This paper explores the complexities and contradictions of frontline practice that pose problems for personalised social care through enhanced choice. It draws on semi-structured interviews with community care workers, social workers, occupational therapists and care managers in a social service department. Practitioners interviewed were asked about their current assessment and documentation system, including the assessment documents currently used; how they approached information gathering and the topics they explored with service users; and their experience of documenting assessment and care management. The paper argues that the validity and sustainability of personalised social care in frontline practice relies on developing a thorough understanding of the complex and implicit assessment processes operating at the service user/practitioner interface and the inevitable tensions that arise for practitioners associated with the organisational context and broader service environment. The findings demonstrate the variability among practitioners in how they collect information and more importantly, the critical role practitioners occupy in determining the kinds of topics to be explored during the assessment process. In so doing, it shows how practitioners can exert control over the decision-making process. More importantly, it provides some insight into how such processes are shaped by the constraints of the organisational context and broader service environment. Complexities and contradictions may be an inherent part of frontline practice. The issues discussed in this paper, however, highlight potential areas that might be targeted in conjunction with implementing personalised social care through enhanced choice for people with disabilities.

Although cytosolic glutathione S-transterase (GST) enzymes occupy a key position in biological detoxification processes, two of the most relevant human isoenzymes. GST1-1 and GSTM1-1, are genetically deleted (non-functional alleles GSTT1*0 and GsTM1*0) in a high percentage of the human population, with major ethnic differences. The structures of the GSTT and GSTM gene areas explain the underlying genetic processes. GSTT1-1 is highly conserved during evolution and plays a major role in phase-II biotransformation of a number of drugs and industrial chemicals. e.g. cytostatic drugs, hydrocarbons and halogenated hydrocarbons. GSTM1-1 is particularly relevant in the deactivation of carcinogenic intermediates of polycyclic aromatic hydrocarbons. Several lines of evidence Suggest that hGSTT1-1 and/or hGSTM1-1 play a role in the deactivation of reactive oxygen species that are likely to be involved in cellular processes of inflammation, ageing and degenerative diseases. There is cumulating evidence that combinations of the GSTM1*0 state with other genetic traits affecting the metabolism of carcinogens (CYP1A1, GSTP1) may predispose the aero-digestivc tract and lung, especially in smokers, to a higher risk of cancer. The GSTM1*0 status appears also associated with a modest increase in the risk of bladder cancer, consistent with a GSTM1 interaction with carcinogenic tobacco smoke constituents. Both human GST deletions, although largely counterbalanced by overlapping substrate affinities within the GST superfamily, have consequences when the organism comes into contact with distinct man-made chemicals. This appears relevant in industrial toxicology and in drug metabolism.

Antarctic bryophyte communities presently tolerate physiological extremes in water availability, surviving both desiccation and submergence events. We investigated the relative ability of three Antarctic moss species to tolerate physiological extremes in water availability and identified physiological, morphological, and biochemical characteristics that assist species performance under such conditions. Tolerance of desiccation and submergence was investigated using chlorophyll fluorescence during a series of field- and laboratory-based water stress events. Turf water retention and degree of natural habitat submergence were determined from gametophyte shoot size and density, and delta C-13 signatures, respectively. Finally, compounds likely to assist membrane structure and function during desiccation events (fatty acids and soluble carbohydrates) were determined. The results of this study show significant differences in the performance of the three study species under contrasting water stress events. The results indicate that the three study species occupy distinctly different ecological niches with respect to water relations, and provide a physiological explanation for present species distributions. The poor tolerance of submergence seen in Ceratodon purpureus helps explain its restriction to drier sites and conversely, the low tolerance of desiccation and high tolerance of submergence displayed by the endemic Grimmia antarctici is consistent with its restriction to wet habitats. Finally the flexible response observed for Bryum pseudotriquetrum is consistent with its co-occurrence with the other two species across the bryophyte habitat spectrum. The likely effects of future climate change induced shifts in water availability are discussed with respect to future community dynamics.

Background. We describe the development, reliability and applications of the Diagnostic Interview for Psychoses (DIP), a comprehensive interview schedule for psychotic disorders. Method. The DIP is intended for use by interviewers with a clinical background and was designed to occupy the middle ground between fully structured, lay-administered schedules, and semi-structured., psychiatrist-administered interviews. It encompasses four main domains: (a) demographic data; (b) social functioning and disability; (c) a diagnostic module comprising symptoms, signs and past history ratings; and (d) patterns of service utilization Lind patient-perceived need for services. It generates diagnoses according to several sets of criteria using the OPCRIT computerized diagnostic algorithm and can be administered either on-screen or in a hard-copy format. Results. The DIP proved easy to use and was well accepted in the field. For the diagnostic module, inter-rater reliability was assessed on 20 cases rated by 24 clinicians: good reliability was demonstrated for both ICD-10 and DSM-III-R diagnoses. Seven cases were interviewed 2-11 weeks apart to determine test-retest reliability, with pairwise agreement of 0.8-1.0 for most items. Diagnostic validity was assessed in 10 cases, interviewed with the DIP and using the SCAN as 'gold standard': in nine cases clinical diagnoses were in agreement. Conclusions. The DIP is suitable for use in large-scale epidemiological studies of psychotic disorders. as well as in smaller Studies where time is at a premium. While the diagnostic module stands on its own, the full DIP schedule, covering demography, social functioning and service utilization makes it a versatile multi-purpose tool.

The thelastomatoid fauna of Macropanesthia rhinoceros was examined from 13 localities across its range in Queensland, Australia. Nine species of thelastomatoids, including two representing new genera, Geoscaphenema megaovum n. g., n. sp. and Jaidenema rhinoceratum n. g., n. sp., were found. Macropanesthia rhinoceros is reported as a new host for seven species previously recorded from Panesthia cribrata (Blaberidae: Panesthiinae) and P. tryoni tryoni, viz, Blattophila sphaerolaima, Leidynemella fusiformis, Cordonicola gibsoni, Travassosinema jaidenae, Coronostoma australiae, Hammerschmidtiella hochi and Desmicola ornata. Overall estimated richness for the system ranged from 10.1-13.5 species. The high degree of parasite faunal overlap between M. rhinoceros and the two Panesthia species is surprising given the disparate ecological niches that they occupy; P. cribrata and P. tryoni tryoni burrow in, and feed upon, moist decaying wood and require a climate that is moist all year round, whereas M. rhinoceros burrows in loose soil, feeds on fallen leaf litter and is tolerant of much drier environments.

A decade after his death, Manfredo Tafuri continues to occupy a pivotal position within contemporary architectural discourse both as a protagonist of the critical debates of the 1970s and as a historian who elucidated every period of architectural history from the fifteenth century to the twentieth. In addition to marking the publication of the English translation of his last work, INTERPRETING THE RENAISSANCE: PRINCES, CITIES, ARCHITECTURES, translated by Daniel Sherer (Yale University Press in association with Harvard GSD, 2006), this two-day conference took this event as an occasion for a new assessment of his critical legacies.

A região do Grande ABC é caracterizada por ser local que retrata, de forma singular, os conflitos, as mudanças e o impacto da globalização no País e no mundo. Diferentes setores, articulados, construíram o Planejamento Regional para a região, que se deparava com a flexibilização, o desemprego estrutural e a diminuição nos postos de trabalho das grandes montadoras, de indústrias de autopeças, entre outras, e descobriu caminhos de enfrentamento para o processo que avassalou o Brasil e o mundo. Isso exigiu a profissionalização de quem ocupava a função de Secretário, para atender ao perfil das multinacionais. A partir daí, o Secretariado Executivo vem se consolidando como uma profissão de caráter inteligente e intelectual, considerado como analítico-simbólico. O Secretário rompeu com seu antigo perfil, enfrentando a concorrência de profissionais de outras áreas, e desempenha hoje um papel de assessoria adjunta, gerenciando processos e facilitando tomadas de decisão. É um profissional de formação intelectual diferenciada, com competências técnicas, comportamentais e estratégicas. Assim, esta pesquisa teve como objetivo analisar o processo de construção da identidade do ser profissional Secretário na região do Grande ABC; levantar os espaços ocupados por esse profissional, através de mapeamento dos postos que ocupa, e construir uma linha do tempo dessa profissão na região. Para atender aos objetivos, foi utilizada pesquisa documental, a construção de uma linha do tempo e a história oral temática, procedendo-se à coleta e à análise dos dados de sete profissionais Secretários atuantes na região do Grande ABC. O diálogo interdisciplinar entre teoria e os discursos evidenciou que ainda existe um estereótipo cultural quanto à profissão nas organizações, mas há um respeito, ainda que velado, dos superiores e colegas, conquistado por sua persistência e profissionalização. Os resultados demonstram que a identidade do ser profissional Secretário é construída em suas experiências vividas no passado e no presente, o que lhe proporciona um esboço de valores individuais e profissionais que são revistos nas diversas organizações em que atuam ou atuaram e dentro do contexto regional estudado, gerando variações em suas identidades.(AU)

A pesquisa, em 5 capítulos, desenvolve o tema dos/as pobres como categoria social nos Sl 3-14, subunidade do primeiro livro do saltério (Sl 3-41). Os Sl 3-14 são atribuídos a Davi, o fato está relacionado com as escolas e suas teologias presentes na edição final do saltério. Esses salmos nasceram nas comunidades camponesas do antigo Israel, posteriormente, foram aperfeiçoados e adaptados por grupos de cantores oficiais no templo de Jerusalém. Os Sl 3-14 se destacam pelo lamento e pela súplica individual. Pertencem a uma coleção pré-exílica, mas concentra textos tardios, do pós-exílio. Os lugares que ocupam foram pensados estrategicamente. Os Sl 9; 10 apresentam conceitos hebraicos que identificam os/as pobres: dak, ani, ebyon. Estes/as designam pequenos/as camponeses/as livres, ainda com acesso à terra. Ao longo do antigo Israel não sofreram mudanças bruscas como categoria social, no entanto, podem assinalar-se algumas características que os/as distinguem nos períodos correspondentes ao primeiro e o segundo templo de Jerusalém. Ademais, os Sl 9; 10 apresentam palavras sinônimas que também os/as identificam: hellkah pobre/infeliz e naqi inocente . Apesar das pequenas variações dos conceitos, todos apontam à uma categoria social, com direito à apelação nos tribunais, embora com fraca influência jurídica. Essa comunidade tem identidade teológica. Javé é apresentado como o seu defensor. A espoliação no saltério é algo dramático, porque o rosto do/a pobre é o próprio rosto de Javé. A pobreza não é um assunto de espiritualidade nem de casualidade. É gerada por um sistema político-social, planejado de forma inteligente, que não permite ao povo da roça progredir como agricultor. Esse setor poderoso, nacional ou estrangeiro, é identificado nos textos, sob os conceitos: goyim nações , sorerim agressores , oyebim inimigos , raxa im injustos . O seu domínio é suportado pela violência e as armas. A ideologia dos sistemas dominantes é fundamental para a interpretação dos textos. A sociedade dos salmistas apresenta crises com relação à identidade humana. A violência e a paz se disputam os espaços. O Sl 8 mostra uma sociedade alternativa pensada a partir daquilo aparentemente fraco: as olelim crianças e os yanaqim lactantes (Sl 8,3). O grito das criancinhas, o grito dos/as oprimidos/as, unido ao grito da criação, se compara à dor de parto, com o qual inicia a vida. Trata-se de um grito que busca transformar os trajetos entortados da história. Esses são indícios da esperança que distingue a teologia dos/as pobres. Os Sl 3-7 e 11-14 continuam a apresentar a situação dos/as pobres. Às vezes, localizam-se os conceitos: ani oprimido e ebyon pobre , outras, recorre-se a novos sinônimos como has͇id fiel e sadiq justo . Esses salmos demonstram que os/as pobres estão presentes também nos textos onde tais conceitos não aparecem. As agrupações (Sl 3-7 e 11-14) são uma pausa na subunidade (Sl 3-14), não uma quebra de sentido com os Sl 8; 9 e 10. Finalmente, se localiza na sociedade dos Sl 3-14, o Modo de Produção Tributário. As teorias das ciências econômica, arqueológica, histórica, contribuem com a compreensão do universo sóciopolítico gerador de pobres.

A dissertação elabora considerações reflexivas a respeito do canto coral como prática cultural social no protestantismo brasileiro. O texto direciona sua atenção para o canto coral e suas idiossincrasias a partir das bases da sociologia de Pierre Bourdieu e de sua aplicação às discussões sociológicas sobre o gosto como realidade social definida por tensões criadas no meio cultural, as quais se aplicam os conceitos de campo, habitus e capital. A prática cultural do canto coral é apresentada e um histórico é traçado desde suas origens até os dias atuais. O foco recai sobre os usos do canto coral pelas mais diferentes vertentes do protestantismo a fim de acompanhar os aspectos que se mostraram preponderantes quando da chegada dessa prática cultural ao Brasil e compreender fatos e personagens que fizeram com que o canto coral evangélico brasileiro tomasse os rumos históricos que o levaram a ocupar um posto de alto valor cultural dentro da cultura protestante no Brasil. Posteriormente, verificam-se os abalos ao canto coral causados por mudanças sociais que recaracterizaram profundamente o gosto musical no protestantismo pátrio. A discussão se encaminha para a análise das lutas que essas novas configurações estão estabelecendo dentro do campo religioso e às expressões dessas batalhas dentro de um mercado de bens simbólicos que as comporta e orienta. Visando determinar os sujeitos envolvidos, são apresentados os principais adversários do canto coral no campo evangélico brasileiro e, por fim, são delineados três projetos que, nas últimas décadas, têm alcançado impacto dentro do meio protestante como defensores do canto coral.

A cidade se desenvolve a partir de um núcleo denominado centro, na direção de círculos periféricos. As populações instaladas nestas zonas de fronteiras vivenciam e estabelecem relações próprias com o espaço que ocupam e com o núcleo denominado centro. Em São Paulo, é plausível que se reconheça não há uma uniformidade na população urbana, mas populações urbanas, marcadas por características plurais e significativos contrastes entre si, inclusive nas manifestações de fenômenos religiosos.A zona leste da cidade de São Paulo é objeto da análise desta pesquisa apresentada em quatro capítulos, observada em seus diferentes aspectos, identificando-se inclusive, que há na própria região geográfica marcada como ZL, duas disposições, a saber: ZL 1 e ZL 2, que por si só evidenciam o contraste na zona de fronteira. O olhar específico para este espaço é delimitado pelo tempo, entre 1990-2000 e pela referência da pesquisa que se propõe identificar e analisar as Práticas Pastorais das Igrejas Batistas residentes nesta Zona. Na análise das Práticas Pastorais no viés da evangelização e expansão missionária, elabora-se a pesquisa especificamente sobre três comunidades, a saber: Primeira Igreja Batista da Penha, Igreja Batista em Vila Salete e Primeira Igreja Evangélica Batista em Guaianases.Parte-se não de um referencial externo à comunidade como instrumental de análise, mas suas respectivas propostas de Práticas Pastorais registradas em seus documentos oficiais como atas, anuários e informativos dominicais, os quais serviram como fonte documental. As fronteiras urbanas são, por natureza, inovadoras e caóticas, sendo exatamente por isso o espaço mais adequado para igrejas criarem e desenvolverem Práticas Pastorais próprias e conseqüentes, sem delimitadores à inovação de toda a ordem, configurando-se como Igreja sem Fronteiras. Em caso contrário à motivação inovadora, rompendo-se o diálogo e a vivência com o meio urbano fronteiriço, excluindo-se o ambiente de contraste que se oferece à igreja, dá-se a gênese ao vazio, à desesperança, a Fronteiras sem Igreja.(AU)

Este estudo analisa a mídia impressa local produzida no lado brasileiro da Tríplice Fronteira, região limítrofe entre Brasil, Paraguai e Argentina e toma como exemplar o jornal brasileiro A Gazeta do Iguaçu, editado na cidade de Foz do Iguaçu, PR, que compõe a área trinacional. Os principais objetivos são analisar as características desta mídia local, a incidência de notícias sobre a fronteira, o relacionamento daquele veículo com as comunidades e com os grupos humanos que vivem na região, bem como o grau de influência e o nível de comprometimento do jornal com a política de Foz do Iguaçu. A base teóricometodológica constituiu-se de bibliografia sobre a questão global- local, dos estudos sobre multiculturalismo e diversidade cultural (estudos culturais), e dos questionamentos acerca de comunicação local e regional. As técnicas utilizadas foram, além da revisão bibliográfica, a análise de conteúdo do jornal A Gazeta do Iguaçu e entrevistas semi-estruturadas. Dentre as conclusões encontradas, verificou-se que a cobertura da fronteira é uma pauta prioritária, mas que ao mesmo tempo, perde espaço para o jornalismo de opinião, através das diversas colunas de opinião e sociais, que ocupam a maior parte do noticiário cotidiano local.(AU)

Automatic Term Recognition (ATR) is a fundamental processing step preceding more complex tasks such as semantic search and ontology learning. From a large number of methodologies available in the literature only a few are able to handle both single and multi-word terms. In this paper we present a comparison of five such algorithms and propose a combined approach using a voting mechanism. We evaluated the six approaches using two different corpora and show how the voting algorithm performs best on one corpus (a collection of texts from Wikipedia) and less well using the Genia corpus (a standard life science corpus). This indicates that choice and design of corpus has a major impact on the evaluation of term recognition algorithms. Our experiments also showed that single-word terms can be equally important and occupy a fairly large proportion in certain domains. As a result, algorithms that ignore single-word terms may cause problems to tasks built on top of ATR. Effective ATR systems also need to take into account both the unstructured text and the structured aspects and this means information extraction techniques need to be integrated into the term recognition process.

The title of Juana Castro’s poetry book publshed in 1978, Cóncava mujer — Concave woman — expresses the hollow nature of the social female subject. From Juana Castro’s point of view, this female social concavity is only allowed to transformed itself into its opposite, the convex women which clearly represents the reproductive role of the female body. These two extreme roles assigned to women, hollowness or maternity, are the poetic paradigms for Juana Castro’s two poetry books analised in this article. As if we were presented with the two sides of a coin, Cóncava mujer and Del dolor y las alas – On anguish and wings--(1982) reflect the author´s concious realisation of the above-mentioned female duality as a defined and percieved subject by male society. Each poetry book, however, respond to two different personal moments, and each result in two different ways of conceiving poetic language. On one hand, the poetic subject of Cóncava mujer emerges with all its force as a feminist voice whose goal is the attack of all aspects of the patriarchal society as the cause of the female concavity. On the other hand, in Del dolor y las alas the poetic voice unfolds her motherhood as both loss and creation: the death of Juana Castro’s son makes the poetic subject incomplete, and therefore a concave one; whereas the poetic discourse appears as the perfect way to occupy the empty space left by the son’s death.

The way in which employed senior elites in English local government exercise their agency in the practice of local democracy and local governance is considered in this thesis. The research posits the notion that elite Officers act as Local Democracy Makers as they draw on their own traditions and ideologies in responding to the dilemmas of changing policy and politics in the public realm. The study is located in the latter part of New Labour?s term of office and applies an interpretive and reflexive approach to three studies of the exercise of well being powers. The approach is one of applied ethnography through the examination of literature reviews, interviews and observations of decisions taken in the exercise of the powers of economic, environmental and social well-being are used to examine how and why the Local Democracy Makers make sense of their world in the way that they do. The research suggests that, despite prevailing narratives, local governance arrangements depend on a system of hierarchy, employed elites and local politics. The challenges of re-configuring local democracy and attempts at "hollowing out" the state have secured an influential role for the non-elected official. How officials interpret, advise, mediate and manage the exercise of local governance and local democracy presents a challenge to assumptions that public services are governed beyond or without local government. New narratives and reflections on the role of the local government Officer and the marginalisation of the elected Councillor are presented in the research. In particular, how the senior elite occupy managerial, strategic and political roles as Local Democracy Makers, offers an insight into the agency of strategic actors in localities. Consequently, the success of changes in public policy is materially influenced by how the practitioner responds to such dilemmas. The thesis concludes by suggesting that integral to the design and success of public policy implementation is the role of the Officer, and especially those practitioners that advise governing arrangements and democratic practice.

Two key issues defined the focus of this research in manufacturing plasmid DNA for use In human gene therapy. First, the processing of E.coli bacterial cells to effect the separation of therapeutic plasmid DNA from cellular debris and adventitious material. Second, the affinity purification of the plasmid DNA in a Simple one-stage process. The need arises when considering the concerns that have been recently voiced by the FDA concerning the scalability and reproducibility of the current manufacturing processes in meeting the quality criteria of purity, potency, efficacy, and safety for a recombinant drug substance for use in humans. To develop a preliminary purification procedure, an EFD cross-flow micro-filtration module was assessed for its ability to effect the 20-fold concentration, 6-time diafiltration, and final clarification of the plasmid DNA from the subsequent cell lysate that is derived from a 1 liter E.coli bacterial cell culture. Historically, the employment of cross-flow filtration modules within procedures for harvesting cells from bacterial cultures have failed to reach the required standards dictated by existing continuous centrifuge technologies, frequently resulting in the rapid blinding of the membrane with bacterial cells that substantially reduces the permeate flux. By challenging the EFD module, containing six helical wound tubular membranes promoting centrifugal instabilities known as Dean vortices, with distilled water between the Dean number's of 187Dn and 818Dn,and the transmembrane pressures (TMP) of 0 to 5 psi. The data demonstrated that the fluid dynamics significantly influenced the permeation rate, displaying a maximum at 227Dn (312 Imh) and minimum at 818Dn (130 Imh) for a transmembrane pressure of 1 psi. Numerical studies indicated that the initial increase and subsequent decrease resulted from a competition between the centrifugal and viscous forces that create the Dean vortices. At Dean numbers between 187Dn and 227Dn , the forces combine constructively to increase the apparent strength and influence of the Dean vortices. However, as the Dean number in increases above 227 On the centrifugal force dominates the viscous forces, compressing the Dean vortices into the membrane walls and reducing their influence on the radial transmembrane pressure i.e. the permeate flux reduced. When investigating the action of the Dean vortices in controlling tile fouling rate of E.coli bacterial cells, it was demonstrated that the optimum cross-flow rate at which to effect the concentration of a bacterial cell culture was 579Dn and 3 psi TMP, processing in excess of 400 Imh for 20 minutes (i.e., concentrating a 1L culture to 50 ml in 10 minutes at an average of 450 Imh). The data demonstrated that there was a conflict between the Dean number at which the shear rate could control the cell fouling, and the Dean number at which tile optimum flux enhancement was found. Hence, the internal geometry of the EFD module was shown to sub-optimal for this application. At 579Dn and 3 psi TMP, the 6-fold diafiltration was shown to occupy 3.6 minutes of process time, processing at an average flux of 400 Imh. Again, at 579Dn and 3 psi TMP the clarification of the plasmid from tile resulting freeze-thaw cell lysate was achieved at 120 Iml1, passing 83% (2,5 mg) of the plasmid DNA (6,3 ng μ-1 10.8 mg of genomic DNA (∼23,00 Obp, 36 ng μ-1 ), and 7.2 mg of cellular proteins (5-100 kDa, 21.4 ngμ-1 ) into the post-EFD process stream. Hence the EFD module was shown to be effective, achieving the desired objectives in approximately 25 minutes. On the basis of its ability to intercalate into low molecular weight dsDNA present in dilute cell lysates, and be electrophoresed through agarose, the fluorophore PicoGreen was selected for the development of a suitable dsDNA assay. It was assesseel for its accuracy, and reliability, In determining the concentration and identity of DNA present in samples that were eleclrophoresed through agarose gels. The signal emitted by intercalated PicoGreen was shown to be constant and linear, and that the mobility of the PicaGreen-DNA complex was not affected by the intercalation. Concerning the secondary purification procedure, various anion-exchange membranes were assessed for their ability to capture plasmid DNA from the post-EFD process stream. For a commercially available Sartorius Sartobind Q15 membrane, the reduction in the equilibriumbinding capacity for  ctDNA in buffer of increasing ionic demonstrated that DNA was being.adsorbed by electrostatic  interactions only. However, the problems associated with fluid distribution across the membrane demonstrated that the membrane housing was the predominant cause of the .erratic breakthrough curves. Consequently, this would need to be rectified before such a membrane could be integrated into the current system, or indeed be scaled beyond laboratory scale. However, when challenged with the process material, the data showed that considerable quantities of protein (1150 μg) were adsorbed preferentially to the plasmid DNA (44 μg). This was also shown for derived Pall Gelman UltraBind US450 membranes that had been functionalised by varying molecular weight poly-L~lysine and polyethyleneimine ligands. Hence the anion-exchange membranes were shown to be ineffective in capturing plasmid DNA from the process stream. Finally, work was performed to integrate a sequence-specific DNA·binding protein into a single-stage DNA chromatography, isolating plasmid DNA from E.coli cells whilst minimising the contamination from genomic DNA and cellular protein. Preliminary work demonstrated that the fusion protein was capable of isolating pUC19 DNA into which the recognition sequence for the fusion-protein had been inserted (pTS DNA) when in the presence of the conditioned process material. Althougth the pTS recognition sequence differs from native pUC19 sequences by only 2 bp, the fusion protein was shown to act as a highly selective affinity ligand for pTS DNA alone. Subsequently, the scale of the process was scaled 25-fold and positioned directly following the EFD system. In conclusion, the integration of the EFD micro-filtration system and zinc-finger affinity purification technique resulted in the capture of approximately 1 mg of plasmid DNA was purified from 1L of E.coli  culture in a simple two stage process, resulting in the complete removal of genomic DNA and 96.7% of cellular protein in less than 1 hour of process time.
