Salmonella Pullorum and Salmonella Gallinarum are classified as biovars of Salmonella enterica subsp. enterica serovar Gallinarum. These salmonellae are the causative agents of Pullorum disease and fowl typhoid, respectively, and are widely distributed throughout the world. Although many developed countries have eradicated these diseases from commercial poultry, they are still the cause of significant economic loss in developing countries. When serovar Gallinarum is isolated, it is difficult to immediately differentiate between biovars because they are antigenically identical by serotyping. However, they cause distinct diseases with different epidemiology, and therefore it is important to differentiate them. This may be done biochemically but takes 2 to 3 days. In the present study, S. Pullorum and S. Gallinarum whole genomes were compared, and 1 genomic region of difference, which is part of the ratA gene, was chosen as a molecular marker for a polymerase chain reaction assay to differentiate rapidly between these organisms. In all, 26 strains of S. Gallinarum and 17 S. Pullorum strains were tested and successfully differentiated by the assay. © 2013 The Author(s).

Calcifying cystic odontogenic tumors (CCOTs) are benign cystic lesions of odontogenic origin characterized by an ameloblastoma-like epithelium and the presence of a group of cells named ghost cells. The pattern of cytokeratin (Ck) expression on these lesions remains unclear and needs to be clarified. To this end, the expression of Ck6, Ck13, Ck14, Ck18, and Ck19 in the epithelium lining of 7 cases of CCOTs was evaluated by immunohistochemistry. For this, the epithelium lining was divided into 3 distinct regions: basal layer, suprabasal layer, and the compartment composed of ghost cells. In this study, 6 cases (85.7%) were classified as type 1 and 1 (14.3%) as type 4. All cases were negative for Ck13 and Ck18, despite the epithelial layer, as well as in the ghost cells. Ck6 was only positive in the ghost cells. Positivity for Ck14 and Ck19 was found in the basal and suprabasal layers, including the ghost cells. The results showing positivity for Ck14 and Ck19 in all of the analyzed cases reinforce CCOT as being of odontogenic origin, and the restricted expression of Ck6 in the ghost cells may be indicative that these cells suffer an altered differentiation into hair follicles in CCOTs. © 2013 Elsevier Inc. The king weakfish (pescada-gó in Portuguese - Macrodon ancylodon (Sciaenidae), a demersal (bottom-feeding) species found in South America Atlantic coastal waters from the Gulf of Paria in Venezuela to Baia Blanca in Argentina, is an economically important species because of its abundance and wide acceptance by consumers. Because of its wide distribution this fish may be subject to geographic isolation and this may have resulted in distinct populations along its coastal range. Considering that this species represents an important economic resource, confirmation of whether M. ancylodon is a single species or there are different genetic stocks spread over its wide distribution would be an important contribution to conservation policies and population management of the king weakfish. To investigate differences between king weakfish populations we used the cytochrome b and 16S rRNA genes to characterize M. ancylodon specimens caught throughout its South American range from Venezuela to Argentina. Our results clearly distinguished two genetically different groups which show nucleotide divergence and genetic structuring patterns that strongly suggest they may be different species, disagreeing with the widely accepted traditional taxonomy that accepts only one species of Macrodon in the western Atlantic.

Four Brazilian populations of Anomalocardia brasiliana were tested for mutual genetic homogeneity, using data from 123 sequences of the mtDNA cytochrome oxidase c subunit I gene. A total of 36 haplotypes were identified, those shared being H3 (Canela Island, Prainha and Acupe) and both H5 and H9 (Prainha and Acupe). Haplotype diversity values were high, except for the Camurupim population, whereas nucleotide values were low in all the populations, except for that of Acupe. Only the Prainha population showed a deviation from neutrality and the SSD test did not reject the demographic expansion hypothesis. Fst values showed that the Prainha and Acupe populations represent a single stock, whereas in both the Canela Island and Camurupim stocks, population structures are different and independent. The observed structure at Canela Island may be due to the geographic distance between this population and the remainder. The Camurupim population does not share any haplotype with the remaining populations in northeastern Brazil. The apparent isolation could be due to the rocky barrier located facing the mouth of the Mamanguape River. The results highlight the importance of wide-scale studies to identify and conserve local genetic diversity, especially where migration is restricted.

O mapeamento geológico realizado à sudeste de Água Azul do Norte/PA, porção norte do Domínio Rio Maria, aliado aos dados petrográficos e geoquímicos permitiram a individualização de associações TTGs e leucogranodioritos. Nesta região, os trabalhos de mapeamento foram realizados apenas em escala regional o que possibilitou a extrapolação da área de ocorrência de rochas similares ao Tonalito Caracol e rondhjemito Mogno. Os TTGs estudados foram individualizados em duas unidades com base no conteúdo de minerais máficos, concentrações de epidoto magmático e no grau de saussuritização (descalcificação) do plagioclásio em: (1) Epidoto-Biotita Tonalito e; (2) Biotita Trondhjemito. Em geral, são rochas que apresentam foliação definida pelo bandamento composicional, localmente pode ser perturbada por dobras e bandas de cisalhamento. Suas características geoquímicas são compatíveis com os TTGs arqueanos do grupo de alto Al2O3, sendo ainda relativamente pobres em elementos ferromagnesianos, com padrões ETRP moderado a fortemente fracionados e anomalias de Eu discretas. As diferenças nas razões La/Yb e anomalia de Eu, possibilitou a discriminação de três grupos distintos de rochas: Os TTGs pertencentes ao grupo de alto La/Yb e Sr/Y são similares às rochas do Trondhjemito Mogno, descritos no Domínio Rio Maria. Estas rochas incluem a maioria das amostras da unidade Biotita Trondhjemito. No caso dos TTGs com médio a baixo La/Yb e Sr/Y quando comparadas com as rochas do Domínio Rio Maria possuem forte correlação com o Tonalito Caracol. Estes grupos são compostos principalmente pela unidade Epidoto-Biotita Tonalito, incluindo também amostras isoladas do Biotita Trondhjemito. Com base nos critérios utilizados acima, os leucogranodioritos da área foram divididos em dois grupos: Biotita Granodiorito e Leucogranodiorito. As rochas do Biotita Granodiorito possuem ampla ocorrência espacial na porção oeste da área, relações de campo mostram que são intrusivas nos granitoides TTGs. Os dados geoquímicos apontam que o Biotita Granodiorito possui padrões de ETR fortemente fracionados, com alta razão La/Yb (33 – 186) e anomalia de Eu positiva (1,11 < Eu/Eu* < 3,26), enquanto os leucogranodioritos mostram padrões levemente fracionados, com moderadas razões La/Yb (24,7 – 34,7) e anomalia de Eu ausente (Eu/Eu*= 1,03). Os diagramas de Harker para elementos maiores e traços não favorecem uma ligação genética por processo de cristalização fracionada entre o Biotita Granodiorito e as associações TTGs, uma vez que apresentam trends de evolução distintos, indicando portanto que as condições de sua gênese e diferenciação foram bem diferentes, tampouco por fusão parcial de uma fonte TTG, pelo fato de não apresentar significante anomalia negativa de Eu, bem como por exibir padrões similares de fracionamento de ETR em relação aos TTGs, atestando que essas rochas provavelmente não foram oriundas de magmas precursores desses TTGs.

Currently, much attention has been devoted to the renewal of knowledge about Stem Cells and Cell Therapy in domestic species. In this sense, the present work aimed to develop a methodology for collecting, processing and cultivation of mesenchymal stem cells obtained from bone marrow of coxal tuberosity in buffaloes. The collection was performed using a Komiyashiki needle, which was introduced in the coxal tuberosity and the bone marrow aspirated into a heparinized syringe with the aid of negative pressure. Directly after collection samples were processed at the laboratory at FMVZ - UNESP. The samples took approximately 32 days to reach 80% confluence, when the first passage and differentiation was performed. To confirm the mesenchymal origin, cells were induced to differentiate into adipogenic and osteogenic lineages. Samples showed morphological changes during differentiation protocol, but not all presented production of extracellular deposits of calcium or intracellular fat droplets, observed after staining with Alizarin Red and Oil Red respectively. Compared with the material obtained from other species and processed in the same laboratory, the primary culture was longer. Therefore, more studies are needed to standardize the age of animals used and to test other inducers of cell differentiation.
