The runtime management of the infrastructure providing service-based systems is a complex task, up to the point where manual operation struggles to be cost effective. As the functionality is provided by a set of dynamically composed distributed services, in order to achieve a management objective multiple operations have to be applied over the distributed elements of the managed infrastructure. Moreover, the manager must cope with the highly heterogeneous characteristics and management interfaces of the runtime resources. With this in mind, this paper proposes to support the configuration and deployment of services with an automated closed control loop. The automation is enabled by the definition of a generic information model, which captures all the information relevant to the management of the services with the same abstractions, describing the runtime elements, service dependencies, and business objectives. On top of that, a technique based on satisfiability is described which automatically diagnoses the state of the managed environment and obtains the required changes for correcting it (e.g., installation, service binding, update, or configuration). "No son necesarios imponentes edificios para dar una buena educación a los niños, mucho menos en zonas de clima suave. En el pasado Filósofos y Santos acostumbraban a sentarse con sus discípulos a la sombra de un árbol, transmitiéndoles su sabiduría sin necesidad de edificaciones de hormigón armado. Pero eran grandes hombres y grandes espíritus que sabían aprovechar el universo entero como material didáctico junto a los simples recursos de su inteligencia y su fantasía". Esta tesis nace con la intención de profundizar en la investigación de los mecanismos arquitectónicos que hicieron posible en un determinado tipo de escuelas la relación entre arquitectura y naturaleza, ya se entienda ésta como paisaje natural o como paisaje artificial creado ex novo. Si desde los tiempos de Lao Tse no había sido superada su definición de Arquitectura: “Arquitectura no son cuatro paredes y un tejado, arquitectura es el ordenamiento de los espacios y el espíritu que se genera dentro”; en realidad dicha definición adolecía de una gran carencia, pues nada decía del “espacio que queda fuera”. Así lo puso de manifiesto D. Rafael de La Hoz Arderius en su discurso de ingreso a la Real Academia de San Fernando5. Hubo que esperar al inicio del siglo XX para que la Arquitectura occidental se centrara de lleno en desmaterializar el límite entre el espacio construido y el “sitio” en el que se inserta, convirtiendo éste en “lugar” habitado. El “dentro” y el “fuera” dejan de entenderse como dos realidades antagónicas para dejar paso a un espacio continuo articulado a través de fructíferas situaciones intermedias. Sin embargo, poco se ha estudiado sobre una tipología arquitectónica : la escuela al aire libre, que fue crucial en la génesis tanto de los espacios educativos, como en la conformación del espacio Moderno así entendido. Éste es por tanto el objeto de la presente Tesis, desde una doble vertiente: por un lado desde la investigación de la evolución de esta tipología en general, y más detenidamente de un caso concreto, el colegio de las Teresianas en Alicante de Rafael de La Hoz Arderius y Gerardo Olivares James. La evolución de la escuela al aire libre se aborda a través de una selección de casos de estudio que ilustran que la regeneración social que pretendía acometerse no podía limitarse sólo a los aspectos higiénicos que centraron su primera etapa, sino que era necesario también reforzar el espíritu comunitario del niño como futuro ciudadano. Por otro lado el Colegio de las Teresianas en Alicante (1964) de Rafael de La Hoz Arderius y Gerardo Olivares James se ha elegido como caso de estudio específico. Este proyecto, siendo uno de los más desconocidos de sus autores, supone la culminación de sus investigaciones en torno a la escuela al aire libre8. Rafael de La Hoz, en línea con los postulados humanistas del Realismo Biológico de Richard Neutra, advertía de la imposibilidad de abordar la ordenación del espacio si desconocemos el proceso perceptivo del ser humano, destinatario de la Arquitectura. Esta dificultad es aún mayor si cabe cuando el destinatario no es el ser humano adulto sino el niño, dada su distinta percepción del binomio “espacio-tiempo”. En este sentido el colegio de las Teresianas en Alicante es además un ejemplo cercano, el único de los incluidos en la presente Tesis del que verdaderamente se ha podido tener un conocimiento profundo tanto por el resultado de su análisis a partir de una investigación de carácter científico, como por la experiencia personal del mismo vivida desde niña, al ser antigua alumna del centro. Tanto en este ejemplo concreto como en el resto de casos analizados la metodología para lograr la educación integral del individuo, reproduciendo el mito de la caverna de Platón revisado a través del Emilio roussoniano, se fundamenta en el contacto directo con el exterior, promoviendo un nuevo modo de vida equilibrado y en armonía con la naturaleza, con uno mismo y con los demás. Desde un primer estadio en el que el espacio exterior sustituye literalmente al aula como lugar para la enseñanza, se evoluciona hacia una tipología más compleja en la que los mecanismos de proyecto habrán de fomentar la continuidad entre interior y exterior en los espacios de aprendizaje, así como reproducir en el interior del aula las ventajas del ambiente exterior evitando algunos de sus inconvenientes. Todo ello con diferentes matices según la edad del alumno y la climatología del lugar. A partir del análisis de los casos de estudio generales y del ejemplo concreto de las Teresianas, se pretende sintetizar cuales fueron los mecanismos de proyecto y los principales temas de reflexión que caracterizaron este tipo de escuelas. ABSTRACT "Imposing buildings are not necessary for children to receive a good education, even less in mild climate areas. In the past, Philosophers and Saints used to sit with their disciples in the shade of a tree, passing on their wisdom without the need of reinforced concrete buildings. But they were great men and great minds who could take advantage of the entire universe as a source of teaching material, together with their intelligence and fantasy." This thesis was undertaken with the purpose of carrying out an in depth analysis of the architectural strategies targeting certain types of schools which have a close relationship between architecture and nature. It is said that since the time of Lao Tzu his definition of architecture had not been surpassed: “architecture is not just four walls and a roof, architecture is the arrangement of the spaces and the spirit that is generated within”. But this definition suffered from a serious lack as the “space left outside” is not mentioned. This was exposed by Rafael de La Hoz Arderius in his speech of entry into the Royal Academy of San Fernando10. It was not until the early twentieth century that Western architecture would squarely focus on dematerializing the boundary between the built environment and the “site” in which it is inserted, turning it into an inhabited “place”. The “inside” and the “outside” are no longer understood as two op-posed realities, instead they make way for a continuous space articulated through fruitful in-between situations. However, little has been studied about an architectural typology: the open air school, which was a turning point in the genesis of both educational, as well as modern space. This is therefore the object of this thesis, having two perspectives. On the one hand the development of this type of school is broadly investigated; on the other hand a specific case is introduced: the school of the Teresian association of Alicante, by Rafael de La Hoz and Gerardo Olivares. The development of the open air school is approached through a selection of case studies. These illustrate that the expected social regeneration could not be limited exclusively by the hygienic aspects targeting its first stage, but it was also necessary to strengthen the community spirit of the child as a future citizen. As previously mentioned the Teresian school of Alicante (1964-1966), has been chosen as a specific case study. Despite being quite a bit less renowned than other projects by the same authors, it represents the culmination of their researches about the open air school. In line with the humanist postulates of Richard Neutra’s Biological Realism, Rafael de La Hoz warned about the inability to deal with the arrangement of space if we are unaware of the perceptive process of the human being, addressee of the architecture. This difficulty becomes greater when the addressee is not the adult human but the child, given his different perception of the binomial “space-time” relationship. In this respect the Teresian school of Alicante is in addition a closely related case study, being the only one of the mentioned cases in this thesis allowing to acquire a deep knowledge, both from the results of its analysis coming from a research of scientific nature, as well as the personal experience lived since I was a child, given that I am a former pupil. Both in this case study and in the other analyzed cases, the methodology implemented to achieve the integral education of the individual is based on the direct contact with the exterior, promoting a balanced and in harmony with nature new way of life, including oneself and the others. Thereby it replicates the Plato’s cavern myth and its roussonian review: Emilio. From the first stage in which the exterior literally substitutes the classroom as the educational space, it is evolved towards a more complex typology in which the project strategies have to promote the continuity between inside and outside learning spaces, as well as to reproduce inside the classroom the advantages of the exterior environment avoiding some of its disadvantages; thereto considering the differing matrixes involving the pupil age and the local climatology. From the analysis of the general case studies and the Teresian school, the main project strategies and elements characterizing the open air school have been synthesized.

Essa pesquisa objetiva a análise da relação entre religião e política, em perspectiva de gênero considerando a atuação de parlamentares evangélicos/as na 54ª Legislatura (de 2011 a 2014) e a forma de intervenção desses atores no espaço político brasileiro quanto à promulgação de leis e ao desenvolvimento de políticas públicas que contemplem, dentre outras, a regulamentação do aborto, a criminalização da homofobia, a união estável entre pessoas do mesmo sexo e os desafios oriundos dessa posição para o Estado Brasileiro que se posiciona como laico. Ora, se laico remete à ideia de neutralidade estatal em matéria religiosa, legislar legitimado por determinados princípios fundamentados em doutrinas religiosas, pode sugerir a supressão da liberdade e da igualdade, o não reconhecimento da diversidade e da pluralidade e a ausência de limites entre os interesses públicos / coletivos e privados / particulares. Os procedimentos metodológicos para o desenvolvimento dessa pesquisa fundamentam-se na análise e interpretação bibliográfica visando estabelecer a relação entre religião e política, a conceituação, qualificação e tipificação do fenômeno da laicidade; levantamento documental; análise dos discursos de parlamentares evangélicos/as divulgados pela mídia, proferidos no plenário e adotados para embasar projetos de leis; pesquisa qualitativa com a realização de entrevistas e observações das posturas públicas adotadas pelos/as parlamentares integrantes da Frente Parlamentar Evangélica - FPE. Porquanto, os postulados das Ciências da Religião devidamente correlacionados com a interpretação do conjunto de dados obtidos no campo de pesquisa podem identificar o lugar do religioso na sociedade de forma interativa com as interfaces da laicidade visando aprofundar a compreensão sobre a democracia, sobre o lugar da religião nas sociedades contemporâneas e sobre os direitos difusos, coletivos e individuais das pessoas.

A presente dissertação analisa como o Partido Social Cristão (PSC), ao longo do tempo, se apropriou da identidade religiosa de seus atores políticos que na sua maioria são membros da Frente Parlamentar Evangélica, os quais defendem no espaço público a “família tradicional”, em detrimento da pluralidade de arranjos familiares na contemporaneidade. Para explicitar o objeto - “família tradicional” e PSC -, foi necessário retroceder no tempo e investigar na historiografia os primórdios da inserção dos evangélicos na política brasileira. Em vista disso, analisamos a participação dos evangélicos nos respectivos períodos do Brasil: Colônia, Império e República. A dificuldade da entrada de evangélicos na política partidária, dentre outros fatores, se deve àinfluência do catolicismo no Estado. Assim sendo, averiguamos em todas as Constituições (1824, 1891, 1934, 1937, 1946, 1967, 1969 e 1988) o que a mesma diz no que tange a proibição e a liberdade religiosa no país. Logo, verificamos entre as Eras Vargas e República Populista, que ocorreu com intensidade a transição do apoliticismo para o politicismo entre os evangélicos brasileiros, porém, eles não recebiam o apoio formal de suas igrejas. Em seguida, a participação dos evangélicos na arena política durante a ditadura militar foi investigada com destaque para o posicionamento de vanguarda da IECLB, através do Manifesto de Curitiba e, também com a presença de parlamentares evangélicos no Congresso Nacional. A politização pentecostal é ressaltada em nosso trabalho, através do pioneirismo de Manoel de Mello e, depois na Redemocratização quando as instituições evangélicas se organizaram para eleger seus candidatos à Assembleia Nacional Constituinte. E, com o fim do regime militar, o PSC surge como partido “nanico”, contudo, deixa o anonimato e ganha visibilidade midiática quando o pastor e deputado, Marco Feliciano, assume a presidência da Comissão de Direitos Humanos e Minorias, em 2013. Esse é o pano de fundo histórico que projetou o PSC e seus atores no pleito de 2014 com o mote “família tradicional”.

Defined model systems consisting of physiologically spaced arrays of H3/H4 tetramer⋅5S rDNA complexes have been assembled in vitro from pure components. Analytical hydrodynamic and electrophoretic studies have revealed that the structural features of H3/H4 tetramer arrays closely resemble those of naked DNA. The reptation in agarose gels of H3/H4 tetramer arrays is essentially indistinguishable from naked DNA, the gel-free mobility of H3/H4 tetramer arrays relative to naked DNA is reduced by only 6% compared with 20% for nucleosomal arrays, and H3/H4 tetramer arrays are incapable of folding under ionic conditions where nucleosomal arrays are extensively folded. We further show that the cognate binding sites for transcription factor TFIIIA are significantly more accessible when the rDNA is complexed with H3/H4 tetramers than with histone octamers. These results suggest that the processes of DNA replication and transcription have evolved to exploit the unique structural properties of H3/H4 tetramer arrays.

The c-Jun N-terminal kinase (JNK), or stress-activated protein kinase plays a crucial role in cellular responses stimulated by environmental stress and proinflammatory cytokines. However, the mechanisms that lead to the activation of the JNK pathway have not been elucidated. We have isolated a cDNA encoding a novel protein kinase that has significant sequence similarities to human germinal center kinase (GCK) and human hematopoietic progenitor kinase 1. The novel GCK-like kinase (GLK) has a nucleotide sequence that encodes an ORF of 885 amino acids with 11 kinase subdomains. Endogenous GLK could be activated by UV radiation and proinflammatory cytokine tumor necrosis factor α. When transiently expressed in 293 cells, GLK specifically activated the JNK, but not the p42/44MAPK/extracellular signal-regulated kinase or p38 kinase signaling pathways. Interestingly, deletion of amino acids 353–835 in the putative C-terminal regulatory region, or mutation of Lys-35 in the putative ATP-binding domain, markedly reduced the ability of GLK to activate JNK. This result indicates that both kinase activity and the C-terminal region of GLK are required for maximal activation of JNK. Furthermore, GLK-induced JNK activation could be inhibited by a dominant-negative mutant of mitogen-activated protein kinase kinase kinase 1 (MEKK1) or mitogen-activated protein kinase kinase 4/SAPK/ERK kinase 1 (SEK1), suggesting that GLK may function upstream of MEKK1 in the JNK signaling pathway.

Spectrin is an important structural component of the plasma membrane skeleton. Heretofore-unidentified isoforms of spectrin also associate with Golgi and other organelles. We have discovered another member of the β-spectrin gene family by homology searches of the GenBank databases and by 5′ rapid amplification of cDNA ends of human brain cDNAs. Collectively, 7,938 nucleotides of contiguous clones are predicted to encode a 271,294-Da protein, called βIII spectrin, with conserved actin-, protein 4.1-, and ankyrin-binding domains, membrane association domains 1 and 2, a spectrin dimer self-association site, and a pleckstrin-homology domain. βIII spectrin transcripts are concentrated in the brain and present in the kidneys, liver, and testes and the prostate, pituitary, adrenal, and salivary glands. All of the tested tissues contain major 9.0-kb and minor 11.3-kb transcripts. The human βIII spectrin gene (SPTBN2) maps to chromosome 11q13 and the mouse gene (Spnb3) maps to a syntenic region close to the centromere on chromosome 19. Indirect immunofluorescence studies of cultured cells using antisera specific to human βIII spectrin reveal a Golgi-associated and punctate cytoplasmic vesicle-like distribution, suggesting that βIII spectrin associates with intracellular organelles. This distribution overlaps that of several Golgi and vesicle markers, including mannosidase II, p58, trans-Golgi network (TGN)38, and β-COP and is distinct from the endoplasmic reticulum markers calnexin and Bip. Liver Golgi membranes and other vesicular compartment markers cosediment in vitro with βIII spectrin. βIII spectrin thus constitutes a major component of the Golgi and vesicular membrane skeletons.

Transmissible spongiform encephalopathies (TSEs) are lethal, infectious disorders of the mammalian nervous system. A TSE hallmark is the conversion of the cellular protein PrPC to disease-associated PrPSc (named for scrapie, the first known TSE). PrPC is protease-sensitive, monomeric, detergent soluble, and primarily α-helical; PrPSc is protease-resistant, polymerized, detergent insoluble, and rich in β-sheet. The “protein-only” hypothesis posits that PrPSc is the infectious TSE agent that directly converts host-encoded PrPC to fresh PrPSc, harming neurons and creating new agents of infection. To gain insight on the conformational transitions of PrP, we tested the ability of several protein chaperones, which supervise the conformational transitions of proteins in diverse ways, to affect conversion of PrPC to its protease-resistant state. None affected conversion in the absence of pre-existing PrPSc. In its presence, only two, GroEL and Hsp104 (heat shock protein 104), significantly affected conversion. Both promoted it, but the reaction characteristics of conversions with the two chaperones were distinct. In contrast, chemical chaperones inhibited conversion. Our findings provide new mechanistic insights into nature of PrP conversions, and provide a new set of tools for studying the process underlying TSE pathogenesis.

We have achieved, to our knowledge, the first high-level heterologous expression of the gene encoding d-ribulose-5-phosphate 3-epimerase from any source, thereby permitting isolation and characterization of the epimerase as found in photosynthetic organisms. The extremely labile recombinant spinach (Spinacia oleracea L.) enzyme was stabilized by dl-α-glycerophosphate or ethanol and destabilized by d-ribulose-5-phosphate or 2-mercaptoethanol. Despite this lability, the unprecedentedly high specific activity of the purified material indicates that the structural integrity of the enzyme is maintained throughout isolation. Ethylenediaminetetraacetate and divalent metal cations did not affect epimerase activity, thereby excluding a requirement for the latter in catalysis. As deduced from the sequence of the cloned spinach gene and the electrophoretic mobility under denaturing conditions of the purified recombinant enzyme, its 25-kD subunit size was about the same as that of the corresponding epimerases of yeast and mammals. However, in contrast to these other species, the recombinant spinach enzyme was octameric rather than dimeric, as assessed by gel filtration and polyacrylamide gel electrophoresis under nondenaturing conditions. Western-blot analyses with antibodies to the purified recombinant enzyme confirmed that the epimerase extracted from spinach leaves is also octameric.

All cloned members of the mammalian Na+/H+ exchanger gene family encode proteins that consist of two functionally distinct domains: a membrane-bound N terminus and a cytoplasmic C terminus, which are required for ion transport and regulation of transport, respectively. Despite their similarity in structure, three members of this family, designated NHE1, NHE2, and NHE3, exhibit different kinetic mechanisms in response to growth factors and protein kinases. For instance, growth factors stimulate NHE1 by a change in the affinity constant for intracellular H+, K'(Hi+), and regulate NHE2 and NHE3 by a change in Vmax. We have constructed chimeric Na+/H+ exchangers by exchanging the N and C termini among three cloned rabbit Na+/H+ exchangers (NHE1 to NHE3) to determine which domain is responsible for the above Vmax-vs.-K'(H(i)+) effect of the Na+/H+ isoforms. All of the chimeras had functional exchange activity and basal kinetic properties similar to those of wild-type exchangers. Studies with serum showed that the N terminus is responsible for the Vmax-vs.-K'(H(i)+) stimulation of the Na+/H+ exchanger isoforms. Moreover, phorbol 12-myristate 13-acetate and fibroblast growth factor altered Na+/H+ exchange only in chimeras that had an epithelial N-terminal domain matched with an epithelial C-terminal domain. Therefore, the protein kinase-induced regulation of Na+/H+ exchangers is mediated through a specific interaction between the N- and C-termini, whcih is restricted so that epithelial N- and epithelial N-and C-terminal portions of the exchangers are required for regulation.
