A espermiogênese nas espécies Steindachnerina insculpta, Cyphocharax gillii, C. modestus, C. spilotus e Potamorhina altamazonica de Curimatidae é caracterizada pelo desenvolvimento lateral do flagelo, rotação do núcleo, formação excêntrica da fossa nuclear e cromatina compactada em fibras espessas. Estes espermatozóides exibem uma cabeça esférica contendo um núcleo com cromatina altamente condensada em fibras espessas com pequenas áreas eletronlúcidas, e sem acrossoma. A fossa nuclear é do tipo moderado e excêntrico, penetrada pelo complexo centriolar. A peça média é pequena, tem muitas vesículas alongadas e um curto canal citoplasmático. Mitocôndrias podem ser alongadas, ramificadas ou em forma de C, e são separadas do segmento inicial do axonema pelo canal citoplasmático. O flagelo contém a estrutura clássica do axonema (9+2) e tem um compartimento membranoso na região inicial; não possui expansões laterais (fins). Somente pequenas diferenças foram observadas entre as espécies e gêneros analisados de Curimatidae. A espermiogênese e os espermatozóides de Curimatidae têm muitas das características encontradas em quase todas as outras espécies de Characiformes. Por outro lado, a presença de um compartimento membranoso na região inicial do flagelo dos curimatídeos, uma estrutura comum nos espermatozóides de muitos cipriniformes, é desconhecida em outros characiformes. Discute-se sobre a espermiogênese e espermatozóides de Characiformes.

Morphological aspects of spermatozoa in marine animals have been used in recent decades as phylogenetic criteria (spermiotaxonomy). This paper presents ultrastructural descriptions of the spermatozoa from Uca maracoani, U. thayeri, and U. vocator. A small portion of the vas deferens of each species was examined under the transmission and scanning electron microscopy. The ultrastructural analysis showed that each spermatophore consists of a varying number of spermatozoa embedded in a dense fibrillar matrix surrounded by a membrane. The spermatozoa of U. maracoani, U. thayeri, and U. vocator are typical of brachyurans. The Voluminous acrosome is characterized by three different layers. The postero-lateral surface of the acrosome is cupped by the reduced cytoplasm, and the anterior surface is covered by the operculum. The perforatorium consists of coiled, helicoidal membranous tubules and is continuous with the cytoplasm. The nucleus is composed by uncondensed chromatin and presents several lateral arms distributed over the entire equatorial plane of the cell. The presence of the apical button is a well defined character among all species of the genus Uca, but in U. thayeri it was not observed. The accessory opercular ring can be found in the three studied species, but in distinct development degree-Two centrioles were detected in U. thayeri and U. vocator, but only one was found in U. maracoani. The presence of centrioles in the mature spermatozoa is the first account for the genus Uca upto-date. Considering the ultrastructure of the spermatozoa of U. maracoani, U. thayeri, and U. vocator, we suggest that these three species partially follow the morphological patterns previously described in other Thoracotremata brachyurans. The absence of the apical button in U. thayeri spermatozoa may represent an evolutionary novelty in the genus Uca. (C) 2007 Elsevier Ltd. All rights reserved.

The digestive tract of insects is an important natural, physical, and chemical defense barrier against pathogen invasion. Certain. lepidopteran caterpillars are serious pests of agricultural crops and their biology has received much attention, but little is known about the larval noctuid gut. The morphological analysis of the digestive tract in Anticarsia gemmatalis under scanning electron microscopy (SEM) is a good model for studies about its defense mechanism. The material was fixed (2,5% glutaraldehyde solution; 0.1 M-phosphate buffer, pH 7.3), post-fixed (1% osmium tetroxide in the same buffer), dried at critical point, gold coated and analyzed in a SEM 515-Philips. A. gemmatalis digestive tract consists of a straight duct of varying length and diameter, subdivided in three main regions: the foregut formed by the oral cavity, pharynx, esophagus, and crop; the midgut that is the largest portion of the digestive tract without noticeable morphological differentiation along its length; and the hindgut that is morphologically differentiated in pylorus, ileum, colon, and rectum. Although the general morphology of the A. gemmatalis digestive tract is quite similar to the other Lepidoptera species, the anatomical array of the crop muscular layers is quite different comparing with the description for other larval insect.

The comparative study of the ultrastructure of the midgut epithelium of stingless bee larvae that eat plant protein (pollen) and animal protein (carrion) throughout the larval phase, shows variations in the digestive cells that are only relative to larval aging and not to the type of larval diet. The cells of older larvae present a cytoplasm with empty spaces that result from emptying of lipid and glycogen stocks, and the presence of autophagic vacuoles. These results are discussed in relation to the hypothesis that variations in the digestive tract of insects may be associated with different diets or phylogeny. We conclude that different diets do not determine cell morphology adaptations in the studied species. As the variations in the ultrastructure of the midgut epithelium are the same in all studied species, including the necrophagous species Trigona hypogea, throughout the larval stage, this sequence of changes seems to be due to different physiological state during larval development.

Fipronil, active ingredient of the acaricide Frontiline (R), is a phenyl-pyrazolic derivative, and its efficacy in the elimination of several plagues, even in low concentrations, has already been demonstrated; however, its effect on nontarget organisms has not been thoroughly explained. In this sense, the objective of this study was to evaluate the effects of different dosages of fipronil on the liver of mice in artificial conditions. Results showed that the animals exposed to fipronil present significant ultrastrucutural changes in hepatic cells with evident cellular and cytoplasm disorganization in hepatocytes characterized by an increase in the number of organelles, mainly mitochondria and rough endoplasmic reticulum, organelles that, in the case of the exposed animals, were probably responsible for the enzymes' synthesis that have the function of inactivating the toxic metabolites. A fat accumulation in the hepatocytes' cytoplasm (steatosis) was observed, in addition to extended vacuolated areas, mainly in regions next to the cell nucleus. Alterations observed in the nuclei of the hepatocytes pointed out cell death processes. Moreover, Kupffer cells increased in number (hyperplasia) suggesting an increase in the phagocytic activity of the liver in the exposed animals. Microsc. Res. Tech., 2011. The salivary glands of Rhipicephalus sanguineus males at stages: unfed (control), at day seven post-attachment, and at days three and seven post-detachment from the host were examined using methods of enzymatic analysis and cell viability. At these stages of feeding, different staining patterns were observed in the cells of type IV, III, II and I acini, which were affected by degeneration in this sequence. Acid phosphatase reaction was inversely proportional to that of ATPase, while ATPase reaction was proportional to membrane integrity.Salivary gland cells of unfed males exhibited intact nucleus and plasma membrane, suggesting that the acid phosphatase detected may participate in the normal physiology of acini. In males at day seven post-attachment, intact membranes were observed in almost all types of acini, as well as stronger reaction for acid phosphatase, nuclear changes, and decrease in ATPase reaction, changes associated with the degenerative process. At days three and seven post-detachment degeneration progress, being observed loss of membrane integrity, nuclear changes, prominent decrease in ATPase reaction, and an increase in acid phosphatase reaction in the first case and a decreased of it at day seven post-detachment from the host. During cell death, alterations occurred in the following sequence: a) nuclear changes, b) loss of ATPase reaction, c) loss of integrity of the plasma membrane, and d) increase of acid phosphatase. The latter might be associated with the late degradation of cytoplasmic remnants, characterizing the process of cell death in glands of R. sanguineus males as atypical or non-classic apoptosis. In insects' oocytes, structures called accessory nuclei can be observed. They are similar to the nucleus of the oocyte and have been detected in Hymenoptera as well as in other groups, with their source and functions still under discussion (Cassidy & King 1972). Through the use of histochemical techniques as well as ultrastructural ones, it was possible to observe several accessory nuclei surrounding the oocyte nucleus in the immature oocytes of Pachycondyla (Neoponera) villosa ants. Morphologically they were seen to be round and smaller than the germinal vesicle. When using these histochemical techniques, the only positive reaction was for proteins. Ultrastructurally they have the same morphology as the oocyte nucleus and a fine and electrondense granulation was observed inside them. The function of accessory nuclei in P (Neoponera) villosa is probably as a source of anullate lamellae and as an RNA reservoir in the oocytes. In mature oocytes (stage III), these structures are not observed, suggesting that they disappeared before this stage by releasing their contents into the oocytes' cytoplasm, according to Camargo-Mathias and Caetano (1993).

The wasps of the genus Polistes have been considered the key to understanding the evolution of social behavior in Hymenoptera. Several studies have shown that the development of organized insect societies was accompanied by the evolution of structures like exocrine glands, which became specialized to perform specific functions. This article investigates the ultrastructural and cytochemical features of the hypopharyngeal glands of Polistes versicolor. These glands have been studied in depth in social bees, where they occur only in nurses and produce the royal jelly. Our results revealed that these glands basically did not vary among individuals or between sexes. They are constituted by spherical cells, each with a large nucleus and well-developed rough endoplasmic reticulum. Secretion vesicles are abundant, but lipid droplets were not observed, indicating that these glands may not have a role in pheromone synthesis. Acid phosphatase was detected in lysosomes, and also free in the cytosol, but did not seem to be related with cell death. Thus, our results suggest that the hypopharyngeal glands of P. versicolor may not have a specialized social role, but could produce digestive enzymes.
