The objective of this study was to evaluate the effect of different interval from the beginning of the heat to the ovulation on the fertility of inseminated mares with diluted equine semen, cooled at 20°C and transported. The mares were grouped with the following interval periods: T1 - period less than five days, T2 - period from five to seven days and T3 - period from 8 to 21 days. The conception rates in the first cycle were 53.85 (7/13), 52.17 (12/23) and 66.67% (10/15) for treatments 1, 2 and 3, respectively, and after three cycles, 50.00 (9/18), 48.15 (13/27) and 64.71% (11/17), in the same preceding order. The duration of heat, in the conditions of this experiment, did not influence the fertility of inseminated mares.

One hundred and one estral cycles of 92 Mangalarga Marchador breed mares were analyzed to study the effect of different spermatic concentrations on different reproductive characteristics. The mares were inseminated with diluted equine semen, cooled at 20°C and transported: T1 - concentrations below 250×10 6 (177.58±41.74×10 6 viable spermatozoa per inseminate dose), T2 - concentrations between 250 and 350 10 6 sptz (315.51±20.78×106), and T3 - concentrations above 350×10 6 sptz (477.71±136.83×10 6). The conception rates in the first cycle were 43.75 (7/16), 57.89 (11/19) and 54.55% (18/33) for treatments 1, 2 and 3, respectively. After five cycles, the conception rates were 36.00 (9/25), 44.12 (15/34) and 54.76% (23/42) for treatments 1, 2 and 3, respectively. Concentrations of 177.58±41,74×10 6 viable spermatozoa per inseminate dose had moderate fertility index in a commercial program using cooled and transported semen.

With the objective of identifying predictors of fertility in bulls, the correlation coefficients of the midpiece length of bovine spermatozoa with semen traits and fertility were estimated. Data were obtained from semen samples of 50 crossbred bulls submitted to a progeny test. The midpiece length was determined in 40 midpieces of each bull through a image analysis system (Videoplan). Besides the physical traits before and after semen freezing, the oxygen consumption rate and the cytochemical activity index were observed. The correlation coefficients were low and non-significant (P> 0.05), indicating that the midpiece length is not a good predictor of fertility.

In this study, the aphid Lipaphis erysimi (Kalt) was reared at different temperatures, under laboratory and field conditions, aiming to obtain age-specific life tables. In the laboratory, L. erysimi was fed on kale, Brassica oleracea L. var acephala, and kept in incubators adjusted to 10, 15, 20, 25 and 30°C, 14h photophase and 70±10% RH. The longest mean generation time (T) was observed at 15°C (23.86 days) and the shortest at 30°C (7.18 days), while the smallest net reproductive rate (R0) occurred at 15°C (4.30) and largest one at 25°C (38.29). For the temperatures of 15, 20, 25 and 30°C, the intrinsic rate of natural increase (rm) and the finite rate of increase (λ) were 0.06/1.06, 0.24/1.27, 0.28/1.33 and 0.23/1.25, respectively. The doubling time (DT) at 15, 20, 25 and 30°C were 11.55, 2.80, 2.47, and 3.01 days, respectively. Under field conditions, the net reproductive rate (R0) of L. erysimi was larger in the winter (53.50) than in the summer (40.99), the same being observed for the mean generation time (T), which was 13.85 days in the winter and 7.57 days in the summer. The intrinsic rate of natural increase (rm) and the finite rate of increase (λ) were 0.29/1.34 and 0.40/1.63 for winter and summer, respectively. The doubling time (DT) observed for winter (2.39 days) was larger than the one observed for summer (1.41 days). The temperature affects longevity of L. erysimi and the best parameters of life table of fertility under laboratory conditions are obtained at 25°C. The data obtained in field conditions reinforced this finding. The daily fecundity was higher and longevity was smaller in the summer than in the winter, thus increasing the innate capacity of increasing in number and duplicating the population in a shorter period of time.

A new protocol using 3-h fast animal for intestinal motility test was developed in our laboratory aiming the 3R's concept to reduce the stress of animals. Our results may aid in formulating recommendations that can be included in revised guidelines with regard to fasting time of mice.

The treatment of animals used in research or in education tests requires proper care, scientific judgment and qualified professionals to understand the needs of the animals and the special requirements of the research, tests, and educational programs. The established guidelines aim the development of knowledge necessary for the improvement of health and wellbeing of humans as well as animals to get precise and accurate results. The techniques should not be performed with inappropriate procedures that cause pain or suffering. Some principles should be considered to have an appropriate animal care and treatment.

A total of 20,065 weights recorded on 3016 Nelore animals were used to estimate covariance functions for growth from birth to 630 days of age, assuming a parametric correlation structure to model within-animal correlations. The model of analysis included fixed effects of contemporary groups and age of dam as quadratic covariable. Mean trends were taken into account by a cubic regression on orthogonal polynomials of animal age. Genetic effects of the animal and its dam and maternal permanent environmental effects were modelled by random regressions on Legendre polynomials of age at recording. Changes in direct permanent environmental effect variances were modelled by a polynomial variance function, together with a parametric correlation function to account for correlations between ages. Stationary and nonstationary models were used to model within-animal correlations between different ages. Residual variances were considered homogeneous or heterogeneous, with changes modelled by a step or polynomial function of age at recording. Based on Bayesian information criterion, a model with a cubic variance function combined with a nonstationary correlation function for permanent environmental effects, with 49 parameters to be estimated, fitted best. Modelling within-animal correlations through a parametric correlation structure can describe the variation pattern adequately. Moreover, the number of parameters to be estimated can be decreased substantially compared to a model fitting random regression on Legendre polynomial of age. © 2004 Elsevier B.V. Fluorescence diagnosis of malignant lesions has been showed as an attractive optical technique due especially to its real-time response and a more objective and quantitative evaluation. Even though the oral cavity allows a direct examination many lesions are diagnosed when it is already in advanced stage, compromising the patient prognosis. In this study, the fluorescence spectroscopy was used to the detection of chemically induced carcinoma at the lateral border of the tongue in a hamster model. Two excitations wavelengths in visible region were applied: 442 and 532 nm. All the spectra results were analyzed comparing with the histopathological diagnosis. The better results were achieved with the 442 nm laser excitation. The spectra from carcinoma showed new emission bands and these were used to determined different ratios for a quantitative analysis. Using the 625-645 nm fluorescence range under 442 nm excitation (A3 coefficient) the percentage of false negative was of 9.1%, however the false positive percentage was of 18.5%. The 532 nm excitation provided a better normal tissue detection compared to 442 nm excitation. The ideal clinical condition is probably the use of multiple wavelengths excitation for a broader tissue fluorescence investigation.

A variety of effects is attributed to the photo stimulation of tissues, such as improved healing of ulcers, analgesic and anti-inflammatory effects, stimulation of the proliferation of cells of different origins and stimulation of bone repair. Some investigations that make qualitative evaluations, like wound healing and evaluation of pain and edema, can be conducted in human subjects. However, deeper investigations on the mechanisms of action of the light stimulus and other quantitative works that requires biopsies or destructive analysis has to be carried out in animal models or in cell cultures. In this work, we propose the use of planarians as a model to study laser-tissue interaction. Contrasting with cell cultures and unicellular organisms, planarians are among the simplest organism having tissue layers, central nerve system, digestive and excretory system that might have been platforms for the evolution of the complex and highly organized tissues and organs found in higher organisms. For the present study, 685 nm laser radiation was employed. Planarians were cut transversally, in a plane posterior to the auricles. The body fragments were left to regenerate and the proliferation dynamics of stem cells was studied by using histological analysis. Maximum cell count was obtained for the laser treated group at the 4th experimental day. At that experimental time, we also had the largest difference between the irradiated and the non-irradiated control group. We concluded that the studied flatworm could be an interesting animal model for in vivo studies of laser-tissue interactions.

Toxoplasmosis is a worldwide zoonosis caused by Toxoplasma gondii and its definitive host is the domestic and wild felids infecting human beings and other warmblooded animals. Dogs are considered a potential risk on the transmission due they can mechanically transmit oocysts to man. In this study, a retrospective analysis of toxoplasmic infection in dog serum samples sent to Serviço de Diagnóstico de Zoonoses/FMVZ-UNESP/Botucatu, SP, in the period of 1998 to 2007 was performed. During this period 1097 serum samples were analyzed by the indirect fluorescent antibody test (IFAT), with 299 (27.25%) positive. The most frequent titer was 16 (42.80%), followed by 64 (37.79%). The results indicate that T.gondii is distributed in the environment showing the role of the dog as sentinel animal to toxoplasmosis to monitor public health actions to the control of this zoonosis.

Leaves from Carpolobia lutea (Polygalaceae) were screened to establish the antiulcer ethnomedicinal claim and to quantitatively isolate, elucidate the active compounds by semi-preparative HPLC. The anti-nociceptive effects of Carpolobia lutea (CL) G. Don (Polygalaceae) organic leaf extracts were tested in experimental models in mice. The anti-nociceptive mechanism was determined using tail-flick test, acetic acid-induced abdominal constrictions, formalin-induced hind paw licking and the hot plate test. The fractions (ethanol, ethyl acetate, chloroform, n-hexane) and crude ethyl acetate extract of CL (770 mg/kg, i.p.) produced significant inhibitions of both phases of the formalin-induced pain in mice, a reduction in acetic acid-induced writhing as well as and an elevation of the pain threshold in the hot plate test in mice. The inhibitions were greater to those produced by indomethacin (5 mg/kg, i.p.). Ethyl acetate fraction revealed cinnamic and coumaric acids derivatives, which are described for the first time in literature. These cinnamalglucosides polyphenols characterised from CL may in part account for the pharmacological activities. These findings confirm its ethnomedical use in anti-inflammatory pain and in pains from gastric ulcer-associated symptoms. © 2011 Springer Basel AG.

Aims: To discuss the importance of studying animal models to test hypotheses about the mechanisms of urinary continence and pathophysiology of diabetes and urinary incontinence. Source of Data: A literature review was conducted in PubMed and SciELO. The key words used were diabetes, urinary incontinence, urethra, human and rats. Summary of Findings: There is a strong relation between the genesis of urinary incontinence and diabetes mellitus. Due to the similarity of normal distribution of skeletal muscle and urethra anatomy between humans and rats, these animal models have been used in current research about these disorders. Conclusions: The use of rats as an animal model is suitable for experimental studies that test hypotheses about the mechanisms of continence and pathophysiology of the binomial diabetes mellitus and urinary incontinence, thus enabling solutions of great value in clinical practice.
