Background: Sensory neurones from the trigeminal nerve innervate the oro-facial region and teeth. Transient receptor potential channels (TRPs) expressed by these neurones are responsible for relaying sensory information such as changes in ambient temperature, mechanical sensations and pain. Study of TRP channel expression and regulation in human sensory neurones therefore merits investigation to improve our understanding of allodynia and hyperalgesia. Objective: The objective of this study was to differentiate human dental pulp stem cells (hDPSCs) towards a neuronal phenotype (peripheral neuronal equivalents; PNEs) and employ this model to study TRP channel sensitisation. Method: hDPSCs were enriched by preferential adhesion to fibronectin, plated on coverslips (thickness 0) coated with poly-l-ornithine and laminin and then differentiated for 7 days in neurobasal A medium with additional supplementation. A whole cell patch clamp technique was used to investigate whether TRP channels on PNE membranes were modulated in the presence of nerve growth factor (NGF). PNEs were treated with NGF for 20 minutes immediately before experimentation and then stimulated for TRPA1 activity using cinnamaldehyde. Peak currents were read at 80 mV and -80 mV and compared to peak currents recorded in untreated PNEs. Data were analysed and plotted using Clampfit9 software (Molecular Devices, Sunnyvale, California, USA). Result: Results showed for the first time that pre-treatment of PNEs by NGF produced significantly larger inward and outward currents demonstrating that TRPA1 channels on PNE membranes were capable of becoming sensitised following treatment with NGF. Conclusion: Sensitisation of TRPA1 by NGF provides evidence of a mechanism for rapid neuronal sensitisation that is independent of TRPA1 gene expression

Background: The oro-facial region is densely innervated by the trigeminal nerve, which when stimulated can induce noxious pain sensation and contribute to neurogenic inflammation in local tissues. Recent research on the expression of specialised ion channels on the trigeminal nerve has highlighted the need to undertake more extensive studies on ion channel expression/functionality with the aim of elucidating their role in pain sensations. A major family of such ion channels is the transient receptor potential (TRP) channels which are activated by a wide variety of thermal, mechanical or chemical stimuli and merit investigation as possible druggable targets for future analgesics.
Objective: Study of TRP channel expression and regulation in oro-facial tissues is hindered by the fact that the cell bodies of neurons innervating these tissues are located in the trigeminal ganglion. Using dental pulp stem cells differentiated towards peripheral neuronal equivalents (PNEs), we sought to determine TRP channel expression, functionality and potential modulation by cytokines in this novel model.
Method: Dental pulp stem cells (DPSCs) were grown on substrate-coated tissue culture plates and differentiated towards a neuronal phenotype using neuronal induction media. Quantitative polymerase chain reaction (qPCR) was performed on PNEs +/-cytokine treatment. Ion channel functionality was investigated using whole cell patch clamping.
Result: qPCR analysis showed that PNEs expressed the TRP channels TRPA1, TRPV1, TRPV4 and TRPM8. TRPA1 was the most abundantly expressed TRP channel studied whereas TRPM8 was lowly expressed. TRP channel expression was shown to be regulated by treatment with inflammatory cytokines. Patch clamp studies using specific agonists and antagonists for TRPA1 and TRPV1 showed these channels were functional.
Conclusion: PNEs differentiated from DPSCs provide a suitable model for TRP channel expression, regulation, and sensistisation in oro-facial tissues. This human neuronal model has potential for use in pre-clinical studies of novel analgesics.

Metabolic networks are highly connected and complex, but a single enzyme, O-GlcNAc transferase (OGT) can sense the availability of metabolites and also modify target proteins. We show that inhibition of OGT activity inhibits the proliferation of prostate cancer cells, leads to sustained loss of c-MYC and suppresses the expression of CDK1, elevated expression of which predicts prostate cancer recurrence (p=0.00179). Metabolic profiling revealed decreased glucose consumption and lactate production after OGT inhibition. This decreased glycolytic activity specifically sensitized prostate cancer cells, but not cells representing normal prostate epithelium, to inhibitors of oxidative phosphorylation (rotenone and metformin). Intra-cellular alanine was depleted upon OGT inhibitor treatment. OGT inhibitor increased the expression and activity of alanine aminotransferase (GPT2), an enzyme that can be targeted with a clinically approved drug, cycloserine. Simultaneous inhibition of OGT and GPT2 inhibited cell viability and growth rate, and additionally activated a cell death response. These combinatorial effects were predominantly seen in prostate cancer cells, but not in a cell-line derived from normal prostate epithelium. Combinatorial treatments were confirmed with two inhibitors against both OGT and GPT2. Taken together, here we report the reprogramming of energy metabolism upon inhibition of OGT activity, and identify synergistically lethal combinations that are prostate cancer cell specific.

As the ESA Rosetta mission approached, orbited, and sent a lander to comet 67P/Churyumov-Gerasimenko in 2014, a large campaign of ground-based observations also followed the comet. We constrain the total activity level of the comet by photometry and spectroscopy to place Rosetta results in context and to understand the large-scale structure of the comet's coma pre-perihelion. We performed observations using a number of telescopes, but concentrate on results from the 8m VLT and Gemini South telescopes in Chile. We use R-band imaging to measure the dust coma contribution to the comet's brightness and UV-visible spectroscopy to search for gas emissions, primarily using VLT/FORS. In addition we imaged the comet in near-infrared wavelengths (JHK) in late 2014 with Gemini-S/Flamingos 2. We find that the comet was already active in early 2014 at heliocentric distances beyond 4 au. The evolution of the total activity (measured by dust) followed previous predictions. No gas emissions were detected despite sensitive searches. The comet maintains a similar level of activity from orbit to orbit, and is in that sense predictable, meaning that Rosetta results correspond to typical behaviour for this comet. The gas production (for CN at least) is highly asymmetric with respect to perihelion, as our upper limits are below the measured production rates for similar distances post-perihelion in previous orbits.

Harmful algal blooms (HABs) are a natural global phenomena emerging in severity and extent. Incidents have many economic, ecological and human health impacts. Monitoring and providing early warning of toxic HABs are critical for protecting public health. Current monitoring programmes include measuring the number of toxic phytoplankton cells in the water and biotoxin levels in shellfish tissue. As these efforts are demanding and labour intensive, methods which improve the efficiency are essential. This study compares the utilisation of a multitoxin surface plasmon resonance (multitoxin SPR) biosensor with enzyme-linked immunosorbent assay (ELISA) and analytical methods such as high performance liquid chromatography with fluorescence detection (HPLC-FLD) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) for toxic HAB monitoring efforts in Europe. Seawater samples (n = 256) from European waters, collected 2009-2011, were analysed for biotoxins: saxitoxin and analogues, okadaic acid and dinophysistoxins 1/2 (DTX1/DTX2) and domoic acid responsible for paralytic shellfish poisoning (PSP), diarrheic shellfish poisoning (DSP) and amnesic shellfish poisoning (ASP), respectively. Biotoxins were detected mainly in samples from Spain and Ireland. France and Norway appeared to have the lowest number of toxic samples. Both the multitoxin SPR biosensor and the RNA microarray were more sensitive at detecting toxic HABs than standard light microscopy phytoplankton monitoring. Correlations between each of the detection methods were performed with the overall agreement, based on statistical 2 × 2 comparison tables, between each testing platform ranging between 32% and 74% for all three toxin families illustrating that one individual testing method may not be an ideal solution. An efficient early warning monitoring system for the detection of toxic HABs could therefore be achieved by combining both the multitoxin SPR biosensor and RNA microarray.

Over the past few decades, there has been an increased frequency and duration of cyanobacterial Harmful Algal Blooms (HABs) in freshwater systems globally. These can produce secondary metabolites called cyanotoxins, many of which are hepatotoxins, raising concerns about repeated exposure through ingestion of contaminated drinking water or food or through recreational activities such as bathing/ swimming. An ultra-performance liquid chromatography tandem mass spectrometry (UPLC–MS/MS) multi-toxin method has been developed and validated for freshwater cyanotoxins; microcystins-LR, -YR, -RR, -LA, -LY and -LF, nodularin, cylindrospermopsin, anatoxin-a and the marine diatom toxin domoic acid. Separation was achieved in around 9 min and dual SPE was incorporated providing detection limits of between 0.3 and 5.6 ng/L of original sample. Intra- and inter-day precision analysis showed relative
standard deviations (RSD) of 1.2–9.6% and 1.3–12.0% respectively. The method was applied to the analysis of aquatic samples (n = 206) from six European countries. The main class detected were the hepatotoxins; microcystin-YR (n = 22), cylindrospermopsin (n = 25), microcystin-RR (n = 17), microcystin-LR (n = 12), microcystin-LY (n = 1), microcystin-LF (n = 1) and nodularin (n = 5). For microcystins, the levels detected ranged from 0.001 to 1.51 mg/L, with two samples showing combined levels above the guideline set by the WHO of 1 mg/L for microcystin-LR. Several samples presented with multiple toxins indicating the potential for synergistic effects and possibly enhanced toxicity. This is the first published pan European survey of freshwater bodies for multiple biotoxins, including two identified for the first time; cylindrospermopsin in Ireland and nodularin in Germany, presenting further incentives for improved monitoring and development of strategies to mitigate human exposure.

The importance of student’s engagement has been recently pointed out in research. However, there has been a lack of engagement assessment instruments, showing psychometric qualities, such as reliability and validity. Objective: This paper presents the Portuguese adaptation of the “Student’s Engagement in Schools International Scale” (SESIS), drawn up from 12 countries international study (Lam et al., 2012; Lam et al., in press). Method: Psychometric properties of this scale were examined using data from 685 students, of both sexes, and from different grades (6th, 7th, 9th and 10th), and regions of the country. Results: Factorial analysis of the results, with varimax rotation, conducted to three different factors which explain 62,19% of the variance. The scale integrates the original 33 items, and cognitive, affective and behavioural dimensions. For the external validity study, the relationship between student’s engagement in school results and other school variables — academic performance, school conduct — was considered, and significant relations were observed, as expected. Conclusion: The data presented highlights the qualities of SESIS, as well as its usefulness for research purpose. Suggestion: It is suggested the investigation of the extension of SESIS’s three-dimensionality, in future studies

Este estudo tem como principal objetivo compreender a complexidade da expressão de preconceitos por parte de professores/as em relação a gays e lésbicas, área em que a investigação parece ser limitada. Neste sentido, procurámos avaliar esta questão com recurso a uma amostra de 204 professores/as de 13 escolas públicas do Distrito de Évora. Na realização deste trabalho, foram aplicados três instrumentos: questionário de caracterização sociodemográfica, Questionário de Discriminação Preconceito Polimorfo (Massey, 2009) e a subescala do Heterossexismo Moderno da Escala Multidimensional de Atitudes em relação a Gays e Lésbicas (Gato, Fontaine & Carneiro, 2012). Os principais resultados encontrados sugerem que o sexo, idade, posicionamentos político e religioso dos/as participantes se encontram relacionados com a expressão de preconceitos em relação a gays e lésbicas.

Este estudo tem como objetivo compreender as diferenças relativamente ao ciúme nas relações amorosas de adolescentes, tendo em conta o género e a orientação sexual. Para tal, foi criado um questionário (Questionário de Ciúme nas Relações Amorosas Adolescentes - QCRAA) que pretende avaliar aspetos do ciúme presentes em relações amorosas adolescentes - Preocupação, Controlo do Comportamento e Comportamentos de Investigação e Agressão. O QCRAA foi aplicado a 316 jovens, tendo sido comparados os resultados em função do género, da orientação sexual, do nível de ensino e da estabilidade da relação amorosa. Os resultados sugerem que as raparigas possuem mais comportamentos de investigação e agressão, e que os níveis de preocupação nas relações amorosas dos/as jovens variam conforme a estabilidade da relação, sendo mais elevados quando a relação não é estável. Neste estudo serão analisados e discutidos os principais resultados obtidos, bem como as suas implicações para a intervenção psicológica na prevenção da violência nas relações amorosas.

A partir de meados do século XX, a investigação qualitativa tem vindo a receber uma aceitação crescente e um maior reconhecimento por parte de investigadores em diferentes áreas de conhecimento psicológico. Dentro dos métodos qualitativos, as entrevistas assumem-se como a técnica mais comummente usada na recolha de dados. Não obstante, a utilização desta abordagem com crianças apresenta alguns desafios para o investigador. Ouvir as crianças e a consciencialização de que as mesmas são cidadãs e membros activos do processo de investigação assenta numa mudança paradigmática, ultrapassando uma perspectiva que as olha como incapazes e imaturas. Neste trabalho, pretende-se apresentar a evolução paradigmática entre a abordagem quantitativa e qualitativa, bem como focar especificamente a técnica da entrevista com crianças. Em termos práticos, pretende-se relatar a experiência de uma recolha de dados recorrendo a essa mesma técnica, com crianças do pré-escolar e do 1º ano do ensino básico.

This work reports the investigation of tolerance to salt in two year-old olive trees (Olea europaea L.) of three varieties, Arbequina, Cobrançosa and Galega vulgar. Plants were grown in 10 L plastic pots containing approximately 9 Kg of a sandy granitic soil, on a greenhouse. For 3 months (from the beginning of February to the end of April 2012), they were subjected to three levels of salinity in the irrigation water, 0 mM, 80 mM and 200 mM NaCl (6 plants per salinity level in a total of 18 plants of each variety),. Stomatal conductance (gs) and relative leaf chlorophyll content were assessed on each plant in February, March and April. Mid-day leaf water potential () and soil salinity were measured at the end of the experiment (April). On average, concerning all treatments and dates of determination, stomatal conductance of Arbequina and Galega vulgar was quite similar, around 40 mmol m-2 s-1, but Cobrançosa had a value of gs 36% higher, almost 50% higher (61 mmol m-2 s-1) when compared with the controls (0 mM salt) of the other two varieties. In percentage of controls, there was little difference in gs between varieties and between salinities during February and March. In contrast, in April, after about 90 days of exposure to salt, there was a clear decrease in gs with salt irrigation, proportional to salt concentration. Compared with controls, plants irrigated with 200 mM salt showed around 80% (Arbequina) or 85% (Cobrançosa and Galega vulgar) decrease in gs. Chlorophyll content of leaves showed less than 5% difference between varieties on the average of all treatments and dates of determination. During the course of this experiment, the salinity levels used did not show any relevant effect on chlorophyll content. Overall, at the end of the experimental period (April), leaf water potential () at midday was significantly higher in Cobrançosa (-1,4 MPa) than in Galega vulgar (-1,7 MPa) or Arbequina (-1,8 MPa), and salt decreased  of control plants (-1,25 MPa) by an average 30% (with 80 mM) and 65% (with 200 mM). At the end of the experiment, salinity in the soil irrigated with 0 mM, 80 mM or 200 mM NaCl was, on average of all varieties, 0,2 mS, 1,0 mS or 2,0 mS, respectively. Soil salinity was quite similar in Arbequina and Galega vulgar but about 35% lower in the pots of Cobrançosa, on average of all salt-irrigation levels. Plants of Cobrançosa had higher stomatal conductance, however they showed higher water potential and lower salinity in the soil. These apparently contradictory results seem to suggest that Cobrançosa responds to salt differently from the other two varieties. This issue needs further investigation.

Background Complex medication regimens may adversely affect compliance and treatment outcomes. Complexity can be assessed with the medication regimen complexity index (MRCI), which has proved to be a valid, reliable tool, with potential uses in both practice and research. Objective To use the MRCI to assess medication regimen complexity in institutionalized elderly people. Setting Five nursing homes in mainland Portugal. Methods A descriptive, cross-sectional study of institutionalized elderly people (n = 415) was performed from March to June 2009, including all inpatients aged 65 and over taking at least one medication per day. Main outcome measure Medication regimen complexity index. Results The mean age of the sample was 83.9 years (±6.6 years), and 60.2 % were women. The elderly patients were taking a large number of drugs, with 76.6 % taking more than five medications per day. The average medication regimen complexity was 18.2 (±SD = 9.6), and was higher in the females (p < 0.001). The most decisive factors contributing to the complexity were the number of drugs and dosage frequency. In regimens with the same number of medications, schedule was the most relevant factor in the final score (r = 0.922), followed by pharmaceutical forms (r = 0.768) and additional instructions (r = 0.742). Conclusion Medication regimen complexity proved to be high. There is certainly potential for the pharmacist’s intervention to reduce it as part as the medication review routine in all the patients.

Inúmeras potenciais funções foram sugeridas para a APP (Proteína Precursora de Amilóide de Alzheimer), contudo, fisiologicamente, a função precisa da APP não foi ainda desvendada. A APP tem características consistentes com a função de molécula-receptora, capaz de reconhecer sinais extracelulares. Também relevante para este trabalho, é o facto de que a sinalização através de RIP (Proteólise Intramembranar Regulada) tem consequências na expressão génica, como no caso da sinalização tipo-Notch. Tal como a proteína Notch, a APP é processada resultando num fragmento C-terminal designado por AICD (Domínio Intracelular da APP). Neste trabalho é focado o papel importante na sinalização nuclear desempenhado pelo fragmento AICD, especificamente através da interacção com proteínas adaptadoras, promovendo a transcrição. Com o objectivo de contribuir para uma melhor compreensão da base molecular da DA (Doença de Alzheimer), torna-se importante investigar as vias de localização nuclear do AICD e o seu envolvimento na transcrição génica, possivelmente afectando proteínas até agora não associadas à DA. Por estes motivos é fundamental a identificação de novas proteínas que interajam com a APP. Um rastreio foi efectuado, utilizando o sistema Dois-Híbrido em Levedura, para identificar interacções específicas do AICD no cérebro humano e, assim, caracterizar o interactoma do AICD. Foi feito o rastreio de aproximadamente 1.1x108 clones de uma biblioteca de cDNA de cérebro humano com o domínio C-terminal da APP com a mutação Y687E, que mimetiza o estado fosforilado. De experiências anteriores deste laboratório sabemos que a tirosina-687 afecta a localização subcelular da APP e é também consensual que a fosforilação é importante nos mecanismos de transdução de sinais, daí a utilização deste mutante parecer apropriada. O rastreio originou 55 clones positivos que foram analisados para identificar proteínas que interagem com a APP. Dois clones são particularmente importantes, a RanBPM e a Transportin-SR2, visto que estão associadas ao transporte de proteínas para o núcleo e confirmam a sinalização nuclear da APP. ABSTRACT: Many putative functions for APP (Alzheimer’s amyloid precursor protein) have been suggested, although the precise physiological function of APP remains to be elucidated. APP has characteristics consistent with it having a role as a receptor, capable of mediating extracellular signals. Also of relevance to the work described here is that RIP (Regulated Intramembrane Proteolysis) signalling can have consequences in gene expression, similar to Notch signalling. Like the latter, APP is processed by RIP resulting in a C-terminal fragment known as AICD. Here we test the hypothesis that the AICD fragment may play an important role in nuclear signalling, specifically by interacting with adaptor proteins potentiating transcription. Therefore, in order to contribute to our understanding of the molecular basis of AD (Alzheimer’s disease) it is important to investigate the pathways of AICD nuclear targeting and its involvement in gene transcription, possibly affecting other proteins hitherto not associated with AD. Thus, it is important to identify AICD binding proteins. A Yeast Two-Hybrid (YTH) screen was performed to identify human brainspecific AICD binding proteins, and thus characterize the AICD interactome. The screen of approximately 1.1 x108 clones from a human brain cDNA library was carried out using the AICD fragment with an Y687E mutation, which mimics phosphorylation on that residue. From previous work carried out in the laboratory we know that tyrosine-687 phosphorylation affects subcellular localization of APP, and it is also recognized that phosphorylation events are important in signal transduction mechanisms, hence the use of this mutant is appropriate. The YTH screen yielded 55 positive clones that were analysed and several novel brain-specific APP binding proteins were identified. Two clones were particularly important, RanBPM and Transportin-SR2, being that they are associated with the nuclear transport of proteins, and support the nuclear signalling for APP.

O presente trabalho é relativo aos resultados das uroculturas realizadas na instituição durante os anos de 2013 e 2014 e respetivos testes de suscetibilidade aos antibióticos (TSA). Todos os resultados se referem a animais de companhia assistidos no âmbito da consulta externa devido a sinais clínicos de afeção do trato genito-urinário. O médico veterinário no decurso da sua atividade profissional deve assegurar-se de que promove uma prática racional e responsável na utilização de antibioterapia, limitando a sua utilização aos casos em que realmente é necessária. Por outro lado, e face ao aumento de estirpes bacterianas multirresistentes, o sucesso terapêutico depende cada vez mais de uma escolha adequada do agente antimicrobiano, que não deverá ser empírica. Apesar de não ser claro se o fluxo de transmissão de resistências microbianas é feito no sentido do animal para o humano ou no sentido inverso, certo é que o fluxo é possível e existe (Ewers, 2012).
