Foeniculum vulgare Mill. (fennel) and Matricaria recutita L. (chamomile) are two examples of plants with reported antioxidant and antimicrobial properties, which can be related with their composition in phenolic compounds [1,2]. Furthermore, according to previous results of our research group, the direct incorporation of the aqueous extracts showed capacity to maintain the nutritional properties of the cottage cheeses, up to 7 days of storage, while improving the antioxidant potential. However, after 14 days, a decrease in the antioxidant properties was observed [1,2], which can be related with factors such as light, moisture, temperature and pH, that can cause bioactive compounds degradation. Therefore, the aim of the present study was to prepare microcapsules with the aqueous extracts of fennel and chamomile for incorporation in cottage cheese samples, in order to protect the bioactive molecules present in the extracts, such as phenolic compounds, and prevent the decrease of the antioxidant activity observed after the 14 days period. The microspheres were prepared using an atomization/coagulation technique. Sodium alginate was used as the matrix material to produce the microspheres that were characterized through optical microscopy (OM), during and after atomization, for inspecting morphology. The encapsulation efficiency (EE) was determined by HPLC-DAD by an indirect method by analysing the coagulation solution. FTIR was also used to attest the presence of the extract inside of the alginate matrix. These microencapsulated extracts were incorporated in cottage cheese samples that were further characterized in terms of nutritional properties and antioxidant potential right after incorporation, and after 7 and 14 days of storage at 4•c. The EE was estimated as -100% and the FTIR analysis confirmed the presence of the extracts inside the microspheres. The results showed that the incorporation of the microencapsulated extracts did not cause changes in the nutritional value of cottage cheeses (through a comparison with control samples without extracts). The predominant fatty acids were palmitic (C16:0) and oleic (CI8:0) acids. The order of abundance of fatty acids was as follows: saturated fatty acids (SF A)> monounsaturatcd fatty acids (MUF A)> polyunsaturated fatty acids (PUF A). Regarding free sugars, lactose was the only sugar identified and quantified in all samples. Regarding the antioxidant activity, the samples functionalized with the microencapsulated extracts showed a higher preservation of this property even after the 7th day of storage. Overall, the incorporation of the protected plant extracts in dairy foods can be a strategy to provide health benefits to consumers.

Atualmente existe um grande interesse da indústria alimentar pela utilização de extratos e produtos naturais em substituição de aditivos sintéticos, por conterem ou serem substâncias biologicamente ativas, nomeadamente do ponto de vista antioxidante [1]. Alguns estudos anteriores da nossa equipa de investigação revelaram que a incorporação de extratos de Foeniculum vulgare Mill. (funcho) e Matricaria recutita L. (camomila) em amostras de requeijão, conferia-lhes propriedades antioxidantes, para além de manterem o seu valor nutricional [2;3]. No entanto, verificou-se que este efeito era limitado a 7 dias. Assim, para prolongar a referida bioatividade, os extratos aquosos das plantas mencionadas foram microencapsulados em matriz de alginato e, posteriormente, incorporados em requeijões. Os produtos finais foram analisados cromatograficamente em termos de ácidos gordos (GC-FID) e açúcares livres (HPLC-RI). As amostras de requeijão contendo os extratos microencapsulados e livres foram comparadas com os requeijões controlo (sem qualquer extrato). Os resultados obtidos revelaram que a incorporação dos extratos não provocou alterações nas características nutricionais dos requeijões. Em todas as amostras, os ácidos gordos predominantes foram os ácidos palmítico (C16:0) e oleico (C18:1). A ordem de abundância em ácidos gordos foi a seguinte: ácidos gordos saturados > ácidos gordos monoinsaturados >ácidos gordos polinsaturados. A lactose foi o único açúcar livre identificado e quantificado em todas as amostras. Adicionalmente, as amostras funcionalizados com extratos microencapsulados apresentaram maior preservação da atividade antioxidante após o sétimo dia.

In a double blind study of ketoprofen and placebo in the treatment of osteoarthritis of the hip, ketoprofen was shown to be significantly more effective. Analysis of results was made using the sequential technique. Major intolerance occurred in two cases and minor intolerance in five cases. Newly diagnosed cases were treated more readily with ketoprofen than chronic cases treated for several months with other drugs which had proved ineffective. There were no changes in biological parameters. Age and sex did not affect the result. The further study of ketoprofen in large open trials appears to be indicated.

A adequação do intervalo entre pastejos (IEP) pode aumentar a produção de leite individual ou por área dos sistemas tropicais de produção de leite. O objetivo do ensaio experimental foi avaliar os efeitos de dois IEP sobre a produção individual e composição do leite, taxa de lotação e produção por área de vacas em lactação mantidas em pastagens de capim Elefante. Os tratamentos testados foram: IEP variável, determinado pela entrada dos animais na área a ser pastejada, quando o dossel atingisse 95% de interceptação de luz e o IEP fixo em 27 dias. O delineamento experimental foi o cross-over com quatro repetições. Não foram observados efeitos dos tratamentos (P>0,05) sobre a produção de leite, produção de leite corrigida para 3,5% de gordura (kg de leite/vaca/dia) e para os parâmetros relacionados à composição nutricional do leite (gordura, proteína, lactose e sólidos totais) tanto em porcentagem quanto em produção diária. Houve tendência para maior produção de leite (P=0,12), maior produção de leite corrigida para 3,5% de gordura (P=0,10) e produção (kg/dia) de gordura (P=0,08), lactose (P=0,11) e sólidos totais (P=0,12). A taxa de lotação (vacas/ha e UA/ha) e a produção de leite por área foram maiores (P<0,05) para o intervalo entre pastejos variáveis. Pode-se concluir que o intervalo entre pastejos variáveis possibilitou maior produção de leite na área, entretanto, mais estudos devem ser conduzidos para a avaliação dessa técnica no manejo do pastejo do capim Elefante.

Background: D-Lactate is normally present in the blood of humans at nanomolar concentrations due to methylglyoxal metabolism; millimolar D-lactate concentrations can arise due to excess gastrointestinal microbial production. Objectives: To examine the levels of plasma D-lactate in the necrotizing enterocolitis in premature infants. Patients and Methods: 128 premature infants were divided into control (group I, n = 69), feeding intolerance (group II, n = 42) and NEC (group III, n = 27) groups. Plasma D-lactate levels were measured at the onset of feeding intolerance or NEC and at weeks 2-3 in control infants (group I) by ELISA. Data were analyzed using descriptive statistics, non-parametric tests and Student’s t-test. Results: In groups I, II, III, median birth weights were 1845.7 ± 267.5 g, 1913.1 ± 306.5 g, and 1898.4 ± 285.3 g, median gestational ages were 34.3 ± 1.7 weeks, 33.9 ± 2.2 weeks and 35.1 ± 2.6 weeks, ages of sampling were 12.3 ± 2.9 days, 14.6 ± 3.7 days and 15.1 ± 1.8 days, respectively. The differences of median birth weights, median gestational ages and ages of sampling were not statistically significant (P > 0.05). The plasma D-lactate levels in groups I, II, III were 3.6 ± 1.9 μg/mL, 12.7 ± 8.3 μg/mL, and 35.4 ± 29.1 μg/mL, respectively, group III had higher plasma D-lactate level than groups I, II, and the difference among these groups was significant (x2 = 21.6, P < 0.01). Conclusions: Plasma D-lactate significantly increased early in NEC. Plasma D-lactate levels were associated with extensive disease in NEC infants. Therefore, it could be used as a diagnosis indicator in the early stage of NEC.

Past research has shown that having a large population of ethnic minorities beyond the neighborhood level arouses intolerance in the majority. However, this paper presents the argument that the effect of minority size on tolerance depends on minority type: the less subject the minority is to negative stereotyping, the more favorable the effect that minority size has on tolerance. In this study, a hierarchical linear model was applied to a dataset on advanced and emerging democracies in Europe. The analysis shows that when the duration and level of democracy are controlled for, ethnic tolerance was associated positively with native minority size and negatively with foreign population size.

McArdle disease is an autosomal recessive disorder caused by inherited deficiency of the muscle isoform of glycogen phosphorylase (or ‘myophosphorylase´), which catalyzes the first step of glycogen catabolism, releasing glucose-1-phosphate from glycogen deposits. As a result, muscle metabolism is impaired, leading to different degrees of exercise intolerance. Patients range from asymptomatic to severely affected, including in some cases limitations in activities of daily living. The PYGM gene codifies myophosphoylase and to date 147 pathogenic mutations and 39 polymorphisms have been reported. Exon 1 and 17 are mutational hot-spots in PYGM and 50% of the described mutations are missense.

A caprinocultura leiteira no Brasil vem se consolidando nos últimos anos, seja pelo aumento na produção no Nordeste alavancada por políticas públicas, seja pelo aumento na oferta de derivados lácteos, com alto valor agregado, observado com maior intensidade nas regiões Sul e Sudeste do País. Esse crescimento, ao exemplo do que ocorreu em outros países tropicais e subtropicais, tem sido sustentado pelo uso de material genético importado, notadamente de países da Europa, de raças como a Saanen, as Alpinas e a Toggenburg, e ainda a Anglo Nubiana. No entanto, a base de dados sobre o potencial de produção e a composição em constituintes lácteos dessas raças puras e de suas cruzas, ainda não está bem caracterizada nos países de clima tropical e subtropical, devido a tais informações não estarem disponíveis em número suficiente, ou de serem avaliadas superficialmente ou ainda não estarem devidamente sistematizadas e disponibilizadas. Baseado nisso, reuniram-se nesta Série Embrapa Documentos informações publicadas na literatura sobre o desempenho produtivo (produção de leite e de seus constituintes) das raças europeias mais difundidas mundialmente, em estado puro ou utilizadas em cruzamentos em ambientes tropical e subtropical. Espera-se que este documento subsidie a tomada de decisão quanto ao uso dos recursos genéticos disponíveis no País, bem como, seja base para disciplinas acadêmicas para técnicos e produtores de caprinos leiteiros.

Hoy se empiezan a sentir con mucho más fuerza las consecuencias de la globalización, entre ellas encontramos la migración. Es esencial debido a que está ocurriendo en los cinco continentes, cada vez con mayor frecuencia, e irónicamente conlleva un problema que “supuestamente“en el siglo XXI no debería estar ocurriendo: la violación a los derechos humanos de los migrantes. Se habla de que las personas migrantes sufren mucho con la finalidad de poder obtener una mejor calidad de vida para sus familias, ya que en sus países de origen no tienen esta oportunidad. Esta no es la excepción de los migrantes centroamericanos quienes tienen que cruzar México para poder llegar al país de los “sueños¨: Estados Unidos. El 100% de los transmigrantes centroamericanos sufre graves vulneraciones a sus derechos lo cual es sumamente preocupante, porque detrás de estas violaciones se encuentran plasmados la corrupción, el crimen organizado, la impunidad y, sobre todo, la falta de tolerancia. Estos fenómenos están gobernando actualmente al mundo.Nowadays, the repercussions of globalization greatly affect many sectors of society and among these, migration is one of them. It is of great importance and value to wake interest in this topic due to the fact that it is happening with more and more frequency in all five continents. Furthermore, it is truly unfortunate that the violation of the human rights of these migrants is still a reality we must face in this 21st century. It is said that those who migrate from a country to another, do it so to guarantee a superior quality of life for themselves and their respective families. Nonetheless, many individuals must suffer significantly before reaching this. Central-American migrants, who must cross Mexican territory in order to attain the “American Dream”, are not the exception in this case. Moreover, up to 100% of Central American migrants, who cross Mexican territory, face a cruel reality marked by transgressions to their human rights. Finally, this issue is of great concern for the region due to the fact that behind this curtain of transgressions, many other tribulations such as corruption, organized crime, impunity and intolerance are involved. Indeed, these evils affect the region and the entire globe.

This PhD project focuses on the study of the early stages of bone biomineralization in 2D and 3D cultures of osteoblast-like SaOS-2 osteosarcoma cells, exposed to an osteogenic cocktail. The efficacy of osteogenic treatment was assessed on 2D cell cultures after 7 days. A large calcium minerals production, an overexpression of osteogenic markers and of alkaline phosphatase activity occurred in treated samples. TEM microscopy and cryo-XANES micro-spectroscopy were performed for localizing and characterizing Ca-depositions. These techniques revealed a different localization and chemical composition of Ca-minerals over time and after treatment. Nevertheless, the Mito stress test showed in treated samples a significant increase in maximal respiration levels associated to an upregulation of mitochondrial biogenesis indicative of an ongoing differentiation process. The 3D cell cultures were realized using two different hydrogels: a commercial collagen type I and a mixture of agarose and lactose-modified chitosan (CTL). Both biomaterials showed good biocompatibility with SaOS-2 cells. The gene expression analysis of SaOS-2 cells on collagen scaffolds indicated an osteogenic commitment after treatment. and Alizarin red staining highlighted the presence of Ca-spots in the differentiated samples. In addition, the intracellular magnesium quantification, and the X-ray microscopy on mineral depositions, suggested the incorporation of Mg during the early stages of bone formation process., SaOS-2 cells treated with osteogenic cocktail produced Ca mineral deposits also on CTL/agarose scaffolds, as confirmed by alizarin red staining. Further studies are underway to evaluate the differentiation also at the genetic level. Thanks to the combination of conventional laboratory methods and synchrotron-based techniques, it has been demonstrated that SaOS-2 is a suitable model for the study of biomineralization in vitro. These results have contributed to a deeper knowledge of biomineralization process in osteosarcoma cells and could provide new evidences about a therapeutic strategy acting on the reversibility of tumorigenicity by osteogenic induction.
