Quelque 30 % de la population neuronale du cortex mammalien est composée d’une population très hétérogène d’interneurones GABAergiques. Ces interneurones diffèrent quant à leur morphologie, leur expression génique, leurs propriétés électrophysiologiques et leurs cibles subcellulaires, formant une riche diversité. Après leur naissance dans les éminences ganglioniques, ces cellules migrent vers les différentes couches corticales. Les interneurones GABAergiques corticaux exprimant la parvalbumin (PV), lesquels constituent le sous-type majeur des interneurones GABAergiques, ciblent spécifiquement le soma et les dendrites proximales des neurones principaux et des neurones PV+. Ces interneurones sont nommés cellules à panier (Basket Cells –BCs) en raison de la complexité morphologique de leur axone. La maturation de la connectivité distincte des BCs PV+, caractérisée par une augmentation de la complexité de l’axone et de la densité synaptique, se déroule graduellement chez la souris juvénile. Des travaux précédents ont commencé à élucider les mécanismes contrôlant ce processus de maturation, identifiant des facteurs génétiques, l’activité neuronale ainsi que l’expérience sensorielle. Cette augmentation marquante de la complexité axonale et de la synaptogénèse durant cette phase de maturation suggère la nécessité d’une synthèse de protéines élevée. La voie de signalisation de la cible mécanistique de la rapamycine (Mechanistic Target Of Rapamycin -mTOR) a été impliquée dans le contrôle de plusieurs aspects neurodéveloppementaux en régulant la synthèse de protéines. Des mutations des régulateurs Tsc1 et Tsc2 du complexe mTOR1 causent la sclérose tubéreuse (TSC) chez l’humain. La majorité des patients TSC développent des problèmes neurologiques incluant des crises épileptiques, des retards mentaux et l’autisme. D’études récentes ont investigué le rôle de la dérégulation de la voie de signalisation de mTOR dans les neurones corticaux excitateurs. Toutefois, son rôle dans le développement des interneurones GABAergiques corticaux et la contribution spécifique de ces interneurones GABAergiques altérés dans les manifestations de la maladie demeurent largement inconnus. Ici, nous avons investigué si et comment l’ablation du gène Tsc1 perturbe le développement de la connectivité GABAergique, autant in vitro que in vivo. Pour investiguer le rôle de l’activation de mTORC1 dans le développement d’une BC unique, nous avons délété le gène Tsc1 en transfectant CRE-GFP dirigé par un promoteur spécifique aux BCs dans des cultures organotypiques provenant de souris Tsc1lox. Le knockdown in vitro de Tsc1 a causé une augmentation précoce de la densité des boutons et des embranchements terminaux formés par les BCs mutantes, augmentation renversée par le traitement à la rapamycine. Ces données suggèrent que l’hyperactivation de la voie de signalisation de mTOR affecte le rythme de la maturation des synapses des BCs. Pour investiguer le rôle de mTORC1 dans les interneurones GABAergiques in vivo, nous avons croisé les souris Tsc1lox avec les souris Nkx2.1-Cre et PV-Cre. À P18, les souris Tg(Nkx2.1-Cre);Tsc1flox/flox ont montré une hyperactivation de mTORC1 et une hypertrophie somatique des BCs de même qu’une augmentation de l’expression de PV dans la région périsomatique des neurones pyramidaux. Au contraire, à P45 nous avons découvert une réduction de la densité des punctas périsomatiques PV-gephyrin (un marqueur post-synaptique GABAergique). L’étude de la morphologie des BCs en cultures organotypiques provenant du knock-out conditionnel Nkx2.1-Cre a confirmé l’augmentation initiale du rythme de maturation, lequel s’effondre ensuite aux étapes développementales tardives. De plus, les souris Tg(Nkx2.1Cre);Tsc1flox/flox montrent des déficits dans la mémoire de travail et le comportement social et ce d’une façon dose-dépendante. En somme, ces résultats suggèrent que l’activation contrôlée de mTOR régule le déroulement de la maturation et la maintenance des synapses des BCs. Des dysfonctions de la neurotransmission GABAergique ont été impliquées dans des maladies telles que l’épilepsie et chez certains patients, elles sont associées avec des mutations du récepteur GABAA. De quelle façon ces mutations affectent le processus de maturation des BCs demeuret toutefois inconnu. Pour adresser cette question, nous avons utilisé la stratégie Cre-lox pour déléter le gène GABRA1, codant pour la sous-unité alpha-1 du récepteur GABAA dans une unique BC en culture organotypique. La perte de GABRA1 réduit l’étendue du champ d’innervation des BCs, suggérant que des variations dans les entrées inhibitrices en raison de l’absence de la sous-unité GABAAR α1 peuvent affecter le développement des BCs. La surexpression des sous-unités GABAAR α1 contenant des mutations identifiées chez des patients épileptiques ont montré des effets similaires en termes d’étendue du champ d’innervation des BCs. Pour approfondir, nous avons investigué les effets de ces mutations identifiées chez l’humain dans le développement des épines des neurones pyramidaux, lesquelles sont l’endroit privilégié pour la formation des synapses excitatrices. Somme toute, ces données montrent pour la première fois que différentes mutations de GABRA1 associées à des syndromes épileptiques peuvent affecter les épines dendritiques et la formation des boutons GABAergiques d’une façon mutation-spécifique.

The present-day condition of bipolar glaciation characterized by rapid and large climate fluctuations began at the end of the Pliocene with the intensification of the Northern Hemisphere continental glaciations. The global cooling steps of the late Pliocene have been documented in numerous studies of Ocean Drilling Program (ODP) sites from the Northern Hemisphere. However, the interactions between oceans and between land and ocean during these cooling steps are poorly known. In particular, data from the Southern Hemisphere are lacking. Therefore I investigated the pollen of ODP Site 1082 in the southeast Atlantic Ocean in order to obtain a high-resolution record of vegetation change in Namibia between 3.4 and 1.8 Ma. Four phases of vegetation development are inferred that are connected to global climate change. (1) Before 3 Ma, extensive, rather open grass-rich savannahs with mopane trees existed in Namibia, but the extension of desert and semidesert vegetation was still restricted. (2) Increase of winter rainfall dependent Renosterveld-like vegetation occurred between 3.1 and 2.2 Ma connected to strong advection of polar waters along the Namibian coast and a northward shift of the Polar Front Zone in the Southern Ocean. (3) Climatically induced fluctuations became stronger between 2.7 and 2.2 Ma and semiarid areas extended during glacial periods probably as the result of an increased pole-equator thermal gradient and consequently globally enhanced atmospheric circulation. (4) Aridification and climatic variability further increased after 2.2 Ma, when the Polar Front Zone migrated southward and the influence of Atlantic moisture brought by the westerlies to southern Africa declined. It is concluded that the positions of the frontal systems in the Southern Ocean which determine the locations of the high-pressure cells over the South Atlantic and the southern Indian Ocean have a strong influence on the climate of southern Africa in contrast to the climate of northwest and central Africa, which is dominated by the Saharan low-pressure cell.

Paleoenvironmental studies and climate models demonstrate that fluvial runoff and moisture availability in the Caribbean hinterland react very sensitively to climatic variations. Late Pleistocene and Holocene climate records document pronounced dry and wet periods over tropical South America mainly caused by shifts of the Intertropical Convergence Zone (ITCZ). However, forcing mechanisms for changes in the ITCZ position remain controversial. Here we present high-resolution foraminiferal Ba/Ca and d18Oseawater records from a core located within the Orinoco River outflow documenting abrupt hydrological changes in the Orinoco catchment area during the deglacial and Holocene. Our data, obtained from the surface-dwelling foraminifera Globigerinoides ruber (pink), show an abrupt increase in Ba/Ca ratios in the early Holocene, starting ~600 yr after the end of the Younger Dryas (YD) cold interval at ca. 10.8 ka and suggesting a massive reorganization of moisture sources in northern South America. In contrast, the salinity dependent d18Oseawater from the same samples shows a gradual decrease starting at the end of the YD. The offset of our Ba/Ca peak excludes meltwater release in conjunction with the northern Andean glacier retreat well before the end of the YD as a forcing mechanism. We suggest that the Ba/Ca record documents an abrupt increase in Ba-rich waters of a northern Andean source caused by the insolation-driven shift of the ITCZ and/or enhanced monsoon activity.

Application of the 230Th normalization method to estimate sediment burial fluxes in six cores from the eastern equatorial Pacific (EEP) reveals that bulk sediment and organic carbon fluxes display a coherent regional pattern during the Holocene that is consistent with modern oceanographic conditions, in contrast with estimates of bulk mass accumulation rates (MARs) derived from core chronologies. Two nearby sites (less than 10 km apart), which have different MARs, show nearly identical 230Th-normalized bulk fluxes. Focusing factors derived from the 230Th data at the foot of the Carnegie Ridge in the Panama Basin are >2 in the Holocene, implying that lateral sediment addition is significant in this part of the basin. New geochemical data and existing literature provide evidence for a hydrothermal source of sediment in the southern part of the Panama Basin and for downslope transport from the top of the Carnegie Ridge. The compilation of core records suggests that sediment focusing is spatially and temporally variable in the EEP. During oxygen isotope stage 2 (OIS 2, from 13-27 ka BP), focusing appears even higher compared to the Holocene at most sites, similar to earlier findings in the eastern and central equatorial Pacific. The magnitude of the glacial increase in focusing factors, however, is strongly dependent on the accuracy of age models. We offer two possible explanations for the increase in glacial focusing compared to the Holocene. The first one is that the apparent increase in lateral sediment redistribution is partly or even largely an artifact of insufficient age control in the EEP, while the second explanation, which assumes that the observed increase is real, involves enhanced deep sea tidal current flow during periods of low sea level stand.

Over the last several decades debates on the 'tempo and mode' of evolution have centered on the question whether morphological evolution preferentially occurs gradually or punctuated, i.e., with long periods of stasis alternating with short periods of rapid morphological change and generation of new species. Another major debate is focused on the question whether long-term evolution is driven by, or at least strongly influenced by changes in the environment, or by interaction with other life forms. Microfossils offer a unique opportunity to obtain the large datasets as well as the precision in dating of subsequent samples to study both these questions.We present high-resolution analyses of selected calcareous nannofossils from the deep-sea section recovered at ODP Site 1262 (Leg 208) in the South-eastern Atlantic. The studied section encompasses nannofossil Zones NP4-NP12 (equivalent to CP3-CP10) and Chrons C27r-C24n. We document more than 70 biohorizons occurring over an about 10 Myr time interval, (~62.5 Ma to ~52.5 Ma), and discuss their reliability and reproducibility with respect to previous data, thus providing an improved biostratigraphic framework, which we relate to magnetostratigraphic information, and present for two possible options of a new Paleocene stratigraphic framework based on cyclostratigraphy. This new framework enabled us to tentatively reconstruct steps in the evolution of early Paleogene calcareous nannoplankton through documentation of transitional morphotypes between genera and/or species and of the phylogenetic relations between the genera Fasciculithus, Heliolithus, Discoasteroides and Discoaster, as well as between Rhomboaster and Tribrachiatus. The exceptional record provided by the continuous, composite sequence recovered at Walvis Ridge allows us to describe the mode of evolution among calcareous nannoplankton: new genera and/or new species usually originated through branching of lineages via gradual, but relatively rapid, morphological transitions, as documented by the presence of intermediate forms between the end-member ancestral and descendant forms. Significant modifications in the calcareous nannofossil assemblages are often "related" to significant changes in environmental conditions, but the appearance of structural innovations and radiations within a single genus also occurred during "stable" environmental conditions. These lines of evidence suggest that nannoplankton evolution is not always directly triggered by stressed environmental conditions but could be also driven by endogenous biotic control.

Planktonic foraminiferal faunas of the southeast Pacific indicate that sea surface temperatures (SST) have varied by as much as 8-10°C in the Peru Current, and by ?5-7°C along the equator, over the past 150,000 years. Changes in SST at times such as the Last Glacial Maximum reflect incursion of high-latitude species Globorotalia inflata and Neogloboquadrina pachyderma into the eastern boundary current and as far north as the equator. A simple heat budget model of the equatorial Pacific shows that observed changes in Peru Current advection can account for about half of the total variability in equatorial SSTs. The remaining changes in equatorial SST, which are likely related to local changes in upwelling or pycnocline depth, precede changes in polar climates as recorded by d18O. This partitioning of processes in eastern equatorial Pacific SST reveals that net ice-age cooling here reflects first a rapid response of equatorial upwelling to insolation, followed by a later response to changes in the eastern boundary current associated with high-latitude climate (which closely resembles variations in atmospheric CO2 as recorded in the Vostok ice core). Although precise mechanisms responsible for the equatorial upwelling component of climate change remain uncertain, one likely candidate that may operate independently of the ice sheets is insolation-driven changes in El Niño/Southern Oscillation (ENSO) frequency. Early responses of equatorial SST detected both here and elsewhere highlight the sensitivity of tropical systems to small changes in seasonal insolation. The scale of tropical changes we have observed are substantially greater than model predictions, suggesting a need for further quantitative assessment of processes associated with long-term climate change.

A 270 cm long sediment sequence was recovered with a piston corer from east lobe Bonney, Taylor Valley, Antarctica, and characterized according to its sedimentological, mineralogical, and geochemical properties. It is the first record of such length recovered from east lobe Bonney. The sediment core is mainly composed of halite crystals of different sizes, water, and a relatively low and stable proportion of clastic particles. Although the sediment surface was probably disturbed by the coring process and absence or low contents of organic material or carbonates hampers the establishment of a robust chronology by radiocarbon dating, the core probably contains at least several hundred years of information about the history of the lake and the Bonney basin. Variations in halite crystal sizes and amount as well as variations in the composition of clastic material can be related to past lake level changes and evaporation cycles.

The breast cancer susceptibility gene Brca1 encodes a large multi-functional protein which is implicated as a caretaker of the genome, through its role in regulation of DNA damage response pathways, including apoptosis. Here we show that in mice expressing a dominant-negative Brca1 transgene on a BALB/c background, vaginal entrance remodeling is inhibited, and that the incidence of this phenotype is increased on a p53 +/- genotype. Given that this developmental process is mediated primarily by apoptosis, we hypothesized that disruption of BRCA1 may confer a resistance to apoptosis in normal epithelial cells. Consistent with this, we show that expression of this transgene in vitro leads to resistance to ionizing radiation induced cell killing in mammary epithelial cells. This is the first time that BRCA1 has been implicated in an apoptosis-mediated normal developmental process.

Stable expression of human groups IIA and X secreted phospholipases A(2) (hGIIA and hGX) in CHO-K1 and HEK293 cells leads to serum- and interleukin-1beta-promoted arachidonate release. Using mutant CHO-K1 cell lines, it is shown that this arachidonate release does not require heparan sulfate proteoglycan- or glycosylphosphatidylinositol-anchored proteins. It is shown that the potent secreted phospholipase A(2) inhibitor Me-Indoxam is cell-impermeable. By use of Me-Indoxam and the cell-impermeable, secreted phospholipase A(2) trapping agent heparin, it is shown that hGIIA liberates free arachidonate prior to secretion from the cell. With hGX-transfected CHO-K1 cells, arachidonate release occurs before and after enzyme secretion, whereas all of the arachidonate release from HEK293 cells occurs prior to enzyme secretion. Immunocytochemical studies by confocal laser and electron microscopies show localization of hGIIA to the cell surface and Golgi compartment. Additional results show that the interleukin-1beta-dependent release of arachidonate is promoted by secreted phospholipase A(2) expression and is completely dependent on cytosolic (group IVA) phospholipase A(2). These results along with additional data resolve the paradox that efficient arachidonic acid release occurs with hGIIA-transfected cells, and yet exogenously added hGIIA is poorly able to liberate arachidonic acid from mammalian cells.

Cementum is known to be growth-hormone (GH)-responsive, but to what extent is unclear. This study examines the effects of extremes of GH status on cementogenesis in three lines of genetically modified mice; GH excess (giant), GH antagonist excess (dwarf), and GH receptor-deleted (GHR-KO) (dwarf). Age-matched mandibular molar tissues were processed for light microscope histology. Digital images of sections of first molar teeth were captured for morphometric analysis of lingual root cementum. The study aimed to examine the factors influencing referral to rehabilitation following traumatic brain injury (TBI) by using social problems theory as a conceptual model to focus on practitioners and the process of decision-making in two Australian hospitals. The research design involved semi-structured interviews with 18 practitioners and observations of 10 team meetings, and was part of a larger study on factors influencing referral to rehabilitation in the same settings. Analysis revealed that referral decisions were influenced primarily by practitioners' selection and their interpretation of clinical and non-clinical patient factors. Further, practitioners generally considered patient factors concurrently during an ongoing process of decision-making, with the combinations and interactions of these factors forming the basis for interpretations of problems and referral justifications. Key patient factors considered in referral decisions included functional and tracheostomy status, time since injury, age, family, place of residence and Indigenous status. However, rate and extent of progress, recovery potential, safety and burden of care, potential for independence and capacity to cope were five interpretative themes, which emerged as the justifications for referral decisions. The subsequent negotiation of referral based on patient factors was in turn shaped by the involvement of practitioners. While multi-disciplinary processes of decision-making were the norm, allied health professionals occupied a central role in referral to rehabilitation, and involvement of medical, nursing and allied health practitioners varied. Finally, the organizational pressures and resource constraints, combined with practitioners' assimilation of the broader efficiency agenda were central factors shaping referral. Elevated plasma homocysteine is recognized as an independent risk factor for cardiovascular disease. Recently, there have been conflicting reports of the relationship between physical activity and homocysteine. A more objective measure of physical activity is cardiorespiratory fitness; however, its relationship with homocysteine has yet to be investigated. The aim of this study was to determine the relationship between cardiorespiratory fitness and plasma homocysteine. Cross-sectional associations between cardiorespiratory fitness (VO(2)max) and plasma homocysteine were examined in 49 men and 11 women. A submaximal bicycle test was used to determine VO(2)max and plasma homocysteine was measured using high performance liquid chromatography with fluorescence detection. Dietary analysis determined B vitamin intake. There was a significant inverse relationship between plasma homocysteine concentration and VO(2)max in women (r = -0.81, P = 0.003) but not in men (r = -0.09, P = 0.95). There were no significant relationships between plasma homocysteine and age, BMI, body fat, total cholesterol, and LDL cholesterol. In summary, elevated cardiorespiratory fitness is associated with decreased plasma homocysteine concentrations in women. (C) 2004 Elsevier Inc. All rights reserved.

Increasingly, large areas of native tropical forests are being transformed into a mosaic of human dominated land uses with scattered mature remnants and secondary forests. In general, at the end of the land clearing process, the landscape will have two forest components: a stable component of surviving mature forests, and a dynamic component of secondary forests of different ages. As the proportion of mature forests continues to decline, secondary forests play an increasing role in the conservation and restoration of biodiversity. This paper aims to predict and explain spatial and temporal patterns in the age of remnant mature and secondary forests in lowland Colombian landscapes. We analyse the age distributions of forest fragments, using detailed temporal land cover data derived from aerial photographs. Ordinal logistic regression analysis was applied to model the spatial dynamics of mature and secondary forest patches. In particular, the effect of soil fertility, accessibility and auto-correlated neighbourhood terms on forest age and time of isolation of remnant patches was assessed. In heavily transformed landscapes, forests account for approximately 8% of the total landscape area, of which three quarters are comprised of secondary forests. Secondary forest growth adjacent to mature forest patches increases mean patch size and core area, and therefore plays an important ecological role in maintaining landscape structure. The regression models show that forest age is positively associated with the amount of neighbouring forest, and negatively associated with the amount of neighbouring secondary vegetation, so the older the forest is the less secondary vegetation there is adjacent to it. Accessibility and soil fertility also have a negative but variable influence on the age of forest remnants. The probability of future clearing if current conditions hold is higher for regenerated than mature forests. The challenge of biodiversity conservation and restoration in dynamic and spatially heterogeneous landscape mosaics composed of mature and secondary forests is discussed. (c) 2004 Elsevier B.V. This study examined relations between stress and coping predictors and negative and positive outcomes in MS caregiving. A total of 222 carers and their care-recipients completed questionnaires at Time 1 and three months later, Time 2 ( n = 155). Predictors included care-recipient characteristics ( age, time since diagnosis, course and life satisfaction), and Times 1 and 2 carer problems, stress appraisal and coping. Dependent variables were Time 2 negative ( anxiety, depression) and positive outcomes ( life satisfaction, positive affect, benefits). Regressions indicated that, overall, the hypothesised direct effects of stress appraisal and coping strategies on positive and negative outcomes were supported. The hypothesised stress-buffering effects of positive reframing coping were also supported. All but one of the coping strategies were related to both positive and negative outcomes; specifically, practical assistance coping emerged as a unique predictor of distress. Of the model predictors, care-recipient life satisfaction emerged as the strongest and most consistent predictor of both positive and negative outcomes except benefit finding. Findings support the role of care-recipient characteristics and the carer's appraisal and coping processes in shaping both positive and negative outcomes. The guiding framework and findings have the potential to inform interventions designed to promote well-being in carers.
