We performed a genome-wide analysis of gene expression in primary human CD15+ myeloid progenitor cells. By using the serial analysis of gene expression (SAGE) technique, we obtained quantitative information for the expression of 37,519 unique SAGE-tag sequences. Of these unique tags, (i) 25% were detected at high and intermediate levels, whereas 75% were present as single copies, (ii) 53% of the tags matched known expressed sequences, 34% of which were matched to more than one known expressed sequence, and (iii) 47% of the tags had no matches and represent potentially novel genes. The correct genes were confirmed by application of the generation of longer cDNA fragments from SAGE tags for gene identification (GLGI) technique for high-copy tags with multiple matches. A set of genes known to be important in myeloid differentiation were expressed at various levels and used different spliced forms. This study provides a normal baseline for comparison of gene expression in myeloid diseases. The strategy of using SAGE and GLGI techniques in this study has broad applications to the genome-wide identification of expressed genes.

Ten novel small nucleolar RNA (snoRNA) gene clusters, consisting of two or three snoRNA genes, respectively, were identified from Arabidopsis thaliana. Twelve of the 25 snoRNA genes in these clusters are homologous to those of yeast and mammals according to the conserved antisense sequences that guide 2′-O-ribose methylation of rRNA. The remaining 13 snoRNA genes, including two 5.8S rRNA methylation guides, are new genes identified from A.thaliana. Interestingly, seven methylated nucleotides, predicted by novel snoRNAs Z41a–Z46, are methylated neither in yeast nor in vertebrates. Using primer extension at low dNTP concentration the six methylation sites were determined as expected. These snoRNAs were recognized as specific guides for 2′-O-ribose methylation of plant rRNAs. Z42, however, did not guide the expected methylation of 25S rRNA in our assay. Thus, its function remains to be elucidated. The intergenic spacers of the gene clusters are rich in uridine (up to 40%) and most of them range in size from 35 to 100 nt. Lack of a conserved promoter element in each spacer and the determination of polycistronic transcription from a cluster by RT–PCR assay suggest that the snoRNAs encoded in the clusters are transcribed as a polycistron under an upstream promoter, and individual snoRNAs are released after processing of the precursor. Numerous snoRNA gene clusters identified from A.thaliana and other organisms suggest that the snoRNA gene cluster is an ancient gene organization existing abundantly in plants.

Changes in polymerized actin during stress conditions were correlated with potato (Solanum tuberosum L.) tuber protein synthesis. Fluorescence microscopy and immunoblot analyses indicated that filamentous actin was nearly undetectable in mature, quiescent aerobic tubers. Mechanical wounding of postharvest tubers resulted in a localized increase of polymerized actin, and microfilament bundles were visible in cells of the wounded periderm within 12 h after wounding. During this same period translational activity increased 8-fold. By contrast, low-oxygen stress caused rapid reduction of polymerized actin coincident with acute inhibition of protein synthesis. Treatment of aerobic tubers with cytochalasin D, an agent that disrupts actin filaments, reduced wound-induced protein synthesis in vivo. This effect was not observed when colchicine, an agent that depolymerizes microtubules, was used. Neither of these drugs had a significant effect in vitro on run-off translation of isolated polysomes. However, cytochalasin D did reduce translational competence in vitro of a crude cellular fraction containing both polysomes and cytoskeletal elements. These results demonstrate the dependence of wound-induced protein synthesis on the integrity of microfilaments and suggest that the dynamics of the actin cytoskeleton may affect translational activity during stress conditions.

Cette thèse analyse la manifestation de la tradition dans certains textes des écrivains migrants au Québec, nommément La Québécoite et L’Immense fatigue des pierres de Régine Robin, Comment faire l’amour avec un nègre sans se fatiguer, Cette grenade dans la main du jeune nègre est-elle une arme ou un fruit ?, Pays sans chapeau et L’Énigme du retour de Dany Laferrière et, enfin L’Ingratitude et Immobile de Ying Chen. Les références à des récits, à des croyances et à des pratiques collectives qui se trouvent dans ces textes signalent une transformation de la transmission de l’héritage culturel d’une génération à l’autre. Afin de parvenir à une meilleure compréhension des liens entre la mémoire collective et la communauté culturelle se définissant dans le présent, je me sers du travail de folkloristes comme Alan Dundes, Simon Bronner et Richard Handler de même que des théories sur la faculté de la tradition proposées par Paul Ricœur, Walter Benjamin et Pierre Nora. Dans la première partie de la thèse, j’étudie l’imaginaire populaire du XIXe siècle et son incorporation dans la littérature canadienne-française de l’époque. Ensuite, j’analyse la remise en question de la tradition – de même que le rôle sous-jacent qu’elle continue de jouer, lors de la Révolution tranquille. Par rapport à ces deux périodes précédentes, l’usage du folklore chez les auteurs migrants laisse présager une nouvelle opération de la tradition dans un contexte où plusieurs groupes culturels se rencontrent. Dans un contexte pluraliste, l’idée de la communauté culturelle est fabriquée en fonction de notre appartenance à plusieurs groupes. Je m’intéresse à comment les gens qui habitent ensemble formulent des liens au passé même dans une situation dans laquelle ils ne partagent pas les mêmes rapports à la mémoire collective. Explorant le renouvellement, qui peut sembler paradoxal, de la tradition, je forme l’hypothèse qu’il est possible pour un groupe de se définir en fonction de valeurs culturelles puisées dans un passé collectif qui s’adaptent constamment à la réalité actuelle.

Questa tesi è costituita dalla traduzione di alcuni estratti del romanzo "Chengwei Heping Fandian", dell'autrice cinese Chen Danyan, corredata da un commento traduttivo incentrato soprattutto sulla traduzione dei realia e dei chengyu dal cinese all'italiano. Il commento prende inoltre in considerazione le ambientazioni e gli aspetti storico-culturali che fanno da sfondo alle vicende del romanzo, in particolare l'Hotel della Pace di Shanghai e la Campagna dei "Cinque anti" portata avanti dal regime maoista.

Marine organic matter (OM) sinks from surface waters to the seafloor via the biological pump. Benthic communities, which use this sedimented OM as energy and carbon source, produce dissolved organic matter (DOM) in the process of remineralization, enriching the sediment porewater with fresh DOM compounds. We hypothesized that in the oligotrophic deep Arctic basin the molecular signal of freshly deposited primary produced OM is restricted to the surface sediment pore waters which should differ from bottom water and deeper sediment pore water in DOM composition. This study focused on: 1) the molecular composition of the DOM in sediment pore waters of the deep Eurasian Arctic basins, 2) whether the signal of marine vs. terrigenous DOM is represented by different compounds preserved in the sediment pore waters and 3) whether there is any relation between Arctic Ocean ice cover and DOM composition. Molecular data, obtained via 15 Tesla Fourier transform ion cyclotron resonance mass spectrometer, were correlated with environmental parameters by partial least square analysis. The fresher marine detrital OM signal from surface waters was limited to pore waters from < 5 cm sediment depth. The productive ice margin stations showed higher abundances of peptides, unsaturated aliphatics and saturated fatty acids formulae, indicative of fresh OM/pigments deposition, compared to northernmost stations which had stronger aromatic signals. This study contributes to the understanding of the coupling between the Arctic Ocean productivity and its depositional regime, and how it will be altered in response to sea ice retreat and increasing river runoff.

Through the processes of the biological pump, carbon is exported to the deep ocean in the form of dissolved and particulate organic matter. There are several ways by which downward export fluxes can be estimated. The great attraction of the 234Th technique is that its fundamental operation allows a downward flux rate to be determined from a single water column profile of thorium coupled to an estimate of POC/234Th ratio in sinking matter. We present a database of 723 estimates of organic carbon export from the surface ocean derived from the 234Th technique. Data were collected from tables in papers published between 1985 and 2013 only. We also present sampling dates, publication dates and sampling areas. Most of the open ocean Longhurst provinces are represented by several measurements. However, the Western Pacific, the Atlantic Arctic, South Pacific and the South Indian Ocean are not well represented. There is a variety of integration depths ranging from surface to 220m. Globally the fluxes ranged from -22 to 125 mmol of C/m**2/d. We believe that this database is important for providing new global estimate of the magnitude of the biological carbon pump.
