Due to the advances in sensor networks and remote sensing technologies, the acquisition and storage rates of meteorological and climatological data increases every day and ask for novel and efficient processing algorithms. A fundamental problem of data analysis and modeling is the spatial prediction of meteorological variables in complex orography, which serves among others to extended climatological analyses, for the assimilation of data into numerical weather prediction models, for preparing inputs to hydrological models and for real time monitoring and short-term forecasting of weather.In this thesis, a new framework for spatial estimation is proposed by taking advantage of a class of algorithms emerging from the statistical learning theory. Nonparametric kernel-based methods for nonlinear data classification, regression and target detection, known as support vector machines (SVM), are adapted for mapping of meteorological variables in complex orography.With the advent of high resolution digital elevation models, the field of spatial prediction met new horizons. In fact, by exploiting image processing tools along with physical heuristics, an incredible number of terrain features which account for the topographic conditions at multiple spatial scales can be extracted. Such features are highly relevant for the mapping of meteorological variables because they control a considerable part of the spatial variability of meteorological fields in the complex Alpine orography. For instance, patterns of orographic rainfall, wind speed and cold air pools are known to be correlated with particular terrain forms, e.g. convex/concave surfaces and upwind sides of mountain slopes.Kernel-based methods are employed to learn the nonlinear statistical dependence which links the multidimensional space of geographical and topographic explanatory variables to the variable of interest, that is the wind speed as measured at the weather stations or the occurrence of orographic rainfall patterns as extracted from sequences of radar images. Compared to low dimensional models integrating only the geographical coordinates, the proposed framework opens a way to regionalize meteorological variables which are multidimensional in nature and rarely show spatial auto-correlation in the original space making the use of classical geostatistics tangled.The challenges which are explored during the thesis are manifolds. First, the complexity of models is optimized to impose appropriate smoothness properties and reduce the impact of noisy measurements. Secondly, a multiple kernel extension of SVM is considered to select the multiscale features which explain most of the spatial variability of wind speed. Then, SVM target detection methods are implemented to describe the orographic conditions which cause persistent and stationary rainfall patterns. Finally, the optimal splitting of the data is studied to estimate realistic performances and confidence intervals characterizing the uncertainty of predictions.The resulting maps of average wind speeds find applications within renewable resources assessment and opens a route to decrease the temporal scale of analysis to meet hydrological requirements. Furthermore, the maps depicting the susceptibility to orographic rainfall enhancement can be used to improve current radar-based quantitative precipitation estimation and forecasting systems and to generate stochastic ensembles of precipitation fields conditioned upon the orography.

Seit den 1990er Jahren werden zunehmend nachhaltige Quartiere realisiert. Dabei besteht häufig eine beachtliche Diskrepanz zwischen den Zielen, die von den beteiligten Akteuren angestrebt werden, deren Umsetzung (Realisierungsphase) und deren Erhalt auf Dauer (Nutzungsphase). Es stellt sich folglich die Frage, auf welche Weise die Projektqualität im Sinne einer nachhaltigen Quartiersentwicklung verbessert werden kann. Diese Projekte sind jedoch enorm komplex aufgrund der großen Interdisziplinarität und Interdependenz ihrer Ziele sowie der vielschichtigen Akteursstrukturen. Sie stellen daher be-sonders hohe Anforderungen an die Projektsteuerung. Das konkrete Ziel dieser Arbeit besteht darin, die Bedeutung einer Prozesssteuerung im Sinne von Urban Governance zur Realisierung und zum Erhalt nachhaltiger Quartiere zu untersuchen. Damit soll einen Beitrag zur Förderung einer nachhalti-gen Stadtentwicklung geleistet werden. Die Arbeit stützt sich auf ein umfassendes theoretisches Fundament zum Thema Governance, wobei die relevanten Elemente für den Kontext nachhaltiger Quartiere herausgearbeitet werden. Die Hypothesen prüfen die Bedeutung der Schlüsselcharakteristika von Urban Governance (Kooperation, Partizipation, Verhandlungen) für die Projektqualität während der Realisierungs- und Nutzungsphase. Eine erste empirische Untersuchung wurde an zwanzig europäischen nachhaltigen Modellquartieren vorgenommen. Stärken und Schwächen aus der Perspektive der Nachhaltigkeit werden analysiert, deren Ursachen identifiziert und Handlungsoptio-nen aufgezeigt. Die Erkenntnisse zeigen die Notwendigkeit einer Verbesserung der Projektsteuerung während der Realisierungs- und der Nutzungsphase. Auf der Grundlage dieser Erkenntnisse wird ein umfassender Ansatz zur empirischen Untersuchung von Urban Governance im Kontext nachhaltiger Quartiere entwickelt. Dieser beruht auf dem akteurzentrierten Institutionalismus und den Merkmalen der Urban Governance. Anhand dieses Ansatzes wird mithilfe von Experteninterviews der Realisierungsprozess des nach-haltigen Quartiers Kronsberg (Hannover) analysiert. Betrachtet werden dabei die beteiligten Akteure und ihre Handlungso-rientierungen, die verwendeten Schlüsselinstrumente sowie aufgetretene Divergenzen zwischen Akteuren und deren Auswirkungen auf die Projekt- und Prozessqualität. Eine Vertiefung relevanter Themenfelder wird anhand der Fallstudie Neu-Oerlikon (Zürich) vorgenommen. Diese empirische Arbeit zeigt, dass eine Prozesssteuerung im Sinne von Urban Governance im Vergleich zu einer klassis-chen hierarchischen Steuerung eine notwendige aber nicht hinreichende Bedingung zur Verbesserung der Projektqualität nachhaltiger Quartiere darstellt. An konkreten Beispielen wird herausgearbeitet, dass der Mehrwert einer solchen Steuerung nur unter bestimmten Voraussetzungen erzielt werden kann: In manchen Situationen ist die Steuerungsform Kooperation und die Interaktionsform Verhandlung in ihrer Wirksamkeit zur Sicherung der Projektqualität begrenzt und hierarchische Interventionen sind notwendig. Nicht ein bestimmtes Steuerungsmodell per se ist geeignet, sondern es kommt auf den Ein-zelfall an: auf die Akteursstruktur, die individuellen und institutionellen Handlungsorientierungen der Akteure und deren Ver-haltensweisen, die Rahmenbedingungen und die Ausgestaltung des Urban Governance-Prozesses. Wenn die Spielregeln dieses Prozesses von den Akteuren nicht wirklich angenommen und gelebt werden, dominieren individuelle und institutio-nelle Akteursinteressen zu Lasten der Projektqualität. Ferner zeigen die Untersuchungen, dass die Partizipation der zukünftigen Quartiersnutzer in der Praxis häufig unzureichend ist. Dies führt zu Einbußen in der Projektqualität. Entscheidend ist auf jeden Fall, dass mindestens ein Akteur, in der Regel die öffentliche Hand, präsent ist, der die Definition anspruchsvoller Nachhaltigkeitsstandards, deren Umsetzung und deren Erhalt sichert sowie die notwendigen Rahmenbedingungen dafür schafft. Diese Arbeit belegt darüber hinaus, dass der Erhalt der Projektqualität während der Nutzungsphase (Faktor Zeit) bisher un-zureichend beachtet und in die Projektplanung einbezogen wird. Gerade dieser Aspekt bestimmt aber, ob das Quartier auch auf Dauer dem Nachhaltigkeitsanspruch gerecht werden kann! Tatsächlich handelt es sich um einen fortlaufenden Prozess, der nicht mit der Einweihung des Quartiers abgeschlossen ist. Vor diesem Hintergrund werden relevante Handlungsfelder beschrieben und die Notwendigkeit der langfristigen Fortsetzung einer Steuerung im Sinne von Urban Governance bzw. der Herausbildung einer Urban Governance-Kultur aufgezeigt. Aus den empirischen Erhebungen werden Erfolgs- und Risikofaktoren für Urban Governance-Prozesse während der Realisierungs- und der Nutzungsphase abgeleitet. Ferner werden bisher vernachlässigte Handlungsfelder (langfristiges Umwelt-management, ökologische Finanzierungsformen, urbane Landwirtschaft, Umweltkommunikation, etc.) eruiert. Die Berücksichtigung dieser Erkenntnisse ist unerlässlich für eine Verbesserung der Projektqualität nachhaltiger Quartiere. ---------------------------------------------- Gouvernance urbaine et quartiers durables: Entre intensions et mise en oeuvre --- Résumé --- Depuis les années 90, la thématique des quartiers durables a gagné en importance, même si leur développement s'est avéré difficile. Le décalage entre les objectifs, leur mise en oeuvre et le projet tel qu'il est vécu par ses habitants est souvent important et nécessite d'être réduit. Un quartier durable est par nature un projet complexe, aux objectifs ambitieux situé à la croisée de multiples champs disciplinaires, mobilisant de nombreux acteurs aux intérêts divergents. De plus, chaque projet, du fait des ses spécificités, requiert un pilotage adapté. L'objectif principal de la recherche vise à analyser la nature du pilotage du processus de conception, de réalisation et d'exploitation des quartiers durables. Ses résultats ont pour ambition de contribuer à optimiser et promouvoir le développement urbain durable. Le fondement théorique de la recherche se base sur le concept de gouvernance urbaine, adapté au contexte particulier de la gouvernance des quartiers durables. La gouvernance urbaine, au sens où nous l'entendons, est un mode de pilotage basé sur la coopération entre les acteurs publics et privés. Les hypothèses centrales du travail testent la portée et les limites des caractéristiques-clefs de la gouvernance urbaine (coopération, participation, négociation), ainsi que l'importance de la notion de pérennité pour la qualité du projet. Dans un premier temps, nous avons analysé vingt quartiers durables modèles européens et identifié leurs atouts et leurs faiblesses en termes de durabilité, ainsi que leurs divers modes de pilotage. Les enseignements tirés de ces exemples révèlent la nécessité d'améliorer le pilotage des projets. Dans un deuxième temps, nous avons élaboré une grille d'analyse fine fondée sur l'approche institutionnelle des acteurs et les caractéristiques-clefs de la gouvernance urbaine. En nous appuyant sur cette grille, nous avons analysé le processus de conception et de réalisation du quartier durable de « Kronsberg » (Hanovre) à l'aide des éléments suivants : les acteurs (avec leurs intérêts et objectifs propres), les instruments d'aménagement du territoire, les modes de pilotage, les zones de divergence et de convergence entre les acteurs, ainsi que leurs impacts sur le processus et le projet. Dans un troisième temps, les hypothèses centrales ont été testées sur le quartier de « Neu-Oerlikon » (Zurich) afin d'approfondir et d'élargir les enseignements tirés de celui de « Kronsberg ». Les résultats des analyses mettent en évidence le fait qu'un pilotage de projet selon le modèle de la gouvernance urbaine est certes une condition nécessaire mais non suffisante pour améliorer la qualité du projet. De plus, la valeur ajoutée de la gouvernance urbaine n'est valable qu'à certaines conditions. En effet, la coopération et la négociation peuvent même, dans certaines situations, réduire la qualité du projet ! Le principal enseignement de la recherche révèle qu'il n'y a pas de mode de pilotage idéal, mais que la qualité d'un projet dépend d'une multitude de facteurs, tels que les constellations d'acteurs, leurs intérêts personnels et institutionnels, les conditions cadres et les « règles du jeu » de la gouvernance urbaine. Si les « règles du jeu » en particulier ne sont pas réellement appropriées par l'ensemble des acteurs, les intérêts et les comportements personnels ou institutionnels prédominent au détriment de la qualité du projet. De même, si la participation des futurs usagers à l'élaboration du projet de quartier durable n'est pas assurée, tant la qualité du projet que sa pérennité en pâtissent. Nous avons également constaté que la présence d'un acteur (en règle générale les autorités publiques) qui veille à la définition d'objectifs ambitieux en matière de développement durable et à leur application constitue un apport essentiel à la qualité du projet. En outre, la recherche met en évidence les carences dans le suivi et le maintien à long terme des qualités de durabilité de la phase d'exploitation des projets de quartiers durables analysés. Dans la phase d'exploitation, le degré de coopération diminue généralement et les modes de fonctionnement et de pilotage sectoriels se mettent en place au détriment de la qualité du projet. Cela confirme la nécessité de poursuivre le processus de pilotage selon le modèle de la gouvernance urbaine au-delà de la phase de réalisation des projets. La recherche précise les enjeux des champs d'action de la phase d'exploitation (domaine encore peu étudié) et démontre la pertinence du mode de pilotage préconisé. Enfin, les analyses permettent d'identifier des facteurs de réussite et de risque susceptibles d'influencer les systèmes de gouvernance urbaine, ainsi que les enjeux des domaines de la durabilité encore négligés (agriculture urbaine, gestion environnementale dans la durée, comportement des usagers, financement équitable, etc.). La prise en compte de ces enseignements est essentielle à l'amélioration de la gestion de futurs projets de quartiers durables. ---------------------------------------------- Urban Governance and Sustainable Neighbourhoods: A Contribution to a Lasting Sustainable Development --- Abstract --- Since the 1990s, sustainable neighbourhoods have become an increasingly important topic. However, their development has proven to be difficult. There is an often considerable gap, which must be reduced, between the initial goals, the way they are implemented and how the project is finally inhabited. A sustainable neighbourhood is inherently a complex project, with ambitious goals that lie at the intersection of multiple disciplines, involving numerous stakeholders with diverging interests. Moreover, each project, due to its specific characteristics, requires an adapted steering. The main goal of this research is to analyse the nature of the steering process during the planning, realisation and use of sustainable neighbourhoods. The results aim to contribute to the promotion of sustainable urban development. The theoretical foundation of this research is based on the concept of urban governance, adapted to the particular context of sustainable neighbourhoods. Urban governance is understood in this work, as a mode of project steering based on the cooperation between public and private stakeholders. The central hypotheses of this work test the importance and the limits of the key characteristics of urban governance (cooperation, participation, negotiation) as well as the importance of continuity for the project quality. To begin with, we surveyed and analysed twenty exemplary European sustainable neighbourhoods and identified their strengths and weaknesses in terms of sustainability, as well as their diverse steering modes. The lessons learned from these examples reveal the need to improve the projects' steering. Secondly we elaborated a detailed framework for analysis founded on stakeholder-centred institutionalism and the key characteristics of urban governance. By systematically applying this framework, we analysed the planning and implementation process of the sustainable neighbourhood "Kronsberg" (Hannover). Our focus was on the following dimensions: the stakeholders (with their particular interests and goals), the instruments of spatial planning, the steering modes, the points of divergence and convergence amongst the stakeholders, as well as their impacts on the process and on the project. The final step was to test the core hypotheses on the neighbourhood "Neu-Oerlikon" (Zürich) in order to broaden the lessons learned from "Kronsberg". The results of the analysis highlight the fact that an urban governance type project steering is certainly a necessary but insufficient condition to improve the project quality. Moreover, the added value of urban governance is only valid under certain conditions. In fact, cooperation and negotiation can even in certain situations reduce the project's quality! The main lesson of this research is that there is not an ideal steering mode, but rather that the quality of the project depends on numerous factors, such as the stakeholder constellation, their individual and institutional interests, the general conditions and the "rules of the game" of urban governance. If these "rules of the game" are not really appropriated by all stakeholders, individual and institutional interests and behaviours predominate at the expense of the project's quality. Likewise, if the future users' participation in the project development is insufficient, both the project's quality and its continuity suffer. We have also observed that the presence of a stakeholder (in general the public authorities) who ensures the definition of ambitious goals in terms of sustainable development and their implementation is crucial for the project's quality. Furthermore, this research highlights the deficiencies in the follow-up and long-term preservation of the sustainability qualities in the neighbourhood projects which we have analysed. In the use phase, the degree of cooperation generally diminishes. Attitudes and project management become more sectorial at the expense of the project's quality. This confirms the need to continue the steering process according to the principles of urban governance beyond the project's implementation phase. This research specifies the challenges that affect the use phase (a still neglected area) and shows the relevance of the recommended steering mode. Finally, the analyses also identify the success and risk factors that may influence urban-governance systems, as well as the challenges of still neglected fields of sustainability (urban agriculture, long-term environmental management, user behaviour, fair funding, etc.). Taking into account these outcomes is essential to improve the management of future sustainable-neighbourhood projects.

The efficacy of inoculation of single pure bacterial cultures into complex microbiomes, for example, in order to achieve increased pollutant degradation rates in contaminated material (that is, bioaugmentation), has been frustrated by insufficient knowledge on the behaviour of the inoculated bacteria under the specific abiotic and biotic boundary conditions. Here we present a comprehensive analysis of genome-wide gene expression of the bacterium Sphingomonas wittichii RW1 in contaminated non-sterile sand, compared with regular suspended batch growth in liquid culture. RW1 is a well-known bacterium capable of mineralizing dibenzodioxins and dibenzofurans. We tested the reactions of the cells both during the immediate transition phase from liquid culture to sand with or without dibenzofuran, as well as during growth and stationary phase in sand. Cells during transition show stationary phase characteristics, evidence for stress and for nutrient scavenging, and adjust their primary metabolism if they were not precultured on the same contaminant as found in the soil. Cells growing and surviving in sand degrade dibenzofuran but display a very different transcriptome signature as in liquid or in liquid culture exposed to chemicals inducing drought stress, and we obtain evidence for numerous 'soil-specific' expressed genes. Studies focusing on inoculation efficacy should test behaviour under conditions as closely as possible mimicking the intended microbiome conditions.

O objetivo deste trabalho foi utilizar o método Bayesiano no ajuste do modelo de Wood a dados de produção de leite de cabras da raça Saanen. Dois grupos de animais da primeira e segunda lactação foram considerados. Amostras das distribuições marginais a posteriori dos parâmetros do modelo de Wood e das funções de produção derivadas desses parâmetros - pico de produção, tempo do pico de produção, persistência e produção total de leite - foram obtidas pelo algoritmo Gibbs Sampler. As inferências foram feitas em cada população e os resultados mostraram diferenças na taxa de decréscimo da produção após o pico e na persistência, indicando maior produção nos animais de segunda lactação. Realizou-se um estudo de simulação de dados para avaliar o método Bayesiano sob diferentes estruturas de matrizes de covariâncias dos parâmetros. Os resultados desse estudo indicam que o método é eficiente no estudo das curvas de lactação quando a matriz de covariância apresenta alta correlação dos parâmetros.

Revision actual de las Uniones Temporales de Empresas (UTE), con el objeto de constatar si la informacion contable que presentan las empresas asociadas mediante esta formula es fidedigna y cual es su situacion ante la reforma contable. La investigacion consta de una parte teorica y una parte empirica. La parte teorica define la union temporal de empresas, sus caracteristicas y objetivos, el origen historico y la regulacion contable, fiscal y juridica que les afecta. Asimismo, se trata con especial relevancia la adaptacion sectorial del PGC a las Empresas Constructoras (PSEC). Por otra parte, se analizan las Normas Internacionales de Informacion Financiera (NIIF) relacionadas con las UTE. Finalmente, se comparan las normas contables españolas, internacionales y francesas referidas a los contratos de larga duracion. La parte empirica, resulta de un analisis de los estados financieras del periodo 2002-2004 de una poblacion de treinta empresas constructoras que habitualmente realizan UTE, la mayoria de las mismas forman parte del SEOPAN (Asociacion de Empresas Constructoras de ambito nacional).

Between 1995 and 2005, the Spanish economy grew at an annual average rate higher than 3,5%. Total employment increased by more than 4.9 millions. Most of this growth was in occupations related with university degrees (more than 890,000, 18% of the total employment increase) and vocational qualifications (more than 855,000, 17.5% of the total employment increase). From a sectoral perspective, the main part of this increase took place in “Real estate, renting and business activities” (K sector in NACE rev.1), “Construction” (F sector) and “Health and social sector” (N sector). This paper analyses this employment growth in an Input-output framework, by means of a structural decomposition analysis (SDA). Two kinds of results have been obtained. From a sectoral perspective we decompose employment growth into Labour requirements change, technical change and demand change. From an occupational perspective, we decompose the employment growth in substitutions effect, labour productivity effect and demand effect. The results show that, in aggregated terms, the main part of this growth is attributable to demand growth, with a small technical improvement. But the results also show that this aggregated behaviour hides important sectoral and occupational variation. The purpose of this paper is to contribute to the ongoing debate over productivity growth and what has been called the “growth model” for the Spanish economy.

A significant number of environmental microorganisms can cause serious, even fatal, acute and chronic infections in humans. The severity and outcome of each type of infection depends on the expression of specific bacterial phenotypes controlled by complex regulatory networks that sense and respond to the host environment. Although bacterial signals that contribute to a successful acute infection have been identified in a number of pathogens, the signals that mediate the onset and establishment of chronic infections have yet to be discovered. We identified a volatile, low molecular weight molecule, 2-amino acetophenone (2-AA), produced by the opportunistic human pathogen Pseudomonas aeruginosa that reduces bacterial virulence in vivo in flies and in an acute mouse infection model. 2-AA modulates the activity of the virulence regulator MvfR (multiple virulence factor regulator) via a negative feedback loop and it promotes the emergence of P. aeruginosa phenotypes that likely promote chronic lung infections, including accumulation of lasR mutants, long-term survival at stationary phase, and persistence in a Drosophila infection model. We report for the first time the existence of a quorum sensing (QS) regulated volatile molecule that induces bistability phenotype by stochastically silencing acute virulence functions in P. aeruginosa. We propose that 2-AA mediates changes in a subpopulation of cells that facilitate the exploitation of dynamic host environments and promote gene expression changes that favor chronic infections.

Background: The ratio of the rates of non-synonymous and synonymous substitution (d(N)/d(S)) is commonly used to estimate selection in coding sequences. It is often suggested that, all else being equal, d(N)/d(S) should be lower in populations with large effective size (Ne) due to increased efficacy of purifying selection. As N-e is difficult to measure directly, life history traits such as body mass, which is typically negatively associated with population size, have commonly been used as proxies in empirical tests of this hypothesis. However, evidence of whether the expected positive correlation between body mass and d(N)/d(S) is consistently observed is conflicting. Results: Employing whole genome sequence data from 48 avian species, we assess the relationship between rates of molecular evolution and life history in birds. We find a negative correlation between dN/dS and body mass, contrary to nearly neutral expectation. This raises the question whether the correlation might be a method artefact. We therefore in turn consider non-stationary base composition, divergence time and saturation as possible explanations, but find no clear patterns. However, in striking contrast to d(N)/d(S), the ratio of radical to conservative amino acid substitutions (K-r/K-c) correlates positively with body mass. Conclusions: Our results in principle accord with the notion that non-synonymous substitutions causing radical amino acid changes are more efficiently removed by selection in large populations, consistent with nearly neutral theory. These findings have implications for the use of d(N)/d(S) and suggest that caution is warranted when drawing conclusions about lineage-specific modes of protein evolution using this metric.

We study energy relaxation in thermalized one-dimensional nonlinear arrays of the Fermi-Pasta-Ulam type. The ends of the thermalized systems are placed in contact with a zero-temperature reservoir via damping forces. Harmonic arrays relax by sequential phonon decay into the cold reservoir, the lower-frequency modes relaxing first. The relaxation pathway for purely anharmonic arrays involves the degradation of higher-energy nonlinear modes into lower-energy ones. The lowest-energy modes are absorbed by the cold reservoir, but a small amount of energy is persistently left behind in the array in the form of almost stationary low-frequency localized modes. Arrays with interactions that contain both a harmonic and an anharmonic contribution exhibit behavior that involves the interplay of phonon modes and breather modes. At long times relaxation is extremely slow due to the spontaneous appearance and persistence of energetic high-frequency stationary breathers. Breather behavior is further ascertained by explicitly injecting a localized excitation into the thermalized arrays and observing the relaxation behavior.

This work consists of three essays investigating the ability of structural macroeconomic models to price zero coupon U.S. government bonds. 1. A small scale 3 factor DSGE model implying constant term premium is able to provide reasonable a fit for the term structure only at the expense of the persistence parameters of the structural shocks. The test of the structural model against one that has constant but unrestricted prices of risk parameters shows that the exogenous prices of risk-model is only weakly preferred. We provide an MLE based variance-covariance matrix of the Metropolis Proposal Density that improves convergence speeds in MCMC chains. 2. Affine in observable macro-variables, prices of risk specification is excessively flexible and provides term-structure fit without significantly altering the structural parameters. The exogenous component of the SDF is separating the macro part of the model from the term structure and the good term structure fit has as a driving force an extremely volatile SDF and an implied average short rate that is inexplicable. We conclude that the no arbitrage restrictions do not suffice to temper the SDF, thus there is need for more restrictions. We introduce a penalty-function methodology that proves useful in showing that affine prices of risk specifications are able to reconcile stable macro-dynamics with good term structure fit and a plausible SDF. 3. The level factor is reproduced most importantly by the preference shock to which it is strongly and positively related but technology and monetary shocks, with negative loadings, are also contributing to its replication. The slope factor is only related to the monetary policy shocks and it is poorly explained. We find that there are gains in in- and out-of-sample forecast of consumption and inflation if term structure information is used in a time varying hybrid prices of risk setting. In-sample yield forecast are better in models with non-stationary shocks for the period 1982-1988. After this period, time varying market price of risk models provide better in-sample forecasts. For the period 2005-2008, out of sample forecast of consumption and inflation are better if term structure information is incorporated in the DSGE model but yields are better forecasted by a pure macro DSGE model.

In the root-colonizing biocontrol strain CHA0 of Pseudomonas fluorescens, cell density-dependent synthesis of extracellular, plant-beneficial secondary metabolites and enzymes is positively regulated by the GacS/GacA two-component system. Mutational analysis of the GacS sensor kinase using improved single-copy vectors showed that inactivation of each of the three conserved phosphate acceptor sites caused an exoproduct null phenotype (GacS-), whereas deletion of the periplasmic loop domain had no significant effect on the expression of exoproduct genes. Strain CHA0 is known to synthesize a solvent-extractable extracellular signal that advances and enhances the expression of exoproduct genes during the transition from exponential to stationary growth phase when maximal exoproduct formation occurs. Mutational inactivation of either GacS or its cognate response regulator GacA abolished the strain's response to added signal. Deletion of the linker domain of the GacS sensor kinase caused signal-independent, strongly elevated expression of exoproduct genes at low cell densities. In contrast to the wild-type strain CHA0, the gacS linker mutant and a gacS null mutant were unable to protect tomato plants from crown and root rot caused by Fusarium oxysporum f. sp. radicis-lycopersici in a soil-less microcosm, indicating that, at least in this plant-pathogen system, there is no advantage in using a signal-independent biocontrol strain.

We initiate a systematic scan of the landscape of black holes in any spacetime dimension using the recently proposed blackfold effective worldvolume theory. We focus primarily on asymptotically flat stationary vacuum solutions, where we uncover large classes of new black holes. These include helical black strings and black rings, black odd-spheres, for which the horizon is a product of a large and a small sphere, and non-uniform black cylinders. More exotic possibilities are also outlined. The blackfold description recovers correctly the ultraspinning Myers-Perry black holes as ellipsoidal even-ball configurations where the velocity field approaches the speed of light at the boundary of the ball. Helical black ring solutions provide the first instance of asymptotically flat black holes in more than four dimensions with a single spatial U(1) isometry. They also imply infinite rational non-uniqueness in ultraspinning regimes, where they maximize the entropy among all stationary single-horizon solutions. Moreover, static blackfolds are possible with the geometry of minimal surfaces. The absence of compact embedded minimal surfaces in Euclidean space is consistent with the uniqueness theorem of static black holes

"MotionMaker (TM)" is a stationary programmable test and training system for the lower limbs developed at the 'Ecole Polytechnique Federale de Lausanne' with the 'Fondation Suisse pour les Cybertheses'.. The system is composed of two robotic orthoses comprising motors and sensors, and a control unit managing the trans-cutaneous electrical muscle stimulation with real-time regulation. The control of the Functional Electrical Stimulation (FES) induced muscle force necessary to mimic natural exercise is ensured by the control unit which receives a continuous input from the position and force sensors mounted on the robot. First results with control subjects showed the feasibility of creating movements by such closed-loop controlled FES induced muscle contractions. To make exercising with the MotionMaker (TM) safe for clinical trials with Spinal Cord Injured (SCI) volunteers, several original safety features have been introduced. The MotionMaker (TM) is able to identify and manage the occurrence of spasms. Fatigue can also be detected and overfatigue during exercise prevented.

Professional cleaning is a basic service occupation with a wide variety of tasks carried out in all kind of different sectors and workplaces by a large workforce. One important risk for cleaning workers is the exposure to chemical substances that are present in cleaning products.Monoethanolamine was found to be often present in cleaning products such as general purpose cleaners, bathroom cleaners, floor cleaners and kitchen cleaners. Monoethanolamine can injure the skin, and exposure to monoethanolamine was associated to asthma even when the air concentrations were low. It is a strong irritant and known to be involved in sensitizing mechanisms. It is very likely that the use of cleaning products containing monoethanolamine gives rise to respiratory and dermal exposures. Therefore there is a need to further investigate the exposures to monoethanolamine for both, respiratory and dermal exposure.The determination of monoethanolamine has traditionally been difficult and analytical methods available are little adapted for occupational exposure assessments. For monoethanolamine air concentrations, a sampling and analytical method was already available and could be used. However, a method to analyses samples for skin exposure assessments as well as samples of skin permeation experiments was missing. Therefore one main objective of this master thesis was to search an already developed and described analytical method for the measurement of monoethanolamine in water solutions, and to set it up in the laboratory. Monoethanolamine was analyzed after a derivatisation reaction with o-pthtaldialdehyde. The derivated fluorescing monoethanolamine was then separated with high performance liquid chromatography and detection took place with a fluorescent detector. The method was found to be suitable for qualitative and quantitative analysis of monoethanolamine. An exposure assessment was conducted in the cleaning sector to measure the respiratory and dermal exposures to monoethanolamine during floor cleaning. Stationary air samples (n=36) were collected in 8 companies and samples for dermal exposures (n=12) were collected in two companies. Air concentrations (Mean = 0.18 mg/m3, Standard Deviation = 0.23 mg/m3, geometric Mean = 0.09 mg/m3, Geometric Standard Deviation = 3.50) detected were mostly below 1/10 of the Swiss 8h time weighted average occupational exposure limit. Factors that influenced the measured monoethanolamine air concentrations were room size, ventilation system and the concentration of monoethanolamine in the cleaning product and amount of monoethanolamine used. Measured skin exposures ranged from 0.6 to 128.4 mg/sample. Some cleaning workers that participated in the skin exposure assessment did not use gloves and had direct contact with the solutions containing the cleaning product and monoethanolamine. During the entire sampling campaign, cleaning workers mostly did not use gloves. Cleaning workers are at risk to be regularly exposed to low air concentrations of monoethanolamine. This exposure may be problematic if a worker suffers from allergic reactions (e.g. Asthma). In that case a substitution of the cleaning product may be a good prevention measure as several different cleaning products are available for similar cleaning tasks. Currently there are no occupational exposure limits to compare the skin exposures that were found. To prevent skin exposures, adaptations of the cleaning techniques and the use of gloves should be considered. The simultaneous skin and airborne exposures might accelerate adverse health effects. Overall the risks caused by exposures to monoethanolamine are considered as low to moderate when the cleaning products are used correctly. Whenever possible, skin exposures should be avoided. Further research should consider especially the dermal exposure routes, as very high exposures might occur by skin contact with cleaning products. Dermatitis but also sensitization might be caused by skin exposures. In addition, new biomedical insights are needed to better understand the risks of the dermal exposure. Therefore skin permeability experiments should be considered.

Abstract : Matrix metalloproteinases (MMPs) are thought to play a major role in the tumor dissemination process as they degrade all components of the extracellular matrix. However, failure of clinical trials testing broad MMP inhibitors in cancer led to the consensus that a better understanding of the MMP biology was required. Using intravital multiphoton laser scanning microscopy, we developed an in vivo model to observe tumor dissemination and extracellular matrix remodeling in real time. We show that the matrix-modifying hormone relaxin increases tumor associated fibroblast interaction with collagen fibers by inducing integrin beta-1 expression. This causes changes in the collagen network that are mediated by MMP-8 and MT1-MMP. Also, we show that MMP-mediated collagen remodeling in vivo requires a direct contact between stationary tumor associated fibroblasts (TAFs) and collagen fibers. As MMPs are expressed in the tumor and stromal compartment of breast cancers we determined the importance of Membrane-type 1 MMP (MT1-MMP) from each compartment for cancer progression. We find that tumor-MT1-MMP promotes the invasion of the blood vasculature and blood-borne metastasis in vivo by enhancing tumor cell migration and endothelial basement membrane degradation. Interestingly, stromal-MT1-MMP cannot compensate for the lack of tumor-MT1-MMP but promotes peritumor collagen I remodeling. Thus, the function of MT1-MMP is context dependent and we identify the different but complementary roles of tumor and stromal MT1-MMP for tumor dissemination. Finally, we translate our preclinical findings in to human breast cancer samples. We show that tumor-MT1-MMP expression correlates with tumor invasion of the blood vasculature in ER-PR-HER2- breast cancers and that MT1-MMP expression increases with cancer progression. MT1-MMP could thus represent an interesting therapeutic target for the prevention of blood vasculature invasion in these tumors. Resumé : Les matrix metalloproteinases (MMPs) semblent jouer un rôle majeur pour la dissémination tumorale en raison de leur capacité à dégrader l'ensemble des composants de la matrice extracellulaire (MEC). Néanmoins, les résultats décevants des études cliniques testant les inhibiteurs des MMP ont conduit à la notion qu'une compréhension plus précise de la biologie des MMP était requise. Dans ce travail de thèse, nous avons développé un modèle murin qui permet d'observer simultanément la dissémination tumorale ainsi que les modifications de la MEC en temps réel. Nous démontrons que le traitement de tumeurs par l'hormone relaxin augmente l'interaction des fibroblastes tumoraux avec les fibres de collagène via l'intégrine beta-1. Nous montrons que cette interaction favorise et est nécessaire à la dégradation des fibres de collagène par MMP-8 et MT1-MMP. Ensuite, étant donné que les MMPs sont exprimées dans les cellules tumorales et stromales des cancers du sein, nous nous sommes intéressés au rôle de la MMP membranaire type 1 (MT1-MMP) exprimée dans chacun de ces compartiments. Nous démontrons que MT1-MMP dérivant des cellules tumorales favorise leur invasion dans les vaisseaux sanguins par la dégradation de la membrane basale vasculaire. De manière inattendue, nous montrons que l'expression de MT1-MMP par le compartiment stromal ne peut compenser le manque de MT1-MMP dans le compartiment tumoral. Néanmoins, nos résultats prouvent que MT1-MMP dérivant du compartiment stromal est impliqué dans la dégradation de collagène peritumorale. La fonction de la protéine MT1-MMP varie donc selon le compartiment tumoral d'origine. Finalement, nous avons testé nos résultats pré cliniques chez l'humain. Dans des biopsies de cancer du sein nous montrons une corrélation entre l'expression de MT1-MMP dans les cellules tumorales et l'invasion de vaisseaux sanguins par des tumeurs ER-PR-HER2-. MT1-MMP pourrait donc être une cible intéressante pour la prévention de dissémination vasculaire de ces tumeurs
