Este trabalho descreve o desenvolvimento de metodologia para o controle de qualidade dos extratos etanólicos e infusão de Turnera diffusa Willd. ex Schult., Turneraceae, usando como marcadores os compostos fenólicos. Essa espécie é usada popularmente como afrodisíaca e antiulcerogênica. Foram usadas técnicas cromatográficas: CLAE acoplada com detector de ultravioleta e cromatografia em camada delgada. Foram detectados flavonóides como os principais componentes da planta.

Enzimas Peroxidases são heme-proteínas encontradas nos diferentes organismos vivos, especialmente vegetais, apresentam importante papel fisiológico/bioquímico como proteção contra microorganismos invasores. A soja, um dos mais importantes produtos para o agronegócio brasileiro apresenta na casca de suas sementes (subproduto) alta atividade de peroxidase, denominada soybean peroxidase,com potencial de utilização em métodos analíticos clínicos. A proposta do trabalho foi aplicar o planejamento fatorial para otimização das condições extração da enzima, definição das condições ótimas de atividade (pH e temperatura), utilizando metodologia de superfície de resposta. Os dados obtidos com clara definição foram: i) extração em pó cetonico, ii) meio reacional: pH 3,3, volume da amostra contendo a enzima 330 µL - 340 µL, peróxido de hidrogênio 4,2 mmol.L-1 150 µL, tempo de reação 20 segundos, temperatura 50º C, substrato guaiacol 30mmol.L-1 300 µL, e 0,1 mol.L-1 de NaCl. O uso da dessa metodologia para definição das condições de extração e estudos cinético-enzimáticos da peroxidase de soja foram eficientes e mais precisos, comparado a metodologia de variações/repetições (tentativa e erro).

A importância do estudo de bactérias acéticas, em especial as do gênero Gluconobacter, está baseada em suas aplicações industriais, pois estas possuem a capacidade de bioconversão de sorbitol a sorbose, viabilizando o processo de produção de vitamina C. O estudo envolveu coletas de amostras em indústrias de refrigerante, flores, frutos e mel, seguidas de purificação, identificação fenotípica e identificação molecular, com a utilização de iniciador definido a partir de consulta ao Nucleotide Sequence Database. Preservaram-se as linhagens identificadas como membros da família Acetobacteriaceae, gênero Gluconobacter. Foi isolado um total de 110 linhagens dos substratos: Pyrostegia venusta (Cipó de São João), mel, Vitis vinifera (uva), Pyrus communis (pêra), Malus sp. (maçã) e de duas amostras de refrigerantes envasados em embalagens de PET de 2 L. Deste total, 57 linhagens foram recuperadas em meio MYP (manitol, extrato de levedura, peptona), 12 em meio YGM (glicose, manitol, extrato de levedura, etanol, ácido acético), 41 em meio de enriquecimento e, posteriormente, em meio GYC (glicose, extrato de levedura e carbonato de cálcio). Obtiveram-se 68 linhagens identificadas como bastonetes Gram negativos. Destas, 31 foram caracterizadas bioquimicamente como pertencentes à família Acetobacteriaceae por serem catalase positivas, oxidase negativas e produtoras de ácido a partir de glicose. A caracterização dessas linhagens foi complementada com os testes bioquímicos: liquefação da gelatina, redução de nitrato, formação de indol e H2S e oxidação de etanol a ácido acético. Métodos moleculares foram aplicados para identificação do gênero Gluconobacter. Finalmente, oito linhagens foram caracterizadas como pertencentes ao gênero Gluconobacter. As linhagens encontram-se depositadas em coleção de cultura do laboratório de Microbiologia do Departamento de Biologia da UNESP, campus de Assis, estocadas em extrato de malte 20 a -196 ºC.

Pós nanométricos SnO2.Nb2O5 foram estudados para o desenvolvimento de sensores de etanol. Estes pós foram preparados pelo método Pechini, caracterizados quanto à sua morfologia por difração de raios X, determinação de área superficial específica por BET e Microscopia Eletrônica de Transmissão e foram submetidos a testes de sensibilidade ao vapor de etanol. Foi estabelecida uma correlação entre a microestrutura do material, os efeitos do dopante e a resposta do sensor.

Cinco éguas mestiças (idade e peso corporal médios de seis anos e 480 kg PV) foram distribuídas em delineamento experimental em quadrado latino para se e avaliar a utilização de dietas contendo grãos de triticale (secos ou ensilados) em substituição ao milho na alimentação de eqüinos, por meio do ensaio de avaliação do trânsito gastrintestinal e da digestibilidade. Os tratamentos consistiram de três níveis de grãos de triticale em substituição aos grãos de milho (0, 50 e 100%) e duas formas de conservação dos grãos de triticale (secos ou ensilados). As dietas foram isoprotéicas (12,5% PB), com ingestão diária de MS pelos animais de 2,0% PV (relação volumoso : concentrado de 50:50). Os coeficientes de digestibilidade aparente (CDa) da MS, MO, PB e FDN foram determinados indiretamente. Os parâmetros de trânsito gastrintestinal avaliados foram: k1 (taxa de passagem pelo intestino grosso), k2 (taxa de passagem pelo estômago), TT (tempo de trânsito), TMR (tempo médio de retenção) e TMRT (tempo médio de retenção total). Não houve diferença para os CDa da MS, MO, PB e FDN entre as dietas experimentais, observando-se valores médios de 64,31; 65,14; 74,13 e 57,25%, respectivamente. Considerando-se a cinética das fases sólida e líquida, notou-se efeito somente para k2 na fase sólida da digesta, cujo valor nas rações contendo 100% de triticale (seco ou ensilado) foi de 19,63%/h e na dieta controle (0% de triticale), de 23,72%/h. Observou-se efeito linear crescente para o TT na fase sólida da digesta, com a elevação dos níveis de substituição dos grãos de milho pelos grãos de triticale ensilados. Concluiu-se que os concentrados para eqüinos podem ser formulados com grãos de triticale secos ou ensilados em substituição total ao milho. A inclusão de grãos de triticale na alimentação desta espécie animal promoveu trânsito lento da fase sólida da digesta.

Droplets formed at the tip of a tube under the same conditions possess extreme uniformity of form, volume and weight. These properties of liquid drop formation have been known for a long time and consequently many applications for the drop have been found in instrumentation and chemical analysis methods. In the present paper, we report on the analytical use of a dynamic LED-based flow-through optical absorption detector with optical path length controlled by continuous dropping of a solution. This arrangement consists of a flow cell built within a high-intensity red LED (lambda (max)=630 nm). The feasibility of the detector is demonstrated by colorimetric determination of methylene blue, and ammonium by Berthelot's reaction, in a flow-injection system. For ammonium, the reaction forms a blue dye (indophenol) with a maximum absorption at 630-650 nm. The detection limit, considered as 3 times the signal of the blank, is better than 125 mu g l(-1). The small flow cell represents a good combination of optical path length, low volume and fast washout. This detector can be used advantageously in automated methods and can represent a solution to problems of optical detection involving gas bubbles and precipitation of particles in turbidimetric applications.

We measured progesterone and estradiol levels from birth to the beginning of adult life in 10 Thoroughbred fillies from the Equilia Stud Farm in Avare SP, Brasil. The animals were measured and weighed monthly for the determination of body development and of a possible correlation between the rate of weight and height gain and the onset of detectable sex hormone levels. Jugular blood was collected twice a week and stored at -20 degrees C until assay of progesterone by a solid phase RIA with a sensitivity of 0.32 nmol/L and of estradiol by liquid phase RIA adapted to low levels (3.67 pmol/L). The fillies were born with high serum levels of both hormones,which fell to undetectable levels by the first week of life. A variation in growth rate was observed, with the highest levels occuring from birth to the 3rd month of life, followed by a reduction until 15 mo of life, when fast growth was resumed. The monthly weight gain was 1.5% when the fillies reached puberty and 5.4% during the next month, (P<0.05, Friedman test). During this second period of accelerated growth after the beginning of progesterone production at detectable levels (above 0.318 nmol/L), the parameters of skeletal growth did not differ (P>0.05). The month of onset of puberty was the month of lowest weight gain in the life of the fillies, and it coincided with the highest insolation period. In conclusion, horses, like all other developed vertebrates, have a double pattern of development, with the acceleration observed at puberty depending on sex steroids, which in turn coincides with the highest insolation period. Gonadal activity characterized by serum progesterone levels was low from birth to the onset of puberty. After puberty the progesterone cycles were similar to those of adult animals with a mature hypothalamic-gonadal axis. (C) 1997 by Elsevier B.V.

The efficiency of different methods for the decontamination of glassware used for the analysis of dissolved organic carbon (DOC) was tested using reported procedures as well as new ones proposed in this work. A Fenton solution bath (1.0 mmol L-1 Fe2+ and 100 mmol L-1 H2O2) for 1 h or for 30 min employing UV irradiation showed to combine simplicity, low cost and high efficiency. Using the optimized cleaning procedure, the DOC for stored UV-irradiated ultrapure water reached concentrations below the limit of detection (0.19 mu mol C L-1). Filtered (0.7 mu m) rain samples maintained the DOC integrity for at least 7 days when stored at 4 degrees C. The volatile organic carbon (VOC) fraction in the rain samples collected at two sites in São Paulo state (Brazil) ranged from 0% to 56% of their total DOC content. Although these high-VOC concentrations may be derived from the large use of ethanol fuel in Brazil, our results showed that when using the high-temperature catalytic oxidation technique, it is essential to measure DOC rather than non-purgeble organic carbon to estimate organic carbon, since rainwater composition can be quite variable, both geographically and temporally. The objective of this study was to evaluate the allelophatic potential ethanol extract of leaves of Anadenanthera macrocarpa (Benth.) Brenan (angico) and Astronium graveolens Jacq (aroeira) on the biomonitor germination of Brassica chinensis (Malaysia's cabbage) and Lactuca sativa (lettuce). For the test of allelopathy was the bioassay of germination of B. chinensis and L. sativa using solutions of ethanol extracts in different concentrations (5, 10 and 20 mg / mL) with continuous light for 11 days. The experimental and control groups were divided into the Petri dish containing 50 seeds of B. chinensis and L. sativa each. Were performed six replicates for each concentration of extract of leaves of A. macrocarpa and A. graveolens and the negative control (water). The rolling action of the treatments was performed by quantitative observation and monitoring of germination every six hours. The results show that the ethanol extract of A. macrocarpa and A. graveolens presents allelopathic potential, regardless of the concentration used, but with answers dose-dependent, and thus able to interfere directly in germination, average speed and time of germination biomonitor of B. chinensis and L. sativa.

Propolis has been used in folk medicine and possesses a broad spectrum of biological activities, specially antibacterial activity. Studies have demonstrated that the composition of propolis extract may have influence in such activity. The goal of this study was to investigate the antibacterial activity of eleven propolis extracts (PE) against sixty one Staphylococcus aureus strains, isolated from newborn clinical specimens. The PE from Apis mellifera were prepared by using pure water and mixtures of water with ethanol at different concentrations (from 0 to 100%), 25g of propolis in 100 mL of solvent, and three days of maceration followed by filtration. Determination of Minimal Inhibitory Concentration (MIC) by agar dilution method was performed and serial concentrations from each PE were achieved (%v/v) in plates containing Mueller Hinton agar. It was possible to verify that the anti S. aureus activity was directly proportional to ethanol concentration and no significant differences were observed among PE with ethanol concentration from 70 to 100%. The MIC 90% values ranged from 0.4 to 0.6% (v/v) and the 70% ethanolic extract were the most efficient to inhibit bacterial growth (MIC 90%=0.42%, v/v). In conclusion, our results suggest that the EP composition and, consequently, the concentration of ethanol used as solvent may influence the antibacterial activity of propolis from A. mellifera.

In this study it is reported the operation of a horizontalflow anaerobic immobilized biomass (HAlB) reactor under sulfate-reducing condition which was also exposed to different amounts of ethanol and benzene. The HAIB reactor comprised of an immobilized biomass on polyurethane foam and ferrous and sodium sulfate solutions were used (91 and 550 mg.l -1, respectively), to promote a sulfate-reducing environment. Benzene was added at an initial concentration of 2.0 mg.l -1 followed by an increased to 9 e 10 mg. l -1, respectively. Ethanol was added at an initial concentration of 170 mg.l -1 followed by an increased range of 960 mg.l -1. The reactor was operated at 30 (± 2) °C with hydraulic detention time of 12 h. Organic matter removal efficiency of 90% with a maximum benzene degradation rate of 0.07 mv, benzene.mg -1 vss.d -1 Thus, this work corroborate the data obtained for Cattony et al (2005) and also demonstrate that compact units of HAIB reactors, under sulfate reducing conditions, are a potential alternative for in situ aromatic compounds bioremediation.

The objective of this work was to make a comparative analysis of germ cell organization at different stages of cellular differentiation in adult males of Dendropsophus nanus (Boulenger, 1889), Pseudis limellum (Cope, 1862), P. paradoxa (Linnaeus, 1758), and Scinax acuminatus (Cope, 1862), belonging to the family Hylidae; and Leptodactylus chaquensis (Cei, 1950) and L. podicipinus (Cope, 1862), belonging to the family Leptodactylidae, collected in the Pantanal and in Serra da Bodoquena, Mato Grosso do Sul State, Brazil. The testes were removed and fixed, dehydrated in a graded series of ethanol and embedded in methacrylate glycol resin (Historesin Leica®). The sections were stained by 1% toluidine blue and observed under light microscope. It was detected that all individual of the Hylidae family show, throughout the year, the presence of all germ cell types of spermatogenesis. However, all Leptodactylidae family individuals only show the presence of all germ cell types during the rainy season. The variations of characteristics in seminiferous epithelium organization, as well as the evident difference in the amount of spermatozoa inside the tubules, are evidence that the anurans in this work show different forms of spermatogenesis development throughout the year: the cycle is continuous for the Hylidae family, and discontinuous with explosive release of spermatozoa for the Leptodactylidae family.
