The aim of this work was to perform a detailed investigation of the use of Selective Laser Melting (SLM) technology to process eutectic silver-copper alloy Ag 28 wt. % Cu (also called AgCu28). The processing occurred with a Realizer SLM 50 desktop machine. The powder analysis (SEM-topography, EDX, particle distribution) was reported as well as the absorption rates for the near-infrared (NIR) spectrum. Microscope imaging showed the surface topography of the manufactured parts. Furthermore, microsections were conducted for the analysis of porosity. The Design of Experiments approach used the response surface method in order to model the statistical relationship between laser power, spot distance and pulse time.

Il Selective Laser Melting è un processo di additive manufacturing che consiste nella realizzazione di componenti metallici tridimensionali, sovrapponendo strati di polvere, che viene via via fusa mediante una sorgente controllata di energia (laser). È una tecnica produttiva che viene utilizzata da più di 20 anni ma solo ora sta assumendo un ruolo rilevante nell’industria. È un processo versatile ma complesso che ad oggi permette di processare solo un numero limitato di leghe. Il presente lavoro di tesi riguarda in particolare lo studio, dal punto di vista microstrutturale, di componenti in acciaio inossidabile austenitico AISI-316L processato mediante Selective Laser Melting, attività svolta in collaborazione con il Gruppo di Tecnologia – Laser del Dipartimento di Ingegneria Industriale. Alla base dell’attività sperimentale è stata svolta anche un’ampia ricerca bibliografica per chiarire lo stato dell’arte sul processo e sulla lega in questione, la microstruttura, i difetti, le proprietà meccaniche e l’effetto dei parametri di processo sul componente finito. Le attività sperimentali hanno previsto una prima fase di caratterizzazione delle polveri di 316L, successivamente la caratterizzazione dei campioni prodotti tramite selective laser melting, in termini di microstruttura e difetti correlati al processo. Le analisi hanno rivelato la presenza di una microstruttura “gerarchica” costituita da melt pool, grani e celle submicrometriche. I difetti rinvenuti sono pori, delaminazione degli strati, particelle di polvere non fuse. Infine è stata eseguita la caratterizzazione frattografica dei campioni sottoposti a prove di trazione e di fatica a flessione rotante (attività condotte dal gruppo Laser) per identificare la morfologia di frattura e i siti di innesco della cricca di fatica.

Near-surface air temperature is an important determinant of the surface energy balance of glaciers and is often represented by a constant linear temperature gradients (TGs) in models. Spatiotemporal variability in 2 m air temperature was measured across the debris-covered Miage Glacier, Italy, over an 89 d period during the 2014 ablation season using a network of 19 stations. Air temperature was found to be strongly dependent upon elevation for most stations, even under varying meteorological conditions and at different times of day, and its spatial variability was well explained by a locally derived mean linear TG (MG–TG) of −0.0088°C m−1. However, local temperature depressions occurred over areas of very thin or patchy debris cover. The MG–TG, together with other air TGs, extrapolated from both on- and off-glacier sites, were applied in a distributed energy-balance model. Compared with piecewise air temperature extrapolation from all on-glacier stations, modelled ablation, using the MG–TG, increased by <1%, increasing to >4% using the environmental ‘lapse rate’. Ice melt under thick debris was relatively insensitive to air temperature, while the effects of different temperature extrapolation methods were strongest at high elevation sites of thin and patchy debris cover.

Different industrial induction melting processes involve free surface and melt-solid interface of the liquid metal subject to dynamic change during the technological operation. Simulation of the liquid metal dynamics requires to solve the non-linear, coupled hydrodynamic-electromagnetic-heat transfer problem accounting for the time development of the liquid metal free boundary with a suitable turbulent viscosity model. The present paper describes a numerical solution method applicable for various axisymmetric induction melting processes, such as, crucible with free top surface, levitation, semi-levitation, cold crucible and similar melting techniques. The presented results in the cases of semi-levitation and crucible with free top surface meltings demonstrate oscillating transient behaviour of the free metal surface indicating the presence of gravity-inertial-electromagnetic waves which are coupled to the internal fluid flow generated by both the rotational and potential parts of the electromagnetic force.

La modélisation de la cryolite, utilisée dans la fabrication de l’aluminium, implique plusieurs défis, notament la présence de discontinuités dans la solution et l’inclusion de la difference de densité entre les phases solide et liquide. Pour surmonter ces défis, plusieurs éléments novateurs ont été développés dans cette thèse. En premier lieu, le problème du changement de phase, communément appelé problème de Stefan, a été résolu en deux dimensions en utilisant la méthode des éléments finis étendue. Une formulation utilisant un multiplicateur de Lagrange stable spécialement développée et une interpolation enrichie a été utilisée pour imposer la température de fusion à l’interface. La vitesse de l’interface est déterminée par le saut dans le flux de chaleur à travers l’interface et a été calculée en utilisant la solution du multiplicateur de Lagrange. En second lieu, les effets convectifs ont été inclus par la résolution des équations de Stokes dans la phase liquide en utilisant la méthode des éléments finis étendue aussi. Troisièmement, le changement de densité entre les phases solide et liquide, généralement négligé dans la littérature, a été pris en compte par l’ajout d’une condition aux limites de vitesse non nulle à l’interface solide-liquide pour respecter la conservation de la masse dans le système. Des problèmes analytiques et numériques ont été résolus pour valider les divers composants du modèle et le système d’équations couplés. Les solutions aux problèmes numériques ont été comparées aux solutions obtenues avec l’algorithme de déplacement de maillage de Comsol. Ces comparaisons démontrent que le modèle par éléments finis étendue reproduit correctement le problème de changement phase avec densités variables.

Self-assembled materials produced in the reaction between alkanethiol and Ag are characterized and compared. It is revealed that the size of the Ag substrate has a significant role in the self-assembly process and determines the reaction products. Alkanethiol adsorbs on the surface of Ag continuous planar thin films and only forms self-assembled monolayers (SAMs), while the reaction between alkanethiol and Ag clusters on inert surfaces is more aggressive and generates a significantly larger amount of alkanethiolate. Two dissimilar products are yielded depending on the size of the clusters. Small Ag clusters are more likely to be converted into multilayer silver-alkanethiolate (AgSR, R = CnH2n+1) crystals, while larger Ag clusters form monolayer-protected clusters (MPCs). The AgSR crystals are initially small and can ripen into large lamellae during thermal annealing. The crystals have facets and flat terraces with extended area, and have a strong preferred orientation in parallel with the substrate surface. The MPCs move laterally upon annealing and reorganize into a single-layer network with their separation distance approximately equal to the length of an extended alkyl chain. AgSR lamellar crystals grown on inert surfaces provide an excellent platform to study the melting characteristics of crystalline lamellae of polymeric materials with the thickness in the nanometer scale. This system is also unique in that each crystal has integer number of layers – magic-number size (thickness). The size of the crystals is controlled by adjusting the amount of Ag and the annealing temperature. X-ray diffraction (XRD) and atomic force microscopy (AFM) are combined to accurately determine the size (number of layers) of the lamellar crystals. The melting characteristics are measured with nanocalorimetry and show discrete melting transitions which are attributed to the magic-number sizes of the lamellar crystals. The discrete melting temperatures are intrinsic properties of the crystals with particular sizes. Smaller lamellar crystals with less number of layers melt at lower temperatures. The melting point depression is inversely proportional to the total thickness of the lamellae – the product of the number of layers and the layer thickness.

High-resolution swath-bathymetry data from inner Kongsfjorden, Svalbard, reveal characteristic landform assemblages formed during and after surges of tidewater glaciers, and provide new insights into the dynamics of surging glaciers. Glacier front oscillations and overriding related to surge activity lead to the formation of overridden moraines, glacial lineations of two types, terminal moraines, associated debris lobes and De Geer moraines. In contrast to submarine landform assemblages from other Svalbard fjords, the occurrence of two kinds of glacial lineations and the presence of De Geer moraines suggest variability in the landforms produced by surge-type tidewater glaciers. All the landforms in inner Kongsfjorden were deposited during the last c. 150 years. Lithological and acoustic data from the innermost fjord reveal that suspension settling from meltwater plumes as well as ice rafting are dominant sedimentary processes in the fjord, leading to the deposition of stratified glacimarine muds with variable numbers of clasts. Reworking of sediments by glacier surging results in the deposition of sediment lobes containing massive glacimarine muds. Two sediment cores reveal minimum sediment accumulation rates related to the Kongsvegen surge from 1948; these were 30 cm a-1 approximately 2.5 km beyond the glacier front shortly after surge termination, and rapidly dropped to an average rate of 1.8 cm a-1 in ∼1950, during glacier retreat.

Abstract Where photosynthetic eukaryotic organisms survived during the Snowball Earth events of the Neoproterozoic remains unclear. Our previous research tested whether a narrow arm of the ocean, similar to the modern Red Sea, could have been a refugium for photosynthetic eukaryotes during the Snowball Earth. Using an analytical ice-flow model, we demonstrated that a limited range of climate conditions could restrict sea-glacier flow sufficiently to allow an arm of the sea to remain partially free from sea-glacier penetration, a necessary condition for it to act as a refugium. Here we expand on the previous study, using a numerical ice-flow model, with the ability to capture additional physics, to calculate sea-glacier penetration, and to explore the effect of a channel with a narrow entrance. The climatic conditions are made selfconsistent by linking sublimation rate to surface temperature. As expected, the narrow entrance allows parts of the nearly enclosed sea to remain safe from sea-glacier penetration for a wider range of climate conditions.

Insulin was used as model protein to developed innovative Solid Lipid Nanoparticles (SLNs) for the delivery of hydrophilic biotech drugs, with potential use in medicinal chemistry. SLNs were prepared by double emulsion with the purpose of promoting stability and enhancing the protein bioavailability. Softisan(®)100 was selected as solid lipid matrix. The surfactants (Tween(®)80, Span(®)80 and Lipoid(®)S75) and insulin were chosen applying a 2(2) factorial design with triplicate of central point, evaluating the influence of dependents variables as polydispersity index (PI), mean particle size (z-AVE), zeta potential (ZP) and encapsulation efficiency (EE) by factorial design using the ANOVA test. Therefore, thermodynamic stability, polymorphism and matrix crystallinity were checked by Differential Scanning Calorimetry (DSC) and Wide Angle X-ray Diffraction (WAXD), whereas the effect of toxicity of SLNs was check in HepG2 and Caco-2 cells. Results showed a mean particle size (z-AVE) width between 294.6 nm and 627.0 nm, a PI in the range of 0.425-0.750, ZP about -3 mV, and the EE between 38.39% and 81.20%. After tempering the bulk lipid (mimicking the end process of production), the lipid showed amorphous characteristics, with a melting point of ca. 30 °C. The toxicity of SLNs was evaluated in two distinct cell lines (HEPG-2 and Caco-2), showing to be dependent on the concentration of particles in HEPG-2 cells, while no toxicity in was reported in Caco-2 cells. SLNs were stable for 24 h in in vitro human serum albumin (HSA) solution. The resulting SLNs fabricated by double emulsion may provide a promising approach for administration of protein therapeutics and antigens.

G-quadruplexes are secondary structures present in DNA and RNA molecules, which are formed by stacking of G-quartets (i.e., interaction of four guanines (G-tracts) bounded by Hoogsteen hydrogen bonding). Human PAX9 intron 1 has a putative G-quadruplex-forming region located near exon 1, which is present in all known sequenced placental mammals. Using circular dichroism (CD) analysis and CD melting, we showed that these sequences are able to form highly stable quadruplex structures. Due to the proximity of the quadruplex structure to exon-intron boundary, we used a validated double-reporter splicing assay and qPCR to analyze its role on splicing efficiency. The human quadruplex was shown to have a key role on splicing efficiency of PAX9 intron 1, as a mutation that abolished quadruplex formation decreased dramatically the splicing efficiency of human PAX9 intron 1. The less stable, rat quadruplex had a less efficient splicing when compared to human sequences. Additionally, the treatment with 360A, a strong ligand that stabilizes quadruplex structures, further increased splicing efficiency of human PAX9 intron 1. Altogether, these results provide evidences that G-quadruplex structures are involved in splicing efficiency of PAX9 intron 1.

With the increase in life expectancy, biomaterials have become an increasingly important focus of research because they are used to replace parts and functions of the human body, thus contributing to improved quality of life. In the development of new biomaterials, the Ti-15Mo alloy is particularly significant. In this study, the Ti-15Mo alloy was produced using an arc-melting furnace and then characterized by density, X-ray diffraction, optical microscopy, hardness and dynamic elasticity modulus measurements, and cytotoxicity tests. The microstructure was obtained with β predominance. Microhardness, elasticity modulus, and cytotoxicity testing results showed that this material has great potential for use as biomaterial, mainly in orthopedic applications.
