmiuksia tukea voimavaraistavaa potilasohjausta, jonka ulottuvuuksien esiintyvyyden arviointiasteikko oli kehitetty tätä tutkimusta varten. Induktiivisella sisällönerittelyllä analysoitiin infektioiden torjuntaan liittyvä sisältö. Potilasohjeiden laadussa oli paljon vaihtelua, ja vain 36 % (n = 18) potilasohjeista oli julkaistu viimeisen kolmen vuoden (2010–2012) aikana. Potilasohjeet olivat ulkoasultaan selkeitä ja johdonmukaisia, jolloin kirjallisten potilasohjeiden ominaisuuksista parhaiten toteutuivat ulkoasun sekä kielen ja rakenteen ominaisuudet. Potilasohjeet kuvasivat toimenpidettä ja siitä selviytymistä, antaen parhaiten valmiuksia tukea toiminnallista ja bio-fysiologista selviytymistä potilasohjauksella. Infektioiden torjuntaan liittyvä sisältö oli pääasiassa esitetty epäsuorasti toisen asian kautta. Infektioiden syntyyn tai torjuntaan ei viitattu, vaan asiaa tarkasteltiin muusta näkökulmasta esimerkiksi ylipainon ja ihon kunnon osalta. Yleisin infektioiden torjuntaan liittyvä sisältö koski käsihygieniaa, vaikka sen toteuttamista ei opastettu tai ohjeistettu. Tutkimus tarjoaa lähtökohdan kehittää potilasohjeiden infektioiden torjuntaan liittyvää sisältöä sekä potilaan osallistuvuuden lisäämistä. Erityisesti tulisi kehittää potilaan osallistuvuuden tukemista voimavaraistavalla potilasohjauksella. Jatkotutkimuksena tulisi kartoittaa potilaiden tarpeet infektioiden torjunnan sisällöstä sekä kehittää kansallinen ohjeistus potilasohjeiden infektioon torjuntaan liittyvästä sisällöstä ja rakenteesta.
