Objective. To access the incidence of infectious problems after liver transplantation (LT). Design. A retrospective, single-center study. Materials and Methods. Patients undergoing LT from January 2008 to December 2011 were considered. Exclusion criterion was death occurring in the first 48 hours after LT. We determined the site of infection and the bacterial isolates and collected and compared recipient’s variables, graft variables, surgical data, post-LT clinical data. Results. Of the 492 patients who underwent LT and the 463 considered for this study, 190 (Group 1, 41%) developed at least 1 infection, with 298 infections detected. Of these, 189 microorganisms were isolated, 81 (51%) gram-positive bacteria (most frequently Staphylococcus spp). Biliary infections were more frequent (mean time of 160.4 167.7 days after LT); from 3 months after LT, gram-negative bacteria were observed (57%). Patients with infections after LT presented lower aminotransferase levels, but higher requirements in blood transfusions, intraoperative vasopressors, hemodialysis, and hospital stay. Operative and cold ischemia times were similar. Conclusion. We found a 41% incidence of all infections in a 2-year follow-up after LT. Gram-positive bacteria were more frequent isolated; however, negative bacteria were commonly isolated later. Clinical data after LT were more relevant for the development of infections. Donors’ variables should be considered in future analyses.

Single organisms of Trypanosoma cruzi of the virulent Peru strain were isolated by direct visualisation and were injected peritoneally into CFI mice. Single trypanomastigotes of different morphology and from different sources (mouse blood, in vitro cufture and bug faeces) were used. Single trypanomastigotes from mouse blood caused parasitaemia and fataiity in a high percentage of mice. This was true irrespective of the morphology (broad or narrow form) of the trypano mastigo tec. Single organisms of the culture forms were also capable of causing infection, although these were less infectious than single trypanomastigotes obtained from mouse blood or the reduviid bug. The difficulties attendant on the performance of the cloning technique are discussed and some indication is given of how these problems can be overcome.

INTRODUCTION: The aim of this preliminary work is to analyze the clinical features of 52 patients with a functional transplanted kidney for >25 years (all first transplant and all deceased donor recipients) and to compare with a similar though more complete study from Hôpital Necker-Paris 2012. METHODS: The mean graft survival at 25 years is 12.7% and at 30 years is 10%. The actual mean serum creatinine concentration is 1.3 mg/L. We analyzed recipient age (mean, 35.9 years) and gender (29 men and 23 women). Donor age was 26.7 ± 10.3 years. Seven patients (13.4%) were transplanted with 1 HLA mismatch, 42.3% with 2 mismatches, and 44.2% with 3 mismatches. Mean cold ischemia time was 15.45 ± 7.7 hours. Of the recipients, 76% had immediate graft function; 38% experienced 1 acute rejection episode and 4 patients had 2 rejection crises. The initial immunosuppressive regimen was azathioprine (AZA) + prednisolone (Pred) in 14 patients, cyclosporin (CSA) + Pred in 13 patients, and CSA + AZA + Pred in 25 patients. Of these patients, 19% maintained their initial regimen, and 54% (28 patients) were very stable on a mixed CSA regimen for >25 years. RESULTS: We present the major complications (diabetes, neoplasia, and hepatitis C virus positivity). CONCLUSION: Our results in deceased donor kidney recipients for >25 years are similar to the mixed population (deceased donors and living donors) presented by the Necker group, although 54% of our patients remain on CSA immunosuppression, contradicting the idea that its use is not compatible with good long-term kidney function in transplant recipients.

INTRODUCTION: New scores have been developed and validated in the US for in-hospital mortality risk stratification in patients undergoing coronary angioplasty: the National Cardiovascular Data Registry (NCDR) risk score and the Mayo Clinic Risk Score (MCRS). We sought to validate these scores in a European population with acute coronary syndrome (ACS) and to compare their predictive accuracy with that of the GRACE risk score. METHODS: In a single-center ACS registry of patients undergoing coronary angioplasty, we used the area under the receiver operating characteristic curve (AUC), a graphical representation of observed vs. expected mortality, and net reclassification improvement (NRI)/integrated discrimination improvement (IDI) analysis to compare the scores. RESULTS: A total of 2148 consecutive patients were included, mean age 63 years (SD 13), 74% male and 71% with ST-segment elevation ACS. In-hospital mortality was 4.5%. The GRACE score showed the best AUC (0.94, 95% CI 0.91-0.96) compared with NCDR (0.87, 95% CI 0.83-0.91, p=0.0003) and MCRS (0.85, 95% CI 0.81-0.90, p=0.0003). In model calibration analysis, GRACE showed the best predictive power. With GRACE, patients were more often correctly classified than with MCRS (NRI 78.7, 95% CI 59.6-97.7; IDI 0.136, 95% CI 0.073-0.199) or NCDR (NRI 79.2, 95% CI 60.2-98.2; IDI 0.148, 95% CI 0.087-0.209). CONCLUSION: The NCDR and Mayo Clinic risk scores are useful for risk stratification of in-hospital mortality in a European population of patients with ACS undergoing coronary angioplasty. However, the GRACE score is still to be preferred.

This work describes an electrochemical and quantum chemical investigation of the fipronil insecticide. Cyclic voltammetry (CV) and square wave voltammetry (SWV) experiments were performed over a graphite-polyurethane (GPU) composite electrode. The fipronil molecule presents an one?electron irreversible oxidation reaction. Profiting the SWV signal a square wave stripping voltammetry (SWSV) procedure to determine the fipronil molecule in a 0.10 mol L-1 Britton-Robinson buffer solution, pH 8.0 was developed with accumulation potential and time of 0.50 V and 120 s, respectively. The limits of detection and quantification were 0.80 and 2.67 ?g L-1, respectively. Recovery tests were performed in three natural waters samples with values ranging from 99.67 to 101.37%. Quantum chemical studies showed that the nitrogen atom of the pyrazole group is the most probable oxidation site of the fipronil molecule.

Paracoccidioidomycosis is a systemic fungal infection caused by Paracoccidioides brasiliensis. As infectious diseases can cause DNA damage, the authors aimed at analyzing DNA breakage in peripheral blood cells of patients with paracoccidioidomycosis by using the comet assay. The results suggested that paracoccidioidomycosis does not cause genotoxicity.

Pneumocystis jirovecii é conhecido por causar infecções específicas no aparelho respiratório de seus hospedeiros, principalmente em doentes imunocomprometidos, manifestando-se por uma pneumonia grave e por vezes fatal, normalmente designada por pneumonia por Pneumocystis. A caracterização da diversidade genética de P. jirovecii tem demonstrado que determinados polimorfismos de base única poderão ser reconhecidos como marcadores moleculares de eleição para o estudo da distribuição geográfica, vias de transmissão, resistência/susceptibilidade a fármacos, factores de virulência e genética populacional de subtipos genéticos. Este estudo teve como objectivo a caracterização de polimorfismos de P. jirovecii, através da metodologia PCR multiplex/Extensão de base única (do inglês single base extension), com a principal finalidade de constatar eventuais associações entre polimorfismos de base única, genótipos multilocus, e dados clínicos e demográficos da infecção. Sessenta e seis espécimes pulmonares, previamente considerados positivos para P. jirovecii, obtidos entre 2001 e 2012, a partir de doentes portugueses imunocomprometidos, foram seleccionados de forma aleatória para este estudo multilocus. PCR multiplex foi utilizada para a amplificação simultânea de três regiões genómicas: subunidade grande do rRNA mitocondrial, superóxido dismutase e dihidropteroato sintetase. Cinco polimorfismos de base única, previamente correlacionados com parâmetros da doença, foram genotipados por extensão de base única: mt85, SOD110, SOD215, DHPS165 e DHPS171. Um total de 330 polimorfismos de base única e 29 genótipos multilocus putativos de P. jirovecii foram identificados e caracterizados nos espécimes pulmonares analisados. Os padrões de distribuição dos polimorfismos foram analisados, sendo considerada a variação temporal e/ou geográfica das suas formas alélicas. Constatou-se grande diversidade genotípica entre os isolados de P. jirovecii que poderá ter influência a nível epidemiológico. Foram observadas associações estatísticas entre mt85/genótipos multilocus e parâmetros demográficos e clínicos. A correlação mais importante verificou-se entre mt85C e cargas parasitárias baixas a moderadas, enquanto mt85T foi associado com cargas parasitárias altas; MLG5, MLG9 e MLG13 foram associados com cargas parasitárias baixas, moderadas e altas, respectivamente. Tais associações demonstram que potenciais marcadores moleculares da infecção por P. jirovecii poderão existir e que polimorfismos/genótipos específicos poderão determinar perfis epidemiológicos da pneumonia por Pneumocystis. A análise genética cruzada permitiu verificar associações entre polimorfismos de base única. Os polimorfismos SOD110T e SOD215C, SOD110C e SOD215T, DHPS165A e DHPS171C, DHPS165G e DHPS171T foram associados estatisticamente. Os genótipos multilocus mais prevalentes foram considerados para o teste recombinatório d1. Dois genótipos multilocus (MLG7 e MLG9) foram observados com elevada frequência, e a análise genética indicou que estes se encontravam sobre-representados na população de P. jirovecii estudada. Estas evidências indicam que o fenómeno de desequilíbrio de ligação e a propagação clonal de subtipos genéticos é frequente, considerando que a espécie P. jirovecii poderá ser representada por uma população com estrutura epidémica. O presente trabalho confirmou a importância do estudo de polimorfismos em P. jirovecii, sugerindo que a caracterização multilocus poderá fornecer informação relevante para a compreensão dos padrões, causas e controlo da infecção, melhorando assim a investigação deste importante patogéneo.

Este livro, agora publicado pelo INIC, é o resultado de um trabalho de investigação realizado entre 1985 e 1987 para a obtenção do grau de Mestre em Estudos Literários Comparados pela Faculdade de Ciências Sociais e Humanas da Universidade Nova de Lisboa. A literatura fantástica é uma área da literatura mundial que recebeu nas últimas décadas um grande interesse por parte da comunidade científica internacional. A multiplicação das associações, reuniões e congressos, a par da crescente produção teórica e científica, dão prova da vitalidade desta área da crítica literária. Parte deste movimento foi precisamente despoletado pelo interesse provocado pela obra de 1. R. R. Tolkien e de outros autores como C. S. Lewis, Charles Williams, etc. Este fenómeno internacional não teve ainda reais desenvolvimentos em Portugal. Cinco anos volvidos sobre a realização deste trabalho verificamos que ele é um dos primeiros (senão o primeiro) editado entre nós que, de uma forma sistemática e científica, analisa a obra de Tolkien, abordando ainda, embora de forma breve, a problemática da ficção fantástica contemporânea.

NAV Portugal is the Air Navigation Service Provider in Portugal, providing air traffic control services in the airspace under the country’s responsibility. Recently, the company has been included in an initiative launched by the European Commission, called the Single European Sky. This aims for a unification of the European airspace, improving it in four main pillars: safety, capacity, environment, and cost-efficiency. To each of them, Key Performance Indicators need to be computed and monitored, all having pre-defined targets. The presented work project will be analyzing how NAV Portugal is doing in the pillar of capacity, proving suggestions if needed.

White Color tuning is an attractive feature that Organic Light Emitting Diodes (OLEDs) offer. Up until now, there hasn’t been any report that mix both color tuning abilities with device stability. In this work, White OLEDs (W-OLEDs) based on a single RGB blend composed of a blue emitting N,N′-Di(1-naphthyl)-N,N′-diphenyl-(1,1′-biphenyl)-4,4′-diamine (NPB) doped with a green emitting Coumarin-153 and a red emitting 4-(Dicyanomethylene)-2-methyl-6-(4-dimethylaminostyryl)-4H-pyran (DCM1) dyes were produced. The final device structure was ITO/Blend/Bathocuproine (BCP)/ Tris(8-hydroxyquinolinato)aluminium (Alq3)/Al with an emission area of 0.25 cm2. The effects of the changing in DCM1’s concentration (from 0.5% to 1% wt.) allowed a tuning in the final white color resulting in devices capable of emitting a wide range of tunes – from cool to warm – while also keeping a low device complexity and a high stabilitty. Moreover, an explanation on the optoelectrical behavior of the device is presented. The best electroluminescense (EL) points toward 160 cd/m2 of brightness and 1.1 cd/A of efficiency, both prompted to being enhanced. An Impedance Spectroscopy (IS) analysis allowed to study both the effects of BCP as a Hole Blocking Layer and as an aging probe of the device. Finally, as a proof of concept, the emission was increased 9 and 64 times proving this structure can be effectively applied for general lighting.

Abstract:INTRODUCTION:The therapeutic scheme of triclabendazole (TCBZ), the recommended anthelmintic against Fasciola hepatica , involves 10mg/kg of body weight administered in a single dose; however, clinical trials in children are scarce. We evaluated the efficacy and tolerability of 2 schemes of TCBZ.METHODS: Eighty-four Peruvian children with F. hepatica eggs in their stools were allocated into 2 groups: 44 received 2 dosages of 7.5mg/kg each with a 12-h interval (Group I), and 40 received a single 10-mg/kg dose (Group II). Evaluation of efficacy was based on the presence of eggs in stools, and tolerability was based on the presence of symptoms and signs post-treatment.RESULTS: A parasitological cure was obtained in 100% of individuals from Group I and 95% of individuals from Group II. The most common adverse event was biliary colic.CONCLUSIONS: The tested scheme was efficacious and tolerable, and it might be an optimal scheme in the region. To the best of our knowledge, this represents the largest series of children treated with TCBZ in a non-hospital setting.

Abstract: INTRODUCTION : Molecular analyses are auxiliary tools for detecting Koch's bacilli in clinical specimens from patients with suspected tuberculosis (TB). However, there are still no efficient diagnostic tests that combine high sensitivity and specificity and yield rapid results in the detection of TB. This study evaluated single-tube nested polymerase chain reaction (STNPCR) as a molecular diagnostic test with low risk of cross contamination for detecting Mycobacterium tuberculosis in clinical samples. METHODS: Mycobacterium tuberculosis deoxyribonucleic acid (DNA) was detected in blood and urine samples by STNPCR followed by agarose gel electrophoresis. In this system, reaction tubes were not opened between the two stages of PCR (simple and nested). RESULTS: STNPCR demonstrated good accuracy in clinical samples with no cross contamination between microtubes. Sensitivity in blood and urine, analyzed in parallel, was 35%-62% for pulmonary and 41%-72% for extrapulmonary TB. The specificity of STNPCR was 100% in most analyses, depending on the type of clinical sample (blood or urine) and clinical form of disease (pulmonary or extrapulmonary). CONCLUSIONS: STNPCR was effective in detecting TB, especially the extrapulmonary form for which sensitivity was higher, and had the advantage of less invasive sample collection from patients for whom a spontaneous sputum sample was unavailable. With low risk of cross contamination, the STNPCR can be used as an adjunct to conventional methods for diagnosing TB.

To find sustainable solutions for the production of energy, it is necessary to create photovoltaic technologies that make every photon count. To pursue this necessity, in the present work photodetectors of zinc oxide embedded with nano-structured materials, that significantly raise the conversion of solar energy to electric energy, were developed. The novelty of this work is on the development of processing methodologies in which all steps are in solution: quantum dots synthesis, passivation of their surface and sol-gel deposition. The quantum dot solutions with different capping agents were characterized by UVvisible absorption spectroscopy, spectrofluorimetry, dynamic light scattering and transmission electron microscopy. The obtained quantum dots have dimensions between 2 and 3nm. These particles were suspended in zinc acetate solutions and used to produce doped zinc oxide films with embedded quantum dots, whose electric response was tested. The produced nano-structured zinc oxide materials have a superior performance than the bulk, in terms of the produced photo-current. This indicates that an intermediate band material should have been produced that acts as a photovoltaic medium for solar cells. The results are currently being compiled in a scientific article, that is being prepared for possible submission to Energy and Environmental Science or Nanoscale journals.
