Sequences of the coat protein amino acids of definitive and tentative species of carlaviruses deposited in GenBank were aligned and a region of seven amino acids (GLGVPTE) was found to be conserved. The corresponding nucleotides were aligned, allowing the design of a degenerate primer that together with an oligo dT anti-sense primer, was effective for the detection of three distinct carlavirus species, two transmitted by aphids and one by whitefly. These primers have the advantage that about 940 nt from the 3'-terminus, comprising part of the CP gene (about 60%), the 11 K gene, and the terminal untranslated region can be amplified for sequencing. The fact that this amino acid sequence is conserved in almost all of the sequenced carlaviruses, allows the prediction that this primer pair will be useful as a diagnostic tool for carlavirus species. Using the numerical solution of the nonlinear Schrodinger equation and a variational method it is shown that (3 + 1)-dimensional spatiotemporal optical solitons can be stabilized by a rapidly oscillating dispersion coefficient in a Kerr medium with cubic nonlinearity. This has immediate consequence in generating dispersion-managed robust optical soliton in communication as well as possible stabilized Bose-Einstein condensates in periodic optical-lattice potential via an effective-mass formulation. We also critically compare the present stabilization with that obtained by a rapid sinusoidal oscillation of the Kerr nonlinearity parameter.

In this paper, we explicitly construct an infinite number of Hopfions (static, soliton solutions with nonzero Hopf topological charges) within the recently proposed (3 + 1)-dimensional, integrable, and relativistically invariant field theory. Two integers label the family of Hopfions we have found. Their product is equal to the Hopf charge which provides a lower bound to the soliton's finite energy. The Hopfions are explicitly constructed in terms of the toroidal coordinates and shown to have a form of linked closed vortices.

Let alpha be a C(infinity) curve in a homogeneous space G/H. For each point x on the curve, we consider the subspace S(k)(alpha) of the Lie algebra G of G consisting of the vectors generating a one parameter subgroup whose orbit through x has contact of order k with alpha. In this paper, we give various important properties of the sequence of subspaces G superset of S(1)(alpha) superset of S(2)(alpha) superset of S(3)(alpha) superset of ... In particular, we give a stabilization property for certain well-behaved curves. We also describe its relationship to the isotropy subgroup with respect to the contact element of order k associated with alpha.

We performed a light microscope and a computer three-dimensional reconstruction study of serial sections of the molar enamel organ of 3- and 5-day-old rats perfused with Indian ink through the arterial system. The tooth germs were fixed in Bouin's solution, embedded in paraffin, sectioned and stained with haematoxylin and eosin. For the three-dimensional reconstruction, light micrographs of the serial sections were digitized, and aligned using the serial EM Align software downloaded from http://synapses.bu.edu/tools/. After alignment, the boundaries of the India-ink-filled blood vessels were manually traced with a mouse using the software IGL trace (version 1.26b), also downloaded from the above website. After tracing, a three-dimensional representation of the blood vessel contours was generated in a VRML format and visualized with the help of the software Cortona Web3D viewer (version 4.0) downloaded from http://www.parallelgraphics.com/products/cortona. Our results showed that in regions where ameloblasts are polarized the capillaries are arranged in three distinct levels: (1) penetrating and leaving capillaries in relation to the outer enamel epithelium; (2) capillaries crossing and branching inside the stellate reticulum; and (3) capillaries branching and anastomosing profusely within the stratum intermedium, thereby forming an extensive capillary plexus intimately associated with the cells of the stratum intermedium. The existence of a conspicuous capillary plexus intermingled with cells of the stratum intermedium, as shown in our results, suggests that some molecules produced by cells of the stratum intermedium could be released into the capillary plexus and thereafter carried to the dental follicle.

The negative-dimensional integration method (NDIM) seems to be a very promising technique for evaluating massless and/or massive Feynman diagrams. It is unique in the sense that the method gives solutions in different regions of external momenta simultaneously. Moreover, it is a technique whereby the difficulties associated with performing parametric integrals in the standard approach are transferred to a simpler solving of a system of linear algebraic equations, thanks to the polynomial character of the relevant integrands. We employ this method to evaluate a scalar integral for a massless two-loop three-point vertex with all the external legs off-shell, and consider several special cases for it, yielding results, even for distinct simpler diagrams. We also consider the possibility of NDIM in non-covariant gauges such as the light-cone gauge and do some illustrative calculations, showing that for one-degree violation of covariance (i.e. one external, gauge-breaking, light-like vector n μ) the ensuing results are concordant with the ones obtained via either the usual dimensional regularization technique, or the use of the principal value prescription for the gauge-dependent pole, while for two-degree violation of covariance - i.e. two external, light-like vectors n μ, the gauge-breaking one, and (its dual) n * μ - the ensuing results are concordant with the ones obtained via causal constraints or the use of the so-called generalized Mandelstam-Leibbrandt prescription. © 1999 Elsevier Science B.V.

Hrp1p is a heterogeneous ribonucleoprotein (hnRNP) from the yeast Saccharomyces cerevisiae that is involved in the cleavage and polyadenylation of the 3'-end of mRNAs and mRNA export. In addition, Hrp1p is one of several RNA-binding proteins that are posttranslationally modified by methylation at arginine residues. By using-functional recombinant Hrp1p, we have identified RNA sequences with specific high affinity binding sites. These sites correspond to the efficiency element for mRNA 3'-end formation, UAUAUA. To examine the effect of methylation on specific RNA binding, purified recombinant arginine methyltransferase (Hmt1p) was used to methylate Hrp1p. Methylated Hrp1p binds with the same affinity to UAUAUA-containing RNAs as unmethylated Hrp1p indicating that methylation does not affect specific RNA binding. However, RNA itself inhibits the methylation of Hrp1p and this inhibition is enhanced by RNAs that specifically bind Hrp1p. Taken together, these data support a model in which protein methylation occurs prior to protein-RNA binding in the nucleus.

Group theoretical-based techniques and fundamental results from number theory are used in order to allow for the construction of exact projectors in finite-dimensional spaces. These operators are shown to make use only of discrete variables, which play the role of discrete generator coordinates, and their application in the number symmetry restoration is carried out in a nuclear BCS wave function which explicitly violates that symmetry. © 1999 Published by Elsevier Science B.V. This paper presents a numerical approach to model the complex failure mechanisms that define the ultimate rotational capacity of reinforced concrete beams. The behavior in tension and compression is described by a constitutive damage model derived from a combination of two specific damage models [1]. The nonlinear behavior of the compressed region is treated by the compressive damage model based on the Drucker-Prager criterion written in terms of the effective stresses. The tensile damage model employs a failure criterion based on the strain energy associated with the positive part the effective stress tensor. This model is used to describe the behavior of very thin bands of strain localization, which are embedded in finite elements to represent multiple cracks that occur in the tensioned region [2]. The softening law establishes dissipation energy compatible with the fracture energy of the concrete. The reinforcing steel bars are modeled by truss elements with elastic-perfect plastic behavior. It is shown that the resulting approach is able to predict the different stages of the collapse mechanism of beams with distinct sizes and reinforcement ratios. The tensile damage model and the finite element embedded crack approach are able to describe the stiffness reduction due to concrete cracking in the tensile zone. The truss elements are able to reproduce the effects of steel yielding and, finally, the compressive damage model is able to describe the non-linear behavior of the compressive zone until the complete collapse of the beam due to crushing of concrete. The proposed approach is able to predict well the plastic rotation capacity of tested beams [3], including size-scale effects.

Background. Characterization of novel rodent models for prostate cancer studies requires evaluation of either spontaneous and carcinogen-induced tumors as well as tumor incidence in different prostatic lobes. We propose a new short-term rodent model of chemically induced prostate carcinogenesis in which prostate cancer progression occurs differentially in the dorsolateral and ventral lobes. Methods. Adult gerbils were treated with MNU alone or associated with testosterone for 3 or 6 months of treatment. Tumor incidence, latency, localization, and immunohistochemistry (AR, PCNA, smooth muscle α-actin, p63, MGMT, and E-cadherin) were studied in both lobes. Results. Comparisons between both lobes revealed that lesions developed first in the DL while the VL presented longer tumor latency. However, after 6 months, there was a dramatic increase in tumor multiplicity in the VL, mainly in MNU-treated groups. Lesions clearly progressed from a premalignant to a malignant phenotype over time and tumor latency was decreased by MNU + testosterone administration. Three-dimensional reconstruction of the prostatic complex showed that the DL developed tumors exclusively in the periurethral area and showed intense AR, PCNA, and MGMT immunostaining. Moreover, VL lesions emerged throughout the entire lobe. MNU-induced lesions presented markers indicative of an aggressive phenotype: lack of basal cells, rupture of the smooth muscle cell layer, loss of E-cadherin, and high MGMT staining. Conclusions. There are distinct pathways involved in tumor progression in gerbil prostate lobes. This animal provides a good model for prostate cancer since it allows the investigation of advanced steps of carcinogenesis with shorter latency periods in both lobes. Este trabalho apresenta um estudo teórico sobre novos circuladores compactos com 3-portas tipos W e Y, baseados em cristais fotônicos bidimensionais. No circulador tipo Y, os guias de onda que o compõem formam ângulos de 120° entre si (com formato assemelhado ao da letra Y). O circulador tipo W é uma modificação do tipo Y, obtido a partir do reposicionamento de uma das portas entre as outras duas com um ângulo de 60° entre os guias de onda (com formato assemelhado ao da letra W). Os parâmetros geométricos dos cristais foram obtidos dos diagramas de bandas proibidas. O circulador de três portas tipo Y, projetado para operar em frequências de micro-ondas, foi investigado com o objetivo de gerar um protótipo inédito, enquanto que o tipo W, para frequências ópticas, foi investigado para demonstrar a possibilidade de desenvolver um circulador mais compacto em comparação com o tipo Y conhecido. O tipo W pode ser também uma alternativa geométrica mais adequada no design de circuitos integrados. Os modelos são bons no sentido em que possuem elevada isolação (maior que -20 dB em ambos os circuladores) e baixa perda de inserção (maior que -0,5 dB no caso do circulador tipo Y). O circulador tipo W apresenta uma largura de banda de operação em torno de 100 GHz para um nível de -20 dB de isolação, centrado no comprimento de onda de 1,5um. As simulações foram feitas utilizando-se o software comercial COMSOL Multiphysics, o qual se baseia no método dos elementos finitos.

This paper aims to contribute to the three-dimensional generalization of numerical prediction of crack propagation through the formulation of finite elements with embedded discontinuities. The analysis of crack propagation in two-dimensional problems yields lines of discontinuity that can be tracked in a relatively simple way through the sequential construction of straight line segments oriented according to the direction of failure within each finite element in the solid. In three-dimensional analysis, the construction of the discontinuity path is more complex because it requires the creation of plane surfaces within each element, which must be continuous between the elements. In the method proposed by Chaves (2003) the crack is determined by solving a problem analogous to the heat conduction problem, established from local failure orientations, based on the stress state of the mechanical problem. To minimize the computational effort, in this paper a new strategy is proposed whereby the analysis for tracking the discontinuity path is restricted to the domain formed by some elements near the crack surface that develops along the loading process. The proposed methodology is validated by performing three-dimensional analyses of basic problems of experimental fractures and comparing their results with those reported in the literature.

Given the spread of antibiotic resistance in bacterial pathogens, antimicrobial peptides that can also modulate the immune response may be a novel approach for effectively controlling periodontal infections. In the present study, we used a three-dimensional (3D) co-culture model of gingival epithelial cells and fibroblasts stimulated with Aggregatibacter actinomycetemcomitans lipopolysaccharide (LPS) to investigate the anti-inflammatory properties of human beta-defensin-3 (hBD-3) and cathelicidin (LL-37) and to determine whether these antimicrobial peptides can act in synergy. The 3D co-culture model composed of gingival fibroblasts embedded in a collagen matrix overlaid with gingival epithelial cells had a synergistic effect with respect to the secretion of IL-6 and IL-8 in response to LPS stimulation compared to fibroblasts and epithelial cells alone. The 3D co-culture model was stimulated with non-cytotoxic concentrations of hBD-3 (10 and 20 mu M) and LL-37 (0.1 and 0.2 mu M) individually and in combination in the presence of A. actinomycetemcomitans LPS. A multiplex ELISA assay was used to quantify the secretion of 41 different cytokines. hBD-3 and LL-37 acted in synergy to reduce the secretion of GRO-alpha, G-CSF, IP-10, IL-6, and MCP-1, but only had an additive effect on reducing the secretion of IL-8 in response to A. actinomycetemcomitans LPS stimulation. The present study showed that hBD-3 acted in synergy with LL-37 to reduce the secretion of cytokines by an LPS-stimulated 3D model of gingival mucosa. This combination of antimicrobial peptides thus shows promising potential as an adjunctive therapy for treating inflammatory periodontitis.

The paper describes the preliminary studies of University of Minho on the use of Electric Impedance/Resistance Tomography to assess masonry structures. The study is focused on the analysis of values of current and voltage resulting from the use of an electrical source with voltage and frequency values from a distribution network. The analysis is made from results obtained through computer simulations, using a three-dimensional model of the idealized masonry structures. A finite element program was used for the simulations. Three types of electrodes were used in simulations, and the analysis of the results led to significant conclusions. Later masonry specimens were built and a series of preliminary tests were carried out in the laboratory. The comparative analysis of simulated and experimental results allowed identifying the factors that have influence on the physical results.
