This thesis starts showing the main characteristics and application fields of the AlGaN/GaN HEMT technology, focusing on reliability aspects essentially due to the presence of low frequency dispersive phenomena which limit in several ways the microwave performance of this kind of devices. Based on an equivalent voltage approach, a new low frequency device model is presented where the dynamic nonlinearity of the trapping effect is taken into account for the first time allowing considerable improvements in the prediction of very important quantities for the design of power amplifier such as power added efficiency, dissipated power and internal device temperature. An innovative and low-cost measurement setup for the characterization of the device under low-frequency large-amplitude sinusoidal excitation is also presented. This setup allows the identification of the new low frequency model through suitable procedures explained in detail. In this thesis a new non-invasive empirical method for compact electrothermal modeling and thermal resistance extraction is also described. The new contribution of the proposed approach concerns the non linear dependence of the channel temperature on the dissipated power. This is very important for GaN devices since they are capable of operating at relatively high temperatures with high power densities and the dependence of the thermal resistance on the temperature is quite relevant. Finally a novel method for the device thermal simulation is investigated: based on the analytical solution of the tree-dimensional heat equation, a Visual Basic program has been developed to estimate, in real time, the temperature distribution on the hottest surface of planar multilayer structures. The developed solver is particularly useful for peak temperature estimation at the design stage when critical decisions about circuit design and packaging have to be made. It facilitates the layout optimization and reliability improvement, allowing the correct choice of the device geometry and configuration to achieve the best possible thermal performance.

The aim of this study is to evaluate the effects of the pulsed electromagnetic fields (PEMFS) on pain relief and functional capacity of dogs with osteoarthritis by which a single centre study prospective clinical trial. PEMFs are non ionized, athermic and time varying electromagnetic fields that has been successfully used for the treatment of osteoarthritis in human thanks to their chondroprotective, antinflammatory and analgesic property. 20 dogs were treated with PEMFs , 3 times per week for a total of 20 sessions.We found beneficial effects on pain relief and lameness in the absence of adverse effect. The decrease of pain impacted positively on the health-dogs related quality of life and the grade of satisfaction of their owner was very high. The benefits were obvious at half therapy and lasted for a medium long time. This is the first published report concerning PEMFs treatment on canine osteoarthtitis. The result of this study proves that PEMFs is a non –invasive remedy, lacking in adverse effect , easy to employ and useful for controlling pain and inflammation associated with osteoarthritis.

Background. Neuroblastoma is the most deadly solid tumor of childhood. In the 25% of cases it is associated with MYCN amplification (MA), resulting in the disregulation of several genes involved in cancer progression, chemotherapy resistance and poor prognosis causing the disregulation of several genes involved in cancer progression and chemotherapy resistance and resulting in a poor prognosis. Moreover, in this contest, therapy-related p53 mutations are frequently found in relapsed cases conferring an even stronger aggressiveness. For this reason, the actual therapy requires new antitumor molecules. Therefore, rapid, accurate, and reproducible preclinical models are needed to evaluate the evolution of the different subtypes and the efficacy of new pharmacological strategies. Procedures. We report the real-time tumorigenesis of MA Neuroblastoma mouse models: transgenic TH-MYCN mice and orthotopic xenograft models with either p53wt or p53mut, by non-invasive micro PET and bioluminescent imaging, respectively. Characterization of MYCN amplification and expression was performed on every collected sample. We tested the efficacy of a new MYCN inhibitor in vitro and in vivo. Results. MicroPET in TH-MYCN mice permitted the identification of Neuroblastoma at an early stage and offered a sensitive method to follow metabolic progression of tumors. The MA orthotopic model harboring multitherapy-related p53 mutations showed a shorter latency and progression and a stronger aggressiveness respect to the p53wt model. The presence of MA and overexpression was confirmed in each model and we saw a better survival in the TH-MYCN homozigous mice treated with the inhibitor. Conclusions. The mouse models obtained show characteristics of non-invasiveness, rapidity and sensitivity that make them suitable for the in vivo preclinical study of MA-NB. In particular, our firstly reported p53mut BLI xenograft orthotopic mouse model offers the possibility to evaluate the role of multitherapy-related p53 mutations and to validate new p53 independent therapies for this highly aggressive Neuroblastoma subtype. Moreover, we have shown potential clinical suitability of an antigene strategy through its cellular and molecular activity, ability to specifically inhibit transcription and in vivo efficacy with no evidence of toxicity.

A numerical investigation of dielectric barrier discharge aimed to simulate the electro hydro dynamic interaction is presented. A discharge drift diffusive model according to the Townsend avalanche is described and used to duplicate the plasma kinetics of a DBD actuator. The discharge characteristics dependence upon dielectric material and applied voltage are simulated and the EHD force field according to a simplified approach is presented and discussed. The coupling of DBD results with a fluid dynamic code is also shown. Finally, a new non invasive diagnostic technique for EHD interaction based on Schlieren imaging is computationally validated.

Im ersten Teil der Arbeit wurde das Bindungsverhalten von Annexin A1 und Annexin A2t an festkörperunterstützte Lipidmembranen aus POPC und POPS untersucht. Für beide Proteine konnte mit Hilfe der Fluoreszenzmikroskopie gezeigt werden, dass irreversible Bindung nur in Anwesenheit von POPS auftritt. Durch rasterkraftmikroskopische Aufnahmen konnte die laterale Organisation der Annexine auf der Lipidmembran dargestellt werden. Beide Proteine lagern sich in Form lateraler Aggregate (zweidimensionale Domänen) auf der Oberfläche an, außerdem ist der Belegungsgrad und die Größe der Domänen von der Membranzusammensetzung und der Calciumkonzentration abhängig. Mit zunehmendem POPS-Gehalt und Calciumkonzentration steigt der Belegungsgrad an und der mittlere Domänenradius wird kleiner. Diese Ergebnisse konnten in Verbindung mit detaillierten Bindungsstudien des Annexins A1 mit der Quarzmikrowaage verwendet werden, um ein Bindungsmodell auf Basis einer heterogenen Oberfläche zu entwickeln. Auf einer POPC-reichen Matrix findet reversible Adsorption statt und auf POPS-reichen Domänen irreversible Adsorption. Durch die Anpassung von dynamischen Monte Carlo-Simulationen basierend auf einer zweidimensionalen zufälligen sequentiellen Adsorption konnten Erkenntnisse über die Membranstruktur und die kinetischen Ratenkonstanten in Abhängigkeit von der Calciumkonzentration und der Inkubationszeit des Proteins gewonnen werden. Die irreversible Bindung ist in allen Calciumkonzentrationsbereichen schneller als die reversible. Außerdem zeigt die irreversible Adsorption eine deutlich stärkere Abhängigkeit von der Calciumkonzentration. Ein kleinerer Belegungsgrad bei niedrigen Ca2+-Gehalten ist hauptsächlich durch die Abnahme der verfügbaren Bindungsplätze auf der Oberfläche zu erklären. Die gute Übereinstimmung der aus den Monte Carlo-Simulationen erhaltenen Domänenstrukturen mit den rasterkraftmikroskopischen Aufnahmen und die Tatsache, dass sich die simulierten Resonanzfrequenzverläufe problemlos an die experimentellen Kurven aus den QCM-Messungen anpassen ließen, zeigt die gute Anwendbarkeit des entwickelten Simulationsprogramms auf die Adsorption von Annexin A1. Die Extraktion der kinetischen Parameter aus dem zweidimensionalen RSA-Modell ist mit Sicherheit einem einfachen Langmuir-Ansatz überlegen. Bei einem Langmuir-Modell erfolgt eine integrale Erfassung einer einzelnen makroskopischen Geschwindigkeitskonstante, während durch das RSA-Modell eine differenzierte Betrachtung des reversiblen und irreversiblen Bindungsprozesses möglich ist. Zusätzlich lassen sich mikroskopische Informationen über die Oberflächenbeschaffenheit gewinnen. Im zweiten Teil der Arbeit wurde das thermotrope Phasenverhalten von festkörperunterstützten Phospholipidbilayern untersucht. Dazu wurden mikrostrukturierte, frei stehende Membranstreifen präpariert und mit Hilfe der bildgebenden Ellipsometrie untersucht. Dadurch konnten die temperaturabhängigen Verläufe der Schichtdicke und der lateralen Membranausdehnung parallel beobachtet werden. Die ermittelten Phasenübergangstemperaturen von DMPC, diC15PC und DPPC lagen 2 - 3 °C oberhalb der Literaturwerte für vesikuläre Systeme. Außerdem wurde eine deutliche Verringerung der Kooperativität der Phasenumwandlung gefunden, was auf einen großen Einfluss des Substrats bei den festkörperunterstützten Lipidmembranen schließen lässt. Zusätzlich wurde ein nicht systematischer Zusammenhang der Ergebnisse von der Oberflächenpräparation gefunden, der es unabdingbar macht, bei Untersuchungen von festkörperunterstützten Substraten einen internen Standard einzuführen. Bei der Analyse des thermotropen Phasenübergangsverhaltens von DMPC/Cholesterol - Gemischen wurde daher die individuelle Adressierbarkeit der strukturierten Lipidmembranen ausgenutzt und ein Lipidstreifen aus reinem DMPC als Standard verwendet. Auf diese Weise konnte gezeigt werden, dass das für Phospholipide typische Phasenübergangsverhalten ab 30 mol% Cholesterol in der Membran nicht mehr vorhanden ist. Dies ist auf die Bildung einer nur durch höhere Sterole induzierten fluiden Phase mit hoch geordneten Acylketten zurückzuführen. Abschließend konnte durch die Zugabe von Ethanol zu einer mikrostrukturierten DMPC-Membran die Bildung eines interdigitierten Bilayers nachgewiesen werden. Die bildgebende Ellipsometrie ist eine sehr gute Methode zur Untersuchung festkörperunterstützter Lipidmembranen, da sie über ein sehr gutes vertikales und ein ausreichendes laterales Auflösungsvermögen besitzt. Sie ist darin zwar einem Rasterkraftmikroskop noch unterlegen, besitzt dafür aber eine einfachere Handhabung beim Umgang mit Flüssigkeiten und in der Temperierung, eine schnellere Bildgebung und ist als optische Methode nicht-invasiv.

Sowohl in der Natur als auch in der Industrie existieren thermisch induzierte Strömungen. Von Interesse für diese Forschungsarbeit sind dabei die Konvektionen im Erdmantel sowie in den Glasschmelzwannen. Der dort stattfindende Materialtransport resultiert aus Unterschieden in der Dichte, der Temperatur und der chemischen Konzentration innerhalb des konvektierenden Materials. Um das Verständnis für die ablaufenden Prozesse zu verbessern, werden von zahlreichen Forschergruppen numerische Modellierungen durchgeführt. Die Verifikation der dafür verwendeten Algorithmen erfolgt meist über die Analyse von Laborexperimenten. Im Vordergrund dieser Forschungsarbeit steht die Entwicklung einer Methode zur Bestimmung der dreidimensionalen Temperaturverteilung für die Untersuchung von thermisch induzierten Strömungen in einem Versuchsbecken. Eine direkte Temperaturmessung im Inneren des Versuchsmaterials bzw. der Glasschmelze beeinflusst allerdings das Strömungsverhalten. Deshalb wird die geodynamisch störungsfrei arbeitende Impedanztomographie verwendet. Die Grundlage dieser Methode bildet der erweiterte Arrhenius-Zusammenhang zwischen Temperatur und spezifischer elektrischer Leitfähigkeit. Während der Laborexperimente wird ein zähflüssiges Polyethylenglykol-Wasser-Gemisch in einem Becken von unten her erhitzt. Die auf diese Weise generierten Strömungen stellen unter Berücksichtigung der Skalierung ein Analogon sowohl zu dem Erdmantel als auch zu den Schmelzwannen dar. Über mehrere Elektroden, die an den Beckenwänden installiert sind, erfolgen die geoelektrischen Messungen. Nach der sich anschließenden dreidimensionalen Inversion der elektrischen Widerstände liegt das Modell mit der Verteilung der spezifischen elektrischen Leitfähigkeit im Inneren des Versuchsbeckens vor. Diese wird mittels der erweiterten Arrhenius-Formel in eine Temperaturverteilung umgerechnet. Zum Nachweis der Eignung dieser Methode für die nichtinvasive Bestimmung der dreidimensionalen Temperaturverteilung wurden mittels mehrerer Thermoelemente an den Beckenwänden zusätzlich direkte Temperaturmessungen durchgeführt und die Werte miteinander verglichen. Im Wesentlichen sind die Innentemperaturen gut rekonstruierbar, wobei die erreichte Messgenauigkeit von der räumlichen und zeitlichen Auflösung der Gleichstromgeoelektrik abhängt.

The present thesis is concerned with the development of novel cocaine-derived dopamine transporter ligands for the non-invasive exploration of the striatal and extra-striatal dopamine transporter (DAT) in living systems. The presynaptic dopamine transporter acquires an important function within the mediation of dopaminergic signal transduction. Its availability can serve as a measure for the overall integrity of the dopaminergic system. The DAT is upregulated in early Parkinson’s disease (PD), resulting in an increased availability of DAT-binding sites in the striatal DAT domains. Thereby, DAT imaging has become an important routine diagnostic tool for the early diagnosis of PD in patients, as well as for the differentiation of PD from symptomatically similar medical conditions. Furthermore, the dopaminergic system is involved in a variety of psychiatric diseases. In this regard, DAT-selective imaging agents may provide detailed insights into the scientific understanding of the biochemical background of both, the progress as well as the origins of the symptoms. DAT-imaging may also contribute to the determination of the dopaminergic therapeutic response for a given medication and thereby contribute to more convenient conditions for the patient. From an imaging point of view, the former demands a high availability of the radioactive probe to facilitate broad application of the modality, whereas the latter profits from short-lived probes, suitable for multi-injection studies. Therefore, labelling with longer-lived 18F-fluoride and in particular the generator nuclide 68Ga is worthwhile for clinical routine imaging. In contrast, the introduction of a 11C-label is a prerequisite for detailed scientific studies of neuronal interactions. The development of suitable DAT-ligands for medical imaging has often been complicated by the mixed binding profile of many compounds that that interact with the DAT. Other drawbacks have included high non-specific binding, extensive metabolism and slow accumulation in the DAT-rich brain areas. However, some recent examples have partially overcome the mentioned complications. Based on the structural speciality of these leads, novel ligand structures were designed and successfully synthesised in the present work. A structure activity relationship (SAR) study was conducted wherein the new structural modifications were examined for their influence on DAT-affinity and selectivity. Two of the compounds showed improvements in in vitro affinity for the DAT as well as selectivity versus the serotonin transporter (SERT) and norepinephrine transporter (NET). The main effort was focussed on the high-affinity candidate PR04.MZ, which was subsequently labelled with 18F and 11C in high yield. An initial pharmacological characterisation of PR04.MZ in rodents revealed highly specific binding to the target brain structures. As a result of low non-specific binding, the DAT-rich striatal area was clearly visualised by autoradiography and µPET. Furthermore, the radioactivity uptake into the DAT-rich brain regions was rapid and indicated fast binding equilibrium. No radioactive metabolite was found in the rat brain. [18F]PR04.MZ and [11C]PR04.MZ were compared in the primate brain and the plasma metabolism was studied. It was found that the ligands specifically visualise the DAT in high and low density in the primate brain. The activity uptake was rapid and quantitative evaluation by Logan graphical analysis and simplified reference tissue model was possible after a scanning time of 30 min. These results further reflect the good characteristics of PR04.MZ as a selective ligand of the neuronal DAT. To pursue 68Ga-labelling of the DAT, initial synthetic studies were performed as part of the present thesis. Thereby, a concept for the convenient preparation of novel bifunctional chelators (BFCs) was developed. Furthermore, the suitability of novel 1,4,7-triazacyclononane based N3S3-type BFCs for biomolecule-chelator conjugates of sufficient lipophilicity for the penetration of the blood-brain-barrier was elucidated.

The evaluation of chronic activity of the hypothalamic-pituitary-adrenal (HPA) axis is critical for determining the impact of chronic stressful situations. The potential use of hair glucocorticoids as a non-invasive, retrospective, biomarker of long term HPA activity is of great interest, and it is gaining acceptance in humans and animals. However, there are still no studies in literature examining hair cortisol concentration in pigs and corticosterone concentration in laboratory rodents. Therefore, we developed and validated, for the first time, a method for measuring hair glucocorticoids concentration in commercial sows and in Sprague-Dawley rats. Our preliminary data demonstrated: 1) a validated and specific washing protocol and extraction assay method with a good sensitivity in both species; 2) the effect of the reproductive phase, housing conditions and seasonality on hair cortisol concentration in sows; 3) similar hair corticosterone concentration in male and female rats; 4) elevated hair corticosterone concentration in response to chronic stress manipulations and chronic ACTH administration, demonstrating that hair provides a good direct index of HPA activity over long periods than other indirect parameters, such adrenal or thymus weight. From these results we believe that this new non-invasive tool needs to be applied to better characterize the overall impact in livestock animals and in laboratory rodents of chronic stressful situations that negatively affect animals welfare. Nevertheless, further studies are needed to improve this methodology and maybe to develop animal models for chronic stress of high interest and translational value in human medicine.

Soltanto interrogando l'edificio si è in grado di comprenderne le singolarità qualitative, la vulnerabilità ed i punti critici, per agire dove necessario con tecniche compatibili e non invasive seguendo il criterio del minimo intervento. Gli interventi progettuali di miglioramento strutturale proposti si pongono a coronamento del percorso conoscitivo di ricerca svolto.

In recent years, thanks to the technological advances, electromagnetic methods for non-invasive shallow subsurface characterization have been increasingly used in many areas of environmental and geoscience applications. Among all the geophysical electromagnetic methods, the Ground Penetrating Radar (GPR) has received unprecedented attention over the last few decades due to its capability to obtain, spatially and temporally, high-resolution electromagnetic parameter information thanks to its versatility, its handling, its non-invasive nature, its high resolving power, and its fast implementation. The main focus of this thesis is to perform a dielectric site characterization in an efficient and accurate way studying in-depth a physical phenomenon behind a recent developed GPR approach, the so-called early-time technique, which infers the electrical properties of the soil in the proximity of the antennas. In particular, the early-time approach is based on the amplitude analysis of the early-time portion of the GPR waveform using a fixed-offset ground-coupled antenna configuration where the separation between the transmitting and receiving antenna is on the order of the dominant pulse-wavelength. Amplitude information can be extracted from the early-time signal through complex trace analysis, computing the instantaneous-amplitude attributes over a selected time-duration of the early-time signal. Basically, if the acquired GPR signals are considered to represent the real part of a complex trace, and the imaginary part is the quadrature component obtained by applying a Hilbert transform to the GPR trace, the amplitude envelope is the absolute value of the resulting complex trace (also known as the instantaneous-amplitude). Analysing laboratory information, numerical simulations and natural field conditions, and summarising the overall results embodied in this thesis, it is possible to suggest the early-time GPR technique as an effective method to estimate physical properties of the soil in a fast and non-invasive way.

Introduction. Glycomic analysis allows investigating on the global glycome within body fluids (as serum/plasma), this could eventually lead to identify new types of disease biomarkers, or as in this study, biomarkers of human aging studying specific aging models. Recent studies demonstrated that the plasma N-glycome is modified during human aging, suggesting that measurements of log-ratio of two serum/plasma N-glycans (NGA2F and NA2F), named GlycoAge test could provide a non-invasive biomarker of aging. Down syndrome (DS) is a genetic disorder in which multiple major aspects of senescent phenotype occur much earlier than in healthy age-matched subjects and has been often defined as an accelerated aging syndrome. The aim of this study was to compare plasma N-glycome of patients affected by DS with age- and sex matched non-affected controls, represented by their siblings (DSS), in order to assess if DS is characterized by a specific N-glycomic pattern. Therefore, in order to investigate if N-glycans changes that occur in DS were able to reveal an accelerated aging in DS patients, we enrolled the mothers (DSM) of the DS and DSS, representing the non-affected control group with a different chronological age respect to DS. We applied two different N-glycomics approaches on the same samples: first, in order to study the complete plasma N-glycome we applied a new high-sensitive protocol based on a MALDI-TOF-MS approach, second, we used DSA-FACE technology. Results: MALDI-TOF/MS analysis detected a specific N-glycomics signature for DS, characterized by an increase of fucosylated and bisecting species. Moreover, in DS the abundance of agalactosylated (as NA2F) species was similar or higher than their mothers. The measurement of GlycoAge test with DSA-FACE, validated also by MALDI-TOF, demonstrated a strongly association with age, moreover in DS, it’s value was similar to their mothers, and significantly higher than their age- and sex matched not-affected siblings

La sindrome di Cushing (SC) è una delle endocrinopatie più comuni nel cane. La diagnosi richiede l'integrazione di anamnesi, segnalamento, segni clinici, esami ematobiochimici, test endocrini specifici e diagnostica per immagini . Nel corso degli anni diversi sono i principi attivi testati per la terapia della SC del cane. In passato, il mitotane è stato il farmaco più utilizzato, sebbene il suo uso risulti complicato e non privo di potenziali effetti collaterali. Recentemente, il trilostano ha dimostrato di essere un trattamento efficace per il controllo dei sintomi ed è stato approvato per tale uso nel cane. Al fine di testare metodiche non invasive per la diagnosi di SC nel cane abbiamo valutato le concentrazioni di cortisolo nel pelo (HCC) .Queste risultavano significativamente più elevate nei cani con SC rispetto ai cani sani e malati. Questo test può essere quindi considerato una procedura diagnostica non invasiva in cani con un elevato sospetto di SC. Inoltre, a causa della difficile reperibilità dell’ACTH esogeno sono state valutate le concentrazioni di cortisolo basale come strumento di monitoraggio in cani con SC trattati con trilostano. Tuttavia la singola valutazione del cortisolo basale non rappresenta un parametro efficace ed accurato per il monitoraggio della terapia con trilostano. Sono stati inoltre valutati i fattori prognostici in cani con SC alla diagnosi. L' iperfosfatemia è risultata un riscontro comune nei cani SC, rappresentando un fattore prognostico negativo. La terapia chirurgica non è una procedura di routine nella SC del cane, tuttavia abbiamo descritto l'approccio di ipofisectomia transsfenoidale in un Galgo spagnolo di 8 anni con SC . Il cane è stato sottoposto per due volte ad ipofisectomia transsfenoidale che ha permesso di rimuovere completamente il macroadenoma ipofisario. In conclusione, questi studi ci hanno permesso di indagare alcuni aspetti patogenetici, clinici e diagnostici della SC del cane.

Gleno-humeral joint (GHJ) is the most mobile joint of the human body. This is related to theincongr uence between the large humeral head articulating with the much smaller glenoid (ratio 3:1). The GHJ laxity is the ability of the humeral head to be passively translated on the glenoid fossa and, when physiological, it guarantees the normal range of motion of the joint. Three-dimensional GHJ linear displacements have been measured, both in vivo and in vitro by means of different instrumental techniques. In vivo gleno-humeral displacements have been assessed by means of stereophotogrammetry, electromagnetic tracking sensors, and bio-imaging techniques. Both stereophotogrammetric systems and electromagnetic tracking devices, due to the deformation of the soft tissues surrounding the bones, are not capable to accurately assess small displacements, such as gleno-humeral joint translations. The bio-imaging techniques can ensure for an accurate joint kinematic (linear and angular displacement) description, but, due to the radiation exposure, most of these techniques, such as computer tomography or fluoroscopy, are invasive for patients. Among the bioimaging techniques, an alternative which could provide an acceptable level of accuracy and that is innocuous for patients is represented by magnetic resonance imaging (MRI). Unfortunately, only few studies have been conducted for three-dimensional analysis and very limited data is available in situations where preset loads are being applied. The general aim of this doctoral thesis is to develop a non-invasive methodology based on open-MRI for in-vivo evaluation of the gleno-humeral translation components in healthy subjects under the application of external loads.

Non-invasive molecular-imaging technologies are playing a key role in drug discovery, development and delivery. Positron Emission Tomography (PET) is such a molecular imaging technology and a powerful tool for the observation of various deceases in vivo. However, it is limited by the availability of vectors with high selectivity to the target and radionuclides with a physical half-life which matches the biological half-life of the observed process. The 68Ge/68Ga radionuclide generator makes the PET-nuclide anywhere available without an on-site cyclotron. Besides the perfect availability 68Ga shows well suited nuclide properties for PET, but it has to be co-ordinated by a chelator to introduce it in a radiopharmaceuticals.rnHowever, the physical half-life of 68Ga (67.7 min) might limit the spectrum of clinical applications of 68Ga-labelled radiodiagnostics. Furthermore, 68Ga-labelled analogues of endoradiotherapeuticals of longer biological half-live such as 90Y- or 177Lu-labeled peptides and proteins cannot be used to determine individual radiation dosimetry directly. rnThus, radionuclide generator systems providing positron emitting daughters of extended physical half-life are of renewed interest. In this context, generator-derived positron emitters with longer physical half-life are needed, such as 72As (T½ = 26 h) from the 72Se/72As generator, or 44Sc (T½ = 3.97 h) from the 44Ti/44Sc generator.rnIn this thesis the implementation of radioactive gallium-68 and scandium-44 for molecular imaging and nuclear medical diagnosis, beginning with chemical separation and purification of 44Ti as a radionuclide mother, investigation of pilot generators with different elution mode, building a prototype generator, development and investigation of post-processing of the generator eluate, its concentration and further purification, the labeling chemistry under different conditions, in vitro and in vivo studies of labeled compounds and, finally, in vivo imaging experiments are described.

Ad oggi, grazie al progresso tecnologico, sono stati realizzati accuratissimi rivelatori di radiazione infrarossa, rendendo possibili misure altamente precise. L’impiego massiccio dell’indagine termografica infrarossa è basato sull’emissione termica da parte della materia. Tutta la materia a temperatura superiore dello zero assoluto emette radiazione elettromagnetica con una certa distribuzione spettrale, dipendente dalla temperatura e dalle caratteristiche costitutive. Le capacità emissive di un corpo sono quantificate da un parametro, l’emissività, che confronta i corpi reali con il corpo nero, modellizzazione matematica di un perfetto emettitore. I corpi a temperatura ambiente emettono principalmente nell’infrarosso. Poichè la radiazione infrarossa non è percepibile dall’occhio umano, i corpi a temperature ordinarie non appaiono luminosi e sono visti brillare unicamente di luce riflessa. Solo fornendo calore al sistema, aumentandone cioè la temperatura, l’emissione di picco si sposta verso le frequenze del visibile ed i corpi appaiono luminosi; si pensi ad esempio alle braci della legna bruciata. La possibilità di rivelare la radiazione infrarossa con opportuni dispositivi permette quindi di vedere la “luce” emessa da corpi relativamente freddi. Per questo motivo la termografia trova applicazione nella visione o videoregistrazione notturna o in condizioni di scarsa visibilità. Poichè la distribuzione spettrale dipende dalla temperatura, oltre al picco di emissione anche la quantità di radiazione emessa dipende dalla teperatura, in particolare è proporzionale alla quarta potenza della temperatura. Rilevando la quantità di radiazione emessa da un corpo se ne può dunque misurare la temperatura superficiale, o globale se il corpo è in equilibrio termico. La termometria, ovvero la misura della temperatura dei corpi, può dunque sfruttare la termografia per operare misure a distanza o non invasive.
