The aim of this study was to evaluate the humoral antibody response, the genome viral excretion and the contact transmission of pathogenic chicken origin Newcastle disease virus (NDV) from experimentally infected pigeons (Columba livia) to in-contact pigeon. The antibody response to infection was assessed by the hemagglutination inhibition (HI) test and the genome viral excretion was detected by RT-PCR. Viral strain induced high antibody levels, both in inoculated and in sentinel birds. The pathogenic viral strain for chickens was unable to produce clinical signs of the disease in experimentally infected pigeons, although it induced the Immoral antibody response and produced NDV genome shedding. NDV genome was detected intermittently throughout the experimental period, from 5 days post-infection (dpi) to 24 dpi. Therefore, viral genome shedding occurred for 20 days. The viral genome was detected in all birds, between I I and 13 dpi. Furthermore, the high infectivity of the virus was confirmed, as all non-inoculated sentinel pigeons showed antibody levels as high as those of inoculated birds. (C) 2007 Elsevier B.V. The demographic and epidemiological transition process caused by a declining in birth rates and in mortality, also changes occurred in morbidity and mortality is represented by the increasing of the aging population and the raising of chronic diseases. These diseases are characterized by multiple etiologies, risk factors, long latency period, a prolonged evolution, non-infectious origin and it has association with functional impairment and disability. Thus, elderly with chronic non-communicable disease has priority because they belong to a vulnerable group to get affection of comorbidities in aging, with increased demand and spending on health services. This study is aimed to analyse the understanding of elderly people with chronic non comunicable disease in the medium complexity service as a contribution to the improvement of health care in the city of Natal / RN. This is a descriptive and exploratory study with a quantitative approach, carried out at the Specialized Center for Elderly Health Care and at the Pescadores Hospital. The population was composed of 4,180 persons with a sample of 124 elderly aged above 60 years, attended in these medium complexity services. The instrument, a structured form, adapted from a questionnaire for monitoring risk and protective factors for chronic disease of the Ministry of Health. To collect data was was used the interview form containing demographic data, habits, health status and health care services. The results were processed using the Statistical Package for Social Science, version 18.0, analyzed by simple statistics. It was found that most seniors were female, predominantly between 70 and 74 years old, married, with a brown skin tone and Catholic religion, more than half had incomplete basic education, family income between one to two minimum wages and living with their families. Regarding the interviewers lifestyle, 94.4%, of them ate chicken and 97.6%, fruits, it was observed a reduction in smoking, alcoholism habits and physical activity according to the increasing age, 58.1 and 18.5% had insomnia18,5 % used sleeping pills. The elderly (51.6%) reported using services in times of sickness, seeking primary care at first (30.6%), 52% did not receive referral and was looking for free demand (38.7%). The most reported morbidity was hypertension, followed by musculoskeletal disorders. Regarding the difficulties in seeking health services, the delay in treatment and the waiting line were the most cited by the elderly. Almost all of them reported no activities to promote health in these services and those who received individual counseling on chronic diseases. Almost always, the health professionals who care of them, were mostly doctors followed by nurses. Based on the results presented, it is considered that the health services of medium complexity must undergone a more continuous dialogue with other attention level and focus on actions of health promotion and prevention. It is also recommended the necessity for qualified professionals to delivery health care to elderly and the implementation of protocols by a multidisciplinary health team, intending to provide better and continous care for the elderly with chronic diseases. The healthcare professionals who served them, were mostly physicians, followed by nurses. Through the results presented, it is considered that the medium complexity healthcare services need to perform a more continuous dialogue with the other levels of attention focusing attention to the health promotion and prevention actions. It is also recommended the necessity for qualified professionals to delivery healthcare for the elderly, in addition, a protocol implementation for the multidisciplinary health care team, to provide better care, and also the care continuity to elderly with chronic diseases

The Bola-DRB3 gene participates in the development of the immune response and is highly polymorphic. For these reasons, it has been a candidate gene in studies of the genetic basis of disease resistance and in population genetic analysis. South American native cattle breeds have been widely replaced by improved exotic breeds leading to a loss of genetic resources. In particular South American native breeds have high levels of fertility and disease resistance. This work describes genetic variability in the BoLA-DRB3 gene in native (Caracu, Pantaneiro, Argentinean Creole) and exotic (Holstein, Jersey, Nelore, Gir) cattle breeds in Brazil and Argentina. PCR-RFLP alleles were identified by combining the restriction patterns for the BoLA-DRB3.2 locus obtained with RsaI, BstY, and HaeIII restriction enzymes. Allelic frequencies and deviations from the Hardy-Weinberg equilibrium were also calculated. Analysis of the 24 BoLA-DRB3 PCR-RFLP alleles identified showed differences in the allele distributions among breeds.

Although numerous studies have reported the production of skeletal muscle alpha -tropomyosin in E. coli, the protein needs to be modified at the amino terminus in order to be active. Without these modifications the protein does not bind to actin, does not exhibit head-to-tail polymerization, and does not inhibit the actomyosin Mg2+-ATPase in the absence of troponin. on the other hand, the protein produced in insect cells using baculovirus as an expression vector (Urbancikova, M., and Hitchcock-DeGregori, S. E., J. Biol. Chem., 269, 24310-24315, 1994) is only partially acetylated at its amino terminal and therefore is not totally functional. In an attempt to produce an unmodified functional recombinant muscle alpha -tropomyosin for structure-function correlation studies we have expressed the chicken skeletal alpha -tropomyosin cDNA in the yeast Pichia pastoris. Recombinant protein was produced at a high level (20 mg/L) and was similar to the wild type muscle protein in its ability to polymerize, to bind to actin and to regulate the actomyosin S1 Mg2+-ATPase. Este trabalho foi desenvolvido com o objetivo de pesquisar o efeito da energia da dieta sobre a temperatura do cólon e concentração de proteína de choque térmico (Hsp70) de frangos à temperatura ambiente, bem como durante o estresse calórico agudo. Os frangos foram criados até 51 dias de idade e alimentados com dietas contendo nível de energia alto (13.186 kJ EM/kg) ou baixo (12.139 kJ EM/kg). No 21º e 51º dias de idade, a temperatura do cólon foi medida e amostras de fígado foram obtidas para quantificação da Hsp70 através da análise por Western Blotting.. Nessas mesmas idades, a resposta das aves ao estresse calórico agudo (37º C/5 h) foi avaliada (temperatura colón e Hsp70 no fígado). Os resultados mostraram que aos 21 dias de idade, à temperatura ambiente, a temperatura do cólon e a concentração de Hsp70 hepática não foram afetadas pela energia da dieta, mas, aos 51 dias de idade, os frangos alimentados com baixos teores de energia apresentaram menores concentrações de Hsp70 no fígado. As respostas ao estresse calórico agudo mostraram que as aves alimentadas com dietas de alta energia tiveram menor incremento na temperatura do cólon, bem como no conteúdo de Hsp70 hepático. Os resultados desse estudo sugerem que a síntese de Hsp70 no fígado pode ser afetada pela energia da dieta e que frangos alimentados com altos níveis de energia podem ter a termotolerância alterada em condições de estresse agudo pelo calor.

Este trabalho foi realizado para avaliar o efeito da adição de enzima e/ou probiótico, bem como do estresse calórico sobre a atividade de enzimas digestivas de frangos de corte aos 7, 14, 28 e 42 dias de idade. O delineamento experimental utilizado foi o inteiramente casualizado em parcelas subdivididas com 6 tratamentos primários, constituindo um esquema fatorial 6x2 (rações x temperatura) e 4 tratamentos secundários (idades de amostragem), com 2 repetições. Não foi verificado efeito significativo dos tratamentos sobre a atividade específica das enzimas, exceto para as atividades de amilase aos 14 dias e tripsina aos 28 dias de idade, nas quais a adição de enzima proporcionou maiores atividades. Já a idade de criação afetou significativamente todas as enzimas quantificadas, sendo que a atividade específica de lipase diminuiu e as atividades específicas de amilase, tripsina e quimotripsina aumentaram com a idade das aves. A temperatura ambiente (calor) também afetou a produção enzimática de acordo com a idade dos frangos de corte, com um aumento na atividade de lipase e redução na tripsina e amilase.

The objective of this work was to assess the functionality of the glycolytic pathways in the bacterium Xylella fastidiosa. To this effect, the enzymes phosphoglucose isomerase, aldolase, glyceraldehyde-3-phosphate dehydrogenase and pyruvate kinase of the glycolytic pathway, and glucose 6-phosphate dehydrogenase of the Entner-Doudoroff pathway were studied, followed by cloning and expression studies of the enolase gene and determination of its activity. These studies showed that X. fastidiosa does not use the glycolytic pathway to metabolize carbohydrates, which explains the increased duplication time of this phytopatogen. Recombinant enolase was expressed as inclusion bodies and solubilized with urea (most efficient extractor), Triton X-100, and TCA. Enolase extracted from X. fastidiosa and from chicken muscle and liver is irreversibly inactivated by urea. The purification of enolase was partial and resulted in a low yield. No enzymatic activity was detected for either recombinant and native enolases, aldolase, and glyceraldehyde-3-phosphate dehydrogenase, suggesting that X. fastidiosa uses the Entner-Doudoroff pathway to produce pyruvate. Evidence is presented supporting the idea that the regulation of genes and the presence of isoforms with regulation patterns might make it difficult to understand the metabolism of carbohydrates in X. fastidiosa.

Data comprising 53,181 calving records were analyzed to estimate the genetic correlation between days to calving (DC), and days to first calving (DFC), and the following traits: scrotal circumference (SC), age at first calving (AFC), and weight adjusted for 550 d of age (W550) in a Nelore herd. (Co)variance components were estimated using the REML method fitting bivariate animal models. The fixed effects considered for DC were contemporary group, month of last calving, and age at breeding season (linear and quadratic effects). Contemporary groups were composed by herd, year, season, and management group at birth; herd and management group at weaning; herd, season, and management group at mating; and sex of calf and mating type (multiple sires, single sire, or AI). In DFC analysis, the same fixed effects were considered excluding the month of last calving. For DC, a repeatability animal model was applied. Noncalvers were not considered in analyses because an attempt to include them, attributing a penalty, did not improve the identification of genetic differences between animals. Heritability estimates ranged from 0.04 to 0.06 for DC, from 0.06 to 0.13 for DFC, from 0.42 to 0.44 for SC, from 0.06 to 0.08 for AFC, and was 0.30 for W550. The genetic correlation estimated between DC and SC was low and negative (-0.10), between DC and AFC was high and positive (0.76), and between DC and W550 was almost null (0.07). Similar results were found for genetic correlation estimates between DFC and SC (-0.14), AFC (0.94), and W550 (-0.02). The genetic correlation estimates indicate that the use of DC in the selection of beef cattle may promote favorable correlated responses to age at first mating and, consequently, higher gains in sexual precocity can be expected.

Ram lambs (n = 356) born to Romney ewes and sired by a range of straightbred or crossbred sires were evaluated for carcass composition and, for a subsample (n = 236), some meat quality characteristics over a period of 4 years. Within each year, 2 to 4 sire genotypes, including Romney controls, were evaluated at 2 slaughter ages (5-9 months and 10-11 months). Weight-adjusted carcass length was greater for lambs with some East Friesian or Finn genes, but no differences were found in dressing-out percent. At a constant carcass weight, carcass fatness (fat depths and leg dissectible fat) was lower in lambs with East Friesian, Texel, or Finn genes. Meat yield at a constant weight tended to be greatest in sire groups that were least fat, and M:B ratio in the femur region was highest in lambs with Texel genes and, to a lesser extent, those with Poll Dorset genes. Leg muscularity in the femur region generally paralleled differences in M:B, except in a comparison of East Friesian-cross and Romney lambs where the former had lower muscularity but slightly better M:B. Lambs slaughtered at the older age for each year had longer and leaner carcasses at the same weight, possibly because they had been taken part way through a winter or because of slower growth rates leading up to slaughter. Meat quality characteristics differed little between the genotype groups, but for 2 of 3 years, meat from the semimembranosus muscle of lambs in the older slaughter lot was significantly tougher.

It is reasonable to assume that the knowledge of suckling behaviour contributes to optimal management and selection of beef cattle. However, there is little information about suckling behaviour of some beef cattle breeds. The aim of this study was to describe the suckling behaviour of two zebu (Bos indicus) and one criollo (Bos taurus) breeds, analysing the potential effects of breed and some environmental factors on suckling frequency and duration. Forty cows, 17 Nelore, 14 Gir (both zebu) and 9 Caracu (criollo) were bred in a diallelic crossing design. The cows and resulting calves were kept on pasture from birth to weaning. Their behaviour was recorded weekly during daylight. Three behavioural traits were considered: number of suckling meals (NSM), duration of each suckling meal (DSM) and total suckling duration (TSD). Allosuckling was not observed. The calves suckled at any time during the daylight and the overall means were: NSM = 2.57 +/- 0.05 meals/12 h (from back transformed data), DSM = 9.25 +/- 0.11 min/suckling meal and TSD = 23.76 +/- 0.47 min/12 h. There was an effect of dam's breed on NSM and DSM; the calf's genetic group within breed of cow influenced NSM and TSD when the dams were from the Nelore breed. The age of calf had significant effects on all traits. Males averaged higher NSM and TSD (2.60 +/- 0.03 meals and 25.05 +/- 1.37 min/12 h, respectively) than females (2.12 +/- 0.04 meals and 21.51 +/- 1.55 min/12 h, respectively). The differences in suckling behaviour seem to be produced by a complex combination of genetic and environmental factors, which result in a particular behavioural relationship within mother-offspring pairs. Várias alterações metabólicas e fisiológicas são desencadeadas em frangos de corte submetidos a altas temperaturas ambientais, o que pode acarretar em grandes perdas no desempenho e na imunocompetência destas aves. Além das técnicas de controle ambiental estarem sendo freqüentemente empregadas para a redução do impacto negativo do estresse calórico sobre o desempenho das aves, outras medidas estão constantemente sendo estudadas. Nos últimos anos, o manejo nutricional adequado também tem demonstrado ser efetivo como medida preventiva para o estresse calórico, pois o funcionamento do sistema termorregulador do frango (produção de calor, rotas evaporativas e não evaporativas de dissipação de calor) pode ser influenciado pela dieta, em especial, o estabelecimento de adequados balanços eletrolíticos, devido a sua importância fisiológica no mecanismo do estresse calórico. Assim, os mecanismos nutricionais devem ser reavaliados como uma ferramenta no controle desta disfunção metabólica das aves.
