Studies in regions of the nuclear chart in which the model predictions of properties of nuclei fail can bring a better understanding of the strong interaction in the nuclear medium. To such regions belongs the so called "island of inversion" centered around Ne, Na and Mg isotopes with 20 neutrons in which unexpected ground-state spins, large deformations and dense low-energy spectra appear. This is a strong argument that the magic N = 20 is not a closed shell in this area. In this thesis investigations of isotope shifts of stable 24,25,26Mg, as well as spins and magnetic moments of short-lived 29,31Mg are presented. The successful studies were performed at the ISOLDE facility at CERN using collinear laser and beta-NMR spectroscopy techniques. The isotopes were investigated as single-charged ions in the 280-nm transition from the atomic ground state 2S1/2 to one of the two lowest excited states 2P1/2,3/2 using continuous wave laser beams. The isotope-shift measurements with fluorescence detection for the three stable isotopes show that it is feasible to perform the same studies on radioactive Mg isotopes up to the "island of inversion". This will allow to determine differences in the mean charge square radii and interpret them in terms of deformation. The high detection efficiency for beta particles and optical pumping close to saturation allowed to obtain very good beta-asymmetry signals for 29Mg and 31Mg with half-lives around 1 s and production yields about 10^5 ions/s. For this purpose the ions were implanted into a host crystal lattice. Such detection of the atomic resonances revealed their hyperfine structure, which gives the sign and a first estimate of the value of the magnetic moment. The nuclear magnetic resonance gave also their g-factors with the relative uncertainty smaller than 0.2 %. By combining the two techniques also the nuclear spin of both isotopes could be unambiguously determined. The measured spins and g-factors show that 29Mg with 17 neutrons lies outside the "island of inversion". On the other hand, 31Mg with 19 neutrons has an unexpected ground-state spin which can be explained only by promoting at least two neutrons across the N = 20 shell gap. This places the above nucleus inside the "island". However, modern shell-model approaches cannot predict this level as the ground state but only as one of the low-lying states, even though they reproduce very well the experimental g-factor. This indicates that modifications to the available interactions are required. Future measurements include isotope shift measurements on radioactive Mg isotopes and beta-NMR studies on 33Mg.

The new family of the anion receptors based on oligoureas with varied flexibility was developed and studied. The preparation of the urea chains containing two different units in various sequences was elaborated. The complete sets of four cyclic trimers and six tetramers based on the two units were prepared. Their conformational and complexation properties were studied with NMR spectroscopy and X-ray structure determinations, their behaviour towards various anions was evaluated and compared. The synthesis and the same studies were performed also with four different cyclic hexamers. During these studies the remarkable templation by two halide anions was observed.

Tethered bilayer lipid membranes (tBLMs) are a promising model system for the natural cell membrane. They consist of a lipid bilayer that is covalently coupled to a solid support via a spacer group. In this study, we developed a suitable approach to increase the submembrane space in tBLMs. The challenge is to create a membrane with a lower lipid density in order to increase the membrane fluidity, but to avoid defects that might appear due to an increase in the lateral space within the tethered monolayers. Therefore, various synthetic strategies and different monolayer preparation techniques were examined. Synthetical attempts to achieve a large ion reservoir were made in two directions: increasing the spacer length of the tether lipids and increasing the lateral distribution of the lipids in the monolayer. The first resulted in the synthesis of a small library of tether lipids (DPTT, DPHT and DPOT) characterized by 1H and 13C NMR, FD-MS, ATR, DSC and TGA. The synthetic strategy for their preparation includes synthesis of precursor with a double bond anchor that can be easily modified for different substrates (e.g. metal and metaloxide). Here, the double bond was modified into a thiol group suitable for gold surface. Another approach towards the preparation of homogeneous monolayers with decreased two-dimensional packing density was the synthesis of two novel anchor lipids: DPHDL and DDPTT. DPHDL is “self-diluted” tether lipid containing two lipoic anchor moieties. DDPTT has an extended lipophylic part that should lead to the preparation of diluted, leakage free proximal layers that will facilitate the completion of the bilayer. Our tool-box of tether lipids was completed with two fluorescent labeled lipid precursors with respectively one and two phytanyl chains in the hydrophobic region and a dansyl group as a fluorophore. The use of such fluorescently marked lipids is supposed to give additional information for the lipid distribution on the air-water interface. The Langmuir film balance was used to investigate the monolayer properties of four of the synthesized thiolated anchor lipids. The packing density and mixing behaviour were examined. The results have shown that mixing anchor with free lipids can homogeneously dilute the anchor lipid monolayers. Moreover, an increase in the hydrophylicity (PEG chain length) of the anchor lipids leads to a higher packing density. A decrease in the temperature results in a similar trend. However, increasing the number of phytanyl chains per lipid molecule is shown to decrease the packing density. LB-monolayers based on pure and mixed lipids in different ratio and transfer pressure were tested to form tBLMs with diluted inner layers. A combination of the LB-monolayer transfer with the solvent exchange method accomplished successfully the formation of tBLMs based on pure DPOT. Some preliminary investigations of the electrical sealing properties and protein incorporation of self-assembled DPOT and DDPTT-based tBLMs were conducted. The bilayer formation performed by solvent exchange resulted in membranes with high resistances and low capacitances. The appearance of space beneath the membrane is clearly visible in the impedance spectra expressed by a second RC element. The latter brings the conclusion that the longer spacer in DPOT and the bigger lateral space between the DDPTT molecules in the investigated systems essentially influence the electrical parameters of the membrane. Finally, we could show the functional incorporation of the small ion carrier valinomycin in both types of membranes.

Bei den schweren Tetrylenen [ERR’, E = Si—Pb; R, R’ = Organyl-, elementorganischer Rest] zeigt sich ein Lewis-amphoterer Charakter, der bei den silylsubstituierten besonders ausgeprägt ist, wodurch sie wertvolle Vorstufen für Folgereaktionen darstellen. Bis zur Fertigstellung der Arbeit waren von den schwersten Vertretern lediglich vier homoleptische silylsubstituierte Stannylene/Distannene und zwei Plumbylene bekannt und strukturell charakterisiert. Im Rahmen dieser Arbeit wurden nun zwei weitere homoleptische silylsubstituierte Stannylene und zwei Plumbylene charakterisiert [E{Si(SiMe3)2SiMe2t-Bu}2, E{Si(SiMe3)2Sii-Pr3}2, E =Sn, Pb]. Diese Stannylene und Sn[Si(SiMe3)3]2 reagieren mit AlH3NMe3 beziehungsweise LiAlH4 in einer α-Addition. Geht man zu dem sterisch weniger anspruchsvollen Silylsubstituenten Si(SiMe3)2Et über, so lassen sich ein neues Kaliumplumbanid und das bislang erste stabile einkernige Blei(III)-Radikal Pb[Si(SiMe3)2Et]3 synthetisieren. Analog lassen sich E[Si(SiMe3)3]3 [E = Sn, Pb] darstellen, von denen bislang nur die Zinnverbindung isoliert werden konnte. Pb[Si(SiMe3)3]2 reagiert mit Lewis-Basen [THF, NH3, KN(SiMe3)2, „KI"] zu den entsprechenden Addukten. Darüberhinaus wurden zwei Aluminiumsilylverbindungen [AlH2[Si(SiMe3)2SiMe2t-Bu]NMe3, LiAlH3[Si(SiMe3)3]] und die bislang ersten röntgenographisch charakterisierten tetrasilylsubstituierten Plumbane [Pb[Si(SiMe3)2Et]3R, R = SiMe3, Si(SiMe3)2Et] isoliert.

Imidazolium types of ionic liquids were immobilized by tethering it to acrylate backbone. These imidazolium salt containing acrylate monomers were polymerize at 70oC by free radical polymerization to give polymers poly(AcIm-n) with n being the side chain lenght. The chemical structure of the polymer electrolytes obtained by the described synthetic routes was investigated by NMR-spectroscopy. The polymers were doped with various amounts of H3PO4 and LiN(SO2CF3)2, to obtain poly(AcIm-n) x H3PO4 and poly(AcIm-2-Li) x LiN(SO2CF3)2. The TG curves show that the polymer electrolytes are thermally stable up to about 200◦C. DSC results indicates the softening effect of the length of the spacers (n) as well as phosphoric acid. The proton conductivity of the samples increase with x and reaches to 10-2 Scm-1 at 120oC for both poly(AcIm-2)2H3PO4 and poly(AcIm-6)2H3PO4. It was observed that the lithium ion conductivity of the poly(AcIm-2-Li) x LiN(SO2CF3)2 increases with blends (x) up to certain composition and then leveled off independently from blend content. The conductivity reaches to about 10-5 S cm-1 at 30oC and 10-3 at 100oC for poly(AcIm-2-Li) x LiN(SO2CF3)2 where x is 10. The phosphate and phosphoric acid functionality in the resulting polymers, poly(AcIm-n) x H3PO4, undergoes condensation leading to the formation of cross-linked materials at elevated temperature which may improve the mechanical properties to be used as membrane materials in fuel cells. High resolution nuclear magnetic resonance (NMR) spectroscopy was used to obtain information about hydrogen bonding in solids. The low Tg enhances molecular mobility and this leads to better resolved resonances in both the backbone region and side chain region. The mobile and immobile protons can be distinguished by comparing 1H MAS and 1H-DQF NMR spectra. The interaction of the protons which may contribute to the conductivity is observed from the 2D double quantum correlation (DQC) spectra.

Im Rahmen der vorliegenden Promotionsarbeit wird das wasserähnliche Lösungsmittel flüssiger Ammoniak verwendet, um die kinetisch instabilen Münzmetallsilyle in hohen Ausbeuten darzustellen. Als Ausgangsverbindungen dienten verschiedene basenfreie Kaliumsilanide, die sich in flüssigem Ammoniak gut lösen ohne nennenswert protolysiert zu werden. Gegenüber konventionellen Synthesen, die in organischen Lösungsmitteln durchgeführt werden, können statt Alkoholate die leicht zugänglichen Münzmetallhalogenide eingesetzt werden. Bei einer Stöchiometrie von 1:1 werden in Abhängigkeit der sterischen Anspruchs des Silylrestes, die cyclischen oder die Ammoniakate der dimeren Kupfer– bzw. Silbersilyle erhalten, während zwei Äquivalente basenfreien Kaliumsilanid und ein Äquivalent Münzmetallhalogenid zu den homologen Homocuprate, -argentate und -aurate führen. Zusätzlich wird die Auswirkung des unterschiedlich sterischen Anspruches der Silylliganden und des gebundenen Münzmetalls auf die Strukturparameter untersucht. Im zweiten Teil dieser Arbeit wird das dargestellte Aurat KAuHyp2 (Hyp = Si(SiMe3)3) mit Trimethylchlorsilan in verschiedenen organischen Lösungsmitteln umgesetzt. In Anhängigkeit von der Stöchiometrie, der Reaktionsdauer, der Temperatur und des verwendeten Lösungsmittels werden erstmalig eine Vielzahl neuer anionischer Goldsilylkomplexe erhalten, genannt sei die Verbindung [K2(Toluol)2][Au4Hyp4], welsches ein Au4-Tetraederskelett mit vier terminalen Hypersilylliganden besitzt. Vom besonderen Interesse ist die Reduktion des Golds. Bemerkenswert sind die zu beobachtenden Silizium-Silizium-Bindungsspaltungen bzw. -Bindungsmetathese bei Raumtemperatur, beispielsweise erkennbar an der Verbindung [K][Au5(Si(SiMe3)2)6]. Auf die Thematik dieser neuartigen strukturell interessanten anionischen Goldsilyle wird in dieser Arbeit näher eingegangen.

The quality of fish products is indispensably linked to the freshness of the raw material modulated by appropriate manipulation and storage conditions, specially the storage temperature after catch. The purpose of the research presented in this thesis, which was largely conducted in the context of a research project funded by Italian Ministry of Agricultural, Food and Forestry Policies (MIPAAF), concerned the evaluation of the freshness of farmed and wild fish species, in relation to different storage conditions, under ice (0°C) or at refrigeration temperature (4°C). Several specimens of different species, bogue (Boops boops), red mullet (Mullus barbatus), sea bream (Sparus aurata) and sea bass (Dicentrarchus labrax), during storage, under the different temperature conditions adopted, have been examined. The assessed control parameters were physical (texture, through the use of a dynamometer; visual quality using a computer vision system (CVS)), chemical (through footprint metabolomics 1H-NMR) and sensory (Quality Index Method (QIM). Microbiological determinations were also carried out on the species of hake (Merluccius merluccius). In general obtained results confirmed that the temperature of manipulation/conservation is a key factor in maintaining fish freshness. NMR spectroscopy showed to be able to quantify and evaluate the kinetics for unselected compounds during fish degradation, even a posteriori. This can be suitable for the development of new parameters related to quality and freshness. The development of physical methods, particularly the image analysis performed by computer vision system (CVS), for the evaluation of fish degradation, is very promising. Among CVS parameters, skin colour, presence and distribution of gill mucus, and eye shape modification evidenced a high sensibility for the estimation of fish quality loss, as a function of the adopted storage conditions. Particularly the eye concavity index detected on fish eye showed a high positive correlation with total QIM score.

In der vorliegenden Arbeit wurde die erste β-1,3-Glucanase aus Delftia beschrieben. Es konnte gezeigt werden, dass das Enzym unter anderem gegen das nur schwer zu hydrolysierende Exopolysaccharid aus Pediococcus parvulus wirkte. rnrnIm Einzelnen wurde zunächst das Exopolysaccharid aus Pediococcus parvulus B399 aus einem eigens zusammengestellten β-Glucan-Synthesemedium (Medium M) isoliert und gereinigt. Anschließend erfolgte eine umfassende Charakterisierung des Biopolymers. Hierzu gehörten neben der sauren Hydrolyse zur Bestimmung der Monomerzusammensetzung des Polymers, auch spektroskopische Methoden, darunter 1H und 13C-NMR. Mithilfe der NMR-Spektroskopie konnte die Struktur des Exopolysaccharids aus Pediococcus parvulus B399 bestimmt werden. Es handelte sich hierbei ebenfalls um ein β-1,3(1,2)-Glucan, wie es bereits für Pediococcus parvulus 2.6 beschrieben wurde. Darüber hinaus wurde erstmals ein ATR-FTIR-Spektrum für ein Exopolysaccharid aus Pediokokken gezeigt. Über GPC-Messungen konnte auch die molekulare Größe des β-1,3(1,2)-Glucans aus Pediococcus parvulus B399 bestimmt werden. Es wurde nachgewiesen, dass sich das Exopolysaccharid bei Anzucht in Medium M aus einer hochmolekularen Fraktion (5*106 g/mol) und vier niedermolekularen Fraktionen (347; 818; 10048 und 20836 g/mol) zusammensetzte. Neben der strukturellen Charakterisierung, wurde das Exopolysaccharid auch rheologisch untersucht. Dabei konnte festgestellt werden, dass es sich durch seine schwach gelbildenen Eigenschaften auch zum Einsatz in der Lebensmittelindustrie als Stabilisator, Fettersatzmittel oder ähnliches eignen würde. Die erwähnte gelbildende Netzwerkstruktur konnte für das Exopolysaccharid aus Pediococcus parvulus B399 auch erstmals im AFM bestätigt werden. rnEin weiterer Teil der Arbeit umfasste ein breites Screeningverfahren nach einem geeigneten Organismus, der das Exopolysaccharid aus Pediococcus parvulus B399 effektiv hydrolysieren sollte. Aus einer Anreicherungskultur des Termitendarms (Wenzel et al., 2002), konnte Delftia sp. MV01 isoliert werden. Dieser Organismus produzierte bei Wachstum in β glucanhaltigem Medium (Exopolysaccharid aus Pediococcus parvulus B399, sowie weitere kommerziell erhältliche β-1,3-Glucane) eine Glucanase, die in folgenden Schritten konventionell gereinigt und charakterisiert wurde.
From amphiphilic block copolymers to ferrocenyl-functionalized polymers for biosensoric applications

The present thesis can be divided in three main parts. In all parts new polymer architecturesrnwere synthesized and characterized concerning their special features.rnThe first part will emphasize the advantage of a polystyrene-block-(hyperbranchedrnpolyglycerol) copolymer in comparison to an analogue polystyrene-block-(linear polyglycerol)rncopolymer. Therefore a synthethic route to prepare linear block copolymersrnhas been developed. Two strategies were examined. One strategy was based on thernclassic, sequential anionic polymerization; the second strategy was based on arn“Click-Chemistry” coupling reaction. In a following step glycidol was hypergraftedrnfrom these block copolymers by applying a hypergrafting reaction with glycidol. Thernbehavior of the amphiphilic block copolymers synthesized was studied in differentrnsolvents. Furthermore the polarity of the solvent was changed to form the correspondingrninverse micelles. DLS, SLS, SEC-MALLS-VISCO, AFM and Cyro TEMrnmeasurements were performed to obtain a visual image from the appearance of thernaggregates. It was found that a linear-hyperbranched architecture is necessary, ifrnwell defined, monodisperse aggregates are required, e.g. for the preparation of orderedrnnanoarrays. Linear-linear block copolymers formed only polydisperse aggregates.rnAdditionally it was found that size distribution could be improved dramaticallyrnby passing the aggregates through a SEC column with large pores. The SEC columnsrnacted like a template in which the aggregates adopt a more stable conformation.rnIn the second part anionic polymerization was employed to synthesize silaneendfunctionalizedrnmacromonomers with different molecular weights based on polybutadienernand polyisoprene. These were polymerized by a hydrosilylation reaction inrnbulk to obtain branched polymers, using Karstedt’s catalyst. Surprisingly the additionrnof monofunctional silanes during the polymerization had only a minimal effect concerningrnthe degree of polymerization. It was possible to introduce silanes without increasingrnthe overall number of reaction steps by a very convenient “pseudo-copolymerization”rnmethod. All branched polymers were analyzed by SEC, SEC-MALLS,rnSEC-viscometry, 1H-NMR-spectroscopy and DSC concerning their branching ratio.rnThe branching parameters for the branched polymers exhibited similar characteristicsrnas hyperbranched polymers based on AB2 monomers. Detailed kinetic study showedrnthat the polymerization occurred very rapidly in comparison to the hydrosilylation polymerizationrnof classical AB2 type carbosilanes monomers.rnThe last part will deal with ferrocenyl-functionalized polymers. On the one hand,rnferrocenyl-functionalized polyglycerols (PG) were studied. Esterification of PGs withrndifferent molecular weight using ferrocenemonocarboxylic acid gave the ferrocenylrnfuntionalized polymers in high yields. On the other hand three different block copolymersrnwere prepared with different ratios of styrene to butadiene units (10:1, 4:1, 2:1).rnThe double bonds of the 1,2-PB block were hydrosilylated using silanes bearing onern(HSiMe2Fc) or two (HSiMeFc2) ferrocene units. High degrees of functionalizationrnwere obtained (up to 83 %). In this manner, six different ferrocenyl-rich block copolymersrnwith different fractions of ferrocene were prepared and analyzed, employingrnNMR-spectroscopy, SEC, SEC/MALLS/viscometry, DLS and cyclic voltammetry. Thernredox properties of the studied polymers varied primarily with the nature of the silanernunit attached. Additionally, the redox properties in solution of the studied polymersrnwere influenced by the block length ratio of the block copolymers. Unexpectedly, withrnincreasing block length of the ferrocenyl block the fraction of active ferrocenes decreased.rnNevertheless, in case of thin monolayer films this behaviour was not observed.rnAll polymers (PG and PS-b-PB based) exhibited good electrochemical propertiesrnin a wide range of solvents, which rendered them very interesting for biosensoricrnapplications.

The present thesis deals with the development of new branched polymer architectures containing hyperbranched polyglycerol. Materials investigated include hyperbranched oligomers, hyperbranched polyglycerols containing functional initiator-cores at the focal point, well-defined linear-hyperbranched block copolymers and also negatively charged hyperbranched polyelectrolytes.rnHyperbranched oligoglycerols (DPn = 7 and 14) have been synthesized for the first time. The materials show narrow polydispersity (Mw/Mn ca. 1.45) and a very low content in cyclic homopolymers. 13C NMR evidences the dendritic structure of the oligomers and the DB could be calculated (44% and 52%). These new oligoglycerols were compared with the industrial products obtained by polycondensation which exhibit narrow polydispersity (Mw/Mn<1.3) butrnmultimodal distribution in SEC. Detailed 13C NMR and Maldi-ToF studies reveal the presence of branched units and cyclic compounds. In comparison, the hyperbranched oligoglycerols comprise a very low proportion of cyclic homopolymer which render them very interesting materials for biomedical applications for example.rnThe site isolation of the core moiety in dendritic structure offers intriguing potential with respect to peculiar electro-optical properties. Various initiator-cores (n-alkyl amines, UVabsorbing amines and benzophenone) for the ROMBP of glycidol have been tested. The bisglycidolized amine initiator-cores show the best control over the molecular weight and the molecular weight distribution. The photochemical analyses of the naphthalene containingrnhyperbranched polyglycerols show a slight red shift, a pronounced hypochromic effect (decrease of the intensity of the band) compared with the parent model compound and the formation of a relative compact structure. The benzophenone containing polymers adopt an open structure in polar solvents. The fluorescence measurements show a clear “dendritic effect” on the fluorescence intensities and the quantum yield of the encapsulated benzophenone.rnA convenient 3-step strategy has been developed for the preparation of well-defined amphiphilic, linear-hyperbranched block copolymers via hypergrafting. The procedure represents a combination of carbanionic polymerization with the alkoxide-based, controlled ring-opening multibranching polymerization of glycidol. Materials consisting of a polystyrene linear block and a hyperbranched polyglycerol block exhibit narrow polydispersity (1.01-1.02rnfor 5.4% to 27% wt. PG and 1.74 for 52% wt. PG) with a high grafting efficiency. The strategy was also extended to materials with a linear polyisoprene block.rnDetailed investigations of the solution properties of the block copolymers with linear polystyrene blocks show that block copolymer micelles are stabilized by the highly branched block. The morphology of the aggregates is depending on the solvent: in chloroform monodisperse spherical shape aggregates and in toluene ellipsoidal aggregates are formed. On graphite these aggregates show interesting features, giving promising potential applications with respect to the presence of a very dense, functional and stable hyperbranched block.rnThe bulk morphology of the linear-hyperbranched block copolymers has been investigated. The materials with a linear polyisoprene block only behave like complex liquids due to the low Tg and the disordered nature of both components. For the materials with polystyrene, only the sample with 27% wt. hyperbranched polyglycerol forms some domains showing lamellae.rnThe preparation of hyperbranched polyelectrolytes was achieved by post-modification of the hydroxyl groups via Michael addition of acrylonitrile, followed by hydrolysis. In aqueous solution materials form large aggregates with size depending on the pH value. After deposition on mica the structures observed by AFM show the coexistence of aggregates andrnunimers. For the low molecular weight sample (PG 520 g·mol-1) extended and highly ordered terrace structures were observed. Materials were also successfully employed for the fabrication of composite organic-inorganic multilayer thin films, using electrostatic layer-bylayer self-assembly coupled with chemical vapor deposition.

Definiert konfigurierte mittelgroße ungesättigte Heterocyclen sind wertvolle Zwischenstufen in der Naturstoff- und Wirkstoffsynthese. Es konnte gezeigt werden, dass 2-alkinyl-substituierte Piperidine und Azepane in einer Aza-Keten-Claisen-Reaktion zu 10- und 11-gliedrigen Allenyllactamen umgelagert werden können. Ein 9-gliedriges Allenyllactam konnte nicht dargestellt werden (Ringspannung). Über eine sechs- bis sieben-stufige Reaktionssequenz konnten optisch aktive, geschützte Piperidinole aufgebaut werden. Es wurden Auxiliar kontrollierte Hetero-Diels-Alder-Reaktionen, diastereoselektive Reduktionen, Bestmann-Ohira Umlagerungen zu Alkinen und verschiedene Alkin-Funktionalisierungen erarbeitet. Eine Aza-Claisen-Umlagerung liefert schließlich optisch aktive Lactame deren absolute Konfiguration des Allensystems mittels NOE-NMR-Spektroskopie untersucht werden kann. Limitierungen und Möglichkeiten der Synthese werden eingehend diskutiert. Sowohl der stereochemische Verlauf der Reaktion als auch die Konformation der Produkte ermöglichen eine Fokussierung auf nachfolgende Naturstoffsynthesen vorzunehmen.

Urease is a nickel-dependent enzyme that catalyzes hydrolysis of urea in the last step of organic nitrogen mineralization. Its active site contains a dinuclear center for Ni(II) ions that must be inserted into the apo-enzyme through the action of four accessory proteins (UreD, UreE, UreF, UreG) leading to activation of urease. UreE, acting as a metallo-chaperone, delivers Ni(II) to the preformed complex of apo-urease-UreDFG and has the capability to enhance the GTPase activity of UreG. This study, focused on characterization of UreE from Sporosarcina pasteurii (SpUreE), represents a piece of information on the structure/mobility-function relationships that control nickel binding by SpUreE and its interaction with SpUreG. A calorimetric analysis revealed the occurrence of a binding event between these proteins with positive cooperativity and a stoichiometry consistent with the formation of the (UreE)2-(UreG)2 hetero-oligomer complex. Chemical Shift Perturbations induced by the protein-protein interaction were analyzed using high-resolution NMR spectroscopy, which allowed to characterize the molecular details of the protein surface of SpUreE involved in the complex formation with SpUreG. Moreover, backbone dynamic properties of SpUreE, determined using 15N relaxation analysis, revealed a general mobility in the nanoseconds time-scale, with the fastest motions observed at the C-termini. The latter analysis made it possible for the first time to characterize of the C-terminal portions, known to contain key residues for metal ion binding, that were not observed in the crystal structure of UreE because of disorder. The residues belonging to this portion of SpUreE feature large CSPs upon addition of SpUreG, showing that their chemical environment is directly affected by protein-protein interaction. Metal ion selectivity and affinity of SpUreE for cognate Ni(II) and non cognate Zn(II) metal ions were determined, and the ability of the protein to select Ni(II) over Zn(II), in consistency with the proposed role in Ni(II) cations transport, was established.

Diese Arbeit legt, im Rahmen eines interdisziplinären BMBF Projekts, die Grundlage für eine neue, potentielle Früherkennungsmethode für Prostatakrebs. Ausgehend von der Idee, einen sensitiv detektierbaren Farbstoff in selektiv, Hepsin-spaltbare Kapseln einzubringen, wurde ein Modellsystem erarbeitet. Als Zielsubstrat für das Enzym Hepsin wurde die Sequenz RQLRVVGG identifiziert. Basierend auf dem hier entwickelten Modell, könnte in Zukunft eine Detektion von Hepsin und damit von Prostatakrebs im Frühstadium in vivo erfolgen. rnUm das Konzept der Enzymspaltbarkeit zu zeigen,wurden, basierend auf einem Modell,enzymatisch-spaltbare, polymere Nanopartikeldargestellt. In dieser Arbeit konnte die Synthese von vier Generationen spezifisch Protease-spaltbarer, hydrophober Nanopartikel gezeigt werden. Die Enzyme Pepsin und Trypsin, die selektiv die Peptidsequenz GFF spalten, wurden als Modellenzyme für das im Prostatakarzinom überexprimierte Hepsin eingesetzt. rnrnAls Ausgangspolymer diente Poly(styrol-co-acrylsäure), das in freier, radikalischer Polymerisation hergestellt und mit Molekulargewichten zwischen 8500 und 49400 g/mol erhalten wurde. Der Funktionalisierungsgrad wurde zwischen 5 und 16%-Gew. variiert. Die Charakterisierung erfolgte mittels GPC und NMR-Spektroskopie. In einer polymeranalogen Reaktion wurden die Säurefunktionen zu Aminogruppen umgesetzt. Die resultierendenPolymere wurden unter Einsatz der über Jahrzehnte entwickelten, effektiven Peptidkupplungschemie mit enzymspaltbarenPeptiden gekuppelt, um dadurch definierte Peptid-Polymer-Konjugate zu erhalten.Als enzymatisch spaltbarer Teil des Konjugats, wurden mithilfe der Festphasenpeptidsynthese (SPPS),FRET(Fluoreszenz-Resonanz-Energie-Transfer)-markiertePeptide synthetisiert, die auf der Trypsin-spaltbaren Sequenz GFF basierten. Zum Nachweis der Vernetzung der späteren Nanopartikel, wurde zusätzlich einern15N-markierte Aminosäure (Fmoc-(15N)-Glycin) am N-Terminus der Peptide eingebaut, um die Reaktion des Amins zu einem Amid verfolgen zu können.Die Charakterisierung der Peptide erfolgte mittels1H-, 13C-NMR-Spektroskopie, MALDI TOF-MS und HPLC. Aus den Polymeren und dem Peptid wurden Peptid-Polymer-Konjugate hergestellt und die erfolgreiche Anbindung mittels DOSY-NMR-Spektroskopie und HPLC nachgewiesen. Aus den Konjugaten konnten im nächsten Schritt, durch Anwendung des inversen Miniemulsionsprozesses, Nanopartikel formuliertund diese in wässriges Milieu überführt werden. Die Dispersionen konnten mittels PCCS, REM und Zetapotentialmessungen charakterisiert werden, wobei Partikeldurchmesser um 230 nm resultierten. Der Nachweis der Vernetzung des Polymers in den Partikeln konnte durch die Reaktion desrn15N-Amins zu einem 15N-Amid mittels 15N-Festkörper-NMR-Spektroskopie nachverfolgt werden. Abschließend konnten die Partikel durch Trypsin gespalten werden, was durch das eingebaute FRET-Paar über eine in situ Detektion der relativen Fluoreszenz erfolgte. rnIn dieser Arbeit konnten Peptid-vernetzte, Polystyrol basierte Nanopartikel hergestellt und in Heterophase von Trypsin gespalten werden.rn

Polycarbosilanes are a class of polymers at the interface between organic and inorganic chemistry. They are characterized by a high thermal and chemical inertness and high flexibility, especially pronounced for branched structures. Linear polycarbosilanes are well known as precursors for the preparation of SiCx ceramics. Additionally, more sophisticated architectures like dendrimers, hyperbranched polymers or block copolymers have been the subject of research for more than a decade. The scope of this work was to expand the properties and fields of application for polycarbosilane-containing structures. Thus, the work is divided in two major parts. The first part covers the synthesis and characterization of hyperbranched polycarbosilanes containing organometallic moieties. Hyperbranched poly-carbosilanes were synthesized using hydrosilylation of diallylmethylsilane and methyldiundecenylsilane. The degree of branching for polydiallymethylsilane was determined using standard 1H-NMR spectroscopy. The functional building blocks ferrocenyldimethylsilane and diferrocenylmethylsilane were synthesized which contain an isolated ferrocene unit or two ferrocenes bridged by silicon, respectively. Hyperbranched polycarbosilanes functionalized with ferrocenyl moieties were synthesized by modification of preformed polymers or by copolymerization of AB2 carbosilane monomers with AX-type ferrocenylsilanes. Polymers with Mn = 2500-9000g/mol and ferrocene contents of up to 67wt% were obtained. Electrochemical characterization by cyclic voltammetry revealed that polymers functionalized with isolated ferrocene units showed a single reversible oxidation wave, while voltammograms for polymers functionalized with diferrocenyl silane exhibited two well-separated reversible oxidation-reduction waves. This shows that the polymer bound ferrocenes bridged by silicon are electronically communicating and thus oxidation of the first ferrocene shifts the oxidation potential for the adjacent one. The polymers were utilized successfully for the preparation of modified electrodes with persistent and reproducible electrochemical response in organic solvents as well as in aqueous solution. The presented work has proven that ferrocenyl-functionalized hyperbranched polymers exhibit similar electrochemical properties as the analogous dendrimers. In a further approach it was shown that hyperbranched polymers containing organometallic moieties can be synthesized by polymerization of a new ferrocene-containing AB2 monomer - diallylferrocenylsilane. The second part of this work is dedicated to the preparation of core-functional hyperbranched polycarbosilanes. Low molecular weight ambifunctional molecules were synthesized that contain double bonds for the attachment of a polycarbosilane polymer as well as a second functionality available for further reaction and modification. Reactive vinyl groups in the core molecule allow an efficient attachment of hyperbranched polycarbosilane which was proven by MALDI-ToF and GPC. In combination with slow monomer addition techniques molecular weight and polydispersity of the polymers were controlled successfully. Core-functional polymers were characterized by NMR-spectroscopy, MALDI-ToF and GPC. Polymers with polydispersities <2 and molecular weights up to 5300g/mol were obtained. Transformation of the double bonds of the carbosilane was demonstrated with various silanes using hydrosilylation reaction or hydrogenation. Additionally, the core-functionality was varied resulting in polymers with bromo-, phthalimide-, amine- or azide moieties. Thus, a versatile synthetic strategy was developed that allows the synthesis of tailor-made polymers.A promising approach is the application of the polymer building blocks in copolymer synthesis. Bisglycidolization of amine-functional polycarbosilanes produces macro-initiators that are suitable for the multibranching-ring opening polymerization of glycidol. This experiments lead to the first example of hyperbranched-hyperbranched amphiphilic block copolymers, hb-PG-b-hb-PCS. Furthermore, the implementation of copper-catalyzed cycloaddition between azide-functional polycarbosilane and alkyne-functional poly(ethoxyethyl glycidylether) resulted in linear-hyperbranched block copolymers. The facile removal of acetal protecting groups provided convenient access to lin-PG-b-hb-PCS.

Monoanionische, stickstoffhaltige Chelatliganden sind sehr populär und werden oft als Baueinheiten zur Synthese von Metallkomplexen eingesetzt. Im Gegensatz zu den bekannten Amidinat- und β-Diketiminatliganden wurde den Triazeniden bisher weniger Aufmerksamkeit geschenkt. Dies liegt hauptsächlich daran, dass wenige, sterisch anspruchsvolle Vertreter dieser Gruppe bekannt waren. Mit der Synthese neuer biphenyl- und terphenylsubstituierter Triazenide sind diese stärker in den Fokus der Forschung gerückt. Durch die Sperrigkeit der Liganden werden unerwünschte Ligandenumverteilungsreaktionen unterdrückt und der Weg zu neuen Metallkomplexen mit bemerkenswerten Eigenschaften geebnet. Diese Arbeit beschäftigt sich mit Triazenidokomplexen der Alkalimetalle, des Magnesiums, des Zinks und des Aluminiums. Zusätzlich zu den monofunktionellen wurden bifunktionelle Triazene, welche aus einer Biphenylgruppe aufgebaut sind und zwei terphenylsubstituierte Triazengruppen tragen, synthetisiert und deren Funktion als Komplexliganden untersucht. Durch das hohe Volumen der Liganden konnten Metallkomplexe hergestellt werden, die bemerkenswerte strukturelle Eigenschaften aufweisen. So bilden beispielsweise alle dargestellten Verbindungen Kristalle, die aus monomeren Baueinheiten aufgebaut sind. Eine homoleptische Magnesiumverbindung mit zwei Triazenidliganden weist eine ungewöhnliche, planare Koordinationssphäre auf, eine Triazenidodiphenylaluminiumverbindung zeigt eine interessante Ausrichtung der Phenylgruppen, die auf π-Stapeleffekte zurückgeführt wird und die Alkalimetalltriazenide weisen sekundäre Metall-π-Aren-Wechselwirkungen auf, womit eine bessere koordinative Absättigung des Koordinationszentrums erreicht wird. Zusätzlich zur strukturellen Charakterisierung mittels Röntgendiffraktometrie wurden, wo es möglich war 1H und 13C NMR-Messungen durchgeführt, Schmelzpunkte bestimmt sowie IR- und UV-VIS-Spektren aufgenommen. Durch die hohe Dichte an aromatischen Systemen innerhalb der Verbindungen, ist es bei einer dreikernigen Zinktriazenidverbindung gelungen mittels 1H NMR-Messungen die starken Einflüsse des Ringstroms nachzuweisen. Außerdem wurden DFT-Berechnungen durchgeführt, um herauszufinden, wie sehr die räumliche Ausrichtung der Bi- bzw. Terphenylsubstituenten gegenüber der N3-Baugruppe der Triazenide die Verteilung der negativen Ladung innerhalb eines monoanionischen Triazenids und somit dessen Koordinationseigenschaften beeinflusst. Diese Effekte konnten durch experimentelle Ergebnisse unterstützt werden.
