“Plasmon” is a synonym for collective oscillations of the conduction electrons in a metal nanoparticle (excited by an incoming light wave), which cause strong optical responses like efficient light scattering. The scattering cross-section with respect to the light wavelength depends not only on material, size and shape of the nanoparticle, but also on the refractive index of the embedding medium. For this reason, plasmonic nanoparticles are interesting candidates for sensing applications. Here, two novel setups for rapid spectral investigations of single nanoparticles and different sensing experiments are presented.rnrnPrecisely, the novel setups are based on an optical microscope operated in darkfield modus. For the fast single particle spectroscopy (fastSPS) setup, the entrance pinhole of a coupled spectrometer is replaced by a liquid crystal device (LCD) acting as spatially addressable electronic shutter. This improvement allows the automatic and continuous investigation of several particles in parallel for the first time. The second novel setup (RotPOL) usesrna rotating wedge-shaped polarizer and encodes the full polarization information of each particle within one image, which reveals the symmetry of the particles and their plasmon modes. Both setups are used to observe nanoparticle growth in situ on a single-particle level to extract quantitative data on nanoparticle growth.rnrnUsing the fastSPS setup, I investigate the membrane coating of gold nanorods in aqueous solution and show unequivocally the subsequent detection of protein binding to the membrane. This binding process leads to a spectral shift of the particles resonance due to the higher refractive index of the protein compared to water. Hence, the nanosized addressable sensor platform allows for local analysis of protein interactions with biological membranes as a function of the lateral composition of phase separated membranes.rnrnThe sensitivity on changes in the environmental refractive index depends on the particles’ aspect ratio. On the basis of simulations and experiments, I could present the existence of an optimal aspect ratio range between 3 and 4 for gold nanorods for sensing applications. A further sensitivity increase can only be reached by chemical modifications of the gold nanorods. This can be achieved by synthesizing an additional porous gold cage around the nanorods, resulting in a plasmon sensitivity raise of up to 50 % for those “nanorattles” compared to gold nanorods with the same resonance wavelength. Another possibility isrnto coat the gold nanorods with a thin silver shell. This reduces the single particle’s resonance spectral linewidth about 30 %, which enlarges the resolution of the observable shift. rnrnThis silver coating evokes the interesting effect of reducing the ensemble plasmon linewidth by changing the relation connecting particle shape and plasmon resonance wavelength. This change, I term plasmonic focusing, leads to less variation of resonance wavelengths for the same particle size distribution, which I show experimentally and theoretically.rnrnIn a system of two coupled nanoparticles, the plasmon modes of the transversal and longitudinal axis depend on the refractive index of the environmental solution, but only the latter one is influenced by the interparticle distance. I show that monitoring both modes provides a self-calibrating system, where interparticle distance variations and changes of the environmental refractive index can be determined with high precision.

The aim of my PhD research project was to investigate new and more sustainable routes, compared to those currently used, for the production of adipic acid (AA). AA is a very important chemical intermediate. The main use of AA is the production of Nylon-6,6 fibers, resins, polyesters, plasticizers. My project was divided into two parts: 1. The two-step oxidation of cyclohexene, where the latter is first oxidized into trans-1,2-cyclohexanediol (CHD) with aqueous hydrogen peroxide, and then the glycol is transformed into AA by reaction with molecular oxygen. Various catalysts were investigated in this process, both heterogeneous (alumina-supported Ru(OH)x and Au nanoparticles supported on TiO2, MgO and Mg(OH)2) and homogeneous (polyoxometalates). We also studied the mechanism of CHD oxidation with oxygen in the presence of these catalysts. 2. Baeyer-Villiger oxidation of cyclohexanone with aqueous hydrogen peroxide into ɛ-caprolactone, as a first step on the way to produce AA. Study on the mechanism of the uncatalyzed (thermal) oxidation of cyclohexanone were also carried out. Investigation on how the different heterogeneous catalysts affect the formation of the reaction products and their distribution was done.

Self-assembled molecular structures were investigated on insulating substrate surfaces using non-contact atomic force microscopy. Both, substrate preparation and molecule deposition, took place under ultra-high vacuum conditions. First, C60 molecules were investigated on the TiO2 (110) surface. This surface exhibits parallel running troughs at the nanometer scale, which strongly steer the assembly of the molecules. This is in contrast to the second investigated surface. The CaF2 (111) surface is atomically flat and the molecular assemblyrnwas observed to be far less affected by the surface. Basically different island structures were observed to what is typically know. Based on extensive experimental studies and theoretical considerations, a comprehensive picture of the processes responsible for the island formation of C60 molecules on this insulating surfaces was developed. The key process for the emergence of the observed novel island structures was made out to be the dewetting of molecules from the substrate. This new knowledge allows to further understand andrnexploit self-assembly techniques in structure fabrication on insulating substrate surfaces. To alter island formation and island structure, C60 molecules were codeposited with second molecule species (PTCDI and SubPc) on the CaF2 (111) surface. Depending on the order of deposition, quiet different structures were observed to arise. Thus, these are the first steps towards more complex functional arrangements consisting of two molecule species on insulating surfaces.

In the last decades noble metal nanoparticles (NPs) arose as one of the most powerful tools for applications in nanomedicine field and cancer treatment. Glioblastoma multiforme (GBM), in particular, is one of the most aggressive malignant brain tumors that nowadays still presents a dramatic scenario concerning median survival. Gold nanorods (GNRs) and silver nanoparticles (AgNPs) could find applications such as diagnostic imaging, hyperthermia and glioblastoma therapy. During these three years, both GNRs and AgNPs were synthesized with the “salt reduction” method and, through a novel double phase transfer process, using specifically designed thiol-based ligands, lipophilic GNRs and AgNPs were obtained and separately entrapped into biocompatible and biodegradable PEG-based polymeric nanoparticles (PNPs) suitable for drug delivery within the body. Moreover, a synergistic effect of AgNPs with the Alisertib drug, were investigated thanks to the simultaneous entrapment of these two moieties into PNPs. In addition, Chlorotoxin (Cltx), a peptide that specifically recognize brain cancer cells, was conjugated onto the external surface of PNPs. The so-obtained novel nanosystems were evaluated for in vitro and in vivo applications against glioblastoma multiforme. In particular, for GNRs-PNPs, their safety, their suitability as optoacoustic contrast agents, their selective laser-induced cells death and finally, a high tumor retention were all demonstrated. Concerning AgNPs-PNPs, promising tumor toxicity and a strong synergistic effect with Alisertib was observed (IC50 10 nM), as well as good in vivo biodistribution, high tumor uptake and significative tumor reduction in tumor bearing mice. Finally, the two nanostructures were linked together, through an organic framework, exploiting the click chemistry azido-alkyne Huisgen cycloaddition, between two ligands previously attached to the NPs surface; this multifunctional complex nanosystem was successfully entrapped into PNPs with nanoparticles’ properties maintenance, obtaining in this way a powerful and promising tool for cancer fight and defeat.

Polymer-nanoparticle hybrids show synergistic effects, demonstrating both, the unique properties of nanosized structures and the good processability and functionalities of polymeric materials. This work shows the synthesis and application of block copolymers containing a soluble, functional block and a short anchor block, which efficiently binds to the surface of nanocrystals. We functionalized anisotropic, semiconducting nanoparticles, which can be dissolved in organic and polymeric matrices upon modification. The modified nanorods have the ability to form liquid crystalline phases, which behave similar to low molecular liquid crystals with a reversible clearing behaviour. These liquid crystalline phases could also be obtained in hole conducting matrices. For a macroscopic orientation of the nanorods, electric fields were applied and a switching (in analogy to known liquid crystals) to a homeotropic orientation was observed.rnBy introduction of dye molecules in the anchor block of a hole conducting block copolymer, all essential components of a solar cell can be combined in a single particle. Light absorption of the dye induces the injection of electrons into the particles, followed by a charging, that was monitored by a special AFM technique.rnLight emitting nanocrystals were functionalized analogously with a hole transporting polymer. The stability of the particles could be enhanced by the sterically stabilizing polymer corona and the particles showed improved properties in terms of processing. We applied these hybrid materials in light emitting devices, which showed better characteristics due to an improved hole injection and well dispersed emitting particles in the active device layer.rnThe work shows the broad spectrum of properties and applications based on the synergistic effects in hybrid and composite materials.

Plasmonen sind die kollektive resonante Anregung von Leitungselektronen. Vom Licht angeregternPlasmonen in subwellenlängen-grossen Nanopartikeln heissen Partikelplasmonen und sind vielversprechende Kandidaten für zukünftige Mikrosensoren wegen der starken Abhängigkeit der Resonanz an extern steuerbaren Parametern, wie die optischen Eigenschaften des umgebenden Mediums und die elektrische Ladung der Nanopartikel. Die extrem hohe Streue_zienz von Partikelplasmonen erlaubt eine einfache Beobachtung einzelner Nanopartikel in einem Mikroskop.rnDie Anforderung, schnell eine statistisch relevante Anzahl von Datenpunkten sammeln zu können,rnund die wachsende Bedeutung von plasmonischen (vor allem Gold-) Nanopartikeln für Anwendungenrnin der Medizin, hat nach der Entwicklung von automatisierten Mikroskopen gedrängt, die im bis dahin nur teilweise abgedeckten spektralen Fenster der biologischen Gewebe (biologisches Fenster) von 650 bis 900nm messen können. Ich stelle in dieser Arbeit das Plasmoscope vor, das genau unter Beobachtung der genannten Anforderungen entworfen wurde, in dem (1) ein einstellbarer Spalt in die Eingangsö_nung des Spektrometers, die mit der Bildebene des Mikroskops zusammenfällt, gesetzt wurde, und (2) einem Piezo Scantisch, der es ermöglicht, die Probe durch diesen schmalen Spalt abzurastern. Diese Verwirklichung vermeidet optische Elemente, die im nahen Infra-Rot absorbieren.rnMit dem Plasmoscope untersuche ich die plasmonische Sensitivität von Gold- und Silbernanostrnäbchen, d.h. die Plasmon-Resonanzverschiebung in Abhängigkeit mit der Änderung des umgebendenrnMediums. Die Sensitivität ist das Mass dafür, wie gut die Nanopartikeln Materialänderungenrnin ihrer Umgebung detektieren können, und damit ist es immens wichtig zu wissen, welche Parameterrndie Sensitivität beein_ussen. Ich zeige hier, dass Silbernanostäbchen eine höhere Sensitivität alsrnGoldnanostäbchen innerhalb des biologischen Fensters besitzen, und darüberhinaus, dass die Sensitivität mit der Dicke der Stäbchen wächst. Ich stelle eine theoretische Diskussion der Sensitivitätrnvor, indenti_ziere die Materialparameter, die die Sensitivität bein_ussen und leite die entsprechendenrnFormeln her. In einer weiteren Annäherung präsentiere ich experimentelle Daten, die die theoretische Erkenntnis unterstützen, dass für Sensitivitätsmessschemata, die auch die Linienbreite mitberücksichtigen, Goldnanostäbchen mit einem Aspektverhältnis von 3 bis 4 das optimalste Ergebnis liefern. Verlässliche Sensoren müssen eine robuste Wiederholbarkeit aufweisen, die ich mit Gold- und Silbernanostäbchen untersuche.rnDie Plasmonen-resonanzwellenlänge hängt von folgenden intrinsischen Materialparametern ab:rnElektrondichte, Hintergrundpolarisierbarkeit und Relaxationszeit. Basierend auf meinen experimentellen Ergebnissen zeige ich, dass Nanostäbchen aus Kupfer-Gold-Legierung im Vergleich zu ähnlich geformten Goldnanostäbchen eine rotverschobene Resonanz haben, und in welcher Weiserndie Linienbreite mit der stochimetrischen Zusammensetzung der legierten Nanopartikeln variiert.rnDie Abhängigkeit der Linienbreite von der Materialzusammensetzung wird auch anhand von silberbeschichteten und unbeschichteten Goldnanostäbchen untersucht.rnHalbleiternanopartikeln sind Kandidaten für e_ziente photovoltaische Einrichtungen. Die Energieumwandlung erfordert eine Ladungstrennung, die mit dem Plasmoscope experimentell vermessen wird, in dem ich die lichtinduzierte Wachstumsdynamik von Goldsphären auf Halbleiternanost äbchen in einer Goldionenlösung durch die Messung der gestreuten Intensität verfolge.rn

Nanotechnology promises huge benefits for society and capital invested in this new technology is steadily increasing, therefore there is a growing number of nanotechnology products on the market and inevitably engineered nanomaterials will be released in the atmosphere with potential risks to humans and environment. This study set out to extend the comprehension of the impact of metal (Ag, Co, Ni) and metal oxide (CeO2, Fe3O4, SnO2, TiO2) nanoparticles (NPs) on one of the most important environmental compartments potentially contaminated by NPs, the soil system, through the use of chemical and biological tools. For this purpose experiments were carried out to simulate realistic environmental conditions of wet and dry deposition of NPs, considering ecologically relevant endpoints. In detail, this thesis involved the study of three model systems and the evaluation of related issues: (i) NPs and bare soil, to assess the influence of NPs on the functions of soil microbial communities; (ii) NPs and plants, to evaluate the chronic toxicity and accumulation of NPs in edible tissues; (iii) NPs and invertebrates, to verify the effects of NPs on earthworms and the damaging of their functionality. The study highlighted that NP toxicity is generally influenced by NP core elements and the impact of NPs on organisms is specie-specific; moreover experiments conducted in media closer to real conditions showed a decrease in toxicity with respect to in vitro test or hydroponic tests. However, only a multidisciplinary approach, involving physical, chemical and biological skills, together with the use of advanced techniques, such as X-ray absorption fine structure spectroscopy, could pave the way to draw the right conclusions and accomplish a deeper comprehension of the effects of NPs on soil and soil inhabitants.

In this thesis, anodic aluminum oxide (AAO) membranes, which provide well-aligned uniform mesoscopic pores with adjustable pore parameters, were fabricated and successfully utilized as templates for the fabrication of functional organic nanowires, nanorods and the respective well-ordered arrays. The template-assisted patterning technique was successfully applied for the realization of different objectives:rnHigh-density and well-ordered arrays of hole-conducting nanorods composed of cross-linked triphenylamine (TPA) and tetraphenylbenzidine (TPD) derivatives on conductive substrates like ITO/glass have been successfully fabricated. By applying a freeze-drying technique to remove the aqueous medium after the wet-chemical etching of the template, aggregation and collapsing of the rods was prevented and macroscopic areas of perfectly freestanding nanorods were feasible. Based on the hole-conducting nanorod arrays and their subsequent embedding into an electron-conducting polymer matrix via spin-coating, a novel routine concept for the fabrication of well-ordered all-organic bulk heterojunction for organic photovoltaic applications was successfully demonstrated. The increased donor/acceptor interface of the fabricated devices resulted in a remarkable increase of the photoluminescence quenching compared to a planar bilayer morphology. Further, the fundamental working principle of the templating approach for the solution-based all-organic photovoltaic device was demonstrated for the first time.rnFurthermore, in order to broaden the applicability of patterned surfaces, which are feasible via the template-based patterning of functional materials, AAO with hierarchically branched pores were fabricated and utilized as templates. By pursuing the common templating process hierarchically polymeric replicas, which show remarkable similarities with interesting biostructures, like the surface of the lotus leaf and the feet of a gecko, were successfully prepared.rnIn contrast to the direct infiltration of organic functional materials, a novel route for the fabrication of functional nanowires via post-modification of reactive nanowires was established. Therefore, reactive nanowires based on cross-linked pentafluorophenylesters were fabricated by utilizing AAO templates. The post-modification with fluorescent dyes was demonstrated. Furthermore, reactive wires were converted into well-dispersed poly(N-isopropylacrylamide) (PNIPAM) hydrogels, which exhibit a thermal-responsive reversible phase transition. The reversible thermal-responsible swelling of the PNIPAM nanowires exhibited a more than 50 % extended length than in the collapsed PNIPAM state. rnLast but not least, the shape-anisotropic pores of AAO were utilized to uniformly align the mesogens of a nematic liquid crystalline elastomer. Liquid crystalline nanowires with a narrow size distribution and uniform orientation of the liquid crystalline material were fabricated. It was shown that during the transition from the nematic to the isotropic phase the rod’s length shortened by roughly 40 percent. As such these liquid crystalline elastomeric nanowires may find application, as wire-shaped nanoactuators in various fields of research, like lab-on-chip systems, micro fluidics and biomimetics.rn

Im Rahmen dieser Arbeit wurden amphiphile Co- und Terpolymere verwendet, um die Grenzflächeneigenschaften anorganischer Nanopartikel zu kontrollieren. Es wurde eine effiziente und vielseitige Methode entwickelt, mit der in-situ hydrophobierte, formanisotrope ZnO-, CdS- und Au-Nanopartikel sowie poröse TiO2-Nanopartikel hergestellt werden konnten. Diese Technik basierte auf der Fällung anorganischer Nanopartikel in einer inversen Emulsion mittels kombinierten Einsatzes zweier maßgeschneiderter amphiphiler Polymere. Ein Copolymer ermöglichte sowohl die Stabilisierung der Emulsion als auch die Hydrophobierung der Partikel, und ein weiteres Struktur-dirigierendes Agens (SDA) kontrollierte den Kristallisationsprozess. Infolge ihrer Form zeigten die Nanopartikel von sphärischen Teilchen abweichende Lagen der Oberflächenplasmonenresonanz und der Bandlücke. Aufgrund der hervorragenden Hydrophobierung dieser Kolloide mittels amphiphiler Copolymere konnten diese homogen in polymere Materialien eingearbeitet werden. Dies erlaubte es die speziellen Eigenschaften von nicht-sphärischen Kolloiden auf Nanokompositmaterialien zu übertragen. Darüber hinaus wurden amphipolare Copolymere genutzt, um superhydrophobe Oberflächen zu generieren. Hierzu wurden Filme bestehend aus rauen SiO2-Nanopartikeln mit fluorierten Emulgatoren beschichtet. In einem dritten Schwerpunkt dieser Arbeit wurden amphiphile Co- und Terpolymere verwendet, um anorganische Nanopartikel zu hydrophilieren. Durch Variation der Emulgatorzusammensetzung konnten die Ladung und Ladungsdichte auf der Partikeloberfläche gezielt gesteuert werden. Darüber hinaus konnte die Partikelhülle zusätzlich mit Farbstoffmolekülen funktionalisiert werden, was den erfolgreichen Einsatz der Kolloide in Zellaufnahmeexperimenten ermöglichte.

Das Silicatein α ist ein 24 kDa großes Enzym, welches im Schwamm Suberites domuncula für die Synthese von Biosilikat verantwortlich ist. Vorhergehende Studien haben gezeigt, dass Silicatein auch die Synthese anderer Metalloxide wie Titandioxid, Galliumoxid und Zirkoniumdioxid katalysieren kann. Diese Fähigkeiten machen das Silicatein α für biomedizinische und biotechnologische Anwendungen interessant, da die Synthese unter nahezu physiologischen Bedingungen ablaufen kann, was die Herstellung neuartiger Kompositmaterialien mit einzigartigen Eigenschaften erleichtern würde. Zur Immobilisierung des Silicatein α auf verschiedenen Oberflächen wurde bislang ein Nickel-NTA-Kopolymer eingesetzt. Diese Art der Immobilisierung bietet eine Reihe von Möglichkeiten in der Nanobiotechnologie, stößt aber in der Biomedizin an ihre Grenzen, da sich nicht alle Oberflächen für ein solches Coating eignen. Zudem können die zur Aktivierung des Polymers nötigen Lösungsmittel und die über die Zeit freigesetzten Monomere aus dem Polymergerüst toxische oder mutagene Wirkung auf das umliegende Gewebe haben. Deshalb wurde das Silicatein α in dieser Arbeit mit zwei Affinitäts-Tags so modifiziert, dass es an verschiedene Oberflächen immobilisiert werden kann und dabei seine Aktivität beibehält. Zuerst wurde das Silicatein mit einem Glu-tag am N-terminalen Ende modifiziert. Dadurch gelang die direkte Immobilisierung an Hydroxyapatit und die folgende, enzymkatalysierte Synthese von Biosilikat-Beschichtungen auf diesem Träger. Die Eigenschaften eines solchen HA-Kompositmaterials können zum Beispiel zu einem verbesserten, schnelleren und stabileren Einwachsen von Knochenimplantaten führen, da Biosilikat die Reifung und Differenzierung von Osteoblasten beschleunigt. rnMit dem an Hydroxyapatit-Plättchen immobilisierten Glu-tag-Silicatein wurde ein modifizierter Pull-down Assay etabliert, wodurch bekannte, aber auch bis dahin noch unbekannte Protein-Interaktionspartner identifiziert werden konnten. rnUm zu zeigen, dass der entwickelte Glu-tag an präformierte, calciumhaltige Oberflächen binden kann, wurden die Nadeln des Kalkschwammes Paraleucilla magna als Modellorganismus verwendet. Die Nadeln konnten durch das immobilisierte Silicatein mit einer Titandioxid-Schicht überzogen werden und unter Verwendung des Interaktionspartners Silintaphin-1 konnte diese Beschichtung noch verstärkt werden. Solche CaCO3-Kompositmaterialien könnten sowohl in der Biomedizin als auch in der Biotechnologie zum Einsatz kommen. Neben den erwähnten calciumhaltigen Materialien finden auch andere Stoffe wie TiO2-Nanodrähte Verwendung in der Forschung. In weiterführenden Experimenten konnte gezeigt werden, dass der entwickelte Glu-tag auch Affinität zu Titandioxid-Oberflächen vermittelt. Auch hier konnte durch das oberflächenimmobilisierte Enzym eine Biosilikatbeschichtung synthetisiert werden. rnMit der zweiten Modifikation - einem Cys-tag - konnte Silicatein direkt auf Goldoberflächen immobilisiert werden. Durch die Verwendung eines Polydimethylsiloxan (PDMS)-Stempels wurde das Cys-getaggte Silicatein in einem linienförmigen Muster auf das Gold übertragen und die Synthese von Titandioxid dort nachgewiesen.rnDie Experimente und Ergebnisse dieser Arbeit haben gezeigt, dass Silicatein α durch einfache Modifikationen an verschiedene Oberflächen immobilisiert werden kann und dabei immer noch seine Aktivität behält. rnHierdurch ergibt sich die Möglichkeit, unter Normalbedingungen verschiedenste Kompositmaterialien herzustellen.rn

This work deals with the oxidation of 5-hydroxymethylfurfural (HMF) to 2,5-furandicarboxylic acid (FDCA) using metal supported catalysts. Catalysts were prepared from the immobilisation of preformed monometallic (Au, Pd) and bimetallic (AuCu, AuPd) nanoparticles on commercial oxides (TiO2, CeO2). Au-TiO2 catalyst was found to be very active for HMF oxidation; however, this system deactivated very fast. For this reason, we prepared bimetallic gold-copper nanoparticles and an increase in the catalytic activity was observed together with an increase in catalyst stability. In order to optimise the interaction of the metal active phase with the support, Au and AuCu nanoparticles were supported onto CeO2. Au-CeO2 catalyst was found to be more active than the bimetallic one, leading to the conclusion that in this case the most important feature is the interaction between gold and the support. Catalyst pre-treatments (calcination and washing) were carried out to maximise the contact between the metal and the oxide and an increase in the FDCA production could be observed. The presence of ceria defective sites was crucial for FDCA formation. Mesoporous cerium oxide was synthesised with the hard template method and was used as support for Au nanoparticles to promote the catalytic activity. In order to study the role of active phase in HMF oxidation, PdAu nanoparticles were supported onto TiO2. Au and Pd monometallic catalysts were very active in the formation of HMFCA (5-hydroxymethyl-2-furan carboxylic acid), but Pd was not able to convert it, leading to a low FDCA yield. The calcination of PdAu catalysts led to Pd segregation on the particles surface, which changed the reaction pathway and included an important contribution of the Cannizzaro reaction. PVP protected PdAu nanoparticles, synthesised with different morphologies (core-shell and alloyed structure), confirmed the presence of a different reaction mechanism when the metal surface composition changes.

This PhD thesis focused on nanomaterial (NM) engineering for occupational health and safety, in the frame of the EU project “Safe Nano Worker Exposure Scenarios (SANOWORK)”. Following a safety by design approach, surface engineering (surface coating, purification process, colloidal force control, wet milling, film coating deposition and granulation) were proposed as risk remediation strategies (RRS) to decrease toxicity and emission potential of NMs within real processing lines. In the first case investigated, the PlasmaChem ZrO2 manufacturing, the colloidal force control applied to the washing of synthesis rector, allowed to reduce ZrO2 contamination in wastewater, performing an efficient recycling procedure of ZrO2 recovered. Furthermore, ZrO2 NM was investigated in the ceramic process owned by CNR-ISTEC and GEA-Niro; the spray drying and freeze drying techniques were employed decreasing NM emissivity, but maintaining a reactive surface in dried NM. Considering the handling operation of nanofibers (NFs) obtained through Elmarco electrospinning procedure, the film coating deposition was applied on polyamide non-woven to avoid free fiber release. For TiO2 NF the wet milling was applied to reduce and homogenize the aspect ratio, leading to a significant mitigation of fiber toxicity. In the Colorobbia spray coating line, Ag and TiO2 nanosols, employed to transfer respectively antibacterial or depolluting properties to different substrates, were investigated. Ag was subjected to surface coating and purification, decreasing NM toxicity. TiO2 was modified by surface coating, spray drying and blending with colloidal SiO2, improving its technological performance. In the extrusion of polymeric matrix charged with carbon nanotube (CNTs) owned by Leitat, the CNTs used as filler were granulated by spray drying and freeze spray drying techniques, allowing to reduce their exposure potential. Engineered NMs tested by biologists were further investigated in relevant biological conditions, to improve the knowledge of structure/toxicity mechanisms and obtain new insights for the design of safest NMs.

Der light harvesting complex II (LHCII) ist ein pflanzliches Membranprotein, das in seiner trimeren Form über 40 Chlorophylle bindet. In der Pflanze kann er besonders effizient Licht sammeln und die Anregungsenergie anschließend fast verlustfrei über andere chlorophyll-bindende Proteine an die Reaktionszentren weiterleiten. Aufgrund dieser besonderen Eigenschaften war es ein Ziel dieser Arbeit, rekombinanten LHCII mit synthetischen Komponenten zu kombinieren, die zur Ladungstrennung befähigt sind. Zu diesem Zweck wurden unter anderem Halbleiternanokristalle (Quantum Dots, QDs) ausgewählt, die je nach Zusammensetzung sowohl als Energieakzeptoren als auch als Energiedonoren in Frage kamen. Durch Optimierung des Puffers gelang es, die Fluoreszenzquantenausbeute der QDs in wässriger Lösung zu erhöhen und zu stabilisieren, so dass die Grundvoraussetzungen für die spektroskopische Untersuchung verschiedener LHCII-QD-Hybridkomplexe erfüllt waren.rnUnter Verwendung bereits etablierter Affinitätssequenzen zur Bindung des LHCII an die QDs konnte gezeigt werden, dass die in dieser Arbeit verwendeten Typ-I QDs aus CdSe und ZnS sich kaum als Energie-Donoren für den LHCII eignen. Ein Hauptgrund lag im vergleichsweise kleinen Försterradius R0 von 4,1 nm. Im Gegensatz dazu wurde ein R0 von 6,4 nm für den LHCII als Donor und Typ-II QDs aus CdTe, CdSe und ZnS als Akzeptor errechnet, wodurch in diesem System eine höhere Effizienz des Energietransfers zu erwarten war. Fluoreszenzspektroskopische Untersuchungen von Hybridkomplexen aus LHCII und Typ-II QDs ergaben eine hohe Plausibilität für einen Fluoreszenz Resonanz Energietransfer (FRET) vom Lichtsammler auf die QDs. Weitere QD-Affinitätssequenzen für den LHCII wurden identifiziert und deren Bindekonstanten ermittelt. Versuche mit dem Elektronenakzeptor Methylviologen lieferten gute Hinweise auf eine LHCII-sensibilisierte Ladungstrennung der Typ-II QDs, auch wenn dies noch anhand alternativer Messmethoden wie z.B. durch transiente Absorptionsspektroskopie bestätigt werden muss. rnEin weiteres Ziel war die Verwendung von LHCII als Lichtsammler in dye-sensitized solar cells (DSSC). Geeignete dotierte TiO2-Platten wurden ermittelt, das Verfahren zur Belegung der Platten optimiert und daher mit wenig Aufwand eine hohe LHCII-Belegungsdichte erzielt. Erste Messungen von Aktionsspektren mit LHCII und einem zur Ladungstrennung fähigen Rylenfarbstoff zeigen eine, wenn auch geringe, LHCII sensibilisierte Ladungstrennung. rnDie Verwendung von Lanthanide-Binding-Tags (LBTs) ist ein potentielles Verfahren zur in vivo-Markierung von Proteinen mit Lanthanoiden wie Europium und Terbium. Diese Metalle besitzen eine überdurchschnittlich lange Lumineszenzlebensdauer, so dass sie leicht von anderen fluoreszierenden Molekülen unterschieden werden können. Im Rahmen der vorliegenden Arbeit gelang es, eine LBT in rekombinanten LHCII einzubauen und einen Lumineszenz Resonanz Energietransfer (LRET) vom Europium auf den LHCII nachzuweisen.rn

In this thesis cholesteric films made of liquid crystalline cellulose derivatives with improved optical properties were prepared. The choice of the solvent, hydrogen bond influencing additives, the synthetic realization of a very high degree of substitution on the cellulosic polymer and the use of mechanical stirring at the upper concentration limit of the liquid crystalline range were the basis for an improved alignment of the applied cellulose tricarbamates. In combination with a tuned substrate treatment and film preparation method, cholesteric films were obtained, with optical properties that were theoretically predicted and only known from low molecular weight liquid crystals so far. Subsequent polymerization allowed a permanent fixing of the alignment and the fabrication of free standing and insensitive films.rnThe incorporation of inorganic nanorods into the cholesteric host material was mediated with tailored block copolymers, available via controlled radical polymerization methods. In addition to the shape match between the rodlike mesogens of the host and the nanorods it was possible to increase the miscibility of both materials. Nevertheless, the size of the nanorods, in comparison to the mesogens, in these densely packed liquid crystalline phases as well as their long equilibration times were the reasons for phase separation. Nanorods are, in principle, valuable substitutes for organics, but their utilization in cellulosic CLC was not to be combined with a high quality alignment of the cholesteric structure.rnA swelling process of polymerized films in a dye solution or dissolving dyes in non-polymerized CLC was used for incorporation of the organic chromophores. With the first method the CLC could be aligned and polymerized without any disturbance due to dye molecules. The optical properties of dye and CLC were matched, with regard to mirrorless lasing devices. The dye was optically excited and laser emission supported by the cholesteric cavity was obtained. The polarization and wavelength of the emitted radiation as well as its bandwidth, the obtained interference pattern and threshold behavior of the emission proofed the feedback mechanism that was not believed to be realizable in liquid crystalline polymers. rnUtilization of a microfluidic co-flow injection device enabled us to transfer the properties of cellulosic CLC from the planar film shape to spherical micrometer sized particles. The pure material yielded particles with distorted mesogen alignment similar to films prepared by capillary flow. Dilution of the CLC with a solvent that migrated into the carrier phase during particle preparation provided the basis for particles with well ordered areas. rnAlthough cellulose derivatives were known for their liquid crystalline behavior for decades and synthesized in mass production, their application as feedback material was affected by bad optical properties. In comparison to low molar mass compounds, the low degree of order in the CLC phase was the cause. With the improved material, defined lasing emission was shown and characterized. Derivatives of cellulose are desirable materials, because, as a renewable resource, they are available in large amounts for a low price and need only simple derivatization reactions. The fabrication of CLC films with tunable lasing emission, for which this thesis can provide a starting point, is in good agreement with today's requirements of modern technology and its miniaturization.rn

This work is focused on the development of high quality nanoporous 1D photonic crystals –so called Bragg stacks – made by spin-coating of approximately 25 nm large SiO2 and TiO2 nanoparticles bearing interparticle voids large enough to infiltrate reactive species. Therefore, the first part of this work describes the synthesis of well-dispersed TiO2 nanoparticles in this size range (the corresponding SiO2 nanoparticles are commercially available). In the second part, a protocol was developed to prepare nanoporous Bragg stacks of up to 12 bilayers with high quality and precision. Tailor-made Bragg stacks were prepared for different applications such as (i) a surface emitting feedback laser with a FWHM of only 6 nm and (ii) an electrochromic device with absorption reversibly switchable by an external electrical bias independently of the Bragg reflection. In the last chapter, the approach to 1D photonic crystals is transferred to 1D phononic crystals. Contrast in the modulus is achieved by spin-coating SiO2 and PMMA as high and low moduli material. This system showed a band gap of fg = 12.6 GHz with a width of Dfg/fg = 4.5 GHz.
