We report an optical sensor based on localized surface plasmon resonance (LSPR) to study small-molecule protein interaction combining high sensitivity refractive index sensing for quantitative binding information and subsequent conformation-sensitive plasmon-activated circular dichroism spectroscopy. The interaction of α-amylase and a small-size molecule (PGG, pentagalloyl glucose) was log concentration-dependent from 0.5 to 154 μM. In situ tests were additionally successfully applied to the analysis of real wine samples. These studies demonstrate that LSPR sensors to monitor small molecule–protein interactions in real time and in situ, which is a great advance within technological platforms for drug discovery.

Astringency is an organoleptic property of beverages and food products resulting mainly from the interaction of salivary proteins with dietary polyphenols. It is of great importance to consumers, but the only effective way of measuring it involves trained sensorial panellists, providing subjective and expensive responses. Concurrent chemical evaluations try to screen food astringency, by means of polyphenol and protein precipitation procedures, but these are far from the real human astringency sensation where not all polyphenol–protein interactions lead to the occurrence of precipitate. Here, a novel chemical approach that tries to mimic protein–polyphenol interactions in the mouth is presented to evaluate astringency. A protein, acting as a salivary protein, is attached to a solid support to which the polyphenol binds (just as happens when drinking wine), with subsequent colour alteration that is fully independent from the occurrence of precipitate. Employing this simple concept, Bovine Serum Albumin (BSA) was selected as the model salivary protein and used to cover the surface of silica beads. Tannic Acid (TA), employed as the model polyphenol, was allowed to interact with the BSA on the silica support and its adsorption to the protein was detected by reaction with Fe(III) and subsequent colour development. Quantitative data of TA in the samples were extracted by colorimetric or reflectance studies over the solid materials. The analysis was done by taking a regular picture with a digital camera, opening the image file in common software and extracting the colour coordinates from HSL (Hue, Saturation, Lightness) and RGB (Red, Green, Blue) colour model systems; linear ranges were observed from 10.6 to 106.0 μmol L−1. The latter was based on the Kubelka–Munk response, showing a linear gain with concentrations from 0.3 to 10.5 μmol L−1. In either of these two approaches, semi-quantitative estimation of TA was enabled by direct eye comparison. The correlation between the levels of adsorbed TA and the astringency of beverages was tested by using the assay to check the astringency of wines and comparing these to the response of sensorial panellists. Results of the two methods correlated well. The proposed sensor has significant potential as a robust tool for the quantitative/semi-quantitative evaluation of astringency in wine.

Hyperamylasemia has been reported in more than 65% of patients with severe leptospirosis, and the true diagnosis of acute pancreatitis is complicated by the fact that renal failure can increase serum amylase levels. Based on these data we retrospectively analyzed the clinical and histopathological features of pancreas involvement in 13 cases of fatal human leptospirosis. The most common signs and symptoms presented at admission were fever, chills, vomiting, myalgia, dehydratation, abdominal pain and diarrhea. Trombocytopenia was evident in 11 patients. Mild increased of AST and ALT levels was seen in 9 patients. Hyperamylasemia was recorded in every patient in whom it was measured, with values above 180 IU/L (3 cases). All patients presented acute renal failure and five have been submitted to dialytic treatment. The main cause of death was acute respiratory failure due to pulmonary hemorrhage. Pancreas fragments were collected for histological study and fat necrosis was the criterion used to classify acute pancreatitis. Histological pancreatic findings were edema, mild inflammatory infiltrate of lymphocytes, hemorrhage, congestion, fat necrosis and calcification. All the patients infected with severe form of leptospirosis who develop abdominal pain should raise the suspect of pancreatic involvement.

Software tools in education became popular since the widespread of personal computers. Engineering courses lead the way in this development and these tools became almost a standard. Engineering graduates are familiar with numerical analysis tools but also with simulators (e.g. electronic circuits), computer assisted design tools and others, depending on the degree. One of the main problems with these tools is when and how to start use them so that they can be beneficial to students and not mere substitutes for potentially difficult calculations or design. In this paper a software tool to be used by first year students in electronics/electricity courses is presented. The growing acknowledgement and acceptance of open source software lead to the choice of an open source software tool – Scilab, which is a numerical analysis tool – to develop a toolbox. The toolbox was developed to be used as standalone or integrated in an e-learning platform. The e-learning platform used was Moodle. The first approach was to assess the mathematical skills necessary to solve all the problems related to electronics and electricity courses. Analysing the existing circuit simulators software tools, it is clear that even though they are very helpful by showing the end result they are not so effective in the process of the students studying and self learning since they show results but not intermediate steps which are crucial in problems that involve derivatives or integrals. Also, they are not very effective in obtaining graphical results that could be used to elaborate reports and for an overall better comprehension of the results. The developed tool was based on the numerical analysis software Scilab and is a toolbox that gives their users the opportunity to obtain the end results of a circuit analysis but also the expressions obtained when derivative and integrals calculations, plot signals, obtain vector diagrams, etc. The toolbox runs entirely in the Moodle web platform and provides the same results as the standalone application. The students can use the toolbox through the web platform (in computers where they don't have installation privileges) or in their personal computers by installing both the Scilab software and the toolbox. This approach was designed for first year students from all engineering degrees that have electronics/electricity courses in their curricula.

The paper presents a multi-robot cooperative framework to estimate the 3D position of dynamic targets, based on bearing-only vision measurements. The uncertainty of the observation provided by each robot equipped with a bearing-only vision system is effectively addressed for cooperative triangulation purposes by weighing the contribution of each monocular bearing ray in a probabilistic manner. The envisioned framework is evaluated in an outdoor scenario with a team of heterogeneous robots composed of an Unmanned Ground and Aerial Vehicle.

To improve surgical safety, and to reduce the mortality and surgical complications incidence, the World Health Organization (WHO) developed the Surgical Safety Checklist (SSC). The SSC is a support of information that aids health professionals to reduce the number of complications, induction of anaesthesia, period before skin incision and period before leaving the operating room (OR). The SSC was tested in several countries of the world and their results shown that after introduction of the SSC the incidence of patient complication lowered from 11.0% to 7.0% (P<0.001), the rate of death declined from 1.5% to 0.8% (P = 0.003) and the nurses recognized that patients identity was more often con rmed (81.6% to 94.2%, P<0.01) in many institutions. Recently the SSC was also implemented in Portuguese hospitals, which led us to its study in the real clinical environment. An observational study was performed: several health professionals were observed and interviewed, to understand the functioning of the SSC in an OR, during the clinical routine. The objective of this study was to understand the current use of the SSC, and how it may be improved in terms of usability, taking advantage of the technological advancements such as mobile applications. During two days were observed 14 surgeries, only 2 surgeries met the requirements for the three phases of the SSC, as de ned by the WHO. Of the remaining 12 observed surgeries, 9 surgeries completed the last phase at the correct time. It was also observed that only in 2 surgeries all the phases of the SSC were read aloud to the team and that, in 7 surgeries, several items were read aloud and answered but no one was checking the SSC, only after the end of the phase. The observational study results disclose that several health professionals do not meet with rules of the WHO manual. This study demonstrates that it is urgent to change the mindset of health professionals, and that di erent features in the SSC may be useful to make it more easy to use. With the results of the observational study, a SSC application proposal was developed with new functionalities to improve and aid the health professional in its use. In this application the user can chose between a SSC already created to a speci c surgery or to create a new SSC, adding and adapting some questions from the WHO standard. To create a new SSC, the application is connected to an online questionnaire builder (JotForm). The choice for this online questionnaire builder went through three essential characteristics: number of types of questions, mainly checkbox, radio button and text; the possibility of to create sections inside sections and the API. In addition, in this proposal the improvements are focused in forcing the user to focus in the work ow of the SSC and to save the input timestamps and any actions made by them. Therefore, the following features was implemented to achieve that goal: display one item of the SSC at a time; display the stage where the SSC is; do not allow going back to the previous step; do not allow going forward to the next item if the current is not lled; do not allow going forward to the next item if the time it took to ll the item was too short and log any action made by the user.

O coaching é um processo que permite ajudar um ou mais indivíduos a definirem e saberem como concretizar os seus objetivos, sejam eles pessoais ou profissionais. Atualmente, existe um interesse e procura crescente de pessoas com experiência nesta área (designados por coaches) por parte de empresas, equipas desportivas, escolas e outras organizações, com a finalidade de obter um maior rendimento. De forma a ajudar os intervenientes no processo, este documento demonstra a necessidade de existir uma ferramenta de apoio que permite aos coaches gerirem melhor a sua atividade profissional. A pesquisa e estudo efetuados procuram responder a este caso, desenvolvendo um sistema informático inteligente de apoio ao coach dotado de uma interface centrada no utilizador. Antes de iniciar o desenvolvimento de um sistema inteligente é necessário realizar e apresentar um levantamento do estado da arte, mais concretamente sobre a interação homem-computador, modelação do perfil de utilizador e processo de coaching, que apresenta os fundamentos teóricos para a escolha da metodologia de desenvolvimento adequado. São apresentadas posteriormente as fases constituintes do modelo de desenvolvimento de interfaces escolhido, a engenharia de usabilidade, que se inicia com uma análise detalhada, permitindo de seguida uma estruturação dos conhecimentos obtidos e a aplicação de linhas de orientação estipuladas, finalizando com testes de utilização e respetivo feedback dos utilizadores. O protótipo desenvolvido distingue utilizadores com diferentes características, através de uma classificação por níveis e permite gerir todo o processo de coaching efetuado a outras pessoas ou ao próprio utilizador. O facto de existir uma classificação dos utilizadores faz com que a interação entre sistema e utilizadores seja diferente e adaptada às necessidades de cada um. O resultado dos testes de utilização com um caso prático e dos questionários efetuados permite detetar se o modelo foi bem-sucedido e funciona corretamente e o que é necessário alterar no futuro para facilitar a interação e satisfazer as necessidades de cada utilizador.

A 64-year-old apparently immunocompetent white man developed lung and brain lesions of disseminated cryptococcosis. The radiologic features mimicked those of lung cancer metastatic to the central nervous system. C. gattii was recovered from cultures of bronchoalveolar lavage fluid, brain biopsy, and blood. The same fungus was recovered from pulmonary and brain specimens at autopsy. Serum and cerebrospinal fluid cryptococcal antigen tests were diagnostic in our case and should be included in the diagnostic evaluation of unexplained pulmonary and cerebral lesions. A literature search showed few reports of fungemia by this species of Cryptococcus, contrasting to C. neoformans.

In Brazil relatively little attention is being paid to the study of the features of the spread of the AIDS epidemic towards small cities and rural areas. We report a descriptive study on the epidemiological features of HIV infection among 208 adult patients seen between July 1999 and May 2006 in the municipal HIV/AIDS Programs of three cities of inner Rio de Janeiro State: Saquarema, Santo Antonio de Pádua and Miracema. A portrait of a heterosexual epidemic emerged, with an overall male to female ratio of 1.1. More than 90% were residents of the studied cities, demonstrating a local demand for HIV-related assistance and the importance of municipal HIV/AIDS Programs. Past or current use of snorted cocaine was reported by a quarter of the patients. Older age and male gender were independent predictors of having a diagnosis of AIDS at presentation. The latter is in accordance with a more recent wave of epidemic spread towards female gender. A low frequency of male circumcision, an important determinant of heterosexual HIV transmission, was recorded. Almost 60% of the patients first presented in advanced stages of HIV infection, suggesting the existence of a large pool of undiagnosed cases in the community.

To identify the clinical and epidemiological profile of cryptococcosis diagnosed at the University Hospital of the Federal University of Mato Grosso do Sul, Brazil, medical records of 123 patients admitted from January 1995 to December 2005 were analyzed. One hundred and four cases (84.5%) had HIV infection, six (4.9%) had other predisposing conditions and 13 (10.6%) were immunocompetent. Male patients predominated (68.3%) and their age ranged from 19 to 69 years (mean: 35.9). Most patients (73.2%) were born and lived lifelong in the state of Mato Grosso do Sul. Involvement of the central nervous system occurred in 103 patients (83.7%) and headache and vomiting were the most frequent symptoms. In 77 cases it was possible to identify the Cryptococcus species: 69 (89.6%) C. neoformans and eight (10.4%) C. gattii. Amphotericin B was the drug of choice for treatment (106/123), followed by fluconazole in 60% of cases. The overall lethality rate was 49.6%, being 51% among the HIV infected patients and 41.2% among the non-HIV infected (p > 0.05). Although cryptococcosis exhibited in our region a similar behavior to that described in the literature, the detection of an important rate of immunocompetent individuals and five C. gattii cryptococcosis in HIV-infected patients is noteworthy.

O âmbito deste trabalho envolve o teste do modelo BIM numa obra em construção pela Mota-Engil – Engenharia, na extração experimental de peças desenhadas de preparação e apoio à execução de obra. No capítulo 1 deste relatório são definidos o âmbito e os objetivos deste trabalho, é feito um enquadramento histórico do tema e abordados conceitos e atividades da preparação de obra, na sua forma tradicional. O estado do conhecimento da preparação de obras e mais em concreto da tecnologia BIM a nível nacional e internacional é abordado no capítulo 2. Nesse sentido procura-se definir os conceitos principais inerentes a esta nova metodologia, que passa por identificar e caraterizar a tecnologia envolvida e o seu nível de desenvolvimento. Com suporte em casos práticos de preparação de obra na sua forma tradicional, identificados e desenvolvidos no capítulo 3, foi compilado um processo tipo de peças desenhadas de suporte identificadas e caracterizadas no capítulo 4, frequentes e comuns à execução de diversos tipos de obras de edifícios. Assente na compilação baseada em casos práticos e no estudo do projeto de execução da empreitada que sustenta o presente trabalho, com base no qual o modelo BIM foi concebido, identificou-se um conjunto de peças desenhadas de preparação e apoio à execução dos trabalhos, em 2D, a extrair do modelo. No capítulo 5, é feita uma descrição do modo como foi estudado o projeto da obra, com evidência para os fatores mais relevantes, especificando os desenhos a extrair. Suportada pelo programa de modelação ArchiCAD, a extração do conjunto de desenhos identificados anteriormente foi conseguida com recurso às funcionalidades disponíveis no software, que permite a criação de desenhos 2D atualizáveis ou não automaticamente a partir do modelo. Qualquer alteração introduzida no modelo virtual é automaticamente atualizada nos desenhos bidimensionais, caso o utilizador assim o pretenda. Ao longo desse trabalho foram detetados e analisados os condicionalismos inerentes ao processo de extração, referidos no capítulo 6, para estabelecimento de regras de modelação padrão a adotar em futuras empreitadas, que possam simplificar a obtenção dos elementos desenhados de preparação necessários à sua execução. No ponto 6.3 são identificadas melhorias a introduzir no modelo. Em conclusão no capítulo 7 são abordadas especificidades do setor da construção que sustentam e evidenciam cada vez mais a necessidade de utilizar as novas tecnologias com vista à adoção de práticas e ferramentas padrão de apoio à execução de obras. Sendo a tecnologia BIM, transversal a todo o setor, a sua utilização com regras padrão na conceção dos modelos e na extração de dados, potencia a otimização dos custos, do tempo, dos recursos e da qualidade final de um empreendimento, ao longo de todo o seu ciclo de vida, para além de apoiar com elevada fiabilidade as tomadas de decisão ao longo desse período. A tecnologia BIM, possibilita a antevisão do edifício a construir com um elevado grau de pormenor, com todas as vantagens que daí advêm.
