The sensitivity of altitudinal and latitudinal tree-line ecotones to climate change, particularly that of temperature, has received much attention. To improve our understanding of the factors affecting tree-line position, we used the spatially explicit dynamic forest model TreeMig. Although well-suited because of its landscape dynamics functions, TreeMig features a parabolic temperature growth response curve, which has recently been questioned. and the species parameters are not specifically calibrated for cold temperatures. Our main goals were to improve the theoretical basis of the temperature growth response curve in the model and develop a method for deriving that curve's parameters from tree-ring data. We replaced the parabola with an asymptotic curve, calibrated for the main species at the subalpine (Swiss Alps: Pinus cembra, Larix decidua, Picea abies) and boreal (Fennoscandia: Pinus sylvestris, Betula pubescens, P. abies) tree-lines. After fitting new parameters, the growth curve matched observed tree-ring widths better. For the subalpine species, the minimum degree-day sum allowing, growth (kDDMin) was lowered by around 100 degree-days; in the case of Larix, the maximum potential ring-width was increased to 5.19 mm. At the boreal tree-line, the kDDMin for P. sylvestris was lowered by 210 degree-days and its maximum ring-width increased to 2.943 mm; for Betula (new in the model) kDDMin was set to 325 degree-days and the maximum ring-width to 2.51 mm; the values from the only boreal sample site for Picea were similar to the subalpine ones, so the same parameters were used. However, adjusting the growth response alone did not improve the model's output concerning species' distributions and their relative importance at tree-line. Minimum winter temperature (MinWiT, mean of the coldest winter month), which controls seedling establishment in TreeMig, proved more important for determining distribution. Picea, P. sylvestris and Betula did not previously have minimum winter temperature limits, so these values were set to the 95th percentile of each species' coldest MinWiT site (respectively -7, -11, -13). In a case study for the Alps, the original and newly calibrated versions of TreeMig were compared with biomass data from the National Forest Inventor), (NFI). Both models gave similar, reasonably realistic results. In conclusion, this method of deriving temperature responses from tree-rings works well. However, regeneration and its underlying factors seem more important for controlling species' distributions than previously thought. More research on regeneration ecology, especially at the upper limit of forests. is needed to improve predictions of tree-line responses to climate change further.

There is mounting evidence that organic or inorganic enrichment of aquatic environments increases the risk of infectious diseases, with disease agents ranging from helminth parasites to fungal, bacterial, and viral pathogens. The causal link between microbial resource availability and disease risk is thought to be complex and, in the case of so-called "opportunistic pathogens," to involve additional stressors that weaken host resistance (e.g., temperature shifts or oxygen deficiencies). In contrast to this perception, our experiment shows that the link between resource levels and infection of fish embryos can be very direct: increased resource availability can transform benign microbial communities into virulent ones. We find that embryos can be harmed before further stresses (e.g., oxygen depletion) weaken them, and treatment with antibiotics and fungicides cancels the detrimental effects. The changed characteristics of symbiotic microbial communities could simply reflect density-dependent relationships or be due to a transition in life-history strategy. Our findings demonstrate that simple microhabitat changes can be sufficient to turn "opportunistic" into virulent pathogens.

ABSTRACT : Background: Inactivation of tumour-related genes by promoter hypermethylation is a common epigenetic event in the development of a variety of tumours. Aim: To investigate in primary uveal melanoma the status of promoter methylation of genes thought to be involved in tumour development: p16, TIMP3, RASSF1, RARB, FHIT, hTERT and APC. Methods: Gene promoter methylation was studied by methylation-sensitive single-strand conformation analysis and dot-blot assay in a series of 23 primary uveal melanomas. All DNA samples were obtained from paraffin-embedded formalin-fixed tissue blocks. Results: hTERT promoter methylation was found with a relatively high frequency (52%). Promoter methylation of p16, TIMP3, RASSF1, RARB, FHIT and APC was a rare event. For none of these genes did promoter methylation exceed 15% of tumour samples, and, for some genes (FHIT and APC), no methylation was found at all. Furthermore, promoter methylation was absent in 39% (9/ 23) of cases. In only 22% (5/23) of cases was hypermethylation of at least two promoters observed. Conclusions: Promoter methylation of hTERT is a regular event in uveal melanoma. Hypermethylation of the other genes studied does not seem to be an essential element in the development of this tumour. As promoter methylation of APC, RASSF1 and RARB is often observed in cutaneous melanoma, these results suggest that different epigenetic events occur in the development of cutaneous and uveal melanoma. RAPPORT DE SYNTHESE : L'inactivation de gènes par une hyperméthylation de leur promoteur apparaît être un événement épigénétique fréquent, se retrouvant dans de nombreuses tumeurs. Dans cette étude, nous avons investigué dans des mélanomes primaires de l'uvée l'état de méthylation du promoteur de gènes fréquemment impliqués dans le développent tumoral tels que p16, TIMP3, RASSFI, RARB, FHIT, hTERT et APC. La méthylation des promoteurs de gènes a été étudiée par methylationsensitive single-strand conformation analysis (MS-SSCA) et dot blot assay (MS-DBA) dans une série de 23 mélanomes primaires de l'uvée. Tous les échantillons tissulaires provenaient de matériel fixé dans le formol et conservé dans des blocs de parraffine. Nous avons identifié une fréquence relativement élevée (52%) pour la méthylation du promoteur de hTERT. En ce qui concerne le reste des gènes étudiés, nous avons retrouvé des fréquences de méthylation de promoteurs relativement basses avec 13% pour RASSF1, 13% pour RARB 13%, 9% pour TIMP3 et 4% pour p16. Nous n'avons pas retrouvé d'hyperméthylation des promoteurs des gènes APC et FRIT. La méthylation de hTERT apparaît être un événement important dans la biologie du mélanome de l'uvée. L'hyperméthylation des autres gènes évalués ne semble pas être cruciale dans le développent de cette tumeur. Comme la méthylation des promoteurs des gènes APC, RASSF1 et RARB a été fréquemment observée dans le mélanome de la peau, notre étude tend à démontrer que des mécanismes épigénétiques différents surviennent dans le développement respectif de ces tumeurs.

Business cycle theory is normally described as having evolved out of a previous tradition of writers focusing exclusively on crises. In this account, the turning point is seen as residing in Clément Juglar's contribution on commercial crises and their periodicity. It is well known that the champion of this view is Schumpeter, who propagated it on several occasions. The same author, however, pointed to a number of other writers who, before and at the same time as Juglar, stressed one or another of the aspects for which Juglar is credited primacy, including the recognition of periodicity and the identification of endogenous elements enabling the recognition of crises as a self-generating phenomenon. There is indeed a vast literature, both primary and secondary, relating to the debates on crises and fluctuations around the middle of the nineteenth century, from which it is apparent that Juglar's book Des Crises Commerciales et de leur Retour Périodique en France, en Angleterre et aux États-Unis (originally published in 1862 and very much revised and enlarged in 1889) did not come out of the blue but was one of the products of an intellectual climate inducing the thinking of crises not as unrelated events but as part of a more complex phenomenon consisting of recurring crises related to the development of the commercial world - an interpretation corroborated by the almost regular occurrence of crises at about 10-year intervals.

The enhanced flow in carbon nanotubes is explained using a mathematical model that includes a depletion layer with reduced viscosity near the wall. In the limit of large tubes the model predicts no noticeable enhancement. For smaller tubes the model predicts enhancement that increases as the radius decreases. An analogy between the reduced viscosity and slip-length models shows that the term slip-length is misleading and that on surfaces which are smooth at the nanoscale it may be thought of as a length-scale associated with the size of the depletion region and viscosity ratio. The model therefore provides a physical interpretation of the classical Navier slip condition and explains why `slip-lengths' may be greater than the tube radius.

La meva investigació s’articula entorn a l’obra del filòsof alemany Jürgen Habermas. L’aportació filosòfica més important de Habermas ha estat l’elaboració d’una racionalitat discursiva. Aquesta nova manera d’entendre la raó com a discurs ha tingut importants repercussions en l’àmbit de la filosofia i de les ciències socials en general. La primera part de la meva investigació analitza l’obra de Habermas, posant èmfasi en el procés de configuració d’aquesta racionalitat discursiva. Aquest primer període culmina amb la presentació de la tesina “Introducció al pensament i l’obra de Jürgen Habermas”. Com a resultat d’aquesta investigació, la meva recerca es centrarà, primer, en les nocions habermasianes de publicitat (Öffentlichkeit) i d’opinió pública (öffentliche Meinung) i, finalment, en l’anomenada democràcia deliberativa. Per tant, la recerca consta de dos part estretament relacionades: (1) un anàlisi del procés de configuració d’una raó discursiva en l’obra de Habermas; (2) una valoració crítica dels límits i les possibilitats de la democràcia deliberativa en les societats contemporànies. Aquesta darrera fase d’investigació –en la qual treballo actualment– s’endinsa en el debat dels “teòrics deliberatius”. Entre d’altres, aprofundeix en les repercussions ètiques i polítiques d’una relació problemàtica entre esfera pública i esfera privada. Desfer aquesta problemàtica passa per repensar l’exigència d’una racionalitat imparcial que, alhora, sigui Desfer aquesta problemàtica capaç de garantir la participació democràtica de la ciutadania en el context actual, on el pluralisme de valors ha esdevingut un dels fenòmens vertebradors de les societats contemporànies.

Human RNA polymerase (Pol) III-transcribed genes are thought to share a simple termination signal constituted by four or more consecutive thymidine residues in the coding DNA strand, just downstream of the RNA 3'-end sequence. We found that a large set of human tRNA genes (tDNAs) do not display any T(≥4) stretch within 50 bp of 3'-flanking region. In vitro analysis of tDNAs with a distanced T(≥4) revealed the existence of non-canonical terminators resembling degenerate T(≥5) elements, which ensure significant termination but at the same time allow for the production of Pol III read-through pre-tRNAs with unusually long 3' trailers. A panel of such non-canonical signals was found to direct transcription termination of unusual Pol III-synthesized viral pre-miRNA transcripts in gammaherpesvirus 68-infected cells. Genome-wide location analysis revealed that human Pol III tends to trespass into the 3'-flanking regions of tDNAs, as expected from extensive terminator read-through. The widespread occurrence of partial termination suggests that the Pol III primary transcriptome in mammals is unexpectedly enriched in 3'-trailer sequences with the potential to contribute novel functional ncRNAs.

This paper explores two major issues, from biophysical and historical viewpoints. We examine land management, which we define as the long-term fertility maintenance of land in relation to agriculture, fishery and forestry. We also explore humans’ positive role as agents aiming to reinforce harmonious materials circulation within the land. Liebig’s view on nature, agriculture and land, emphasizes the maintenance of long-term land fertility based on his agronomical thought that the circulation of matter in agricultural fields must be maintained with manure as much as possible. The thoughts of several classical economists, on nature, agriculture and land are reassessed from Liebig’s view point. Then, the land management problem is discussed at a much more fundamental level, to understand the necessary conditions for life in relation to land management. This point is analyzed in terms of two mechanisms: entropy disposal on the earth, and material circulation against gravitational field. Finally from the historical example of the metropolis of Edo, it is shown that there is yet another necessary condition for the sustainable management of land based on the creation of harmonious material cycles among cities, farm land, forests and surrounding sea areas in which humans play a vital role as agent.

El laboratori de física és el lloc que permet apropar la Física i la seva realitat quotidiana, tant la tecnològica com la científica, a l'estudiantat mitjançant experiències i demostracions motivadores. En general, les pràctiques de laboratori se solen realitzar per parelles, encara que no necessàriament, propiciant un treball en equip. Ara bé, pensant en aquelles estudiantes i aquells estudiants que no poden seguir el calendari de sessions presencials establert, per raons justificables, s’ha generat un material docent innovador consistent en la filmació de vídeos d’algunes de les pràctiques que actualment s'estan duent a terme en el laboratori de Física de l'EPSEM. Les filmacions van acompanyades per uns tutorials que permeten introduir la pràctica i il·lustrar tots els conceptes teòrics que hi intervenen i algunes simulacions. A més, també hi ha disponible uns tests d'autoavaluació i d'avaluació que acreditin l'aprenentatge de l'estudiant. L’objectiu és procurar, a partir d’una experimentació virtual, minimitzar la manca d’adquisició d’algunes habilitats pròpies de l’experimentació real al laboratori. Els productes creats han estat inclosos en el web del Departament de Física Aplicada de la UPC a l'EPSEM, on es proporciona tot un conjunt d’eines i informacions pensades per facilitar una millor forma de treballar en un laboratori. En aquest web es poden trobar, les normes generals per a la realització de les pràctiques i per a l’elaboració dels informes preceptius, el guió i l’esquema del muntatge de cada experimentació, així com enllaços a webs relacionats amb la física, que poden resultar molt útils per a l’alumnat. El pas endavant que suposa el material desenvolupat, ampliable en un futur, és sens dubte una alternativa educativa complementaria per garantir una formació més integral de les nostres estudiantes i dels nostres estudiants, en correspondència amb l’esperit de fomentar l'autoaprenentatge que traspua el procés d'integració a l'EEES. El projecte ha tingut com a finalitat cobrir un buit en la formació dels estudiants pel que respecta a les seves limitacions en l’aprenentatge de les diferents estratègies i metodologies de treball. Aquest buit ens ha fet pensar en l’elaboració d’un sistema didàctic que possibiliti una comunicació fluida amb l’alumne i que pugui anar més enllà de l’àmbit específic de les aules. Es per això que la nostra proposta té com a referent un aprofitament més racional del temps de treball de l’estudiant. Per tal d’arribar a aquesta finalitat ens hem plantejat un grup d’objectius concrets ordenats segons els dos nivells d’assoliment següents: Nivell 1: - Definir un material didàctic que pugui transmetre de la manera més objectiva possible les eines necessàries per tal que l’alumne resolgui un conjunt de problemes concrets. - Proporcionar a l’estudiant un procediment didàctic basat en un grup de tasques amb un material de suport on ha de posar a prova l’aplicabilitat dels conceptes donats a les classes teòriques. - Partir d’un sistema que permeti a l’alumne tenir una referència constant de recolzament teòric fora de les aules quan està resolent les pràctiques assignades en el curs. - Definir unes característiques funcionals del material utilitzat en l’activitat pràctica de tal manera que sigui un instrument efectiu per al control del propi estudiant sobre el seu procés d’avaluació. Nivell 2: - Fer servir una metodologia d’activitats pràctiques on es puguin accentuar les capacitats de resposta creativa individual de l’estudiant. - Plantejar l’intercanvi d’experiències i propostes d’estratègies per a la resolució dels temes pràctics mitjançant sessions de seminaris prèvies a les pràctiques. - Promoure unes relacions transversals amb altres àrees de coneixement de tal manera que l’alumne pugui vincular la seva resposta a les activitats pràctiques proposades per altres assignatures, amb els coneixements adquirits a la nostra assignatura.

El Laboratori Virtual de Psicologia Bàsica (LVPB) és una eina que acompanya a l’estudiant en un recorregut a través d’experiments que s’han convertit en clàssics de la Psicologia Cognitiva. La plataforma reprodueix experiments provinents de l’estudi de l’atenció, l’emoció, el llenguatge, la memòria, la motivació, el pensament i la percepció. En cada experiment, el LVPB presenta un text introductori amb alguns articles de referència on es van proposar per primera vegada aquests experiments. El LVPB es basa en la plataforma d’e-learning Moodle, el que permet que cada estudiant tingui el seu propi espai en el qual guardar les dades, els resultats i les anàlisis que hagi realitzat. Tota la interacció amb el sistema queda enregistrada i pot ésser consultada tant per l’estudiant com pel seu professor en qualsevol moment. New anti-cancer agents are being developed that specifically recognise tumour cells. Recognition is dependent upon the enhanced expression of antigenic determinants on the surface of tumour cells. The tumour exposure and the extracellular accessibility of the mucin MUC-1 make this marker a suitable target for tumour diagnosis and therapy. We isolated and characterised six human scFv antibody fragments that bound to the MUC-1 core protein, by selecting a large naive human phage display library directly on a MUC-1-expressing breast carcinoma cell line. Their binding characteristics have been studied by ELISA, FACS and indirect immunofluorescence. The human scFv antibody fragments were specific for the tandem repeat region of MUC-1 and their binding is inhibited by soluble antigen. Four human scFv antibody fragments (M2, M3, M8, M12) recognised the hydrophilic PDTRP region of the MUC-1 core protein, which is thought to be an immunodominant region. The human scFv antibody fragments were stable in human serum at 37 degrees C and retained their binding specificity. For imaging or targeting to tumours over-expressing MUC-1, it might be feasible to use these human scFv, or multivalent derivatives, as vehicles to deliver anti-cancer agents.

Human Fas ligand (L) (CD95L) and tumor necrosis factor (TNF)-alpha undergo metalloproteinase-mediated proteolytic processing in their extracellular domains resulting in the release of soluble trimeric ligands (soluble [s]FasL, sTNF-alpha) which, in the case of sFasL, is thought to be implicated in diseases such as hepatitis and AIDS. Here we show that the processing of sFasL occurs between Ser126 and Leu127. The apoptotic-inducing capacity of naturally processed sFasL was reduced by >1,000-fold compared with membrane-bound FasL, and injection of high doses of recombinant sFasL in mice did not induce liver failure. However, soluble FasL retained its capacity to interact with Fas, and restoration of its cytotoxic activity was achieved both in vitro and in vivo with the addition of cross-linking antibodies. Similarly, the marginal apoptotic activity of recombinant soluble TNF-related apoptosis-inducing ligand (sTRAIL), another member of the TNF ligand family, was greatly increased upon cross-linking. These results indicate that the mere trimerization of the Fas and TRAIL receptors may not be sufficient to trigger death signals. Thus, the observation that sFasL is less cytotoxic than membrane-bound FasL may explain why in certain types of cancer, systemic tissue damage is not detected, even though the levels of circulating sFasL are high.

1. Wind pollination is thought to have evolved in response to selection for mechanisms to promote pollination success, when animal pollinators become scarce or unreliable. We might thus expect wind-pollinated plants to be less prone to pollen limitation than their insect-pollinated counterparts. Yet, if pollen loads on stigmas of wind-pollinated species decline with distance from pollen donors, seed set might nevertheless be pollen-limited in populations of plants that cannot self-fertilize their progeny, but not in self-compatible hermaphroditic populations.2. Here, we test this hypothesis by comparing pollen limitation between dioecious and hermaphroditic (monoecious) populations of the wind-pollinated herb Mercurialis annua.3. In natural populations, seed set was pollen-limited in low-density patches of dioecious, but not hermaphroditic, M. annua, a finding consistent with patterns of distance-dependent seed set by females in an experimental array. Nevertheless, seed set was incomplete in both dioecious and hermaphroditic populations, even at high local densities. Further, both factors limited the seed set of females and hermaphrodites, after we manipulated pollen and resource availability in a common garden experiment.4. Synthesis. Our results are consistent with the idea that pollen limitation plays a role in the evolution of combined vs. separate sexes in M. annua. Taken together, they point to the potential importance of pollen transfer between flowers on the same plant (geitonogamy) by wind as a mechanism of reproductive assurance and to the dual roles played by pollen and resource availability in limiting seed set. Thus, seed set can be pollen-limited in sparse populations of a wind-pollinated species, where mates are rare or absent, having potentially important demographic and evolutionary implications.

Brain invasion is a biological hallmark of glioma that contributes to its aggressiveness and limits the potential of surgery and irradiation. Deregulated expression of adhesion molecules on glioma cells is thought to contribute to this process. Junctional adhesion molecules (JAMs) include several IgSF members involved in leukocyte trafficking, angiogenesis, and cell polarity. They are expressed mainly by endothelial cells, white blood cells, and platelets. Here, we report JAM-C expression by human gliomas, but not by their normal cellular counterpart. This expression correlates with the expression of genes involved in cytoskeleton remodeling and cell migration. These genes, identified by a transcriptomic approach, include poliovirus receptor and cystein-rich 61, both known to promote glioma invasion, as well as actin filament associated protein, a c-Src binding partner. Gliomas also aberrantly express JAM-B, a high affinity JAM-C ligand. Their interaction activates the c-Src proto-oncogene, a central upstream molecule in the pathways regulating cell migration and invasion. In the tumor microenvironment, this co-expression may thus promote glioma invasion through paracrine stimuli from both tumor cells and endothelial cells. Accordingly, JAM-C/B blocking antibodies impair in vivo glioma growth and invasion, highlighting the potential of JAM-C and JAM-B as new targets for the treatment of human gliomas.
