Three-dimensional positioning of the nuclear genome plays an important role in the epigenetic regulation of genes. Although nucleographic domain compartmentalization in the regulation of epigenetic state and gene expression is well established in higher organisms, it remains poorly understood in the pathogenic parasite Plasmodium falciparum. In the present study, we report that two histone tail modifications, H3K9Ac and H3K14Ac, are differentially distributed in the parasite nucleus. We find colocalization of active gene promoters such as Tu1 (tubulin-1 expressed in the asexual stages) with H3K9Ac marks at the nuclear periphery. By contrast, asexual stage inactive gene promoters such as Pfg27 (gametocyte marker) and Pfs28 (ookinete marker) occupy H3K9Ac devoid zones at the nuclear periphery. The histone H3K9 is predominantly acetylated by the PCAF/GCN5 class of lysine acetyltransferases, which is well characterized in the parasite. Interestingly, embelin, a specific inhibitor of PCAF/GCN5 family histone acetyltransferase, selectively decreases total H3K9Ac acetylation levels (but not H3K14Ac levels) around the var gene promoters, leading to the downregulation of var gene expression, suggesting interplay among histone acetylation status, as well as subnuclear compartmentalization of different genes and their activation in the parasites. Finally, we found that embelin inhibited parasitic growth at the low micromolar range, raising the possibility of using histone acetyltransferases as a target for antimalarial therapy.

Robotic surgical tools used in minimally invasive surgeries (MIS) require miniaturized and reliable actuators for precise positioning and control of the end-effector. Miniature pneumatic artificial muscles (MPAMs) are a good choice due to their inert nature, high force to weight ratio, and fast actuation. In this paper, we present the development of miniaturized braided pneumatic muscles with an outer diameter of similar to 1.2 mm, a high contraction ratio of about 18%, and capable of providing a pull force in excess of 4 N at a supply pressure of 0.8 MPa. We present the details of the developed experimental setup, experimental data on contraction and force as a function of applied pressure, and characterization of the MPAM. We also present a simple kinematics and experimental data based model of the braided pneumatic muscle and show that the model predicts contraction in length to within 20% of the measured value. Finally, a robust controller for the MPAMs is developed and validated with experiments and it is shown that the MPAMs have a time constant of similar to 10 ms thereby making them suitable for actuating endoscopic and robotic surgical tools.

How similar species co-exist in nature is a fundamental question in community ecology. Resource partitioning has been studied in desert lizard communities across four continents, but data from South Asia is lacking. We used area-constrained visual encounter surveys to study community composition and spatial and temporal resource partitioning in a lizard community during summer in the Thar Desert, western India, addressing an important biogeographic gap in knowledge. Twelve one-hectare grids divided into 25 m x 25 m plots were placed across four habitats barren dunes, stabilized dunes, grassland, and rocky hills. We recorded 1039 sightings of 12 species during 84 sampling sessions. Lizard abundance decreased in the order stabilized dunes > grassland > barren dunes > rocky hills; richness was in roughly the opposite order. Resource partitioning was examined for the seven commonest species. Overall spatial overlap was low (<0.6) between species pairs. Overlap was higher within habitats, but species showed finer separation through use of different microhabitat categories and specific spatial resources, as well as by positioning at different distances to vegetation. Diurnal species were also separated by peak time of activity. Space appears to be an important resource dimension facilitating coexistence in this desert lizard community. (C) 2015 Elsevier Ltd. Novel BioBr/Cd(OH)(2) heterostructures were synthesized by a facile chemical bath method under ambient conditions. A series of BiOBr/Cd(OH)(2) heterostructures were obtained by tuning the Bi/Cd molar ratios. The obtained heterostructures were characterized by powder X-ray diffraction (PXRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and energy dispersive spectroscopy (EDS). Optical properties were studied by UV-visible spectroscopy, diffuse reflectance spectroscopy and photoluminescence (PL). Photocatalytic studies on rhodamine B (RhB) under visible light irradiation showed that the heterostructures are very efficient photocatalysts in mild basic medium. Scavenger test studies confirmed that the photogenerated holes and superoxide radicals (O-2(center dot-)) are the main active species responsible for RhB degradation. Comparison of photoluminescence (PL) intensity suggested that an inhibited charge recombination is crucial for the degradation process over these photocatalysts. Moreover, relative positioning of the valence and conduction band edges of the semiconductors, O-2/O-2(center dot-) and (OH)-O-center dot/H2O redox potentials and HOMO-LUMO levels of RhB appear to be responsible for the hole-specificity of degradation. Photocatalytic recycling experiments indicated the high stability of the catalysts in the reaction medium without any significant loss of activity. This study hence concludes that the heterojunction constructed between Cd(OH)(2) and BiOBr interfaces play a crucial role in influencing the charge carrier dynamics and subsequent photocatalytic activity.

Measurement of out-of-plane linear motion with high precision and bandwidth is indispensable for development of precision motion stages and for dynamic characterization of mechanical structures. This paper presents an optical beam deflection (OBD) based system for measurement of out-of-plane linear motion for fully reflective samples. The system also achieves nearly zero cross-sensitivity to angular motion, and a large working distance. The sensitivities to linear and angular motion are analytically obtained and employed to optimize the system design. The optimal shot-noise limited resolution is shown to be less than one angstrom over a bandwidth in excess of 1 kHz. Subsequently, the system is experimentally realized and the sensitivities to out-of-plane motions are calibrated using a novel strategy. The linear sensitivity is found to be in agreement with theory. The angular sensitivity is shown to be over 7.5-times smaller than that of conventional OBD. Finally, the measurement system is employed to measure the transient response of a piezo-positioner, and, with the aid of an open-loop controller, reduce the settling time by about 90%. It is also employed to operate the positioner in closed-loop and demonstrate significant minimization of hysteresis and positioning error.

Despite extensive research into triosephosphate isomerases (TIMs), there exists a gap in understanding of the remarkable conjunction between catalytic loop-6 (residues 166-176) movement and the conformational flip of Glu165 (catalytic base) upon substrate binding that primes the active site for efficient catalysis. The overwhelming occurrence of serine at position96 (98% of the 6277 unique TIM sequences), spatially proximal to E165 and the loop-6 residues, raises questions about its role in catalysis. Notably, Plasmodium falciparum TIM has an extremely rare residuephenylalanineat this position whereas, curiously, the mutant F96S was catalytically defective. We have obtained insights into the influence of residue96 on the loop-6 conformational flip and E165 positioning by combining kinetic and structural studies on the PfTIM F96 mutants F96Y, F96A, F96S/S73A, and F96S/L167V with sequence conservation analysis and comparative analysis of the available apo and holo structures of the enzyme from diverse organisms.

Resumen: El artículo afirma que en la Teatrología argentina de los últimos años ha crecido la conciencia y la responsabilidad de los investigadores respecto de la pregunta epistemológica. Se llama base epistemológica para el estudio teatral a la elección de las condiciones de conocimiento que determinan los marcos, las capacidades y las limitaciones teóricas, metodológicas, historiológicas, analíticas, críticas y pedagógicas de un investigador ante su objeto de estudio. La determinación de la base epistemológica depende del posicionamiento consciente del investigador respecto de su relación científica con el teatro. El artículo afirma que diferentes líneas científicas construyen concepciones del teatro diferentes. Finalmente se describe la base epistemológica de la Filosofía del Teatro, disciplina de innovación en los estudios teatrales de la Argentina.

Resumen: El clima emocional no es la simple suma de las emociones individuales sino un afecto colectivo generado por cómo los individuos interactúan unos con otros como respuestas colectivas a sus condiciones económicas, políticas y sociales (de Rivera, 2014). Por su parte, en el contexto argentino la inseguridad es el principal problema social percibido en los últimos años. Sobre este marco, se realizó un estudio con el objetivo de analizar el clima emocional, la percepción de inseguridad y el miedo al delito junto a otros factores psicosociales asociados, y de explorar los perfiles perceptivos diferenciales según el auto-posicionamiento ideológico. La muestra, intencional, estuvo compuesta por 516 estudiantes universitarios. Los resultados dan cuenta de un clima emocional negativo (enfado y desesperanza), baja confianza institucional, frustración anómica y alta percepción de inseguridad. Se observan diferencias al comparar a los participantes en función de su autoposicionamiento ideológico. La percepción del clima emocional es más positivo cuánto más a la izquierda se ubican los participantes, manifestando mayor seguridad, menor desesperanza y enfado. Exhiben además menos miedo al delito, menor preocupación por la inseguridad y menor probabilidad de victimización. Sin embargo, quienes se posicionan ideológicamente hacia la derecha muestran mayores niveles de frustración anómica, y la confianza (o desconfianza) institucional varía por el posicionamiento ideológico en función de la institución. Finalmente, la heteropercepción de inseguridad es mayor que la autopercepción, surgiendo de este modo mecanismos defensivos como la ilusión de invulnerabilidad, que conllevan mayor riesgo.

Resumen: En el presente trabajo se analiza el aporte que un grupo de poetas, políticos e intelectuales argentinos en la revista Latinidad –creada por el impresor francés Mauricio Bouxin en 1920 y reeditada entre 1939 y 1947–, para dar cuenta de una posible apertura en la recepción de colaboraciones. Esto, en una variedad de sentidos: en cuanto a la nacionalidad y pertenencia de quienes escribían, su espectro ideológico y militancia, el posicionamiento en cuanto a la Segunda Guerra Mundial –tópico principal de la publicación– y, en cuanto a opiniones sobre política interna argentina. Plantearemos cómo la latinidad fue propuesta como un factor cohesionante, no sólo dentro de la propia comunidad francesa, sino en el contexto latinoamericano con el que pretendían estrechar lazos de identidad compartida, en el marco del conflicto bélico mundial.

Using fluorescence microscopy with single molecule sensitivity it is now possible to follow the movement of individual fluorophore tagged molecules such as proteins and lipids in the cell membrane with nanometer precision. These experiments are important as they allow many key biological processes on the cell membrane and in the cell, such as transcription, translation and DNA replication, to be studied at new levels of detail. Computerized microscopes generate sequences of images (in the order of tens to hundreds) of the molecules diffusing and one of the challenges is to track these molecules to obtain reliable statistics such as speed distributions, diffusion patterns, intracellular positioning, etc. The data set is challenging because the molecules are tagged with a single or small number of fluorophores, which makes it difficult to distinguish them from the background, the fluorophore bleaches irreversibly over time, the number of tagged molecules are unknown and there is occasional loss of signal from the tagged molecules. All these factors make accurate tracking over long trajectories difficult. Also the experiments are technically difficulty to conduct and thus there is a pressing need to develop better algorithms to extract the maximum information from the data. For this purpose we propose a Bayesian approach and apply our technique to synthetic and a real experimental data set.

[ES] A lo largo de la última década, se ha originado un importante debate acerca de lo que debe entenderse por valor de marca. En la delimitación de este concepto se han utilizado varias perspectivas de análisis y criterios de estimación muy diferentes, de ahí que aún exista sobre este tema una excesiva ambigüedad. En este trabajo se presenta un marco teórico donde se contemplan distintas perspectivas de estudio y varios criterios de estimación del valor de marca. A partir de este marco, se analizan empíricamente siete criterios de estimación de acuerdo con la información recogida de una muestra de usuarios sobre seis marcas de zapatillas de deporte. Con ello se pretende contribuir a un mejor conocimiento del concepto de valor de marca.

[ES] El objetivo principal de este trabajo es confirmar si la ocupación de espacio por parte de las marcas de distribuidor en los lineales es excesivo, tal y como parecen afirmar los productores de marcas de fabricante. Para ello, se presentan los resultados de una observación del espacio ocupado por estas marcas en una muestra representativa de 40 categorías en 55 establecimientos. Con los datos obtenidos, se muestran las diferencias existentes entre categorías y entre enseñas. Además se observa si los establecimientos han llegado a un punto de lineal máximo y si el espacio ocupado por sus marcas propias es desproporcionado en relación a su cuota de mercado.

[ES] La apuesta de la distribución por la extensión de su marca a las categorías de productos duraderos, como electrodomésticos y productos electrónicos para el hogar, ha demostrado ser acertada, a pesar de las críticas en contra de esta estrategia. Posiblemente, la mayor familiaridad del consumidor con las marcas del distribuidor, el cambio de actitud favorable hacia éstas, la disponibilidad de la tecnología, y por qué no, la crisis económica internacional que afecta a las economías domésticas, pueden ser factores explicativos de este éxito.

[ES] Las redes virtuales de fabricación global (RVFGs) están formadas por empresas independientes las cuales establecen entre sí relaciones de tipo horizontal y vertical, pudiendo incluso ser competidores, donde no es necesario mantener internamente grandes recursos fabriles sino gestionar y compartir eficientemente los recursos de la red.
