One hundred and twenty two strains of Staphylococcus aureus isolated from throats and palms of 39 workers from 6 fish processing factories situated in and around Cochin were tested for their sensitivity to nine commonly used antibiotics-ampicillin, chloramphenicol, erythromycin, kanamycin, neomycin, penicillin, polymyxin-B, streptomycin and tetracycline. Highest percentage of resistance was observed towards ampicillin followed by penicillin i.e. 64.75% and 59.84%. Resistance towards other antibiotics like tetracycline, polymyxin-B, erythromycin, kanamycin, neomycin, chloramphenicol and streptomycin were shown by 22.95, 16.39, 7.38, 5.74, 3.28 and 1.64% of the isolates respectively.

The authors report on inoculation experiments of Artemia nauplii and young adults of the San Francisco Bay strains in earthen fish ponds. The test inoculated proved successful where water salinity ranges from 20 to 32 o/oo during the start of the rainy season in the Philippines.

An experiment of 120 days of culture was conducted in brackishwater earthen ponds having an area of 0.2ha each. The hatchery produced shrimp (Penaeus monodon) post larvae were stocked in the 40m² fine meshed nylon net nursery enclosures were fed with commercial pellet feed. After two weeks of nursing, juveniles were allowed to spread in cultural pond by opening the fence. Fingerlings of three different strain of tilapia were stocked as shrimp and Strain-1 all male (monosex) (T1), shrimp and Strain-2 all male (T2), shrimp and Strain-3 mixed sex population (T3) @ 20.000/ha and 10.000/ha, respectively and shrimp only (monoculture) (T4) @ 20.000/ha. The shrimp and fish were fed with farm made feed consisting of a mixture of fishmeal 29%, MOC 15%, rice bran 30%, soybean meal 16%, wheat flour 9% and vitamin premix 0.1%. The average final weight of shrimp was 24.9±1.13g, 23.41±3.26g and 26.67±1.89g that stocked with tilapia in treatments T1, T2, and T3 respectively. The final average weight of shrimp in monoculture (T4) was 27.41±0.76g, apparently higher but insignificant in treatments. The survival of shrimp was 42.17%, 32.38%, 39.45% and 61.98% in treatments T1 T2, T3 and T4 respectively. The production of shrimp in concurrent culture was 193.67, 154.26 and 210.41kg/ha in T1, T2 and T3, respectively, while in monoculture (T4) was 339.77 kg/ha. The growth and survival of tilapia among the treatments was insignificant. The growth of monosex tilapia ranged 225.29 and 291.31g and survival 62.77 and 72.20% in T1 and T2, respectively, in mixed sex was 193.0g and 83.20% (T3). The production of tilapia monosex strains was 1676.69kg/ha (Strain-2 all male) and 1668.98 kg/ha (Strain-1 all male) while that of Strain-3 mixed sex population was 1622.92 kg/ha.

Iron is required for many microbes and pathogens for their survival and proliferation including Leishmania which cause leishmaniasis. Leishmaniasis is an increasingly serious infectious disease with a wide spectrum of clinical manifestations. These range from localized cutaneous leishmaniasis (CL) lesions to a lethal visceral form. Certain strains such as BALB/c mice fail to control L. major infection and develop progressive lesions and systemic disease. These mice are thought to be a model of non-healing forms of the human disease such as kala-azar or diffuse cutaneous leishmaniasis. Progression of disease in BALB/c mice has been associated with the anemia, in last days of their survival, the progressive anemia is considered to be one of the reasons of their death. Ferroportin (Fpn), a key regulator of iron homeostasis is a conserved membrane protein that exports iron across the duodenal enterocytes as well as macrophages and hepatocytes into the blood circulation. Fpn has also critical influence on survival and proliferation of many microorganisms whose growth is dependent upon iron, thus preparation of Fpn is needed to study the role of iron in immune responses and pathogenesis of micoorganisms. To prepare and characterize a recombinant ferroportin, total RNA was extracted from Indian zebrafish duodenum, and used to synthesize cDNA by RT-PCR. PCR product was first cloned in Topo TA vector and then subcloned into the GFP expression vector pEGFP–N1. The final resulted plasmid (pEGFP-ZFpn) was used for expression of FPN-EGFP protein in Hek 293T cells. The expression was confirmed by fluorescence microscopy and flow cytometery. Recombinant Fpn was further characterized by submission of its predicted amino acid sequences to the TMHMM V2.0 prediction server (hidden Markov model), NetOGlyc 3.1 server and NetNGlyc 3.1 server. Data emphasised that obtained Fpn from indian zebrafish contained eight transmembrane domains with N- and C-termini inside the cytoplasm and harboured 78 mucin-type glycosylated amino acid. The results indicate that the prepared and characterized recombinant Fpn protein has no membrane topology difference compared to other Fpn described by other researcher. Our next aim was to deliver recombinant plasmid (pEGFP-ZFpn) to entrocyte cells. However, naked therapeutic genes are rapidly degraded by nucleases, showing poor cellular uptake, nonspecificity to the target cells, and low transfection efficiency. The development of safe and efficient gene carriers is one of the prerequisites for the success of gene therapy. Chitosan and alginate 139 polymers were used for oral gene carrier because of their biodegradability, biocompatibility and their mucoadhesive and permeability-enhancing properties in the gut. Nanoparticles comprising Alginate/Chitosan polymers were prepared by pregel preparation method. The resulting nanoparticles had a loading efficiency of 95% and average size of 188 nm as confirmed by PCS method and SEM images had showed spherical particles. BALB/c mice were divided to three groups. The first and second group were fed with chitosan/alginate nanoparticles containing the pEGFP-ZFpn and pEGFP plasmid, respectively (30 μgr/mice) and the third group (control) didn’t get any nanoparticles. The result showed BALB/c mice infected by L.major, resulted in higher hematocryte and iron level in pEGFP-ZFpn fed mice than that in other groups. Consentration of cytokines determined by ELISA showed lower levels of IL-4 and IL-10 and higher levels of IFN-γ/IL-4 and IFN-γ/IL-10 ratios in pEGFP-ZFpn fed mice than that in other groups. Morover more limited increase of footpad thickness and significant reduction of viable parasites in lymph node was seen in pEGFP-ZFpn fed mice. The results showed the first group exhibited a highr hematocryte and iron compared to the other groups. These data strongly suggests the in vivo administration of chitosan/alginate nanoparticles containing pEGFP-ZFpn suppress Th2 response and may be used to control the leishmaniasis .

Genetic biodiversity is the vaflatlOn among individuals within and between units of interbreeding individuals (populations) of a species. It includes inheritable and transmittable differences that occur between individuals andlor popuhitions of a given species through reproductive interaction. There exists enormous variability among individuals andlor populations of a species for most living organisms, and most of this variation is inheritable. differences among individuals arise through mutation and via recombination of genes during meiosis. These ifferences are then transmitted to successive generations through sexual reproduction and maintained in the populations through processes such as natural selection and genetic drift. Unfortunately much of this variation is normally threatened and often in danger of extinction because most focus in conservation of natural resources is put at saving species or habitats than varieties or strains of a species

The sequences of the 16S rRNA genes from 38 strains of the family Thermaceae were compared by alignment analysis. The genus-specific and species-specific base substitutions or base deletions (signature positions) were found in three hypervariable regions (in the helices 6, 10 and 17). The differentiation of secondary structures of the high variable regions in the 5' end (38-497) containing several signature positions further supported the concept. Based on the comparisons of the secondary structures in the segments of 16S rRNAs, a key to the species of the family Thermaceae was proposed. We identified a new class of human immunodeficiency virus type 1 (HIV-1) recombinants (00CN-HH069 and 00CN-HH086) in which further recombination occurred between two established circulating recombinant forms (CRFs). These two isolates were found among 57 HIV-1 samples from a cohort of injecting drug users in eastern Yunnan Province of China. Informative-site analysis in conjunction with bootscanning plots and exploratory tree analysis revealed that these two strains were closely related mosaics comprised of CRF07_BC and CRF08_BC, which are found in China. The genotype screening based on gag-reverse transcriptase sequences if 57 samples from eastern Yunnan identified 47 CRF08_BC specimens (82.5%), 5 CRF07_BC specimens (8.8%), and 3 additional specimens with the novel recombinant structure. These new "second-generation" recombinants thus constitute a substantial proportion (5 of 57; 8.8%) of HIV-1 strains in this population and may belong to a new but yet-undefined class of CRF. This might be the first example of CRFs recombining with each other, leading to the evolution of second-generation inter-CRF recombinants.

The chemokine receptor CCR5 is the receptor for several chemokines and major coreceptor for R5 human immunodeficiency virus type-1 strains entry into cell. Three-dimensional models of CCR5 were built by using homology modeling approach and 1 ns molecular dynamics (MD) simulation, because studies of site-directed mutagenesis and chimeric receptors have indicated that the N-terminus (Nt) and extracellular loops (ECLs) of CCR5 are important for ligands binding and viral fusion and entry, special attention was focused on disulfide bond function, conformational flexibility, hydrogen bonding, electrostatic interactions, and solvent-accessible surface area of Nt and ECLs of this protein part. We found that the extracellular segments of CCR5 formed a well-packet globular domain with complex interactions occurred between them in a majority of time of MID simulation, but Nt region could protrude from this domain sometimes. The disulfide bond Cys20-Cys269 is essential in controlling specific orientation of Nt region and maintaining conformational integrity of extracellular domain. RMS comparison analysis between conformers revealed the ECL1 of CCR5 stays relative rigid, whereas the ECL2 and Nt are rather flexible. Solvent-accessible surface area calculations indicated that the charged residues within Nt and ECL2 are often exposed to solvent. Integrating these results with available experimental data, a two-step gp120-CCR5 binding mechanism was proposed. The dynamic interaction of CCR5 extracellular domain with gp120 was emphasized. Compensation grouting is increasingly employed as a mitigation technique of settlements induced by tunnelling and its effectiveness both in clayey and sandy soils is reported in a wide number of case histories. However, the results are highly dependent on grout properties, injection characteristics and soil properties. An experimental study was conducted to investigate the parameters that control grout injections in silty soils. The results from one injection test in a large sample of silty soil show that the compensation efficiency, defined as the ratio of the volume of heave obtained at ground surface and the injected grout volume, is much lower than one and tends to decrease with time, while the initial volume of grout lost due to pressure filtration is small. Finally, results from finite elements back analyses of the laboratory test show that a good agreement with the experimental data can be obtained if the development of large strains is taken into account. © 2012 Taylor & Francis Group.

Differential growth of thin elastic bodies furnishes a surprisingly simple explanation of the complex and intriguing shapes of many biological systems, such as plant leaves and organs. Similarly, inelastic strains induced by thermal effects or active materials in layered plates are extensively used to control the curvature of thin engineering structures. Such behaviour inspires us to distinguish and to compare two possible modes of differential growth not normally compared to each other, in order to reveal the full range of out-of-plane shapes of an initially flat disk. The first growth mode, frequently employed by engineers, is characterised by direct bending strains through the thickness, and the second mode, mainly apparent in biological systems, is driven by extensional strains of the middle surface. When each mode is considered separately, it is shown that buckling is common to both modes, leading to bistable shapes: growth from bending strains results in a double-curvature limit at buckling, followed by almost developable deformation in which the Gaussian curvature at buckling is conserved; during extensional growth, out-of-plane distortions occur only when the buckling condition is reached, and the Gaussian curvature continues to increase. When both growth modes are present, it is shown that, generally, larger displacements are obtained under in-plane growth when the disk is relatively thick and growth strains are small, and vice versa. It is also shown that shapes can be mono-, bi-, tri- or neutrally stable, depending on the growth strain levels and the material properties: furthermore, it is shown that certain combinations of growth modes result in a free, or natural, response in which the doubly curved shape of disk exactly matches the imposed strains. Such diverse behaviour, in general, may help to realise more effective actuation schemes for engineering structures. © 2012 Elsevier Ltd. All rights reserved.

This paper discusses road damage caused by heavy commercial vehicles. Chapter 1 presents some important terminology and a brief historical review of road construction and vehicle-road interaction, from ancient times to the present day. The main types of vehicle-generated road damage, and the methods that are used by pavement engineers to analyze them are discussed in Chapter 2. Attention is also given to the main features of the response of road surfaces to vehicle loads and mathematical models that have been developed to predict road response. Chapter 3 reviews the effects on road damage of vehicle features which can be studied without consideration of vehicle dynamics. These include gross vehicle weight, axle and tire configurations, tire contact conditions and static load sharing in axle group suspensions. The dynamic tire forces generated by heavy vehicles are examined in Chapter 4. The discussion includes their simulation and measurement, their principal characteristics, the effects of tires and suspension design on dynamic forces, and the potential benefits of using advanced suspensions for minimizing dynamic tire forces. Chapter 5 discusses methods for estimating the effects of dynamic tire forces on road damage. The two main approaches are either to examine the statistics of the forces themselves; or to calculate the response of a pavement model to the forces, and to calculate the resulting wear using a material damage model. The issues involved in assessing vehicles for 'road friendliness' are discussed in Chapter 6. Possible assessment methods include measuring strains in an instrumented pavement traversed by the vehicle, measuring dynamic tire forces, or measuring vehicle parameters such as the 'natural frequency' and 'damping ratio'. Each of these measurements involves different assumptions and analysis methods for converting the results into some measure of road damage. Chapter 7 includes a summary of the main conclusions of the paper and recommendations for tire and suspension design, road design and construction, and for vehicle regulations.

An analysis is presented of a database of 67 tests on 21 clays and silts of undrained shear stress-strain data of fine-grained soils. Normalizations of secant G in terms of initial mean effective stress p9 (i.e., G=p9 versus log g) or undrained shear strength cu (i.e., G=cu versus log g) are shown to be much less successful in reducing the scatter between different clays than the approach that uses the maximum shear modulus,Gmax, a technique still not universally adopted by geotechnical researchers and constitutive modelers. Analysis of semiempirical expressions forGmax is presented and a simple expression that uses only a void-ratio function and a confining-stress function is proposed. This is shown to be superior to a Hardin-style equation, and the void ratio function is demonstrated as an alternative to an overconsolidation ratio (OCR) function. To derive correlations that offer reliable estimates of secant stiffness at any required magnitude of working strain, secant shear modulus G is normalized with respect to its small-strain value Gmax, and shear strain g is normalized with respect to a reference strain gref at which this stiffness has halved. The data are corrected to two standard strain rates to reduce the discrepancy between data obtained from static and cyclic testing. The reference strain gref is approximated as a function of the plasticity index.Aunique normalized shear modulus reduction curve in the shape of a modified hyperbola is fitted to all the available data up to shear strains of the order of 1%. As a result, good estimates can be made of the modulus reduction G/Gmax ±30% across all strain levels in approximately 90% of the cases studied. New design charts are proposed to update the commonly used design curves. © 2013 American Society of Civil Engineers.

One of the biggest issues for underground construction in a densely built-up urban environment is the potentially adverse impact on buildings adjacent to deep excavations. In Singapore, a building damage assessment is usually carried out using a three-staged approach to assess the risk of damage caused by major underground construction projects. However, the tensile strains used for assessing the risk of building damage are often derived using deflection ratios and horizontal strains under 'greenfield' conditions. This ignores the effects of building stiffness and in many cases may be conservative. This paper presents some findings from a study on the response of buildings to deep excavations. Firstly, the paper discusses the settlement response of an actual building - the Singapore Art Museum - adjacent to a deep excavation. By comparing the monitored building settlement with the adjacent ground settlement markers, the influence of building stiffness in modifying the response to excavation-induced settlements is observed. Using the finite element method, a numerical study on the building response to movements induced by deep excavations found a consistent relationship between the building modification factor and a newly defined relative bending stiffness of the building. This relationship can be used as a design guidance to estimate the deflection ratio in a building from the greenfield condition. By comparing the case study results with the design guidance developed from finite element analysis, this paper presents some important characteristics of the influence of building stiffness on building damages for deep excavations.

This paper describes first some of the recent performance checks on the high performance fibre-reinforced cementitious composite CARDIFRC and then its application to the retrofitting of damaged concrete beams. It is shown that an even distribution of fibres throughout the bulk of the material is crucial to its excellent fatigue performance and to the reduction in the autogenous shrinkage strains. The distribution of fibres in beams, cylinders and strips is examined using computerised tomography imaging and traditional image analysis. Thin strips of CARDIFRC are used to retrofit damaged concrete beams which are subjected to thermal cycling. It is shown that neither the load carrying capacity of the retrofitted beams nor the bond between retrofit strips and concrete deteriorates with thermal cycling. The load carrying capacity of retrofitted beams is predicted with a model based on fracture mechanics, and the predictions are shown to be in good agreement with test data. © 2006 Elsevier Ltd. A class of ultra-high-performance fibre-reinforced cementitious composites (UHPFRCC) has been developed at Cardiff university and registered under the trade name CARDIFRC. The method of its production and its mechanical and fracture properties were reported previously in a series of papers in Magazine of Concrete Research. Here the results of recent fatigue and shrinkage tests on this material are reported. As with the mechanical and fracture properties, it is shown that an even and uniform distribution of fibres throughout the bulk of the material is crucial to its superior fatigue performance and to the reduction in the shrinkage strains. © 2010 Thomas Telford Ltd.
