Diffuse cutaneous leishmaniasis (DCL) is a rare form of tegumentary leishmaniasis (TL) and 350 cases have been reported world wide. Presently in Brazil, there were 31 cases reported corresponding to 8.9% of the world cases. The state of Maranhao contributed with ten cases (32.3%) of national reports. Recently the authors had opportunity to follow two new cases of DCL, both from the countryside of that state totaling 12 cases. The parasite Leishmania L. amazonensis was incriminated in the disease, confirming the available data as being the unique species to produce DCL in this state. The therapy associating meglumine anthimoniate with paramomicine sulphate (Gabbrox®) during 90 days, showed good tolerance and favorable response, being another option for fills form of DCL that constituted a challenge to science so far.

Toxocariasis is caused by infection of man by Toxocara canis and Toxocara, cati larvae, the common roundworm of dogs and cats. Because larvae are difficult to detect in tissues, diagnosis is mostly based on serology. Non specific reactions are observed mainly due to cross-reactivity with Ascaris sp antigens. This investigation aimed at developing and evaluating an indirect antibody competition ELISA (IACE) employing a specific rabbit IgG anti-Toxocara canis excretory-secretory antigens as the competition antibody. in order to improve indirect ELISA specificity performed for toxocariasis diagnosis. For that, the rabbit IgG was previously absorbed by Ascaris suum adult antigens. Sensitivity and specificity of IACE were first evaluated in 28 serum samples of mice experimentally infected with T. canis embryonated eggs. Adopting cut-off value established in this population before infection, sensitivity and specificity were 100% after 20 days post-inoculation. For human population IACE was evaluated using sera from 440 patients with clinical signs of toxocariasis and the cut-off value was established with 60 serum samples from apparently healthy individuals. Using as reference test the indirect ELISA performed by Adolfo Lutz Institute, sensitivity was 60.2%, specificity was 98% and concordance was 77.3%. Repeatability of IACE was evaluated by the inter-reactions variation coefficient (2.4%).

Irritable bowel syndrome is a high prevalent functional disorder. Its diagnosis is based in symptoms and the Rome Criteria. The treatment takes in account psychosocial and biologic factors. The therapeutic measures aim to correct the predominant symptoms: constipation, diarrhea and abdominal pain.

The pregnant women presenting Diabetes mellitus develop metabolic alterations, that may cause damage to the fetal well-being and provoke anomalies and/or malformations. The antioxidant treatment has improved the embryonic development from streptozotocin diabetic rats. Several studies have shown that a Gingko biloba extract presents antioxidant effects and, in the present study, one of the G. biloba extract formulations was used (EGb761) - Tebonin (200 mg/Kg/day), given to the diabetic pregnant female rats. The aim was to evaluate the effect of the EGb761 treatment on the of anomalies and/ or malformations incidence of the offspring. Diabetes was induced in female rats using streptozotocin in a dose of 40 mg/kg. The rats were mated, and the pregnant animals were divided in two groups: Control (water) and experimental (G. biloba). At day 21 of pregnancy, the rats were killed, and their fetuses were analyzed and processed for anomalies and/or malformations incidence. The results demonstrated that control and experimental groups presented no external anomalies and malformations; increased incidence of skeletal anomalies and of visceral malformations, and lower rate of visceral anomalies and skeletal malformations. These data confirm no statistical difference and, therefore, EGb761 treatment did not cause changes. Thus, a dose of 200 mg/Kg/day of a Gingko biloba extract given during the pregnancy rat was ineffective in the prevention of the anomalies and/or malformations related to the diabetes.

In the renal and femoral arteries of rabbit was verified that both vessels had walls structured by myostromal components, despite of their different distributive of blood, being the renal artery a visceral blood vessel and the femoral artery a parietal vessel. This wall pattern in these vessels concerned to presence of connective stromal elements (collagen and elastic fibres and lamellae) and smooth muscle cells coexisting with some equilibrium in the wall structure of the renal and femoral arteries, mainly in the medial layer architecture. An intimal folding pattern was verified around the vascular lumen, possibly related to capacitance of the both arteries regarding to variability of pressure levels in cardiac cycle. Furthermore, myostromal relations of connective elements and smooth muscle cells verified in the medial layer and the network formed by connective elements in the adventitial layer of these arteries contributed to maintenance of wall viscoelasticity properties of the vessels.

Male juveniles of Nile tilapia, Oreochromis niloticus, were stocked in 12, 300-m 2 ponds at a density of 1.7 fish/m 2 to evaluate the effects of different food management methods (natural foods and diets in pellet, floating or minced form) on fish production and carcass characteristics. Water quality variables monitored during the period were within acceptable levels for the species. Total fish production was significantly different (P < 0.05) and the highest values were obtained with diets in pellet (5,997 kg/ha) and floating (5,441 kg/ha) form. The fish fed diets had higher contents of body fat (1.57 to 1.98%) and visceral fat (12.64 to 25.04%) than fish fed natural food, which had levels between 0.17 and 0%, respectively. Natural food treatment yielded lower fish production and fish with lower body fat, while treatments that yielded higher fish production (rations) higher percentage of visceral fat. © 2004 by The Haworth Press, Inc. Feeding, seasonal changes in visceral fat and condition factor were compared in two species of characidiin fishes, Characidium lauroi and C. alipioi from Ribeirão Grande system, southeastern Brazil. Five streams of Ribeirão Grande system were sampled (22° 47' 08 S, 45° 28' 17W). The samples were taken four times per site, from July, 2001 to April, 2002: winter (July 2001), spring (October 2001), summer (February 2002) and autumn (April 2002). Quantitative collections were made with an electro-fishing device powered by a generator with maximum capacity of 1,500 V and 8.7 A of 60 Hz alternating current. Ephemeroptera nymphs, Diptera larvae (Chironomidae, Simuliidae), Plecoptera nymphs, Trichoptera larvae (Hydroptilidae, Psychoyiidae), terrestrial insects (Coleoptera, Isoptera, Hemiptera [Heteroptera, Homoptera]), Megaloptera larvae (Corydalidae), Arachnida, Ostracoda and vegetal debris were found in both species' diets. Visceral fat declined in February, coinciding with the decline of the condition factor in both species. The increased feeding from summer to fall provides fat accumulation. During subsequent seasons, fish may utilize visceral fat reserves for maintenance and reproduction.

Distribution and ecology knowledge of sandfly species is essential for epidemiology vigilance and risk determination for transmission of leishmaniasis. Sandfly trapping was carried out in a cerrado strictu sensu vegetation fragment in a rural area of Corumbataí Municipality, São Paulo State, Brazil, during July to November 2004. Two CDC light traps were used from 18h to 8h, once a month, resulting in 112 total hours of capture. During this period, 60 sandfly specimens of ten different species were sampled. The most abundant species and the one found in all captures was Pintomyia monticola totalizing 15 (25.0%) specimens, the second more abundant one was P. pessoai with 14 (23.3%) and the third one was Brumptomyia guimaraesi with 12 (20.0%) individuals collected. Other captured species were Psathyromyia aragaoi, B. avellari, B. brumpti, B. cunhai, P. bianchigalatiae, Evandromyia termitophila and Nyssomyia whitmani which corresponded to 19 (31.2%) specimens captured. Pintomyia pessoai and N. whitmani presence indicates transmission risk of American Cutaneous Leishmaniasis in the area.

During the structural designing of new drugs, it is possible predict the influence of specific chemical groups on pharmacological activity. Among these, the nitro group has potential antiparasitic activity, being present in many antimicrobial drugs, such as metronidazole, nitrofurazone, furazolidone, oxamniquine and chloramphenicol. Also, the introduction of the nitro group into a molecule can modify the physicochemical and electronic properties of the substance. Besides antimicrobial drugs, this group is also found in other drug classes, such as antiulcer, anti-inflamatory and anxiolytic. However, the use of the nitro group in drug design has encountered restrictions, due to the associated toxicity. This article is a review of the toxicity of nitrofuran compounds, as well the possible mechanisms involved and the strategy of latentiation by molecular modification to decrease their toxicity.

The curimbata constitutes species of fish of wide distribution in the Neotropical area and high economic value. This work focused to evaluate processing yield and the chemical composition of the fillet in the male and female migratories and residents stocks of Prochilodus lineatus. The using the plotting was completely randomized by 4x2 factorial scheme, having as factors four stocks (one resident and three migratories) and two genders (male and female) with thirty replicates, considering each fish as an experimental unit. Each individual was submitted to the manual filleted and determination of the corporal composition (texts of moisture, crude protein, lipids, ashes and energy). They following parameters of yield had been evaluated (%): visceral entire fish (RPIE %); yield of boneless fillet (RFILE %) and processing yield of fillet with bone (RFILEC %). There was a significant difference (p<0.05) between the stocks and the studied samples. The biggest yield of boneless fillet were observed for the fish of the migrator stock I (45.77%). Males and females of the migrator stock I presented biggest fillet yield (46.79% and 43.93%, respectively), while the migrator III presented smaller yield (37.29% for males and 38.44% for females). The migratories stocks I, II and III presented values significantly smaller (p<0.05) of lipids level (0.98%; 2.01% and 1.97%, respectively) and an increase in the content of corporal protein in relation to the resident stock. It concluded that the male weighing 591g on average, belonging of the stock migrator I (captured in December), presented better thread yield and the stocks migratories are fish that has low fat tenor and high protein tenor.

Desmoplastic small round cell tumor (DSRCT) is a rare, aggressive, malignant neoplasm usually present with the widespread abdominal serosal involvement and affects mainly adolescents and young adults. When presenting within visceral organs, as kidney, the diagnosis of DSRCT imposes significant difficulties. We present a case of primary DSRCT of the kidney in a 10-year-old boy mimicking clinically and pathologically Wilms tumor. The tumor showed morphologic and immunohistochemical features of DSRCT and the presence of the Ewing sarcoma and Wilm tumor 1 fusion transcripts resulting from the t(11;22) (p13;q12) reciprocal translocation. DSRCT should be considered in the differential diagnosis of Wilm tumor and other small blue-round cell tumors of the kidney. © 2009 by Lippincott Williams & Wilkins.

The aim of this study was to detect the presence of IgG antibodies anti-Leishmania spp., Toxoplasma gondii and Neospora caninum in dogs from a Veterinary Hospital from Universidade Estadual de Londrina. Blood samples from 112 animals were obtained by jugular venipuncture to obtain sera. The samples were tested by indirect immunofluorescence to detect antibodies anti-Leishmania spp., anti-N. caninum and anti-T. gondii. Thirteen (11.61%), 25 (22.32%), and 57 (50.89%) samples were positive for Leishmania spp., N. caninum, and T. gondii, respectively. The co-presence of anti-Leishmania spp. and N. caninum was observed in 6 (5.36%), anti-Leishmania spp. and anti-T. gondii in 8 (14.7%), and anti-N. caninum and anti-T. gondii in 18 (16.07%) samples. The co-presence of anti-Leishmania spp., anti-N. caninum and anti-T. gondii was observed in 5 (4.46%) dogs. There was a higher prevalence of Leishmania in Toxoplasma and Neospora positive animals, however, these results were not statistically significant (range p = 0.052 p = 0.06). The dogs have an important role in the epidemiological cycle of these diseases, which are important in animal and public health. The northern state of Paraná is an endemic area for human cutaneous leishmaniasis, therefore, studies should be conducted to uncover the real role of dogs as reservoirs of Leishmania to humans in the state.

The present study describes the leishmanicidal and trypanocidal activities of two quinonemethide triterpenes, maytenin (1) and pristimerin (2), isolated from Maytenus ilicifolia root barks (Celastraceae). The compounds were effective against the Trypanosomatidae Leishmania amazonensis and Leishmania chagasi and Trypanosoma cruzi, etiologic agents of leishmaniasis and Chagas' disease, respectively. The quinonemethide triterpenes 1 and 2 exhibited a marked in vitro leishmanicidal activity against promastigotes and amastigotes with 50% inhibitory concentration (IC50) values of less than 0.88 nM. Both compounds showed IC50 lower than 0.3 nM against Trypanosoma cruzi epimastigotes. The selectivity indexes (SI) based on BALB/c macrophages for L. amazonensis and L. chagasi were 243.65 and 46.61 for (1) and 193.63 and 23.85 for (2) indicating that both compounds presented high selectivity for Leishmania sp. The data here presented suggests that these compounds should be considered in the development of new and more potent drugs for the treatment of leishmaniasis and Chagas' disease. © 2013 by the authors.

The mortality caused by snakebites is more damaging than many tropical diseases, such as dengue haemorrhagic fever, cholera, leishmaniasis, schistosomiasis and Chagas disease. For this reason, snakebite envenoming adversely affects health services of tropical and subtropical countries and is recognized as a neglected disease by the World Health Organization. One of the main components of snake venoms is the Lys49-phospholipases A2, which is catalytically inactive but possesses other toxic and pharmacological activities. Preliminary studies with MjTX-I from Bothrops moojeni snake venom revealed intriguing new structural and functional characteristics compared to other bothropic Lys49-PLA2s. We present in this article a comprehensive study with MjTX-I using several techniques, including crystallography, small angle X-ray scattering, analytical size-exclusion chromatography, dynamic light scattering, myographic studies, bioinformatics and molecular phylogenetic analyses.Based in all these experiments we demonstrated that MjTX-I is probably a unique Lys49-PLA2, which may adopt different oligomeric forms depending on the physical-chemical environment. Furthermore, we showed that its myotoxic activity is dramatically low compared to other Lys49-PLA2s, probably due to the novel oligomeric conformations and important mutations in the C-terminal region of the protein. The phylogenetic analysis also showed that this toxin is clearly distinct from other bothropic Lys49-PLA2s, in conformity with the peculiar oligomeric characteristics of MjTX-I and possible emergence of new functionalities inresponse to environmental changes and adaptation to new preys. © 2013 Salvador et al.
