We have shown Galanin(GAL) and Neuropeptide Y Y1(NPYY1) interactions at behavioural, cellular and receptor levels through GALR2/NPYY1R heterodimers in the amygdala. The aim of this work was to analyze GAL/NPYY1R interactions in the Dentate Gyrus(DG) of the Hippocampus, using autoradiographic, in situ hybridization and in situ proximity ligation assay(PLA). Rats(n=6) were sacrificed 15 minutes or 5 hours after icv injections of GAL(3nmol) and DG sections were incubated with NPYY1R agonist [I125]-[Leu31,Pro34]PYY(25 pM) or NPYY1R-33PdATP specific probe, for autoradiography and in situ hybridization respectively. Autoradiograms were analyzed using NIH image analysis system and Student’s unpaired t-test was used. For PLA, DG sections were incubated with anti-GALR2 Rabbit(1:100) and anti-NPYY1R Goat(1:200). PLA signals were detected with PLA PLUS or MINUS probes for rabbit or goat/mouse antibodies. PLA signals were visualized by using a confocal microscope Leica TCS-SL confocal microscope(Leica). We observed that GAL significant increased the NPYY1R agonist [I125]-[Leu31,Pro34]PYY binding in the DG by 20% (p<0,05) and the NPYY1R mRNA expression in the granular layer of DG by 31% (p<0,001). Moreover, PLA-positive red clusters were found specifically in the polymorphic layer and subgranular zone of the DG. No PLA clusters were observed neither in the molecular layer of the DG nor in the corpus callosum, an area that seems to lack of GALR2 receptor. These results demonstrate a novel mechanism of interaction between GAL and NPY1R in the DG at receptor level, probably involving the formation of GALR2/NPYY1R heteroreceptor complexes. Ongoing quest for finding treatment against memory loss seen in aging and in many neurological and neurodegenerative diseases, so far has been unsuccessful and memory enhancers are seen as a potential remedy against this brain dysfunction. Recently, we showed that gene corresponding to a protein called regulator of G-protein signaling 14 of 414 amino acids (RGS14414) is a robust memory enhancer (Lopez-Aranda et al. 2009: Science). RGS14414-treatment in area V2 of visual cortex caused memory enhancement to such extent that it converted short-term object recognition memory (ORM) of 45min into long lasting long-term memory that could be traced even after many months. Now, through targeting of multiple receptors and molecules known to be involved in memory processing, we found that GluR2 subunit of AMPA receptor might be key to memory enhancement in RGS-animals. RGS14-animals showed a progressive increase in GluR2 protein expression while processing an object information which reached to highest level after 60min of object exposure, a time period required for conversion of short-term ORM into long-term memory in our laboratory set up. Normal rats could retain an object information in brain for 45min (short-term) and not for 60min. However, RGS-treated rats are able to retain the same information for 24h or longer (long-term). Therefore, highest expression of GluR2 subunit seen at 60min suggests that this protein might be key in memory enhancement and conversion to long-term memory in RGS-animals. In addition, we will also discuss the implication of Hebbian plasticity and interaction of brain circuits in memory enhancement.

Several studies show that morphological changes of microglia over the course of inflammation are tightly coupled to function. However the progressive transformation into activated microglia is poorly characterized. AIMS: This study aimed to establish a spatiotemporal correlation between quantifiable morphological parameters of microglia and the spread of an acute ventricular inflammatory process. METHODS: Inflammation was induced by a single injection of the enzyme neuraminidase within the lateral ventricle of rats. Animals were sacrificed 2, 4 and 12 hours after injection. Coronal slices were immunostained with Iba1 to label microglia and with IL1β to delimit the spread of inflammation. Digital images were obtained by scanning the labelled sections. Single microglia images were randomly selected from periventricular areas of caudate putamen, hippocampus and hypothalamus. FracLac for ImageJ software was used to measure the following morphological parameters: fractal dimension, lacunarity, area, perimeter and density. RESULTS: Significant differences were found in fractal dimension, lacunarity, perimeter and density of microglia cells of neuraminidase injected rats compared to sham animals. However no differences were found in the parameter “area”. In hipoccampus there was a delay in the significant change of the measured parameters. These morphological changes correlated with IL1β-expression in the same areas. CONCLUSIONS: Ventricular inflammation induced by neuraminidase provokes quantifiable morphological changes in microglia restricted to areas labelled with IL1β. Morphological parameters of microglia such as fractal dimension, lacunarity, perimeter and density are sensitive and valuable tools to quantify activation. However, the extensively used parameter “area” did not change upon microglia activation.

Systematic literature review and consumer research in Belgium and in Denmark (Es necesario el chocolate “saludable”?

Haemoglobins constitute a set of proteins with interesting structural and functional properties, especially when the two large animal groups reptiles and fishes are focused on. Here, the crystallization and preliminary X-ray analysis of haemoglobin-II from the South American fish matrinxa (Brycon cephalus) is reported. X-ray diffraction data have been collected to 3.0 Angstrom resolution using synchrotron radiation (LNLS). Crystals were determined to belong to space group P2(1) and preliminary structural analysis revealed the presence of two tetramers in the asymmetric unit. The structure was determined using the standard molecular-replacement technique.

AIMS: Cognitive decline in Alzheimer's disease (AD) patients has been linked to synaptic damage and neuronal loss. Hyperphosphorylation of tau protein destabilizes microtubules leading to the accumulation of autophagy/vesicular material and the generation of dystrophic neurites, thus contributing to axonal/synaptic dysfunction. In this study, we analyzed the effect of a microtubule-stabilizing compound in the progression of the disease in the hippocampus of APP751SL/PS1M146L transgenic model. METHODS: APP/PS1 mice (3 month-old) were treated with a weekly intraperitoneal injection of 2 mg/kg epothilone-D (Epo-D) for 3 months. Vehicle-injected animals were used as controls. Mice were tested on the Morris water maze, Y-maze and object-recognition tasks for memory performance. Abeta, AT8, ubiquitin and synaptic markers levels were analyzed by Western-blots. Hippocampal plaque, synaptic and dystrophic loadings were quantified by image analysis after immunohistochemical stainings. RESULTS: Epo-D treated mice exhibited a significant improvement in the memory tests compared to controls. The rescue of cognitive deficits was associated to a significant reduction in the AD-like hippocampal pathology. Levels of Abeta, APP and ubiquitin were significantly reduced in treated animals. This was paralleled by a decrease in the amyloid burden, and more importantly, in the plaque-associated axonal dystrophy pathology. Finally, synaptic levels were significantly restored in treated animals compared to controls. CONCLUSION: Epo-D treatment promotes synaptic and spatial memory recovery, reduces the accumulation of extracellular Abeta and the associated neuritic pathology in the hippocampus of APP/PS1 model. Therefore, microtubule stabilizing drugs could be considered therapeutical candidates to slow down AD progression. Supported by FIS-PI12/01431 and PI15/00796 (AG),FIS-PI12/01439 and PI15/00957(JV)

Summary: Climate change has a potential to impact rainfall, temperature and air humidity, which have relation to plant evapotranspiration and crop water requirement. The purpose of this research is to assess climate change impacts on irrigation water demand, based on future scenarios derived from the PRECIS (Providing Regional Climates for Impacts Studies), using boundary conditions of the HadCM3 submitted to a dynamic downscaling nested to the Hadley Centre regional circulation model HadRM3P. Monthly time series for average temperature and rainfall were generated for 1961-90 (baseline) and the future (2040). The reference evapotranspiration was estimated using monthly average temperature. Projected climate change impact on irrigation water demand demonstrated to be a result of evapotranspiration and rainfall trend. Impacts were mapped over the target region by using geostatistical methods. An increase of the average crop water needs was estimated to be 18.7% and 22.2% higher for 2040 A2 and B2 scenarios, respectively. Objective ? To analyze the climate change impacts on irrigation water requirements, using downscaling techniques of a climate change model, at the river basin scale. Method: The study area was delimited between 4º39?30? and 5º40?00? South and 37º35?30? and 38º27?00? West. The crop pattern in the target area was characterized, regarding type of irrigated crops, respective areas and cropping schedules, as well as the area and type of irrigation systems adopted. The PRECIS (Providing Regional Climates for Impacts Studies) system (Jones et al., 2004) was used for generating climate predictions for the target area, using the boundary conditions of the Hadley Centre model HadCM3 (Johns et al., 2003). The considered time scale of interest for climate change impacts evaluation was the year of 2040, representing the period of 2025 to 2055. The output data from the climate model was interpolated, considering latitude/longitude, by applying ordinary kriging tools available at a Geographic Information System, in order to produce thematic maps.

Mastoparans are tetradecapeptides found to be the major component of vespid venoms. A mastoparan toxin isolated from the venom of Anterhynchium flavomarginatum micado has been crystallized and X-ray diffraction data collected to 2.7 Angstrom resolution using a synchrotron-radiation source. Crystals were determined to belong to the space group P6(2)22 (P6(4)22). This is the first mastoparan to be crystallized and will provide further insights into the conformational significance of mastoparan toxins with respect to their potency and activity in G-protein regulation.

On categorization and symbolic power in social work: The myth of the resourceful parents to children with neuro-psychiatric diagnoses Social workers often use the term resourceful about a certain group of parents to children with neuro-psychiatric diagnoses, e.g. autism spectrum disorder and ADHD. This paper discusses the consequences of this sort of stereotyping/categorization with particular regard to the collaboration between these parents and the social workers as they meet in the social services system, when the parents apply for help for their children. Drawing upon Pierre Bourdieu, it is suggested that the categorization resourceful is not only overly simplistic and a myth, but is straight out misguiding and complicates the interaction – in fact it represents an act of symbolic power. Based upon a six months long sociological field-work study it is shown, that even though social workers seem to acknowledge the difficult and grueling life-situation of the parents, they are first and foremost perceived as resourceful and knowledgeable but hence also as annoying, insufferable, demanding and basically unjustified, even though they have obvious legal rights. Parents and social workers alike described the collaboration as being conflictual and a struggle and the complex power-relations are discussed in the light of Bourdieu.

This paper describes three metaphors for time drawn from contemporary and historical literature on knowledge organization systems (KOS). It then links these metaphors to the evaluation of knowledge organization by describing the dominant paradigm in KOS evaluation to be judging whether a KOS is correct. We conclude by saying a foundational view of evaluating and theorizing about KOS must account for change and time in order for us to take a long view of improving knowledge organization and our understanding of KOS.
