Tumor-reactive T cells play an important role in cancer immunosurveillance. Applying the multimer technology, we report here an unexpected high frequency of Melan-A-specific CTLs in a melanoma patient with progressive lymph node metastases, consisting of 18 and 12.8% of total peripheral blood and tumor-infiltrating CD8+ T cells, respectively. Melan-A-specific CTLs revealed a high cytolytic activity against allogeneic Melan-A-expressing target cells but failed to kill the autologous tumor cells. Loading of the tumor cells with Melan-A peptide reversed the resistance to killing, suggesting impaired function of the MHC class I antigen processing and presentation pathway. Mutations of the coding region of the HLA-A2 binding Melan-A26-35 peptide or down-regulation of the MHC class I heavy chain, the antigenic peptide TAP, and tapasin could be excluded. However, PCR and immunohistochemical analysis revealed a deficiency of the immunoproteasomes low molecular weight protein 2 and low molecular weight protein 7 in the primary tumor cells, which affects the quantity and quality of generated T-cell epitopes and might explain the resistance to killing. This is supported by our data, demonstrating that the resistance to killing can be partially reversed by pre-exposure of the tumor cells to IFN-gamma, which is known to induce the immunoproteasomes. Overall, this is the first report of an extremely high frequency of tumor-specific CTLs that exhibit competent T-cell-effector functions but fail to lyse the autologous tumor cells. Immunotherapeutic approaches should not only focus on the induction of a robust antitumor immune response, but should also have to target tumor immune escape mechanisms.

El estudio se realizó a bordo de dos barcos y en forma simultánea: BIC Humboldt que cubrió de 100 a 200 mn y BIC Olaya, dentro de las 100 mn. Las observaciones más importantes fueron: (a) presencia de las aguas subtropicales superficiales (ASS) de Pimentel (8°S) hasta la frontera sur (18°S), con una notable advección a la costa de Huarmey (10°S), al Callao (12°S) y al sur de los 15°S (San Juan de Marcona); (b) proyección de aguas ecuatoriales superficiales (AES) hasta los 5°20’S, por el arribo de una nueva onda Kelvin; (c) registro de aguas ecuatoriales superficiales (AES) entre 6°30’ y 8°S por fuera de las 140 mn, constituyendo rezagos de las ondas que arribaron en agosto; (d) predominancia de anomalías térmicas positivas de hasta +1,5 °C, relacionadas con la persistencia de las ondas Kelvin y el relajamiento de los sistemas atmosféricos que debilitaron los vientos Alisios del sudeste, y que redujeron las áreas de afloramiento costero; (e) registro de dos pequeños núcleos con anomalías negativas.

El crucero se desarrolló en dos etapas, la primera en primavera 2010, de 4° a 11°25’S y la segunda en verano 2011, de 10°10’ a 17°23’S, entre 50 a 200 mn de la costa. La captura del recurso en la primera etapa fue 35.079,6 kg (25.669,1 kg de producción) y en la segunda 165.955,7 kg (123.229,5 kg de producción). La captura por unidad de esfuerzo por día de trabajo fluctuó de 2,9 a 4.292,6 kg/hora; 0,68 a 948,4 kg/línea; 0,07 a 99,8 kg/ línea*hora y 0,0017 a 2,4957 kg/pot*hora. La longitud del manto varió de 17 a 119 cm. Se registró hembras en estadio desovante III (51,5%), madurante II (21,6%); y machos en estadio evacuación III (85,1%) y virginales I (9,7%). Los grupos tróficos más importantes fueron: cefalópodos (% IRI= 66,4), crustáceos (% IRI= 23,7), peces (% IRI= 9,9). Las hembras presentaron: L∞ =111,233 cm LM, K=0,016, t0= 235 y los machos L∞ =99,718 cm LM, K=0,167 y t0=228,4; esta especie tiene una longevidad próxima o poco mayor a un año.

Coagulation factor V (FV) deficiency is characterised by variable bleeding phenotypes and heterogeneous mutations. To add new insights into the FV genotype-phenotype relationship, we characterised the R1698W change in the A3 domain, at the poorly investigated interface with the A2 domain. The FV R1698W mutation was responsible for a markedly reduced expression level (10% of FV-WT) and specific activity in thrombin generation (0.39). Interestingly, the FVa1698W showed rapid activity decay upon activation due to increased dissociation rate between the heavy and light chains. The importance of the size and charge of the residue at position 1698 was investigated by three additional recombinant mutants, FVR1698A, FVR1698Q, and FVR1698E. FVR1698A and FVR1698Q expression (30 and 45% of FV-WT), specific activity (both 0.57) and stability were all reduced. Noticeably, FVR1698E showed normal activity and stability despite poor expression (10% of FV-WT). These data indicate the essential role of R1698 for normal biosynthetic process and support local flexibility for positively or negatively charged residues to produce stable and functional A3-A2 domain interactions. Their experimental alteration produces a gradient of FV defects, which help to interpret the wide spectrum of phenotypes in FV-deficient patients.

BAFF deficiency in mice impairs B cell development beyond the transitional stage 1 in the spleen and thus severely reduces the size of follicular and marginal zone B cell compartments. Moreover, humoral immune responses in these mice are dramatically impaired. We now addressed the question whether the decrease in mature B cell numbers and the reduced humoral immune responses in BAFF-deficient mice could be overcome by the injection of recombinant BAFF. We therefore engineered a recombinant protein containing the human IgG1 Fc moiety fused to receptor-binding domain of human BAFF (Fc-BAFF). At 1 week after the second injection of this fusion protein a complete rescue of the marginal zone B cell compartment and a 50% rescue of the follicular B cell compartment was observed. Moreover these mice mounted a T cell-dependent humoral immune response indistinguishable from wild-type mice. By day 14 upon arrest of Fc-BAFF treatment mature B cell numbers in the blood dropped by 50%, indicating that the life span of mature B cells in the absence of BAFF is 14 days or less. Collectively these findings demonstrate that injection of Fc-BAFF in BAFF-deficient mice results in a temporary rescue of a functional mature B cell compartment.

High-molecular-weight (HMW) penicillin-binding proteins (PBPs) are divided into class A and class B PBPs, which are bifunctional transpeptidases/transglycosylases and monofunctional transpeptidases, respectively. We determined the sequences for the HMW PBP genes of Streptococcus gordonii, a gingivo-dental commensal related to Streptococcus pneumoniae. Five HMW PBPs were identified, including three class A (PBPs 1A, 1B, and 2A) and two class B (PBPs 2B and 2X) PBPs, by homology with those of S. pneumoniae and by radiolabeling with [3H]penicillin. Single and double deletions of each of them were achieved by allelic replacement. All could be deleted, except for PBP 2X, which was essential. Morphological alterations occurred after deletion of PBP 1A (lozenge shape), PBP 2A (separation defect and chaining), and PBP 2B (aberrant septation and premature lysis) but not PBP 1B. The muropeptide cross-link patterns remained similar in all strains, indicating that cross-linkage for one missing PBP could be replaced by others. However, PBP 1A mutants presented shorter glycan chains (by 30%) and a relative decrease (25%) in one monomer stem peptide. Growth rate and viability under aeration, hyperosmolarity, and penicillin exposure were affected primarily in PBP 2B-deleted mutants. In contrast, chain-forming PBP 2A-deleted mutants withstood better aeration, probably because they formed clusters that impaired oxygen diffusion. Double deletion could be generated with any PBP combination and resulted in more-altered mutants. Thus, single deletion of four of the five HMW genes had a detectable effect on the bacterial morphology and/or physiology, and only PBP 1B seemed redundant a priori.

BACKGROUND: By analyzing human immunodeficiency virus type 1 (HIV-1) pol sequences from the Swiss HIV Cohort Study (SHCS), we explored whether the prevalence of non-B subtypes reflects domestic transmission or migration patterns. METHODS: Swiss non-B sequences and sequences collected abroad were pooled to construct maximum likelihood trees, which were analyzed for Swiss-specific subepidemics, (subtrees including ≥80% Swiss sequences, bootstrap >70%; macroscale analysis) or evidence for domestic transmission (sequence pairs with genetic distance <1.5%, bootstrap ≥98%; microscale analysis). RESULTS: Of 8287 SHCS participants, 1732 (21%) were infected with non-B subtypes, of which A (n = 328), C (n = 272), CRF01_AE (n = 258), and CRF02_AG (n = 285) were studied further. The macroscale analysis revealed that 21% (A), 16% (C), 24% (CRF01_AE), and 28% (CRF02_AG) belonged to Swiss-specific subepidemics. The microscale analysis identified 26 possible transmission pairs: 3 (12%) including only homosexual Swiss men of white ethnicity; 3 (12%) including homosexual white men from Switzerland and partners from foreign countries; and 10 (38%) involving heterosexual white Swiss men and females of different nationality and predominantly nonwhite ethnicity. CONCLUSIONS: Of all non-B infections diagnosed in Switzerland, <25% could be prevented by domestic interventions. Awareness should be raised among immigrants and Swiss individuals with partners from high prevalence countries to contain the spread of non-B subtypes.

Cognitive efficacy of quetiapine in early-onset first-episode psychosis: a 12-week open label trial.

Twenty-three adolescents with psychotic disorders, aged from 13 to 18 years, participated in a 12-week open label trial (17 adolescents completed the study) in order to examine the impact of quetiapine on clinical status and cognitive functions (encompassing processing speed, attention, short-term memory, long-term memory and executive function). An improvement in Clinical Global Impression and Positive and Negative Symptom Scale (P's ≤ 0.001) was observed. In addition, after controlling for amelioration of symptoms, a significant improvement was observed on one executive function (P = 0.044; Trail Making Part B). The remaining cognitive abilities showed stability. In addition, we observed an interaction between quetiapine doses (>300 mg/day or <300 mg/day) and time, where lower doses showed more improvement in verbal short-term memory (P = 0.048), inhibition abilities (P = 0.038) and positive symptoms (P = 0.020). The neuropsychological functioning of adolescents with psychotic disorders remained mainly stable after 12 weeks of treatment with quetiapine. However, lower doses seemed to have a better impact on two components of cognition (inhibition abilities and verbal short-term memory) and on positive symptoms.

Na região de Lavras (MG), foram selecionados perfis de solos com horizonte B textural para a avaliação micropedológica dos processos de alteração e investigação de características de evolução pedológica vinculadas ao material de origem. Os solos com horizonte B textural foram selecionados para tal estudo, dado seu grau de evolução que permite avaliar características herdadas da rocha original. Foram escolhidas classes de solos com B textural, desenvolvidos em relevo ondulado a forte ondulado, a partir de materiais geológicos litoquimicamente distintos: Perfil 1 - Argissolo Vermelho-Amarelo distrófico, originado por alteração de rochas do domínio geológico Granito de Itutinga; Perfil 2 - Argissolo Vermelho eutrófico, desenvolvido a partir de rochas do domínio geológico Diorito do Rosário; e Perfil 3 - Chernossolo Argilúvico férrico, desenvolvido a partir de rochas do domínio geológico Greenstone-belt de Lavras. Ao longo dos perfis de alteração selecionados, realizaram-se análises petrográficas dos fácies rocha fresca e rocha alterada, bem como análises micromorfológicas dos fácies alterito, de transição e sólum. Para avaliar os processos de alteração, observaram-se as reações de alteração e feições texturais, com auxílio da difração de raios-X com vistas em identificar a mineralogia da fração argila, numa discussão envolvendo material de origem versus processos de alteração pedogenética, possibilitando estabelecer a ordem de destruição dos minerais primários. A evolução dos processos de alteração ao longo dos perfis mostrou-se diretamente vinculada à textura, composição química e mineralógica dos materiais originais, refletindo-se nas características micromorfológicas e composição mineralógica dos horizontes que compõem cada perfil, especialmente o horizonte diagnóstico - B textural.
