The search for new instruments to promote an appropriate cervical preparation has led to the development of new rotary instruments such as TripleGates. However, to the best of the authors' knowledge, there is no study evaluating TripleGates effect on the “risk zone” of mandibular molars. Objectives: The aim of this study was to evaluate the effects of a crown-down sequence of Gates-Glidden and TripleGates burs on the remaining cervical dentin thickness and the total amount of dentin removed from the root canals during the instrumentation by using cone beam computed tomography. The number of separated instruments was also evaluated. Material and Methods: Mesial roots of 40 mandibular first molars were divided into 2 equal groups: crown-down sequence of Gates-Glidden (#3, #2, #1) and TripleGates burs. Cervical dentin thickness and canal area were measured before and after instrumentation by using cone beam computed tomography and image analysis software. Student’s t-test was used to determine significant differences at p<0.05. Results: No significant differences (p>0.05) were observed between the instruments, regarding the root canal area and dentin wall thickness. Conclusion: Both tested instruments used for cervical preparation were safe to be used in the mesial root canal of mandibular molars.

Chikungunya virus (CHIKV) is a mosquito-borne arthrogenic alphavirus that causes acute febrile illness in humans accompanied by joint pains and in many cases, persistent arthralgia lasting weeks to years. The re-emergence of CHIKV has resulted in numerous outbreaks in the eastern hemisphere, and threatens to expand in the foreseeable future. Unfortunately, no effective treatment is currently available. The present study reports the use of resazurin in a cell-based high-throughput assay, and an image-based high-content assay to identify and characterize inhibitors of CHIKV-infection in vitro. CHIKV is a highly cytopathic virus that rapidly kills infected cells. Thus, cell viability of HuH-7 cells infected with CHIKV in the presence of compounds was determined by measuring metabolic reduction of resazurin to identify inhibitors of CHIKV-associated cell death. A kinase inhibitor library of 4,000 compounds was screened against CHIKV infection of HuH-7 cells using the resazurin reduction assay, and the cell toxicity was also measured in non-infected cells. Seventy-two compounds showing >= 50% inhibition property against CHIKV at 10 mu M were selected as primary hits. Four compounds having a benzofuran core scaffold (CND0335, CND0364, CND0366 and CND0415), one pyrrolopyridine (CND0545) and one thiazol-carboxamide (CND3514) inhibited CHIKV-associated cell death in a dose-dependent manner, with EC50 values between 2.2 mu M and 7.1 mu M. Based on image analysis, these 6 hit compounds did not inhibit CHIKV replication in the host cell. However, CHIKV-infected cells manifested less prominent apoptotic blebs typical of CHIKV cytopathic effect compared with the control infection. Moreover, treatment with these compounds reduced viral titers in the medium of CHIKV-infected cells by up to 100-fold. In conclusion, this cell-based high-throughput screening assay using resazurin, combined with the image-based high content assay approach identified compounds against CHIKV having a novel antiviral activity -inhibition of virus-induced CPE - likely by targeting kinases involved in apoptosis.

Animal behavioral parameters can be used to assess welfare status in commercial broiler breeders. Behavioral parameters can be monitored with a variety of sensing devices, for instance, the use of video cameras allows comprehensive assessment of animal behavioral expressions. Nevertheless, the development of efficient methods and algorithms to continuously identify and differentiate animal behavior patterns is needed. The objective this study was to provide a methodology to identify hen white broiler breeder behavior using combined techniques of image processing and computer vision. These techniques were applied to differentiate body shapes from a sequence of frames as the birds expressed their behaviors. The method was comprised of four stages: (1) identification of body positions and their relationship with typical behaviors. For this stage, the number of frames required to identify each behavior was determined; (2) collection of image samples, with the isolation of the birds that expressed a behavior of interest; (3) image processing and analysis using a filter developed to separate white birds from the dark background; and finally (4) construction and validation of a behavioral classification tree, using the software tool Weka (model 148). The constructed tree was structured in 8 levels and 27 leaves, and it was validated using two modes: the set training mode with an overall rate of success of 96.7%, and the cross validation mode with an overall rate of success of 70.3%. The results presented here confirmed the feasibility of the method developed to identify white broiler breeder behavior for a particular group of study. Nevertheless, more improvements in the method can be made in order to increase the validation overall rate of success. (C) 2013 Elsevier B.V. All rights reserved.
