In their survey/study - Adult Alternatives for Social Drinking: A Direction - by John Dienhart and Sandra Strick, Assistant Professors, Department of Restaurant, Hotel and Institutional Management, Purdue University, Dienhart and Strick begin with: “Changes in consumer habits have brought about a change in the business of selling alcoholic drinks and have impacted upon hotel food and beverage operations. The authors surveyed a sample of hotel corporate food and beverage directors to ascertain how they are handling this challenge.” Dienhart and Strick declare that the alcoholic beverage market, sale and consumption thereof, has taken a bit of a hit in contemporary society. “Even to the casual observer, it's obvious that the bar and beverage industry has undergone a great deal of change in the past few years,” say the authors. “Observations include a change in the types of drinks people are ordering, as well as a decrease in the number of drinks being sold,” they qualify. Dienhart and Strick allude to an increase in the federal excise tax, attacks from alcohol awareness groups, the diminished capacity of bars and restaurants to offer happy hours, increased liability insurance premiums as well as third-party liability issues, and people’s awareness of their own mortality as some of the reasons for the change. To quantify some empirical data on beverage consumption the Restaurant, Hotel, and Institutional Management Department of Purdue University conducted a study “… to determine if observed trends could be documented with hard data.” In regards to the subject, the study asks and answers a lot of interesting questions with the results presented to concerned followers via percentages. Typical of the results are: “When asked whether the corporation experienced a change in alcoholic sales in the past year, 67 percent reported a decrease in the amount of alcohol sold.” “Sixty-two percent of the respondents reported an increase in non-alcoholic sales over the past year. The average size of the increase was 8 percent. What Dienhart and Strick observe is that the decrease in alcoholic beverage consumption has resulted in a net increase for non-alcoholic beverage consumption. What are termed specialty drinks are gaining a foothold in the market, say the authors. “These include traditional cocktails made with alcohol-free products, as well as creative new juice based drinks, cream based drinks, carbonated beverages, and heated drinks,” say Dienhart and Strick by way of citation . Another result of the non-alcoholic consumption trend is the emergence of some novel marketing approaches by beer, wine, and spirits producers, including price increases on their alcohol based beverages as well as the introduction of faux alcoholic drinks like non-alcoholic beer and wine. Who or what is the big winner in all of this? That distinction might go to bottled water!

Actually, Brazil is one of the larger fruit producer worldwide, with most of its production being consumed in nature way or either as juice or pulp. It is important to highlig ht in the fruit productive chain there are a lot lose due mainly to climate reasons, as well as storage, transportation, season, market, etc. It is known that in the pulp and fruit processing industy a yield of 50% (in mass) is usually obtained, with the other part discarded as waste. However, since most this waste has a high nutrient content it can be used to generate added - value products. In this case, drying plays an important role as an alternative process in order to improve these wastes generated by the fruit industry. However, despite the advantage of using this technique in order to improve such wastes, issues as a higher power demand as well as the thermal efficiency limitation should be addressed. Therefore, the control of the main variables in t his drying process is quite important in order to obtain operational conditions to produce a final product with the target specification as well as with a lower power cost. M athematical models can be applied to this process as a tool in order to optimize t he best conditions. The main aim of this work was to evaluate the drying behaviour of a guava industrial pulp waste using a batch system with a convective - tray dryer both experimentally and using mathematical modeling. In the experimental study , the dryin g carried out using a group of trays as well as the power consume were assayed as response to the effects of operational conditions (temperature, drying air flow rate and solid mass). Obtained results allowed observing the most significant variables in the process. On the other hand, the phenomenological mathematical model was validated and allowed to follow the moisture profile as well as the temperature in the solid and gas phases in every tray. Simulation results showed the most favorable procedure to o btain the minimum processing time as well as the lower power demand.

In order to evaluate the effect of levels of dehydrated cane juice sugar (DCJS) (0.0, 1.5, 3.0 and 4.5%) in the diet of European quail (Coturnix coturnix coturnix) on performance on performance, carcass characteristics and economical at 22 days of age indices, 192 quails cutting, were distributed in a completely randomized design with four treatments and six replicates of eight birds each There was no significant difference to the performance of quail in any of the periods. Quadratic effects of levels of DCJS on carcass weight (y = 173.71 + 4.2767x – 1.2644x², R² = 0.99), thigh-thigh more about (y = 36,055 + 1,1263x – 0,2256x², R² = 0.91) and abdominal fat (y = 3,3295 - + 0.8903x 0,19x2, R ² = 0,97) where the optimum levels were estimated 1,69; 2.50 and 2.34%, respectively. There was a linear effect descending of DCJS levels on weight breast with skin (y = 66.267 – 0.5653x, R² = 0.83) and without skin (y = 60.286 – 0.7193x, R² = 0.58). In economic analysis, one can observe higher profit to the producer with the use of conventional feed. However, between the levels of inclusion of sugar cane juice is observed that the level of 1.5% DCJS obtained the best results in economic analysis, obtaining only a difference of relative gross margin of 0.47% compared to conventional. It is recommended 1.69 and 2.50% DCJS for higher carcass, thigh + drumstick weight and lower percentage of abdominal fat quails, respectively.

Camu-camu (Myrciaria dubia H.B.K. (McVaugh)) is a native Amazon fruit, recognized worldwide as one of the main natural sources of ascorbic acid. Due to its great acidity, this fruit is generally consumed after processing into juice or as ingredient in food preparations. As a co-product of the camu-camu processing, a significant amount of agroindustrial residue is generated. Despite the studies showing the bioactive value and biological potential of the fruit, few studies have approached the possible processing techniques, transformation and preservation of camu-camu fruits and its agroindustrial pomace. Therefore, the present work has the objective of evaluating two different drying processes applied to camu-camu pomace (peel and seeds with residual pulp), freeze drying and hot air drying, in order to obtain a functional fruit product. This thesis was divided into three stages: the first one shows the studies related to the freeze drying and hot air drying, where we demonstrated the impact of the selected drying techniques on the bioactive components of camu-camu, taking the fresh pomace as the control group. Among the investigated conditions, the groups obtained at 50ºC and 4 m/s (SC50) and 80ºC and 6 m/s (SC80) were selected as for further studies, based on their ascorbic acid final content and Folin-Ciocalteau reducing capacity. In addition to SC50 and SC80, the fresh pomace (RF) and freeze dried (RL) samples were also evaluated in these further stages of the research. Overall, the results show higher bioactive concentration in the RF samples, followed by RL, SC50 and SC80. On the second step of the research, the antioxidant, antimicrobial and antienzymatic activities were evaluated and the same tendency was observed. It was also reported, for the first time in the literature, the presence of syringic acid in dried camu-camu pomace. In the third and final stage of the research, it was investigated the effect of dried camu-camu on aging and neuroprotective disorders, using the in vivo model C.elegans. It was observed that camu-camu extracts were able to modulate important signaling genes relevant to thermal and oxidative stresses (p < 0.05). The polar acid, polar basic and polar neutral fractions obtained from the low molecular extracts of SC50 were able to extend the lifespan of wild type N2 C. elegans in 20% and 13% (p < 0.001). Results also showed that the paralysis induced by the β1-42 amyloid was significantly (p < 0.0001) retarded in CL4176 worms. Similarly, the camu-camu extracts attenuated the dopaminergic induction associated to Parkinson’s disease. Finally, a global analysis of the data presented here reveal that the camu-camu pomace, a co-product obtained from the industrial processing of a native Brazilian fruit, is a relevant natural source of health relevant compounds. This thesis, shows for the first time, the multifunctionality of camu-camu pomace, a natural resource still underexploited for scientific, commercial and technological purposes.

Much of what is known about word recognition in toddlers comes from eyetracking studies. Here we show that the speed and facility with which children recognize words, as revealed in such studies, cannot be attributed to a task-specific, closed-set strategy; rather, children's gaze to referents of spoken nouns reflects successful search of the lexicon. Toddlers' spoken word comprehension was examined in the context of pictures that had two possible names (such as a cup of juice which could be called "cup" or "juice") and pictures that had only one likely name for toddlers (such as "apple"), using a visual world eye-tracking task and a picture-labeling task (n = 77, mean age, 21 months). Toddlers were just as fast and accurate in fixating named pictures with two likely names as pictures with one. If toddlers do name pictures to themselves, the name provides no apparent benefit in word recognition, because there is no cost to understanding an alternative lexical construal of the picture. In toddlers, as in adults, spoken words rapidly evoke their referents.

The abuse of antibiotics and the emergence of multi-drug resistant bacterial strains have created the need to explore alternative methods of controlling microbial pathogens. The bacteriocin family of antimicrobial peptides has been proposed as one such alternative to classic antibiotics. Nisin A belongs to the subgroup of bacteriocins called the lantibiotics, which contain several unusual amino acids as a consequence of enzyme-mediated post-translational modifications. As nisin is produced by generally regarded as safe (GRAS) microorganisms, it could potentially be applied in a clinical setting. However, as lantibiotics are naturally produced in such small quantities, this can hinder their industrial potential. In order to overcome this, several approaches can be utilised. For example, given the gene encoded nature of lantibiotics, genetic engineering approaches can be implemented in order to yield variants with enhanced properties. Here, the use of mutagenesis-based strategies was employed to obtain a derivative of nisin with enhanced bioactivity in vitro. Investigations with purified peptide highlighted the enhanced specific activity of this variant, nisin M21V, against food-borne Listeria monocytogenes strains. Furthermore, this specific enhanced bioactivity was evident in a mouse model of listeriosis. Reductions in bioluminescence and microbial counts in organs from infected mice were observed following treatment with nisin M21V compared to that of wild-type nisin A. Peptide bioengineering approaches were also implemented to obtain additional novel derivatives of nisin. The generation of “S5X” and “S33X” banks (representing a change of natural serines at positions 5 and 33 to all possible alternative residues) by a combination of site-saturation and site-directed mutagenesis led to the identification of several derivatives exhibiting improved stability. This allowed the rational design of variants with enhanced stability compared to that of wild type nisin. Another means of tackling issues associated with lantibiotic yield is to combine lantibiotics with other antimicrobials. This could circumvent the need for enhanced production while also reducing concentrations of the peptide antimicrobials. We observed that combinations of nisin variants and low levels of plant essential oils (thymol, carvacrol, trans-cinnamaldehyde) significantly controlled Gram negative foodborne pathogens in in vitro assays compared to nisin A-essential oil combinations. This enhanced control was also evident in model food systems. Nisin variants used in conjunction with carvacrol significantly reduced numbers of E. coli O157:H7 in apple juice while a commercial nisin preparation used in combination with citric acid significantly controlled C. sakazakii in infant milk formula. It is noteworthy that while nisin is generally associated with Gram positive targets, upon combination with plant essential oils the spectrum of inhibition was broadened to Gram negative targets.

Background: Various factors have been investigated to account for the higher premature death rates in Scotland compared to England. Higher levels of deprivation in Scotland provide a partial explanation for these differences but recent work comparing areas of the UK with similar deprivation profiles and low life expectancy has shown that this is not the only reason. One hypothesis yet to be tested adequately is differences in diet and nutrition. Objective: To conduct a comparative analysis of dietary intake between Scotland and England using pooled food purchase data from the Living Costs and Food Survey (LCFS) from 2001 to 2012 and to assess differences in equivalised income quintiles (controlling for survey year, age of household reference person, and age household reference person left full-time education). Results: Lower intakes of fruit and vegetables, oil rich fish, fibre, vitamin A, folate, vitamin C and vitamin D and higher intakes of red and processed meat, whole milk, butter, savoury snacks, confectionary, soft drinks, saturated fat and NMES (added sugar and sugar in fruit juice), sodium and alcohol were found for Scotland compared to England. Differences between Scotland and England were higher for those on lower incomes for dietary components known to be related to health outcomes. For example fruit consumption was 14g/day lower for the lowest income quintile compared to 4 g/day lower in the highest quintile for Scotland versus England. Conclusions: A poorer diet in Scotland compared to England, particularly among disadvantaged groups, is likely to be one of the reasons for excess mortality. The current evidence on the continued poor diet in Scotland, particularly in disadvantaged groups, should not be ignored. Identifying effective, culturally appropriate approaches to improve diet across the population and notably in the most deprived areas needs further investment. Funded by NHS Health Scotland. Brazilian sweet sorghum is used to generate ethanol and the bagasse is burned in industrial boilers or deposited on soil polluting the environment. This study evaluated the performance of sorghum plants and its bagasse silage nutritional value aiming to use it in the ruminant nutrition. Experiments were set up on the UTFPR campus at Dois Vizinhos-PR. The first trial was established on October 2nd, 2012 using the genotypes ADV 2010, Hunnigreen, Sugargraze, Volumax, BR 505, 503, 501 and the second trial at 2013 on November 27th assessing the materials ADV 2010, Sugargraze, Hunnigreen, EX 5110, BR 506, 508, 509 and 511. Experimental was laid out as a randomized block design with three replications. Results were analyzed through ANOVA comparing the averages by Duncan test at 5% error probability. As field variables were evaluated: plant height (Pl hei), green mass production (GM Prod), percentage of leaves, stems and panicles in relation to the plant, stems production without straw (Prod stems with straw), whole stems production (Who stems Prod), stem diameter (Stem diam), juice production (Juice prod) and Brix degree (oBrix). After juice plant extraction, forage bagasse was crushed and packed in silos for 60 days and green matter yield was estimated (GM). In bagasse silage gauged to buffer capacity (BC), dry matter (DM), mineral matter (MM), crude protein (CP), ether extract (EE), total carbohydrates (TC), non-fibrous carbohydrates (NFCH), neutral detergent fiber (NDF), acid detergent fiber (ADF), lignin (LIG ) and digestibility "in vitro" (DIGIV). There was no significant difference (P> 0.05) between genotypes regarding to green matter production and on average, the second crop yield was lower and reflected in the reduction of bagasse production. Bagasse silage DM was of 32.3% and 33.1%; NDF 73% and 65.8%, crude protein 3.8% and 5.9; pH 3.7 and 3.7; TC and 9.8 and 10.7. mg MS-1; the amount of NFCH was 11.1 and 13.5%; DIG of DM 36.9 and 62.4% for the respectively to the bagasse produced from materials grown in the 2012/2013 and 2013/2014 seasons. Hybrid genotypes had a better agronomic performance while the varieties were more efficient in bromatological indexes. And despite the high percentage of NDF and the low protein level, it is possible to feed ruminants with this coproduct.

A ocratoxina A é um composto formado a partir do metabolismo secundário de fungos dos gêneros Aspergillus e Penicillium. Uma vez que a presença dessa micotoxina nos alimentos causa sérios danos à saúde humana e animal, surge o interesse pelo desenvolvimento de métodos que visem a redução dos seus níveis em diferentes matrizes. Diversos processos de descontaminação têm sido propostos, sendo que os métodos de redução biológica tem recebido destaque. Esses métodos consistem na aplicação de micro-organismos ou de suas enzimas, o que gera a biotransformação ou degradação da toxina produzindo metabólitos com menor ou nenhuma toxicidade. Diante disso, o objetivo geral do trabalho foi avaliar o efeito da peroxidase na redução dos níveis de ocratoxina A. As enzimas peroxidases testadas foram a comercial e a obtida do farelo de arroz. Para a extração enzimática foram utilizadas as frações granulométricas do farelo de arroz de 48 a 100 mesh, sendo estas frações caracterizadas quimicamente. A peroxidase foi extraída do farelo de arroz em tampão 10 mM pH 5,0 e purificada por partição trifásica, obtendo 77,1% de recuperação e 9,2 para o fator de purificação. O método utilizado para a extração da ocratoxina A do sistema aquoso foi por partição líquido-líquido utilizando como solvente o clorofórmio, sendo esse método validado segundo os parâmetros de linearidade (0,1 a 20 ng mL-1), coeficientes de correlação (0,9997) e de determinação (0,9994), e limites de detecção (0,02) e quantificação (0,03). A afinidade entre as peroxidases e a ocratoxina A foi verificada segundo os parâmetros de KM e Vmáx, resultando em 0,00027 mM e 0,000015 mM min-1, respectivamente, para a peroxidase comercial, e 0,0065 mM e 0,000031 mM min-1 para a obtida do farelo de arroz. Com relação aos percentuais de redução de ocratoxina A, foram avaliadas 3 proporções enzima:substrato (1:10, 1:5 e 8:1 para a comercial e 1:10, 1:5 e a com atividade de 0,063 U mL-1 para a do farelo), sendo que as proporções que forneceram maior redução foi a de 8:1 para a enzima comercial (0,063 U mL-1) e a correspondente a 0,063 U mL-1 para a enzima obtida do farelo. Os percentuais de redução de ocratoxina A foram de 59% para a peroxidase comercial em 300 min e 41% para a peroxidase do farelo de arroz em 1440 min. O efeito de adsorção da ocratoxina A pela enzima peroxidase foi descartado uma vez que foi realizada a sua hidrólise com a enzima pepsina e verificado um percentual de 2,7% de adsorção, demonstrando que a redução foi por ação enzimática. A enzima obtida de farelo de arroz com atividade de 0,063 U mL-1 foi aplicada em suco de uva tinto e branco. Observou-se que para o primeiro não houve redução significativa, enquanto que para o segundo a redução foi de 17%. Neste trabalho, então, foi possível verificar a capacidade de redução dos níveis da ocratoxina A pela enzima peroxidase, tanto em sistema aquoso como no suco de uva integral branco.

The routine analysis for quantization of organic acids and sugars are generally slow methods that involve the use and preparation of several reagents, require trained professional, the availability of special equipment and is expensive. In this context, it has been increasing investment in research whose purpose is the development of substitutive methods to reference, which are faster, cheap and simple, and infrared spectroscopy have been highlighted in this regard. The present study developed multivariate calibration models for the simultaneous and quantitative determination of ascorbic acid, citric, malic and tartaric and sugars sucrose, glucose and fructose, and soluble solids in juices and fruit nectars and classification models for ACP. We used methods of spectroscopy in the near infrared (Near Infrared, NIR) in association with the method regression of partial least squares (PLS). Were used 42 samples between juices and fruit nectars commercially available in local shops. For the construction of the models were performed with reference analysis using high-performance liquid chromatography (HPLC) and refractometry for the analysis of soluble solids. Subsequently, the acquisition of the spectra was done in triplicate, in the spectral range 12500 to 4000 cm-1. The best models were applied to the quantification of analytes in study on natural juices and juice samples produced in the Paraná Southwest Region. The juices used in the application of the models also underwent physical and chemical analysis. Validation of chromatographic methodology has shown satisfactory results, since the external calibration curve obtained R-square value (R2) above 0.98 and coefficient of variation (%CV) for intermediate precision and repeatability below 8.83%. Through the Principal Component Analysis (PCA) was possible to separate samples of juices into two major groups, grape and apple and tangerine and orange, while for nectars groups separated guava and grape, and pineapple and apple. Different validation methods, and pre-processes that were used separately and in combination, were obtained with multivariate calibration models with average forecast square error (RMSEP) and cross validation (RMSECV) errors below 1.33 and 1.53 g.100 mL-1, respectively and R2 above 0.771, except for malic acid. The physicochemical analysis enabled the characterization of drinks, including the pH working range (variation of 2.83 to 5.79) and acidity within the parameters Regulation for each flavor. Regression models have demonstrated the possibility of determining both ascorbic acids, citric, malic and tartaric with successfully, besides sucrose, glucose and fructose by means of only a spectrum, suggesting that the models are economically viable for quality control and product standardization in the fruit juice and nectars processing industry.

A concentração de seis elementos: Cd, Cr, Pb, Ni, Zn e Fe foi medida em sessenta e sete amostras de sumos de fruta 100 %, duas amostras de refrigerantes, dez amostras de concentrados de sumos e sete amostras de águas de diluição utilizadas no processamento dos sumos. As amostras de sumos representam numa prespectiva bastante abrangente o mercado Português de sumos de fruta 100 %. Os refrigerantes concentrados e águas de diluição foram cedidos por duas empresas fabricantes de sumos Portuguesas. As concentrações elementares foram medidas pelas técnicas de FAAS e GFAAS e foi medido também o grau Brix dos sumos. Os factores: fruta, percentagem de fruta, origem, agricultura, tratamento, embalagem, conservação e processo foram obtidos por informação do fabricante nos rótulos dos produtos e por contacto directo. Caracterizou-se o mercado em termos da concentração desses elementos e caracterizou-se a sua diluição comparando-a com valores de referência do mercado Europeu. Mediu-se o grau de associação entre os diversos parâmetros e a concentração final elementar dos sumos e utilizou-se a análise de agrupamentos, a análise de correspondência múltipla e a análise factorial para reestruturar a matriz de dados. Este estudo envolve o controlo e a optimização das condições de culturas dos microrganismos: Saccharomyces cerevisiae CCMI 396, S. cerevisiae v. lab., Aspergillus oryzae CCMI 125, Aspergillus japonicus CCMI 443, Fusarium oxysporum CCMI 866, Aspergillus niger CCMI 296 com vista à produção de oligossacáridos. Determinaram-se os parâmetros característicos das culturas de duas diferentes estirpes de Saccharomyces com diferentes fontes de carbono e em diferentes condições ambientais. O perfil de crescimento da S. cerevisiae CCMI 396 foi semelhante nos diferentes meios de cultura estudados, sendo a velocidade específica de crescimento mais elevada no meio com glucose a pH 5 e a 30°C (0,36h-1). A S. cerevisiae v. lab. Teve velocidade específica de crescimento idêntica nas mesmas condições da outra estirpe, no entanto, o perfil de crescimento foi diferente nos outros meios de cultura. Estudou-se o efeito da adição de sumo de laranja ou de tomate ao meio de cultura com sacarose e avaliou-se a evolução glucídica no meio de cultura durante o ensaio por HPLC com detector RI. Determinou-se a frutosiltransferase no sobrenadante e na fracção intracelular e determinou-se a evolução dos oligossacáridos. Numa segunda parte deste trabalho efectuaram-se culturas dos quatro fungos filamentosos com vista a avaliar a capacidade de produção, nomeadamente, de fruto­oligassacáridos. Os resultados mostraram que a espécie Aspergillus japonicus CCMI 443 originou, nas mesmas condições de cultura, valores superiores, sendo a percentagem de produção FOStotais/GluCtotais de 61% para as enzimas intracelulares e 40% para as enzimas no sobrenadante. ABSTRACT; This study involves control and optimization of the cultures of microorganisms: Saccharomyces cerevisiae CCMI 396, S. cerevisiae v. lab., Aspergillus oryzae CCMI 125, Aspergillus japonicus CCMI 443, Fusarium oxysporum CCMI 866, Aspergillus níger CCMI 296 for oligosaccharides production. Were determined the parameters characteristic of the cultures of two different strains of Saccharomyces with different sources of carbon and in different environmental conditions. The growth profile of S. cerevisiae CCMI 396 was similar in different cultures media, but the highest specific growth was obtained in a medium with glucose, pH 5, at 30°C (0.36h-1). S. cerevisiae v. lab. had similar growth profile in a medium with glucose but with others culture media was different. We studied the effect of adding orange juice or tomato to the culture medium with sucrose and evaluated the evolution glucidic in the culture medium during the test by HPLC with RI detector. Fructosyltransferase was determined in the extracellular and the intracellular fractions and determined the evolution of oligosaccharides. ln the second part of this work were carried out cultures of four filamentous fungi in order to assess production capacity, in particular, fructoligosaccharides. The results showed that the specie Aspergillus japonicus CCMI 443 originated in the same culture conditions, higher values and the percentage of production FOStotal/Guctotal of 61% for intracellular enzymes and 40% for extracellular enzymes.
