Eukaryotic translation initiation factor 5A (eIF5A) is a protein that is highly conserved and essential for cell viability. This factor is the only protein known to contain the unique and essential amino acid residue hypusine. This work focused on the structural and functional characterization of Saccharomyces cerevisiae eIF5A. The tertiary structure of yeast eIF5A was modeled based on the structure of its Leishmania mexicana homologue and this model was used to predict the structural localization of new site-directed and randomly generated mutations. Most of the 40 new mutants exhibited phenotypes that resulted from eIF-5A protein-folding defects. Our data provided evidence that the C-terminal alpha-helix present in yeast eIF5A is an essential structural element, whereas the eIF5A N-terminal 10 amino acid extension not present in archaeal eIF5A homologs, is not. Moreover, the mutants containing substitutions at or in the vicinity of the hypusine modification site displayed nonviable or temperature-sensitive phenotypes and were defective in hypusine modification. Interestingly, two of the temperature-sensitive strains produced stable mutant eIF5A proteins - eIF5A(K56A) and eIF5A(Q22H,L93F)- and showed defects in protein synthesis at the restrictive temperature. Our data revealed important structural features of eIF5A that are required for its vital role in cell viability and underscored an essential function of eIF5A in the translation step of gene expression.

We report an experimental and theoretical study of magnetic properties of synthetic eumelanin. The magnetization curves are determined by using both a vibrating sample magnetometer and a superconducting quantum interferometer device in an extended range of magnetic fields ranging from -10 kOe to 10 kOe at different temperatures. We find that the eumelanin magnetization can be qualitatively explained in terms of a simple model of dipolar spheres with an intrinsic magnetic moment. The latter one is experimentally measured by using X-band electron paramagnetic resonance. Our findings indicate that synthetic melanins are superparamagnetic.

Tropicalia concerns the attempt of understanding of Brazil and a national identity characterized by the transience of time and space, rhizomatic action, perpetual laceration of cultural boundaries and at the same time act synchronously to instances such as policy and the social. A word created by artist Hélio Oiticica and registered by him on the National Register of Trademarks and Patents, was later used to a name the eponymous song by Caetano Veloso and also in this fertile cultural impulse at the turn of the sixties to the seventies on last century, who had the intention to make creating new spaces and simultaneously rethink the spatial cuts of Brazilian culture, encouraging them to be much more than a myth of a paradise within a lush new look of a tropical truth.

Este artigo pretende fazer um levantamento bibliográfico sobre a neurose profissional, cujos sintomas são entendidos como a expressão simbólica de um conflito psíquico que se desenvolve a partir de uma situação organizacional ou profissional determinada. O diagnóstico é formado a partir das queixas mencionadas, da história de trabalho e da análise da situação de trabalho atual. Entende-se que condições estressantes de trabalho, associadas ao contexto social de desemprego e competitividade, contribuem para o aparecimento de doenças mentais como a neurose profissional. As formas de tratamento para essa doença, além da psicoterapia individual, envolvem a ação integrada de uma equipe multiprofissional capacitada para lidar com o sofrimento psíquico do trabalhador e com os aspectos sociais e de intervenção nos ambientes de trabalho. Ressalta-se a carência de literatura sobre o tema estudado e a importância de se ampliar a discussão sobre os fatores de risco no trabalho, os determinantes psíquicos para o desenvolvimento da neurose profissional e o papel do psicólogo no contexto de prevenção e promoção da saúde mental.

As más condições laborais e a forma de organização do trabalho dentro do sistema capitalista frequentemente têm se mostrado nocivas para o trabalhador. Além disso, a chamada síndrome da fadiga crônica é um dos males que acomete a classe trabalhadora. Por meio de revisão teórica buscou-se caracterizar a síndrome, suas possíveis causas e consequências e o papel do psicólogo nas organizações. A fadiga pode ser compreendida como um esgotamento físico e mental, culminando em consequências para a saúde do trabalhador. Excesso de trabalho, pressão por produção, turnos alternados, riscos físicos e falta de autonomia são alguns fatores apontados como prováveis desencadeadores da síndrome, e as consequências encontradas vão desde baixo rendimento e maior risco de acidentes de trabalho até dificuldade para relaxar em momentos de lazer. É papel do psicólogo do trabalho e organizacional realizar ações remediadoras e preventivas que impeçam ou dificultem o aparecimento da fadiga no trabalho.

Este estudo teórico resgata a concepção processual e contextual de resiliência, a qual compreende esse fenômeno enquanto relação complexa entre indivíduo e ambiente na produção de fatores de risco e no provimento de fatores protetores da subjetividade do indivíduo, buscando aplicá-la à realidade organizacional, haja vista que o mundo do trabalho contemporâneo demanda uma força de trabalho específica, qual seja, o trabalhador flexível, polivalente, sujeito a mudanças - portanto, um trabalhador resiliente. A partir dessas considerações, realiza-se um processo de reflexão sobre as situações em que a resiliência pode ser promotora de saúde mental, bem como acerca dos contextos que contribuem para o processo de adoecimento do trabalhador, buscando aventar possibilidades de atuação do profissional psicólogo diante de um contexto de trabalho adverso, considerando-se que o principal objetivo dessa atuação profissional é garantir a proteção/promoção da saúde e qualidade de vida no ambiente de trabalho.

Different species of Leishmania can cause a variety of medically important diseases, whose control and treatment are still health problems. Telomere binding proteins (TBPs) have potential as targets for anti-parasitic chemotherapy because of their importance for genome stability and cell viability. Here, we describe LaTBP1 a protein that has a Myb-like DNA-binding domain, a feature shared by most double-stranded telomeric proteins. Binding assays using full-length and truncated LaTBP1 combined with spectroscopy analysis were used to map the boundaries of the Myb-like domain near to the protein only tryptophan residue. The Myb-like domain of LaTBP1 contains a conserved hydrophobic cavity implicated in DNA-binding activity. A hypothetical model helped to visualize that it shares structural homology with domains of other Myb-containing proteins. Competition assays and chromatin immunoprecipitation confirmed the specificity of LaTBP1 for telomeric and GT-rich DNAs, suggesting that LaTBP1 is a new TBP. (C) 2007 Elsevier B.V. Leishmania amazonensis causes a wide spectrum of leishmaniasis. There are no vaccines or adequate treatment for leishmaniasis, therefore there is considerable interest in the identification of new targets for anti-leishmania drugs. The central role of telomere-binding proteins in cell maintenance makes these proteins potential targets for new drugs. In this work, we used a combination of purification chromatographies to screen L. amazonensis proteins for molecules capable of binding double-stranded telomeric DNA. This approach resulted in the purification of a 38 kDa polypeptide that was identified by mass spectrometry as Rbp38, a trypanosomatid protein previously shown to stabilize mitochondrial RNA and to associate with nuclear and kinetoplast DNAs. Western blotting and supershift assays confirmed the identity of the protein as LaRbp38. Competition and chromatin immunoprecipitation assays confirmed that LaRbp38 interacted with kinetoplast and nuclear DNAs in vivo and suggested that LaRbp38 may have dual cellular localization and more than one function. Studies of telomere structure and maintenance in trypanosomatids have provided insights into the evolutionary origin and conservation of some telomeric components shared by trypanosomes and vertebrates. For example, trypanosomatid telomeres are maintained by telomerase and consist of the canonical TTAGGG repeats, which in Trypanosoma brucei can form telomeric loops (t-loops). However, the telomeric chromatin of trypanosomatids is composed of organ ism-specific proteins and other proteins that share little sequence similarity with their vertebrate counterparts. Because telomere maintenance mechanisms are essential for genome stability, we propose that the particular features shown by the trypanosome telomeric chromatin hold the key for the design of antiparasitic drugs.

Replication protein A (RPA) is a highly conserved heterotrimeric single-stranded DNA-binding protein involved in different events of DNA metabolism. In yeast, subunits 1 (RPA-1) and 2 (RPA-2) work also as telomerase recruiters and, in humans, the complex unfolds G-quartet structures formed by the 3' G-rich telomeric strand. In most eukaryotes, RPA-1 and RPA-2 bind DNA using multiple OB fold domains. In trypanosomatids, including Leishmania, RPA-1 has a canonical OB fold and a truncated RFA-1 structural domain. In Leishmania amazonensis, RPA-1 alone can form a complex in vitro with the telomeric G-rich strand. In this work, we show that LaRPA-1 is a nuclear protein that associates in vivo with Leishmania telomeres. We mapped the boundaries of the OB fold DNA-binding domain using deletion mutants. Since Leishmania and other trypanosomatids lack homologues of known telomere end binding proteins, our results raise questions about the function of RPA-1 in parasite telomeres.
