Euterpe (Martius, 1823), a genus from Central and South America, has species with high economic importance in Brazil, because of their palm heart and fruits, known as açaí berries. Breeding programs have been conducted to increase yield and establish cultivation systems to replace the extraction of wild material. These programs need basic information about the genome of these species to better explore the available genetic variability. The aim of this study was to compare E. edulis (Martius, 1824), E. oleracea (Martius, 1824) and E. precatoria (Martius, 1842), with regard to karyotype, type of interphase nucleus and nuclear DNA amount. Metaphase chromosomes and interphase nuclei from root tip meristematic cells were obtained by the squashing technique and solid stained for microscope analysis. The DNA amount was estimated by flow cytometry. There were previous reports on the chromosome number of E. edulis and E. oleracea, but chromosome morphology of these two species and the whole karyotype of E. precatoria are reported for the first time. The species have 2n=36, a number considered as a pleisomorphic feature in Arecoideae since the modern species, according to floral morphology, have the lowest chromosome number (2n=28 and 2n=30). The three Euterpe species also have the same type of interphase nuclei, classified as semi-reticulate. The species differed on karyotypic formulas, on localization of secondary constriction and genome size. The data suggest that the main forces driving Euterpe karyotype evolution were structural rearrangements, such as inversions and translocations that alter chromosome morphology, and either deletion or amplification that led to changes in chromosome size.

Genome editing is becoming an important biotechnological tool for gene function analysis and crop improvement, being the CRISPR-Cas9 (Clustered Regularly Interspaced Short Palindromic Repeat-CRISPR associated protein 9) system the most widely used. The natural CRISPR/Cas9 system has been reduced to two components: a single-guide RNA (sgRNA) for target recognition via RNA-DNA base pairing, which is commonly expressed using a promoter for small-RNAs (U6 promoter), and the Cas9 endonuclease for DNA cleavage (1). To validate the CRISPR/Cas9 system in strawberry plants, we designed two sgRNAs directed against the floral homeotic gene APETALA3 (sgRNA-AP3#1 and sgRNA-AP3#2). This gene was selected because ap3 mutations induce clear developmental phenotypes in which petals and stamens are missing or partially converted to sepals and carpels respectively (2). In this work, we used two different U6 promoters to drive the sgRNA-AP3s expression: AtU6-26 from Arabidopsis (4), and a U6 promoter from Fragaria vesca (FvU6) (this work). We also tested two different coding sequences of Cas9: a human- (hSpCas9) (3) and a plant-codon optimized (pSpCas9) (this work). Transient expression experiments using both CRISPR/Cas9 systems (AtU6-26:sgRNA-AP3#1_35S:hSpCas9_AtU6-26:sgRNA-AP3#2 and FvU6:sgRNA-AP3#1_35S:pSpCas9_FvU6:sgRNA-AP3#2) were performed infiltrating Agrobacterium tumefaciens into F. vesca fruits. PCR amplification and sequencing analyses across the target sites showed a deletion of 188-189 bp corresponding to the region comprised between the two cutting sites of Cas9, confirming that the CRISPR/Cas9 system is functional in F. vesca. Remarkably, the two systems showed different mutagenic efficiency that could be related to differences in expression of the U6 promoters as well as differences in the Cas9 transcripts stability and translation. Stable transformants for both F. vesca (2n) and Fragaria X anannassa (8n) are currently being established to test whether is possible to obtain heritable homozygous mutants derived from CRISPR/Cas9 strategies in strawberry. Thus, our work offers a promising tool for genome editing and gene functional analysis in strawberry. This tool might represent a more efficient alternative to the sometimes inefficient RNAi silencing methods commonly used in this species.

Although Mauritia flexuosa (Arecaceae) plays a pivotal role in the ecology and economy of the Amazon, and occurs in a variety of habitats, little is known about the influence of habitat on the reproductive biology of this palm. My dissertation focuses on the reproductive biology of M. flexuosa in three habitats in Roraima, Brazil: undisturbed forest, undisturbed forest-savanna ecotone, and savanna disturbed by plantations of the exotic tree, Acacia mangium. First, I calculated sex ratios and linked precipitation patterns with phenology. Sex ratios were female-biased. Precipitation was negatively associated with flowering, and positively associated with fruiting. Habitat appears to have no significant influence on phenology of M. flexuosa, although short-term climate variation may affect phenology of this species. Second, I examined floral biology, observed floral visitors, and performed exclusion experiments to determine the pollination system of M. flexuosa. Fruit set did not differ significantly between the visitor exclusion treatment and the control, but was significantly lowest in the wind + visitor exclusion treatment, suggesting that this dioecious palm is anemophilous, independent of habitat. Third, I identified the abiotic and biotic factors explaining variation in fruit mass, seed mass, seed number per fruit, and total fruit yield among habitats. Soil moisture and flooding during the wet season were the best predictors of fruit and seed output. The number of leaves, diameter at breast height, and height were all accurate predictors of reproductive output, but crown volume did not accurately predict fruit yields. Results re-evaluate traditional assumptions about wind-pollination in the tropics, and highlight abiotic and biotic factors responsible for variation in reproductive output of M. flexuosa, with implications for effective management of this palm. Finally, I interviewed harvesters and vendors to document the traditional knowledge and market dynamics of the fruit of M. flexuosa, buriti. Traditional knowledge corroborated results from scientific studies. Vendors argued that the price of buriti must increase, and must fluctuate with varying supply. With appropriate economic incentives to vendors/harvesters, Roraima may expand its market infrastructure for buriti, effectively stimulating the regional economy and practicing sustainable harvesting.

This thesis is composed of two parts, encompassed in a third: a poetry collection; a critical dissertation; and an artist’s book. The thesis as a whole is entitled Florilegium. This title, from the Latin flos, or ‘flower’, and legere, ‘to gather’, refers to the medieval system of collecting extracts from various authors to form a larger body of work. It is also applicable to flower-treatises, dedicated to their ornamental nature rather than medicinal or scientific. The critical dissertation comes in the form of a glossary. It intends to show that the flower plays an essential role in linking Modernist poetics with that of its Romantic predecessors and beyond. In isolated and ‘illuminated’ examples from Aristotle to Zukofsky, it examines the lineage of botanical poetry, in the light of its unique linguistic makeup: a vernacularized scientific lexicon established in the Latin of Carl Linnaeus. While the critical component of the thesis is an interrogation of botanical language, the poetry collection is its living representation. To enhance the living nature of the text, I have designed and printed an artist’s book, which also acts as an herbarium for floral specimens collected and pressed over the duration of my degree. The design of the book is in keeping with traditional florilegia, incorporating historic binding techniques, typography, paper, and size.

Olive fruit fly Bactrocera oleae (Rossi) (Diptera: Tephritidae) is a major olive pest in the Mediterranean basin where increasing insecticide resistance has enhanced damage and necessitates more reliance on other control strategies, such as biological control. Provision of floral resources has been reported to improve the effectiveness of natural enemies. Here, we tested the effect of six plant nectars and two honeydew sources on the survival of Psyttalia concolor (Szépligeti) (Hymenoptera: Braconidae), a parasitoid wasp used in the biological control of olive fruit fly. Our results showed a positive effect on survival associated with nectars of Anchusa azurea Mill., Rosmarinus officinalis L., Lavatera cretica L. and Calamintha nepeta (L.) Savi, while honeydew proved to be a valuable alternative food source. When offering flowers directly to insects, Anchusa azurea, Lavatera cretica, and Foeniculum vulgare L. were found to be the most beneficial species, indicating also that P. concolor feeds predominantly on shallow corollas.

Abstract: In this text, we deal with proccesses of appropriation of space in the Dam of Póvoa (Portugal) using a dwelling perspective (Ingold 2009) over the territory. The reconversion of innactive spaces in rural areas into leisure-related ones in a quite common practice in contemporary times. The Spaniards beach is one of the local names given by locals to the Dam of Póvoa (Castelo de Vide), in northern Alentejo (Portugal). In the collective memory of the residents, the Dam (built in the 1920’s) is remembered for the many flowers surrounding the area. This Garden-style aesthetics has outlived the initial function of the Dam (to produce electric power). From the 1960’s to the 1980’s, it was also a popular leisure space for Spanish and Portuguese people. It is not a beach, but being inland, it was the closest thing to a beach the residents had. The centrality of this leisure space in the area only decayed after the construction of swimming pools in the nearby towns. Early in the begginning of 21st century a fire destroyed part of the natural floral landscape of the dam. Abandoned for some years, the place has still been appropriated by leisured people in day-trips (for sight-seeing, pic-nics), fishing competitions, caravan soujorns, and, since 2013, in a music and dance festival named Andanças (www.andancas.net). The research that underlies this paper was carried out between 2013 and 2016 by a team of anthropologists. An ethnography of the Dam and its uses, and of the festival and its participants was carried out. In our working-paper we higlight the main research findings achieved. Key-words: Leisure; Landscape; Tourism; Music; Space; Portugal.
