Summary : A large body of evidence indicates that the innate immune system plays a key role in host response to viral infection. Recently, Toll-like receptors (TLRs), RIG-I-like receptors (RLRs), and NOD-like receptor receptors (NLRs) have emerged as key innate immune sensors of microbial products, eliciting intracellular signaling and leading to the production of chemokines, cytokines and interferons (IFNs) that shape innate immune responses and coordinate the development of adaptive immunity. Poxviruses are currently developed as vaccines vectors for infectious diseases such as HIV, tuberculosis and malaria. Modified vaccinia virus Ankara (MVA) and New York vaccinia virus (NWAC) are attenuated, replication deficient strains of poxvirus. The mechanisms underlying innate immune responses to MVA and NYVAC are poorly characterized. Thus, the objectives of the project were to determine the innate immune profile stimulated by poxviruses in innate immune cells and to evaluate the impact of modifications in the viral genome on MVA and NYVAC immunogenicity. MVA stimulated the production of abundant amounts of chemokines and IFNß but low levels of cytokines by human macrophages. In contrast, NYVAC weakly stimulated the production of all mediators. Interestingly, MVA and NYVAC strongly stimulated innate immune responses in vivo and in human whole blood, suggesting that a soluble factors}, possibly a complement component, was required for optimal activation of innate immune cells by poxviruses. Modified MVA and NYVAC produced by single or multiple deletions of viral genes targeting crucial pathways of host innate immunity, and mutant poxviruses with limited replication capacity, increased the production of pro-inflammatory molecules by human whole blood. Gene expression profiling in human macrophages confirmed the increased immunologic stimulatory capacity of modified poxviruses. The pathways activated by MVA and NYVAC in innate immune cells were described by analysing the response of knockdown or shRNA transduced macrophages with impaired expression of TLRs and their adaptors (MyD8$ and TRIF), RLRs (RIG-I, MDA-5 and the adaptor IPS-1) and the NALP3 inflammasome composed óf the NLR NALP3, caspase-1 and ASC. These experiments revealed a critical role for TLR2-TLR6-MyD88 in the production of tFNß-independent chemokines and of MDA-5-IPS-1 in the production of IFNß and IFNßdependent chemokines. The transcription of the iL1b gene encoding for the IL-1ß cytokine was initiated through TLR2-MyD88, whereas the maturation and the secretion of IL-1ß were controlled by the NALP3 inflammasome. Finally, we analyzed the role of macrophage migration inhibitory factor (MIF), a mediator of inflammation and innate immune responses, in MVA infection. We observed that MVA infection increased MIF production by innate immune cells and that MIF deficiency impaired macrophage and dendritic cell responses (ie migration, maturation, cytokine and IFN production) to MVA infection in vitro and in vivo. Moreover, MIF-deficiency resulted in delayed anti-MVA specific antibody production in mice immunized with the virus. In conclusion, we demonstrate. that poxviruses can be modified genetically to improve their immunogenicity. We also report the first comprehensive analysis of poxvirus sensing by innate immune cells, showing that the TLR, RLR and NLR pathways play specific and coordinated roles in regulating cytokine, chemokine and IFN response to poxvirus infection. Finally, we show that MIF is an integral host component involved in innate and adaptive immune responses to MVA infection. The present findings provide important information relevant to the study of the pathogenesis of poxvirus infections and allow a better understanding of the immunogenic potential of vaccine vectors, which is required for the development of optimized modìfied pox-vaccine vectors.

A identificação de dois recipientes colocados ritualmente em posição invertida, conotáveis com a derradeira fase de ocupação do povoado calcolítico do Outeiro Redondo (Sesimbra), e deste modo com uma cerimónia de abandono do próprio sítio arqueológico, conduziu à procura de exemplos comparáveis. Tal procura estendeu‑se, depois, às evidências de utilização ritual, em contextos habitacionais, de recipientes cerâmicos, desde a época a que pertencem os exemplares em causa, até ao Período Romano. Teve‑se ainda em consideração o contributo da Etnografia e da Arqueologia africana, através da experiência do signatário, relacionada com intervenção arqueológica que dirigiu em unidade doméstica do século XVII/XVIII da ilha de São Vicente (República de Cabo Verde). Sem se pretender assumir a continuidade desta prática ritual no actual território português, desde o Calcolítico até pelo menos à Época Romana, por poder tratar‑se de uma simples convergência, recorrentemente verificada ao longo da Idade do Bronze e do Ferro. Como se deverá interpretar o achado do Outeiro Redondo? As evidências assinaladas através do uso ritual de recipientes cerâmicos em contextos habitacionais podem ter um significado ligado ao próprio quotidiano, como no povoado de Fraga da Pena (Fornos de Algodres). Aquelas afiguram‑se, contudo, mais abundantes e expressivas no que se refere quer às práticas de natureza fundacional, quer às relacionadas com o abandono; por vezes, dada a presença de aves sacrificadas, tais rituais poderiam também revestir uma função propiciatória, que não é incompatível com as duas funções mencionadas, também elas não incompatíveis entre si.

Apresentam‑se os principais resultados obtidos nas duas missões arqueológicas patrocinadas pelo Centro Português de Actividades Subaquáticas (CPAS) à ilha de São Vicente (República de Cabo Verde), em 1998 e em 2005. Em 1998, confirmou‑se o efectivo interesse arqueológico do sítio, localizado sobre o mar, em local abrigado da vasta baía de Salamansa, situada na parte setentrional da ilha, tendo‑se registado a respectiva extensão e estratigrafia e procedido à colheita de amostras para datação. Embora os resultados dessa campanha tivessem sido publicados, indicando estação de carácter habitacional, revelada pela notável acumulação de conchas, acompanhada de abundantes fragmentos de cerâmicas manuais, de produção africana, mantinha‑se indefinida a sua verdadeira natureza. Impunha‑se, assim, proceder à escavação integral da área que ainda subsistia da estação — sujeita de forma contínua a forte erosão marinha — bem como à colheita de novos materiais para datação, de forma a confirmar as conclusões preliminares anteriormente obtidas, objectivos que se concretizaram em 2005. Deste modo, foi possível concluir que, contrariando a hipótese, de início considerada, de poder corresponder a um testemunho da ocupação da ilha em época anterior à chegada dos Portugueses — hipótese que já as primeiras datas de radiocarbono contradiziam — se trata de um sítio onde uma unidade habitacional construída por muros de pedra seca, de planta ortogonal, revela inspiração europeia, aliás sublinhada pelos materiais exumados, onde estão representados produtos com tal origem, como cachimbos de caulino, vidros, faianças portuguesas, e projécteis de armas de fogo, a par de objectos oriundos do Extremo Oriente, num quadro dominado pelas produções cerâmicas africanas. Esta situação evidencia um estabelecimento cuja ocupação se centrou no século XVII, conforme indicam os materiais recolhidos e os resultados das datações obtidas, francamente aberto aos contactos de longa distância, apesar do isolamento do local escolhido. Os restos faunísticos recolhidos, com a presença deburro e de boi, sugerem um estacionamento sedentário, sendo a alimentação assegurada essencialmente pela captura de tartarugas, pela pesca e pela recolecção de moluscos marinhos (especialmente grandes lapas) e complementada pelo consumo de cabra, que poderia ser doméstica ou caçada, dado o estado selvagem a que retornou ali esta espécie. na última parte do trabalho, discutem‑se as diversas hipóteses susceptíveis de explicar esta estação — desde um entreposto comercial relacionado com a exploração agro‑pecuária da ilha de Santo Antão, passando por pequeno estabelecimento especializado de apoio à navegação, com a produção de carne salgada de tartaruga, até ter constituído refúgio relacionado com a intensa pirataria vigente à época no arquipélago, tendo presente os elementos históricos conhecidos, que, aliás, indicam que o início da ocupação permanente de São Vicente só se produziu a partir da segunda década do século XIX. Seja como for, a forte componente cultural africana revelada pelo espólio destes primeiros ocupantes da ilha expressa‑se também pelos rituais que terão envolvido o abandono do estabelecimento, com o enterramento de dois vasos emborcados sob o chão da habitação explorada, e a deposição de uma pequena taça, nas mesmas circunstâncias, junto à parede da mesma, do lado externo.

The aims of this thesis were to better characterize HIV-1 diversity in Portugal, Angola, Mozambique and Cape Verde and to investigate the origin and epidemiological history of HIV-1 in these countries. The impact of these issues in diagnosis, disease progression and susceptibility to ARV therapy was also investigated. Finally, the nature, dynamics and prevalence of transmitted drug resistance (TDR) was determined in untreated HIV-1 infected patients. In Angola, practically all HIV-1 genetic forms were found, including almost all subtypes, untypable (U) strains, CRFs and URFs. Recombinants (first and second generation) were present in 47.1% of the patients. HIV/AIDS epidemic in Angola probably started in 1961, the major cause being the independence war, subsequently spreading to Portugal. In Maputo, 81% of the patients were infected with subtype C viruses. Subtype G, U and recombinants such as CRF37_cpx, were also present. The results suggest that HIV-1 epidemic in Mozambique is evolving rapidly in genetic complexity. In Cape Verde, where HIV-1 and HIV-2 co-circulate, subtype G is the prevailed subtype. Subtypes B, C, F1, U, CRF02_AG and other recombinant strains were also found. HIV-2 isolates belonged to group A, some being closely related to the original ROD isolate. In all three countries numerous new polymorphisms were identified in the RT and PR of HIV-1 viruses. Mutations conferring resistance to the NRTIs or NNRTIs were found in isolates from 2 (2%) patients from Angola, 4 (6%) from Mozambique and 3 (12%) from Cape Verde. None of the isolates containing TDR mutations would be fully sensitive to the standard first-line therapeutic regimens used in these countries. Close surveillance in treated and untreated populations will be crucial to prevent further transmission of drug resistant strains and maximize the efficacy of ARV therapy. In Portugal, investigation of a seronegative case infection with rapid progression to AIDS and death revealed that the patient was infected with a CRF14_BG-like R5-tropic strain selectively transmitted by his seropositive sexual partner. The results suggest a massive infection with a highly aggressive CRF14_BG like strain and/or the presence of an unidentified immunological problem that prevented the formation of HIV-1-specific antibodies. Near full-length genomic sequences obtained from three unrelated patients enabled the first molecular and phylogenomic characterization of CRF14_BG from Portugal; all sequences were strongly related with CRF14_BG Spanish isolates. The mean date of origin of CRF14_BG was estimated to be 1992. We propose that CRF14_BG emerged in Portugal in the early 1990s, spread to Spain in late 1990s as a consequence of IDUs migration and then to the rest of Europe. Most CRF14_BG strains were predicted to use CXCR4 and were associated with rapid CD4 depletion and disease progression. Finally, we provide evidence suggesting that the X4 tropism of CRF14_BG may have resulted from convergent evolution of the V3 loop possibly driven by an effective escape from neutralizing antibody response.

Several marine deposits in the southern Costa Brava shoreline have been studied. They appear at different heights above and below the present sea level. Three groups are defined in relation to their origin: conglomerate levels at the bottom of the cliffs, not-cementedemerged beaches and cemented submerged beaches. The age of the emerged beaches has been accurately determined by means of the ceramic content and radiocarbon dating. Chronological succession of the deposits and their stratigraphic and paleontological characteristics allow to define a sequence of the sea level changes during the latest stages of the versiliane transgression in the studied area

The chemical resistance of ceramic tiles is the subject of the European Standard UNE-EN ISO 10545-13. In order to evaluate the effect of aqueous solutions of several chemicals agents on the aspect of the tile surface, this standard establishes a series of tests at room temperature followed by visual inspection. According to this standard the tiles of this study are classified as being of maximum resistance (UHA). However operating conditions can be more aggressive than those detailed in the standard. So, a systematic study has been undertaken. In the present work, the effect of aqueous solutions of several organic and inorganic acids on the tile surface is evaluated. Samples immersed in different solutions are subjected to the following conditions: T= 60º C; pH=2 and to agitation processes. Visual analysis, as well as optical microscopy and scanning electron microscopy (SEM) were performed in order to determine the possible variation of the superficial aspect of tiles. Moreover, atomic absorption spectrophotometry has been used in order to obtain quantitative information concerning the solubility of system M (III)-L (M= Fe; L= H2O or L= ligand). The results obtained show, in all cases, a progressive dissolution of iron oxide precipitates presents in the ceramic body

L'Institut de Police Scientifique (IPS) de Lausanne a développé et emploie une méthode de profilage de l'héroïne qui permet d'établir des liens entre les saisies de différentes affaires. La comparaison s'appuie sur des caractéristiques chimiques. Au cours du trafic, les spécimens sont transportés, stockés et dilués. Leurs caractéristiques chimiques sont alors susceptibles de varier. L'objectif de ce travail consiste à évaluer les conséquences d'une dilution importante sur l'évolution des liens chimiques établis, puis d'observer l'influence de la température et de l'humidité sur le profil chimique d'échantillons d'héroïne placés dans des conditions de vieillissement accéléré. On remarque que la dilution n'influence pas directement le profilage, mais l'inférence d'une source commune entre les spécimens dépend de la différence entre les concentrations des échantillons comparés. Dans le cadre de l'étude de vieillissement, tous les échantillons ont pu être liés et aucune variation significative du profil n'a été constatée. Cependant, les quantités de certains composés ont plus tendance à varier et pourraient être à l'origine d'une modification du profil à long terme.

Recientes investigaciones en el campo de los materiales cerámicos han dado cuenta de la importancia de la metaestabilidad para obtener estructuras con características singulares. Durante la consolidación del material las fases mestaestables se transforman en una estructura donde se produce la inhibición del crecimiento de grano. Este efecto es una consecuencia directa de la inmiscibilidad de dos fases en estado sólido. Los nanocomposites conseguidos, gracias a su pequeño tamaño de grano y a su estructura uniforme, exhiben unas interesantes propiedades como elevada dureza y tenacidad. Estas fases metaestables pueden ser producidas por diversas técnicas entre las que se encuentra la proyección térmica. En concreto en este trabajo se ha empleado la Proyección por plasma (APS). Las fases de partida inmiscibles, son fundidas y homogeneizadas durante su corta estancia en la zona caliente del plasma. Seguidamente, las partículas fundidas y aceleradas por el plasma, se someten a un enfriamiento rápido o temple (quenching) en un medio líquido, como el agua o en un substrato enfriado con nitrógeno líquido, formándose a través de este proceso las fases metaestables. El principal objetivo de este trabajo ha sido la obtención de polvos cerámicos metastables a través de la aplicación de APS y el establecimiento de un proceso de temple conducente a la formación de fases metastables así como la caracterización estructural de éstas. Como última etapa del trabajo se han estudiado los materiales nanoestructurados conseguidos tras realizar tratamientos térmicos.

Este trabajo tiene por objeto la caracterizaci6n mineralbgica y fsico-qumica de las arcillas terciarias explotadas en el yacimiento de Sitjar, cercano a la localidad de Onda (Castellbn), utilizadas como materia prima por la industria ceramica castellonense. Se estudian tambitn algunas propiedades ceramicas de estas arcillas, incidiendo en el comportamiento ttrmico, para su correcta aplicaci6n. La composici6n qumica y mineral6gica posibilita su utilizaci6n como aditivo para la fabricaci6n de materiales porosos y refractarios.

This year the development of our project can be divided into two main clearly different parts, on one hand the laboratory work, where the sampled ceramic individuals has been prepared and analyzed and the elaboration of the data obtained during the excavation of 2007 has been finished and, on the other hand, the field work developed at the archaeological site during this specific year (2008).In the mark of the analytical work a significant number of ceramic individuals (144) from the different stratigraphical units from various areas of the excavation of Termez and Tchinguiz Tepe sampled duringthe new and previous field works has been archaeometrically characterized. This specific material includedindividuals dated into the Hellenistic and Sassanian period, which has been confirmed by C14dating upon organic samples.At the same time, in the mark of the field work of 2008 the archaeological record, already started tobe under study during the excavation of 2007, has been completed and two new archaeological recordshave been registered on of which is located in the area of Tchinguiz Tepe. For the archeological studythe information of the previous geophysical prospecting has been indisputably taken into considerationand the same methodology has been applied to crosscheck the latter archaeological results.

Tobacco farmers of southern Brazil use high levels of fertilizers, without considering soil and environmental attributes, posing great risk to water resources degradation. The objective of this study was to monitor nitrate and ammonium concentrations in the soil solution of an Entisol in and below the root zone of tobacco under conventional tillage (CT), minimum tillage (MT) and no-tillage (NT). The study was conducted in the small-watershed Arroio Lino, in Agudo, State of Rio Grande do Sul, Brazil. A base fertilization of 850 kg ha-1 of 10-18-24 and topdressing of 400 kg ha-1 of 14-0-14 NPK fertilizer were applied. The soil solution was sampled during the crop cycle with a tension lysimeter equipped with a porous ceramic cup. Ammonium and nitrate concentrations were analyzed by the distillation and titration method. Nitrate concentrations, ranging from 8 to 226 mg L-1, were highest after initial fertilization and decreased during the crop cycle. The average nitrate (N-NO3-) concentration in the root zone was 75 in NT, 95 in MT, and 49 mg L-1 in CT. Below the root zone, the average nitrate concentration was 58 under NT, 108 under MT and 36 mg L-1 under CT. The nitrate and ammonium concentrations did not differ significantly in the management systems. However, the nitrate concentrations measured represent a contamination risk to groundwater of the watershed. The ammonium concentration (N-NH4+) decreased over time in all management systems, possibly as a result of the nitrification process and root uptake of part of the ammonium by the growing plants.

The outcome of a viral infection depends on the interplay between the host's capacity to trigger potent antiviral responses and viral mechanisms that counteract them. Although Toll-like receptor (TLR)-3, which recognizes virally derived double-stranded (ds) RNA, transmits downstream antiviral signaling through the TIR adaptor Trif (TICAM-1), viral RNA-sensing RIG-like helicases (RLHs) use the mitochondrial-bound CARD protein Cardif (IPS-1/MAVS/VISA). The importance of these two antiviral signaling pathways is reflected by the fact that both adaptors are inhibited through specific cleavage triggered by the hepatitis C virus serine protease NS3-4A. Here, we show that inactivation can also occur through cellular caspases activated by various pro-apoptotic signals. Upon caspase-dependent cleavage both adaptors loose their capacity to activate the transcription factors interferon regulatory factors (IRF) and NF-kappaB. Importantly, poliovirus infection triggers a caspase-dependent cleavage of Cardif, suggesting that some viruses may activate caspases not only as a mean to facilitate shedding and replication, but also to impair antiviral responses

Quantitative approaches in ceramology are gaining ground in excavation reports, archaeological publications and thematic studies. Hence, a wide variety of methods are being used depending on the researchers' theoretical premise, the type of material which is examined, the context of discovery and the questions that are addressed. The round table that took place in Athens on November 2008 was intended to offer the participants the opportunity to present a selection of case studies on the basis of which methodological approaches were discussed. Many cell types are currently being studied as potential sources of cardiomyocytes for cell transplantation therapy to repair and regenerate damaged myocardium. The question remains as to which progenitor cell represents the best candidate. Bone marrow-derived cells and endothelial progenitor cells have been tested in clinical studies. These cells are safe, but their cardiogenic potential is controversial. The functional benefits observed are probably due to enhanced angiogenesis, reduced ventricular remodeling, or to cytokine-mediated effects that promote the survival of endogenous cells. Human embryonic stem cells represent an unlimited source of cardiomyocytes due to their great differentiation potential, but each step of differentiation must be tightly controlled due to the high risk of teratoma formation. These cells, however, confront ethical barriers and there is a risk of graft rejection. These last two problems can be avoided by using induced pluripotent stem cells (iPS), which can be autologously derived, but the high risk of teratoma formation remains. Cardiac progenitor cells have the advantage of being cardiac committed, but important questions remain unanswered, such as what is the best marker to identify and isolate these cells? To date the different markers used to identify adult cardiac progenitor cells also recognize progenitor cells that are outside the heart. Thus, it cannot be determined whether the cardiac progenitor cells identified in the adult heart represent resident cells present since fetal life or extracardiac cells that colonized the heart after cardiac injury. Developmental studies have identified markers of multipotent progenitors, but it is unknown whether these markers are specific for adult progenitors when expressed in the adult myocardium. Cardiac regeneration is dependent on the stability of the cells transplanted into the host myocardium and on the electromechanical coupling with the endogenous cells. Finally, the promotion of endogenous regenerative processes by mobilizing endogenous progenitors represents a complementary approach to cell transplantation therapy.
