My thesis consists of three essays that investigate strategic interactions between individuals engaging in risky collective action in uncertain environments. The first essay analyzes a broad class of incomplete information coordination games with a wide range of applications in economics and politics. The second essay draws from the general model developed in the first essay to study decisions by individuals of whether to engage in protest/revolution/coup/strike. The final essay explicitly integrates state response to the analysis. The first essay, Coordination Games with Strategic Delegation of Pivotality, exhaustively analyzes a class of binary action, two-player coordination games in which players receive stochastic payoffs only if both players take a ``stochastic-coordination action''. Players receive conditionally-independent noisy private signals about the normally distributed stochastic payoffs. With this structure, each player can exploit the information contained in the other player's action only when he takes the “pivotalizing action”. This feature has two consequences: (1) When the fear of miscoordination is not too large, in order to utilize the other player's information, each player takes the “pivotalizing action” more often than he would based solely on his private information, and (2) best responses feature both strategic complementarities and strategic substitutes, implying that the game is not supermodular nor a typical global game. This class of games has applications in a wide range of economic and political phenomena, including war and peace, protest/revolution/coup/ strike, interest groups lobbying, international trade, and adoption of a new technology. My second essay, Collective Action with Uncertain Payoffs, studies the decision problem of citizens who must decide whether to submit to the status quo or mount a revolution. If they coordinate, they can overthrow the status quo. Otherwise, the status quo is preserved and participants in a failed revolution are punished. Citizens face two types of uncertainty. (a) non-strategic: they are uncertain about the relative payoffs of the status quo and revolution, (b) strategic: they are uncertain about each other's assessments of the relative payoff. I draw on the existing literature and historical evidence to argue that the uncertainty in the payoffs of status quo and revolution is intrinsic in politics. Several counter-intuitive findings emerge: (1) Better communication between citizens can lower the likelihood of revolution. In fact, when the punishment for failed protest is not too harsh and citizens' private knowledge is accurate, then further communication reduces incentives to revolt. (2) Increasing strategic uncertainty can increase the likelihood of revolution attempts, and even the likelihood of successful revolution. In particular, revolt may be more likely when citizens privately obtain information than when they receive information from a common media source. (3) Two dilemmas arise concerning the intensity and frequency of punishment (repression), and the frequency of protest. Punishment Dilemma 1: harsher punishments may increase the probability that punishment is materialized. That is, as the state increases the punishment for dissent, it might also have to punish more dissidents. It is only when the punishment is sufficiently harsh, that harsher punishment reduces the frequency of its application. Punishment Dilemma 1 leads to Punishment Dilemma 2: the frequencies of repression and protest can be positively or negatively correlated depending on the intensity of repression. My third essay, The Repression Puzzle, investigates the relationship between the intensity of grievances and the likelihood of repression. First, I make the observation that the occurrence of state repression is a puzzle. If repression is to succeed, dissidents should not rebel. If it is to fail, the state should concede in order to save the costs of unsuccessful repression. I then propose an explanation for the “repression puzzle” that hinges on information asymmetries between the state and dissidents about the costs of repression to the state, and hence the likelihood of its application by the state. I present a formal model that combines the insights of grievance-based and political process theories to investigate the consequences of this information asymmetry for the dissidents' contentious actions and for the relationship between the magnitude of grievances (formulated here as the extent of inequality) and the likelihood of repression. The main contribution of the paper is to show that this relationship is non-monotone. That is, as the magnitude of grievances increases, the likelihood of repression might decrease. I investigate the relationship between inequality and the likelihood of repression in all country-years from 1981 to 1999. To mitigate specification problem, I estimate the probability of repression using a generalized additive model with thin-plate splines (GAM-TPS). This technique allows for flexible relationship between inequality, the proxy for the costs of repression and revolutions (income per capita), and the likelihood of repression. The empirical evidence support my prediction that the relationship between the magnitude of grievances and the likelihood of repression is non-monotone.

Esta pesquisa, intitulada CRIATIVIDADE PARA QUÊ? CONVERSAS, PROCESSOS E PRODUÇÕES NA FORMAÇÃO DE PROFESSORES, investiga a criatividade na formação de professores. Sua fundamentação teórica baseia-se, principalmente, na atividade criadora de Fayga Ostrower e é contextualizada nas relações existentes entre imaginação e criatividade, propostas por Vigostky. A investigação intentou compreender as concepções de criatividade de um grupo de estudantes de licenciatura da Universidade Federal do Rio Grande - FURG. Os objetivos específicos consistiram em investigar as condições inibidoras e estimuladoras da criatividade no ensino; identificar as necessidades, desejos e possibilidades dos estudantes de licenciatura em relação ao ensino atual; e discutir as evidências e as características do professor criativo. A produção de dados foi realizada a partir da execução de um projeto de extensão, no qual os participantes realizaram atividades com temáticas relacionadas ao tema de pesquisa, tais como a conceituação de criatividade; inibidores e potencializadores da criatividade; a escola criativa e o professor criativo. Os dados foram produzidos por meio de questionários, diários, áudio e criação de objetos tridimensionais, confeccionados com tecido e outros materiais. O projeto de extensão foi realizado em duas ocasiões com públicos distintos. A primeira versão foi realizada em 2013, com nove alunos de diversos cursos de licenciatura. A segunda ocorreu em 2014, com acadêmicos do curso de Artes Visuais - Licenciatura. A análise de dados foi inspirada na Análise Textual Discursiva - ATD, de Moraes e Galiazzi, e os resultados foram apresentados a partir de histórias ficcionais, contadas por professores, personagens fictícios. Os personagens foram compostos por aspectos relacionados à criatividade, apresentados e discutidos pelos grupos durante o desenvolvimento do projeto, e as histórias identificam características que inibem ou potencializam a criatividade no processo de ensino-aprendizagem. Entre os aspectos apontados como inibidores do processo criativo estão o excesso de atividades ao qual estão submetidos os professores e alunos na universidade; o uso de vocabulário prolixo pelo professor; a repressão do sistema educacional, representada pela imposição de regras e pelo excesso de atividades que impedem os alunos de utilizarem a livre expressão. Como elementos que estimulam a criatividade, foram identificados o desejo pelo novo ou desconhecido; a contextualização dos conteúdos, relacionando-os ao cotidiano dos alunos; aulas realizadas em ambientes externos à sala de aula, entre outros. Os aspectos indicados pelos sujeitos da pesquisa como sendo criativos não são surpreendentes ou inovadores. Poucos professores universitários foram lembrados pelos alunos como sendo criativos, e muitas foram as críticas em relação à universidade e aos métodos de ensino utilizados pelos professores que, segundo os sujeitos, inibem o processo criativo. Embora a criatividade seja considerada vital ao homem, constata-se que, no ambiente universitário, há muitas barreiras, dificultando e inibindo o processo criativo dos futuros professores.

As Microcistinas são heptapeptídios cíclicos produzidos como metabólitos secundários por diferentes espécies de cianobactérias, sendo relevantes pelo seu potencial hepatotóxico. Peixes apresentam estratégias bioquímicas para detoxificar contaminantes ambientais, incluindo a ativação de enzimas de fase II de biotransformação, que incluem as isoformas de glutationa S-transferase (GST). As GST catalizam a conjugação de glutationa reduzida (GSH) com uma variedade de xenobióticos, incluindo as microcistinas. O presente estudo avaliou os níveis transcricionais de quinze isoformas de GST a fim de identificar isoformas possivelmente envolvidas na detoxificação de contaminantes ambientais como a microcistina-LR (MC-LR) em Danio rerio. A técnica de PCR em tempo real (RT-qPCR) foi utilizada para avaliação dos níveis transcricionais, permitindo análise das GST em diferentes órgãos, abundância e a ativação/repressão das isoformas de GST pela exposição à MC-LR. Foram avaliados os possíveis efeitos causados em brânquia e fígado após exposição por 24 hs às concentrações de 5 µg.L-1 e 50 µg.L-1 de MC-LR. Baseado nos scores de estabilidade para oito genes normalizadores, foram selecionados glicose-6-fosfato desidrogenase (g6pdh), β-actina1 e beta-2-microglobulina (b2m); b2m, alfa-tubulina 1 (tuba) e β- actin1; e tuba, b2m e g6pdh, para normalização dos níveis trancricionais de GST para distribuição órgão-específica, abundância e efeito da MC-LR em brânquia e fígado, respectivamente. A avaliação transcricional da distribuição órgão-específica revelou níveis significativos de gstal e gstk1.1 no fígado; gstp1 e gstp2 em brânquia; mgst3a, gstr1, gstm2, gstm33, gstp1, gstp2 e gstk1.1 no intestino; gstm2, gstm3 e gstal no olho e gstt1a e gsta2.1 no cérebro. Considerando os níveis de transcritos para um dado órgão, gstk1.1, gstal, gstp1 e gstt2 foram mais abundantes nos órgãos de detoxificação, tais como o fígado, brânquias e intestino, enquanto gstt1a e gsta2.1 foram mais abundantes no rim. Em brânquia, gsta2.1 e gstt1b foram reprimidas por 5 µg.L-1 de MC-LR e mgst1.1 foi reprimida em 50 µg.L-1 de MC-LR. No fígado, as isoformas gst2.2 e gstp2 foram reprimidas em ambas as concentrações, gstal foi reprimida em 5 µg.L-1, e gstt1a e gstk1.1 foram reprimidas em 50 µg.L-1 de MC-LR. As isoformas gstal, gstr1, gstp1, mgst3a, gstm1, gstm2 e gstm3 não foram alteradas pela exposição a MC-LR. Os resultados obtidos fornecem informações para a escolha de isoformas específicas de GST possivelmente envolvidas na detoxificação/toxicidade de MC-LR, a serem melhores caracterizadas ao nível protéico e também contribui para a escolha de genes normalizadores a serem utilizados em outros estudos da mesma natureza

A morte suscita-nos grande inquietação (e.g., Oliveira, 2008). Especialmente entre as crianças, perdidas na incompreensão, silêncio e repressão psicológica por parte dos adultos. Nesta investigação averiguamos as representações sociais, da morte e da vida, entre crianças dos 8 aos 11 anos, escolarizadas de ambos os sexos. As meninas representam a morte pelo medo que suscita, e pela sua dimensão ritualista, enquanto que os meninos a percecionam sobretudo por sentimento de mal-estar e impotência. Sobre a vida, os meninos evidenciam uma visão mais hedonista do que elas, que salientam a interação com o outro e as questões afetivo-emocionais. As representações das raparigas aproximam-se mais das dos sujeitos de 10-11 anos, enquanto que as dos rapazes se aproximam das dos participantes de 8-9 anos. Esperamos contribuir para o modo como as crianças representam a morte e, para uma educação para a morte e para a vida. ABSTRACT; Death rouses great uneasiness (e.g., Oliveira, 2008). Especially among children who are lost amid the incomprehension, silence and psychological repression of adults. This research examines social representations of life and death among school attending children of both sexes, between the ages of 8 and 11 years old. Girls represent death for both the fear it rouses and its ritualistic dimension, while boys mostly perceive it as a feeling of uneasiness and impotence. Regarding life, boys show a more hedonistic vision than girls, who give relevance to interacting with. others, and to affective and emotional issues. The representations of girls are closer to those of the 10-11-year-old subjects, while the representations of boys are closer to those of the 8-9-year-old participants. We hope to contribute to the way children represent death, and to an education on death and life.

O contrabando é uma actividade comercial ilícita que não tem uma origem definida, uma vez que, ultrapassa todas as fronteiras espaciais e temporais. No entanto, a partir do século XIX e ao longo do período da Primeira Guerra Mundial, o tema constitui uma das principais preocupações dos países Aliados. Nas Conferências de Haia (1899 e 1907) e Londres (1908-09), o conceito é discutido e são promulgadas listas com as mercadorias consideradas contrabando de guerra. Durante a Primeira Guerra Mundial é criado o bloqueio económico com o intuito de reprimir o contrabando e a circulação de mercadorias com destino ao inimigo. Esta dissertação de mestrado tem como principal objectivo compreender o significado, a importância e o impacto do contrabando num contexto local e inserido numa conjuntura de guerra e pós-guerra. Destacando os actores, as dinâmicas de resistência e repressão, assim como as relações entre o poder central, poder local e contrabandistas.

[ES]El siglo XX fue caracterizado por un desarrollo de las dictaduras, que, a través de un marco legal, realizaron políticas de exclusión y de represión contra aquellos que se mostraban opuestos al régimen. A partir de una definición de los “enemigos” como “peligrosos”, y a través de una lucha por la defensa de la comunidad, los gobiernos totalitarios crearon una legislación en base a estos términos, que propició el control social y en última instancia, el holocausto. El presente Trabajo tratará de abordar los orígenes de este Derecho, tanto en la dictadura nacionalsocialista, como en la franquista, la teoría y dogmática que las fundó y las políticas que se llevaron a cabo. Se realizará un análisis del mismo y una perspectiva actual a raíz de estos hechos.

The neural crest is a group of migratory, multipotent stem cells that play a crucial role in many aspects of embryonic development. This uniquely vertebrate cell population forms within the dorsal neural tube but then emigrates out and migrates long distances to different regions of the body. These cells contribute to formation of many structures such as the peripheral nervous system, craniofacial skeleton, and pigmentation of the skin. Why some neural tube cells undergo a change from neural to neural crest cell fate is unknown as is the timing of both onset and cessation of their emigration from the neural tube. In recent years, growing evidence supports an important role for epigenetic regulation as a new mechanism for controlling aspects of neural crest development. In this thesis, I dissect the roles of the de novo DNA methyltransferases (DNMTs) 3A and 3B in neural crest specification, migration and differentiation. First, I show that DNMT3A limits the spatial boundary between neural crest versus neural tube progenitors within the neuroepithelium. DNMT3A promotes neural crest specification by directly mediating repression of neural genes, like Sox2 and Sox3. Its knockdown causes ectopic Sox2 and Sox3 expression at the expense of neural crest territory. Thus, DNMT3A functions as a molecular switch, repressing neural to favor neural crest cell fate. Second, I find that DNMT3B restricts the temporal window during which the neural crest cells emigrate from the dorsal neural tube. Knockdown of DNMT3B causes an excess of neural crest emigration, by extending the time that the neural tube is competent to generate emigrating neural crest cells. In older embryos, this resulted in premature neuronal differentiation. Thus, DNMT3B regulates the duration of neural crest production by the neural tube and the timing of their differentiation. My results in avian embryos suggest that de novo DNA methylation, exerted by both DNMT3A and DNMT3B, plays a dual role in neural crest development, with each individual paralogue apparently functioning during a distinct temporal window. The results suggest that de novo DNA methylation is a critical epigenetic mark used for cell fate restriction of progenitor cells during neural crest cell fate specification. Our discovery provides important insights into the mechanisms that determine whether a cell becomes part of the central nervous system or peripheral cell lineages.

O Policiamento de Proximidade é o modelo de policiamento atual em Portugal, privilegiando uma postura preventiva e dando relevo à pró-atividade das forças policiais. À Guarda Nacional Republicana, enquanto força policial, cabe-lhe zelar pelos direitos, liberdades e garantias dos cidadãos, constitucionalmente protegidos, através dessa atuação preventiva, procurando intensificar pró-ativamente a sua presença no seio da sociedade. Contudo, por vezes é necessário recorrer a uma intervenção de caráter repressivo, a fim de repor a ordem e visando uma prevenção futura, evitando os comportamentos antissociais. Torna-se assim pertinente estudar de que forma se articulam a atuação preventiva e repressiva da GNR, de modo a perceber se é possível existir um equilíbrio entre elas, ou se, por alguma razão, existem limitações no seu emprego. O Destacamento Territorial de Coimbra constitui o estudo de caso. Esta investigação para além de descrever a articulação entre ambas as formas de atuação, pretende ainda identificar aquilo que as caracteriza bem como as suas potencialidades e vulnerabilidades. De forma a recolher a informação pretendida, foram privilegiadas a análise documental, entrevistas ao Comandante de Destacamento Territorial de Coimbra e aos respetivos Comandantes dos Postos Territoriais, bem como questionários aos militares do destacamento responsáveis por exercerem o patrulhamento. Seguidamente procedeu-se à análise de conteúdo dos dados recolhidos. Das conclusões retiradas, salienta-se que a falta de recursos principalmente humanos, corresponde a um problema do destacamento, que dificulta uma ação de patrulhamento contínuo junto do cidadão e que contribui para o Policiamento de Proximidade. Denota-se também a intensa fiscalização rodoviária que, apesar de ter uma natureza preventiva, é interpretada pela população com uma imagem de repressão.

The apparent simplicity of viruses hides the complexity of their interactions with their hosts. Viruses are masters at circumventing host defenses and manipulating the cellular environment for their own benefit. The replication of the largest known family of single-stranded DNA viruses, Geminiviridae, is impaired by DNA methylation and Arabidopsis mutants affected in cytosine methylation are hypersusceptible to geminivirus infection. This implies that plants might use methylation as a defense against geminiviruses and that the viral genome is a target for plant DNA methyltransferases. We have found a novel counter-defense strategy used by geminiviruses, that reduces the expression of the plant maintenance DNA methyltransferases, MET1 and CMT3, in both, locally and systemically infected tissues. Furthermore, we demonstrated that the virus-mediated repression of these two maintenance DNA methyltransferases is widely spread among different geminivirus species. Additionally, we identified Rep as the geminiviral protein responsible for the repression of MET1 and CMT3, and another viral protein, C4, as an ancillary player in MET1 downregulation. The presence of Rep, suppresses TGS of an Arabidopsis transgene and of host loci whose expression is strongly controlled by CG methylation. Bisulfite sequencing analyses showed that the expression of Rep caused a substantial reduction in the levels of DNA methylation at CG sites. Our findings suggest that Rep, the only viral protein essential for geminiviral replication, displays TGS suppressor activity through a mechanism distinct from the one thus far described for geminiviruses.

Streptococcus suis is an emerging zoonotic pathogen. With the lack of an effective vaccine, antibiotics remain the main tool to fight infections caused by this pathogen. We have previously observed a reserpine-sensitive fluoroquinolone (FQ) efflux phenotype in this species. Here, SatAB and SmrA, two pumps belonging to the ATP binding cassette (ABC) and the major facilitator superfamily (MFS), respectively, have been analyzed in the fluoroquinolone-resistant clinical isolate BB1013. Genes encoding these pumps were overexpressed either constitutively or in the presence of ciprofloxacin in this strain. These genes could not be cloned in plasmids in Escherichia coli despite strong expression repression. Finally, site-directed insertion of smrA and satAB in the amy locus of the Bacillus subtilis chromosome using ligated PCR amplicons allowed for the functional expression and study of both pumps. Results showed that SatAB is a narrow-spectrum fluoroquinolone exporter (norfloxacin and ciprofloxacin), susceptible to reserpine, whereas SmrA was not involved in fluoroquinolone resistance. Chromosomal integration in Bacillus is a novel method for studying efflux pumps from Gram-positive bacteria, which enabled us to demonstrate the possible role of SatAB, and not SmrA, in fluoroquinolone efflux in S. suis.

Culture, history, and biology are inseparable. Cultural manifestations are necessarily immersed in a context, originate in the embodied minds that create them, and are directed to the embodied minds that receive them and recreate them within their contexts (individual and collective). The novel and the film of historical memory in Spain aim to connect their audiences with a problem that has not been solved, as the Civil War, the postwar, and the pact of forgetfulness left a wide sector of the Spanish society voiceless. During the last few years, a series of initiatives coming from the arts, as well as other realms such as the legal, have sought to reexamine the unhealed wound that still haunts Spanish subjects. La voz dormida [The Sleeping Voice] is one of those initiatives. It begins as testimony, develops into a hybrid and intertextual novel, and later becomes a film. It constitutes an inclusive project, one of offering an alternative version to the “official history”, while incorporating the marginal voices of women that had been left out of the memory of the war and the dictatorship. Objective and Results By examining both the literary and the cinematic versions of Chacón’s work I aimed to evidence the connections that exist between the artistic portrayal of the postwar repression (particularly how it affects women) and the current movement of recovery of historical memory in Spain. Specifically, I was interested in showing how both the novel and the film employ a series of narrative strategies that emphasize the body and intentionality, with the purpose of creating in readers and spectators an empathetic response that may lead to prosocial behavior. In order to carry out this interdisciplinary study, which relates fiction, mind, and socio-historical context, I draw on cognitive theories of literature and film, as well as theories from social and developmental psychology, such as the Richard Gerrig’s theory of narrative experience, Keith Oatley’s psychology of fiction, Suzanne Keen’s theory of narrative empathy, and the empathy-altruism hypothesis, derived form the ideas of Jean Decety, among others...

Power flow calculations are one of the most important tools for power system planning and operation. The need to account for uncertainties when performing power flow studies led, among others methods, to the development of the fuzzy power flow (FPF). This kind of models is especially interesting when a scarcity of information exists, which is a common situation in liberalized power systems (where generation and commercialization of electricity are market activities). In this framework, the symmetric/constrained fuzzy power flow (SFPF/CFPF) was proposed in order to avoid some of the problems of the original FPF model. The SFPF/CFPF models are suitable to quantify the adequacy of transmission network to satisfy “reasonable demands for the transmission of electricity” as defined, for instance, in the European Directive 2009/72/EC. In this work it is illustrated how the SFPF/CFPF may be used to evaluate the impact on the adequacy of a transmission system originated by specific investments on new network elements

This paper extends the symmetric/constrained fuzzy powerflow models by including the potential correlations between nodal injections. Therefore, the extension of the model allows the specification of fuzzy generation and load values and of potential correlations between nodal injections. The enhanced version of the symmetric/constrained fuzzy powerflow model is applied to the 30-bus IEEE test system. The results prove the importance of the inclusion of data correlations in the analysis of transmission system adequacy.
