The environmental impact of agro-chemicals for fish production was extensively reviewed. The positive contribution of agro- chemicals and the devastating effect on aquaculture was x-rayed to alert users to this obvious environmental problem. Lime and fertilizers are commonly used in fish farming to increase pH of pond soil and water and to increase alkalinity and hardness, reduce humic acid content and to initiate primary and secondary productivity. Devastating effect of lime on environment is likely to be minimal. In the case of fertilizers, over utilization of this agro-chemical could impair water quality as phytoplankton bloom become excessive which consequently raises BOD. The use of Therapeutants in aquaculture was discovered to be more popular in Europe and North America than in the tropics (Africa). Commonly used therapeutants include antibiotics and antimicrobials. For fish pathology chemicals like formalin, potassium permanganate, Dipterex and malachite green are widely in use. Effluent from farms where these chemicals are commonly in use can distort the aquatic ecosystem. The changes in water quality, aquatic community structure and productivity caused by intensive aquaculture are typical of the impacts of pollution from a wide variety of sources like sewage, agricultural run-off and effluent discharges from industry

This study focuses on mechanism of ceramic coating on Al-Si alloys with bulk primary Si using plasma electrolytic oxidation (PEO) technology. Al-Si alloys with 27-32% Si in weight were used as substrates. The morphologies, composition and microstructure of PEO coatings were investigated by scanning electron microscopy (SEM) with energy dispersive X-ray system (EDX). Results showed that the PEO process had four different stages. The effect of bulk Si is greatly on the morphology and composition of coatings at first three stages. Anodic oxide films formed on Al and Si phases, respectively. When the voltage exceeded 40 V, glow appeared and concentrated on the localized zone of interface of Al and Si phase. Al-Si-O compounds formed and covered on the dendrite Si phase surface, and the coating on bulk Si, which was silicon oxide, was rougher than that on other phase. If the treatment time was long enough, the coatings with uniform surface morphologies and elements distribution will be obtained but the microstructure of inner layer is looser due to the bulk Si.

The environmental conditions of Kainji Lake from 1971/72 to date appear to have stabilized to a large extent, judging from the similarity of physico-chemical parameters investigated in this study over the period. Solar radiation (as reflected in variation in temperature) and pH have remained largely constant over the years, while conductivity (index of nutrient enrichment), though significantly higher in 1995/96, could be described as sporadic and needs further monitoring to ascertain its trend in the lake. While water transparency and dissolved oxygen were higher in 1971/72 compared to the other years, these increases cannot be said to be overwhelming. The lower transparency in 1995/96 was due to the exceptional flood of that year and may have also accounted for the poorer dissolved oxygen concentration compared to the other years due to its impact on photosynthesis. There is no evidence from this study to indicate that primary productivity has increased over the years. Consequently, the observed increase in fish yield by the KLFPP from CAS, which is corroborated by estimates from the MEI, cannot be supported on the basis of improved photosynthetic production. The phenomenal high levels of conductivity recorded during certain periods in 1995 (600 mu mhos cm super(-1)) are hitherto unknown in the lake and may indicate a trend towards nutrient enrichment. However, it is premature at this stage to conclude on its long-term impact on primary production and consequently, on fish yield. Secondly, the notion of overfishing in the 80s (Ita, 1993), may need to be further examined as low or dwindling catches could be due to a number of factors among which are the level of fishing effort, the type and efficiency of gears and the intensity of sampling. It would appear that with the intervention of KLFPP, the better management of the lake's fisheries would increase the current level of catch. It also needs to be examined how much of the clupeid fisheries, which is now known to account for a substantial proportion of the total fish yield in Kainji Lake, was included in the sampling of the 80s. There is a growing amount of experimental evidence that suggests people often deviate from the predictions of game theory. Some scholars attempt to explain the observations by introducing errors into behavioral models. However, most of these modifications are situation dependent and do not generalize. A new theory, called the rational novice model, is introduced as an attempt to provide a general theory that takes account of erroneous behavior. The rational novice model is based on two central principals. The first is that people systematically make inaccurate guesses when they are evaluating their options in a game-like situation. The second is that people treat their decisions similar to a portfolio problem. As a result, non optimal actions in a game theoretic sense may be included in the rational novice strategy profile with positive weights.

The rational novice model can be divided into two parts: the behavioral model and the equilibrium concept. In a theoretical chapter, the mathematics of the behavioral model and the equilibrium concept are introduced. The existence of the equilibrium is established. In addition, the Nash equilibrium is shown to be a special case of the rational novice equilibrium. In another chapter, the rational novice model is applied to a voluntary contribution game. Numerical methods were used to obtain the solution. The model is estimated with data obtained from the Palfrey and Prisbrey experimental study of the voluntary contribution game. It is found that the rational novice model explains the data better than the Nash model. Although a formal statistical test was not used, pseudo R^2 analysis indicates that the rational novice model is better than a Probit model similar to the one used in the Palfrey and Prisbrey study.

The rational novice model is also applied to a first price sealed bid auction. Again, computing techniques were used to obtain a numerical solution. The data obtained from the Chen and Plott study were used to estimate the model. The rational novice model outperforms the CRRAM, the primary Nash model studied in the Chen and Plott study. However, the rational novice model is not the best amongst all models. A sophisticated rule-of-thumb, called the SOPAM, offers the best explanation of the data.

The study was carried to examine the contribution of women to fisheries development in the Kainji basin. The sample was drawn in some of the major fishing villages in the sub strata of the basin. A total of 120 respondents were used. The findings revealed that women were involved in fisheries activities most especially in the processing and marketing aspects. Many of the respondents happened to be wives of the fishermen. Few bought fishing equipment with a special arrangement for the supply of fish while some buy from fishermen. This revealed the willingness of the women to be empowered in the economic activities in the area to meet some basic household. Major constrains to the women in this area were the scarcity of fish, no support from the government or private agencies. Recommendations were made on how to enhance women participation in fisheries development

The model dependence inherent in hadronic calculations is one of the dominant sources of uncertainty in the theoretical prediction of the anomalous magnetic moment of the muon. In this thesis, we focus on the charged pion contribution and turn a critical eye on the models employed in the few previous calculations of $a_\mu^{\pi^+\pi^-}$. Chiral perturbation theory provides a check on these models at low energies, and we therefore calculate the charged pion contribution to light-by-light (LBL) scattering to $\mathcal{O}(p^6)$. We show that the dominant corrections to the leading order (LO) result come from two low energy constants which show up in the form factors for the $\gamma\pi\pi$ and $\gamma\gamma\pi\pi$ vertices. Comparison with the existing models reveal a potentially significant omission - none include the pion polarizability corrections associated with the $\gamma\gamma\pi\pi$ vertex. We next consider alternative models where the pion polarizability is produced through exchange of the $a_1$ axial vector meson. These have poor UV behavior, however, making them unsuited for the $a_\mu^{\pi^+\pi^-}$ calculation. We turn to a simpler form factor modeling approach, generating two distinct models which reproduce the pion polarizability corrections at low energies, have the correct QCD scaling at high energies, and generate finite contributions to $a_\mu^{\pi^+\pi^-}$. With these two models, we calculate the charged pion contribution to the anomalous magnetic moment of the muon, finding values larger than those previously reported: $a_\mu^\mathrm{I} = -1.779(4)\times10^{-10}\,,\,a_\mu^\mathrm{II} = -4.892(3)\times10^{-10}$.

The contributions of hematological factors to the distribution and estimations of Eustrongylides africanus larvae densities in Clarias gariepinus and C. anguillaris of Bida floodplain of Nigeria were documented for the first time. The hematological factors making the most important contributions to the distributions of E. africanus larvae infections in clarias species are mean corpuscular haemoglobin concentration (MCHC), mean corpuscular haemoglobin (MCH), mean corpuscular volume (MCV) and neutrophils count, in descending order of magnitude; having the manifestations for the months of January, March, September, and December of the year being closely related. Five haematological factors (neutrophils, lymphocytes and eosinophils counts; MCH and MCV) having positive or negative correlation coefficient (r) between 0.50 and 0.85 contributed to the estimated of E.africanus larvae densities in the wild population of Clarias species

This thesis presents the development of chip-based technology for informative in vitro cancer diagnostics. In the first part of this thesis, I will present my contribution in the development of a technology called “Nucleic Acid Cell Sorting (NACS)”, based on microarrays composed of nucleic acid encoded peptide major histocompatibility complexes (p/MHC), and the experimental and theoretical methods to detect and analyze secreted proteins from single or few cells.

Secondly, a novel portable platform for imaging of cellular metabolism with radio probes is presented. A microfluidic chip, so called “Radiopharmaceutical Imaging Chip” (RIMChip), combined with a beta-particle imaging camera, is developed to visualize the uptake of radio probes in a small number of cells. Due to its sophisticated design, RIMChip allows robust and user-friendly execution of sensitive and quantitative radio assays. The performance of this platform is validated with adherent and suspension cancer cell lines. This platform is then applied to study the metabolic response of cancer cells under the treatment of drugs. Both cases of mouse lymphoma and human glioblastoma cell lines, the metabolic responses to the drug exposures are observed within a short time (~ 1 hour), and are correlated with the arrest of cell-cycle, or with changes in receptor tyrosine kinase signaling.

The last parts of this thesis present summaries of ongoing projects: development of a new agent as an in vivo imaging probe for c-MET, and quantitative monitoring of glycolytic metabolism of primary glioblastoma cells. To develop a new agent for c-MET imaging, the one-bead-one-compound combinatorial library method is used, coupled with iterative screening. The performance of the agent is quantitatively validated with cell-based fluorescent assays. In the case of monitoring the metabolism of primary glioblastoma cell, by RIMChip, cells were sorting according to their expression levels of oncoprotein, or were treated with different kinds of drugs to study the metabolic heterogeneity of cancer cells or metabolic response of glioblastoma cells to drug treatments, respectively.

Interleukin 2 (IL2) is the primary growth hormone used by mature T cells and this lymphokine plays an important role in the magnification of cell-mediated immune responses. Under normal circumstances its expression is limited to antigen-activated type 1 helper T cells (TH1) and the ability to transcribe this gene is often regarded as evidence for commitment to this developmental lineage. There is, however, abundant evidence than many non-TH1 T cells, under appropriate conditions, possess the ability to express this gene. Of paramount interest in the study of T-cell development is the mechanisms by which differentiating thymocytes are endowed with particular combinations of cell surface proteins and response repertoires. For example, why do most helper T cells express the CD4 differentiation antigen?

As a first step in understanding these developmental processes the gene encoding IL2 was isolated from a mouse genomic library by probing with a conspecific IL2 cDNA. The sequence of the 5' flanking region from + 1 to -2800 was determined and compared to the previously reported human sequence. Extensive identity exists between +1 and -580 (86%) and sites previously shown to be crucial for the proper expression of the human gene are well conserved in both sequence location in the mouse counterpart.

Transient expression assays were used to evaluate the contribution of various genomic sequences to high-level gene expression mediated by a cloned IL2 promoter fragment. Differing lengths of 5' flanking DNA, all terminating in the 5' untranslated region, were linked to a reporter gene, bacterial chloramphenicol acetyltransferase (CAT) and enzyme activity was measured after introduction into IL2-producing cell lines. No CAT was ever detected without stimulation of the recipient cells. A cloned promoter fragment containing only 321 bp of upstream DNA was expressed well in both Jurkat and EL4.El cells. Addition of intragenic or downstream DNA to these 5' IL2-CAT constructs showed that no obvious regulatory regions resided there. However, increasing the extent of 5' DNA from -321 to -2800 revealed several positive and negative regulatory elements. One negative region that was well characterized resided between -750 and -1000 and consisted almost exclusively of alternating purine and pyrimidines. There is no sequence resembling this in the human gene now, but there is evidence that there may have once been.

No region, when deleted, could relax either the stringent induction-dependence on cell-type specificity displayed by this promoter. Reagents that modulated endogenous IL2 expression, such as cAMP, cyclosporin A, and IL1, affected expression of the 5' IL2-CAT constructs also. For a given reagent, expression from all expressible constructs was suppressed or enhanced to the same extent. This suggests that these modulators affect IL2 expression through perturbation of a central inductive signal rather than by summation of the effects of discrete, independently regulated, negative and positive transcription factors.

Aegagropila sauteri is a peculiar green algae, the branched thalli of which, according to the amount of growth, forms velvety spheres of a diameter of 3-4, sometimes to 5-6 cm. and bigger. Investigators attribute it to a special genus of green algae. The authors examine Aegagropila sauteri in Lake Markakol (Kazakhstan).

The temporoammonic (TA) pathway is the direct, monosynaptic projection from layer III of entorhinal cortex to the distal dendritic region of area CA1 of the hippo­ campus. Although this pathway has been implicated in various functions, such as memory encoding and retrieval, spatial navigation, generation of oscillatory activity, and control of hippocampal excitability, the details of its physiology are not well understood. In this thesis, I examine the contribution of the TA pathway to hippocampal processing. I find that, as has been previously reported, the TA pathway includes both excitatory, glutamatergic components and inhibitory, GABAergic components. Several new discoveries are reported in this thesis. I show that the TA pathway is subject to forms of short-term activity-dependent regulation, including paired-pulse and frequency­ dependent plasticity, similar to other hippocampal pathways such as the Schaffer collateral (SC) input from CA3 to CA1. The TA pathway provides a strongly excitatory input to stratum radiatum giant cells of CA1. The excitatory component of the TA pathway undergoes a long-lasting decrease in synaptic strength following low-frequency stimulation in a manner partially dependent on the activation of NMDA receptors. High­ frequency activation of the TA pathway recruits a feedforward inhibition that can prevent CA1 pyramidal cells from spiking in response to SC input; this spike-blocking effect shows that the TA pathway can act to regulate information flow through the hippocampal trisynaptic pathway.

Glandular cells, other than the mucous cells, have been described in the skin of various groups of fish (Teleosts, Ganoids, Selachii) and they have been called 'albuminose' by various authors. The authors propose to study the albuminose cells in the skin of Torpedo ocellata Raf. from a histochemical point of view. The albuminose cells have a complex morphological structure and a correspondingly complicated histochemical make-up. One must treat them as an example of cell with secretions of a particular type, which must and will be better incorporated when more is known of characteristics existent in other species.
