Aim. To investigate the biomechanical effects of zoledronic acid (ZA) on femurs of female osteoporotic rats after follow-up periods of 9 and 12 months. Methods. Eighty female Wistar rats were prospectively assessed. At 60 days of age, the animals were randomly divided into two groups: bilateral oophorectomy (O) (n=40) and sham surgery (S) (n=40). At 90 days of age, groups O and S were randomly subdivided into four groups, according to whether 0.1 mg/kg of ZA or distilled water (DW) was intraperitoneally administered: OZA (n=20), ODW (n=20), SZA (n=20), and SDW (n=20). The animals were sacrificed at 9 and 12 months after the administration of the substances, and then their right femurs were removed and analyzed biomechanically. Axial compression tests that focused on determining the maximum load (N), yield point (N), and stiffness coefficient (N/mm) of the proximal femur were performed in the biomechanical study. Results. ZA significantly increased the maximum load and yield point, reducing the stiffness coefficient concerning the oophorectomy status and follow-up period. Conclusion. Zoledronic acid, at a dose of 0.1 mg/kg, significantly increased the maximum loads and yield points and reduced the stiffness coefficients in the femurs of female rats with osteoporosis caused by bilateral oophorectomy. © 2012 Evandro Pereira Palacio et al.

This study aimed to validate the enzyme immunoassay (EIA) for fecal progestin quantification of the species Mazama americana, define its excretion profile during periods of gestation and postpartum and determine the gestation period and resumption of postpartum ovarian activity in this species in captivity Fecal samples were collected twice a week during gestation and every day in the postpartum period, and analyzed using EIA The mean concentrations (±SEM) of fecal progestins during gestation were 2180.0 ± 299.1 ng/g in early pregnancy (week 1-11), 3271.4 ± 406.9 ng/g in middle pregnancy (week 12-22) and 5592.0 ± 1125.8 ng/g in late pregnancy (week 23-32) The gestation period determined for the species was 220.9 ± 1.2 days The concentration of progestins reached its peak prior to parturition and returned to baseline levels in 4 ± 0.31 days after parturition In the postpartum period, the mean concentrations of fecal progestins were 1564.2 ± 182.6 ng/g in the interval between parturition and resumption of ovarian activity, 469.8 ± 24.5 ng/g in the inter-luteal phase and 2401.7 ± 318.5 ng/g during the luteal phase, such that the postpartum period and the luteal phase differed from the inter-luteal phase Fecal progestin profiling permitted the detection of ovulation 26.9 ± 3.4 days after parturition in all the hinds studied and estimation of the mean duration of the estrous cycle, 21.3 ± 1.1 days Analysis established that concentrations of progestins above 3038.76 ng/g diagnosed pregnancy, a value determined from the week 12 of gestation Moreover, the quantification of fecal progestins by EIA proved to be an important tool for noninvasive endocrine monitoring and to obtain reproductive data on the species M americana in captivity © 2013 Elsevier B.V.

Objectives: This study aimed to evaluate the characteristics of the brain and vascular indicesof the middle cerebral artery of canine foetuses. Methods: Twenty-five bitches were selected. Tissue development, echogenicity, echotexture and brain echobiometric data were studied, and the major structures were identified between the 5th and 8th gestational weeks. The area and volume of the brain mass (BMA and BMV), cranial area and volume (AC and VC), brain mass index (BMI) and brain volume index (BVI) were determined. A single ultrasound examination was performed during each studied week (6th, 7th and 8th). Doppler ultrasonography was performed to assess the maximum and minimum velocity, resistance and pulsatility index of middle cerebral artery of the foetuses. Results: Echoencephalography was performed to evaluate the morphological characteristics of the central nervous system. Cerebral echobiometry indicated an increase in area and volume of the hemispheres and cranium (P<0·001) but no changes in BMI or BVI over the gestational period studied. Doppler ultrasonography identified increases in peak systolic velocity (P=0·0188) and end diastolic velocity (P=0·0274) and decreases in resistance index (P=0·0002) and pulsatility index (P<0·001). Clinical Significance: Echoencephalography and spectral Doppler ultrasonography of the middle cerebral artery in canine foetuses might be a useful technique for prenatal care. © 2013 British Small Animal Veterinary Association.

The objective was to evaluate when the LH reserve was re-established in postpartum Nellore (Bos indicus) cows by evaluating the response of the hypothalamic-pituitary axis responsiveness to exogenous GnRH or estradiol benzoate (EB). Additionally, we tested the influence of dietary supplementation (SUPL) and calf removal (CR) on the duration of postpartum anestrus. Ninety multiparous lactating Nellore cows were randomly assigned to eight groups. The EB and GnRH groups received 1.0 mg EB (N = 7), and 50 μg lecireline (N = 16), respectively. Additional cows were given the same hormones, and subjected to either nutritional supplementation (EB-SUPL, N = 9; GnRH-SUPL, N = 16), or calf removal at 72 hours after calving (EB-CR, N = 4; GnRH-CR, N = 13). The remaining two groups were the LH (12.5 mg, N = 14) and control groups (saline, N = 11). Hormones were administered weekly from 7 (±5) days postpartum to first ovulation (detection of a CL during a weekly ultrasonographic examination). Blood samples were collected just before and 2 hours (GnRH, LH, and control groups) or 18 hours (EB groups) after hormone or saline (control) administration. Ovulation occurred as early as 15 days postpartum in the GnRH group. The mean ± SEM intervals (days) from calving to first ovulation were EB, 87.7 ± 4.2; EB-CR, 20.3 ± 1.2; EB-SUPL, 60.3 ± 3.2; GnRH, 40.4 ± 2.1; GnRH-CR, 21.0 ± 1.1; GnRH-SUPL, 26.4 ± 1.1; LH, 35.6 ± 1.1; and control, 60.9 ± 2.1. We concluded that there was sufficient LH in the pituitary gland (of Nellore cows) from the second week postpartum to induce ovulation in response to exogenous GnRH. Additionally, calf removal and nutritional supplementation reduced, by 2 to 4 weeks, the interval from calving to an LH increase and ovulation induced by GnRH or EB. © 2013.

There is species divergence in control of DNA methylation during preimplantation development. The exact pattern of methylation in the bovine embryo has not been established nor has its regulation by gender or maternal signals that regulate development such as colony stimulating factor 2 (CSF2). Using immunofluorescent labeling with anti-5-methylcytosine and embryos produced with X-chromosome sorted sperm, it was demonstrated that methylation decreased from the 2-cell stage to the 6-8 cell stage and then increased thereafter up to the blastocyst stage. In a second experiment, embryos of specific genders were produced by fertilization with X- or Y-sorted sperm. The developmental pattern was similar to the first experiment, but there was stage × gender interaction. Methylation was greater for females at the 8-cell stage but greater for males at the blastocyst stage. Treatment with CSF2 had no effect on labeling for DNA methylation in blastocysts. Methylation was lower for inner cell mass cells (i.e., cells that did not label with anti-CDX2) than for trophectoderm (CDX2-positive). The possible role for DNMT3B in developmental changes in methylation was evaluated by determining gene expression and degree of methylation. Steady-state mRNA for DNMT3B decreased from the 2-cell stage to a nadir for D 5 embryos >16 cells and then increased at the blastocyst stage. High resolution melting analysis was used to assess methylation of a CpG rich region in an intronic region of DNMT3B. Methylation percent decreased between the 6-8 cell and the blastocyst stage but there was no difference in methylation between ICM and TE. Results indicate that DNA methylation undergoes dynamic changes during the preimplantation period in a manner that is dependent upon gender and cell lineage. Developmental changes in expression of DNMT3B are indicative of a possible role in changes in methylation. Moreover, DNMT3B itself appears to be under epigenetic control by methylation. © 2013 Dobbs et al.

This study examined the effect of fast-velocity concentric isokinetic resistance training (FV) on the rate of force development (RFD) at early (<100 ms) and late phases (>100 ms) of rising muscle force. Nine men participated in a 6-week resistance training intervention for the lower body, and nine matched subjects participated as controls (CON). During concentric isokinetic (180°s-1) knee extension training, subjects were instructed to do each contraction 'as fast and forcefully as possible'. Maximal muscle strength (MVC) and RFD (0-10, 0-20, ..., 0-250 ms from the onset of contraction) were measured during maximal voluntary isometric contraction of the knee extensors (KE). There were no significant changes in MVC of KE in both groups after intervention (FV = 314·2 ± 101·1 versus 338·7 ± 88·0 N{bullet operator}m, P>0·05; CON = 293·3 ± 94·8 versus 280·0 ± 72·2 N{bullet operator}m, P>0·05). The RFD increased 39-71% at time intervals up to 90 ms from the onset of the contraction (P<0·05), whereas no change occurred at later time intervals. Similarly, relative RFD (i.e.%MVC{bullet operator}s-1) (RFDr) increased 33-56% at time intervals up to 70 ms from the onset of the contraction (P<0·05). It can be concluded that a short period of resistance training performed with concentric fast-velocity isokinetic muscle contractions is able to enhance RFD and RFDr obtained at the early phase of rising muscle force. © 2013 The Authors Clinical Physiology and Functional Imaging © 2013 Scandinavian Society of Clinical Physiology and Nuclear Medicine.

Achados acidentais de instrumentos líticos em região de terra firme junto ao rio Curuá, no médio curso da Bacia do Xingu, sugere uma ocupação disseminada por caçador-coletores pré-cerâmicos na região, contrariamente a expectativas de que a floresta tropical teria recursos alimentares insuficientes para a ocupação humana longe da várzea. Os artefatos líticos incluem pontas de projétil de lascamento cuidadoso, possivelmente relacionados a alguns artefatos do Pleistoceno final encontrados na Caverna da Pedra Pintada, em Monte Alegre. Os resíduos alimentares encontrados com os artefatos de Monte Alegre eram de uma economia de coleta de ambientes rupestres e ribeirinhos. As pontas do Xingu foram recolhidas por garimpeiros nas reias e cascalhos no leito do rio Curuá. Os garimpeiros encontraram os artefatos enquanto escavavam e peneiravam sedimentos auríferos. Tais depósitos algumas vezes também contêm remanescentes de plantas e artefatos de madeira pré-históricos, fontes de informação potencial sobre antigos habitats, subsistência e tecnologia. O grupo de pesquisa do Projeto Baixo Amazonas viajou a diversos dos sítios submersos com os garimpeiros para preparar escavações para o futuro. Em um sítio, Curupité, onde os garimpeiros encontraram uma grande ponta com pedúnculo e um arpão de madeira inteiro em 1986, a equipe utilizou equipamento de mergulho para prospectar o leito do rio e os barrancos, e mapearam a topografa com um teodolito a laser.

Os estágios iniciais de lagosta tropical Panulirus echinatus eclodiram e foram cultivados em laboratório. Fêmeas ovígeras foram capturadas em seu habitat e cuidadosamente transportadas para o laboratório. As larvas foram cultivadas em tanques de recirculação de água e alimentadas com náuplios de Artemia e gônadas mexilhão Brachydonts sp. Microalga Dunaliella viridis foi adicionada em cada tanque de cultivo na concentração de 150 x 104 cel.mL–1. Os filosomas mudaram oito vezes e o período de intermuda de cada instar foi de cerca de 7 a 10 dias. As principais mudanças morfológicas de cada apêndice foram descritas, ilustradas e comparadas com trabalhos anteriores.

Os estágios iniciais da lagosta tropical Enoplometopus antillensis Lütken, 1865 foram descritos e ilustrados a partir de espécimes cultivados em laboratório. Fêmeas ovígeras foram capturadas em seu habitat, na profundidade cerca de 15 metros e transportadas para o laboratório. As larvas foram cultivadas em tanques de água recirculante por aproximadamente 15 dias e, então transferidas para quatro aquários (capacidade 10 litros). As larvas foram alimentadas com náuplios de Artemia sp. recém eclodidos. A microalga Dunaliella viridis AUTOR foi diariamente adicionada no cultivo. As larvas mudaram sete vezes alcançando o zoea VIII. O estágio megalopa não foi obtido. O período de intermuda de cada estágio variou de cerca de oito a 12 dias. Comparações morfológicas com trabalhos anteriores são brevemente discutidas.

Este trabalho compara as mudanças morfológicas e vegetacionais ocorridas ao longo da zona costeira da Ilha de Marajó, litoral amazônico, e da planície costeira do Rio Doce, sudeste do Brasil, durante o Holoceno e Pleistoceno tardio/Holoceno, respectivamente, com foco especificamente sobre a resposta dos manguezais para as flutuações do nível do mar e mudanças climáticas, já identificadas em vários estudos ao longo da costa brasileira. Esta abordagem integra datações por radiocarbono, descrição de características sedimentares, dados de pólen, e indicadores geoquímicos orgânicos (δ13C, δ1₵N e C/N). Na planície costeira do Rio Doce entre ~47.500 e 29.400 anos cal AP, um sistema deltaico foi desenvolvido em resposta principalmente à diminuição do nível do mar. O aumento do nível do mar pós-glacial causou uma incursão marinha com invasão da zona costeira, favorecendo a evolução de um sistema estuarino/lagunar com planícies lamosas ocupadas por manguezais entre pelo menos ~7400 e ~5100 anos cal AP. Considerando a Ilha de Marajó durante o Holoceno inicial e médio (entre ~7500 e ~3200 anos cal AP) a área de manguezal aumentou nas planícies de maré lamosas com acúmulo de matéria orgânica estuarina/marinha. Provavelmente, isso foi resultado da incursão marinha causada pela elevação do nível do mar pós-glacial associada a uma subsidência tectônica da região. As condições de seca na região amazônica durante o Holoceneo inicial e médio provocou um aumento da salinidade no estuário, que contribuiu para a expansão do manguezal. Portanto, o efeito de subida do nível relativo do mar foi determinante para o estabelecimento dos manguezais na sua atual posição nas regiões norte e sudeste do Brasil. Entretanto, durante o Holoceno tardio (~3050-1880 anos cal AP) os manguezais em ambas as regiões retrairam para pequenas áreas, com algumas delas substituídas por vegetação de água doce. Isso foi causado pelo aumento da vazão dos rios associada a um período mais úmido registrado na região amazônica, enquanto que na planície costeira do Rio Doce, os manguezais encolheram em resposta a um aumento da entrada de sedimento fluvial associado a uma queda no nível relativo do mar.
