The zinc hydrolase superfamily is a group of divergently related proteins that are predominantly enzymes with a zinc-based catalytic mechanism. The common structural scaffold of the superfamily consists of an eight-stranded β-sheet flanked by six α-helices. Previous analyses, while acknowledging the likely divergent origins of leucine aminopeptidase, carboxypeptidase A and the co-catalytic enzymes of the metallopeptidase H clan based on their structural scaffolds, have failed to find any homology between the active sites in leucine aminopeptidase and the metallopeptidase H clan enzymes. Here we show that these two groups of co-catalytic enzymes have overlapping dizinc centers where one of the two zinc atoms is conserved in each group. Carboxypeptidase A and leucine aminopeptidase, on the other hand, no longer share any homologous zinc-binding sites. At least three catalytic zinc-binding sites have existed in the structural scaffold over the period of history defined by available structures. Comparison of enzyme-inhibitor complexes show that major remodeling of the substrate-binding site has occurred in association with each change in zinc ligation in the binding site. These changes involve re-registration and re-orientation of the substrate. Some residues important to the catalytic mechanism are not conserved amongst members. We discuss how molecules acting in trans may have facilitated the mutation of catalytically important residues in the active site in this group.

A new method to synthesize conducting oxide nanoparticles with low photocatalytic activity was investigated. Initially, the preparation of amorphous ZnO-SnO2 solid solution nanoparticles was studied using a sol-gel technique. It was found that X-ray amorphous nanopowders with low photocatalytic activity were produced when the precipitates were heat treated below 500 °C. However, FT-IR data showed that the sample may not be an oxide semiconductor. A mixture of ZnO and SnO2 crystalline nanoparticles was also produced at 800 °C and found to have much reduced photoactivity than commercial ZnO nanoparticles having a similar specific surface area.

In situ prepared zinc disorbate (ZDS) in natural rubber (NR) by the reaction of zinc oxide and sorbic acid was used to reinforce the dicumyl peroxide-cured NR vulcanizate. The changes in mechanical properties of NR vulcanizates after ageing and were determined and the structures and thermal stability of vulcanizates were also analyzed using scanning electron microscope and thermal gravimetric analyzer. The change ratios in tensile strength and elongation at break of NR vulcanizate with theoretic formation of ZDS of 21phr can be increased to -33 from -44 and -27 from -38 after ageing and the initial weight loss temperature of NR vulcanizate can be increased for about 7°C as compared to un-reinforced NR vulcanizate, indicating that the antioxidative behavior and thermal stability of NR can be improved significantly with theoretic formation of ZDS of 21phr.

The apical cytoplasm of airway epithelium (AE) contains abundant labile zinc (Zn) ions that are involved in the protection of AE from oxidants and inhaled noxious substances. A major question is how dietary Zn traffics to this compartment. In rat airways, in vivo selenite autometallographic (Se-AMG)-electron microscopy revealed labile Zn-selenium nanocrystals in structures resembling secretory vesicles in the apical cytoplasm. This observation was consistent with the starry-sky Zinquin fluorescence staining of labile Zn ions confined to the same region. The vesicular Zn transporter ZnT4 was likewise prominent in both the apical and basal parts of the epithelium both in rodent and human AE, although the apical pools were more obvious. Expression of ZnT4 mRNA was unaffected by changes in the extracellular Zn concentration. However, levels increased 3-fold during growth of cells in air liquid interface cultures and decreased sharply in the presence of retinoic acid. When comparing nasal versus bronchial human AE cells, there were significant positive correlations between levels of ZnT4 from the same subject, suggesting that nasal brushings may allow monitoring of airway Zn transporter expression. Finally, there were marked losses of both basally-located ZnT4 protein and labile Zn in the bronchial epithelium of mice with allergic airway inflammation. This study is the first to describe co-localization of zinc vesicles with the specific zinc transporter ZnT4 in airway epithelium and loss of ZnT4 protein in inflamed airways. Direct evidence that ZnT4 regulates Zn levels in the epithelium still needs to be provided. We speculate that ZnT4 is an important regulator of zinc ion accumulation in secretory apical vesicles and that the loss of labile Zn and ZnT4 in airway inflammation contributes to AE vulnerability in diseases such as asthma.

A novel sensitive humidity nanosensor based on Na1-doped ZnO nanofiber membrane has been prepared via electrospinning and calcination. The product was characterized by scanning electron microscopy and X-ray diffraction. During the whole relative humidity (11%–95%) measurement, the response and recovery time is about 3 and 6 s, respectively, with good linearity, and reproducibility. These remarkable and sensitive sensing performances make our product a good candidate in fabricating humidity sensors.

This thesis examines the direct interaction of Docosahexaenoic acid (DHA, an omega-3 fatty acid) against zinc-induced mitochondrial dysfunction and involvement of bioenergetic regulation as a zinc toxicity target, which may be the initiator of oxidative stress, caspase cascade, alteration in epigenetic patterns and therefore gene expression in human neuronal cells.

The UVA performances of two all-mineral zinc oxide sunscreens are measured following Colipa and ISO procedures and compared to a sunscreen containing only organic actives. It is found that the two sunscreen types yield very different in vitro SPF and UVA results. It shown that the results can be rationalized in terms of the differences in the monochromatic extinction spectra of the two types of sunscreens.
