Hidrolases englobam um grupo de enzimas que catalisam a quebra de ligações covalentes em reação com água; entre elas estão as proteases, amilases, lipases, pectinases, celulases e catalases. Essas enzimas são muito importantes, com ampla utilização na indústria em geral. O solo é um ambiente muito rico e diverso em microrganismos, sendo considerado a maior fonte para obtenção de substâncias, enzimas e antibióticos, por exemplo. Com a metagenômica, passou a ser possível acessar melhor esse potencial microbiano, permitindo a descoberta de novos genes e biomoléculas. Neste estudo foram coletadas amostras de solos (0-10 cm de profundidade) do norte do Paraná visando buscar hidrolases microbianas funcionais. Foi realizada a extração do DNA de um Latossolo Vermelho Eutroférrico sob quatro manejos de solo e de culturas distintos e as amostras foram submetidas ao sequenciamento utilizando a plataforma 454 (Applied Science). As sequências de DNA foram comparadas com o banco de dados não redundante (NR) do NCBI (National Center for Biotechnology Information) e KEGG (Kyoto Encyclopedia of Genes and Genomes) para busca de similaridade com proteases, amilases, lipases, pectinases, celulases e catalases. A partir do DNA total foram realizadas reações de PCR (Polymerase Chain Reaction) com primers degenerados direcionados para a amplificação de pectinases, celulases e lacases e os produtos de PCR foram purificados com Purelink kit (Invitrogen®) e sequenciados (ABI 3500xL, Aplied Biosystems®). A comparação com as sequências do NCBI e KEGG resultou na identificação de 1.137 sequências com grande similaridade com a enzima lacase; 16.883 sequências para celulase; 2.001 para pectinase; 1.006 para amilase; e 3.725 para lipase. Esses resultados mostram que esses solos agrícolas representam uma fonte importante de recursos biológicos para aplicação industrial, principalmente de enzimas celulases. Até o presente momento, o sequenciamento de 26 produtos amplificados por PCR apresentou identidade para uma amostra, que foi identificada como a enzima celulase.

O Azoto (N): da ciência para a sociedade é um projecto de comunicação de ciência que tem por objecNvo consciencializar os jovens para as ameaças que o azoto (N) em excesso traz para a humanidade. Pode ser dividido em duas partes. Uma, de invesNgação, sobre a análise de resultados de uma consulta pública realizada entre professores, usando o método qualitaNvo do focus group, para compreender a sua sensibilidade e propostas de solução para minimizar o excesso de N no ambiente. Os resultados obNdos foram instrumentais para o desenvolvimento da segunda parte. Esta segunda parte é uma proposta de projecto a submeter ao Horizon 2020, no âm-­‐ bito da “Science with and for Society “. Nela se propõe uma abordagem educaNva trans-­‐disciplinar, conseguida através da interacção entre docentes do secundário, e do ensino superior, associação de pais e organizações cívicas não governamentais, com vista à consciencialização dos jovens para as ameaças do N em excesso no meio ambiente, fazendo o enquadramento cien@fico e fornecendo abordagens tecnológi-­‐ cas. A inovação desta proposta baseia-­‐se: (i) no acompanhamento e desenvolvimen-­‐ to profissional dos docentes do secundário, (ii) na moNvação dos estudantes a de-­‐ senvolver o seu próprio estudo e pesquisa com a tutoria dos docentes, da escola e do ensino superior, e (iii) no desenvolvimento de capacidades de comunicação dos jo-­‐ vens para exercer uma cidadania acNva em prol da minimização das ameaças do N.

Leptospirosis is a zoonotic disease that has emerged to cause epidemics in urban communities in developing countries. However, little is known about the infection in the general population. A seroprevalence survey was performed on a random sample of 1,390 subjects in Salvador, Brazil. Data on environmental and socioeconomic factors were collected. The microagglutination test of serum samples was used to show any prior Leptospira infection. The overall seroprevalence was 12.4%. Among the seropositive individuals, 111 (61%) had high titers for serovars of the Icterohaemorrhagiae serogroup. Seroprevalence increased with age and was similar for males and females. A positive correlation between Leptospira infection and low educational level was found. These findings indicate that a significant proportion of this urban population is exposed to pathogenic Leptospira. Public health actions for leptospirosis control will need to target not only the traditional groups at risk of infection with severe forms of this disease, but also the general population that is at risk.

The aim of the present study was to outline the serological profile of hepatitis C among blood donors seen at the Uberaba Regional Blood Center, Hemominas Foundation, over the last 14 years. The frequency of hepatitis C was compared between first-time and repeat donors and the epidemiological characteristics of those with positive and indeterminate ELISA anti-HCV (third and fourth generation) were analyzed based on the donor histories kept in the archives of the Uberaba Regional Blood Center. The serological ineligibility rate was 0.3%, with higher prevalence in the group of first-time donors. We did not find any significant differences regarding age, skin color, marital status or place of residence between eligible and ineligible donors; however, the frequency of positive serology was higher among men. The lower (0.3%) rate of ineligibility due to hepatitis C that was observed at the Uberaba Regional Blood Center, in relation to most Brazilian blood centers, is probably due to the large number of repeat donors (83.3%). This reinforces the importance of achieving donor commitment for increasing transfusion safety.

O estágio que aqui se relata enquadra-se no âmbito do mestrado em Comunicação de Ciência, da Faculdade de Ciências Sociais Humanas (FCSH) da Universidade Nova de Lisboa (UNL) e do Instituto de Tecnologia Química e Biológica (ITQB). Foi realizado no projeto STOL – Science Through Our Lives, do Centro de Biologia Molecular e Ambiental (CBMA) da Escola de Ciências da Universidade do Minho (ECUM), e teve a duração de três meses. Esta componente prática do mestrado permitiu desenvolver um trabalho in loco, com o objetivo de servir, sobretudo, a comunicação de ciência informal. As principais atividades desenvolvidas foram exposições (Homo numericus; “Ponto a Ponto Enche a Ciência o Espaço), produção de materiais educativos (Objetos Educativos para a Casa das Ciências), campanhas de sensibilização (Comemoração do Dia Mundial da Água), auscultação do público (Vox Pop), crónicas de jornal (coluna “Aqui há Ciência”) e estabelecimento de parcerias estratégicas entre o STOL e outras forças vivas da região (AVianense e “Diário do Minho”). Ao integrar as atividades a decorrer no projeto STOL, foi possível contribuir com sugestões e novos materiais, e implementar projetos cujo principal objetivo foi o contacto direto com o público. De todo o trabalho se obteve um feedback positivo, conforme se documenta neste relatório e respetivos anexos. Se alguns constrangimentos podem ser enunciados (falta de recursos económicos e humanos, fraca tradição de envolvimento da população com a ciência e vice-versa, e a considerável falta de apoio institucional a atividades de divulgação de ciência), também devem ser evidenciados aspetos muito positivos como a dinâmica do grupo STOL, a capacidade de envolver diferentes públicos nas suas atividades, diferenciar estratégias para esses grupos, e finalmente, avaliar, sempre que possível, o seu desempenho, incluindo os resultados nas atividades seguintes. Neste contexto foram aplicados os conhecimentos adquiridos durante o primeiro ano curricular do mestrado, permitindo o ganho de experiência na área de trabalho onde a autora se sente realizada e pretende permanecer.

The project of writing to a fictional Portuguese-speaking Queen on the crises proceeding since 2008 builds on Letters to Queen Elizabeth written by the British Academy and was first published in 2013. This expanded edition signals greater awareness of the complementarity between economic potential and cultural legacy in the Community of Portuguese-speaking Countries (CPLP) insofar as its members, observers and their areas of economic integration encompass the globe. The edition is dedicated to the memory of Manuel Jacinto Nunes, who supported the project as dean of the economics section at the Lisbon Academy of Science. The cover shows a Crown with nine CPLP flags as jewels in the shape of a 7 rising from the waves. The waves of lusophonia appear far gentler than Poe’s maelstrom, reproduced in the back flap. This material, inserted in the proceedings published on IICT’s 130th anniversary, is used at NOVASBE through its Center for Globalization and Governance (CG&G).

INTRODUCTION: Zoonotic kala-azar, a lethal disease caused by protozoa of the genus Leishmania is considered out of control in parts of the world, particularly in Brazil, where transmission has spread to cities throughout most of the territory and mortality presents an increasing trend. Although a highly debatable measure, the Brazilian government regularly culls seropositive dogs to control the disease. Since control is failing, critical analysis concerning the actions focused on the canine reservoir was conducted. METHODS: In a review of the literature, a historical perspective focusing mainly on comparisons between the successful Chinese and Soviet strategies and the Brazilian approach is presented. In addition, analyses of the principal studies regarding the role of dogs as risk factors to humans and of the main intervention studies regarding the efficacy of the dog killing strategy were undertaken. Brazilian political reaction to a recently published systematic review that concluded that the dog culling program lacked efficiency and its effect on public policy were also reviewed. RESULTS: No firm evidence of the risk conferred by the presence of dogs to humans was verified; on the contrary, a lack of scientific support for the policy of killing dogs was confirmed. A bias for distorting scientific data towards maintaining the policy of culling animals was observed. CONCLUSIONS: Since there is no evidence that dog culling diminishes visceral leishmaniasis transmission, it should be abandoned as a control measure. Ethical considerations have been raised regarding distorting scientific results and the killing of animals despite minimal or absent scientific evidence

INTRODUCTION: To describe the clinical and epidemiological profile of pregnant women and children treated at a reference outpatient clinic for congenital toxoplasmosis. METHODS: Pregnant women potentially exposed to Toxoplasma gondii were observed. Diagnoses were made using serologic tests compatible with acute toxoplasmosis. Children presenting with: Toxoplasma-specific antibodies (IgM or IgA or ascending IgG titers higher than maternal titers in the first 3 months of life) coupled with toxoplasmosis symptoms; intracranial calcifications (by transfontanelar ultrasound or cephalic segment tomography); or retinochoroiditis (by fundoscopy examination) in the first 8 months of life were also included in the study. RESULTS: Fifty-eight mother-child pairs were observed (mean age of the mothers was 22.1 years). Most patients lived in urban areas (86.2%) and had attended less than 8 years of school (51.7%). Diagnosis was made after birth in 19 (32.8%) children. Thirty-four (58.6%) women received some type of treatment during pregnancy. Most (72.4%) of the children did not present with clinical alterations at birth. The main findings were ophthalmological: 20 (34.5%) children with retinochoroiditis, 17 (29.3%) with strabismus, and 7 (12.1%) with nystagmus. Of the children with retinochoroiditis, 9 presented with subnormal vision. Ten (32.3%) out of 31 children presented with intracranial calcifications by cephalic segment congenital toxoplasmosis, and 9 (42.9%) children presented with delayed psychomotor development. CONCLUSIONS: Our results highlight a critical situation. Protocols for follow-up of pregnant women and their children must be created to improve medical care and minimize sequelae.

INTRODUCTION: A retrospective study was conducted to assess the occurrence of blood donations that were ineligible due to Chagas disease infection from 1995 to 2009 at the Uberaba Regional Blood Center (HRU), Brazil, verify the tendency of this ineligibility, and describe the epidemiologic profile of the donors. METHODS: Retrospective studies of serological ineligibility due to Chagas disease, statistical analysis by means of the chi-square test and odds ratio, study of the tendencies using a dispersion graph and the linear correlation coefficient (r) were performed. RESULTS: In the period under study, a 0.2% serum prevalence of ineligibility due to Chagas disease was found, with a significant drop in ineligible donations from 2001 to 2009. Among the serum positive-donors, there was a significant predominance among those aged 30 years or above and non-single individuals. CONCLUSIONS: The results show a rate of occurrence that is lower than that described in literature, as well as a progressive drop during the 15 years under assessment. Such results are a consequence of systematic combat of the vector since the 70s and the progressive and consistent increase of returning donors, resulting in a drop of the contamination risk factor by means of blood transfusion and in the improvement of the quality of hemotherapy practices in the HRU.
