We studied the eutrophication history of a tropical shallow reservoir in the So Paulo metropolitan region, southeast Brazil. We analyzed grain size, geochemistry, diatom assemblages, and land-use records in a sediment core from the reservoir to infer its trophic state history during the last similar to 110 years (1894-2005). Eighty diatom species were observed in the core and shifts in the relative abundances of planktonic and benthic taxa indicate major limnological changes associated with complex interactions between hydrologic factors and eutrophication. Discostella stelligera was associated with deforestation and water physical changes whereas Aulacoseira granulata, a species abundant throughout the core, was mostly associated with high flux conditions and erosion events, regardless of trophic state. Eutrophication was triggered by construction of the city zoo (1958) and installation of the So Paulo State Department of Agriculture (1975) within the Gar double dagger as watershed, and increasing loads of untreated sewage from these institutions. The data suggest that deterioration in water quality began after similar to 1975 and markedly accelerated after similar to 1990. The reservoir has been hypereutrophic since 1999. Steady increases in geochemical proxies for trophic state, along with a decrease in C/N ratios, indicated higher nutrient concentrations and the prevalence of autochthonous production towards the core top. Appearance of Achnanthidium catenatum similar to 1993 highlighted the onset of a marked eutrophication phase. The subsequent dominance of Planothidium rostratum and Cyclotella meneghiniana suggested a sharp shift to a hypereutrophic state since 1999. Land-use history proved valuable for validating the chronology and interpreting anthropogenic impacts. Multi-proxy analysis of the sediment record provided an effective tool for tracking ecological shifts in the reservoir ecosystem. This study provides the first reconstruction of lake eutrophication history in Brazil and highlights the importance of hydrological/physical changes as drivers of diatom assemblage shifts in reservoirs, which may confound trophic state inferences based on shifts in the planktonic/benthic diatom ratio.

Recent reports have shown an increase in potentially harmful phytoplankton in Santos bay (Southeastern Brazilian Coast), located in a highly urbanised estuarine complex. Prediction of blooms is, thus, essential but the phytoplankton community structure in very dynamic regions is difficult to determine. In the present work, we discriminate bloom forming microphytoplankton dominance and their relationship to physical and meteorological variables to look for patterns observed in different tides and seasons. Comparing 8 distinct situations, we found five scenarios of dominance that could be related to winds, tides and rainfall: i) Surfers, diatoms occurring during high surf zone energies; ii) Sinkers, represented by larger celled diatoms during spring tide, after periods of high precipitation rates; iii) Opportunistic mixers, composed of chain forming diatoms with small or elongate cells occurring during neap tides; iv) Local mixers, microplanktonic diatoms and dinoflagellates which occurred throughout the 298 sampling stations; and v) Mixotrophic dinoflagellates, after intense estuarine discharges. Results suggest alterations in the temporal patterns for some bloom-forming species, while others appeared in abundances above safe limits for public health. This approach can also illustrate possible impacts of changes in freshwater discharge in highly urbanised estuaries.

Estuaries are extremely dynamic environments that are vulnerable to anthropogenic alterations. Thus, monitoring phytoplankton abundances and composition is an essential tool for the prediction of eutrophication and its effects on coastal ecosystems. Phytoplankton biomass, as chlorophyll-a, in the São Vicente estuary (Brazil) varies in response to tidal cycles and seasonal rainfall. Objectives. To present two datasets designed to assess the relationship between chlorophyll-a and changes in water turbidity driven by tide and rain. Methods. Weekly observations were made in the shallow embayment (February to September 2008; site 1) and observations recorded on alternate days (summer 2010, site 2). Results. At site 1, turbidity differed between high and low tides, but on most days was over 3000 RU, maintaining moderate chlorophyll-a levels (4 mg.m-3) and only two blooms developed during low turbidity. Site 2 mean turbidity was 1500 RU, nutrient level was higher during neap tides and phytoplankton blooms were mainly observed at the end of neap tides at 15-day intervals, dominated by chain-forming diatoms and occasionally flagellates and pennate diatoms. Conclusions. Taxonomic composition of the blooms was different and their frequency altered by events characterized by intense freshwater discharges from the Henry Borden Hydroelectric Dam (> 9*106.m³), inhibiting phytoplankton accumulation during neap tide periods.

A low content of organic matter, which is largely refractory in nature, is characteristic of most sediments, meaning that aquatic deposit-feeders live on a very poor food source. The food is derived mainly from sedimenting phytodetritus, and in temperate waters like the Baltic Sea, from seasonal phytoplankton blooms. Deposit-feeders are either bulk-feeders, or selective feeders, which preferentially ingest the more organic-rich particles in the sediment, including phytodetritus, microbes and meiofauna. The soft-bottom benthos of the Baltic Sea has low species biodiversity and is dominated by a few macrobenthic species, among which the most numerous are the two deposit-feeding amphipods Monoporeia affinis and Pontoporeia femorata, and the bivalve Macoma balthica. This thesis is based on laboratory experiments on the feeding of these three species, and on the priapulid Halicryptus spinulosus. Feeding by benthic animals is often difficult to observe, but can be effectively studied by the use of tracers. Here we used the radioactive isotope 14C to label food items and to trace the organic matter uptake in the animals, while the stable isotopes 13C and 15N were used to follow feeding on aged organic matter in the sediment. The abundance of M. balthica and the amphipods tends to be negatively correlated, i.e., fewer bivalves are found at sites with dense populations of amphipods, with the known explanation that newly settled M. balthica spat are killed by the amphipods. Whether the postlarvae are just accidentally killed, or also ingested after being killed was tested by labelling the postlarvae with 14C and Rhodamine B. Both tracer techniques gave similar evidence for predation on and ingestion of postlarval bivalves. We calculated that this predation was likely to supply less than one percent of the daily carbon requirement for M. affinis, but might nevertheless be an important factor limiting recruitment of M. balthica. The two amphipods M. affinis and P. femorata are partly vertically segregated in the sediment, but whether they also feed at different depths was unknown. By adding fresh 14C-labelled algae either on the sediment surface or mixed into the sediment, we were able to distinguish surface from subsurface feeding. We found M. affinis and P. femorata to be surface and subsurface deposit-feeders, respectively. Whether the amphipods also feed on old organic matter, was studied by adding fresh 14C-labelled algae on the sediment surface, and using aged, one-year-old 13C- and 15N-labelled sediment as deep sediment. Ingestion of old organic matter, traced by the stable isotopes, differed between the two species, with a higher uptake for P. femorata, suggesting that P. femorata utilises the older, deeper-buried organic matter to a greater extent. Feeding studies with juveniles of both M. affinis and P. femorata had not been done previously. In an experiment with the same procedure and treatments as for the adults, juveniles of both amphipod species were found to have similar feeding strategies. They fed on both fresh and old sediment, with no partitioning of food resources, making them likely to be competitors for the same food resource. Oxygen deficiency has become more wide-spread in the Baltic Sea proper in the last half-century, and upwards of 70 000km2 are now devoid of macrofauna, even though part of that area does not have oxygen concentrations low enough to directly kill the macrofauna. We made week-long experiments on the rate of feeding on 14C-labelled diatoms spread on the sediment surface in different oxygen concentrations for both the amphipod species, M. balthica and H. spinulosus. The amphipods were the most sensitive to oxygen deficiency and showed reduced feeding and lower survival at low oxygen concentrations. M. balthica showed reduced feeding at the lowest oxygen concentration, but no mortality increase. The survival of H. spinulosus was unaffected, but it did not feed, showing that it is not a surface deposit-feeder. We conclude that low oxygen concentrations that are not directly lethal, but reduce food intake, may lead to starvation and death in the longer term.

Sinking particles through the pelagic ocean have been traditionally considered the most important vehicle by which the biological pump sequesters carbon in the ocean interior. Nevertheless, regional scale variability in particle flux is a major outstanding issue in oceanography. 5 Here, we have studied the regional and temporal variability of total particulate organic matter fluxes, as well as chloropigment and total hydrolyzed amino acid (THAA) compositions and fluxes in the Canary Current region, between 20–30 N, during two contrasting periods: August 2006, characterized by warm and stratified waters, but also intense winds which enhanced eddy development south of the Canary Islands, 10 and February 2007, characterized by colder waters, less stratification and higher productivity. We found that the eddy-field generated south of the Canary Islands enhanced by >2 times particulate organic carbon (POC) export with respect to stations (FF; farfield) outside the eddy-field influence. We also observed flux increases of one order of magnitude in chloropigment and 70% in THAA in the eddy-field relative to FF stations. 15 Principal Components Analysis (PCA) was performed to assess changes in particulate organic matter composition between stations. At eddy-field stations, higher chlorophyll enrichment reflected “fresher” material, while at FF stations a higher proportion of pheophytin indicated greater degradation due to microbes and microzooplankton. PCA also suggests that phytoplankton community structure, particularly the dominance of 20 diatoms versus carbonate-rich plankton, is the major factor influencing the POC export within the eddy field. In February, POC export fluxes were the highest ever reported for this area, reaching values of 15 mmolCm−2 d−1 at 200m depth. Compositional changes in pigments and THAA indicate that the source of sinking particles varies zonally and meridionally and suggest that sinking particles were more degraded at 25 near-coastal stations relative to open ocean stations.

[EN] Diatom cell quantity and their biochemical composition vary among species and are greatly affected by harvest stage or culture conditions. Biometric parameters, growth, attachment capacity and variations in biochemical composition of four species of benthic diatoms (Amphora sp., Navicula incerta, Nitzschia sp. and Proschkinia sp.) were studied. For biochemical analysis the diatoms were harvested at different stages, in log and stationary phase of growth. The culture conditions were identical for all the experiments, benthic diatoms were cultured during 7 days in F/2 medium at 28.5 ± 1.4 ºC, at different original inoculating densities (50000, 100000, and 250000 cell mL-1), under continuous light of 5403 ± 649 Lux provided by cool white fluorescent lighting. The cultures were neither aerated nor agitated. These results show that the specific density of 10000 cell mL-1 was the best for weekly production: Proschkinia sp. reached the highest cell density of 5.81 x 106 cells mL-1 and Amphora sp. had the highest cell attachment capacity with 12000 cell mm-2, in stationary phase of growth. Protein and lipid content were higher in log phase than in stationary phase for the four diatoms. Amphora sp. in log phase of growth had the highest lipid content of 9.74% dry weight (DW). Polyunsaturated fatty acid (PUFA) content ranged from 23.25% to 38.62% of the total fatty acids (TFA), and the four diatoms tested were richer in n-3 PUFA than in n-6 PUFA. All the diatoms had significant quantities of 20:5n-3 (EPA) ranging between 12.69% and 17.68% of TFA. Benthic diatoms play an important and critical role in abalone culture as they are the principal food source of abalone post-larvae. Therefore, it is necessary to improve diatom quantity and quality to optimize post-larval nutrition and the consistency of production, resulting in an increase in growth and survival of abalones.

[EN] Size-abundance spectra (SAS) were analyzed in different phytoplankton assemblages of the Alborán Sea collected in areas with contrasting hydrological features (upwelling areas and anticyclonic gyre). Abundance of micro-plankton cells decreased following the hydrological gradient from the most productive stations towards offshore stations. This size-fraction of phytoplankton was dominated by diatoms. Pico-plankton followed an opposite gradient. Concordantly, the slope of the SAS trended to decrease from coastal areas (upwelling) towards the anticyclonic gyre. However, phytoplankton assemblages featuring similar SAS values presented a quite different taxonomical composition of their diatom communities. According to the previous available information about the taxonomical composition of the phytoplankton communities in the Alborán Sea, these differences in diatom composition are indicative of differences in productivity of the phytoplankton. Consequently, the utility of phytoplankton SAS as an indicator of changes in the phytoplankton communities of the Alborán Sea is discussed.

[EN] Fe(II) oxidation kinetics were studied in seawater and in seawater enriched with exudates excreted by Phaeodactylum tricornutum as an organic ligand model. The exudates produced after 2, 4, and 8 days of culture at 6.21 .. 107, 2.29 .. 108, and 4.98 .. 108 cell L?1 were selected. The effects of pH (7.2?8.2), temperature (5?35 ºC), and salinity (10?36.72) on the Fe(II) oxidation rate were studied. All the data were compared with the results for seawater without exudates (control). The Fe(II) rate constant decreased as a function of culture time and cell concentration in the culture at different pH, temperature, and salinity. All the experimental data obtained in this study were fitted to a polynomial function in order to quantify the fractional contribution of the organic exudates from the diatoms to the Fe(II) oxidation rate in natural seawater. Experimental results showed that the organic exudates excreted by P. tricornutum affect Fe(II) oxidation, increasing the lifetime of Fe(II) in seawater. A kinetic model approach was carried out to account for the speciation of each Fe(II) type together with its contribution to the overall rate.

Zusammenfassung:In Chlorophyll(Chl) a/c-haltigen Algen leisten Xanthophylle einen wesentlichen Beitrag zur Lichtsammlung. Daneben finden sich weitere Xanthophylle, die an einem Schutzmechanismus bei überoptimalem Lichtangebot beteiligt sind, dem sog. Xanthophyllzyklus. Ein Teil der Chl a/c-haltigen Algen besitzt den auch bei Höheren Pflanzen anzutreffenden Violaxanthin/Antheraxanthin/Zeaxanthin-(Vx/Ax/Zx-)Zyklus. In anderen Gruppen wie den Dinophyta, Haptophyta und den Kieselalgen (Bacillariophyceae) ist statt dessen der Diadinoxanthin/Diatoxanthin-(Ddx/Dtx-)Zyklus zu finden. Die vorliegende Arbeit zeigt, daß schwachlichtadaptierte Turbidostatkulturen der Kieselalge Phaeodactylum tricornutum unter mehrstündiger Starklichtinkubation neben den Pigmenten des Ddx/Dtx-Zyklus auch die des Vx/Ax/Zx-Zyklus akkumulieren. Außerdem läßt sich ein dritter Xanthophyllzyklus zwischen beta-Cryptoxanthin (Cx) und beta-Cryptoxanthin-Epoxid (CxE) nachweisen, doch liegen diese beiden Pigmente nur in sehr geringen Konzentrationen vor. Für die Starklichtakkumulation von Zx ist eine hohe Deepoxidase-Aktivität und die de-novo-Synthese von Carotinoiden erforderlich. Aus Zx wird im anschließenden Schwachlicht über die Intermediate Vx und Ddx das Lichtsammelxanthophyll Fucoxanthin (Fx) synthetisiert. Dies bestätigt auch ein Vergleich der Kinetiken der einzelnen Umwandlungsschritte mit den anhand eines Modells der Xanthophyllbiosynthesewege ermittelten theoretischen Ratenkonstanten. Dieser Vergleich legt jedoch nahe, daß bei der Vx-Synthese aus beta-Carotin CxE anstelle von Zx involviert sein könnte. Eine Untersuchung weiterer Chl a/c-haltiger Algen mit Ddx/Dt-Zyklus ergab, daß sie unter Starklicht ebenfalls den Vx/Ax/Zx-Zyklus akkumulieren. Weiterhin sind, mit Einschränkungen bei den Dinophyten und Xanthophyceen, alle untersuchten Algen in der Lage, die unter Starklicht akkumulierten Xanthophyllzykluspigmente im nachfolgenden Schwachlicht zur Synthese des jeweiligen Lichtsammelxanthophylls zu nutzen. Unter energetischen Gesichtspunkten stellt dieses Pigment-Recycling insbesondere für die Fx-haltigen Algen einen Vorteil dar, da ihre Lichtsammelkomplexe im Vergleich zu denen der Höheren Pflanzen etwa die doppelte Anzahl an Xanthophyllen binden.

Since the industrial revolution, the ocean has absorbed around one third of the anthropogenic CO2, which induced a profound alteration of the carbonate system commonly known as ocean acidification. Since the preindustrial times, the average ocean surface water pH has fallen by 0.1 units, from approximately 8.2 to 8.1 and a further decrease of 0.4 pH units is expected for the end of the century. Despite their microscopic size, marine diatoms are bio-geo-chemically a very important group, responsible for the export of massive amount of carbon to deep waters and sediments. The knowledge of the potential effects of ocean acidification on the phytoplankton growth and on biological pump is still at its infancy. This study wants to investigate the effect of ocean acidification on the growth of the diatom Skeletonema marinoi and on its aggregation, using a mechanistic approach. The experiment consisted of two treatments (Present and Future) representing different pCO2 conditions and two sequential experimental phases. During the cell growth phase a culture of S. marinoi was inoculated into transparent bags and the effect of ocean acidification was studied on various growth parameters, including DOC and TEP production. The aggregation phase consisted in the incubation of the cultures into rolling tanks where the sinking of particles through the water column was simulated and aggregation promoted. Since few studies investigated the effect of pH on the growth of S. marinoi and none used pH ranges that are compatible with the OA scenarios, there were no baselines. I have shown here, that OA does not affect the cell growth of S. marinoi, suggesting that the physiology of this species is robust in respect to the changes in the carbonate chemistry expected for the end of the century. Furthermore, according to my results, OA does not affect the aggregation of S. marinoi in a consistent manner, suggesting that this process has a high natural variability but is not influenced by OA in a predictable way. The effect of OA was tested over a variety of factors including the number of aggregates produced, their size and sinking velocity, the algal, bacterial and TEP content. Many of these variables showed significant treatment effects but none of these were consistent between the two experiments.

Die für Metazoen einzigartige Fähigkeit, hochdifferenzierte Silikatstrukturen herzustellen und als Gerüstsubstanz zu verwenden, steht bei den Porifera in einem scheinbaren Gegensatz zu der niedrigen Konzentration an Silizium in dem die Schwämme umgebenden Medium. In der zweiten bedeutenden silikatpolymerisierenden Species, den einzelligen Kieselalgen (Diatomeen), konnte bereits ein Silikattransporter identifiziert werden, dessen Sequenzdaten jedoch aufgrund der phylogenetisch geringen Verwandtschaft der Demospongien mit den Diatomeen keine Verwendung finden konnte Im Zuge der Suche nach einem Silikat-Transportsystem im Schwamm Suberites domuncula wurde ein potentielles Kandidatengen mittels molekularbiologischer Techniken aus einer cDNA Bank des Instituts isoliert, vervollständigt und analysiert. Es zeigte sich, dass dieser Transporter durch seine Sequenzdaten der Familie der Bikarbonattransporter angehörte, und somit membranständig war. Seine Transportfunktion zeigte sich mittels spezifischer Inhibitoren hemmbar. Damit der Schwamm in der Lage ist, eine regulierbare und schnelle Anreicherung von Silikat durchführen zu können, lag eine Annahme einer Induzierbarkeit der Transportergene durch das Substrat Silikat nahe. Mittels Northern-Blot Analyse konnte in einem Primmorphensystem des Schwammes eine Hochregulation der Transkription der Transportergene festgestellt werden. Die Lokalisation der Exprimierung des Transporters innerhalb des Schwammgewebes konnte mittels In situ Hybridisierung untersucht werden und zeigte eine direkte Nähe zu den Polysilikatstrukturen des Schwammes. Um Hinweise auf eine Bifunktionalität des Transporters aufgrund der Ähnlichkeit von Carbonat und Silikat zu erhärten, wurden fluoreszenzmikroskopische Studien an isolierten Zellkulturen des Schwammes durchgeführt. Es kam zu einer intensive Reaktion der Zellen auf Silikat als Substrat. Dieser Effekt konnte nicht nur durch einen spezifischen Transportinhibitor (DIDS) gehemmt werden, sondern zeigt auch eine deutliche Temperaturabhängigkeit. Um den potentiellen Silikattransporter in Zusammenhang mit dem Gesamtmechanismus der Silikatnadelherstellung in Schwämmen zu bringen, wurden zusätzliche elektronenmikroskopische Studien angestellt. Hier konnte zunächst gezeigt werden, wie sich das die Polykondensation auslösende und dirigierende Proteinfilament des Schwammes bei der Nadelbildung entwickelt. Mittels einer darauf folgenden Immunogold-Markierung des Hauptaxialfilamentproteins des Schwammes in elektronenmikroskopischen Gewebepräparaten, konnte dessen Vorkommen nicht nur im Zentrum der Silikatnadel, sondern auch in den die Nadel umgebenden Strukturen nachgewiesen werden

The most ocean - atmosphere exchanges take place in polar environments due to the low temperatures which favor the absorption processes of atmospheric gases, in particular CO2. For this reason, the alterations of biogeochemical cycles in these areas can have a strong impact on the global climate. With the aim of contributing to the definition of the mechanisms regulating the biogeochemical fluxes we have analyzed the particles collected in the Ross Sea in different years (ROSSMIZE, BIOSESO 1 and 2, ROAVERRS and ABIOCLEAR projects) in two sites (mooring A and B). So it has been developed a more efficient method to prepare sediment trap samples for the analyses. We have also processed satellite data of sea ice, chlorophyll a and diatoms concentration. At both sites, in each year considered, there was a high seasonal and inter-annual variability of biogeochemical fluxes closely correlated with sea ice cover and primary productivity. The comparison between the samples collected at mooring A and B in 2008 highlighted the main differences between these two sites. Particle fluxes at Mooring A, located in a polynia area, are higher than mooring B ones and they happen about a month before. In the mooring B area it has been possible to correlate the particles fluxes to the ice concentration anomalies and with the atmospheric changes in response to El Niño Southern Oscillations. In 1996 and 1999, years subjected to La Niña, the concentrations of sea ice in this area have been less than in 1998, year subjected to El Niño. Inverse correlation was found for 2005 and 2008. In the mooring A area significant differences in mass and biogenic fluxes during 2005 and 2008 has been recorded.
