This work consisted of determining the degree of humification of humic substances (HS) extracted from six different Amazonian soils collected from flooded and unflooded regions at different depths (0-10, 10-20, 20-40, and 60 cm). The humic substances were extracted according to procedures recommended by the International Humic Substances Society and characterized using elemental analysis, electron paramagnetic resonance (EPR), and fluorescence spectroscopy. The findings on semiquinone-type free radical concentrations in HS showed variations of 0.10-7.55x10(18) spins g(-1) of carbon (g C)(-1), indicating considerable differences between the humification levels of HS extracted from Amazonian soils. The results showed an average of 1.71 +/- 0.04 x 10(18) spins (g C)(-1), which is congruent with other data reported in the literature on Tropical soils. It was found that, on average, HS extracted from flooded soil contained higher semiquinone-type free radical concentrations than HS extracted from unflooded soils, indicating the influence of humidity in the humification process of organic matter. The humification process varies according to the profile, and the 10-20- and 0-10-cm profiles generally showed more humified HS. The degree of humification of the HS studied here displayed a similar behavior when exposed to fluorescence (excitation at 465 nm) and EPR (R=0.85). However, the low correlation between the C/H, C/O, and C/N atomic ratios and the semiquinone-type free radical concentration/fluorescence intensities indicated that data obtained by these techniques with regard to the degree of humification of HS may lead to different conclusions. Objectives: To investigate the presence of mutations in the pncA gene in 31 pyrazinamide-resistant Mycobacterium tuberculosis and 5 susceptible strains. MICs and pyrazinamidase (PZase) activity were also determined.Methods: All 36 M. tuberculosis clinical isolates were genotyped by mycobacterial interspersed repetitive units (MIRUs) and most were also typed by spoligotyping. The MIC value necessary to inhibit 99% of the resistant mycobacterial isolates was determined by microplate Alamar Blue assay (MABA) and by Lowenstein-Jensen assay (LJA). The PZase activity was measured by pyrazinamide deamination to pyrazinoic acid and ammonia, and the entire pncA sequence including the 410 by upstream from the start codon was determined by DNA sequencing of purified PCR products.Results: of the 31 isolates resistant to pyrazinamide, 26 (83.9%) showed at least one mutation in the pncA gene or in its putative regulatory region: Among the 22 different mutations detected in the pncA gene and in its regulatory region, 9 (40.9%) mutations (consisting of six substitutions, two insertions and one deletion) have not been described in previous studies. Three pyrazinamide-resistant isolates, confirmed by MIC varying from 800 to 1600 mg/L, carried the wild-type pncA sequence and retained PZase activity.Conclusions: These results contribute to the knowledge of the molecular mechanism of pyrazinamide resistance in Brazil and also expand the profile of pncA mutations worldwide. The MABA was successfully used to determine the MICs of pyrazinamide.

Extracts and pure amenthoflavone isolated from Byrsonima crassa (Malpighiaceae), a shrub growing in the semi-arid region of Brazil Cerrado, were evaluated in vitro, at different doses, for their effects on tomato seed germination and subsequent growth of seedlings. A hydromethanolic extract showed general stimulatory effects. The EtOAc extract stimulated root elongation and root weight of tomato; shoot elongation was inhibited, while shoot weight was not altered. The pure amenthoflavone isolated from the plant, stimulated shoot elongation at concentrations ranging between 10(-4) M and 10(-6) M.

We present evidence for Plasmodium vivax infection among Duffy blood group-negative inhabitants of Brazil. The P. vivax identification was determined by both genotypic and non-genotypic screening tests. The Duffy blood group was genotyped by PCR/RFLP and phenotyped using a microtyping kit. We detected two homozygous FY*B-33 carriers infected by P vivax, whose circumsporozoite protein genotypes were VK210 and/or P. vivax-like. Additional efforts are necessary in order to clarify the evidence that P. vivax is being transmitted among Duffy blood group-negative patients from the Brazilian Amazon region. The objective of the present study was to characterize the fecundity of Callinectes ornatus from the northern coast of São Paulo, Brazil. To study the fecundity and its relationships to size, brood weight, and egg size, the ovigerous females were collected at 2-month intervals for two consecutive years (January, 1991 to November, 1992) in the Ubatuba region using a fishing boat equipped with an otter-trawl type of net. After collecting, the animals were screened, bagged, labeled, and stored frozen. Only data from 38 females carrying early stage eggs were considered. In the laboratory, the specimens were thawed at room temperature, the pleopod structure with the egg mass was removed and the eggs were carefully removed from the pleopods. The samples were then fixed in 10% formalin and stored in 70% ethanol, until the time for processing. Processing followed the method of Hines (1982, 1988). Following the frequency determination of carapace width and fecundity, mean egg number, mean volume and mean dry weight of brooded egg mass were determined for each class obtained. Data were analyzed by regressions of log-log transformations for allometric plots (Y = aX(b)) of reproductive variables versus carapace width. Carapace width was one of the main factors for the determination of fecundity, which ranged 171 570 +/- 94 634 eggs, with females of the same size class presenting a wide amplitude of variation. This supports the hypothesis that portunid females present staggered spawning, possibly presenting more than one period of reproduction within one year. The present species showed lower fecundity than the remaining portunids studied, but they did show a rapid and efficient embryonic development, presumably capable of assuring reproductive success.

This work aimed to evaluate the gastrointestinal helminthfauna composition of six-banded armadillos from the Brazilian semi-arid region. Gastrointestinal contents of six road-killed adult animals from Patos County, Paraiba State, were analyzed. Six species of nematodes, comprising five genera and four families, were recovered from the analyzed animals. New morphological data on Trichohelix tuberculata is given, along with a new taxonomical proposal for Hadrostrongylus ransomi (Travassos, 1935) n. comb. This is the first record for parasitic helminths in this host from the Brazilian semi-arid.

Three-hundred faecal swabs were obtained from pigs with diarrhoea in farms located in different areas of the Ribeirao Preto region in the State of Sao Paulo. One-hundred Escherichia coli strains were isolated and tested for production of thermolabile (TL) and thermostable (STRa and STb) enterotoxins, and for the presence of colonization factors F4, F5 and F6. The strains were also tested for sensitivity to 14 antibiotics and chemotherapeutic agents. Twenty-four Escherichia coli strains produced enterotoxin STb, 5 produced LT and 3 produced STa. In the mannose-resistant haemagglutination reaction, one strain reacted positively with sheep, chicken, horse and human red blood cells and another reacted positively with guinea pig, sheep, chicken, horse and human red cells. However, both strains were negative for colonization factors F4, F5 and F6 when submitted to the slide agglutination test. All Escherichia coli strains were resistant to at least one antibiotic, the highest percentages being obtained for resistance to penicillin, tetracycline and cephalotin. In addition to the importance of the virulence factors normally encountered in enterotoxigenic Escherichia coli strains from pigs, the present results show the possible existence of new colonization factors other than F4, F5 and F6 participating in E. coli-induced pigs colibacillosis in the Ribeirao Preto region.

A cave bat community from a semiarid region in eastern Brazil was studied during the dry season. A high diversity was observed, with thirteen species recorded. Except for Micronycteris minuta and Lionycteris spurrelli, these bats were also found in other Brazilian karstic areas. The commonest species were the hematophague Desmodus rotundus, the omnivore Carollia perspicillata, and the insectivore Natalus stramineus. The bats formed itinerant colonies, moving frequently inside the cave. L. spurrelli was previously known from Amazonia, usually associated with evergreen forests. The present record extends its range 1600 km to the south. A disjunct distribution explains some morphological differentiation between Amazonia and Olhos d'Agua cave populations. The latter may be a relict of former continuous forest vegetation, which has been fragmented after the climatic optimum. Dispersion through the Cerrado forest gallery is also possible.
