Mutations on TP53 gene are common in human cancer but not in cervical cancer where they are rarely found and the inactivation and degradation of p53 protein are attributed to the action of E6 viral oncogene from high risk human papillomavirus (HPV). Analysis of cervical cancer cell lines suggests that HPV negative samples shows mutation on TP53, but clinical approaches didn t confirmed this hypothesis. However, in most TP53 mutations studies on cervical cancer, only the exons 5 to 8 were analyzed. Approximately 90% of mutations described are on this region. Recent studies on several cancer suggests that mutation frequency in the other exons must be considered. The aim of this work was to verify whether mutations on coding and non-coding regions occur in cancer tissue from cervical cancer in patients from Rio Grande do Norte using Denaturing Gradient Gel Electrophoresis (DGGE) as screening tool. Exons 8 to 11 were analyzed including some introns from 80 tumor samples and 8 peripheral blood samples from healthy women. DNA were submitted to PCR using primers with GC clamp on the end of one of them. The results were observed for each region after DGGE and silver staining. It was observed no amplified fragment with different migration profile from those obtained from DNA of peripheral blood. These results agree with those from literature where TP53 mutations in cervical cancer have been described in a very low frequency

Recently, genetically encoded optical indicators have emerged as noninvasive tools of high spatial and temporal resolution utilized to monitor the activity of individual neurons and specific neuronal populations. The increasing number of new optogenetic indicators, together with the absence of comparisons under identical conditions, has generated difficulty in choosing the most appropriate protein, depending on the experimental design. Therefore, the purpose of our study was to compare three recently developed reporter proteins: the calcium indicators GCaMP3 and R-GECO1, and the voltage indicator VSFP butterfly1.2. These probes were expressed in hippocampal neurons in culture, which were subjected to patchclamp recordings and optical imaging. The three groups (each one expressing a protein) exhibited similar values of membrane potential (in mV, GCaMP3: -56 ±8.0, R-GECO1: -57 ±2.5; VSFP: -60 ±3.9, p = 0.86); however, the group of neurons expressing VSFP showed a lower average of input resistance than the other groups (in Mohms, GCaMP3: 161 ±18.3; GECO1-R: 128 ±15.3; VSFP: 94 ±14.0, p = 0.02). Each neuron was submitted to current injections at different frequencies (10 Hz, 5 Hz, 3 Hz, 1.5 Hz, and 0.7 Hz) and their fluorescence responses were recorded in time. In our study, only 26.7% (4/15) of the neurons expressing VSFP showed detectable fluorescence signal in response to action potentials (APs). The average signal-to-noise ratio (SNR) obtained in response to five spikes (at 10 Hz) was small (1.3 ± 0.21), however the rapid kinetics of the VSFP allowed discrimination of APs as individual peaks, with detection of 53% of the evoked APs. Frequencies below 5 Hz and subthreshold signals were undetectable due to high noise. On the other hand, calcium indicators showed the greatest change in fluorescence following the same protocol (five APs at 10 Hz). Among the GCaMP3 expressing neurons, 80% (8/10) exhibited signal, with an average SNR value of 21 ±6.69 (soma), while for the R-GECO1 neurons, 50% (2/4) of the neurons had signal, with a mean SNR value of 52 ±19.7 (soma). For protocols at 10 Hz, 54% of the evoked APs were detected with GCaMP3 and 85% with R-GECO1. APs were detectable in all the analyzed frequencies and fluorescence signals were detected from subthreshold depolarizations as well. Because GCaMP3 is the most likely to yield fluorescence signal and with high SNR, some experiments were performed only with this probe. We demonstrate that GCaMP3 is effective in detecting synaptic inputs (involving Ca2+ influx), with high spatial and temporal resolution. Differences were also observed between the SNR values resulting from evoked APs, compared to spontaneous APs. In recordings of groups of cells, GCaMP3 showed clear discrimination between activated and silent cells, and reveals itself as a potential tool in studies of neuronal synchronization. Thus, our results indicate that the presently available calcium indicators allow detailed studies on neuronal communication, ranging from individual dendritic spines to the investigation of events of synchrony in neuronal networks genetically defined. In contrast, studies employing VSFPs represent a promising technology for monitoring neural activity and, although still to be improved, they may become more appropriate than calcium indicators, since neurons work on a time scale faster than events of calcium may foresee

The industry's interest in having a greater control of the deformations caused by welding is due to the geometric and dimensional tolerances been more and more precise in the project specifications, motivating the manufacturing engineering to develop stable processes and to ensure routine production. Aiming at it, the main goal of this present work is to analyze how much routine situations used in automatic aluminum welding can influence on the angular deformations of this material. Using the alloy AA 5052 H34, and the automatic welding in pulsed GMAW process, three types of weaving were applied throughout the length of the weld, in butt joints assembled without groove and with 60 degrees single-V-groove, arranged transversely as well as longitudinally to the rolling direction of the plate. The measurement of the deformations was made by a three-dimensional equipment, before and after the welding, in three distinct regions in the specimens. The profile of the weld bead was the main factor for the different types of deformations found, as revealed by macrographical analysis. The 60 degrees single-V-groove had higher amplitudes of deformations as the joint without groove. The torch oscillation wasn't a variable of statistically significant influence on this amplitudes.

The results of dynamic instrumentation in two construction sites that used steel rails as pile foundation are presented in this paper. The first dynamic load tests were executed in piles made by single steel rails of the type TR32 and TR37. In the second group of dynamic tests, the piles were made by a composition of two and three steel rails of the same type TR37. The difficulties in placing the sensors, the effect of hammer blow eccentricity and the influence of the non-uniform welding along the pile length are presented in detail and discussed.

In this work, three welding programs for orbital TIG previously developed were used, using pulsed current and increasing speed (M), constant current (#B) and pulsed current and decreasing current (#C). One of those should be used for the propulsion system of the satellite CBERS (CHINA-BRAZIL EARTH RESOURCES SATELLITE). Welded joints using tubes of commercially pure titanium were obtained with these procedures, which were characterized by means of mechanical and metallographic tests. The obtained results show that the three welding procedures produce welded joints free of defects and with adequate shape. Although small differences on mechanical properties and microstructure have been observed, the three welding programs attained compatible results with international standards used in the aerospace segment. The welding program #B, due the reduced heat input used, was considered to obtain slightly advantage over the others.

The aim of this study is to analyze the effects of corrosion and successive tungsten inert gas (TIC) welding repairs on the reverse bending fatigue strength of AISI 4130 steel used in components critical to the flight-safety. The tests were performed on hot-rolled steel plate specimens, 1.10 mm and 1.60 mm thick, by means of a SCHENK PWS equipment, with load ratio R = -1, constant amplitude, 30 Hz frequency and room temperature. It was observed that the reverse bending fatigue strength of AISI 4130 steel decreases due to the corrosion and the TIC welding and re-welding processes. (C) 2010 Elsevier Ltd. All rights reserved.

The conventional radiographic technique using industrial radiographic films is with the days finishing. The Digital Radiography is taking place in several sectors, e.g., the medical, aerospace, security, automotive, etc. In addition to representing a technological trend, it has been demonstrated that digital radiography offers a series of benefits in terms of productivity, sensitivity, environmental aspects, image treatment tools, cost reduction, etc. If one weld seam to be inspected is from a serried product, as example a pipe, the best option to be implemented is the Flat Panel Detector with this equipment is possible to reduce the obtaining digital radiographic images in place of films and reducing the inspection cycle time due to its high degree of automation. In the experiments described in this paper this new technique was tested and the results were compared with those obtained by the conventional radiography. The welded specimens were prepared using the submerged-arc welding process and small artificial cracks of the most varied dimensions, present in the specimens, were used to establish a comparison of the sensitivities presented by the techniques employed After conducting several experiments, the digital method presented the highest sensitivity to the wire-type Image Quality Indicator (IQI) and in the detection of small defects, leading to the conclusion that the use of digital radiography using the flat-panel detector offers advantages over the conventional technique [1, 2]. This work was carried out based on the API 5L Edition 2004 [3] and ISO 3183 Edition 2007 [4] specifications.

In this work the effect of Gas Tungsten Arc Welding (GTAW) repairs on the axial fatigue strength of an AISI 4130 steel welded joint used in airframe critical to the flight-safety was investigated. Fatigue tests were performed at room temperature on 0.89 mm thick hot-rolled plates with constant amplitude and load ratio of R = 0.1, at 20 Hz frequency. Monotonic tensile tests, optical metallography and microhardness, residual stress and weld geometric factors measurements were also performed. The fatigue strength decreased with the number of GTAW repairs, and was related to microstructural and microhardness changes, as well as residual stress field and weld profile geometry factors, which gave origin to high stress concentration at the weld toe. OBJETIVO: Avaliar a eficiência do clampe de Ganz na estabilização e redução dos deslocamentos da pelve, quando utilizado no tratamento de urgência, além de aquilatar eventuais dificuldades e facilidades do método. MÉTODOS: O clampe de Ganz foi utilizado no tratamento de urgência em 31 pacientes com graves lesões do anel pélvico (Tile C) associadas a importante instabilidade hemodinâmica. RESULTADO: Entre os pacientes, 27 (87,1%) apresentaram evolução favorável, com estabilização, redução dos deslocamentos e compressão da região posterior do anel, além de estabilização hemodinâmica, e quatro (12,9%) evoluíram para óbito. CONCLUSÃO: O clampe de Ganz mostrou-se eficiente ferramenta no tratamento de urgência das lesões do anel pélvico, por ser de concepção simples, de rápida colocação, não impedir ou dificultar procedimentos no abdome e, principalmente, por permitir a estabilização da pelve, redução dos deslocamentos e compressão na região posterior do anel, local onde ocorrem os maiores sangramentos, reduzindo-os ou eliminando-os.

Aim. Lower-limb traumatic injury associated with ischemia and followed by reperfusion (I/R) is a common severe situation in muscle lesions due to trauma and hypoxia followed by local and systemic injuries induced by oxygen-derived free radical release during reperfusion. The aim of this study was to evaluate the attenuating effects of trimetazidine (TMZ) and N-acetylcysteine (NAC) in such situation.Methods. The muscles at the root of the right hind limb of Wistar rats were cross-sectioned, preserving femoral vessels and nerves and clamping the femoral artery for four hours. The clamp was then released and the femoral artery has been reperfused for 2 hours. Rats were randomly divided in groups of ten as follows: Group 1: sham I/R, treated with saline; Group 2: I/R, treated with saline; Group 3: sham I/R, treated with TMZ (7.5 mg/kg/dose); Group 4: sham I/R, treated with NAC (375 mg/kg/dose); Group 5: I/R treated with TMZ (7.5 mg/kg/dose); Group 6: I/R treated with NAC (375 mg/kg/dose). All rats received two intravenous bolus injections of the drugs, one before ischemia and one before reperfusion. Oxidative stress in plasma (MDA, total, oxidized and reduced glutathione), creatinephosphokinase (CPK), optical and electron microscopy and pelvic extremity circumference and volume were studied.Results. No statistical differences were found between the groups for MDA or total and reduced glutathione. Oxidized glutathione increased significantly in groups 5 and 2. Limb circumference as well as limb volume increased in all groups over time, mainly in groups 5, 2 and 1. CPK increased in all groups, being highest in groups 5, 6 and 2. Histological lesions were present in all but sham groups, being less severe in group 6. Soleus muscle analyses at electron microscopy exhibit some degree of alteration in all groups.Conclusion. This experimental model simulated severe limb trauma associated with ischemia and reperfusion, and, as such, it was aggressive, causing severe injury and local inflammatory reaction. The model did not show antioxidant action from NAC, and possible antioxidant action from TMZ was insufficient to attenuate tissue injuries. [Int Angiol 2009;28:412-7]

To identify early metabolic abnormalities in type 2 diabetes mellitus, we measured insulin secretion, sensitivity to insulin, and hepatic insulin extraction in 48 healthy normal glucose-tolerant Brazilians, first-degree relatives of type 2 diabetic patients (FH+). Each individual was matched for sex, age, weight, and body fat distribution with a person without history of type 2 diabetes (FH-). Both groups were submitted to a hyperglycemic clamp procedure (180 mg/dl). Insulin release was evaluated in its two phases. The first was calculated as the sum of plasma insulin at 2.5, 5.0, 7.5, and 10.0 min after the beginning of glucose infusion, and the second as the mean plasma insulin level in the third hour of the clamp procedure. Insulin sensitivity index (ISI) was the mean glucose infusion rate in the third hour of the clamp experiment divided by the mean plasma insulin concentration during the same period of time. Hepatic insulin extraction was determined under fasting conditions and in the third hour of the clamp procedure as the ratio between C-peptide and plasma insulin levels. FH+ individuals did not differ from FH- individuals in terms of the following parameters [median (range)]: a) first-phase insulin secretion, 174 (116-221) vs 207 (108-277) µU/ml, b) second-phase insulin secretion, 64 (41-86) vs 53 (37-83) µU/ml, and c) ISI, 14.8 (9.0-20.8) vs 16.8 (9.0-27.0) mg kg-1 min-1/µU ml-1. Hepatic insulin extraction in FH+ subjects was similar to that of FH- ones at basal conditions (median, 0.27 vs 0.27 ng/µU) and during glucose infusion (0.15 vs 0.15 ng/µU). Normal glucose-tolerant Brazilian FH+ individuals well-matched with FH- ones did not show defects of insulin secretion, insulin sensitivity, or hepatic insulin extraction as tested by hyperglycemic clamp procedures.

The Brasiliano Cycle in the Seridó Belt (NE Brazil) is regarded mostly as a crustal reworking event, characterized by transcurrent or transpressional shear zones which operated under high temperature and low pressure conditions. In the eastern domain of this belt- the so-called São José de Campestre Massif (SJCM), a transtensional deformation regime is evidenced by extensional components or structures associated to the strikeslip shear zones. The emplacement of the Neoproterozoic Brasiliano granitoids is strongly controled by these discontinuities. Located in the southern border of the SJCM, the Remígio-Pocinhos shear zone (RPSZ) displays, in its northern half, top to the SW extensional movement which progressively grade, towards its southern half, to a dextral strike-slip kinematics, defining a negative semi-flower structure. This shear zone is overprinted upon allocthonous metasediments of the Seridó Group and an older gneiss-migmatite complex, both of which containing metamorphic parageneses from high amphibolite to granulite facies (the latter restricted to the strike-slip zone), defining the peak conditions of deformation. Several granitoid plutons are found along this structure, emplaced coeval with the shearing event. Individually, such bodies do not exceed 30 km2 in outcropping area and are essentially parallel to the trend of the shear zone. Petrographic, textural and geochemical data allow to recognize five different granitoid suites along the RPSZ: porphyritic granites (Serra da Boa Vista and Jandaíra), alkaline granites (Serra do Algodão and Serra do Boqueirão) and medium to coarse-grained granites (Olivedos) as major plutons, while microgranite and aluminous leucogranite sheets occur as minor intrusions. The porphyritic granites are surrounded by metasediments and present sigmoidal or en cornue shapes parallel to the trend of the RPSZ, corroborating the dextral kinematics. Basic to intermediate igneous enclaves are commonly associated to these bodies, frequently displaying mingling textures with the host granitoids. Compositionally these plutons are made up by titanite-biotite monzogranites bearing amphibole and magnetite; they are peraluminous and show affinities to the monzonitic, subalkaline series. Peraluminous, ilmenite-bearing biotite monzogranites and titanite-biotite monzogranites correspond, respectivally, to the Olivedos pluton and the microgranites. The Olivedos body is hosted by metasediments, while the microgranites intrude the gneiss-migmatite complex. Being highly evolved rocks, samples from these granites plot in the crustal melt fields in discrimination diagrams. Nevertheless, their subtle alignment also looks consistent with a monzonitic, subalkaline affinity. These chemical parameters make them closer to the I-type granites. Alkaline, clearly syntectonic granites are also recognized along the RPSZ. The Serra do Algodão and Serra do Boqueirão bodies display elongated shapes parallel to the mylonite belt which runs between the northern, extensional domain and the southern strike-slip zone. The Serra do Algodão pluton shows a characteristic isoclinal fold shape structure. Compositionally they encompass aegirine-augite alkali-feldspar granites and quartz-bearing alkaline syenite bearing garnet (andradite) and magnetite plus ilmenite as opaque phases. These rocks vary from meta to peraluminous, being correlated to the A-type granites. Aluminous leucogranites bearing biotite + muscovite ± sillimanite ± garnet (S-type granites) are frequent but not volumetrically important along the RPSZ. These sheet-like bodies may be folded or boudinaged, representing partial melts extracted from the metasediments during the shear zone development. Whole-rock Rb-Sr isotope studies point to a minimum 554��10 Ma age for the crystalization of the porphyritic granites. The alkaline granites and the Olivedos granite produced ca. 530 Ma isochrons which look too young; such values probably represent the closure of the Rb-Sr radiometric clock after crystallization and deformation of the plutons, at least 575 Ma ago (Souza et al. 1998). The porphyritic and the alkaline granites crystallized under high oxygen fugacity conditions, as shown by the presence of both magnetite and hematite in these rocks. The presence of ilmenite in the Olivedos pluton suggests less oxidizing conditions. Amphibole and amphibole-plagioclase thermobarometers point to minimum conditions, around 750°C and 6 Kbars, for the crystallization of the porphyritic granites. The zirconium geothermometer indicates higher temperatures, in the order of 800°C, for the porphyritic granites, and 780°C for the Olivedos pluton. Such values agree with the thermobarometric data optained for the country rocks (5,7 Kbar and 765°C; Souza et al. 1998). The geochemical and isotope data set point to a lower crustal source for the porphyritic and the alkaline granites. Granulite facies quartz diorite to tonalite gneisses, belonging or akin to the gneiss-migmatite complex, probably dominate in the source regions. In the case of the alkaline rocks, subordinate contributions of mantle material may be present either as a mixing magma or as a previously added component to the source region. Tonalite to granodiorite gneisses, with some metasedimentary contribution, may be envisaged for the Olivedos granite. The diversity of granitoid rocks along the RPSZ is explained by its lithospheric dimension, allowing magma extraction at different levels, from the middle to lower crust down to the mantle. The presence of basic to intermediate enclaves, associated to the porphyritic granites, confirm the participation of mantle components in the magma extraction system along the RPSZ. This mega-structure is part of the network of Brasiliano-age shear zones, activated by continental collision and terrane welding processes at the end of the Neoproterozoic

The collection of epididymal sperm may be a valuable tool for canine reproduction especially since it can enable collection of cells after death of a valuable dog. The aim of the present study was to evaluate the viability of epididymal sperm after freeze-thawing. Epididymides were obtained from four adult dogs by elective orchiectomy. The caudal portion of the epididymides and part of the deferential ducts were squeezed by means of an anatomic clamp into a Petri dish containing either 0.9% saline solution (Group 1) or Ringer solution without lactate (Group 2). Samples were centrifuged at 800 x g for 10 min, the supernatant was removed and the pellet was diluted in one step with a Tris/citric acid/OEP (Orvus Es Paste) extender containing 7% glycerol and subjected to semen freezing. Oocytes were obtained from canine ovaries, after ovariohysterectomy. Only oocytes that were approximately 100 mu m in diameter, with a dark ooplasm surrounded by three-or four-well formed cumulus cell layers were used for sperm testing. Frozen semen samples were thawed in a water bath at 70 degrees C for 8 s and analysed at room temperature for sperm motility and velocity. Oocytes were incubated with spermatozoa in humidified atmosphere containing 5% CO(2) at 38 degrees C for 18 h. Morphological and functional characteristics of spermatozoa were similar in both groups. However, the percentage of sperm cells bound to oocytes was significantly higher in Group 2 than in Group 1. This result suggests that the Ringer solution without lactate was a more suitable medium for collecting epididymal canine sperm than 0.9% saline.
