This project would target Norfolk Creek Subwatershed for land treatment practices. The Norfolk Creek Subwatershed is 14,035 acres located southwest of Waukon. The landscape is characterized by rugged karst topography and is marked with hundreds of sinkholes, providing direct drainage into the water table, affecting wells, springs, and community water sources. The surface groundwater runoff from this karst landscape eventually flows into the Yellow River. The potential point and non-point pollution sources are complicated and expensive to resolve. Extensive water quality monitoring has been completed on Norfolk Creek and has tested high in many parameters. We hope that with the upland treatment included in this grant request, terraces, grade stabilization structures, sediment control basins, and livestock manure management systems, these will improve. Continued water quality sampling will monitor this. This application has been reviewed and approved by the Allamakee County Soil and Water Conservation District Commissioners.

Two sections of the Yellow River have been named to the State of Iowa’s 303d list of impaired waters. The listing reflects streams with pollution problems linked to habitat alterations, in addition to those with potential disease causing bacteria, viruses and parasites. This fact, combined with local knowledge of water quality problems, shows the need for land treatment practices and habitat improvement measures. This project would target the Yellow River watershed, which totals approximately 49,800 acres. Areas that drain directly into the Yellow River mainstream will be targeted. Individually, these areas are too small to be considered sub-watersheds. This project will assess the drainage areas for active gullies and prioritize grade stabilization structures based upon severity and impact on the fishery. Funding would be utilized to target high priority grade stabilization structure sites and provide cost-share for those projects. A prerequisite for cost-share allocation is 75% of the land contributing to the drainage area must have some form of treatment in place. The Allamakee SWCD has received an EPA Region 7 Grant toward grade stabilization structures in the same area. Landowners have indicated that 75% cost-share is necessary to implement practices. To meet this request, the EPA funding would be used at a 15% cost-share rate if matched with 60% cost-share from WIRB funding. If matched with Federal EQIP funds, 25% of funds obtained from WIRB would be used. If other funds were depleted, WIRB funds would be utilized for the entire 75% cost-share.

Farmers Creek is a moderately flowing stream that winds through seventeen miles of central Jackson County, encompassing a watershed area of 30,590 acres. Due to nutrient loading and sedimentation, the stream was placed on Iowa’s 303(d) List of Impaired Waters in 2002. A three year grant project was initiated in January 2005 to reduce both sediment delivery and phosphorus levels by 40% in critical areas along the stream corridor. Over thirty-five BMP’s were started in the first nine months of this project. Funding through WIRB is being requested specifically for streambank stabilization and protection projects not covered by other cost-share programs. Innovative project designs and techniques will be installed and act as demonstration sites. Projects may include jetties, weirs, cedar revetments, cattle crossings, and fencing. To assist in excluding cattle from the stream, watering systems such as slingpumps, pasture pumps, or water rams will be installed, in conjunction with filter strips and riparian buffers.

With WIRB funding the Fox River Ecosystem Development board will continue to install prioritized practices identified by assessments within the impaired segment of Fox River. The FRED board is also asking to continue funding for a new 5 year position for assessment, planning and technical assistance. With new assessments and water quality monitoring already being done on the impaired segment of the Fox River a lot of valuable information is at hand. Ecosystem Development board is requesting funding from WIRB to install grade stabilization structures, water sediment basins, and terraces to reduce sediment delivery to Fox River. The FRED board in both Iowa and Missouri are committed not only to seek funding to continue water quality efforts for more practices but also to enhance and protect existing practices and investments that protect our water quality and economic viability in both states. We are off to a good start and want to continue our progress on the Fox River.

The O6-methylguanine-DNA-methyltransferase (MGMT) promoter methylation status is a predictive parameter for the response of malignant gliomas to alkylating agents such as temozolomide. First clinical trials with temozolomide plus bevacizumab therapy in metastatic melanoma patients are ongoing, although the predictive value of the MGMT promoter methylation status in this setting remains unclear. We assessed MGMT promoter methylation in formalin-fixed, primary tumor tissue of metastatic melanoma patients treated with first-line temozolomide and bevacizumab from the trial SAKK 50/07 by methylation-specific polymerase chain reaction. In addition, the MGMT expression levels were also analyzed by MGMT immunohistochemistry. Eleven of 42 primary melanomas (26%) revealed a methylated MGMT promoter. Promoter methylation was significantly associated with response rates CR + PR versus SD + PD according to RECIST (response evaluation criteria in solid tumors) (p<0.05) with a trend to prolonged median progression-free survival (8.1 versus 3.4 months, p>0.05). Immunohistochemically different protein expression patterns with heterogeneous and homogeneous nuclear MGMT expression were identified. Negative MGMT expression levels were associated with overall disease stabilization CR + PR + SD versus PD (p=0.05). There was only a poor correlation between MGMT methylation and lack of MGMT expression. A significant proportion of melanomas have a methylated MGMT promoter. The MGMT promoter methylation status may be a promising predictive marker for temozolomide therapy in metastatic melanoma patients. Larger sample sizes may help to validate significant differences in survival type endpoints.

Waterloo Creek Watershed is a 30,610 acre area that straddles the Iowa and Minnesota border. The lower 43% of the watershed is in Iowa. Bee and Duck Creeks in Minnesota flow into Waterloo Creek in Iowa. Designated as a primary contact recreational stream as well as a high-quality, cold water stream in Iowa, Waterloo Creek is a popular destination for anglers and other nature enthusiasts. The stream was on the Iowa DNR’s “Impaired Waters List” in 2008 and 2010 for Escherichia coli (E. coli) bacteria. Samples collected in 2010 and 2011 showed higher levels of E. coli. at sites with cattle in close proximity to the stream and were generally greater after high rainfall events. Other factors affecting water quality are high turbidity levels and frequent flooding. There is a deficiency in upland land treatment and an abundance of conventional tillage which increases the amount of erosion and potential for surface runoff to carry sediment to the stream. A comprehensive watershed assessment and management plan have been completed for the watershed which identify the causes of and solutions to water quality impairments. The goals of this project are to 1) develop a formal working relationship between technical staff in Iowa and Minnesota, 2) identify specific locations for Best Management Practice (BMP) implementation, 3) reduce sediment loading to Waterloo Creek to improve aquatic habitat and decrease bacteria delivery, and 4) reduce flooding potential in the watershed. The following BMPs will be implemented to reach these goals: terraces, grade stabilization structures, pasture management, stream buffers, stream bank stabilization, and agricultural waste structures.

The purposes of this study were to prospectively determine changes in rotator cuff strength before and after surgical shoulder stabilization by Bristow-Latarjet procedure and to better estimate time needed for rotator cuff strength recovery. 20 patients with recurrent anterior posttraumatic shoulder dislocation underwent internal (IR) and external (ER) rotator isokinetic evaluation before and 3, 6 and 21 months after Bristow-Latarjet surgery. In a seated position with 45° of shoulder abduction in the scapular plane, both shoulders were evaluated concentrically with a Con-Trex® isokinetic dynamometer at 180°∙s - 1, 120°∙s - 1 and 60°∙s - 1. 3 months post-surgery, IR and ER strength of the operated shoulder were significantly lower than before surgery ( - 28±20% for IR, - 17±17% for ER) (P<0.05). At 6 and 21 months post-surgery, IR and ER strength were comparable to strength before surgery; strength recovery is seen at 6 months post-surgery with long-term maintenance at 21 months. Given the weakness 3 months post-surgery, return to sports (including overhead and contact sports) should be discussed, and 6 months post-surgery may be a better point for an athlete to resume practicing sports. Isokinetic rotator cuff strength evaluation appears to be relevant in helping to determine the need of continuing strength rehabilitation. Pre-surgical evaluation contributes to the relevance of later comparisons.

Manufacturing of glass from tin mining tailings in Bolivia Tailings from mining activities in Bolivia represent an environmental problem. In the vicinity of the tin mines of Llallagua,Potosí department, there are large dumps and tailings. We present a study of the use of these wastes as raw materials for the manufacture of glass. This procedure aims to contribute to environmental remediation of mining areas through the vitrification, a process which offers an alternative for stabilization of hazardous waste. In addition, the marketing of the obtained product would provide an additional income to the mining areas. For this study three samples of mining waste, with grain size between sand and silt, were used. The chemical composition of these raw materials, determined by X-ray fluorescence, is granitic, with high contents of heavy metals. On the basis of its composition, glass were made from silica glass by adding CaCO3 and Na2CO3. The thermal cycle has been determined from TDA. Tg values of glass range from 626º to 709 °C. Leaching tests of the obtained glasses confirm their capacity to retain heavy metals.

Tenascin-C is an adhesion-modulating extracellular matrix molecule that is highly expressed in tumor stroma and stimulates tumor cell proliferation. Adhesion of T98G glioblastoma cells to a fibronectin substratum is inhibited by tenascin-C. To address the mechanism of action, we performed a RNA expression analysis of T89G cells grown in the presence or absence of tenascin-C and found that tenascin-C down-regulates tropomyosin-1. Upon overexpression of tropomyosin-1, cell spreading on a fibronectin/tenascin-C substratum was restored, indicating that tenascin-C destabilizes actin stress fibers through down-regulation of tropomyosin-1. Tenascin-C also increased the expression of the endothelin receptor type A and stimulated the corresponding mitogen-activated protein kinase signaling pathway, which triggers extracellular signal-regulated kinase 1/2 phosphorylation and c-Fos expression. Tenascin-C additionally caused down-regulation of the Wnt inhibitor Dickkopf 1. In consequence, Wnt signaling was enhanced through stabilization of beta-catenin and stimulated the expression of the beta-catenin target Id2. Finally, our in vivo data derived from astrocytoma tissue arrays link increased tenascin-C and Id2 expression with high malignancy. Because increased endothelin and Wnt signaling, as well as reduced tropomyosin-1 expression, are closely linked to transformation and tumorigenesis, we suggest that tenascin-C specifically modulates these signaling pathways to enhance proliferation of glioma cells.

In order to prevent adjacent segment degeneration following spinal fusion new techniques are being used. Lumbar disc arthroplasty yields mid term results equivalent to those of spinal fusion. Cervical disc arthroplasty is indicated in the treatment of cervicobrachialgia with encouraging initial results. The ability of arthroplasty to prevent adjacent segment degeneration has yet to be proven. Although dynamic stabilization had not been proven effective in treating chronic low back pain, it might be useful following decompression of lumbar spinal stenosis in degenerative spondylolisthesis. Interspinal devices are useful in mild lumbar spinal stenosis but their efficacy in treating low back pain is yet to be proven. Confronted with a growing number of new technologies clinicians should remain critical while awaiting long term results.

Eukaryotic mRNA transcription and turnover is controlled by an enzymatic machinery that includes RNA polymerase II and the 3' to 5' exosome. The activity of these protein complexes is modulated by additional factors, such as the nuclear RNA polymerase II-associated factor 1 (Paf1c) and the cytoplasmic Superkiller (SKI) complex, respectively. Their components are conserved across uni- as well as multi-cellular organisms, including yeast, Arabidopsis, and humans. Among them, SKI8 displays multiple facets on top of its cytoplasmic role in the SKI complex. For instance, nuclear yeast ScSKI8 has an additional function in meiotic recombination, whereas nuclear human hSKI8 (unlike ScSKI8) associates with Paf1c. The Arabidopsis SKI8 homolog VERNALIZATION INDEPENDENT 3 (VIP3) has been found in Paf1c as well; however, whether it also has a role in the SKI complex remains obscure so far. We found that transgenic VIP3-GFP, which complements a novel vip3 mutant allele, localizes to both nucleus and cytoplasm. Consistently, biochemical analyses suggest that VIP3-GFP associates with the SKI complex. A role of VIP3 in the turnover of nuclear encoded mRNAs is supported by random-primed RNA sequencing of wild-type and vip3 seedlings, which indicates mRNA stabilization in vip3. Another SKI subunit homolog mutant, ski2, displays a dwarf phenotype similar to vip3. However, unlike vip3, it displays neither early flowering nor flower development phenotypes, suggesting that the latter reflect VIP3's role in Paf1c. Surprisingly then, transgenic ScSKI8 rescued all aspects of the vip3 phenotype, suggesting that the dual role of SKI8 depends on species-specific cellular context.

Työn loppuosassa on esitelty Kymenlaakson alueen laitosten mahdollisuuksia käsitellä öljyvahinkojätettä. Tietoa onkerätty haastattelemalla puhelimitse laitosten edustajia keväällä 2007. Alueella voidaan polttaa leijupedissä puhtaaseen polttoaineeseen sekoitettua öljyistä orgaanista ainesta ja puhdistustyössä käytettyjä varusteita arviolta 19 000 t/a, homogenoitua öljyistä orgaanista ainesta voidaan polttaa rumpu-uunissa arviolta 1200 t/a.

- Atherosclerosis is a complex inflammatory process, leading cause of morbidity and mortality in France. Inflammation is essential in initiation, progression and atherosclerosis plaque destabilization leading to acute cardiovascular events. Recent studies suggest that pharmacological PPAR activation prevents ΑΤΗ développement and progression and decreased cardiovascular mortality. Compared to pharmacological treatment, physical exercise also significantly prevents cardiovascular mortality. The aim of the first study was to investigate the influence of physical exercise on ATS development and PPAR expression in arterial wall. Our results had shown that physical exercise decrease ΑΤΗ progression and increase PPAR-γ expression in arterial wall. Interestingly, PPAR-γ inhibition with BADGE, a PPAR-γ antagonist abolishes these antiatherogenic effects. Hypertension increase ΑΤΗ complication such as plaque rupture. The aim of the second study were to inves¬tigate PPAR-γ implication in progression and stabilization of ΑΤΗ lesions in hypercholesterolemic and hypertensive ApoE-/- mice (2K1C) submitted to different exercises (voluntary wheel running and submitted treadmill running) or treated with telmisartan an anti-hypertensive drug. Our results shown that, physical exercise prevents ATS cardiovascular events by several mechanisms. Similarly to telmisartan, physical exercises stabilize ΑΤΗ lesion. Moreover results shown that, submitted exercise and telmisartan have an comparable mechanism. In fact, they significantly decrease pro-inflammatory cytokines expression and in the same time activated PPAR-γ expression in arterial wall. Contrary to submitted exercise, voluntary exercises increases expression of anti-inflammatory cytokines IL-1ra and increase M2 marker CD206. These results suggest that voluntary and submitted exercise have two different mechanism of action. Moreover, M2 surexpression in response to voluntary exercise shift the inflammatory balance in favor to M2. Further, this change of balance in favor to M2, is significantly correlated to decrease of ΑΤΗ progression. Voluntary exercises significantly decreases ΑΤΗ progression in the same levels like telmisartan treatment. Voluntary and submitted exercise has two different mechanisms, submitted exercise decrease proinflammatory cytokines expression whereas voluntary exercise increase anti-inflammatory cytokines expression and promote an anti-inflammatory phenotype of macrophages M2. The shift of M1/M2 balance towards M2 decreases atherosclerosis progression.
