Background Dengue epidemics have been reported in Brazil since 1981. In Manaus, a large city in the Amazon region, dengue is endemic with all four-virus serotypes (DENV-1, -2, -3, and -4) simultaneously causing human disease. In 2008, during a surveillance of dengue virus in mosquitoes in the district of Tancredo Neves in Manaus, 260 mosquitoes of Aedes genus were captured, identified and grouped into pools of 10 mosquitoes. Findings RNA extracts of mosquito pools were tested by a RT-Hemi-Nested-PCR for detection of flaviviruses. One amplicon of 222 bp, compatible with dengue virus serotype 4, was obtained from a pool of Aedes aegypti. The nucleotide sequence of the amplicon indicated that the mosquitoes were infected with DENV-4 of genotype I. This virus of Asian origin has been described in Manaus in 2008 infecting acute febrile illness patients. Conclusion This is the first report of dengue virus serotype 4 genotype I infecting Aedes aegypti in the Americas.

Sulfur (S) deficiency in soils is becoming increasingly common in many areas of the world as a result of agronomic practices, high biomass exportation and reduced S emissions to the atmosphere. In this review, the incidence and commercial exploitation of S pools in nature are discussed, as well as the importance of S for plants and the organic and inorganic S forms in soil and their transformations, especially the process of microbiological oxidation of elemental sulfur (S0) as an alternative to the replenishment of S levels in the soil. The diversity of S0-oxidizing microorganisms in soils, in particular the genus Thiobacillus, and the biochemical mechanisms of S0 oxidation in bacteria were also addressed. Finally, the main methods to measure the S0 oxidation rate in soils and the variables that influence this process were revised.

Se estudia la relación de dominancia entre dos especies de peces (Parablennius parvicornis y Mauligobius maderensis) que conviven en los charcos del intermareal rocoso de las Islas Canarias. Ambas especies mostraron un patrón de jerarquía lineal basado en ataques. Se discute la jerarquía interespecífica y el cambió de rol al enfrentarse individuos de la misma especie e individuos de especies diferentes. Parablennius parvicornis mostró un comportamiento de cooperación intraespecífica y de codominancia frente a Mauligobius maderensis.It is studied the relationship of dominance between two fish species (Mauligobius maderensis and Parablennius parvicornis) which coexist in the same intertidal pools at the rocky shore of the Canary Islands. Both species showed a pattern of linear hierarchy based in attacks. It is discussed the intraspecific hierarchy and variations in the role of individuals when confronted against coespecifics or individuals of a different species. Parablennius parvicornis showed intraspecific cooperation and codominance against Mauligobius maderensis.

[ES] Respiration is a key ecological index. For either individuals or communities, it can be use to assess carbon and energy, demand and expenditure as well as carbon flow rates through food webs. When combined with productivity measurements it can establish the level of metabolic balance. When combined with measurements of respiratory capacity, it can indicate physiological state. Here, we report pilot studies the metabolism of the green algae, Ulva rotundata that inhabits intertidal pools of Gran Canaria. As a starting point we used the electron transport system (ETS) to differentiate between different growing conditions in the natural environment. We suspected different levels of stress associated with these conditions and looked for the influence of this stress in the ETS measurements. This technique has been successfully applied to study bacteria, phytoplankton and zooplankton in the ocean, but it has not been used to study sessile marine macroalgae. These neritic and littoral macrophytes have major ecological and industrial importance, yet little is known about their respiratory physiology. Such knowledge would strengthen our understanding of the resources of the coastal ocean and facilitate its development and best use. Here, we modified the ETS methodology for Ulva rotundata. With this modified ETS assay we investigated the capacity of Ulva to resist anoxia. We measured respiration with optodes (Fibox 4, Presens) in the dark to the point of oxygen exhaustion and through 24 h of anoxia. Then we exposed the Ulva to light and followed the oxygen increase due to photosynthesis. We discuss here the capacity of Ulva to survive during anoxia.

L’infezione da virus dell’ epatite E (HEV) nei suini e nell’uomo è stata segnalata in diversi Paesi. Nei suini, il virus causa infezioni asintomatiche, mentre nell’uomo è responsabile di epidemie di epatite ad andamento acuto nei Paesi a clima tropicale o subtropicale con condizioni igieniche scadenti, di casi sporadici in quelli sviluppati. HEV è stato isolato anche in diversi animali e l’analisi nucleotidica degli isolati virali di origine animale ha mostrato un elevato grado di omologia con i ceppi di HEV umani isolati nelle stesse aree geografiche, avvalorando l’ipotesi che l'infezione da HEV sia una zoonosi. In America del Sud HEV suino è stato isolato per la prima volta in suini argentini nel 2006, mentre solo dal 1998 esistono dati sull’ infezione da HEV nell’uomo in Bolivia. In questa indagine è stato eseguito uno studio di sieroprevalenza in due comunità rurali boliviane e i risultati sono stati confrontati con quelli dello studio di sieroprevalenza sopra menzionato condotto in altre zone rurali della Bolivia. Inoltre, mediante Nested RT-PCR, è stata verificata la presenza di HEV nella popolazione umana e suina. La sieroprevalenza per anticorpi IgG anti-HEV è risultata pari al 6,2%, molto simile a quella evidenziata nello studio precedente. La prevalenza maggiore (24%) si è osservata nei soggetti di età compresa tra 41 e 50 anni, confermando che l’ infezione da HEV è maggiore fra i giovani-adulti. La ricerca di anticorpi anti HEV di classe IgM eseguita su 52 sieri ha fornito 4 risultati positivi. Il genoma virale è stato identificato in uno dei 22 pool di feci umane e l'esame virologico di 30 campioni individuali fecali e 7 individuali di siero ha fornito rispettivamente risultati positivi in 4/30 e 1/7. La Nested RT-PCR eseguita sui 22 pool di feci suine ha dato esito positivo in 7 pool. L’analisi delle sequenze genomiche di tutti gli amplificati ha consentito di stabilire che gli isolati umani appartenevano allo stesso genotipo III di quelli suini e presentavano con questi una elevata omologia aminoacidica (92%).

The thesis describes the molluscan biodiversity of the infralittoral off-shore reefs in the "Secche di Tor Paterno" marine protected area lying in the Central Tyrrhenian Sea off the coasts of Lazio south of Roma. Data originate from underwater sampling activities carried out by SCUBA diving in four biocoenoses: Posidonia oceanica leaves and rhizomes, coralligenous concretions and detritic pools. The representativeness of molluscs as descriptors of biocoenoses is evaluated by preliminary comparisons with data about Polychaeta, Pleocyemata (Crustacea) and Brachiopoda obtained in the same survey. The malacocoenoses of the four biocoenoses are treated in detail. Then data are compared with other data sets to assess differences and similarities with other communities. The agreement between death and living assemblages in the reefs is evaluated for the Posidonia oceanica and the coralligenous biocoenosis and was carried out by a set of standard metrics and some benthic ecology methods. Molluscs perform very well as descriptors of biocoenoses, better than the other phyla. The molluscan assemblages of the reefs are very rich in species despite richness is mainly concentrated in the coralligenous and in the rhizomes of Posidonia oceanica. The leaves of Posidonia oceanica host a rather poor assemblage. Detritic pools host a poor but peculiar species assemblage. The dead-live agreement showed that death assemblages are highly representative of sediments of nearby biocoenoses as a result of low bottom transport. Fidelity metrics suggest a good agreement between the living and death assemblages when species richness and taxonomic composition are considered. The study suggests that fidelity is lower when considering the species dominance. These differences could be associated to the trophism of species and possibly to the species life span.

Zusammenfassung Diese Arbeit beschreibt Untersuchungen über die zellulären Mechanismen, die zur Bildung dieser DNA-Schäden führen, sowie über die biologischen Auswirkungen dieser Schäden. Die Untersuchungen zu Uracil in der DNA wurden in ung-knockout-MEFs und Mäusen durchgeführt, die es erlauben, die Konsequenzen eines Ausfalls der wichtigsten Reparaturglykosylase für Uracil zu beleuchten. Die Ergebnisse zeigen eine deutliche Akkumulation von Uracil in den ung-/--Mausfibroblasten im Vergleich zum Wildtyp. In frisch isolierten Leber- und Milzzellen der Mäuse konnte dieser genotypspezifische Unterschied, wenn auch weniger ausgeprägt, ebenso beobachtet werden, nicht jedoch in reifen Spermien. Dieser gewebespezifische Unterschied und die quantitativ stärker ausgeprägte Akkumulation in ung-/--Mausfibroblasten im Vergleich zu den Mäusegeweben gab Anlass zur Vermutung, dass die Proliferation der Zellen für den Haupteintrag an Uracil in die DNA verantwortlich ist. Erstmals konnte in Versuche mit konfluenten (nicht mehr proliferierenden) ung-/--Mausfibroblasten gezeigt werden, dass nicht die spontane hydrolytische Desaminierung von Cytosin, sondern der Fehleinbau von dUMP während der DNA-Replikation die Hauptquelle für Uracil in der DNA von Säugerzellen darstellt. Da der Uracilmetabolismus ein wichtiges Target in der Chemotherapie ist, lag es nahe, das zur Verfügung stehende ung-knockout-Modell der MEFs zur Untersuchung mit Fluorpyrimidinen, die als Zytostatika verwendet werden, einzusetzen. Da bisher die Ursachen der beobachteten Apoptose der Tumorzellen und aller anderen metabolisch hochaktiven Zellen eines behandelten Organismus noch nicht vollständig verstanden ist, wurden diese Zellen mit verschiedenen Fluorpyrimidinen behandelt, die als Thymidylatsynthasehemmer die de novo Synthese von Thymidin unterbinden. Es konnte gezeigt werden, dass ung-/- Mausfibroblasten, im Gegensatz zu ung+/+ Mausfibroblasten, verstärkt Uracil in der DNA akkumulieren. Obwohl die ung+/+ Mausfibroblasten keine erhöhten Uracil-Spiegel in der DNA aufwiesen, zeigten sie bei Inkubation mit einem der beiden Thymidylatsynthasehemmern, 5-Fluoruracil (5-FU), die gleiche Sensitivität in einem nachfolgenden Proliferationsversuch wie die ung-/- Mausfibroblasten. Dies lässt darauf schließen, dass weder Reparatur noch Einbau von Uracil in die DNA für die beobachtete Toxizität dieser Zytostatika notwendig sind. Ein weiterer Schwerpunkt dieser Arbeit war die Untersuchung des DNA-schädigenden Potenzials endogener ROS, die aus dem Fremdstoffmetabolismus stammen. Dazu wurden V79-Zellen verwendet, die mit dem humanen Enzym Cytochrom 2E1 (CYP2E1) transfiziert wurden (V79 CYP2E1) sowie Zellen, die ebenfalls durch Transfektion das humane Enzym Cytochromreduktase (auch Oxidoreduktase genannt) überexprimieren (V79 hOR). Beide Enzyme sind zusammen an der Hydroxylierung von Fremdstoffen beteiligt, bei der die Reduktion von molekularem Sauerstoff durch Übertragung von zwei Elektronen notwendig ist. Wird anstatt zweier Elektronen in Folge nur eines auf den Sauerstoff übertragen, so führt dieser von der Substratoxygenierung enkoppelte Vorgang zur Bildung von Superoxid. Daher galt es zu klären, ob das so erzeugte Superoxid und daraus gebildete ROS in der Lage sind, die DNA zu schädigen. Es konnte gezeigt werden, dass die Überexpression von CYP2E1 nicht zu einem erhöhten basalen Gleichgewichtsspiegel oxidativer DNA-Schäden führt und die Metabolisierung von Ethanol durch dieses Enzym ebenfalls keine DNA-Modifikationen verursacht. Die Überexpression der Cytochromreduktase hingegen führte gegenüber dem Wildtyp zu einem erhöhten basalen Gleichgewichtsspiegel oxidativer Basenmodifikationen nach Depletion von Glutathion, einem wichtigen zellulären Antioxidans. Im Mikrokerntest, der gentoxische Ereignisse wie Chromosomenbrüche in Zellen aufzeigt, zeigte sich schon ohne Glutathion-Depletion eine doppelt so hohe Mikrokernrate im Vergleich zum Wildtyp. In weiteren Versuchen wurden die V79-hOR-Zellen mit dem chinoiden Redoxcycler Durochinon inkubiert, um zu untersuchen, ob das vermutlich durch die Reduktase vermittelte Redoxcycling über Generierung von ROS in der Lage ist, einen oxidativen DNA-Schaden und Toxizität zu verursachen. Hier zeigte sich, dass die Überexpression der Reduktase Voraussetzung für Toxizität und den beobachteten DNA-Schaden ist. Die Wildtyp-Zellen zeigten weder einen DNA-Schaden noch Zytotoxizität, auch eine zusätzliche Glutathion-Depletion änderte nichts an dem Befund. Die V79-hOR-Zellen hingegen reagierten auf die Inkubation mit Durochinon mit einer konzentrationsabhängigen Zunahme der Einzelstrangbrüche und oxidativen Basenmodifikationen, wobei sich der DNA-Schaden durch vorherige Glutathion-Depletion verdoppeln ließ.

Population genetic and phylogeography of two common mediterranean species were studied in 10 localities located on the coasts of Toscana, Puglia and Calabria. The aim of the study was to verify the extent of genetic breaks, in areas recognized as boundaries between Mediterranean biogeographic sectors. From about 100 sequences obtained from the mitochondrial Cytochrome Oxidase subunit I (COI) gene of Halocynthia papillosa and Hexaplex trunculus genetic diversity, genetic structure at small and large distances and demographic history of both specieswere analyzed. No evidences of genetic breaks were found for the two species in Toscana and Puglia. The genetic structure of H. trunculus evidences the extent of a barrier to gene flow localized in Calabria, which could be represented by the Siculo-Tunisian Strait and the Strait of Messina. The observed patterns showed similar level of gene flow at small distances in both species, although the two species have different larval ecology. These results suggest that other factors, such as currents, local dynamics and seasonal temperatures, influence the connectivity along the Italian peninsula. The geographic distribution of the haplotypes shows that H. papillosacould represent a single genetic pool in expansion, whereas H. trunculus has two distinct genetic pools in expansion. The demographic pattern of the two species suggests that Pleistocene sea level oscillations, in particular of the LGM, may have played a key role in shaping genetic structure of the two species. This knowledge provides basic information, useful for the definition of management plans, or for the design of a network of marine protected areas along the Italian peninsula.

The aim of this thesis was to apply the techniques of the atomic force microscope (AFM) to biological samples, namely lipid-based systems. To this end several systems with biological relevance based on self-assembly, such as a solid-supported membrane (SSM) based sensor for transport proteins, a bilayer of the natural lipid extract from an archaebacterium, and synaptic vesicles, were investigated by the AFM. For the characterization of transport proteins with SSM-sensors proteoliposomes are adsorbed that contain the analyte (transport protein). However the forces governing bilayer-bilayer interactions in solution should be repulsive under physiological conditions. I investigated the nature of the interaction forces with AFM force spectroscopy by mimicking the adsorbing proteoliposome with a cantilever tip, which was functionalized with charged alkane thiols. The nature of the interaction is indeed repulsive, but the lipid layers assemble in stacks on the SSM, which expose their unfavourable edges to the medium. I propose a model by which the proteoliposomes interact with these edges and fuse with the bilayer stacks, so forming a uniform layer on the SSM. Furthermore I characterized freestanding bilayers from a synthetic phospholipid with a phase transition at 41°C and from a natural lipid extract of the archaebacterium Methanococcus jannaschii. The synthetic lipid is in the gel-phase at room temperature and changes to the fluid phase when heated to 50°C. The bilayer of the lipid extract shows no phase transition when heated from room temperature to the growth temperature (~ 50°C) of the archeon. Synaptic vesicles are the containers of neurotransmitter in nerve cells and the synapsins are a family of extrinsic membrane proteins, that are associated with them, and believed to control the synaptic vesicle cycle. I used AFM imaging and force spectroscopy together with dynamic light scattering to investigate the influence of synapsin I on synaptic vesicles. To this end I used native, untreated synaptic vesicles and compared them to synapsin-depleted synaptic vesicles. Synapsin-depleted vesicles were larger in size and showed a higher tendency to aggregate compared to native vesicles, although their mechanical properties were alike. I also measured the aggregation kinetics of synaptic vesicles induced by synapsin I and found that the addition of synapsin I promotes a rapid aggregation of synaptic vesicles. The data indicate that synapsin I affects the stability and the aggregation state of synaptic vesicles, and confirm the physiological role of synapsins in the assembly and regulation of synaptic vesicle pools within nerve cells.

Le malattie trasmesse da zecche sono un importante problema sia per la salute animale che per quella umana e negli ultimi decenni hanno aumentato notevolmente la loro diffusione, in seguito ai cambiamenti climatici, che hanno permesso la distribuzione delle zecche in aree prima non interessate. Per tale motivo si è deciso di effettuare un’indagine sulla diffusione delle zecche e sui patogeni da loro trasmessi, mediante campionamenti sia a livello ambientale, sia su animali e umani infestati in quattro siti di tre parchi dell’Emilia Romagna, dove non risultavano precedenti segnalazioni, nelle province di Bologna e Ravenna, da Aprile a Ottobre 2010. In totale sono state raccolte 8212 zecche. Dall’ambiente sono state campionate 6734 larve, 1344 ninfe, 61 adulti; dagli animali e da persone sono stati raccolti 68 adulti e 5 ninfe appartenenti a diverse specie di Ixodidae. Sono state condotte analisi sull’abbondanza delle zecche nelle diverse aree di raccolta, in funzione del periodo di campionamento, della temperatura e dell’umidità relativa misurata a 5 cm dal suolo al momento del campionamento e della vegetazione. Su tutti gli individui adulti e su pool di ninfe e di larve, per un totale di 393 campioni, sono state condotte analisi di tipo molecolare per la ricerca di piroplasmi, Anaplasma phagocytophilum e Borrelia burgdorferi s.l. Attraverso la PCR e il sequenziamento, è emerso che il 7,6% dei campioni era positivo per piroplasmi, tra i quali è stata riscontrata anche la presenza delle specie zoonosiche Babesia EU1 e B. divergens. La real-time PCR eseguita solo sui campioni costituiti da ninfe e adulti ha evidenziato una prevalenza del 9,2% per A. phagocytophilum e del 21,6% per B. burgdorferi s.l. Su questi patogeni sono state quindi condotte analisi di tipo filogenetico. In alcuni campioni sono state riscontrate coinfezioni con combinazioni di due patogeni contemporaneamente.

The study aims at providing a framework conceptualizing patenting activities under the condition of intellectual property rights fragmentation. Such a framework has to deal with the interrelated problems of technological complexity in the modern patent landscape. In that respect, ex-post licensing agreements have been incorporated into the analysis. More precisely, by consolidating the right to use patents required for commercialization of a product, private market solutions, such as cross-licensing agreements and patent pools help firms to overcome problems triggered by the intellectual property rights fragmentation. Thereby, private bargaining between parties as such cannot be isolated from the legal framework. A result of this analysis is that policies ignoring market solutions and only focusing on static gains can mitigate the dynamic efficiency gains as induced by the patent system. The evidence found in this thesis supports the opinion that legal reforms that aim to decrease the degree of patent protection or to lift it all together can hamper the functioning of the current system.

In the present work, a multi physics simulation of an innovative safety system for light water nuclear reactor is performed, with the aim to increase the reliability of its main decay heat removal system. The system studied, denoted by the acronym PERSEO (in Pool Energy Removal System for Emergency Operation) is able to remove the decay power from the primary side of the light water nuclear reactor through a heat suppression pool. The experimental facility, located at SIET laboratories (PIACENZA), is an evolution of the Thermal Valve concept where the triggering valve is installed liquid side, on a line connecting two pools at the bottom. During the normal operation, the valve is closed, while in emergency conditions it opens, the heat exchanger is flooded with consequent heat transfer from the primary side to the pool side. In order to verify the correct system behavior during long term accidental transient, two main experimental PERSEO tests are analyzed. For this purpose, a coupling between the mono dimensional system code CATHARE, which reproduces the system scale behavior, with a three-dimensional CFD code NEPTUNE CFD, allowing a full investigation of the pools and the injector, is implemented. The coupling between the two codes is realized through the boundary conditions. In a first analysis, the facility is simulated by the system code CATHARE V2.5 to validate the results with the experimental data. The comparison of the numerical results obtained shows a different void distribution during the boiling conditions inside the heat suppression pool for the two cases of single nodalization and three volume nodalization scheme of the pool. Finaly, to improve the investigation capability of the void distribution inside the pool and the temperature stratification phenomena below the injector, a two and three dimensional CFD models with a simplified geometry of the system are adopted.

Salmonella and Campylobacter are common causes of human gastroenteritis. Their epidemiology is complex and a multi-tiered approach to control is needed, taking into account the different reservoirs, pathways and risk factors. In this thesis, trends in human gastroenteritis and food-borne outbreak notifications in Italy were explored. Moreover, the improved sensitivity of two recently-implemented regional surveillance systems in Lombardy and Piedmont was evidenced, providing a basis for improving notification at the national level. Trends in human Salmonella serovars were explored: serovars Enteritidis and Infantis decreased, Typhimurium remained stable and 4,[5],12:i:-, Derby and Napoli increased, suggesting that sources of infection have changed over time. Attribution analysis identified pigs as the main source of human salmonellosis in Italy, accounting for 43–60% of infections, followed by Gallus gallus (18–34%). Attributions to pigs and Gallus gallus showed increasing and decreasing trends, respectively. Potential bias and sampling issues related to the use of non-local/non-recent multilocus sequence typing (MLST) data in Campylobacter jejuni/coli source attribution using the Asymmetric Island (AI) model were investigated. As MLST data become increasingly dissimilar with increasing geographical/temporal distance, attributions to sources not sampled close to human cases can be underestimated. A combined case-control and source attribution analysis was developed to investigate risk factors for human Campylobacter jejuni/coli infection of chicken, ruminant, environmental, pet and exotic origin in The Netherlands. Most infections (~87%) were attributed to chicken and cattle. Individuals infected from different reservoirs had different associated risk factors: chicken consumption increased the risk for chicken-attributed infections; animal contact, barbecuing, tripe consumption, and never/seldom chicken consumption increased that for ruminant-attributed infections; game consumption and attending swimming pools increased that for environment-attributed infections; and dog ownership increased that for environment- and pet-attributed infections. Person-to-person contacts around holiday periods were risk factors for infections with exotic strains, putatively introduced by returning travellers.

Das humane Enzym PON2 ist in eine Vielzahl pathophysiologischer Prozesse involviert und ist durch zwei Funktionen gekennzeichnet - eine enzymatische Laktonase-Aktivität und eine anti-oxidative Aktivität. Durch die Laktonase-Aktivität hydrolysiert PON2 vorwiegend das bakterielle Signalmolekül 3oxoC12. PON2 ist als Bestandteil des angeborenen Immunsystems anzusehen und trägt wahrscheinlich zur Immunabwehr gegen Infektionen mit den human-pathogenen Pseudomonas aeruginosa Bakterien bei. Durch die anti-oxidative Aktivität vermindert PON2 oxidative Schäden und verringert redox-abhängige pro-apoptotische Stimulation. Diese einzigartige Funktion von PON2 ist jedoch ambivalent zu betrachten, da hohe PON2-Spiegel zwar Arteriosklerose reduzieren können, aber im Verdacht stehen Tumorzellen zu stabilisieren.rnIn dieser Arbeit wurden die noch unbekannten Mechanismen und der Zusammenhang der enzymatischen und der anti-oxidativen Aktivität analysiert. In diesem Rahmen wurde gezeigt, dass PON2 spezifisch die Superoxidfreisetzung an Komplex I und III der Atmungskette in der inneren Mitochondrienmembran reduzieren kann. PON2 veränderte dabei weder die Aktivitäten der Superoxiddismutasen noch die Cytochrom C-Expression. Weiterhin konnte in dieser Arbeit erstmals gezeigt werden, dass PON2 O2- nicht direkt abbaut, sondern vielmehr dessen Bildung verhindert. Diese Erkenntnisse implizieren, dass PON2 die anti-oxidative Aktivität über eine Beeinflussung des Quinon-Pools vermittelt. Anhand von verschiedenen Punktmutationen konnte gezeigt werden, dass die Histidinreste-114 und -133 für die Laktonase-Aktivität essentiell sind. Weiterhin wurden die Glykosylierungsstellen von PON2 identifiziert und gezeigt, dass die Glykosylierung, nicht aber der natürliche Polymorphismus Ser/Cys311 für die Laktonase-Aktivität von Bedeutung ist. Von besonderer Bedeutung ist, dass keine dieser Mutationen die anti-oxidative Aktivität beeinflusste, wodurch erstmals die Unabhängigkeit der beiden Funktionen von PON2 gezeigt werden konnte. rnEs war bekannt, dass PON2 gegen intrinsische und ER-Stress-induzierte Apoptose schützt. Die Spezifität der anti-oxidativen / anti-apoptotischen Wirkung wurde hier an einem weiteren pathophysiologischen Modell untersucht. 7-Ketocholesterol (7-KC) ist der Hauptbestandteil des pro-arteriosklerotischen oxLDL und verursacht in Zellen des Gefäßsystems ER-Stress, oxidativen Stress und Apoptose. Unerwarteterweise konnte PON2 Endothelzellen nicht gegen den 7-KC-induzierten Zelltod schützen. Mehrere unabhängige experimentelle Ansätze belegen, dass 7-KC in Endothelzellen im Gegensatz zu Gefäßmuskelzellen den Zelltod über Autophagie und nicht über ER-Stress oder intrinsische Apoptose bewirkt. Weiterhin führt 7-KC, wie auch 3oxoC12 und Thapsigargin zu einem Abbau der PON2-mRNA, die über die 5’UTR der PON2-mRNA vermittelt wird. Diese Arbeit vermittelt detaillierte mechanistische Einsichten in die Funktionen von PON2, die für ihre Rolle bei Arteriosklerose, in der körpereigenen Immunabwehr und bei Krebs entscheidend sind.rn

The t(8;21) (q22;q22) translocation fusing the ETO (also known as MTG8) gene on human chromosome 8 with the AML1 (also called Runx1 or CBFα) gene on chromosome 21 is one of the most common genetic aberrations found in acute myeloid leukemia (AML). This chromosomal translocation occurs in 12 % of de novo AML cases and in up to 40 % of the AML-M2 subtype of the French-American-British classification. To date, the in vivo function of aberrant AML1-ETO fusion protein expression has been investigated by several groups. However, in these studies, controversial results were reported and some key issues remain unknown. Importantly, the consequences of aberrant AML1-ETO expression for self-renewing hematopoietic stem cells (HSCs), multipotent hematopoietic progenitors (MPPs) and lineage-restricted precursors are not known. rn The aim of this thesis was to develop a novel experimental AML1-ETO in vivo model that (i) overcomes the current lack of insight into the pre-leukemic condition of t(8;21)-associated AML, (ii) clarifies the in vivo consequences of AML1-ETO for HSCs, MPPs, progenitors and more mature blood cells and (iii) generates an improved mouse model suitable for mirroring the human condition. For this purpose, a conditional tet on/off mouse model expressing the AML1-ETO fusion protein from the ROSA26 (R26) locus was generated. rn Aberrant AML1-ETO activation in compound ROSA26/tetOAML1-ETO (R26/AE) mice caused high rates of mortality, an overall disruption of hematopoietic organs and a profound alteration of hematopoiesis. However, since the generalized activity of the R26 locus did not recapitulate the leukemic condition found in human patients, it was important to restrict AML1-ETO expression to blood cell lineages. Therefore, bone marrow cells from non-induced R26/AE mice were adoptively transplanted into sublethal irradiated RAG2-/- recipient mice. First signs of phenotypical differences between AML1-ETO-expressing and control mice were observed after eight to nine months of transgene induction. AML1-ETO-expressing mice showed profound changes in hematopoietic organs accompanied by manifest extramedullary hematopoiesis. In addition, a block in early erythropoiesis, B- and T-cell maturation was observed and granulopoiesis was significantly enhanced. Most interestingly, conditional activation of AML1-ETO in chimeric mice did not increase HSCs, MPPs, common lymphoid precursors (CLPs), common myeloid progenitors (CMPs) and megakaryocyte-erythrocyte progenitors (MEPs) but promoted the selective amplification of granulocyte-macrophage progenitors (GMPs). rn The results of this thesis provide clear experimental evidence how aberrant AML1-ETO modulates the developmental properties of normal hematopoiesis and establishes for the first time that AML1-ETO does not increase HSCs, MPPs and common lineage-restricted progenitor pools but specifically amplifies GMPs. The here presented mouse model not only clarifies the role of aberrant AML1-ETO for shaping hematopoietic development but in addition has strong implications for future therapeutic strategies and will be an excellent pre-clinical tool for developing and testing new approaches to treat and eventually cure AML.rn
