This study investigates thermally induced tensile stresses in ceramic tilings. Daily and seasonal thermal cycles, as well as, rare but extreme events, such as a hail-storm striking a heated terrace tiling, were studied in the field and by numerical modeling investigations. The field surveys delivered temperature– time diagrams and temperature profiles across tiling systems. These data were taken as input parameters for modeling the stress distribution in the tiling system in order to detect potential sites for material failure. Dependent on the thermal scenario (e.g., slow heating of the entire structure during morning and afternoon, or a rapid cooling of the tiles by a rain storm) the modeling indicates specific locations with high tensile stresses. Typically regions along the rim of the tiling field showed stresses, which can become critical with respect to the adhesion strength. Over the years, ongoing cycles of thermal expansion–contraction result in material fatigue promoting the propagation of cracks. However, the installation of flexible waterproofing membranes (applied between substrate and tile adhesive) represents an efficient technical innovation to reduce such crack propagation as confirmed by both numerical modeling results and microstructural studies on real systems.

Understanding the preferential timescales of variability in the North Atlantic, usually associated with the Atlantic meridional overturning circulation (AMOC), is essential for the prospects for decadal prediction. However, the wide variety of mechanisms proposed from the analysis of climate simulations, potentially dependent on the models themselves, has stimulated the debate of which processes take place in reality. One mechanism receiving increasing attention, identified both in idealized models and observations, is a westward propagation of subsurface buoyancy anomalies that impact the AMOC through a basin-scale intensification of the zonal density gradient, enhancing the northward transport via thermal wind balance. In this study, we revisit a control simulation from the Institut Pierre-Simon Laplace Coupled Model 5A (IPSL-CM5A), characterized by a strong AMOC periodicity at 20 years, previously explained by an upper ocean–atmosphere–sea ice coupled mode driving convection activity south of Iceland. Our study shows that this mechanism interacts constructively with the basin-wide propagation in the subsurface. This constructive feedback may explain why bi-decadal variability is so intense in this coupled model as compared to others.

Today, there is little knowledge on the attitude state of decommissioned intact objects in Earth orbit. Observational means have advanced in the past years, but are still limited with respect to an accurate estimate of motion vector orientations and magnitude. Especially for the preparation of Active Debris Removal (ADR) missions as planned by ESA’s Clean Space initiative or contingency scenarios for ESA spacecraft like ENVISAT, such knowledge is needed. ESA's “Debris Attitude Motion Measurements and Modelling” project (ESA Contract No. 40000112447), led by the Astronomical Institute of the University of Bern (AIUB), addresses this problem. The goal of the project is to achieve a good understanding of the attitude evolution and the considerable internal and external effects which occur. To characterize the attitude state of selected targets in LEO and GTO, multiple observation methods are combined. Optical observations are carried out by AIUB, Satellite Laser Ranging (SLR) is performed by the Space Research Institute of the Austrian Academy of Sciences (IWF) and radar measurements and signal level determination are provided by the Fraunhofer Institute for High Frequency Physics and Radar Techniques (FHR). The In-Orbit Tumbling Analysis tool (ιOTA) is a prototype software, currently in development by Hyperschall Technologie Göttingen GmbH (HTG) within the framework of the project. ιOTA will be a highly modular software tool to perform short-(days), medium-(months) and long-term (years) propagation of the orbit and attitude motion (six degrees-of-freedom) of spacecraft in Earth orbit. The simulation takes into account all relevant acting forces and torques, including aerodynamic drag, solar radiation pressure, gravitational influences of Earth, Sun and Moon, eddy current damping, impulse and momentum transfer from space debris or micro meteoroid impact, as well as the optional definition of particular spacecraft specific influences like tank sloshing, reaction wheel behaviour, magnetic torquer activity and thruster firing. The purpose of ιOTA is to provide high accuracy short-term simulations to support observers and potential ADR missions, as well as medium-and long-term simulations to study the significance of the particular internal and external influences on the attitude, especially damping factors and momentum transfer. The simulation will also enable the investigation of the altitude dependency of the particular external influences. ιOTA's post-processing modules will generate synthetic measurements for observers and for software validation. The validation of the software will be done by cross-calibration with observations and measurements acquired by the project partners.

The movement of ions across specific channels embedded on the membrane of individual cardiomyocytes is crucial for the generation and propagation of the cardiac electric impulse. Emerging evidence over the past 20 years strongly suggests that the normal electric function of the heart is the result of dynamic interactions of membrane ion channels working in an orchestrated fashion as part of complex molecular networks. Such networks work together with exquisite temporal precision to generate each action potential and contraction. Macromolecular complexes play crucial roles in transcription, translation, oligomerization, trafficking, membrane retention, glycosylation, post-translational modification, turnover, function, and degradation of all cardiac ion channels known to date. In addition, the accurate timing of each cardiac beat and contraction demands, a comparable precision on the assembly and organizations of sodium, calcium, and potassium channel complexes within specific subcellular microdomains, where physical proximity allows for prompt and efficient interaction. This review article, part of the Compendium on Sudden Cardiac Death, discusses the major issues related to the role of ion channel macromolecular assemblies in normal cardiac electric function and the mechanisms of arrhythmias leading to sudden cardiac death. It provides an idea of how these issues are being addressed in the laboratory and in the clinic, which important questions remain unanswered, and what future research will be needed to improve knowledge and advance therapy.

Prostate cancer is the second leading cause of male cancer-related deaths in the United States. Interestingly, prostate cancer preferentially metastasizes to skeletal tissue. Once in the bone microenvironment, advanced prostate cancer becomes highly resistant to therapeutic modalities. Several factors, such as extracellular matrix (ECM) components, have been implicated in the spread and propagation of prostatic carcinoma. In these studies, we have utilized the PC3 cell line, derived from a human bone metastasis, to investigate the influence of the predominant bone ECM protein, type I collagen, on prostate cancer cell proliferation and gene expression. We have also initiated the design and production of ribozymes to specific gene targets that may influence prostate cancer bone metastasis. ^ Our results demonstrate that PC3 cells rapidly adhere and spread on collagen I to a greater degree than on fibronectin (FN) or poly-L-lysine (PLL). Flow cytometry analysis reveals the presence of the α1, α2 and α3 collagen binding integrin subunits. The use of antibody function blocking studies reveals that PC3 cells can utilize α2β 1 and α3β1 integrins to adhere to collagen I. Once plated on collagen I, the cells exhibit increased rates of proliferation compared with cells plated on FN or tissue culture plastic. Additionally, cells plated on collagen I show increased expression of proteins associated with progression through G1 phase of the cell cycle. Inhibitor studies point to a role for phosphatidylinositol 3-kinase (PI3K), MAP kinase (MAPK), and p70 S6 kinase in collagen I-mediated PC3 cell proliferation and cyclin D1 expression. To further characterize the effect of type I collagen on prostate cancer bone metastasis, we utilized a cDNA microarray strategy to monitor type I collagen-mediated changes in gene expression. Results of this analysis revealed a gene expression profile reflecting the increased proliferation occurring on type I collagen. Microarray analysis also revealed differences in the expression of specific gene targets that may impact on prostate cancer metastasis to bone. ^ As a result of our studies on the interaction of prostate cancer cells and the skeletal ECM, we sought to develop novel molecular tools for future gene therapy of functional knockdown experiments. To this end, we developed a series of ribozymes directed against the α2 integrin and at osteopontin, a protein implicated in the metastasis of various cancers, including prostate. These ribozymes should facilitate the future study of the mechanism of prostate cancer cell proliferation, and disease progression occurring at sites of skeletal metastasis where a type I collagen-based environment predominates. ^ Together these studies demonstrate the involvement of bone ECM proteins on prostate cancer cell proliferation and suggest that they may play a significant role on the growth of prostate metastases once in the bone microenvironment. ^

p53 functions as a tumor suppressor through its ability to initiate either growth arrest or apoptosis in cells which have sustained DNA damage. p53 elicits these cellular phenotypes through its biochemical function as a transcriptional activator. By inducing the expression of a battery of target genes, p53 is able to prevent the propagation of cells with damaged DNA. However, the genes transcriptionally induced by p53 which have been identified to date do not fully explain p53 function. p53 has been demonstrated to activate genes involved in cell cycle inhibition, apoptosis and cell proliferation. The reasons for simultaneous activation of p53 targets with disparate, opposing functions are not clear, but may be due to the use of transformed cell lines in previous experiments. In the studies presented in this thesis, the pathway of p53 tumor suppression has been studied in detail in two systems chosen for their relevance to the natural cell environment. One utilizes a normal, unaltered cultured cell system; the other the whole mouse. In order to better understand the role of the known p53 targets in effecting p53 function in normal cells, early rat embryo fibroblasts were irradiated with ultraviolet light to induce DNA damage. It was discovered that p53 protein levels increased in response to irradiation. The known targets of p53, namely, $p21\sp{WAF1/CIP1},\ mdm2,\ cyclin\ G,$ and bax, were shown for the first time to have a differential temporal induction. The growth suppressor $p21\sp{WAF1/CIP1}$ was induced first, followed by cyclin G then mdm2, which is involved in proliferation through its inactivation of p53, and finally, the apoptosis promoter, bax. These findings indicated that p53 activates its target genes in a manner to allow maximum effectiveness of target function. The rat embryo fibroblasts were shown to undergo apoptosis 24 h after irradiation. Additionally, investigation of these cells for cell cycle alterations demonstrated a brief arrest in G1. In the second study, thymocytes from mice with wild type p53 were shown to undergo apoptosis and activate p53 target genes upon ionizing radiation treatment, while thymocytes from mice deficient in p53 could not. The p53 target genes mdm2 and fas were tested in vivo for their ability to mediate p53-regulated apoptosis, and were found dispensible for that cellular function. Therefore, the p53 targets identified to date do not fully explain the ability of p53 to function as a tumor suppressor. Potentially, functional redundancy between the known targets would account for the data seen in these experiments. Additionally, identification of additional target genes should add further understanding of the p53 pathway of tumor suppression. ^

Variations of acoustic properties within the sediment column may significantly affect the propagation of acoustic energy in the upper portion of the oceanic crust. Moreover, the acoustic properties of sediments reflect their mineral compositions, fabrics, and degrees of compaction and cementation. Hence, the physical properties of indurated deep-sea sediments are of considerable geophysical and geological interest. Chalks and limestones are particularly important because substantial accumulations of biogenic carbonates are generally present at the base of the deep-sea sediment column, and high-standing features such as Hess Rise are capped by calcareous deposits. This paper constitutes a preliminary report of the compressional-wave velocities and densities of 31 indurated calcareous sediment samples recovered at DSDP Sites 463 and 465, in the Mid-Pacific Mountains and on Hess Rise, respectively. The sample set includes nine pairs of samples in which velocities were measured parallel and perpendicular to bedding to determine the velocity anisotropy of the sediment. This research is part of an ongoing study of the seismic properties of indurated deep-sea carbonates.

The concentration and isotopic composition of Nd in water and particles collected in the western Mediterranean Sea are studied by two complementary approaches. The first examines local vertical profiles and time series; the second considers the global Nd budget of the whole western Mediterranean Sea. These two approaches are used to quantify the Nd inputs and the dissolved/particulate exchange processes in the water column. Two profiles of Nd in seawater in the Ligurian Sea taken in May and October 1992 show an average epsilon-Nd(0) = -9.6 ± 0.5. Seawater from the Strait of Sicily, representative of the eastern waters flowing into the western basin, is more radiogenic [epsilon-Nd(0) = -7.7 ± 0.6]. Profiles of particulate matter collected in sediment traps in coastal (Gulf of Lions) and offshore (Ligurian Sea) environments are also shown. Particles are enriched in Nd and are more radiogenic near the coast than offshore. Measurements of Nd concentration and epsilon-Nd(0) of external sources to the western Mediterranean Sea compared with the literature data demonstrate that particulate flux of atmospheric Saharan origin are more rich ([Nd] = 38 ± 10 µg/g) and less radiogenic [epsilon-Nd(0) = -13.0 ± 1.0] than riverine particulate discharge ([Nd] = 21.5 ± 4.4 µg/g; epsilon-Nd(0) = -10.1 ± 0.5), allowing to trace Nd particulate inputs in the water column. Nd atmospheric flux appears to be the major source into the whole western basin, although lateral advection of riverine material is the prevailing process in the coastal environment. Offshore, the vertical propagation of an important Saharan dust event has been recorded for two months in sediment traps at 80, 200 and 1000 m. The evolution of the resulting negative epsilon-Nd(0) peak along depth and time shows that the particles reach 200 m on a time scale of one week. For the first time, the Nd budget in the western Mediterranean basin is constrained by both concentrations and isotopic compositions measured in particles and seawater. Surface budget requires a remobilization of 30 ± 20% of particulate Nd input. In deep water, dissolved Nd concentrations are balanced by a scavenging of 10 ± 20% of the sinking particulate flux. On the other hand, the deep isotopic compositions suggest an exchange between 30 ± 20% of the sinking particles and the deep waters. The hypothesis of a non-stationary regime for the surface waters in the Ligurian Sea is also considered.

An analysis of a stretch of coastline shows multiple alterations through environmental climate actions. The narrow, fragile line displays singularities due to three basic causes. The first is the discontinuity in feed or localised loss of solid coastal material. Called massics, their simplest examples are deltas and undersea canyons. The second is due to a brusque change in the alignment of the shoreline’s edge, headlands, groins, harbour and defence works. Given the name of geometric singularities, their simplest uses are artificial beaches in the shelter of a straight groin or spits. The third is due to littoral dynamics, emerged or submerged obstacles which diffract and refract wave action, causing a change in the sea level’s super-elevation in breaker areas. Called dynamics, the simplest examples are salients, tombolos and shells. Discussion of the causes giving rise to variations in the coastline and formation of singularities is the raison d’être of investigation, using actual cases to check the suitability of the classification proposed, the tangential or differential action of waves on the coastal landscape in addition to possible simple, compound and complex shapes detected in nature, both in erosion and deposit processes

The assessment of structural timber was requested in the rehabilitation project of the Naves of the chemical industry "CROS". The criterion was to maintain as much as possible timber of the structure and to make only partial replacements. In order not to damage the existing structure and to assess the quality of the existing timber, a series of non-destructive testing (NDT) in the entire structure were performed: (1) Identification of the species by anatomical techniques, (2) Strength grading by visual method, (3) Estimation of moisture content by the technique of electrical resistance, (4) Acquisition of ultrasonic propagation velocities (5) Resistography and (6) Record of the propagation of electromagnetic waves by means of Ground-penetrating radar. Following, a sample group was chose to carry out destructive testing in the lab and compare the NDT results with those obtained with the standard UNE-EN408 (modules of strength, stiffness and density). In the present work, the results provided by each of the NDT techniques are detailed and above all, what is more important, the validity of these after they have been contrasted with the destructive standard tests.
