Questa dissertazione si concentra sul dibattito circa l’essenza della matematica a partire dalla nascita delle geometrie non euclidee. Essa è una scienza della natura o una costruzione (e dunque una libera invenzione) della mente umana? Trattando altresì della crisi dei fondamenti, e tenendo a mente le posizioni platoniste e costruttiviste, questa tesi analizza le risposte che, da fine Ottocento in poi, diedero Jules-Henri Poincaré, Bertrand Russell, L.E.J. Brouwer e David Hilbert, e con loro le varie correnti convenzionaliste, logiciste, intuizioniste e formaliste.

This work contributes to the field of spatial economics by embracing three distinct modelling approaches, belonging to different strands of the theoretical literature. In the first chapter I present a theoretical model in which the changes in urban system’s degree of functional specialisation are linked to (i) firms’ organisational choices and firms’ location decisions. The interplay between firms’ internal communication/managing costs (between headquarters and production plants) and the cost of communicating with distant business services providers leads the transition process from an “integrated” urban system where each city hosts every different functions to a “functionally specialised” urban system where each city is either a primary business center (hosting advanced business services providers, a secondary business center or a pure manufacturing city and all this city-types coexist in equilibrium.The second chapter investigates the impact of free trade on welfare in a two-country world modelled as an international Hotelling duopoly with quadratic transport costs and asymmetric countries, where a negative environmental externality is associated with the consumption of the good produced in the smaller country. Countries’ relative sizes as well as the intensity of negative environmental externality affect potential welfare gains of trade liberalisation. The third chapter focuses on the paradox, by which, contrary to theoretical predictions, empirical evidence shows that a decrease in international transport costs causes an increase in foreign direct investments (FDIs). Here we propose an explanation to this apparent puzzle by exploiting an approach which delivers a continuum of Bertrand- Nash equilibria ranging above marginal cost pricing. In our setting, two Bertrand firms, supplying a homogeneous good with a convex cost function, enter the market of a foreign country. We show that allowing for a softer price competition may indeed more than offset the standard effect generated by a decrease in trade costs, thereby restoring FDI incentives.

During my PhD, starting from the original formulations proposed by Bertrand et al., 2000 and Emolo & Zollo 2005, I developed inversion methods and applied then at different earthquakes. In particular large efforts have been devoted to the study of the model resolution and to the estimation of the model parameter errors. To study the source kinematic characteristics of the Christchurch earthquake we performed a joint inversion of strong-motion, GPS and InSAR data using a non-linear inversion method. Considering the complexity highlighted by superficial deformation data, we adopted a fault model consisting of two partially overlapping segments, with dimensions 15x11 and 7x7 km2, having different faulting styles. This two-fault model allows to better reconstruct the complex shape of the superficial deformation data. The total seismic moment resulting from the joint inversion is 3.0x1025 dyne.cm (Mw = 6.2) with an average rupture velocity of 2.0 km/s. Errors associated with the kinematic model have been estimated of around 20-30 %. The 2009 Aquila sequence was characterized by an intense aftershocks sequence that lasted several months. In this study we applied an inversion method that assumes as data the apparent Source Time Functions (aSTFs), to a Mw 4.0 aftershock of the Aquila sequence. The estimation of aSTFs was obtained using the deconvolution method proposed by Vallée et al., 2004. The inversion results show a heterogeneous slip distribution, characterized by two main slip patches located NW of the hypocenter, and a variable rupture velocity distribution (mean value of 2.5 km/s), showing a rupture front acceleration in between the two high slip zones. Errors of about 20% characterize the final estimated parameters.

Die Mikrophysik in Wolken bestimmt deren Strahlungseigenschaften und beeinflusst somit auch den Strahlungshaushalt des Planeten Erde. Aus diesem Grund werden im Rahmen der vorliegenden Arbeit die mikrophysikalischen Charakteristika von Cirrus-Wolken sowie von arktischen Grenzschicht-Wolken behandelt. Die Untersuchung dieser Wolken wurde mithilfe verschiedener Instrumente verwirklicht, welche Partikel in einem Durchmesserbereich von 250nm bis zu 6.4mm vermessen und an Forschungsflugzeugen montiert werden. Ein Instrumentenvergleich bestätigt, dass innerhalb der Bereiche in denen sich die Messungen dieser Instrumente überlappen, die auftretenden Diskrepanzen als sehr gering einzustufen sind. Das vorrangig verwendete Instrument trägt die Bezeichnung CCP (Cloud Combination Probe) und ist eine Kombination aus einem Instrument, das Wolkenpartikel anhand von vorwärts-gerichtetem Streulicht detektiert und einem weiteren, das zweidimensionale Schattenbilder einzelner Wolkenpartikel aufzeichnet. Die Untersuchung von Cirrus-Wolken erfolgt mittels Daten der AIRTOSS-ICE (AIRcraft TOwed Sensor Shuttle - Inhomogeneous Cirrus Experiment) Kampagne, welche im Jahr 2013 über der deutschen Nord- und Ostsee stattfand. Parameter wie Partikeldurchmesser, Partikelanzahlkonzentration, Partikelform, Eiswassergehalt, Wolkenhöhe und Wolkendicke der detektierten Cirrus-Wolken werden bestimmt und im Kontext des aktuellen Wissenstandes diskutiert. Des Weiteren wird eine beprobte Cirrus-Wolke im Detail analysiert, welche den typischen Entwicklungsprozess und die vertikale Struktur dieser Wolkengattung widerspiegelt. Arktische Grenzschicht-Wolken werden anhand von Daten untersucht, die während der VERDI (VERtical Distribution of Ice in Arctic Clouds) Kampagne im Jahr 2012 über der kanadischen Beaufortsee aufgezeichnet wurden. Diese Messkampagne fand im Frühling statt, um die Entwicklung von Eis-Wolken über Mischphasen-Wolken bis hin zu Flüssigwasser-Wolken zu beobachten. Unter bestimmten atmosphärischen Bedingungen tritt innerhalb von Mischphasen-Wolken der sogenannte Wegener-Bergeron-Findeisen Prozess auf, bei dem Flüssigwassertropfen zugunsten von Eispartikeln verdampfen. Es wird bestätigt, dass dieser Prozess anhand von mikrophysikalischen Messungen, insbesondere den daraus resultierenden Größenverteilungen, nachweisbar ist. Darüber hinaus wird eine arktische Flüssigwasser-Wolke im Detail untersucht, welche im Inneren das Auftreten von monomodalen Tröpfchen-Größenverteilungen zeigt. Mit zunehmender Höhe wachsen die Tropfen an und die Maxima der Größenverteilungen verschieben sich hin zu größeren Durchmessern. Dahingegen findet im oberen Übergangsbereich dieser Flüssigwasser-Wolke, zwischen Wolke und freier Atmosphäre, ein Wechsel von monomodalen zu bimodalen Tröpfchen-Größenverteilungen statt. Diese weisen eine Mode 1 mit einem Tropfendurchmesser von 20μm und eine Mode 2 mit einem Tropfendurchmesser von 10μm auf. Das dieses Phänomen eventuell typisch für arktische Flüssigwasser-Wolken ist, zeigen an dem Datensatz durchgeführte Analysen. Mögliche Entstehungsprozesse der zweiten Mode können durch Kondensation von Wasserdampf auf eingetragenen Aerosolpartikeln, die aus einer Luftschicht oberhalb der Wolke stammen oder durch Wirbel, welche trockene Luftmassen in die Wolke induzieren und Verdampfungsprozesse von Wolkentröpfchen hervorrufen, erklärt werden. Unter Verwendung einer direkten numerischen Simulation wird gezeigt, dass die Einmischung von trockenen Luftmassen in den Übergangsbereich der Wolke am wahrscheinlichsten die Ausbildung von Mode 2 verursacht.

OBJECTIVES: Hypoglycaemia (glucose <2.2 mmol/l) is a defining feature of severe malaria, but the significance of other levels of blood glucose has not previously been studied in children with severe malaria. METHODS: A prospective study of 437 consecutive children with presumed severe malaria was conducted in Mali. We defined hypoglycaemia as <2.2 mmol/l, low glycaemia as 2.2-4.4 mmol/l and hyperglycaemia as >8.3 mmol/l. Associations between glycaemia and case fatality were analysed for 418 children using logistic regression models and a receiver operator curve (ROC). RESULTS: There was a significant difference between blood glucose levels in children who died (median 4.6 mmol/l) and survivors (median 7.6 mmol/l, P < 0.001). Case fatality declined from 61.5% of the hypoglycaemic children to 46.2% of those with low glycaemia, 13.4% of those with normal glycaemia and 7.6% of those with hyperglycaemia (P < 0.001). Logistic regression showed an adjusted odds ratio (AOR) of 0.75 (0.64-0.88) for case fatality per 1 mmol/l increase in baseline blood glucose. Compared to a normal blood glucose, hypoglycaemia and low glycaemia both significantly increased the odds of death (AOR 11.87, 2.10-67.00; and 5.21, 1.86-14.63, respectively), whereas hyperglycaemia reduced the odds of death (AOR 0.34, 0.13-0.91). The ROC [area under the curve at 0.753 (95% CI 0.684-0.820)] indicated that glycaemia had a moderate predictive value for death and identified an optimal threshold at glycaemia <6.1 mmol/l, (sensitivity 64.5% and specificity 75.1%). CONCLUSIONS: If there is a threshold of blood glucose which defines a worse prognosis, it is at a higher level than the current definition of 2.2 mmol/l.

BPAG1-b is the major muscle-specific isoform encoded by the dystonin gene, which expresses various protein isoforms belonging to the plakin protein family with complex, tissue-specific expression profiles. Recent observations in mice with either engineered or spontaneous mutations in the dystonin gene indicate that BPAG1-b serves as a cytolinker important for the establishment and maintenance of the cytoarchitecture and integrity of striated muscle. Here, we studied in detail its distribution in skeletal and cardiac muscles and assessed potential binding partners. BPAG1-b was detectable in vitro and in vivo as a high molecular mass protein in striated and heart muscle cells, co-localizing with the sarcomeric Z-disc protein alpha-actinin-2 and partially with the cytolinker plectin as well as with the intermediate filament protein desmin. Ultrastructurally, like alpha-actinin-2, BPAG1-b was predominantly localized at the Z-discs, adjacent to desmin-containing structures. BPAG1-b was able to form complexes with both plectin and alpha-actinin-2, and its NH(2)-terminus, which contains an actin-binding domain, directly interacted with that of plectin and alpha-actinin. Moreover, the protein level of BPAG1-b was reduced in muscle tissues from plectin-null mutant mice versus wild-type mice. These studies provide new insights into the role of BPAG1-b in the cytoskeletal organization of striated muscle.

OBJECTIVE: To evaluate the ease of application of two-piece, graduated, compression systems for the treatment of venous ulcers. METHODS: Four kits used to provide limb compression in the management of venous ulcers were evaluated. These have been proven to be non-inferior to various types of bandages in clinical trials. The interface pressure exerted above the ankle by the under-stocking and the complete compression system and the force required to pull the over-stocking off were assessed in vitro. Ease of application of the four kits was evaluated in four sessions by five nurses who put stockings on their own legs in a blinded manner. They expressed their assessment of the stockings using a series of visual analogue scales (VASs). RESULTS: The Sigvaris Ulcer X((R)) kit provided a mean interface pressure of 46 mmHg and required a force in the range of 60-90 N to remove it. The Mediven((R)) ulcer kit exerted the same pressure but required force in the range of 150-190 N to remove it. Two kits (SurePress((R)) Comfort and VenoTrain((R)) Ulcertec) exerted a mean pressure of only 25 mmHg and needed a force in the range of 100-160 N to remove them. Nurses judged the Ulcer X and SurePress kits easiest to apply. Application of the VenoTrain kit was found slightly more difficult. The Mediven kit was judged to be difficult to use. CONCLUSIONS: Comparison of ease of application of compression-stocking kits in normal legs revealed marked differences between them. Only one system exerted a high pressure and was easy to apply. Direct comparison of these compression kits in leg-ulcer patients is required to assess whether our laboratory findings correlate with patient compliance and ulcer healing.

OBJECTIVE: To compare the proportion and rate of healing, pain, and quality of life of low-strength medical compression stockings (MCS) with traditional bandages applied for the treatment of recalcitrant venous leg ulcers. METHODS: A single-center, randomized, open-label study was performed with consecutive patients. Sigvaris prototype MCS providing 15 mm Hg-25 mm Hg at the ankle were compared with multi-layer short-stretch bandages. In both groups, pads were placed above incompetent perforating veins in the ulcer area. The initial static pressure between the dressing-covered ulcer and the pad was 29 mm Hg and 49 mm Hg with MCS and bandages, respectively. Dynamic pressure measurements showed no difference. Compression was maintained day and night and changed every week. The primary endpoint was healing within 90 days. Secondary endpoints were healing within 180 days, time to healing, pain (weekly Likert scales), and monthly quality of life (ChronIc Venous Insufficiency Quality of Life [CIVIQ] questionnaire). RESULTS: Of 74 patients screened, 60 fulfilled the selection criteria and 55 completed the study; 28 in the MCS and 27 in the bandage group. Ulcers were recurrent (48%), long lasting (mean, 27 months), and large (mean, 13 cm2). All but one patient had deep venous reflux and/or incompetent perforating veins in addition to trunk varices. Characteristics of patients and ulcers were evenly distributed (exception: more edema in the MCS group; P = .019). Healing within 90 days was observed in 36% with MCS and in 48% with bandages (P = .350). Healing within 180 days was documented in 50% with MCS and in 67% with bandages (P = .210). Time to healing was identical. Pain scored 44 and 46 initially (on a scale in which 100 referred to maximum and 0 to no pain) and decreased within the first week to 20 and 28 in the MCS and bandage groups, respectively (P < .001 vs .010). Quality of life showed no difference between the treatment groups. In both groups, pain at 90 days had decreased by half, independent of completion of healing. Physical, social, and psychic impairment improved significantly in patients with healed ulcers only. CONCLUSION: Our study illustrates the difficulty of bringing large and long-standing venous ulcers to heal. The effect of compression with MCS was not different from that of compression with bandages. Both treatments alleviated pain promptly. Quality of life was improved only in patients whose ulcers had healed.

Paraneoplastic pemphigus (PNP) is a devastating autoimmune blistering disease, involving mucocutaneous and internal organs, and associated with underlying neoplasms. PNP is characterized by the production of autoantibodies targeting proteins of the plakin and cadherin families involved in maintenance of cell architecture and tissue cohesion. Nevertheless, the identity of an antigen of Mr 170,000 (p170), thought to be critical in PNP pathogenesis, has remained unknown.

TRPV6 belongs to the vanilloid family of the transient receptor potential channel (TRP) superfamily. This calcium-selective channel is highly expressed in the duodenum and the placenta, being responsible for calcium absorption in the body and fetus. Previous observations have suggested that TRPV6 is not only permeable to calcium but also to other divalent cations in epithelial tissues. In this study, we tested whether TRPV6 is indeed also permeable to cations such as zinc and cadmium. We found that the basal intracellular calcium concentration was higher in HEK293 cells transfected with hTRPV6 than in non-transfected cells, and that this difference almost disappeared in nominally calcium-free solution. Live cell imaging experiments with Fura-2 and NewPort Green DCF showed that overexpression of human TRPV6 increased the permeability for Ca(2+), Ba(2+), Sr(2+), Mn(2+), Zn(2+), Cd(2+), and interestingly also for La(3+) and Gd(3+). These results were confirmed using the patch clamp technique. (45)Ca uptake experiments showed that cadmium, lanthanum and gadolinium were also highly efficient inhibitors of TRPV6-mediated calcium influx at higher micromolar concentrations. Our results suggest that TRPV6 is not only involved in calcium transport but also in the transport of other divalent cations, including heavy metal ions, which may have toxicological implications.

STUDY DESIGN: Retrospective case review. OBJECTIVES: In the present study, the neurological outcome, retirement and prognostic factors of patients with spinal cord injury without radiographic abnormality (SCIWORA) were evaluated. SETTING: Swiss national work accident insurance database. METHODS: The medical histories of 32 patients who were insured by the Swiss Accident Insurance Fund (SUVA) and had SCIWORA between 1995 and 2004 were evaluated thoroughly. Moreover, all available magnetic resonance imaging (MRI) scans were evaluated. RESULTS: At the last follow-up, none of the patients had complete spinal cord injury, only 4 patients had severe deficits and 12 patients had normal motor and sensory function in the neurological examination. However, only 7 out of 32 patients had returned to full-time work and 10 out of 32 patients were fully retired. Both the presence of spinal cord change (ρ=0.51) and higher maximum spinal cord compression (ρ=0.57) in MRI scan correlated with the likelihood for retirement; older age (ρ=0.38) and physical load of work (ρ=0.4) correlated with retirement to a lesser extent. CONCLUSION: Although the neurological outcome of SCIWORA is mostly good, the retirement rate is high. Presence of spinal cord change and severity of cord compression are the best predictors for the degree of retirement.

Based on the Ricker/Witmer survey on Library Support for Science Research and Education, a brief statistical analysis of the Bucknell University community and library support for science and engineering research and education is provided. The position and responsibilities of Reference Librarian/Coordinator of Science and Engineering Resources in the Ellen Clarke Bertrand Library are detailed. Throughout the article, I describe the motivation and justification for an integrated university library collection, which serves not only the Science and Engineering faculty and students, but the entire Bucknell University community. The issues of finance and budget, public service, and information access and delivery in relation to a central university library are discussed.
