Esta dissertação tem como principal objectivo comparar os métodos propostos pelo Eurocódigo 2, através de uma análise paramétrica dos efeitos de segunda ordem em pilares de betão armado. Os métodos estudados foram o método simplificado baseado na Curvatura Nominal e o Método Geral. Através da comparação destes dois métodos verificou-se se o dimensionamento efectuado para o primeiro método é correcto e se encontra do lado da segurança. Para este efeito são analisado noventa e seis pórticos, onde setenta e dois são pórticos simples e vinte e quatro são pórticos parede. Todos os pórticos apresentam uma estrutura simples, constituída por dois elementos verticais, pilar ou parede, e um elemento horizontal, viga. Os noventa e seis pórticos são divididos em dois grupos. Um grupo em que a ligação entre pilar viga é rotulada e o outro grupo onde a ligação é rígida. Dentro de cada grupo é definido oito conjuntos de pórticos cuja diferença reside na percentagem de carga que cada um dos elementos verticais recebe, sendo estas características iguais para os dois grupos. Cada conjunto é constituído por seis pórticos em que é variada a esbelteza. Em todos os modelos analisados manteve-se as mesmas características de materiais e acções. Efectuou-se um primeiro dimensionamento através do método baseado na curvatura nominal, onde obteve-se os esforços e armaduras, que serão utilizados para a modelação no método geral. Este método geral, consiste numa análise não linear, e é efectuado num programa de análise não linear de elementos finitos, Atena. A modelação efectuou-se da seguinte forma: aplicou-se numa primeira fase a totalidade da carga vertical aplicada à estrutura de forma incremental, e quando esta atingiu a carga de dimensionamento, aplicou-se a carga horizontal também de forma incremental até ao colapso da estrutura. De acordo com os resultados obtidos, pode concluir-se que o dimensionamento efectuado pelo método simplificado está do lado da segurança, sendo em alguns casos em que possui pilares esbeltos demasiado conservativo, onde obteve-se mais 200% de carga horizontal.

Conhecer a natureza e as características geomorfológicas e geológicas, que potenciam situações de instabilidade, é um passo importante a fim de determinar as medidas de prevenção e de estabilização, que se podem adaptar a um determinado local. Várias situações de instabilidade, que não se compreendem numa observação expedita e que ocorrem nos taludes do Funchal, despertaram o interesse de realizar um trabalho profundo, centrado na avaliação dos processos de instabilidade do talude. Na caracterização dos maciços rochosos para obtenção do índice RMR (Bieniawsky), fez-se um levantamento das descontinuidades com caracterização espacial do preenchimento e rugosidade. A resistência dos maciços rochosos e avaliação da resistência à compressão uniaxial dos taludes foi conseguido através de vários ensaios in situ, utilizando o martelo de Schmidt. Identificaram-se os possíveis factores que podem estar na base da instabilidade do talude. Dos resultados que se obtiveram, destacam-se a geometria do talude, as propriedades geomecânicas, a distribuição das descontinuidades, a influência da vegetação e da água. Os resultados permitiram propor medidas de prevenção e estabilização do talude, conforme os resultados da classificação geomecânica SMR (Slope Mass Rating) de Romana. Por fim, estabeleceu-se a comparação dos diferentes resultados obtidos com resultados publicados, sobre o mesmo tipo de material para a Ilha da Madeira.

O presente trabalho pretende contribuir para o aprofundamento do conhecimento geológico e geotécnico das formações vulcânicas da Ilha da Madeira, com especial destaque para os parâmetros geomecânicos obtidos através de retroanálise, tendo por base as leituras em obra do Túnel do Cortado (Faial, Santana; Madeira). Assim, o trabalho incide inicialmente sobre os túneis rodoviários na Ilha, com principal atenção nas características das suas secções transversais e emboquilhamentos. Segue-se um levantamento de todos os túneis construídos entre os anos de 1990 e 2013. Uma vez terminada esta apresentação, é feita uma caracterização geológica-geotécnica dos maciços da região, com especial atenção para as zonas geotécnicas afetas ao túnel em questão. São ainda abordadas metodologias de classificações geomecânicas, de retroanálise e modelação computacional. Segue-se um processo de retroanálise através do qual foram gerados vários modelos por forma a efetuar uma análise bidimensional das secções do túnel, onde se procurou estabelecer uma relação entre as leituras de obra e os parâmetros de resistência e deformabilidade do maciço em questão. Por fim, os resultados referentes aos parâmetros de resistência obtidos através dos modelos são analisados, procedendo-se à sua comparação com outros estudos existentes para a Ilha.

Pretende-se determinar os parâmetros geomecânicos de um maciço atravessado por um túnel construído na ilha da Madeira, através de retroanálise. Para tal, recorre-se a um modelo numérico tridimensional para simular o avanço da escavação e a dados obtidos a partir da instrumentação da obra, nomeadamente, através da medição de convergências e deslocamentos superficiais. A análise numérica é aplicada a um problema de identificação de parâmetros geomecânicos, em três secções localizadas muito próximas do emboquilhamento do Faial, do túnel do Faial/Cortado. Este problema rege-se por um comportamento elástico perfeitamente plástico e um critério de rotura de Mohr-Coulomb. Para a resolução do problema recorreu-se à utilização do software de diferenças finitas, FLAC3D, tendo-se usufruído da sua base de programação para modelar o processo complexo de escavação e suporte do túnel. A modelação tridimensional no FLAC3D é conseguida através da sua linguagem de programação interna, FISH, criando funções capazes de simular o processo da escavação parcial, instalação dos elementos de suporte, e monitorizar e extrair as variáveis pretendidas. Numa primeira fase realizou-se uma análise paramétrica relativamente ao comportamento do maciço fazendo variar alguns parâmetros geomecânicos deste. Numa segunda fase do estudo, procedeu-se à otimização de um conjunto de parâmetros elásticos e resistentes, intrínsecos ao material envolvente do túnel, através de retroanálise. A otimização dos parâmetros foi realizada por meio da utilização de expressões de cálculo de medida de dispersão estatística entre os dados obtidos a partir da instrumentação em obra e os resultados correspondentes obtidos a partir da análise numérica. Os valores obtidos para os diferentes parâmetros do maciço rochoso foram comparados com os respetivos valores recolhidos na bibliografia para maciços da ilha da Madeira, bem como com os valores previstos pela caracterização geomecânica efetuada durante a fase de construção do túnel objeto de estudo.

OBJETIVO: Neste trabalho avaliou-se a ação da glucana ? ? 1-3 insolúvel, um polissacarídeo extraído da parede celular interna do fungo Saccharomyces cerevisae, como agente imunoestimulante inespecífico em camundongos submetidos a modelo de sepse experimental. MÉTODOS: Foram utilizados 73 camundongos Swiss, os quais receberam glucana em diferentes doses pelas vias intraperitoneal (i.p.). Sepse difusa foi induzida através da técnica de ligadura e punção do ceco, transfixado e drenado com fio multifilamentar. RESULTADOS: Os animais tratados com glucana apresentaram um aumento significante no número de leucócitos no lavado peritoneal, diminuição no número de unidades formadoras de colônias bacterianas. Observou-se um aumento significante na sobrevida dos animais tratados com glucana insolúvel, a qual proporcionou um maior controle da infecção bacteriana por aumentar o número de células de defesa. CONCLUSÃO: A glucana insolúvel, quando usada em camundongos por via intraperitoneal, em modelo de sepse abdominal, contribuiu para melhorar a sobrevida, induziu proteção contra a formação de colônias bacterianas no líquido peritoneal e aumentou a migração leucocitária

This master thesis aims to assess the influence of the design decisions on the energy building performance of hotels. The research is based on the integration of field study and computer simulation. Firstly, a detailed field study is carried out to identify the characteristics of hotels in Natal, Rio Grande do Norte. The items assessed are occupancies, light and equipment densities, types of air conditioning, total and monthly energy consumption, among others. A second and more comprehensive field study is carried out to identify the range of occurrence of architectural variables, with a larger number of buildings. A base case is modelled in VisualDOE, based on the first field study. Then, a first set of simulations are run to explore the sensitivity of the variables on the energy consumption. The results analyses were the base of a second set of simulations, which combined the most influential variables. The results of 384 models were assessed, and the impacts of design decisions were quantified. The study discusses tendencies and recommendations, as well as the methods advantages and disadvantages

Chitin is an important structural component of the cellular wall of fungi and exoskeleton of many invertebrate plagues, such as insects and nematodes. In digestory systems of insects it forms a named matrix of peritrophic membrane. One of the most studied interaction models protein-carbohydrate is the model that involves chitin-binding proteins. Among the involved characterized domains already in this interaction if they detach the hevein domain (HD), from of Hevea brasiliensis (Rubber tree), the R&R consensus domain (R&R), found in cuticular proteins of insects, and the motif called in this study as conglicinin motif (CD), found in the cristallography structure of the β-conglicinin bounded with GlcNac. These three chitin-binding domains had been used to determine which of them could be involved in silico in the interaction of Canavalia ensiformis and Vigna unguiculata vicilins with chitin, as well as associate these results with the WD50 of these vicilins for Callosobruchus maculatus larvae. The technique of comparative modeling was used for construction of the model 3D of the vicilin of V. unguiculata, that was not found in the data bases. Using the ClustalW program it was gotten localization of these domains in the vicilins primary structure. The domains R&R and CD had been found with bigger homology in the vicilins primary sequences and had been target of interaction studies. Through program GRAMM models of interaction ( dockings ) of the vicilins with GlcNac had been gotten. The results had shown that, through analysis in silico, HD is not part of the vicilins structures, proving the result gotten with the alignment of the primary sequences; the R&R domain, although not to have structural similarity in the vicilins, probably it has a participation in the activity of interaction of these with GlcNac; whereas the CD domain participates directly in the interaction of the vicilins with GlcNac. These results in silico show that the amino acid number, the types and the amount of binding made for the CD motif with GlcNac seem to be directly associates to the deleterious power that these vicilins show for C. maculatus larvae. This can give an initial step in the briefing of as the vicilins interact with alive chitin in and exert its toxic power for insects that possess peritrophic membrane

Polymers of mushroom cellular wall are recognized for presenting a lot of biological activities such as anti-inflammatory, antioxidant and anti-tumoral action. Polysaccharides from mushrooms of different molecular mass obtained mushrooms can activate leucocytes, stimulate fagocitic, citotoxic and antimicrobial activity including oxygen reactive species production. In this study were investigated chemical characteristics, in vitro antioxidant activity and anti-inflammatory action in an acute inflammation model of the polysaccharides extracted from Tylopilus ballouii. Results showed that were mainly extracted polysaccharides and that it primarily consisted of mannose and galactose with variable amounts of xylose and fucose. Infrared analysis showed a possible interation between this polysaccharides and proteins. In addition, molecular mass was about 140KDa. Antioxidant activity was tested by superoxide and hydroxyl radical scavenging assay, total antioxidant activity and lipid peroxidation assay. For superoxide and hydroxyl radical generation inhibition, polysaccharides have an IC50 of 2.36 and 0.36 mg/mL, respectively. Lipid peroxidation assay results showed that polysaccharides from Tylopilus ballouii present an IC50 of 3.42 mg/mL. Futhermore, anti-inflammatory assay showed that polysaccharides cause an paw edema decreasing in 32.8, 42 and 56% in 30, 50 and 70 mg/Kg dose, respectively. Thus, these results can indicate a possible use for these polysaccharides from Tylopilus ballouii as an anti-inflammatory and antioxidant.

Galactans are polysaccharides sulfated present in the cell wall of red algae. Carrageenans are galactans well known in the food industry as gelling polysaccharides and for induce inflammatory process in rodents as animal model. The extraction of polysaccharides from A. multifida has been carried out by proteolysis and precipitation in different volumes of acetone, which produced three fractions (F1, F2, and FT). Chemical and physical analyses revealed that these fractions are sulfated galactan predominantly. Results of the antioxidant activity assays showed that all of these fractions have antioxidant activity and that was associated with sulfate content of the analysis of reducing power and total antioxidant capacity. However, these fractions were not effective against lipid peroxidation. The fraction FT presented higher activity on the APTT test at 200 μg (> 240 s). The assessment of the hemolytic activity showed that the FT fraction has the best activity, increasing lyses by the complement system to 42.3% (50 μg) (p< 0,001). The fraction FT showed the best yield, anticoagulant and hemolytic activity between the three fractions and therefore it was choose for the in vivo studies. The Inflammation assessment using the FT fraction (50 mg / kg MB) showed that the cellular migration and the IL-6 production increased 670.1% (p< 0,001) and 531.8% (p< 0,001), respectively. These results confirmed its use as an inflammation inducer in animal model. Cytotoxicity assay results showed that all fractions have toxic effects on 3T3 and HeLa cells after exposition of 48 hours, except when 100 μg for both F1 and FT were used. These results arise the discussion whether these polysaccharides it should be used as additive in foods, cosmetics and medicines.

The present work is to study the characteristics and technological properties of soil-cement bricks made from binary and ternary mixtures of Portland cement, sand, water, with or without addition of gravel from the drilling of oil wells, which could be used by industry, aiming to improve its performance and reduce cost by using the residue and, consequently, increasing its useful life. The soil-cement bricks are one of the alternatives to masonry construction. These elements, after a short curing period, provide compressive strength similar to that of solid bricks and ceramic blocks, and the higher the resistance the higher the amount of cement used. We used the soil from the city of São José do Mipibu / RN, the banks of the River Baldun, cement CPIIZ-32 and residue of drill cuttings from oil wells drilling onshore wells in the town of Mossley, RN, provided Petrobras. To determine the optimum mix, we studied the inclusion of different residues (100%, 80%, 70%, 60% and 50%) where 15 bodies were made of the test piece. The assessment was made of bricks made from simple compression tests, mass loss by immersion and water absorption. The experimental results proved the efficiency and high utilization of the waste from the drilling of oil wells, making the brick-cement-soil residue with a higher strength and lower water absorption. The best result in terms of mechanical strength and water absorption for the ternary mixture was 10% soil, 14% cement and 80% residue. In terms of binary mixtures, we obtained the best result for the mix-cement residue, which was 14% cement incorporated in the residue
Desenvolvimento de bloco de vedação com barita na composição de partida para blindagem de radiação X

This work main objective is to study the use of bricks in barium X-rays rooms in order to contribute to the optimization of shielding rooms diagnosis. The work was based on experimental measurements of X-ray attenuation (40 to 150 kV), using ceramic seal bearing the incorporation of barium sulfat (BaSO4). Different formulations were studied in three different firing temperatures and evaluated for incorporation in the ceramic body. The composition of 20% of barite processed at a temperature of 950 ° C showed better physical and mechanical properties, is considered the most suitable for the purpose of this work. Were produced bricks sealing composition formulated based on that presented the best technological features. These blocks were tested physically as a building material and wall protective barrier. Properties such as visual, deviation from the square, face flatness, water absorption and compressive strength were evaluated for all the blocks produced. The behavior of this material as attenuator for X-rays was investigated by experimental results which take into account mortar manufacturers barium through the different strains and compared with the reference material (Pb). The simulation results indicated that the ceramic block barium shows excellent properties of attenuation equivalence lead taking into account the energy used in diagnostic X-ray

The oil wells cementing is a critical step during the phase of well drilling, because problems during the operation of slurry pumping and an incomplete filling of the annular space between the metal casing and the formation can cause the slurry loss. Therefore, the slurry adopted in primary cementing an oil well must be properly dosed so that these problems are avoided during its pumping. When you drill a well in a weak rock formation requires even more careful, because should be a limit of hydrostatic pressure exerted during cementation, that does not occur rock collapse. With the objective of performing the cementing of a well whose formation is weak or unconsolidated are employed lighter slurries. Thus, this study used slurries with sodium silicate and nano silica in concentrations of 0,1; 0,4; 0,7 e 1,0 gpc, in which the slurries with nano silica showed the rheological parameters higher concentrations of up to 0.7 gpc and for concentration of 1.0 the slurry with sodium silicate obtained the highest values, remaining above the limits for application in fields, mainly wells with low fracture gradient, because a significant increase in viscosity may result in an increase in pressure pumping in operations of secondary cementations. Furthermore, there was no decrease in strength with increasing concentration of additive. Then, it is possible use of these additives to formulate Lighter slurry

The coatings mortars are essential elements of building structures because they execute an important role in protecting walls and are particularly exposed to aggressive action responsible for its degradation over time. The importance of wall coverings has been the subject of discussion and analysis in the conservation and rehabilitation of old buildings. Are sometimes removed and replaced with inappropriate solutions of constructive point of view or architecture. The most commonly used coatings on walls of old buildings is based on traditional hydraulic lime mortars. The present study aims at the formulation of new lime- based mortars and aerial fine aggregate, in order to contribute to a better field of conservation and restoration mortar coating of old buildings. Residue was used for polishing porcelain as fine aggregate, replacing the aggregate (sand), in percentages 05-30% by mass. We conducted a thorough evaluation of the mortar properties in fresh and hardened state by comparing the performance of the same with a reference mortar. The residue used was characterized as the density, bulk density, and particle size laser, scanning electron microscopy, X-ray diffraction and X-ray fluorescence. Formulations were produced 7, 6 with residue and one commonly used formulation, which served as a reference. In the formulations of lime mortars air (hydrated lime powder CH-I) has been adopted a stroke volume (1:3) with constant binder, was varied and the water / binder and aggregate and waste. For evaluation of mortars fresh, proceeded to consistency analysis, specific gravity, water retention and air content embedded. In the hardened state assays were performed in specific gravity, water retention, modulus of elasticity, tensile strength in bending, compressive strength, water absorption by capillary action, adhesion, tensile strength, resistance to shrinkage and salts by of crystallization trials with resources chloride solution, nitrate and sulfate all sodium in prismatic at 90 days of age, in addition to the micro structural analysis of mortars. Based on the results we can see that the mortar formulated with 10% content of waste and the reference free retraction feature more stable closer to neutrality. The composition of 10% was obtained better performance against the action of the salt crystallization. The mortar with 15% residue obtained better density, lower air content embedded and high capacity for water retention developing good workability. The replacement of 20% of waste generates a satisfactory utilization of resistance to compression, flexion and traction grip the base. And, finally, it can be seen that the mortar with 10, 15 and 20% residual show, in principle, good suitability as coatings, thus enabling a final result consistent with durability, workability and aesthetics developing therefore a material with better performance to repair or replace existing mortars in old buildings

Food habits and morpho-histology of the digestive tract of marbled swamp eel, Synbranchus marmoratus (Block, 1917) were investigated. The fish samples were captured during August, 2007 to July, 2008 in the Marechal Dutra reservoir, Acari, Rio Grande do Norte. The rain fall data was obtained from EMPARN. The fish captured, were measured, weighed, dissected, eviscerated and individual stomach weights were registered. The stomach contents analyses were carried out based on volumetric method, points, frequency of occurrence and applying the Index of Relative Importance. The degrees of repletion of the stomachs were determined besides the Index of Repletion relating to feeding activity variations and frequency of ingestion during the rainy and dry seasons. The rainfall varied from 0 mm a 335 mm with a mean value of 71.62 mm. Highest rainfall of 335.5 mm was registered in March, 2008 and August to December was the dry period. During the dry period the study species presented high degrees of repletion of the stomachs, with a peak value in the month of September (mean = 4.54; ± SD = 0.56). The minimum mean value of = 3.99 ± SD = 0.25 was registered in the month of May during the rainy period. The stomach contents of S. marmoratus registered show that this fish prefers animals, 78.22% of crustaceans 2.85% of mollusks, 3.25% of fish, 1.4% of insects and 13.5% of semi-digested organic matter, thus characterizing the study species as a carnivore with a preference for crustaceans. The morpho-histological aspects of the digestive tract of S. marmoratus indicate that the mouth is terminal adapted to open widely, thin lips with taste buds, small villiform teeth forming a single series on maxillas, four pairs of branchial arches with short and widely spaced branchial rays. The oesophagus is short and cylindrical with a small diameter. The oesophagus wall is thick with mucas surface and internal parallel folds. The stomach is retilinical in form, presenting cardiac, caecal and pyloric portions. The caecal portion is long and is intermediary in position between the cardiac and pyloric portions. The cardiac portion of the stomach is short and cylindrical formed of simple epithelial cylindrical mucus cells. The caecal portion is long with narrow walls, a big cavity and smaller folds which give rise to gastric glands. The phyloric portion has no glands and primary or secondary mucas folds. The morphohistological aspects of the digestive tract of S. marmoratus indicate its adaptation to a carnivorous feeding habit
