Introduction: The HELLP syndrome is a severe complication of pregnant women with preeclampsia (PE), characterized by association of hemolysis, changes in liver enzymes and thrombocytopenia. Hemolysis, defined by the presence of microangiopathic hemolytic anemia, is one of the characteristics in this syndrome. However, as hemolysis occurs in a short time there is some difficulty in its laboratory diagnosis. Therefore, the search for a more sensitive and specific method for hemolysis determination may help in the early diagnosis of the HELLP syndrome. Objectives: a) To determine the plasma concentration of haptoglobin in normotensive pregnant women and in pregnant women with PE, classified into mild PE, severe PE and HELLP/partial HELLP syndrome; b) To compare the efficacy of haptoglobin plasma concentration and serum total bilirubin as criteria for hemolysis diagnosis in HELLP/partial HELLP syndrome. Methods: We conducted a cross-sectional analytical and comparative study involving 66 pregnant women diagnosed with PE, being 25 cases with mild PE, 28 with severe PE, and 13 with HELLP/partial HELLP syndrome. Twenty-one normotensive pregnant women were included for comparison of haptoglobin plasma concentration between the groups and to determine the normal values for pregnant women. The variables studied were: maternal age, gestational age, systolic and diastolic blood pressure, proteinuria, hematocrit and hemoglobin values, platelet count, serum total bilirubin, lactate dehydrogenase (LDH), glutamic oxaloacetic transaminase (AST) and glutamic-pyruvic transaminase (ALT), urea, creatinine and uric acid, and also plasma concentrations of haptoglobin. The results were analyzed by nonparametric tests, with a significance level of 5%. A atorvastatina, amplamente utilizada no tratamento de dislipidemias, é fármaco inibidor da enzima 3-hidroxi-3-metilglutaril coenzima A redutase (HMG-CoA) redutase que atua na etapa limitante da síntese do colesterol nas células hepáticas. O transporte hepatocelular da atorvastatina é mediado principalmente pelo polipeptídeo transportador de ânions orgânicos (OATP) 1B1, o qual é altamente polimórfico geneticamente e está sujeito a interação com outros fármacos usados na clínica. O presente estudo teve como objetivo revisar a literatura científica baseada em estudos clínicos, in vitro e experimentais a fim de avaliar o impacto de polimorfismos genéticos de OATP1B1 na disposição cinética, eficácia e toxicidade da atorvastatina. Os estudos clínicos demonstram que o polimorfismo c.521T>C, presente nos alelos SLCO1B1 *5 e *15, resulta em alteração funcional da atividade transportadora, sendo que indivíduos homozigotos para o alelo raro C, apresentam maiores concentrações plasmáticas de atorvastatina e seus metabólitos ativos 2-hidroxiatorvastatina e 4- hidroxiatorvastatina. Entretanto, o aumento na concentração plasmática da atorvastatina e seus metabólitos ativos em indivíduos c.521 CC parece não ser acompanhado de redução na concentração dos mesmos no sítio alvo, tendo em vista que não foram encontradas diferenças significativas na redução da LDL-C ou colesterol total quando comparados aos indivíduos homozigotos para o alelo selvagem. O alelo SLCO1B1*15 mostrou-se significativamente associado a miopatia induzida pela atorvastatina, apesar do pequeno número de pacientes investigados. Estudos in vitro em células expressando OATP1B1 são necessários a fim de investigar o impacto do polimorfismo c.521T>C na concentração de atorvastatina nos hepatócitos. The routine semen evaluation assessing sperm concentration, motility and morphology, does not identify subtle defects in sperm chromatin architecture. Bulls appear to have stable chromatin, with low levels of DNA fragmentation. However, the nature of fragmentation and its impact on fertility remain unclear and there are no detailed reports characterizing the DNA organization and damage in this species. The intensive genetic selection, the use of artificial insemination and in vitro embryo production associated to the cryopreservation process can contribute to the chromatin damage and highlights the importance of sperm DNA integrity for the success of these technologies. Frozen-thawed semen samples from three ejaculates from a Nellore bull showed high levels of morphological sperm abnormalities (55.8±5.1%), and were selected for complementary tests. Damage of acrosomal (76.9±8.9%) and plasma membranes (75.7±9.3%) as well as sperm DNA strand breaks (13.8±9.5%) and protamination deficiency (3.7±0.6%) were significantly higher compared to the values measured in the semen of five Nellore bulls with normospermia (24.3±3.3%; 24.5±6.1%; 0.6±0.5%; 0.4±0.6% for acrosome, plasma membrane, DNA breaks and protamine deficiency, respectively) (P<0.05). Motility and percentage of spermatozoa with low mitochondrial potential showed no differences between groups. This study shows how routine semen analyses (in this case morphology) may point to the length and complexity of sperm cell damage emphasizing the importance of sperm function testing.

The aim of this study was to compare the following four genetic groups of hair sheep: Santa Inês (SI), Morada Nova (MN), Brazilian Somali (BS), and the F1 1/2Dorper x 1/2Morada Nova crossbreed on traits related to growth and parasitic infection. Thirty-three male lambs of the same age and of simple birth, under the same pre-weaning management conditions were used in the experiment. After weaning the animals were housed in a completely randomized design in paddocks made of Panicum maximum cv. Tanzania. Along the course of the research, the performance of the four groups of sheep was observed to be negatively affected by gastrointestinal parasites, but there was a genotype effect to the average daily weight gain (ADWG), where the SI and F1 genotypes presented higher values. The effects of genotype, time and genotype x time interaction were significant in weight and corporal score (CS) measurements. The BS lambs had the highest CS values throughout the experiment despite not presenting greater weight gain when compared to the SI and F1 breeds. There were also significant effects of time and genotype x time interaction for packed cell volume (PCV) and FAMACHA© score (FAM) and only the time effect was significant in the total number of eggs per gram (EPG) and total plasma protein (TPP). The MN lambs showed higher PCV values and unlike the other groups, presented a FAMACHA© score below 3 and PCV above 23% even having a higher EPG tendency, especially in the initial phase, indicating a possible higher resilience to infection caused by gastrointestinal parasites.

The goal of this paper was to determine the effects of seven diets with differents fatty acids sources in hematology of Nile tilapia before and after cold stimulus. Seven diets, with 3% of fatty acid source, were formulated: soybean oil (OS); fish oil (OP); beef tallow (SB) and mixtures of OS+OP; OS+SB; OP+SB e OS+OP+SB. Fish (112 male of Nile tilapia of 30,0 +/- 2,65g) were totally randomly distributed, with four replicates, into 28 plastic aquaria (40 L; 4 fish per aquarium). Fish were fed four times per day for 35 days. At 35th day and also after seven days cold stimulus blood was collected and the profile hematological was evaluated. Leucocytes numbers were reduced in fish fed a SB and fish oil combination diet and were highest in fish fed SB diet before cold stimulus. Plasma protein was reduced in fish fed soybean oil diets and highest in fishes fed a combination of soybean oil, fish oil and SB diets. Anything fatty acids source promotes effective changes in fish health and cold tolerance promotion. Cold don't drastically reduced fish general health status.

Different parasites that commonly occur concomitantly can influence one another, sometimes with unpredictable effects. We evaluated pathological aspects of dogs naturally co-infected with Leishmania infantum and Ehrlichia canis. The health status of the dogs was investigated based on histopathological, hematological and biochemical analyses of 21 animals infected solely with L. infantum and 22 dogs co-infected with L. infantum and E. canis. The skin of both groups showed chronic, predominantly lymphohistioplasmacytic inflammatory reaction. The plasmacytosis in the lymphoid tissues was likely related with the hypergammaglobulinemia detected in all the dogs. The disorganization of extracellular matrix found in the reticular dermis of the inguinal region and ear, characterized by the substitution of thick collagen fibers for thin fibers, was attributed to the degree of inflammatory reaction, irrespective of the presence of parasites. In addition, the histopathological analysis revealed that twice as many dogs in the co-infected group presented Leishmania amastigotes in the ear skin than those infected solely with Leishmania, increasing the possibility of becoming infected through sand fly vectors. Our findings highlight the fact that the health of dogs infected concomitantly with L. infantum and E. canis is severely compromised due to their high levels of total plasma protein, globulins, alkaline phosphatase and creatine kinase, and severe anemia.

The long-term effects of five different treatments of diabetes were evaluated in alloxan-induced diabetic rats. Seven experimental groups, with 50 rats each (GN--normal control; GD--untreated diabetic control; GI, GA, GIA--treated groups with insulin, acarbose, and insulin plus acarbose, respectively; GTIL, GTPD--treated groups with islet of Langerhans and pancreas transplantation) were studied. Clinical (body weight, water intake, food intake and urine output) and laboratory (blood and urinary glucose, and plasma insulin) parameters were analyzed at the beginning of the study, and after 1, 3, 6, 9 and 12 months of follow-up. Mortality was observed in all groups, except GN, during 12 months (GD = 50%; GI = 20%; GA = 26%; GIA = 18%; GTIL = 4%; GTPD = 20%). Rats from the GD, GI, and GIA groups died due to metabolic or hydrossaline disbalance, and/or pneumonia, diarrhoea, and cachexy. All deaths observed in GTIL and GTPD groups were in decorrence of technical failure at the immediate postoperative, until 72h. Animals from the GI, GA and GIA had significative improving of the clinical and laboratory parameters (p < 0,05) observed in diabetic rats, being the efficacy of theses treatments equal. However, rats from the GTIL and GTPD groups had better control of these parameters than GI, GA, and GIA groups. Transplanted rats had complete restoration, at the normal levels, of all analyzed variables (p < 0.01). Conventional treatments with insulin, acarbose, and insulin plus acarbose improved the severe diabetic state of the alloxan-diabetic rats, but pancreas and islet transplantation have a better performance for treatment of diabetes.

A manutenção do balanço hídrico ou de água é particularmente desafiadora para os anfíbios em virtude de seu tegumento altamente permeável e o risco associado de perda evaporativa de água. Neste sentido, anuros terrestres exibem uma grande diversidade de mecanismos que atuam na diminuição da perda evaporativa de água e contribuem para a manutenção de um balanço hídrico adequado. Ademais, algumas espécies de anuros são conhecidas por tolerar altos níveis de desidratação e por engajar-se em atividades de rotina na natureza, ainda que sob níveis variáveis de desidratação. Finalmente, o próprio nível de desidratação pode acarretar alterações nos fatores envolvidos na manutenção do balanço de água dos anuros, aspecto muito pouco explorado em espécies terrestres neotropicais. Dessa forma, no presente estudo, mensuramos as taxas de perda evaporativa de água (PEA), reidratação a partir de água livre (Re) e calculamos a resistência da pele a desidratação (RP) em indivíduos adultos de Rhinella schneideri submetidos experimentalmente a diferentes níveis de desidratação (99%, 90%, 80% e 70% da massa corpórea inicial). Ademais, medimos as alterações causadas pela desidratação na osmolalidade plasmática (Osm), hematócrito (Htc), concentração de hemoglobina no sangue ([Hb]) e massa percentual dos órgãos viscerais. O objetivo central deste estudo foi avaliar os efeitos potenciais de diferentes níveis de desidratação sobre indicadores e repostas fisiológicas relevantes para a manutenção do balanço hídrico em uma espécie de anfíbio anuro terrestre. Nossos resultados mostram que quanto mais desidratados os animais, maiores a RP, a Re, a Osm, o Htc e a [Hb], e menores a PEA, e a massa percentual do fígado e do rim.
