Dextran-based polymers are versatile hydrophilic materials, which can provide functionalized surfaces in various areas including biological and medical applications. Functional, responsive, dextran based hydrogels are crosslinked, dextran based polymers allowing the modulation of response towards external stimuli. The controlled modulation of hydrogel properties towards specific applications and the detailed characterization of the optical, mechanical, and chemical properties are of strong interest in science and further applications. Especially, the structural characteristics of swollen hydrogel matrices and the characterization of their variations upon environmental changes are challenging. Depending on their properties hydrogels are applied as actuators, biosensors, in drug delivery, tissue engineering, or for medical coatings. However, the field of possible applications still shows potential to be expanded. rnSurface attached hydrogel films with a thickness of several micrometers can serve as waveguiding matrix for leaky optical waveguide modes. On the basis of highly swelling and waveguiding dextran based hydrogel films an optical biosensor concept was developed. The synthesis of a dextran based hydrogel matrix, its functionalization to modulate its response towards external stimuli, and the characterization of the swollen hydrogel films were main interests within this biosensor project. A second focus was the optimization of the hydrogel characteristics for cell growth with the aim of creating scaffolds for bone regeneration. Matrix modification towards successful cell growth experiments with endothelial cells and osteoblasts was achieved.rnA photo crosslinkable, carboxymethylated dextran based hydrogel (PCMD) was synthesized and characterized in terms of swelling behaviour and structural properties. Further functionalization was carried out before and after crosslinking. This functionalization aimed towards external manipulation of the swelling degree and the charge of the hydrogel matrix important for biosensor experiments as well as for cell adhesion. The modulation of functionalized PCMD hydrogel responses to pH, ion concentration, electrochemical switching, or a magnetic force was investigated. rnThe PCMD hydrogel films were optically characterized by combining surface plasmon resonance (SPR) and optical waveguide mode spectroscopy (OWS). This technique allows a detailed analysis of the refractive index profile perpendicular to the substrate surface by applying the Wentzel Kramers Brillouin (WKB) approximation. rnIn order to perform biosensor experiments, analyte capturing units such as proteins or antibodies were covalently coupled to the crosslinked hydrogel backbone by applying active ester chemistry. Consequently, target analytes could be located inside the waveguiding matrix. By using labeled analytes, fluorescence enhancement was achieved by fluorescence excitation with the electromagnetic field in the center of the optical waveguide modes. The fluorescence excited by the evanescent electromagnetic field of the surface plasmon was 2 3 orders of magnitude lower. Furthermore, the signal to noise ratio was improved by the fluorescence excitation with leaky optical waveguide modes.rnThe applicability of the PCMD hydrogel sensor matrix for clinically relevant samples was proofed in a cooperation project for the detection of PSA in serum with long range surface plasmon spectroscopy (LRSP) and fluorescence excitation by LRSP (LR SPFS). rn

Regenerative medicine and tissue engineering attempt to repair or improve the biological functions of tissues that have been damaged or have ceased to perform their role through three main components: a biocompatible scaffold, cellular component and bioactive molecules. Nanotechnology provide a toolbox of innovative scaffold fabrication procedures in regenerative medicine. In fact, nanotechnology, using manufacturing techniques such as conventional and unconventional lithography, allows fabricating supports with different geometries and sizes as well as displaying physical chemical properties tunable over different length scales. Soft lithography techniques allow to functionalize the support by specific molecules that promote adhesion and control the growth of cells. Understanding cell response to scaffold, and viceversa, is a key issue; here we show our investigation of the essential features required for improving the cell-surface interaction over different scale lengths. The main goal of this thesis has been to devise a nanotechnology-based strategy for the fabrication of scaffolds for tissue regeneration. We made four types of scaffolds, which are able to accurately control cell adhesion and proliferation. For each scaffold, we chose properly designed materials, fabrication and characterization techniques.

This PhD Thesis is focused on the development of fibrous polymeric scaffolds for tissue engineering applications and on the improvement of scaffold biomimetic properties. Scaffolds were fabricated by electrospinning, which allows to obtain scaffolds made of polymeric micro or nanofibers. Biomimetism was enhanced by following two approaches: (1) the use of natural biopolymers, and (2) the modification of the fibers surface chemistry. Gelatin was chosen for its bioactive properties and cellular affinity, however it lacks in mechanical properties. This problem was overcome by adding poly(lactic acid) to the scaffold through co-electrospinning and mechanical properties of the composite constructs were assessed. Gelatin effectively improves cell growth and viability and worth noting, composite scaffolds of gelatin and poly(lactic acid) were more effective than a plain gelatin scaffold. Scaffolds made of pure collagen fibers were fabricated. Modification of collagen triple helix structure in electrospun collagen fibers was studied. Mechanical properties were evaluated before and after crosslinking. The crosslinking procedure was developed and optimized by using - for the first time on electrospun collagen fibers - the crosslinking reactant 1,4-butanediol diglycidyl ether, with good results in terms of fibers stabilization. Cell culture experiments showed good results in term of cell adhesion and morphology. The fiber surface chemistry of electrospun poly(lactic acid) scaffold was modified by plasma treatment. Plasma did not affect thermal and mechanical properties of the scaffold, while it greatly increased its hydrophilicity by the introduction of carboxyl groups at the fiber surface. This fiber functionalization enhanced the fibroblast cell viability and spreading. Surface modifications by chemical reactions were conducted on electrospun scaffolds made of a polysophorolipid. The aim was to introduce a biomolecule at the fiber surface. By developing a series of chemical reactions, one oligopeptide every three repeating units of polysophorolipid was grafted at the surface of electrospun fibers.

Ziel der Arbeit war es, Sialyl-LewisX-Mimetika auf Basis ortho-C-glycosylierter Phenole als Inhibitoren für die Selektin-Ligand-Wechselwirkungen zu synthetisieren. Dazu wurde zunächst die Stereoselektivität der ortho-C-Mannosylierung untersucht. Dabei wurde gezeigt, dass bei der Umsetzung von Phenolen mit dem benzylgeschützten Mannosyl-trichloracetimidat in Gegenwart von TMSOTf selektiv das β-C-Mannosid erhalten wurde. Gleichzeitig konnte anhand der NMR-spektroskopischen Untersuchungen nachgewiesen werden, dass die in der Literatur beschriebenen α-C-Mannoside von Phenolen tatsächlich β-konfiguriert sind. Wenn Naphthole als Glycosylakzeptoren verwendet wurden, konnten durch Modifikation des Promotors auch die für die Synthese der Mimetika benötigten α-C-Mannoside erhalten werden, wobei ZnCl2 als Promotor die besten Ergebnisse lieferte. Allerdings zeigten die synthetisierten α-C-Mannoside und α-C-Galactoside eine Inversion des Pyranoseringes und lagen in der ungewöhnlichen 1C4-Konformation vor.rnAnschließend konnte auf diese Weise das durch Docking-Studien gefundene Mimetikum (2S)-3-Cyclohexyl-2-[7-hydroxy-8-(α-D-mannosyl)naphthalin-2-yloxy]propionsäure syntheti-siert werden. Es besaß jedoch in Zelladhäsionstests keine ausreichende Aktivität bei der Inhibierung der Selektin-Ligand-Wechselwirkung. Bei den ursprünglichen Dockingstudien war allerdings von der gewohnten 4C1-Konformation ausgegangen worden. Spätere NMR-Experimente und DFT-Berechnungen zeigten, dass das Mimetikum tatsächlich in der 1C4-Konformation vorlag und es deshalb nicht aktiv war. Die synthetisierten Stereo- und Regioisomere zeigten in Zelladhäsionstests ebenfalls keine Aktivität.rnVersuche, die α-1-C-Mannosylnaphthole zu den benötigten 1-C-2-O-Diglycosyl-naphthalinen umzusetzen waren nicht erfolgreich, da die phenolische OH-Gruppe sterisch zu sehr abgeschirmt war, um unter milden Reaktionsbedingungen glycosyliert zu werden, bzw. die α-1-C-Mannosylnaphthaline unter drastischeren Reaktionsbedingungen nicht stabil waren. Daher wurde 1-(2′,3′,4′,6′-Tetra-O-benzyl-β-D-galactopyranosyl)-2-naphthol mit 2,3,4,6-Tetra-O-acetyl-α-D-mannopyranosyl-trichloracetimidat in Gegenwart von TMSOTf zum ersten synthetischen 1-C-2-O-Diglycosyl-phenol umgesetzt. Nach Abspaltung der Schutzgruppen sollte das erhaltene 1-Galactosyl-2-O-mannosyl-naphthalin enzymatisch zum Sialyl-LewisX-Mimetikum verlängert werden. Es wurde vom Enzym jedoch nicht als Substrat erkannt. Versuche zur chemischen Anbindung des Säurebausteins stehen noch aus.rn

In the central nervous system (CNS), oligodendrocytes form the multilamellar and compacted myelin sheath by spirally wrapping around defined axons with their specialised plasma membrane. Myelin is crucial for the rapid saltatory conduction of nerve impulses and for the preservation of axonal integrity. The absence of the major myelin component Myelin Basic Protein (MBP) results in an almost complete failure to form compact myelin in the CNS. The mRNA of MBP is sorted to cytoplasmic RNA granules and transported to the distal processes of oligodendrocytes in a translationally silent state. A main mediator of MBP mRNA localisation is the trans-acting factor heterogeneous nuclear ribonucleoprotein (hnRNP) A2 which binds to the cis-acting A2 response element (A2RE) in the 3’UTR of MBP mRNA. A signalling cascade had been identified that triggers local translation of MBP at the axon-glial contact site, involving the neuronal cell adhesion molecule (CAM) L1, the oligodendroglial plasma membrane-tethered Fyn kinase and Fyn-dependent phosphorylation of hnRNP A2. This model was confirmed here, showing that L1 stimulates Fyn-dependent phosphorylation of hnRNP A2 and a remodelling of A2-dependent RNA granule structures. Furthermore, the RNA helicase DDX5 was confirmed here acting together with hnRNP A2 in cytoplasmic RNA granules and is possibly involved in MBP mRNA granule dynamics.rnLack of non-receptor tyrosine kinase Fyn activity leads to reduced levels of MBP and hypomyelination in the forebrain. The multiadaptor protein p130Cas and the RNA-binding protein hnRNP F were verified here as additional targets of Fyn in oligodendrocytes. The findings point at roles of p130Cas in the regulation of Fyn-dependent process outgrowth and signalling cascades ensuring cell survival. HnRNP F was identified here as a novel constituent of oligodendroglial cytoplasmic RNA granules containing hnRNP A2 and MBP mRNA. Moreover, it was found that hnRNP F plays a role in the post-transcriptional regulation of MBP mRNA and that defined levels of hnRNP F are required to facilitate efficient synthesis of MBP. HnRNP F appears to be directly phosphorylated by Fyn kinase what presumably contributes to the initiation of translation of MBP mRNA at the plasma membrane.rnFyn kinase signalling thus affects many aspects of oligodendroglial physiology contributing to myelination. Post-transcriptional control of the synthesis of the essential myelin protein MBP by Fyn targets is particularly important. Deregulation of these Fyn-dependent pathways could thus negatively influence disorders involving the white matter of the nervous system.rnrn

CD99, glicoproteina di membrana codificata dal gene MIC2, è coinvolta in numerosi processi cellulari, inclusi adesione, migrazione, apoptosi, differenziamento e regolazione del trafficking intracellulare di proteine, in condizioni fisiologiche e patologiche. Nell’osteosarcoma risulta scarsamente espressa ed ha ruolo oncosoppressivo. L’isoforma completa (CD99wt) e l’isoforma tronca (CD99sh), deleta di una porzione del dominio intracellulare, influenzano in modo opposto la malignità tumorale. In questo studio, comparando cellule di osteosarcoma caratterizzate da differenti capacità metastatiche e diversa espressione di CD99, abbiamo valutato la modulazione dei contatti cellula-cellula, la riorganizzazione del citoscheletro di actina e la modulazione delle vie di segnalazione a valle del CD99, al fine di identificare i meccanismi molecolari regolati da questa molecola e responsabili del comportamento migratorio e invasivo delle cellule di osteosarcoma. L'espressione forzata di CD99wt induce il reclutamento di N-caderina e β-catenina a livello delle giunzioni aderenti ed inibisce l'espressione di molecole cruciali nel processo di rimodellamento del citoscheletro di actina, come ACTR2, ARPC1A, Rho-associated, coiled–coil-containing protein kinase 2 (ROCK2), nonché di ezrina, membro della famiglia ezrin/radixin/moesin e chiaramente associata con la progressione tumorale e la metastatizzazione dell’OS. Gli studi funzionali identificano ROCK2 come mediatore fondamentale nella regolazione della migrazione e della diffusione metastatica dell’osteosarcoma. Mantenendo cSRC in una conformazione inattiva, CD99wt inibisce la segnalazione mediata da ROCK2 inducendo una diminuzione dell’ezrina a livello della membrana accompagnata dalla traslocazione in membrana di N-caderina e β-catenina, principali ponti molecolari per il citoscheletro di actina. La ri-espressione di CD99wt, generalmente presente negli osteoblasti, ma perso nelle cellule di osteosarcoma, attraverso l'inibizione dell'attività di cSrc e ROCK2, aumenta la forza di contatto e riattiva i segnali anti-migratori ostacolando l’azione pro-migratoria, altrimenti dominante, dell’ezrina nell’osteosarcoma. Abbiamo infine valutato la funzione di ROCK2 nel sarcoma di Ewing: nonostante il ruolo oncogenico esercitato da CD99, ROCK2 guida la migrazione cellulare anche in questa neoplasia.

Kohlenhydrate dienen nicht nur der Ernährung oder der Stabilisierung von Zellwänden, sondern sie sind auch von essentieller Bedeutung für Zell-Zell-Wechselwirkungen im Organismus. Damit sind sie von besonderem Interesse für die Behandlung von Entzündungen sowie tumorösen Erkrankungen, die auf Wechselwirkungen zwischen bestimmten Kohlenhydraten und Rezeptoren, Selektine genannt, basieren. Die vorliegende Dissertation mit dem Titel „Synthese von polymergebundenen Sialyl-Lewis x-Strukturen und deren Mimetika als Zelladhäsionsinhibitoren für E-, L- und P-Selektin“ befasst sich mit der Synthese und der Modifizierung des natürlich vorkommenden Selektin-Liganden Sialyl-Lewisx und seinen potentiell mimetischen Strukturen auf Basis von Gemischen der an der Bindung beteiligten Kohlenhydrate sowie von sulfatierten Einheiten. Um im Organismus eine verstärkte Bindung an die zu adressierenden Rezeptoren und eine erhöhte Zirkulationsdauer zu erreichen, wurden die geeignet funktionalisierten Liganden mittels reaktiver Polymere auf Basis von Pentafluorphenyl-Reaktivestern im Rahmen einer polymeranalogen Umsetzung an ein definiertes Methacrylamid-Polymer gebunden.Um die biologische Wirksamkeit der synthetisierten Polymere zu überprüfen, wurden Oberflächenplasmonenresonanz-Messungen (SPR-Messungen) durchgeführt. Dabei zeigte sich bei den verschiedenen Selektinen eine unterschiedlich hohe Affinität der Sialyl-Lewis x sowie der mimetischen Polymere. Es konnten bei E-Selektin die geringsten, bei L-Selektin mittlere und bei P-Selektin die höchsten Affinitäten im unteren nanomolaren Bereich festgestellt werden.

Exposition von Endothelzellen mit ionisierender Strahlung (IR) oder Behandlung mit inflammatorischen Zytokinen (z. B. TNFa) induziert über eine Rho-GTPasen abhängige NF-kB-Aktivierung die Expression verschiedener Zelladhäsionsmoleküle, u. a. auch von E-Selektin. E-Selektin vermittelt die Adhäsion von Tumorzellen (TC) an Endothelzellen und ist daher vermutlich an der Extravasation von zirkulierenden Tumorzellen beteiligt. HMG-CoA-Reduktase-Inhibitoren (Statine), welche eine breite klinische Anwendung als Lipidsenker erfahren, sind in der Lage, Rho-GTPasen und die durch sie vermittelten Signalwege zu hemmen. Daher sollten Statine wie Lovastatin auch Zell-Zell-Adhäsionsvorgänge beeinflussen. Die vorliegende Arbeit widmet sich den Mechanismen, mit denen IR und TNF in Endothel- und/oder Tumorzellen pro-adhäsive Faktoren induzieren können und ob diese Effekte durch Lovastatin beeinflussbar sind. Zu diesem Zweck wurde mittels eines ELISA-basierenden Zelladhäsions-Assays die Auswirkung von IR und TNF auf Zell-Zell-Kontakte zwischen humanen Tumorzellen (u. a. Kolonkarzinomzellen (HT29)) und humanen, venösen Nabelschnurendothelzellen (HUVEC) analysiert. Zudem wurden die Effekte einer Lovastatinvorbehandlung von TC und/oder HUVEC auf TC-HUVEC-Adhäsion untersucht. Des Weiteren wurden die Wirkungen des sLex-Mimetikums Glycyrrhizin und des Rac1-spezifischen „small-molecule“ Inhibitors NSC23766 auf TC-HUVEC-Adhäsion überprüft. Zusätzlich wurde die strahleninduzierbare mRNA-Expression von diversen Zelladhäsionsmolekülen, Metastasierungsfaktoren und DNA-Reparatur-Genen mittels qRT-PCR (Real-Time Analysen) quantitativ erfasst. Um die erhaltenen in vitro Ergebnisse auch in vivo zu bestätigen, untersuchten wir den Effekt einer Ganzkörperbestrahlung (TBI) von BALB/c-Mäusen auf die Expression von pro-adhäsiven Faktoren. Zur Analyse der Tumorzell-Extravasation wurden Tumorzellen in die laterale Schwanzvene immundefizienter Mäuse injiziert und anschließend eine Ganzkörperbestrahlung durchgeführt (4 Gy). Nach einer Wartezeit von 4 Wochen wurde ein erhöhtes Auftreten von Lungenmetastasen beobachtet, welches durch Vorbehandlung der Tiere mit Statinen, NSC23766 oder Glycyrrhizin blockiert werden konnte. Zusammenfassend konnte somit ein Einfluss von IR auf die Expression verschiedener Zelladhäsionsmoleküle in vitro und auf die Extravasation zirkulierender Tumorzellen in vivo festgestellt werden. Diese pro-metastatischen Strahleneffekte konnten durch pharmakologische Hemmung Rho-regulierter Signalwege abgeschwächt werden.

Das Chemokin CXCL12 (auch bekannt als SDF-1) ist ein kleines Protein (8-14) KDa, das in sechs Isoformen exprimiert wird (SDF-1α, SDF-1β, SDF-1γ, SDF- 1δ, SDF-1ε und SDF-1θ) von einem einzigen Gen, dass die Leukozyten-Wanderung regelt und variabel in einer Reihe von normalen und Krebsgeweben exprimiert wird.rnCXCL12 spielt verschiedene Rollen in der Tumorpathogenese. Es wurde nachgewiesen, dass CXCL12 das Tumorwachstum und die Malignität fördert, die Tumorangiogenese stärkt, sich an der Metastasierung beteiligt und zu immunsuppressiven Netzwerken innerhalb des Tumormikromilieus beiträgt. Daher liegt es nahe, dass der CXCL12/CXCR4-Signalweg ein wichtiges Ziel ist für die Entwicklung von neuartigen Krebstherapien.rnUm Licht auf die Rolle der Chemokin CXCL12 Splicevarianten in der Entwicklung von Krebs zu werfen und die mögliche physiologische Relevanz und ihre möglichen funktionellen Unterschiede bei Darmkrebs zu verstehen, haben wir alle CXCL12 Splicevarianten (alpha, beta, gamma, delta, epsilon und theta) in die kolorektalen Zelllinie SW480 und die Melanomzellinie D05 transfiziert und exprimiert.rnrnDiese Arbeit wurde erstellt, um die folgenden Ziele zu erreichen. Untersuchung der Rolle von CXCL12 Splicevarianten bei der Vermittlung von Tumorprogression, Adhäsion, Migration, Invasion und Metastasierung von Darmkrebs. Untersuchung, ob die CXCL12 Variantenwege ein wichtiges Ziel für die Entwicklung von Krebstherapien darstellen.rn• Um eine in vivo Mausmodell zu entwickeln, um die Rolle der CXCL12 Varianten im Rahmen des Tumorwachstums zu verstehen.rnrnUnsere Ergebnisse zeigen, dass:Der CXCL12 G801A Polymorphismus ist ein Low-Penetranz Risikofaktor für die Entwicklung von Darmkrebs. Der CXCL12-Gen-Polymorphismus rs1801157 ist mit dem T-Status (Tumor-node-Metastasen) assoziiert. Es gab keine Beziehung zwischen CXCL12-Gen-Polymorphismus rs1801157 und Fernmetastisen oder LN metastasen. Alle sechs CXCL12 Splicevarianten werden im Darmkrebs und in gesunder Kolon mucosa exprimiert. Die höchste Expression wird bei SDF-1alpha, dann SDF-1 beta gefunden. Alle sechs CXCL12 Varianten zeigen erhöhte Tumorzellproliferation in vitro. SDF-1beta, gefolgt von SDF-1alpha zeigte die größte Aktivität im Proliferationsassay.rn• Alle sechs CXCL12 Varianten induzieren die Tumorzelladhäsion.SDF-1beta dann SDF-1alpha zeigte die größte Aktivität im Rahmen des Adhäsionsassay. Alle sechs CXCL12 Varianten erhöhten die Zellmigration und Invasion von Tumorzellen in vitro. SDF-1theta und SDF-1epsilon 1theta zeigten die größte Aktivität, während die schwächste Aktivität mit SDF-1alpha und SDF-1beta beobachtet wurde. Alle sechs CXCL12 Varianten aktivieren Akt und (MAPK) Mitogen- acktivatedierte Protein kinase Wege und damit die Regulierung viele essentieller Prozesse in Tumorzellen, wie Proliferation, Migration, Invasion und Adhäsion. Es ist interessant festzustellen, dass AMD3100 die CXCL12 Splicevarianten inhibriert, die AKT-MEK-1/2-Phosphorylierung induzieren.rnDer Inhibitor AMD3100 unterdrückt stark die CXCL12 Varianten -delta, -epsilon und theta-und unterdrückt schwach CXCL12-gamma. während es keine signifikante Wirkung auf CXCL12-alpha und beta hatte. Es hat möglicherweise Auswirkungen auf mehrere große Signalwage in Bezug auf Proliferation, Migration und Invasions.rn• Es ist wichtig anzumerken, dass die Hemmung von CXCL12-Varianten durch AMD3100 einen der möglichen Ansaätze in der Krebstherapie darstellen kann.Wir schlagen vor, dass weitere Studien erwogen werden, die wir brauchen, um die biologische Aktivität dieser neuen CXCL12 Varianten bei verschiedenen Arten von Krebs klar zu verstehen.

Resistance of cancer cells towards chemotherapy is the major cause of therapy failure. Hence, the evaluation of cellular defense mechanisms is essential in the establishment of new chemotherapeutics. In this study, classical intrinsic and acquired as well as new resistance mechanisms relevant in the cellular response to the novel vacuolar H+-ATPase inhibitor archazolid B were investigated. Archazolid B, originally produced by the myxobacterium Archangium gephyra, displayed cytotoxicity in the low nanomolar range on a panel of cancer cell lines. The drug showed enhanced cytotoxic activity against nearly all cancerous cells compared to their non-cancerous pendants. With regards to ABC transporters, archazolid B was identified as a moderate substrate of ABCB1 (P-glycoprotein) and a weak substrate of ABCG2 (BCRP), whereas hypersensitivity was observed in ABCB5-expressing cells. The cytotoxic effect of archazolid B was shown to be independent of the cellular p53 status. However, cells expressing constitutively active EGFR displayed significantly increased resistance. Acquired drug resistance was studied by establishing an archazolid B-resistant MCF-7 cell line. Experiments showed that this secondary resistance was not conferred by aberrant expression or DNA mutations of the gene encoding vacuolar H+-ATPase subunit c, the direct target of archazolid B. Instead, a slight increase of ABCB1 and a significant overexpression of EGFR as well as reduced proliferation may contribute to acquired archazolid B resistance. For identification of new resistance strategies upon archazolid B treatment, omics data from bladder cancer and glioblastoma cells were analyzed, revealing drastic disturbances in cholesterol homeostasis, affecting cholesterol biosynthesis, uptake and transport. As shown by filipin staining, archazolid B led to accumulation of free cholesterol in lysosomes, which triggered sterol responses, mediated by SREBP-2 and LXR, including up-regulation of HMGCR, the key enzyme of cholesterol biosynthesis. Furthermore, inhibition of LDL uptake as well as impaired LDLR surface expression were observed, indicating newly synthesized cholesterol to be the main source of cholesterol in archazolid B-treated cells. This was proven by the fact that under archazolid B treatment, total free cholesterol levels as well as cell survival were significantly reduced by inhibiting HMGCR with fluvastatin. The combination of archazolid B with statins may therefore be an attractive strategy to circumvent cholesterol-mediated cell survival and in turn potentiate the promising anticancer effects of archazolid B.

Im Fokus dieser Studie stehen die zu den Gliazellen zählenden OPC, sowie das von diesen exprimierte Typ-1 Membranprotein NG2. Dieses wird auf eine Prozessierung durch α- und γ-Sekretase, in Analogie zu Proteinen wie Notch oder APP, untersucht.rnEine solche Prozessierung ginge mit zusätzlichen intrazellulären Spaltprodukten neben der bekannten Ektodomäne einher. Da OPC mit dem Neuronalen Netzwerk durch synaptische Innervierungen in Verbindung stehen, stellt sich die Frage, ob diese mit der Spaltung von NG2 in Verbindung gebracht werden können. Dazu käme mechanistisch beispielsweise eine aktivitätsabhängige Regulierung der Proteolyse, wie sie jüngst für das neuronale synaptische cell adhesion molecule Neuroligin gezeigt werden konnte, in Frage. Zudem werden eine physiologische Rolle der NG2 Ektodomäne bzw. der möglichen intrazellulären Fragmente untersuchen. Insbesondere potentielle neuromodulatorische Funktionen sind hier von Interesse, da diese die OPC tiefer in das Neuronale Netzwerk integrieren würden. Die Existenz eines NG2 Homologes in D. melanogaster, wirft weiterhin die Frage auf, in wie weit diese Mechanismen in diesem Modellsystem konserviert sind.rnIn Analogie zur Lokalisierung von Markerproteinen an Neuron-Neuron Synapsen in vivo, ergibt sich die Frage ob sich die synaptischen Verbindungen zwischen Neuronen und OPC in ähnlicher Weise darstellen lassen.rnEin Charakteristikum von OPC ist die Teilungsaktivität in sich entwickelnden und adulten Säugern. Zudem gibt es Evidenzen für direkte funktionelle Verknüpfungen zwischen dem NG2 Protein und dem Teilungsmodus der OPC. Deshalb war ein weiteres Ziel mögliche Änderungen in der Zellteilung der OPC, die mit dem NG2 Protein in Verbindung stehen könnten, in NG2 -/- Mäusen zu untersuchen.rn

Although it is well established that stromal intercellular adhesion molecule-1 (ICAM-1), ICAM-2, and vascular cell adhesion molecule-1 (VCAM-1) mediate lymphocyte recruitment into peripheral lymph nodes (PLNs), their precise contributions to the individual steps of the lymphocyte homing cascade are not known. Here, we provide in vivo evidence for a selective function for ICAM-1 > ICAM-2 > VCAM-1 in lymphocyte arrest within noninflamed PLN microvessels. Blocking all 3 CAMs completely inhibited lymphocyte adhesion within PLN high endothelial venules (HEVs). Post-arrest extravasation of T cells was a 3-step process, with optional ICAM-1-dependent intraluminal crawling followed by rapid ICAM-1- or ICAM-2-independent diapedesis and perivascular trapping. Parenchymal motility of lymphocytes was modestly reduced in the absence of ICAM-1, while ICAM-2 and alpha4-integrin ligands were not required for B-cell motility within follicles. Our findings highlight nonredundant functions for stromal Ig family CAMs in shear-resistant lymphocyte adhesion in steady-state HEVs, a unique role for ICAM-1 in intraluminal lymphocyte crawling but redundant roles for ICAM-1 and ICAM-2 in lymphocyte diapedesis and interstitial motility.

To test the hypothesis that the pericellular fibronectin matrix is involved in mechanotransduction, we compared the response of normal and fibronectin-deficient mouse fibroblasts to cyclic substrate strain. Normal fibroblasts seeded on vitronectin in fibronectin-depleted medium deposited their own fibronectin matrix. In cultures exposed to cyclic strain, RhoA was activated, actin-stress fibers became more prominent, MAL/MKL1 shuttled to the nucleus, and mRNA encoding tenascin-C was induced. By contrast, these RhoA-dependent responses to cyclic strain were suppressed in fibronectin knockdown or knockout fibroblasts grown under identical conditions. On vitronectin substrate, fibronectin-deficient cells lacked fibrillar adhesions containing alpha5 integrin. However, when fibronectin-deficient fibroblasts were plated on exogenous fibronectin, their defects in adhesions and mechanotransduction were restored. Studies with fragments indicated that both the RGD-synergy site and the adjacent heparin-binding region of fibronectin were required for full activity in mechanotransduction, but not its ability to self-assemble. In contrast to RhoA-mediated responses, activation of Erk1/2 and PKB/Akt by cyclic strain was not affected in fibronectin-deficient cells. Our results indicate that pericellular fibronectin secreted by normal fibroblasts is a necessary component of the strain-sensing machinery. Supporting this hypothesis, induction of cellular tenascin-C by cyclic strain was suppressed by addition of exogenous tenascin-C, which interferes with fibronectin-mediated cell spreading.

Cleft palate is a common birth defect in humans. Elevation and fusion of paired palatal shelves are coordinated by growth and transcription factors, and mutations in these can cause malformations. Among the effector genes for growth factor signaling are extracellular matrix (ECM) glycoproteins. These provide substrates for cell adhesion (e.g., fibronectin, tenascins), but also regulate growth factor availability (e.g., fibrillins). Cleft palate in Bmp7 null mouse embryos is caused by a delay in palatal shelf elevation. In contrast, palatal shelves of Tgf-β3 knockout mice elevate normally, but a cleft develops due to their failure to fuse. However, nothing is known about a possible functional interaction between specific ECM proteins and Tgf-β/Bmp family members in palatogenesis. To start addressing this question, we studied the mRNA and protein distribution of relevant ECM components during secondary palate development, and compared it to growth factor expression in wildtypewild type and mutant mice. We found that fibrillin-2 (but not fibrillin-1) mRNA appeared in the mesenchyme of elevated palatal shelves adjacent to the midline epithelial cells, which were positive for Tgf-β3 mRNA. Moreover, midline epithelial cells started expressing fibronectin upon contact of the two palatal shelves. These findings support the hypothesis that fibrillin-2 and fibronectin are involved in regulating the activity of Tgf-β3 at the fusing midline. In addition, we observed that tenascin-W (but not tenascin-C) was misexpressed in palatal shelves of Bmp7-deficient mouse embryos. In contrast to tenascin-C, tenascin-W secretion was strongly induced by Bmp7 in embryonic cranial fibroblasts in vitro. These results are consistent with a putative function for tenascin-W as a target of Bmp7 signaling during palate elevation. Our results indicate that distinct ECM proteins are important for morphogenesis of the secondary palate, both as downstream effectors and as regulators of Tgf-β/Bmp activity.
