Four trials of identical experimental design were conducted to determine the effects of temperature, dietary Lys level, and dietary Arg:Lys ratios on performance and carcass yield of male broilers. Birds of a commercial strain were grown from 21 to 42 d of age in wire-floored finishing batteries placed in environmental chambers. The chambers were programmed to provide either a constant thermoneutral temperature (21.1 C), a constant cold temperature (15.5 C), or a cycling hot diurnal temperature (25.5 to 33.3 C). Within each environment there was a factorial arrangement of three Lys levels (1.0, 1.1, and 1.2%) with four Arg:Lys ratios (1.1:1, 1.2:1, 1.3:1, and 1.4:1). Environmental temperature significantly influenced virtually every characteristic examined. Hot cyclic temperatures reduced weight gain, feed intake, and breast meat yield, and increased feed conversion, dressing percentage, leg quarter yield, and abdominal fat content. The cold environment promoted increased feed intake and mortality. Ascites and cardiomyopathy were the leading causes of death under cold exposure and thermoneutral conditions, whereas complications arising from heat exposure were the main cause of death under hot cyclic conditions. Levels of Lys affected leg quarter yield and abdominal fat content over all environments but increased breast meat yield only under cold conditions. Increasing Arg: Lys ratios improved feed conversion and dressing percentage and reduced abdominal fat content; it could not be determined whether these responses were consistent with Arg per se or were due to a nonspecific N response. As increasing Lys levels or Arg:Lys ratios did not improve weight gain, increase breast meat yield, or attenuate adverse effects due to heat or cold exposure, it is concluded that the levels of Lys and Arg suggested for 21 to 42 d by the NRC are adequate for birds of this age under the environmental conditions encountered.

F-4 generation of human growth hormone (hGH) gene-transgenic red common carp, and the non-transgenic controls were fed for 8 weeks on purified diets with 20%, 30% or 40% protein. Analysis of whole-body amino acids showed that the proportions of lysine, leucine, phenylalanine, valine and alanine, as percentages of body protein, increased significantly, while those of arginine, glutamic acid and tyrosine decreased, with increases in dietary protein level in at least one strain of fish. Proportions of the other amino acids were unaffected by the diets. The proportions of lysine and arginine were significantly higher, while those of leucine and alanine were lower in the transgenics than in the controls in at least one diet group. Proportions of the other amino acids were unaffected by strain. The results suggest that the whole-body amino acid profile of transgenic carp, when expressed as proportions of body protein, was in general, similar to that of the non-transgenic controls. AIMS/HYPOTHESIS: Recent studies suggest that oxidative stress should be monitored alongside HbA(1c) to identify subgroups of diabetic patients at high risk of initiation or progression of retinopathy. The acrolein-derived advanced lipoxidation end-product (ALE), [Formula: see text]-(3-formyl-3,4-dehydropiperidino)lysine (FDP-lysine), is a useful biomarker that reflects the cumulative burden of oxidative stress over long periods of time. The purpose of the present study was to investigate whether serum and haemoglobin levels of FDP-lysine are associated with the severity of diabetic retinopathy in type 1 and type 2 diabetic patients.
METHODS: Serum and haemoglobin levels of FDP-lysine were measured by competitive ELISA in 59 type 1 and 76 type 2 diabetic patients with no retinopathy, non-proliferative retinopathy or proliferative retinopathy (mean age [+/-SEM] 54.3 +/- 1.3 years), and in 47 non-diabetic control individuals (mean age 51.9 +/- 2.1 years).
RESULTS: Serum and haemoglobin levels of FDP-lysine were significantly increased in diabetic patients compared with control individuals (p = 0.04 and p = 0.002, respectively). However, no significant association was found between levels of serum FDP-lysine and the severity of diabetic retinopathy (p = 0.97). In contrast, increased haemoglobin FDP-lysine levels were observed in patients with proliferative retinopathy compared with patients without retinopathy and with non-proliferative retinopathy (p = 0.04). The relationship of FDP-lysine with proliferative retinopathy was unaltered after adjustment for HbA(1c), or other clinical parameters.
CONCLUSIONS/INTERPRETATION: Our data suggest that haemoglobin FDP-lysine may provide a useful risk marker for the development of proliferative diabetic retinopathy independently of HbA(1c), and that elevated intracellular ALE formation may be involved in the pathogenesis of this sight-threatening complication of diabetes.

The potential adverse effects on health of diet-derived advanced glycation end-products (AGEs) is of current interest, due to their proposed involvement in the disease progression of diabetic and uraemic conditions. However, accurate information about levels of AGEs in foods is lacking. The objective of this investigation was to determine the level of one particular AGE, N-epsilon-(carboxymethyl)lysine (CML), a marker of AGE formation, in a wide range of foods commonly consumed in a Western style diet. Individual foods (n = 257) were mixed, lyophilised, ground, reduced, fat-extracted, hydrolysed, and underwent solid-phase extraction. Extracts were analysed by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Cereal (2.6 mg/100 g food) and fruit and vegetable (0.13 mg/100 g food) categories had the highest and lowest mean level of CML, respectively, when expressed in mg/100 g food. These data can be used for estimating potential consumer intakes, and provide information that can be used to educated consumers on how to reduce their CML intake. (C) 2011 Elsevier Ltd. N epsilon-(Carboxymethyl)lysine (CML) is formed on oxidative cleavage of carbohydrate adducts to lysine residues in glycated proteins in vitro [Ahmed et al. (1988) J. Biol. Chem. 263, 8816-8821; Dunn et al. (1990) Biochemistry 29, 10964-10970]. We have shown that, in human lens proteins in vivo, the concentration of fructose-lysine (FL), the Amadori adduct of glucose to lysine, is constant with age, while the concentration of the oxidation product, CML, increases significantly with age [Dunn et al. (1989) Biochemistry 28, 9464-9468]. In this work we extend our studies to the analysis of human skin collagen. The extent of glycation of insoluble skin collagen was greater than that of lens proteins (4-6 mmol of FL/mol of lysine in collagen versus 1-2 mmol of FL/mol of lysine in lens proteins), consistent with the lower concentration of glucose in lens, compared to plasma. In contrast to lens, there was a slight but significant age-dependent increase in glycation of skin collagen, 33% between ages 20 and 80. As in lens protein, CML, present at only trace levels in neonatal collagen, increased significantly with age, although the amount of CML in collagen at 80 years of age, approximately 1.5 mmol of CML/mol of lysine, was less than that found in lens protein, approximately 7 mmol of CML/mol of lysine. The concentration of N epsilon-(carboxymethyl)hydroxylysine (CMhL), the product of oxidation of glycated hydroxylysine, also increased with age in collagen, in parallel with the increase in CML, from trace levels at infancy to approximately 5 mmol of CMhL/mol of hydroxylysine at age 80.(ABSTRACT TRUNCATED AT 250 WORDS)

Carboxymethyllysine (CML) has been identified as a modified amino acid that accumulates with age in human lens proteins and collagen. CML may be formed by oxidation of fructoselysine (FL), the Amadori adduct formed on nonenzymatic glycosylation of lysine residues in protein, or by reaction of ascorbate with protein under autoxidizing conditions. We proposed that measurements of tissue and urinary CML may be useful as indices of oxidative stress or damage to proteins in vivo. To determine the extent to which oxidation of nonenzymatically glycosylated proteins contributes to urinary CML, we measured the urinary concentrations of FL and CML in diabetic (n = 26) and control (n = 28) patients. The urinary concentration of FL correlated strongly with HbA1 measurements and was significantly higher in diabetic compared with control samples (9.2 +/- 6.5 and 4.0 +/- 2.8 micrograms/mg creatinine, respectively; P less than 0.0001). There was also a strong correlation between the concentrations of CML and FL in both diabetic and control urine (r = 0.67, P less than 0.0001) but only a weakly significant increase in the CML concentration in diabetic compared with control urine (1.2 +/- 0.5 and 1.0 +/- 0.3 micrograms/mg creatinine, respectively; P = 0.05). The molar ratio of CML to FL was significantly lower in diabetic compared with control patients (0.25 +/- 0.12 and 0.43 +/- 0.16, respectively; P less than 0.0001).(ABSTRACT TRUNCATED AT 250 WORDS)

Epidemiological studies show that elevated plasma levels of advanced glycation end products (AGEs) are associated with diabetes, kidney disease, and heart disease. Thus AGEs have been used as disease progression markers. However, the effects of variations in biological sample processing procedures on the level of AGEs in plasma/serum samples have not been investigated. The objective of this investigation was to assess the effect of variations in blood sample collection on measured Ne_(carboxy-methyl)lysine (CML), the best characterised AGE, and its homolog, Ne_(carboxyethyl)lysine (CEL). The investigation examined the effect on CML and CEL of different blood collection tubes, inclusion of a stabilising cocktail, effect of freeze thaw cycles, different storage times and temperatures, and effects of delaying centrifugation on a pooled sample from healthy volunteers. CML and CEL were measured in extracted samples by ultra_performance liquid chromatography-tandem mass spectrometry. Median CML and CEL ranged from 0.132 to 0.140 mM/M lys and from 0.053 to 0.060 mM/M lys, respectively. No significant difference was shown CML or CEL in plasma/serum samples. Therefore samples collected as part of epidemiological studies that do not undergo specific sample treatment at collection are suitable for measuring CML and CEL.

Chez la levure Saccharomyces cerevisiae, l'acétylation de l'histone H3 sur la lysine 56 (H3K56ac) est présente sur les histones néo-synthétisées déposées derrière les fourches de réplication et est essentielle pour préserver la viabilité cellulaire en réponse au dommage à l'ADN. La désacétylation d'H3K56 sur l'ensemble du génome catalysée par Hst3 et Hst4 et a lieu en phase G2 ou M. H3K56ac est une lame à double tranchant. L'absence d'H3K56ac rend les cellules sensibles aux dommages à l'ADN. En revanche, un excès d'acétylation d'H3K56 dans un mutant hst3Δ hst4Δ a des conséquences encore plus sévères tels que la thermo-sensibilité, l'hypersensibilité aux agents génotoxiques, l'instabilité génomique ainsi qu'une courte durée de vie réplicative. Les désacétylases Hst3 et Hst4 sont étroitement régulées au cours du cycle cellulaire afin de permettre à l'H3K56ac d'exercer son rôle en réponse aux dommages à l'ADN tout en évitant les conséquences néfastes de l'hyperacétylation d'H3K56. Dans cette thèse, nous avons identifié la machinerie moléculaire responsable de la dégradation de Hst3. De plus, nous avons exploré les raisons pour lesquelles l'absence de désacétylation donne lieu aux phénotypes du mutant hst3Δ hst4Δ. Au chapitre 2, nous démontrons que la dégradation d'Hst3 peut être complétée avant l'anaphase. Ceci suggère que la désacétylation de H3K56 a lieu durant une courte fenêtre du cycle cellulaire se situant entre la complétion de la phase S et la métaphase. De plus, nous avons identifié deux sites de phosphorylation d'Hst3 par la kinase cycline-dépendante 1 (Cdk1) et démontré que ces évènements de phosphorylation conduisent à la dégradation d'Hst3 in vivo. Nous avons aussi démontré que l'ubiquityltransférase Cdc34 et l'ubiquitine ligase SCFCdc4 sont requises pour la dégradation d'Hst3. Finalement, nous avons montré que la phosphorylation d'Hst3 par la kinase mitotique Clb2-Cdk1 peut directement entraîner l'ubiquitylation d'Hst3 par SCFCdc4 in vitro. Au chapitre 3, nous avons étudié les mécanismes moléculaires sous-jacents à la sensibilité extrême du mutant hst3Δ hst4Δ aux agents qui endommagent l'ADN. Nous avons établi qu'en raison de la présence anormale d'H3K56ac devant les fourches de réplication, le mutant hst3Δ hst4Δ exhibe une forte perte de viabilité lorsqu'exposé au méthyl méthanesulfonate (MMS) durant un seul passage à travers la phase S. Nous avons aussi découvert que, malgré le fait que le point de contrôle de réponse aux dommages à l'ADN est activé normalement dans le mutant hst3Δ hst4Δ, ce mutant est incapable de compléter la réplication de l'ADN et d'inactiver le point de contrôle pour une longue période de temps après exposition transitoire au MMS. L'ensemble de nos résultats suggère que les lésions à l'ADN induites par le MMS dans le mutant hst3Δ hst4Δ causent une forte perte de viabilité parce que ce mutant est incapable de compléter la réplication de l'ADN après une exposition transitoire au MMS. Dans la deuxième section du chapitre 3, nous avons employé une approche génétique afin d'identifier de nouveaux mécanismes de suppression de deux phénotypes prononcés du mutant hst3Δ hst4Δ. Nous avons découvert que la délétion de plusieurs gènes impliqués dans la formation de frontières entre l'hétérochromatine et de l'euchromatine atténue les phénotypes du mutant hst3Δ hst4Δ sans réduire l'hyperacétylation d'H3K56. Nos résultats indiquent aussi que l'abondante acétylation de l'histone H4 sur la lysine 16 (H4K16ac) est néfaste au mutant hst3Δ hst4Δ. Ce résultat suggère un lien génétique intriguant entre l'acétylation d'H3K56 et celle d'H4K16. L'existence de ce lien était jusqu'à présent inconnu. Nous avons identifié un groupe de suppresseurs spontanés où H3K56ac est indétectable, mais la majorité de nos suppresseurs ne montrent aucune réduction flagrante d'H3K56ac ou d'H4 K16ac par rapport aux niveaux observés dans le mutant hst3Δ hst4Δ. Une étude plus approfondie de ce groupe de suppresseurs est susceptible de mener à la découverte de nouveaux mécanismes génétiques ou épigénétiques permettant d'éviter les conséquences catastrophiques de l'hyperacétylation d'H3K56 chez le mutant hst3Δ hst4Δ. En résumé, cette thèse identifie la machinerie moléculaire responsable de la dégradation d'Hst3 (une désacétylase d'H3K56) durant une fenêtre de temps situées entre la fin de la phase S et la métaphase. Nos résultats permettent aussi d'expliquer pourquoi la dégradation d'Hst3 précède le début de la phase S durant laquelle l'acétylation d'H3K56 s'accumule derrière les fourches de réplication afin d'exercer son rôle de mécanisme de défense contre le dommage à l'ADN. De plus, nous avons identifié plusieurs suppresseurs qui permettent de contourner le rôle important d'Hst3 et Hst4 en réponse au dommage à l'ADN. Plusieurs suppresseurs révèlent un lien génétique inattendu entre deux formes abondantes d'acétylation des histones chez Saccharomyces cerevisiae, soit H3K56ac et H4K16ac.
